1385320 Determination of cholesterol NATIONAL RESEARCH DEVELOPMENT CORP 18 Sept 1972 [22 Sept 1971 19 June 1972] 33069/74 and 33068/74 Divided out of 1385319 Heading G1B A method of assaying for cholesterol in a liquid (e.g. serum) comprises incubating the liquid with an enzyme preparation capable of oxidizing the cholesterol into #4-cholestenone and hydrogen peroxide and determining the amount of cholesterol present by measuring the amount of hydrogen peroxide or #4-cholestenone produced or the amount of oxygen absorbed in the reaction. Preferably the enzyme is obtained from a microorganism of the group Mycobacterium rhodocrous e.g. Nocardia species NCIB 10554 or NCIB 10555 wherein the enzyme preparation has a cholesterol oxidase specific activity of at least 1 unit per 5mg of protein nitrogen and a potency of at least 10<SP>-2</SP> units/ml. The amount of hydrogen peroxide may be measured fluorimetrically or by means of a chromogenic reagent, for example by means of quadrivalent titanium and xylenol oranges or by reaction of the hydrogen peroxide with 4-aminophenazone in the presence of phenol and peroxidase, the product being measured colorimetrically. Other reagents are mentioned. The amount of oxygen taken up may be measured by means of an oxygen electrode and the amount of #<SP>4</SP>-cholestenone may be determined by its carbonyl absorption or a derivative of #4-cholestenone such as its 2, 4 diphenyl hydrazone or iso nicotinic acid hydrazone is formed and is measured colorimetrically. A kit for use in assaying the amount in a liquid comprises in association: (i) an enzyme preparation capable of oxidizing cholesterol into #4-cholestenone and hydrogen peroxide preferably as defined above and (ii) at least one reagent which is capable of being used in the determination of the amount in which hydrogen peroxide or #<SP>4</SP>-cholestenone is formed. The reagent (ii) may be a reagent which takes part in a reaction by means of which hydrogen peroxide may be determined fluorimetrically or colorimetrically and the kit may comprise additionally at least one reagent capable of being used in hydrolyzing serum for serum cholesterol assays.
