Assembly (1) for closing a tube (3), said assembly (1) comprising: - a tube head (5) comprising a neck (7) and a lid (15) closing said neck (7) - a cap (9) comprising a punch (17) adapted to cut out said lid (15), said neck (7) and said stopper (9) being configured to make it possible to maintain said stopper (9) fixed to said neck (7) in a first position, called the waiting position in which the punch (17) is held at a distance of the lid (15) and in a second position, called the use position, in which said punch (17) pierces said lid (15), and in a third position, called the closed position, in which said neck (7 ) and said cap (9) seal said tube, said plug (9) being configured to pass from the standby position to the use position and/or from the use position to the closed position by at least one rotational movement, wherein said cap (9) and said neck (7) define a longitudinal axis ZZ, and wherein said cap (9) is configured to pass from the standby position to the use position and/or from the use in the closed position also by an axial movement along the ZZ axis, said neck (7) comprising at least one first blocking means cooperating with said stopper (9) to hold said stopper (9) in the waiting position, said neck (7) comprising at least one guide groove (41) configured to cooperate with said stopper (9) to allow a rotational movement and/or an axial movement for the passage of said stopper (9) from the waiting position to the use position and/or from the use position to the closed position, the said first blocking means(s) being protrusions (37a, 37b, 37c) of the edges of the said groove (41) extending towards the inside of the groove (41), said groove (41) having successive guide portions.
