The present invention relates to an inhaler for the gaseous or aerosol administration of a liquid active substance or active substance mixture, in particular of nicotine, a method for operating the inhaler and container for use with the inhaler.
In January 2005, the Health Committee of the European Parliament issued a public statement welcoming the Commission's intention to continue its efforts to ban tobacco smoke in confined spaces. The Committee decided to classify tobacco smoke in the environment as a Category 1 carcinogen as soon as possible. Although the opinion was adopted by a narrow majority, it shows clearly that in the context of the improvement of indoor air quality and measures against passive smoking, it is likely that non-smoker protection laws, such as those in Ireland some time ago and now in Italy Came into force in other EU Member States.
In some US states, legislation on tobacco consumption is even more restrictive. After a smoke-free transatlantic flight, a nicotine-starved US traveler expects smoking bans in virtually all public buildings and offices, restaurants, bars and even some outdoors. At least since non-smoking leases or intolerant neighbors even limit smoking in the privacy, the need for smokeless tobacco products has become a priority.
In addition to nicotine patches, nicotine gums and nasal sprays, nicotine inhalers are currently available on the market. As with smoking a cigar or cigarette while a plastic tube is guided with a mouthpiece to the mouth and sucked on it. A small amount of nicotine is desorbed from a cartridge of porous material inside the tube containing about 10 mg of nicotine by vigorous suction. Usually, a lower amount of nicotine is inhaled per puff than in a conventional puff on a cigarette, so that much more often and more vigorously must be sucked in to compensate for a normal pull on the cigarette. The dosage of nicotine is inaccurate and the cartridges tend to dry out.
The scope of these nicotine-containing products is clearly in the smoking cessation and they usually require the medical prescription.
Since 2004, a product has been advertised under the brand NicStic <(R)>, but not yet marketed, which is essentially a further development of the known nicotine inhalers. It is preheated in this case, the sucked air to achieve according to advertising claims, a close approximation to the normal smoking. The classic cigarette shape, the usual filter mouthpiece, the pleasant feeling of warmth resulting from the inhalation and the tobacco taste by supplying a variety of flavors should be retained. A disposable filter is preceded by a cartridge of porous material serving as a flavor and nicotine depot. The filter with the depot can be inserted in a cylindrical reusable part, so that the NicStic <(R)> corresponds in shape and dimensions of a conventional cigarette.
The reusable part is made of plastic and has an internal passive heating element that must be heated via an external charging station before each use. For heating, the reusable part is plugged into a special plug-in connection at the external charging station, and the heating element is brought to the desired operating temperature. By sucking heavily on the filter, an air flow is passed along the heating element and heated, before the nicotine depot again desorbs nicotine and flavorings. With each draft, the passive heating element is further cooled so that the temperature of the air taken in by the user decreases from train to train.
The dosage of nicotine is therefore even more inaccurate than in the inhalers described above, and the acceptance of the device among the users will certainly not be increased by the compulsion to carry a charging station and the heating before use.
In US-A-4 947 874 it has already been proposed to integrate the power supply into the cigarette replacement device. The voltage source is a conventional 9V battery, from which a highly porous heating element is fed. The heating element is impregnated with the nicotine mixture to be evaporated and, on heating, emits an aerosol mixture which is sucked in by the user. The battery is housed in a housing to which a replaceable disposable mouthpiece can be attached. The mouthpiece essentially comprises a filter and an upstream tobacco portion and the heating element with the solution to be evaporated. After 6 to 10 strokes, the reservoir is depleted and the mouthpiece with the heating element must be discarded.
An apparatus in which the heating element is usable several times is described in US-A-5 666 977. A single resistance heating element is powered by an internal voltage source. The liquid mixture to be evaporated is stored in a reservoir and is fed via a supply line to a spray head. The spray head works with well-known inkjet printing technology and produces fine droplet jets from the medium to be evaporated in up to 100 nozzles. These jets are sprayed onto the 15 to 25 mm 2 heating element, so that between 1 and 10 mg of liquid are evaporated to between 300 and 450 ° C. per heating element.
Although the device according to US-A-5 666 977 is independent of an external power supply and allows the heating element to be used multiple times and to dose more precisely the amount of aerosol formed, the energy consumption during operation is very high and would be due to the complicated structure the device probably synonymous very expensive, if it came on the market.
It is therefore an object of the present invention to provide an inhaler and a method for gaseous or aerosol administration of a liquid active ingredient or active ingredient mixture available, which do not have the disadvantages of the products available on the market and the known methods. It is also the object of the present invention to provide a drug container for use with the inhaler, which is preferably designed as a disposable element and contains the liquid active substance or the active substance mixture for gaseous or aerosol administration.
These objects are achieved by an inhaler, a method and a drug container according to the characterizing features of claims 1, 15 and 17.
Further embodiments will become apparent from the dependent claims.

Fig. 1 shows an inhaler (1) for gaseous or aerosol administration of a liquid active ingredient or active ingredient mixture, in particular of nicotine, according to a preferred embodiment of the invention in the assembled state. In Fig. 2, the inhaler 1 is an exploded view in which a mouthpiece 2 is withdrawn from a front end of a housing 4 and a bottom lid 6 from a rear end of the housing. In the exploded view of Fig. 3, a container 3 is pulled from the front end of the housing 4 and a battery 5 from the rear end of the housing 5. Two air apertures 7, 7 can be inserted into corresponding receiving openings 63, 63. In the assembled state, openings 71, 71 that penetrate the air apertures 7, 7 define the air inlet openings in the inhaler 1.
At the opposite end of the inhaler 1, an opening 21 in a tip 20 of the mouthpiece 2 defines an outlet opening. Between the front opening 21 and the rear inlet openings 71 there is a communicating connection which allows a user of the inhaler 1 to suck air through the device. Since the remaining parts of the device, which form the outer surface are preferably approximately gas-tight in the assembled state and are also detachably connected to each other, the volume of the sucked through the device air flow by the strength of the suction and the size of the inlet openings 71 can be determined. It is advantageous that in the entire inhaler 1, the air flow is defined with a plurality of apertures and / or openings.
The defined air flow ensures that the vaporized active substance is entrained, whereby the mixing ratio air / active substance can be defined. The size of the inlet openings 71 mainly define the air supply and thus suction resistance and the mixing ratio between air and active substance.
In Fig. 7 a equipped with a Mikroheizplatte 11 support plate 10 is shown, which represents the actual heart of the inhaler 1. The Mikroheizplatte, abbreviated hereafter also as MHP, evaporated inside the inhaler 1 extremely energy efficient a drug solution, or transferred this into an aerosol form. Micro hotplates are small hotplate membranes, typically made of silicon. In the illustrated embodiment, they preferably have a diameter of about 500 * 500 [micro] m and a thickness of about 20 [micro] m. In order to keep active substances or solutions with low surface tension or those which tend to spread on the MHP on the MHP, according to a preferred embodiment, a small amount of silica gel (SiO 2 in powder form) is applied to the MHP.
Nicotine is easily bound on such treated MHPs and can be well vaporized.
Their small thermal mass allows the membrane in a short time (<0.1 s) to high temperatures (> 250 ° C) to bring. Heating capacities of approx. 100 mW or more are possible. With the illustrated MHP used in the experiments, temperatures between 170 and 310 ° C. can be achieved at a supply voltage of 5.5 V. The thermal resistance [eta] 0 [deg.] C / mW] in a preferred embodiment is 5.8 and the time constant is 10 ms. These values are valid for air. It is possible to apply liquids to the MHP. This reduces the thermal resistance and increases the time constant.
Micro heating plates, which are advantageously used in the inventive inhalers, are available, for example, from AppliedSensor GmbH, D-72770 Reutlingen. In functional samples MHPs of the type MOS sensors AS-MLC were successfully used.
The use of the micro-hot plates allows, thanks to very high evaporation temperatures of 170 to 310 ° C., preferably 250 to 280 ° C., more preferably about 270 ° C., the active substances or mixtures of active ingredients in the smallest amount To convert area and in the finest dosage in a gaseous or aerosol-like state.
At a power of MHP 11 of 100 mW, it takes about 20 to 30 seconds until the 100 nl of nicotine are completely evaporated. If the temperature is lowered by sucking in the air, the nicotine begins to condense again under 35 ° C. The condensation does not cause any great problems, since the steam generated after evaporation reaches the smoker quickly enough. The nicotine can also form a stable aerosol during evaporation, which in turn can be led to the user with low losses. Smaller amounts of the vapor or aerosol may condense on the cooler surfaces of retainer 3 or mouthpiece 2.
Since the holding element is usually disposed of after use and the mouthpiece is very easy to clean, this condensation leads to no adverse taste consequences or incorrect dosage even after prolonged use. Assuming an MHP efficiency of 50%, a heating energy of about 250 mJ is needed to vaporize the amount of nicotine, which is about the same as a conventional cigarette. At a preferred power of 100mW and a voltage of 5V, 20mA of current is required per train for 2.5 seconds. The MHPs according to the invention are selected with a power of 10 to 300 mW, preferably of 50 to 100 mW. If a plurality of MHPs are arranged in an array, then the power can be lowered even further.
If the device 1 with conventional batteries, such as those used for cameras, operated with a capacity of 500-2000 mAh, so more than 30 000 heating cycles can be performed. This not only makes the operation of the device very cheap, but it gives the user a maximum of mobility and independence from external power sources.
The MHPs are preferably operated pulsed, with a frequency of 1 to 100 Hz, preferably of 10 Hz has proven.
If other active ingredients are vaporized as nicotine, then permanent operation is possible in place of the pulsed operation.
The electrical lines are, as well as the control electronics, omitted throughout the figures.
From Figs. 3 and 2 it is clear how in unscrewed mouthpiece 2 of the container 3, which is formed in the embodiment shown by a substantially cylindrical support member 3 with a capillary 34, in a corresponding cylindrical receiving space 44 in the housing 4th can be inserted. A positioning pin 43 on the housing leading edge 44 can be used to position the holding element 3 in a defined angular position in the housing 4 in cooperation with a positioning groove 32. The holding element 3 has on its front side a circumferential flange 31, and the positioning groove 32 is preferably mounted in this flange. In the fully inserted state, the retaining element rests on the support plate 10.
A retaining element 3 according to a preferred embodiment of the invention is shown in detail in FIGS. 5 a to 5 f. The cylindrical base body 30 is penetrated lengthwise by six air channels 36. These establish a communicating connection between an evaporation space 38 and a front end face 35 of the holding element 3.
The evaporation chamber 38 is laterally delimited by Umkragungselement 33, so that it surrounds the Mikroheizplatte 11 in the assembled state and seals and together with the support plate 10 of the MHP 11, the evaporation chamber 38 defined in the Mikroheizplatte 11 and spaced from the rear end of Capillary 34 come to rest. The air inlet to and the air outlet from the evaporation chamber is ensured by the ventilation openings in the support plate 10 and the air channels 36 in the holding element.
Centered between the air channels 36, a cylinder chamber 37 opens at the end face 35 of the retaining element. Starting from the bottom of the cylinder chamber 37, a capillary 34 passes through the retaining element 3 and protrudes with its rear end into the evaporation chamber 38. The capillary 34 is arranged eccentrically and axially parallel in the retaining element 3. It is clear from the detail enlargement of FIG. 4c and FIG. 8 that this positioning of the capillary 34 results in that its rear end in use does not come to lie exactly over the center of the MHP. Retaining element, length of the capillary and the position of the MHP in the housing are coordinated so that the end of the capillary comes to lie in the assembled state with a distance D of preferably 0.2 mm above the MHP.
The eccentric arrangement of the capillary serves as an additional protective measure for the MHP as it prevents the capillary from being pushed directly onto the sensitive center of the MHP. The capillary 34 represents the depot for the active substance or the active substance mixture to be vaporized. In the embodiment shown, the inner diameter of the capillary is 0.1-2 mm, preferably 0.52 mm. It has a volume of 0.1 [micro] l, which is equivalent to the nicotine dose of a light cigarette, up to 40 [micro] l, which corresponds to a whole pack of strong cigarettes. Preferably, it has a volume of 3 [micro] l, so that it is easy to deliver 1 [micro] l nicotine, which corresponds to 1 mg nicotine and thus the maximum permissible value for a cigarette.
Due to the capillary action, the nicotine or the tobacco flavor solution to be vaporized is sucked into the capillary, which is preferably a plastic-coated fused silica capillary. The capillary may also be made of stainless steel, PEEK, Teflon or a molded or extruded plastic material.
The air flow does not necessarily have to go through the retaining element. In the embodiment, as disclosed in the figures and the above description, an axial arrangement is chosen. According to a further preferred embodiment, the capillary can also be arranged parallel to the MHP surface, and the air flow is conducted past the holding element.
After the holding element 3 is inserted into the housing 4, the mouthpiece 2 is screwed or plugged. From Figs. 4 and 8 it can be seen that while a piston 23 of the mouthpiece 2 is pushed into the cylinder chamber. The piston is slightly sealed against the inner wall of the cylinder chamber, so that in the cylinder chamber, a pressure is built up, which presses the capillary in the active ingredient or the drug solution from the capillary to the MHP. The mechanical release of the drug solution is extremely energy efficient.
The pressure buildup does not necessarily have to take place via the piston 23 of the mouthpiece 2. It can be used according to a further embodiment of the invention, the kinetic energy of the pressure switch for the heating program to build up the pressure that promotes the active ingredient on the Mikroheizplatte.
In a further embodiment, a piezo element is used to exert direct pressure on a filled capillary. However, the piezo element requires a high supply voltage of about 50 V.
Also, the resulting negative pressure during suction can be used to apply the active ingredient metered applied to the heating element. A dosage is also possible by the lifting movement of the mouthpiece when pressed against the mouth.
To protect the substances to be vaporized during storage, the container filled with nicotine or another active ingredient is preferably packed gas-tight. In the present example, the capillary is closed in a preferred embodiment on both sides, for example with a septum. The septum is removed before or during insertion of the holding element into the housing and / or when screwing the mouthpiece.
The holding elements can also be sold together with a reservoir in which there is a larger volume of nicotine for reloading the capillary.
After screwing the mouthpiece of the inhaler is ready for use, and it can be applied by pressing an operating knob 41 voltage to the MHP. The user can suck the vaporized by the MHP drug, for example, the nicotine, by normal pulling on the mouthpiece from the evaporation chamber 38 through the air channels 36 in the mouthpiece 2 and inhale through the mouthpiece tip 20. Instead of the operating button 41, a vacuum sensor may be mounted in the inhaler 1, which generates a vaporization signal when sucking in the air through the mouthpiece 2, which triggers the heating of the MHP.
As a further alternative trigger signal, the pressure of the lips can be used on a suitably ausgestaltetes mouthpiece to trigger the heater.
According to a preferred embodiment, the holding element is made of transparent plastic, for example a PMMA material. A viewing window 42, which is arranged in the region of the evaporation space in the housing 4, allows a visual control of the evaporation.
The evaporation of nicotine is controlled primarily by the heating time. Measurements have shown that the released nicotine concentration in ten moves within 3 minutes corresponds to a light cigarette with a nicotine content of about 0.1 mg.
Instead of the capillaries described in the above examples, one or more fillable storage elements can also be used. It may be a thin plastic tube or a one-piece injection molding element provided with a corresponding recess with a discharge opening. The capillary need not necessarily be arranged in the holding element, but it can be fixedly mounted in a further embodiment in the housing, in which case it is filled via a storage element and then emits the active ingredient on the MHP.
It is also the use of multiple capillaries possible, which can be loaded with different agents and may also have different volumes. Holding element and mouthpiece can be advantageously arranged in a single component. The mouthpiece may have a special shape, for example it may be formed like a cigar or a cigarette holder. It may also contain an aroma filter through which nicotine or drug vapor is drawn.
According to a further embodiment, the MHP is mounted in a so-called flip-chip position. The MHP is mounted upside down, so that the electrical contacts on the, the capillary facing away from the bottom. In this embodiment, can be dispensed with an eccentric positioning of the capillary to the MHP, since the sensitive bonding wires can not be damaged by a faulty manipulation with the capillary. This mounting method has the further advantage that the active substance to be evaporated is brought from behind onto the membrane. The etched out membrane of the MHP mounted upside down additionally forms a depression, which serves as a small container for holding the active substances to be evaporated.
In a further embodiment according to the present invention, a receptacle, for example a small cylinder or a torus, is mounted around the MHP and holds the volume of liquid to be evaporated, for example the nicotine over the heating plate.
An indicator LED can be arranged to display the switched-on main power supply to a well-visible location of the device. For example, this LED indicates when a battery change is needed. With a second LED can be displayed when the MHP is heated and also serves as an indicator for the function of the MHP.
