PATENTANSPRÜCHE
1. Pharmazeutische Retard-Tablette bestehend aus einem stäbchenförmigen Pressling enthaltend eine Retard-Tablettenmasse auf Matrixbasis und ein- oder mehrere Wirkstoffe homogen oder in Längsrichtung schichtweise verteilt, welcher Pressling in Querrichtung und senkrecht zum kleinsten Durchmesser eine oder mehrere ein- oder beidseitig angeordnete relativ tiefe Bruchrillen aufweist.
2. Pharmazeutische Retard-Tablette gemäss Patentanspruch 1, dadurch gekennzeichnet, dass die Tiefe der einseitigen Bruchrillen oder die Gesamttiefe der beidseitigen gegen überliegenden Bruchrillen 1/3 bis 2/3, vorzugsweise 2/5 bis lfi, des kleinsten Durchmessers des Stäbchens beträgt.
Gegenstand der Erfindung ist eine teilbare pharmazeutische Retard-Tablette in Stäbchenform.
Pharmazeutische, perorale Darreichungsformen mit verzögerter Wirkstoffabgabe zwecks Aufrechterhaltung einer möglichst konstanten und langandauernden Wirkstoffkonzentration im Blutkreislauf, also sogenannte Retard Formen, sind schon lange bekannt, insbesondere in der Form von überzogenen Tabletten und Kapseln. Weiter sind auch gewöhnliche Tabletten mit tiefen Bruchrillen als solche bekannt und demgemäss auch gewisse Vorteile der tiefen Bruchrillen, nämlich leichtere Brechbarkeit und im Ergebnis präziser dosierte Bruchteile. In diesem Zusammenhang kann auf folgende Patentschriften verwiesen werden: US 3 883 647, US D 201 497, US D 202467 und DE-AS 1 200 790.
Retard-Tabletten, die sich ohne wesentlichen Verlust der Retard-Wirkung in vorbestimmte gleichmässige Teile brechen lassen, wie dies für gewöhnliche Tabletten bekannt ist, hat es bis jetzt nicht gegeben, obwohl ein Bedarf dafür zweifellos besteht.
Die erfindungsgemässe pharmazeutische Retard-Tablette ist dadurch gekennzeichnet, dass sie aus einem stäbchenförmigen Pressling besteht, der eine Retard-Tablettenmasse auf Matrixbasis und ein oder mehrere Wirkstoffe homogen oder in Längsrichtung schichtweise verteilt enthält, welcher Pressling in Querrichtung und senkrecht zum kleinsten Durchmesser eine oder mehrere ein- oder beidseitig angeordnete, relativ tiefe Bruchrillen aufweist.
Die Tiefe der einseitigen Bruchrillen oder die Gesamttiefe der beidseitig gegenüberliegenden Bruchrillen beträgt vorteilhafterweise 1/3 bis 1/2, vorzugsweise 2/5 bis 1/2 des kleinsten Durchmessers des Stäbchens. Die Seitenflächen der Bruchrillen sind vorzugsweise gewölbt.
Die Tablette kann ein- oder beidseitig biplan oder gewölbt und ein- oder beidseitig mit gegenüberliegenden oder versetzten Bruchrillen versehen werden und kann demgemäss in zwei oder mehrere vorbestimmte gleiche oder ungleiche Teile zerlegt werden. Dies erlaubt eine individuellere und somit genauere Dosierbarkeit des Arzneimittels je nach Krankheitsbild und Patient (Erwachsene gemäss Gewicht, Kinder, etc.).
Das Wesen der Erfindung besteht darin, dass durch die besondere Formgebung der vorliegenden Tablette (Stäbchenform mit relativ tiefen Bruchrillen) die Bruchfläche im Vergleich zur Gesamtoberfläche so klein wie möglich gehalten werden kann, so dass der Retard-Effekt nur geringfügig verändert wird und die Gewichtsstreuung der Bruchteile erheblich verringert wird. Überraschenderweise wurde zudem festgestellt, dass die Verminderung der Retard-Wirkung bei den Bruchstücken geringer ist, als eigentlich auf Grund der um die unregelmässig strukturierten und porösen Bruchflächen vergrösserten Gesamtoberfläche erwartet werden müsste.
Die erfindungsgemässen Retard-Tabletten haben ausser den Hauptvorteilen der leichten Teilbarkeit und des im Vergleich zur ganzen Retard-Tablette nur geringfügig veränderten Retard-Effekts der Bruchhälften noch folgende Vorteile: sie sind leicht markierbar durch Prägedruck oder Bedrukkung und zwar beidseitig, z. B. Fabrikantennamen auf der einen Seite und Markenname und/oder eine Code-Bezeichnung des Arzneimittels auf der anderen; sie sind leicht schluckbar, sowohl als Ganzes, als auch als Bruchstücke, d.h. wesentlich leichter als runde Tabletten, bzw. deren Bruchstücke. Es lassen sich auch Mehrschicht-Tabletten mit verschiedenen Wirkstoffen bzw. verschiedenen Wirkstofffreigaben herstellen, wobei gewisse Schichten so angeordnet werden können, dass sie beim Brechen keine Bruchstellen aufweisen.
Durch zusätzliche seitliche Erweiterung der Bruchrillen lässt sich die Bruchfläche nochmals verkleinern, doch wird die Tablette dadurch sehr zerbrechlich, ganz abgesehen von den Herstellungsproblemen, die dadurch entstehen.
Für die Herstellung der erfindungsgemässen Retard Tabletten können die üblichen, gegebenenfalls auf die verwendeten Wirkstoffe abgestimmten, Hilfsstoffe verwendet werden. Aus der leichten Brechbarkeit und der Neigung zum Deckeln (engl.: capping) der Tabletten ergibt sich, dass man vorzugsweise von fest zusammenhängenden Tablettenmassen ausgeht.
Das Matrixmaterial kann aus einem an sich inerten bzw.
unverdaulichen Gemisch bestehen, z. B. aus Kunststoffen wie PVC, Acrylate und Methacrylate. Es kann aber auch ein Material sein, das einer fortschreitenden Aufweichung (z. B.
hydrophile Gelbildner) oder einer Erosion im Verlauf der Magen-Darmpassage unterliegt (z. B. Lipide im Gemisch mit inerten Trägermaterialien oder verdaubare Di- und Triglyceride). Vielfach ist auch die Verwendung von an sich nicht retardierenden Füll- oder Trägerstoffen wie z. B. Bentonit, Talk, Di- und Tricalciumphosphate, Lactose, Kieselsäure, Cellulose u. dgl. gleichzeitig mit den retardierenden Materialien erforderlich oder von Vorteil.
Als retardierende Materialien können im Einzelnen folgende verwendet werden: a) Essentiell wasserunlösliche:
Lipide: Fettalkohole, z. B. Cetylalkohol, Stearylalkohol, Cetostearylalkohol, Glyceride, z. B. Glycerinmonostearat, hydriertes Rizinusöl, hydriertes Baumwollsamenöl, Gemische von Mono-, Di- und Triglyceriden pflanzlicher Ole; Wachse, z.B. Bienenwachs, Carnaubawachs; Paraffine, z. B.
Paraffin, Erdwachs; Fettsäuren, z.B. Stearinsäure. Cellulosederivate, z. B. Äthylcellulose, Acetylcellulose. Polyvinylverbindungen, z. B. PVC, Polyvinylacetat und Copolymere mit Crotonsäure. Polyäthylen. Vinylchlorid-Vinylacetat Copolymere. Polymere und Copolymere und von Acrylaten und Methacrylaten, z. B. Copolymerisate von Acrylsäure äthylester und Methacrylsäure-methylester.
b) Wasserlösliche bzw. mit Wasser quellbare:
Cellulosederivate, z.B. Methylcellulose, Hydroxypropylcellulose, Hydroxypropylmethylcellulose, Na-carboxymethylcellulose (vorzugsweise Verbindungen mit höherer Viskosität). Polyacrylsäure (und Salze). Natürliche (anionische) Schleimstoffe: z. B. Xanthan Gummi, Guar Gummi, Traganth, Alginsäure und Salze.
Wirkstoffe, die im neutralen Darmmilieu nicht besonders gut, beim sauren pH des Magens aber besser löslich sind, können auch mit Zusätzen retardiert werden, die funktionelle Carboxylgruppen aufweisen (lösen sich im neutralen Be
reich), z. B. Schellack, Cellulose-acetatphathalat, Hydroxypropylmethylcellulosephthalat, Halbester von Maleinsäureanhydrid-Copolymeren .
Um die erfindungsgemässen Tabletten noch weiter zu festigen, können die Presslinge noch mit einem löslichen Film überzug versehen werden. Er soll aber nicht so dick sein, dass er die Brechbarkeit übermässig erschweren und gleichzeitig auch die Freigabe des Wirkstoffes steuern bzw. erheblich beeinflussen würde. Die bevorzugte Dicke ist 20-50 llm.
Da viele Wirkstoffe bekanntlich einen unangenehmen bitteren Geschmack aufweisen, dient der Filmüberzug auch der Geschmacksabdeckung. Als Filmüberzugsmaterialien eignen sich besonders Methylcellulose, Hydroxypropylcellulose, Hydroxypropylmethylcellulose (vor allem das letztere), z.T.
im Gemisch mit Talk, Netzmitteln, Pigmenten (zur Erleichterung des Auftrages bzw. zur Kennzeichnung); ein Gemisch von Polyvinylpyrrolidon bzw. Copolymerisat von Polyvinylpyrrolidon und Polyvinylacetat mit Hydroxypropylmethylcellulose, Gemische von Schellack mit Hydroxypropylmethylcellulose oder Polyvinylacetat bzw. dessen Copolymeren mit Polyvinylpyrrolidon und Gemische von wasserlöslichen Cellulosederivaten (wie Hydroxypropylmethylcellulose) und wasserunlöslicher Äthylcellulose. Diese Überzüge werden, je nach Löslichkeit der Komponenten, in wässriger Lösung oder in organischer Lösung (Mischungen mit Schellack, Äthylcellulose) aufgetragen. Ferner können folgende Materialien verwendet werden: Mischungen von an sich wasserunlöslichen Arylaten (z. B.
Copolymerisat von Acrylsäure äthylester und Methacrylsäuremethylester), die in wässriger Dispersion verwendet werden, mit wasserlöslichen Bestandteilen, z. B. Lactose, Polyvinylpyrrolidon, Polyäthylenglykol.
Hydroxypropylmethylcellulose.
Die Herstellung der Tablettenmasse kann durch Mischen des Feststoffteilchens (Wirkstoff und gegebenenfalls Füllstoff) mit Retardierungsmitteln oder durch Mischen bereits überzogener Wirkstoffteilchen mit gewöhnlichen Tablettenhilfsstoffen erfolgen. Das Überziehen kann erfolgen in Wirbelschichten, in Dragierkesseln, in schnellaufenden Mischern, aber auch mit Hilfe des Verfahrens der Mikroverkapselung.
Die Herstellung der Presslinge kann mit den für die Herstellung von stäbchenförmigen Presslingen bzw. Mehrschichttabletten bekannten Tablettiermaschinen erfolgen.
Andere Ziele und Vorteile der Erfindung ergeben sich aus der folgenden Beschreibung in Verbindung mit der Zeichnung, die verschiedene Ausführungsformen erläutert.
In den Figuren 1-3 ist jeweils eine Seitenansicht, b eine Draufsicht und c eine Bruchstelle einer Tablette gemäss der Erfindung.
Figur 1 zeigt eine Tablette mit einer einseitigen Bruchrille, wobei wie oben angegeben die Schicht Sl eine andere Zusammensetzung haben kann als die Schicht S2. Beim Brechen der Tablette bleibt die Schicht S1 ohne Bruchteile. Bei einem Kombinationspräparat befindet sich z. B. der retardierte Wirkstoff in Schicht S1 und der nicht retardierte in Schicht S2. Bei einer Tablette mit nur einem (retardierten) Wirkstoff kann die Schicht S2, auch als Placeboschicht vorhanden sein. Die Rillentiefe t beträgt z.B. 1/3 der Höhe d des Presslings (d = kleinster Durchmesser des Presslings).
Figur 2 zeigt eine Tablette mit zwei einseitigen Bruchrillen, die sich entsprechend in 2 ungleiche oder 3 gleiche Teile zerlegen lässt. Die Bruchrillen können von gleichen oder ungleichen Tiefen sein.
Figur 3 zeigt eine Tablette mit 2 Paaren von beidseitig gegenüberliegenden Bruchrillen. Auch diese Tablette lässt sich entsprechend in 2 ungleiche oder 3 gleiche Teile zerlegen. Bei beidseitig gegenüberliegenden Bruchrillen ist die Gewichtsstreuung der Bruchteile am geringsten. Die Rillentiefe t, auch der beidseitig gegenüberliegenden Rillen kann verschieden sein. Sie ist z. B. bei beidseitig gegenüberliegenden Bruchrillen je 1/5 der Höhe d des Presslings oder gesamthaft 1/3 bis 2/5 des Presslings.
Beispiel 1
2,0 kg Metoprolol-tartrat, 0,1 kg kolloidales Siliciumdioxid, 0,2 kg Calciumhydrogenphosphat und 0,25 kg mikrokristalline Cellulose werden gemischt und mit 0,6 kg einer wässrigen, 30%-igen Dispersion von Acrylsäureäthylester Methacrylsäuremethylester 70:30 Copolymerisat in der Wirbelschicht granuliert. Die Einsprühgeschwindigkeit beträgt 300 ml pro Minute, die Zulufttemperatur 30 "C. Anschliessend wird während 20 Minuten bei 40 "C Zulufttemperatur in der gleichen Apperatur getrocknet. Das Granulat wird in einen Planetenmischer gebracht und mit 0,8 kg geschmolzenem und auf 60 C erwärmtem Stearylalkohol versetzt und während 15 Minuten geknetet.
Nach dem Abkühlen wird das Granulat durch ein Sieb mit Maschenweite 1 mm gedrückt und in einem Taumelmischer mit 0,05 kg Magnesiumstearat, 0,05 kg kolloidalem Siliciumdioxid und 0,4 kg Hydroxypropylmethylcellulose Viskosität 15 000 cps während 10 Minuten gemischt.
Das Verpressen dieses Metoprolol-Retardgranulates zu Tabletten zuje 445 mg Bruttogewicht erfolgt auf einer Rundläufer-Tablettenpresse mit geführten Stempeln folgender Dimensionen: Länge 17,0 mm, Breite 8,0 mm. Die Stempel sind gewölbt (Wölbungsradius 4,8 mm), und auf einem der beiden ist eine 2,0 mm tiefe (bezogen auf die Kalottenhöhe) auslaufende Bruchkerbe (Öffnungswinkel 45"60") angebracht. Die resultierenden Presslinge haben eine Höhe von insgesamt 4,6 mm.
Die Lackierung erfolgt in einem mit Schikanen ausgerüsteten Dragierkessel von 55 cm Durchmesser. Dabei werden 5 kg Presslinge mit einer Lacklösung bzw. Suspension, gemäss nachfolgender Rezeptur, mit Hilfe einer Zweistoffdüse kontinuierlich besprüht.
0,1 kg Hydroxypropylmethylcellulose (Viskosität 5 cps) werden in 1,2 kg entmineralisiertem Wasser gelöst, dazu werden unter Rühren 0,005 kg Polysorbat 80 und 0,05 kg Talk sowie 0,1 kg einer 20%igen homogenen Suspension von Titandioxid in einer Lösung von 0,007 kg Hydroxypropylmethylcellulose (5 cps) in 90%-igem Athanol gegeben.
Die aufgesprühte Menge beträgt 19 mg (Trockengewicht) pro Pressling. Die Zulufttemperatur beträgt 60"C, die Temperatur der Presslinge im Kessel wird auf ca. 35 C gehalten.
Die Auflösungsgeschwindigkeit der filmüberzogenen Tabletten wird mit der Durchmessermethode bestimmt (F.
Langebucher, H. Rettig, Drug. Dev. Ind. Pharm. 3, 241 [19771), und zwar mit einer Durchflussgeschwindigkeit von 16 ml pro Minute mit künstlichem Magensaft (pH 1,2, ohne Enzyme) während der ersten Stunde, anschliessend mit künstlichem Darmsaft (pH 7,5, ohne Enzyme) bei 37 C. Für die Freisetzung von Metoprololtartrat in % des Sollgehaltes aus ganzen bzw. halbierten Filmtabletten sind folgende Ergebnisse typisch: Zeit: ganze Tablette: halbierte Tablette

Beispiel 2
Retard-Granulat von Metoprolol-tartrat wird wie in Beispiel 1 angegeben hergestellt.
Daneben wird ein nicht-retardiertes Chlortalidon Granulat wie folgt hergestellt:
0,25 kg Chlortalidon, 1,75 kg Lactose und 0,5 kg Maisstärke werden gemischt und mit 0,3 kg eines Kleisters aus 0,1 kg Maisstärke und 0,2 kg Wasser in einem Planetenmischer zu einer plastischen Masse verformt. Die feuchte Masse wird durch ein Sieb von 2 mm Maschenweite getrieben und in der Wirbelschicht während 20 Minuten bei 60 "C getrocknet. Das durch ein Sieb von 1 mm Maschenweite getriebene, getrocknete Granulat wird mit 0,1 kg Talk, 0,01 kg Magnesiumstearat und 0,29 kg mikrokristalliner Cellulose vermischt.
Die Verpressung der beiden Granulate erfolgt auf einer Rundläufer-Tablettenmaschine mit geführten Stempeln, die die Herstellung von Schichttabletten erlaubt. Zunächst wird das nicht retardierte Chlortalidon-Granulat zudosiert, anschliessend aus einem zweiten Fülltrichter das Metoprolol Retardgranulat. Zum Verpressen werden 2 Stempel für 2 verschiedene Bruchkerben mit folgenden Abmessungen verwendet: Länge 19,0 mm, Breite 7,0 mm. Der Wölbungsradius beträgt 4,2 mm. Die auslaufenden Bruchkerben im Pressling haben eine Tiefe von 1,7 mm (Kalottentiefe), auf der Seite der Metoprolol-Schicht und von 0,8 mm auf der Seite der Chlortalidon-Schicht. Es resultiert eine Tablettenhöhe von 6 mm.
Die Freisetzung von Metoprol-tartrat erfolgt in der unter Beispiel 1 angegebenen Weise, die Zerfallzeit der nicht retardierten Chlortalidon-Schicht beträgt 2-3 Minuten (Zerfallsprüfgerät nach USP, künstlicher Magensaft bei 37 ).

Retard tablets, which can be broken into predetermined uniform parts without a significant loss of the retarding effect, as is known for ordinary tablets, have not existed until now, although there is undoubtedly a need for them.

The prolonged-release tablets according to the invention have the following advantages in addition to the main advantages of easy divisibility and the retard effect of the broken halves, which is only slightly changed in comparison to the entire prolonged-release tablet. It is also possible to produce multilayer tablets with different active substances or different active substance releases, it being possible for certain layers to be arranged in such a way that they do not have any breaking points when broken.
The fracture surface can be further reduced by additionally expanding the fracture grooves laterally, but this makes the tablet very fragile, quite apart from the manufacturing problems that arise as a result.
The customary auxiliaries which may be matched to the active ingredients used can be used to produce the retard tablets according to the invention. From the easy breakability and the tendency to capping the tablets, it follows that one preferably starts from firmly connected tablet masses.
The matrix material can be made from an inert or
indigestible mixture exist, e.g. B. from plastics such as PVC, acrylates and methacrylates. However, it can also be a material that is subject to progressive softening (e.g.
hydrophilic gel formers) or erosion in the course of the gastrointestinal passage (e.g. lipids in a mixture with inert carrier materials or digestible di- and triglycerides). In many cases, the use of non-retarding fillers or carriers such. B. bentonite, talc, di- and tricalcium phosphates, lactose, silica, cellulose u. Like. Polymers and copolymers and of acrylates and methacrylates, e.g. B. copolymers of acrylic acid ethyl ester and methacrylic acid methyl ester.
b) Water-soluble or water-swellable:
Cellulose derivatives, e.g. Methyl cellulose, hydroxypropyl cellulose, hydroxypropyl methyl cellulose, Na-carboxymethyl cellulose (preferably compounds with higher viscosity). Polyacrylic acid (and salts). Natural (anionic) mucilages: e.g. B. xanthan gum, guar gum, tragacanth, alginic acid and salts.
Active ingredients that are not particularly good in the neutral intestinal environment, but are more soluble in the acidic pH of the stomach, can also be retarded with additives that have functional carboxyl groups (dissolve in the neutral loading
rich), e.g. B. shellac, cellulose acetate phthalate, hydroxypropyl methyl cellulose phthalate, half esters of maleic anhydride copolymers.
In order to further consolidate the tablets according to the invention, the compacts can also be provided with a soluble film. However, it should not be so thick that it would make the breakability excessively difficult and at the same time would control or significantly influence the release of the active ingredient. The preferred thickness is 20-50 llm.
Since many active ingredients are known to have an unpleasant bitter taste, the film coating also serves to cover the taste. Particularly suitable film coating materials are methyl cellulose, hydroxypropyl cellulose, hydroxypropyl methyl cellulose (especially the latter), in some cases.

The tablet mass can be produced by mixing the solid particle (active ingredient and optionally filler) with retardants or by mixing already coated active ingredient particles with ordinary tablet excipients. The coating can take place in fluidized beds, in coating pans, in high-speed mixers, but also with the aid of the microencapsulation process.
The compacts can be produced using the tabletting machines known for the production of rod-shaped compacts or multilayer tablets.

FIG. 1 shows a tablet with a break groove on one side, whereby, as indicated above, the layer S1 can have a different composition than the layer S2. When the tablet is broken, the layer S1 remains without fractions. In a combination preparation is z. B. the retarded drug in layer S1 and the non-retarded in layer S2. In the case of a tablet with only one (retarded) active ingredient, layer S2 can also be present as a placebo layer. 2.0 kg of metoprolol tartrate, 0.1 kg of colloidal silicon dioxide, 0.2 kg of calcium hydrogen phosphate and 0.25 kg of microcrystalline cellulose are mixed and copolymerized with 0.6 kg of an aqueous, 30% dispersion of methyl acrylate, methyl methacrylate 70:30 granulated in the fluidized bed. The spraying rate is 300 ml per minute, the supply air temperature is 30 "C. Then the mixture is dried for 20 minutes at 40" C supply air temperature in the same apparatus. The granules are placed in a planetary mixer and mixed with 0.8 kg of melted stearyl alcohol heated to 60 ° C. and kneaded for 15 minutes.
After cooling, the granules are pressed through a sieve with a mesh size of 1 mm and mixed in a tumble mixer with 0.05 kg of magnesium stearate, 0.05 kg of colloidal silicon dioxide and 0.4 kg of hydroxypropylmethyl cellulose viscosity 15,000 cps for 10 minutes.
These metoprolol prolonged-release granules are compressed into tablets with a gross weight of 445 mg each on a rotary tablet press with guided punches of the following dimensions: length 17.0 mm, width 8.0 mm. The stamps are domed (radius of curvature 4.8 mm), and on one of the two is a 2.0 mm deep (based on the calotte height) tapered break notch (opening angle 45 "60"). The resulting compacts have a total height of 4.6 mm.
The painting takes place in a coating pan equipped with chicanes of 55 cm diameter. 5 kg of pellets are continuously sprayed with a lacquer solution or suspension according to the following recipe using a two-component nozzle.
0.1 kg of hydroxypropylmethylcellulose (viscosity 5 cps) are dissolved in 1.2 kg of demineralized water, with stirring 0.005 kg of polysorbate 80 and 0.05 kg of talc and 0.1 kg of a 20% homogeneous suspension of titanium dioxide in a solution of 0.007 kg of hydroxypropylmethyl cellulose (5 cps) in 90% ethanol.
The amount sprayed on is 19 mg (dry weight) per compact. The supply air temperature is 60 "C, the temperature of the pellets in the boiler is kept at approx. 35 C.
The dissolution rate of the film-coated tablets is determined using the diameter method (F.
Langebucher, H. Rettig, Drug. Dev. Ind. Pharm. 3, 241 [19771), namely at a flow rate of 16 ml per minute with artificial gastric juice (pH 1.2, without enzymes) for the first hour, then with artificial intestinal juice (pH 7.5, without enzymes) at 37 C. Example 2
Retard granules of metoprolol tartrate are produced as indicated in Example 1.
In addition, non-retarded chlorotalidone granules are produced as follows:
0.25 kg of chlorotalidone, 1.75 kg of lactose and 0.5 kg of corn starch are mixed and shaped into a plastic mass with 0.3 kg of paste made from 0.1 kg of corn starch and 0.2 kg of water in a planetary mixer. The moist mass is forced through a sieve with a mesh size of 2 mm and dried in the fluidized bed for 20 minutes at 60 ° C. The dried granules which are passed through a sieve with a mesh size of 1 mm are mixed with 0.1 kg talc, 0.01 kg magnesium stearate and 0.29 kg of microcrystalline cellulose mixed.
The two granules are pressed on a rotary tablet machine with guided punches, which allows the production of layered tablets. First the non-retarded chlortalidone granules are metered in, then the metoprolol prolonged-release granules from a second filling funnel. For pressing, 2 punches are used for 2 different notches with the following dimensions: length 19.0 mm, width 7.0 mm. The radius of curvature is 4.2 mm. The leaking notches in the compact have a depth of 1.7 mm (dome depth) on the side of the metoprolol layer and 0.8 mm on the side of the chlorotalidone layer. The result is a tablet height of 6 mm.
Metoprol tartrate is released in the manner given in Example 1, the disintegration time of the non-retarded chlorotalidone layer is 2-3 minutes (disintegration tester according to USP, artificial gastric juice at 37).
