Abstract

1,222,810. Prothrombin complex. BAXTER LABORATORIES Inc. 16 Sept., 1969 [2 Oct., 1968], No. 45582/69. Heading A5B. A method of preparing a prothrombin complex comprises adjusting the pH of an aqueous suspension of Cohn Plasma Fraction III to within a range of from about 7.0 to about 7.4, separating the resulting suspension and thoroughly mixing the supernatant with tribasic calcium phosphate to adsorb the coagulation factors, thoroughly mixing the tribasic calcium phosphate adsorbed - protein precipitate with trisodium citrate to elute the coagulation factors, separating the resulting suspension and subjecting the supernatant to a succession of two cold ethanol precipitations, first at a pH of about 6.8 and a final concentration of from about 14% to about 16% ethanol at about -5‹C with retention of the supernatant, and then at a pH of about 5.2 and a final concentration of about 25% ethanol at about -7‹C with retention of the precipitate as the active prothrombin complex. After removal of the tribasic calcium phosphate precipitate, an anticoagulant such as heparin may be added to the solution.
