Abstract

Solid porous materials for use in chromatography columns comprise a porous mineral support coated directly with an adsorbed antibody layer stabilised by crosslinking. The support can be a refractory oxide (e.g. SiO2, Al2O3, MgO or TiO2) or a glass, silicate, borosilicate or kaolin. The antibody is pref. an antibody to cholera toxin, but can also be an antibody to diphtheria toxin, tetanus toxin, virus HB, influenza, virus, hepres virus, alpha-foetoprotein, gamma-globulin, albumin, etc. The materials are esp. useful for affinity-chromatographic isolation of cholera toxin, e.g. for vaccin prodn. The antibody is strongly bound to the support without the use of intermediate layers of polymers or coupling agents.
