Conventional absorption methods are used to detect absorbing substances in liquids, gases and solids (measuring volume). Radiation of a defined wavelength is coupled into the measurement volume. On their way through the Meßvolu men, the coupled radiation is weakened by absorbing substances. After a defined distance, the coupling radiation is coupled out again and directed to an optoelectronic receiver which registers the weakened intensity I. The quotient of the weakened and non-weakened intensity I 0 is the transmission T:
T = I/I0 = exp (-αTx) (1)T = I / I 0 = exp (-α T x) (1)
Optik. Berlin - New York, Walter de Gruyter, 1993, S. 244-246 und SCHMIDT, W.: Optische Spektroskopie. Weinheim-New York-Basel-Cambridge-Tokyo, VCH Verlagsgesellschaft, 1994, S. 117-121)This law by Bouguer-Beer-Lambert describes the relationship between transmission and the total absorption coefficient α T (for the sake of simplicity, the scatter has been neglected here). The term x is the path that the coupling radiation travels in the measurement volume. (BERGMANN and SCHAEFER: Textbook of Experimental Physics. Optics. Berlin - New York, Walter de Gruyter, 1993, pp. 244-246 and SCHMIDT, W .: Optical Spectroscopy. Weinheim-New York-Basel-Cambridge-Tokyo, VCH Verlaggesellschaft, 1994, pp. 117-121)
Berlin-Heidelberg-New York, Springer Verlag, 1992, S. 178). For measuring volumes with very low optical densities (e.g. gases), the distance of the Coupled radiation increased in the measurement volume in order to obtain evaluable signals (BAUMBACH, G .: Air pollution control. Berlin-Heidelberg-New York, Springer Verlag, 1992, p. 178). Long distances can be achieved, for example, with the help of reflective elements will. DE 41 04 316 A1 presents an internally mirrored spherical cell, in which the coupled radiation is reflected back and forth several times and then coupled out again and aimed at a recipient. DE 41 24 545 A1 describes a gas absorption cell wrote.
In DD 3 01 863 A7, DE 42 37 227 A1 und MITTENZWEY, K.-H., J. RAUCHFUß, G. SINN, H.-D. KRONFELDT: A new fluorescence technique to measure the total absorption coefficient in fluids. Fres. J. Anal. In DD 3 01 863 A7, DE 42 37 227 A1 and MITTENZWEY, K.-H., J. RAUCHFUß, G. SINN, H.-D. KRONFELDT: A new fluorescence technique to measure the total absorption coefficient in fluids. Fres. J. Anal. Chem., 354 (1996) 159-162 describes a method for determining the total absorption is proposed, in which not that after passing through a defined Path weakened coupling radiation is measured, but by the coupling interaction radiation (fluorescence and scattering). The special It is a feature of the fact that the coupled radiation due to long distances from the measurement volume is almost completely absorbed.
klassischen Sinne gestreute Photonen.
The reflectance R is the diffuse reflection of radiation from matter (measuring volume). It is a measure of the intensity of the photons reflected against the direction of incidence. In the classic sense, these are scattered photons. The remission is determined by the scattering capacity (scattering coefficient β) and absorption capacity (total absorption coefficient α T ,) of the measuring volume. For the sake of simplicity, the absorption will dominate in the following. The theory of Kubelka and Munk is used to describe the remission mathematically. In the case of an infinitely extended measurement volume (e.g. deep water), the remission is proportional to the quotient of the scattering coefficient and absorption coefficient,
Rs ~ β/αT (2).Rs ~ β / α T (2).
If fluorescence is also generated by the radiation incident in the measurement volume, then the remission is determined in the broader sense not only by the scattering but also by the fluorescence ability, which is determined by the product of the fluorescence quantum yield Q F and the absorption coefficient of the fluorophores α F of the measurement volume (Q F α F ) is characterized. The fluorescence contribution to the remission of large measurement volumes is determined by the quotient
RF ~ QFαF(λE)/[αT(λE)+(αT(λF)] (3)
RF ~ Q F α F (λ E ) / [α T (λ E ) + (α T (λ F )] (3)
controlled, where λ E and λ F are the wavelengths of the incident radiation and fluorescence. In many cases of transmitting measurement volumes, the absorption at the wavelength of the incident radiation is greater than the absorption at the fluorescence wavelength (e.g. in the case of eutrophicated surface waters). Then (3) merges into (4):
RF ∼ QFαF(λE)/αT(λE) (4)R F ∼ Q F α F (λ E ) / α T (λ E ) (4)
The formulas (2) and (4) are characterized by the same mathematical structure. The In both cases, reflectance is proportional to the scattering and fluorescence ability and on the other hand, inversely proportional to the total absorption.
The reflectance spectroscopy is e.g. B is fundamental to remote sensing and is used in both optically very dense as well as applied to transmitting measurement volumes. examples for The first case is remission measurements on vegetation (leaves or needles) around which determine physiological condition or measurements on soils to determine example wise moisture and structure.
(COLWELL, R. N.: Manual of remote sensing. Falls Church, The Sheridan Press, 1983, S. 1415-1423 u. 2136- 2143, SCHMIDT, W.: Optische Spektroskopie. Weinheim-New York-Basel-Cambridge-Tokyo, VCH Verlagsgesellschaft, 1994, S. 316-322 und MÜTZE, K.: ABC der Optik. Brockhaus Verlag Leipzig, 1961, S 751-752).The second case of the transmitting measuring volumes include atmosphere, water and Oceans. Comparatively simple relationships are given when the incident Radiation (global radiation, lidar) can run dead in the measurement volume, d. H. that in the example of Water bodies the incident radiation does not reach the water floor. (COLWELL, R. N .: Manual of remote sensing. Falls Church, The Sheridan Press, 1983, pp. 1415-1423 u. 2136- 2143, SCHMIDT, W .: Optical Spectroscopy. Weinheim-New York-Basel-Cambridge-Tokyo, VCH Verlagsgesellschaft, 1994, pp. 316-322 and MÜTZE, K .: ABC der Optik. Brockhaus Verlag Leipzig, 1961, S 751-752).
A major advantage of classic absorption spectrometry compared to the Fluo Resence and scattering is that the injected radiation falls directly on the receiver and thus significantly more photons are available for the measurement. High signal / noise Relationships are the result. Radiation sources with low photon flux densities and simple Semiconductor receivers can be used. The technical expenditure on equipment is relatively small.
A major disadvantage of classic absorption spectrometry is that it is relatively small Sensitivity due to the exponential relationship between the measurement volume weakened coupling radiation and absorption coefficient.
A major advantage of reflectance spectroscopy is that the relationship between is inversely proportional to the total absorption coefficient and the reflectance (see Formulas (2) - (4)). This makes the remission more sensitive than the classic absorption spectra trometry. The remission also contains information about the scatter and fluorescence ability of the measuring volume.
A disadvantage of remission spectroscopy, however, is that the relationship between R and α T , β, Q F α F is ambiguous. This means that an exact separation of the scattering, fluorescence and absorption ability is difficult and in many cases not possible. Furthermore, the use of remission for a sensitive determination of the absorption capacity of transmitting measurement volumes is tied to extended measurement volumes, since the radiation runs dead in the measurement volume only after longer distances (e.g. 10-230 cm with absorption coefficients of 1-23m typical for surface waters -1 ). This is not possible for samples with smaller layer thicknesses (e.g. classic cuvettes). In addition, when light beams with a finite cross-section are irradiated into the extended measurement volume (e.g. lidar), the photometric distance law has a disruptive effect on the signal-to-noise ratio.
Proceeding from this, the task arises, the sensitivity of the absorption spectrometry increase and at the same time eliminate the ambiguity of reflectance spectroscopy, the advantages such as the large signal-to-noise ratio in the absorption and the high Sensitivity of remission, should be maintained as well, absorption and remission to determine synchronously.
The object is achieved by the features of claims 1 and 11.
Vorteilhafte Ausgestaltungen sind in den Unteransprüchen angegeben.Advantageous refinements are specified in the subclaims.
Claim 1 is explained. Radiation of a defined wavelength is coupled into the measuring volume to be examined, which is transmissive. The measuring volume is located between two opposing mirrors. The mirrors are designed in such a way that, due to a sufficiently high number of reflections, the path of the radiation coupled in via the coupling mirror is so long that it can be completely absorbed in the measurement volume. Complete absorption is a prerequisite for the occurrence of a remission according to the formulas given in the prior art (term here: saturated long-path remission). The multiple reflection eliminates the need for extended measurement volumes. In addition, the photometric distance law no longer interferes with the signal-to-noise ratio. The saturated long-path remission is measured with a photoelectronic receiver arranged on the coupling mirror and aligned with the measurement volume in the usual reflectance measurement geometry, that is to say at the rear. The measurement signal is described according to formula ( 2 ) in the case of scattering and in the case of fluorescence according to formula (4).
Synchron dazu erfolgt ein zweiter wesentlicher Meßvorgang. Der Einkoppelspiegel (oder auch
der Gegenspiegel) ist teildurchlässig, z. B. 5% Transmission und 95% Reflektivität. Folglich
A second essential measuring process takes place synchronously with this. The coupling mirror (or also the counter mirror) is partially transparent, e.g. B. 5% transmission and 95% reflectivity. Consequently, after each reflection or after each revolution, part of the coupling radiation transmitted by the measuring volume passes through the coupling mirror and arrives at a second receiver arranged immediately behind the coupling mirror. If the fluorescence and scattering photons passing through the partially transparent mirror are neglected, the intensity I Tr of the transmitted radiation is approximated by the following formula:
ITr ~ m/αT (5)I Tr ~ m / α T (5)
The term m is a constant that is characteristic and known for the permeability of the coupling mirror. The total absorption coefficient α T can thus be determined directly from (5). Compared to classic absorption spectrometry (Lambert-Beer Exponential Law) (5) is characterized by a higher sensitivity, which leads to deeper detection limits and higher accuracies. With increasing α T , I Tr decreases. This makes sense, since with increasing α T the mean path length of the injected radiation decreases until it is almost completely absorbed in the measuring volume and thus the number of reflections or revolutions decreases. This also reduces the intensity I Tr of the coupling radiation passing through the partially transparent mirror. In addition, the intensity I Tr is also determined by the mirror constant m. The larger m, ie the smaller the reflectivity or the greater the permeability of the coupling mirror, the higher I Tr .
By inserting α T in the formulas (2) and (4), the scattering and fluorescence ß and Q F α F can thus be determined indirectly, taking into account claim 2 (see below). The ambiguity of classic reflectance spectroscopy is eliminated by the combination with the absorption spectroscopy presented above. For many applications it will be sufficient to determine only the absorption capacity of the measurement volume according to claim 1 via I Tr . In these cases, the remission measurement can be omitted, which leads to a lower expenditure on equipment. The same applies vice versa if only the scattering and fluorescence ability is required and the determination of the absorption ability is therefore not necessary.
If the reflectance R is significantly influenced by both fluorescence and scatter flows, then a separation between fluorescence and scattering with the help of filters Suppression of fluorescence or scattering can be carried out (claim 2).
Claim 3 solves a problem that occurs with scattering or fluorescent measurement volumes when determining the absorption capacity by means of I Tr (formula (5)). The receiver located immediately behind the coupling mirror not only registers the coupling radiation transmitted by the measuring volume, but also scatter and fluorescence photons of the measuring volume. The overall signal is therefore larger. Consequently, smaller total absorption coefficients are simulated according to formula (5). A correction is therefore necessary. For this purpose, an intensity is subtracted from the total intensity of the photons transmitted through the partially transparent mirror (direct coupling radiation plus scattering or fluorescence), which is proportional to the saturated long-term remission (scattering or fluorescence). The proportionality factor is, among other things, a function of the reflectivity and thus the transmission of the partially transparent mirror.
According to claim 4, the scattering and fluorescence ability of the measurement volume can also be direct be determined. For this purpose, the remission is measured that from the part of the meas lumens, which is located directly behind the coupling point (name here: linear short-path remission). In this small part of the measuring volume immediately behind the Coupling can therefore only travel a short distance to the injected radiation. This leads to a complete absorption of the coupling radiation in this part of the Measurement volume does not occur. The connection between remission and litter and fluo Resistance is linear and independent of the total absorption coefficient. Litter and fluorescence ability can be directly with the help of the linear short-path remission be determined. By the quotient of this linear short-path remission and the saturated long-term remission also indirectly results in the absorption capacity or the total absorption coefficient. This is another independent factor Comparison with the absorption coefficient determined via formula (5) is available here too in the case of scattering or fluorescent measurement volumes for a correction is useful.
Die Absorptionsfähigkeit wird also nicht nach Formel (5) ermittelt, sondern klassisch nach dem Lambert-Beerschen Gesetz (Formel (1)).Claim 5 has a favorable effect on optically thin measurement volumes in which none Saturated long-term remissions can be generated because those are almost full constant absorption of the coupling radiation necessary path lengths not with the help of simple multiple reflection devices can be generated. The optically thin Measurement volumes generated reflectance is due to a linear relationship to the scattering and Fluorescence capability is characterized and is un of the total absorption coefficient dependent (term here: linear long-term remission). So that the litter and Fluorescence ability can be determined directly from the linear long-term remission. Synchronous for this purpose, the absorption capacity or the total absorption coefficient is determined by the radiation coupled into the measurement volume after a defined number of reflections or revolutions coupled out of the multiple reflection device and onto a receiver is judged. The absorption capacity is therefore not determined according to formula (5), but rather classic according to the Lambert-Beer law (formula (1)).
For the generation of saturated remissions and optimization of the signal / noise ratio in the case of optically thin measurement volumes, artificial absorbers (e.g. gray filters) are added subject reflection device used (claim 6). This leads to a defined shortening the path of the injected radiation to its complete absorption. As a measure of the path length to be set is the reciprocal of the smallest possible total absorption coefficients of the measurement volume.
This has the advantage, on the one hand, of the technical expenditure for producing more subject reflection units is minimized and secondly regardless of the optical thickness of the measurement volume always saturated remissions with maximum signal / noise ratio nis can be generated.
Claim 7 represents a favorable specification of claim 6 by the transmit coupling mirror acts as an artificial absorber itself. The greater the permeability of this partially transparent mirror, the shorter the average path length of the injected radiation up to its complete absorption in the measuring volume. The mean path length is thus influenced by the permeability. Because the mean path length of the coupled radiation in the measurement volume inversely proportional to the total absorption coefficients, can from the knowledge of the smallest possible absorption coefficient the largest possible path length to be investigated and from it adjusted the permeability of the partially permeable layer of the coupling mirror will.
The claim 8 represents a solution that with optically denser (but still transmit turing) measurement volumes is applied. The number of rounds to complete Absorption of the incident radiation is small with denser measuring volumes. Determining the Absorbance with high sensitivity over that through the coupling mirror Coupling radiation entering and transmitted by the measuring volume would not be according to formula (5) more is possible. Therefore, it is proposed that the absorption capacity according to the classic Determine method by using either a receiver instead of the counter mirror direct measurement of the coupling radiation weakened by the measuring volume or the coupling mirror is removed and the receiver located immediately behind it Measurement of the weakened coupling intensity can be used. The scatter and fluorescence ability is determined from the linear short-path remission.
According to claim 9, for example when coupling very short pulses of less than 1 ns, the saturated long-path remission and the linear short-path remission time measured resolved. Their latitudes are determined. The reciprocal of the difference The latitude is a measure of the absorption capacity. This method has the advantage that different device-specific properties such as irradiation intensity, filter transmission, Aperture, radiation and receiver area have no disruptive effects. The direct one Comparison between saturated long-term remission and linear short-term remission is possible without extensive correction.
Based on this, the content of claim 10 is that the remission once in a short time window (measurement of the linear short-path remission) and the other in a long time window (measurement of saturated long-term remission) is registered in an integral manner will. Short distances in the measuring volume to be examined become short and long Paths traveled in long times by the coupled radiation. That is, are short times for linear short-path remission and long times for saturated long-path remission typical. The quotient of the short time window and long time window is a measure of that Absorbance of the measuring volume. This has the advantage that both remissions without great effort can be measured purely electronically, resulting in a robust construction and easy handling. The widths to be set Time windows are made depending on the measurement volume to be examined. At optically denser measurement volumes, the time windows are shorter than those with optically thinner ones Measuring volumes set. This leads to the correct measurement of saturated long-term remissions and linear short-path remissions as well as to optimize the signal-to-noise ratios.
Fig. 1 shows an example of a spectrometer that is suitable for carrying out the described method. The essential features are documented in claims 11-20.
Module A ( 1 ) is equipped with light emitting diodes LED ( 5 ), photodiodes as receivers ( 9 , 10 ) and a lock-in amplifier ( 4 ). Instead of the LED, other radiation sources can also be arranged. The radiation from the LED is divergent via optical fibers ( 6 ) into the measuring volume ( 11 ) to be examined. The coupling takes place over the edge of the coupling mirror ( 7 ), the optical waveguide being slightly inclined against the mirror normal and the exit surface of the optical waveguide lying approximately in the plane of the coupling mirror. This mirror is flat. The mirror ( 8 ) is located in module B ( 2 ). This mirror is a concave mirror. The diameter of the concave mirror is larger than that of the opposite plane mirror. In contrast to the coupling mirror, the counter mirror is movably mounted for adjustment purposes. When adjusting, the distance between the two mirrors is particularly important, since this has a significant influence on the number of revolutions of the injected radiation. Coupling and counter mirrors lie on an optical axis. In between is the measuring volume to be examined. Concave and plane mirrors are used for multiple reflection of the coupled LED radiation. This is necessary in order to achieve the long path lengths for the generation of saturated signals. Modules A and B are rigidly connected to each other after adjustment ( 12 ) and can easily be designed as a submersible probe, as a device for measuring on a free liquid jet or with a classic cuvette. The photodiode ( 9 ) is directly behind the transmitting coupling mirror ( 7 ) angeord net. The diameter of the mirror is similar to that of the photosensitive surface of the photodiode. An optics between mirror and photodiode is not necessary. The transmission of the mirror is z. B. 5%. If real absorption is neglected, its reflectivity is 95%. This ensures that after each reflection only a small part of the coupling radiation reaches the receiver through the mirror and thus the coupling radiation is available for further runs through the measurement volume. In the case of optically very thin measurement volumes, this mirror can be designed as a defined absorber in such a way that an almost complete absorption of the injected radiation is always achieved. Depending on the application, an edge or bandpass filter can be placed between the mirror and the photodiode using a filter module. The photodiode ( 10 ) is aligned with the measurement volume and measures the interaction between incident LED radiation and measurement volume (remission). With sufficiently small mirror distances, optics in front of this photodiode to increase the number of photons are not necessary. Depending on the application, an edge or bandpass filter can also be placed in front of the photodiode using a filter module. LED and receiver are coupled with a lock-in amplifier. The LEDs are current modulated with a defined frequency, the lock-in amplifies the useful signal. Module C ( 3 ) contains the controller and a standard interface. The power consumption is low due to the use of LEDs and semiconductor receivers.
Alternatively, the coupling can also take place via a single optical waveguide by spectrally split radiation is coupled into the optical waveguide at different times. Of In addition, the radiation can also be via an optical coupling instead of the optical waveguide consisting of lenses or mirrors are brought into the measuring volume. In addition, the Coupling via an opening in the coupling mirror. The coupling can also be done with parallel radiation (e.g. laser).
