Experiment 2
Zebra fish anti-wrinkle efficacy test
The method comprises the following steps: standard operating procedures for evaluation of anti-wrinkle efficacy of Zebra fish
System and sample size: test system: wild type AB strain zebrafish.
The fish age of the zebra fish is as follows: 4 days after fertilization (4 dpf).
Amount of each experimental sample: 30 tails (three biological replicates, N = 3)
Adult fish feeding and breeding method: the breeding and reproduction method meets the requirements of the international AAALAC certification (certification number: 001458) according to the laboratory standard of the company.
The principle method comprises the following steps: skin growth, repair, nutrition, elasticity and tension are all related to collagen, and its loss causes skin smoothness to decrease, resulting in wrinkles. In quadrupeds, type I collagen is a trimer, composed primarily of two α 1 chains and one α 2 chain, encoded by the col1a1a and col1a2 genes, respectively, and performs collagen-related biological functions in connective tissues and bones. There are three types of collagen type I genes in zebrafish, encoding col1a1a, col1a1b and col1a2 of the α 1 (I), α 2 (I) and α 3 (I) chains, respectively. Therefore, whether the sample has the anti-wrinkle effect can be shown by detecting the relative expression quantity of the col1a1a or (and) col1a1b or (and) col1a2 gene.
In the experiment, the anti-wrinkle effect of the firming anti-wrinkle emulsion in application examples 1-9 is detected by detecting the relative expression quantity of the col1a1a gene, and the detection result is detailed in table 5.
Experiment 3
Zebra fish tightening effect experiment
The method comprises the following steps: standard operating procedures for Zebra fish tightening efficacy evaluation experiment

The principle method comprises the following steps: elastin is the main component of elastic fiber in skin tissues, can provide structural support for skin, and prevents skin from aging and relaxation. The elastin is composed of two types of short peptide segments which are arranged alternately, eln1 and eln2 are responsible for coding different peptide segments of elastin, and the two genes are jointly responsible for regulating and controlling the expression level of elastin, so that the skin is compact and elastic. Zebrafish have an elastin (eln 1, eln 2) gene similar to that of humans. Therefore, whether the sample has the compact efficacy can be indicated by detecting the relative expression amount of the eln1 or (and) eln2 gene.
In the experiment, the tightening effect of the tightening anti-wrinkle emulsion applied in the examples 1-9 is detected by detecting the relative expression quantity of the eln1 gene, and the detection result is detailed in table 5.

As can be seen from table 5, the relative expression amount of the compact anti-wrinkle emulsion col1a1a gene prepared in application example 3 is 2.36, and the relative expression amount of eln1 gene is 2.56, which is significantly increased compared to application example 7, showing that in application example 3, the anti-wrinkle and compact effects added by adding acetyl tetrapeptide-2 and dipeptide diaminobutyrylbenzylamide diacetate on the basis of the ginseng root extract are significantly improved compared to application example 7.
From table 5 in combination with the comparison of the experimental results of fig. 1-2, the significant increase of the relative expression level of col1a1a gene and the relative expression level of eln1 gene measured by the firming and anti-wrinkle emulsion of application example 3 compared with that of application examples 8-9 can be obtained, which indicates that the relative expression level of col1a1a gene and the relative expression level of eln1 gene are reduced significantly after acetyl tetrapeptide-9 is used to replace acetyl tetrapeptide-2 or acetyl hexapeptide-8 is used to replace dipeptide diaminobutyryl benzylamide diacetate, and proves that the ginseng extract, acetyl tetrapeptide-2 and dipeptide diaminobutyryl benzylamide diacetate have better synergistic anti-wrinkle and synergistic firming effects.

Claims (9)

1. A firming and anti-wrinkle composition, comprising: the paint comprises the following components in parts by mass: 0.2-0.5 part of ginseng root extract, 20.001-0.005 part of acetyl tetrapeptide and 0.001-0.005 part of dipeptide diaminobutyrylbenzylamide diacetate.
2. The tightening anti-wrinkle composition according to claim 1, wherein: the firming anti-wrinkle composition further comprises 0.0005-0.003 part by mass of palmitoyl pentapeptide-4.
3. The tightening anti-wrinkle composition according to claim 1, characterized in that: the firming and anti-wrinkle composition also comprises one or more of rosa damascena flower water and centella asiatica leaf extract.
4. The tightening anti-wrinkle composition according to claim 3, wherein: the firming and anti-wrinkle composition further comprises 0.05-0.3 part by mass of rosa damascena flower water and 0.05-0.3 part by mass of centella asiatica leaf extract.
5. A method of preparing the firming and anti-wrinkle composition as claimed in any one of claims 1 to 3, wherein: mixing the components, and stirring uniformly to obtain the firming and anti-wrinkle composition.
6. Use of the firming anti-wrinkle composition according to any one of claims 1 to 3, characterized in that: the firming anti-wrinkle composition is added into a skin care product, and the weight percentage of the firming anti-wrinkle composition is 0.3-1.6%.
7. The firming and anti-wrinkle skin lotion is characterized in that: the composite material comprises the following components in percentage by mass:
the tightening anti-wrinkle composition as claimed in any one of claims 1 to 3 in an amount of 0.3 to 1.6%
3-8% of glycerol
1 to 5 percent of butanediol
EDTA disodium 0.01-0.05%
Sodium hyaluronate 0.03-0.07%

0.3 to 0.6 percent of p-hydroxyacetophenone
0.3 to 0.7 percent of 1, 2-hexanediol
