Verfahren nach einem der Ansprüche 1, 3 und 4, dadurch gekennzeichnet, dass R2 2-Thiazolyl, 5-Methyl-3-isoxazolyl oder 2-Pyridyl, dass R2 4-Fluorphenyl, 3-Trifluormethylphenyl, 2,4-Dichlorphenyl, 4-Bromphenyl, 4-Nitrophenyl, 3 Chlorphenyl, 2-Tolyl, 2,5-Dichlorphenyl, 4-Nitro-2-tolyl, 4 Jodphenyl oder 4-n-Butyl-phenyl bedeutet.
6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man 3,4-Dihydro-2-methyl-3-oxo-4-(2-pyridyl carbamoyl)-2H-thieno[2,3-e] 1 ,2-thiazin- 1 ,1-dioxid herstellt.
7. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man 3,4-Dihydro-2-methyl-3-oxo-4-(2-thiazolyl carbamoyl)-2H-thienol[2,3-e] 1,2-thiazin-1, 1-dioxid herstellt
8. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man 3-Hydroxy-2-methyl-N-pyrazinyl-2H-thieno [2,3-e]-1,2-thiazin-4-carboxamid-1,1-dioxid herstellt.
9. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man 4'-Brom-3-hydroxy-2-methyl-2H-thieno- [2,3-e]-1,2-thiazin-4-carboxanilid-1,1-dioxid herstellt.
10. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man 4'-Chlor-3-hydroxy-2-methyl-2H-thieno [2,3-e]-1 ,2-thiazin-4-carboxanilid- l,l-dioxid herstellt.

Der in dieser Beschreibung verwendete Ausdruck niederes Alkyl bezeichnet geradkettige oder verzweigte gesättigte Kohlenwasserstoffgruppen mit 1-4 Kohlenstoffatomen, wie z.B. Methyl, Äthyl, Propyl, Isopropyl, t-Butyl und dgl. Der Ausdruck niederes Alkoxy bezieht sich auf Hydrocarbonoxygruppen mit bis zu 4 C-Atomen. Die Bezeichnung Halogen bezieht sich auf die 4 Halogene Chlor, Brom, Fluor, Jod.

Die Thienothiazinderivate der Formel I und ihre Salze werden erfindungsgemäss dadurch hergestellt, dass man eine

Die Umsetzung der Verbindungen der Formel II und III kann in An- oder Abwesenheit eines inerten Lösungsmittels erfolgen. Als Lösungsmittel eignen sich Alkohole, wie Äthanol etc., Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, etc., halo genierte Kohlenwasserstoffe, wie Chloroform, Chlorbenzol,
Methylenchlorid, Tetrachlorkohlenstoff, etc., oder Dimethyl formamid oder Dioxan. Die Umsetzung erfolgt vorzugsweise durch Erwärmen, wobei Schmelz- bzw. Rückflusstemperatur des Reaktionsgemisches besonders bevorzugt ist.
Die Ausgangsstoffe der Formel II können gemäss dem nachfolgenden Reaktionsschema hergestellt werden, in wel chem Hal Halogen bedeutet und R, Rl, R2, R3 und R4 obige

Von den Verbindungen der Formel IV ist die 3-Chlorthiophen-2-carbonsäure bekannt, wobei diese aber auf relativ komplizierte Weise hergestellt wurde. Eine einfachere Methode zur Herstellung von 3 -Chlor-thiophen-2-carbonsäure besteht darin, dass man den bekannten 3-Hydroxythiophen-2carbonsäure-methylester in einem inerten, über 80"C siedenden Lösungsmittel, wie Chloroform oder Dioxan, mit einem Chlorierungsmittel, wie Phosphorpentachlorid, intermediär in das 3-Chlor-thiophen-2-carbonsäurechlorid überführt und dieses zur entsprechenden Säure hydrolysiert. In analoger Weise können auch substituierte 3-Chlor-thiophen-2-carbonsäuren (Verbindungen der Formel IV, worin Hal Chlor bedeutet und R3 und/oder R4 von Wasserstoff verschieden ist) hergestellt werden.
Obwohl zur nachstehend beschriebenen Herstellung einer Verbindung der Formel V grundsätzlich auch eine Bromverbindung als Ausgangsmaterial eingesetzt werden kann, empfiehlt sich die Verwendung der entsprechenden Chlorverbindung.
Die Überführung einer Halothiophencarbonsäure entsprechend der Formel IV in ein Kaliumsalz einer Sulfo-thiophencarbonsäure der Formel V erfolgt nach an sich bekannten Methoden durch Umsetzen der Verbindung der Formel IV mit Natriumhydrogensulfit in Gegenwart eines Kupfer(I)salz Katalysators, insbesondere Kupfer(I)chlorid, und Umsetzen des erhaltenen Reaktionsproduktes mit Kaliumchlorid. Beim Umsatz mit Natriumhydrogensulfit sollte zur Erzielung optimaler Ausbeuten eine Temperatur von 143"C eingehalten werden.
Die Überführung der Verbindung der Formel V in die entsprechende freie Säure der Formel VI erfolgt in an sich bekannter Weise, z.B. mit einem starken Ionenaustauscher.
Die Veresterung der Säure der Formel VI zum Ester der Formel VII erfolgt autokatalytisch (Anwesenheit der Sulfogruppe) in einem Alkohol/Chloroform-Gemisch. Zur Bildung des Methylesters wird die Säure in Methanol/Chloroform gelöst und das Reaktionsgemisch zum Siedepunkt des ternären Azeotrops (Methanol/Chloroform/Reaktionswasser) erhitzt.
Die Überführung einer Verbindung der Formel VII in das Säurehalogenid der Formel VIII erfolgt in an sich bekannter Weise mit einem Halogenierungsmittel, vorzugsweise mit einem Chlorierungsmittel, wie Thionylchlorid oder Phosphorpentachlorid. Die Chlorierung mit Thionylchlorid kann ohne Lösungsmittel durch Rückflusserhitzen erfolgen. Bei Verwendung von Phosphorpentachlorid kann in Gegenwart eines inerten Lösungsmittels, wie Chloroform, Tetrachlorkohlenstoff, Dioxan, und bei einer Temperatur zwischen 50"C und Rückflusstemperatur des Reaktionsgemisches gearbeitet werden.
Die Verbindung der Formel VIII kann aber ausgehend von dem weiter oben erwähnten Kaliumsalz einer Sulfo-thiophencarbonsäure entsprechend der Formel V auch über eine Verbindung der Formel IX hergestellt werden. Hierzu wird das erwähnte Kaliumsalz z.B. mit 2 Mol Phosphorpentachlorid und in Gegenwart von Phosphoroxychlorid als Lösungsmittel bei einer Temperatur zwischen 30"C und dem Siedepunkt von Phosphoroxychlorid umgesetzt. Anstelle von Phosphoroxy chlorid kann aber auch ein inertes organisches Lösungsmittel, wie Dioxan, Chloroform, Tetrachlorkohlenstoff, Benzol, Toluol und dgl. verwendet werden.
Die Veresterung der Verbindung der Formel IX zum entsprechenden Ester der Formel VIII erfolgt mit dem entsprechenden Alkohol, insbesondere Methanol, bei einer Temperatur zwischen Raumtemperatur und Rückflusstemperatur. Als Lösungsmittel kann der Alkohol oder ein inertes Lösungsmittel, wie Chloroform, Tetrachlorkohlenstoff, Dioxan oder Benzol, dienen.
Die Verbindung der Formel VIII wird dann zu einer Verbindung der Formel X aminoalkyliert.
Die Aminoalkylierung einer Verbindung der Formel VIII erfolgt in an sich bekannter Weise durch Umsetzen mit einem Alkylamin der allgemeinen Formel Ri-NH2 XIII worin Rl obige Bedeutung besitzt, in Gegenwart eines inerten organischen Lösungsmittels, wie Chloroform, Methylenchlorid, Tetrachlorkohlenstoff, Benzol oder Dioxan und bei Raumtemperatur.
Die Verbindung der Formel X wird dann zur entsprechenden freien Carbonsäure verseift, beispielsweise durch Erhitzen mit Alkali,wie Kaliumhydroxyd, vorzugsweise in wässrig/ alkoholischem Medium. Die erhaltene freie Carbonsäure wird hierauf in das entsprechende Säurechlorid übergeführt, dieses wird mit Diazomethan in das entsprechende Diazomethylketon umgewandelt, und letzteres liefert bei der Behandlung mit Silbernitrat und einem niederen Alkanol die entsprechende Verbindung der Formel XI (wobei das Produkt in zwei konformationsisomeren Formen auftreten kann, wovon die eine der Formel XI entspricht und die andere infolge einer chelatartigen Wasserstoffbrücke eine zyklische Struktur aufweist und wobei zuweilen auch Nebenprodukte entstehen können).
Durch Umsetzung der Verbindung XI bzw. ihres Konformationsisomeren mit einem Chlorameisensäureester der allgemeinen Formel C > CO-OR XIV worin R obige Bedeutung besitzt, in Gegenwart einer starken Base, wie Natriummethylat, erhält man eine Verbindung der Formel XII, welche durch Cyclisation (z.B. durch Erwärmen mit Natriumhydrid in Dioxan) in die entsprechende Verbindung der Formel II übergeführt werden kann.
Die Verbindungen der Formel I sind sauer und können mit entsprechenden Basen pharmazeutisch annehmbare Salze bilden. Als Basen eignen sich z.B. Alkalimetalle, wie Lithium, Natrium, Kalium; Erdalkalien, wie Magnesium und Calcium sowie Amine, wie Triäthanolamin, Diäthylaminoäthanol, Triäthylamin, Trimethylamin oder Diäthylamin und dgl. Verbindungen der Formel I mit einem basischen Heterocyclus R2 können mit starken Säuren auch pharmazeutisch annehmbare Säureadditionssalze bilden, wozu insbesondere Mineralsäuren, wie Salzsäure in Frage kommen.
Die Verbindungen der allgemeinen Formel I sowie deren Salze haben eine antiinflammatorische, analgetische und antirheumatische Wirkung. Diese wertvollen pharmakologischen Eigenschaften können unter Verwendung von Standardmethoden bestimmt werden, beispielsweise im bekannten Kaolin Pfotenoedemtest (an der Ratte). In diesem Test wird in der rechten Hinterpfote der Ratte durch intradermale Injektion von 0,1 ml einer 10%gen Kaolinsuspension (bolus alba) eine akute lokale Entzündung erzeugt. Die zu untersuchende Substanz wird auf oralem Wege verabreicht, und die folgenden Parameter werden gemessen:
1. Durchmesser der Pfote in mm (als Ausdruck der Heftigkeit der Entzündung);
2. Druck (in g) auf die Pfote (zur Ermittlung der Schmerzschwelle).
112 Stunde vor und 31/2 Stunden nach der Kaolininjektion wird die zu untersuchende Substanz verabreicht, und 4 Stunden nach der Kaolin-Injektion werden die oben erwähnten Parameter gemessen. Der oedemhemmende Effekt wird in Prozenten angegeben, basierend auf der Differenz der Oedemintensität zwischen unbehandelten und mit der zu untersuchenden Substanz behandelten Tieren, die antinoziceptiva Aktivität durch die prozentuale Erhöhung der Schmerzschwelle.
In diesem Test zeigen Verbindungen der Formel I eine Oedemhemmung und eine Erhöhung der Schmerzschwelle.
Ausserdem hemmen sie - wie dies in einem entsprechenden Standard-Test gezeigt werden kann - die Blutplättchenaggregation und haben demnach auch antithrombotische Eigenschaften.
Die Verbindungen der Formel I besitzen qualitativ eine ähnliche Wirkung wie Phenylbutazon, welches für seine therapeutische Verwendung und Eigenschaften bekannt ist.
Die Verbindungen der Formel I können als Heilmittel, z.B.
in Form pharmazeutischer Präparate Verwendung finden, welche sie in Mischung mit einem für die enterale oder parenterale Applikation geeigneten, pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z.B. Wasser,
Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline usw. enthalten.
Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln, in halbfester Form, z.B. als Salben, oder in flüssiger Form, z.B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und/oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Die nachfolgenden Beispiele, in welchen alle Temperaturen in Celsiusgraden angegeben sind, erläutern die Erfindung.
Beispiel 1
52,1 g Phosphorpentachlorid werden in 600 ml abs. Tetrachlorkohlenstoff gelöst und zum Sieden erhitzt, worauf während 3 Stunden eine Lösung von 15,8 g 3-Hydroxy-2-methoxycarbonyl-thiophen in 200 ml Tetrachlorkohlenstoff zugetropft wird. Es wird 13 Stunden am Rückfluss gekocht, dann wird der Tetrachlorkohlenstoff abdestilliert und das Reaktionsgemisch im Vakuum bis fast zur Trockene eingedampft.
Unter Kühlen werden 450 ml Wasser zugetropft, worauf man zum Sieden erhitzt und dann abkühlen lässt. Das ausgefallene Produkt wird abgesaugt und in einer Lösung von 25 g Natriumhydrogencarbonat mit 10 g Aktivkohle aufgekocht, worauf die Aktivkohle abgesaugt und die erkaltete Lösung mit Salzsäure angesäuert wird, wobei man 3-Chlorthiophen-2-carbonsäure vom Smp. 185-186 erhält.
In einem Glasautoklaven werden 8,6 g 3-Chlorthiophen-2carbonsäure in 23 ml 2,1 g Natriumhydroxid enthaltendem Wasser gelöst, worauf eine Lösung von 5,6 g Natriumhydrogensulfit in 16 ml Wasser zugegeben und die Lösung mit 30%iger Natronlauge gerade alkalisch gemacht wird. Dann wird mit 0,43 g Kupfer(I)chlorid versetzt und 16 Stunden auf 143" erhitzt. Nach dem Erkalten wird das rote Kupferoxid abgesaugt. Hierauf wird mit 7 ml konz. Salzsäure angesäuert, wobei nicht umgesetztes Ausgangsmaterial ausfällt, das durch Ausschütteln mit Methylenchlorid entfernt wird. Die saure Lösung wird unter Erwärmen mit 12 g Kaliumchlorid versetzt, wobei sich nach Kühlen auf 0 das Mono-Kaliumsalz der 3 Sulfo-thiophen-2-carbonsäure in farblosen Kristallen abscheidet.
8,2 g des obigen Mono-Kaliumsalzes der 3-Sulfo-thiophen2-carbonsäure werden in 50 ml Wasser gelöst. Diese Lösung wird durch eine Ionenaustauschersäule, die mit Protonen beladen ist, fliessen gelassen, wobei so lange mit Wasser nachgespült wird, bis der pH-Wert der ausfliessenden Lösung 5 beträgt. Die Lösung wird im Vakuum zur Trockene eingedampft, und der kristalline Rückstand, bestehend aus 3-Sulfothiophen-2-carbonsäure, wird aus wenig Wasser umkristallisiert.
7,6 g 3-Sulfo-thiophen-2-carbonsäure werden in 140 ml abs.
Methanol und 65 ml abs. Chloroform gelöst und am Rückfluss gekocht, wobei das Reaktionswasser als ternäres Azeotrop (Chloroform, Methanol, Wasser) über eine Füllköperkolonne (1 m) abdestilliert wird. Dann wird im Vakuum abgedampft.
Zur Entfernung von Spuren Methanol wird der Rückstand mit 100 ml Chloroform versetzt, worauf bei Normaldruck eingedampft wird. Das verbleibende braune Öl, bestehend aus 3 Sulfo-thiophen-2-carbonsäuremethylester, kristallisiert nach dem Erkalten sofort aus. Die Kristalle sind jedoch hygroskopisch und zerfliessen an der Luft.
7,4 g roher 3-Sulfo-thiophen-2-carbonsäuremethylester werden in 50 ml Thionylchlorid gelöst und 16 Stunden Rückfluss gekocht. Dann wird im Vakuum zur Trockene eingedampft, worauf das verbleibende hellgelbe Öl, bestehend aus 3-Chlorosulfonylthiophen-2-carbonsäuremethylester, mit Petroläther zur Kristallisation gebracht wird.
Die Überführung des Mono-Kaliumsalzes der 3-Sulfothiophen-2-carbonsäure in den 3-Chlorsulfonylthiophen-2-carbonsäuremethylester kann auch wie folgt durchgeführt werden:
50 g (0,203 Mol) Mono-Kaliumsalz der 3-Sulfo-thiophen-2carbonsäure werden in 250 ml Phosphoroxychlorid suspendiert, und unter Rühren werden 85 g (0,406 Mol) Phosphorpentachlorid zugegeben (heftige HCl-Entwicklung setzt ein).
Dann wird noch 90 Minuten auf dem Wasserbad unter Rühren erhitzt, auf Raumtemperatur abgekühlt, von anorganischen Salzen abgesaugt und das Phosphoroxychlorid im Vakuum so gut wie möglich abdestilliert. Der ölige Rückstand wird zur Entfernung von noch vorhandenen anorganischen Salzen in 400 ml trockenem Chloroform gelöst, filtriert und eingedampft. Der ölige Rückstand, bestehend aus 3-Chlorsulfonylthiophenyl-2-carbonsäurechlorid, kristallisiert beim Abkühlen aus und wird ohne weitere Reinigung in die nächste Stufe eingesetzt.
48 g (0,196 Mol) des so erhaltenen 3-Chlorsulfonylthiophen-2-carbonsäurechlorids werden in 500 ml abs. Chloroform gelöst, 9,6 g (0,3 Mol) abs. Methanol zugegeben und 3 Stunden am Rückfluss erhitzt (bis zum Ende der HCl-Entwicklung). Dann wird im Vakuum zur Trockene eingedampft, wobei der aus reinem 3-Chlor-sulfonyl-thiophen-2-carbonsäu- remethylester bestehende Rückstand auskristallisiert. Das Rohprodukt kann in die nächste Stufe eingesetzt werden.

Der kristalline Rückstand wird zur Reinigung mit Äther digeriert. Man erhält 3-Methylsulfamoyl-thiophen-2-carbonsäuremethylester vom Schmelzpunkt 115-122".

Nach dem Trocknen mit Natriumsulfat und Abdampfen des Lösungsmittels erhält man farblose Kristalle, die für die weitere Umsetzung rein genug sind. Die erhaltenen 3-Methylsulfamoyl-thiophen-2-carbonsäure kann aus Wasser umkristallisiert werden. Smp. 182-184".
11,06 g (0,05 Mol) 3-Methylsulfamoyl-thiophen-2-carbonsäure und 15,8 g (0,075 Mol) Phosphorpentachlorid werden in 150 ml abs. Chloroform aufgeschlämmt und 30 Minuten bei 40 gerührt. Dann wird schonend im Vakuum auf das halbe Volumen eingedampft. Man verdünnt mit 400 ml abs. Äther und tropft die erhaltene Lösung im Verlauf von 1 Stunde zu einer auf -20' gekühlten, aus 50 g Nitrosomethylharnstoff hergestellten ätherischen Diazomethanlösung. Nach Entfernen des Kühlbades wird noch 1'/2 Stunden ausreagieren gelassen und dann auf - 40' abgekühlt, worauf das ausgefallene Diazo methyl-(3-methylsulfamoyl-thiophen-2-yl)-keton abgesaugt wird; Smp. 110111' (aus Äthanol).
3 g (0,0122 Mol) Diazomethyl-(3-methylsulfamoyl-thiophen-2-yl)-keton werden in 30 ml abs. Methanol gelöst und nach Zugabe von 50 mg Silbernitrat 45 Minuten am Rückfluss erhitzt. Dann wird im Vakuum eingedampft, worauf man den Rückstand mit Äther mehrmals auskocht, die vereinigten Ätherphasen mit Aktivkohle rührt und filtriert. Das Filtrat enthält zur Hauptsache 3-Methylsulfamoyl-2-thiophenessig- säuremethylester neben wenig 3-Methylsulfamyol-2-thiophenessigsäure und 3,4-Dihydro-2-methyl-3-oxo-2H-thieno[2,3- e]1,2-thiazin-1,1-dioxid. Zur Entfernung der 3-Methylsulfamoyl-2-thiophenessigsäure wird die Ätherlösung mehrmals mit Natriumhydrogencarbonatlösung ausgeschüttelt, getrocknet und eingedampft.
Zur Ringöffnung von 3,4-Dihydro-2 methyl-3-oxo-2H-thieno[2,3-ej 1 ,2-thiazin-1, 1-dioxid wird der soeben erwähnte Eindampfrückstand mit 12 ml in methanolischer Natriummethylatlösung aufgenommen, worauf die Lösung 20 Minuten bei Raumtemperatur stehengelassen, dann mit konz. Salzsäure eben angesäuert und im Vakuum zur Trockene eingedampft wird. Der Eindampfrückstand wird mit Methylenchlorid und Wasser aufgenommen; die organische Phase wird getrocknet und eingedampft. Der ölige Rückstand besteht zu etwa gleichen Teilen aus zwei konformationsisomeren Formen des 3-Methylsulfamoyl-2-thiophenessigsäureme thylesters und kann ohne weitere Reinigung in die nächste Stufe eingesetzt werden.
Die beiden Formen kann man säulenchromatographisch trennen (Kieselgel, Korngrösse 0,063-0,2 mm; Eluens: Benzol/Eisessig = 4:1): Schneller eluierbare Form: 1H-NMR (CDCl3): b = 7,2-7,4 (qas, 2H, H-4Thiophen und

1,15 g (0,00461 Mol) roher 3-Methylsulfamoyl-2-thiophenessigsäuremethylester werden in 4,7 ml In methanolischer Natriummethylatlösung gelöst, worauf man die Lösung im Vakuum zur Trockene eindampft, den Rückstand mit 10 ml abs. Der sofort kristallisierende Rückstand besteht aus reinem 3,4-Dihydro-4-methoxycarbonyl-2-methyl3-oxo-2H-thieno[2,3-e] 1,2-thiazin-1, 1-dioxid; Smp. 124 126 (aus Äther).

Carbon tetrachloride dissolved and heated to boiling, whereupon a solution of 15.8 g of 3-hydroxy-2-methoxycarbonylthiophene in 200 ml of carbon tetrachloride is added dropwise over 3 hours. The mixture is boiled under reflux for 13 hours, then the carbon tetrachloride is distilled off and the reaction mixture is evaporated to dryness in vacuo.
450 ml of water are added dropwise while cooling, and the mixture is heated to boiling and then allowed to cool. The precipitated product is filtered off with suction and boiled in a solution of 25 g of sodium bicarbonate with 10 g of activated carbon, whereupon the activated carbon is filtered off and the cooled solution is acidified with hydrochloric acid, giving 3-chlorothiophene-2-carboxylic acid of mp. 185-186.
In a glass autoclave, 8.6 g of 3-chlorothiophene-2-carboxylic acid are dissolved in 23 ml of water containing 2.1 g of sodium hydroxide, whereupon a solution of 5.6 g of sodium bisulfite in 16 ml of water is added and the solution is made alkaline with 30% sodium hydroxide solution becomes. 8.2 g of the above mono-potassium salt of 3-sulfo-thiophene-2-carboxylic acid are dissolved in 50 ml of water. This solution is allowed to flow through an ion exchange column which is loaded with protons, rinsing with water until the pH of the outflowing solution is 5. The solution is evaporated to dryness in vacuo, and the crystalline residue, consisting of 3-sulfothiophene-2-carboxylic acid, is recrystallized from a little water.
7.6 g of 3-sulfo-thiophene-2-carboxylic acid in 140 ml abs.
Methanol and 65 ml abs. Chloroform dissolved and boiled under reflux, the water of reaction being distilled off as a ternary azeotrope (chloroform, methanol, water) over a packed column (1 m). Then it is evaporated in vacuo.
To remove traces of methanol, the residue is mixed with 100 ml of chloroform and then evaporated at normal pressure. The remaining brown oil, consisting of 3 sulfo-thiophene-2-carboxylic acid methyl ester, crystallized immediately after cooling. However, the crystals are hygroscopic and flow in the air.
7.4 g of crude 3-sulfo-thiophene-2-carboxylic acid methyl ester are dissolved in 50 ml of thionyl chloride and refluxed for 16 hours. The mixture is then evaporated to dryness in vacuo, whereupon the remaining light yellow oil, consisting of 3-chlorosulfonylthiophene-2-carboxylic acid methyl ester, is brought to crystallization using petroleum ether.
The conversion of the mono-potassium salt of 3-sulfothiophene-2-carboxylic acid into the 3-chlorosulfonylthiophene-2-carboxylic acid methyl ester can also be carried out as follows:
50 g (0.203 mol) of mono-potassium salt of 3-sulfo-thiophene-2-carboxylic acid are suspended in 250 ml of phosphorus oxychloride, and 85 g (0.406 mol) of phosphorus pentachloride are added with stirring (vigorous evolution of HCl sets in).
The mixture is then heated on the water bath with stirring for a further 90 minutes, cooled to room temperature, filtered off with inorganic salts and the phosphorus oxychloride is distilled off in vacuo as far as possible. To remove any inorganic salts still present, the oily residue is dissolved in 400 ml of dry chloroform, filtered and evaporated. The oily residue, consisting of 3-chlorosulfonylthiophenyl-2-carboxylic acid chloride, crystallizes on cooling and is used in the next step without further purification.
48 g (0.196 mol) of the 3-chlorosulfonylthiophene-2-carboxylic acid chloride thus obtained are abs in 500 ml. Chloroform dissolved, 9.6 g (0.3 mol) abs. Methanol was added and the mixture was heated under reflux for 3 hours (until the end of the HCl evolution). The mixture is then evaporated to dryness in vacuo, the residue consisting of pure 3-chlorosulfonyl-thiophene-2-carboxylic acid methyl ester crystallizing out. The raw product can be used in the next stage.
43.5 g (0.18 mol) of the 3-chlorosulfonylthiophene-2-carboxylic acid methyl ester thus obtained are dissolved in 450 ml of abs. Chloroform dissolved and introduced at 10 dried methylamine until a moistened pH paper with the solution shows an alkaline reaction. Then the mixture is left to react for a further 2 hours at room temperature, the solution always being to remain alkaline. It is then shaken with 500 ml of water and 500 ml of 5% but sodium hydrogen carbonate solution (the aqueous phases are each shaken back once with chloroform, the combined organic phases are dried with sodium sulfate and evaporated. The crystalline residue is digested with ether for purification. 3-Methylsulfamoyl is obtained -thiophene-2-carboxylic acid methyl ester of melting point 115-122 ".
23.5 g (0.1 mol) of 3-methylsulfamoyl-thiophene-2-carboxylic acid methyl ester are dissolved in 50 ml of ethanol with heating, 50 ml of 3N potassium hydroxide solution are added and the mixture is heated under reflux for 3 hours. After cooling, it is diluted with water and shaken out with methylene chloride. The aqueous phase is acidified with hydrochloric acid and extracted several times with 50 ml ether. After drying with sodium sulfate and evaporation of the solvent, colorless crystals are obtained which are pure enough for the further reaction. The 3-methylsulfamoyl-thiophene-2-carboxylic acid obtained can be recrystallized from water. M.p. 182-184 ".
11.06 g (0.05 mol) of 3-methylsulfamoyl-thiophene-2-carboxylic acid and 15.8 g (0.075 mol) of phosphorus pentachloride are dissolved in 150 ml of abs. Chloroform slurried and stirred at 40 for 30 minutes. Then it is gently evaporated to half the volume in a vacuum. It is diluted with 400 ml of abs. Ether and dropwise the solution obtained in the course of 1 hour to an ethereal diazomethane solution cooled to -20 'and made from 50 g nitrosomethyl urea. After the cooling bath has been removed, the mixture is left to react for a further 1 1/2 hours and then cooled to -40 ', whereupon the precipitated diazo methyl- (3-methylsulfamoyl-thiophene-2-yl) ketone is suctioned off; M.p. 110111 '(from ethanol).
3 g (0.0122 mol) of diazomethyl- (3-methylsulfamoyl-thiophen-2-yl) ketone in 30 ml of abs. Dissolved methanol and heated to reflux for 45 minutes after adding 50 mg of silver nitrate. Then it is evaporated in vacuo, whereupon the residue is boiled out several times with ether, the combined ether phases are stirred with activated carbon and filtered. The filtrate mainly contains 3-methylsulfamoyl-2-thiopheneacetic acid methyl ester in addition to a little 3-methylsulfamyol-2-thiopheneacetic acid and 3,4-dihydro-2-methyl-3-oxo-2H-thieno [2,3- e] 1, 2-thiazine-1,1-dioxide. To remove the 3-methylsulfamoyl-2-thiophenacetic acid, the ether solution is shaken out several times with sodium hydrogen carbonate solution, dried and evaporated.
To open 3,4-dihydro-2 methyl-3-oxo-2H-thieno [2,3-ej 1,2-thiazine-1,1-dioxide, the evaporation residue just mentioned is taken up in 12 ml in methanolic sodium methylate solution, whereupon the solution was left at room temperature for 20 minutes, then with conc. Hydrochloric acid is acidified and evaporated to dryness in vacuo. The evaporation residue is taken up with methylene chloride and water; the organic phase is dried and evaporated. The oily residue consists in approximately equal parts of two conformationally isomeric forms of the 3-methylsulfamoyl-2-thiopheneacetic acid methyl ester and can be used in the next step without further purification.
The two forms can be separated by column chromatography (silica gel, particle size 0.063-0.2 mm; eluent: benzene / glacial acetic acid = 4: 1): Form which can be eluted more quickly: 1H-NMR (CDCl3): b = 7.2-7.4 ( qas, 2H, H-4thiophen and
H-SThiophen), Ï = 4.85.3 (m, 1H, NH, D20 interchangeable), Ï = 4.15 (s, 2H, -CH2-COOCH3), b = 3.7 (s, 3H, OCH3 ), b = 2.65 (d, 3H, NH-CH3,
J = 6Hz).
Slower elutable form: 1H-NMR (CDCl 3): Ï = 7.65 (s, 2H, H-4thiophene and
H-SThiophen), ô = 5.80-6.20 (m, 1H, -NH, D20 interchangeable), b = 4.40 (s, 2H, -CH2-COOCH3), o = 3.40 (s, 3H, OCH3), ö = 2.6 ppm (d, 3H, NH-CH3, J = 6Hz).
Both forms decompose when distilled in a high vacuum.
1.15 g (0.00461 mol) of crude methyl 3-methylsulfamoyl-2-thiophenoacetate are dissolved in 4.7 ml in methanolic sodium methylate solution, whereupon the solution is evaporated to dryness in vacuo, the residue with 10 ml of abs. Dimethylformamide takes up, cooled to 0 "and 0.50 g (0.00525 mol) methyl chloroformate is added dropwise. After stirring for 1/2 hour at room temperature, the mixture is evaporated in vacuo, whereupon the residue is taken up in methylene chloride and water and the organic phase is separated off, dried and The oily residue consisting of pure 3- (N-methoxycarbonyl-N-methyl-sulfamoyl) -2-thiophene-acetic acid methyl ester can be used in the next step without further purification.

1.42 g (0.00461 mol) of crude 3- (N-methoxycarbonyl-Nmethylsulfamoyl) -2-thiophene-acetic acid methyl ester are dissolved in 10 ml of abs. Dissolved dioxane and mixed with 0.21 g (0.00485 mol) of 55% sodium hydride suspension (in mineral oil), whereupon 50 mg of methanol were added and the mixture was stirred at 50-60 for 1 hour. Then it is cooled, acidified with glacial acetic acid and evaporated in vacuo. The residue is taken up in methylene chloride and saturated sodium bicarbonate solution; the organic phase is shaken out several times with sodium bicarbonate solution, whereupon the aqueous phases are combined and concentrated. Hydrochloric acid are acidified. The precipitate is shaken out with methylene chloride, whereupon the organic phase is dried and evaporated.
The immediately crystallizing residue consists of pure 3,4-dihydro-4-methoxycarbonyl-2-methyl3-oxo-2H-thieno [2,3-e] 1,2-thiazine-1,1-dioxide; Mp 124 126 (from ether).
0.3 g (1.1 mmol) of 3,4-dihydro-4-methoxycarbonyl-2-methyl-3-oxo-2H-thieno [2,3-el1,2-thiazine-1,1-dioxide and 0, 13 g (1.38 mmol) of 2-aminopyridine are abs in 30 ml.
Dissolved xylene and heated to reflux for 1 hour. After cooling, the crystals are filtered off and recrystallized from glacial acetic acid; 3,4-dihydro-2-methyl-3-oxo-4- (2 pyridyl-carbamoy -2H-thieno [2,3-e] 1,2-thiazine-1,1-dioxide of mp 255- 256 (dec.).
Example 2
0.15 g (0.55 mmol) of 3,4-dihydro-4-methoxycarbonyl-2-methyl-3-oxo-2H-thieno [2,3-e] 1,2-thiazine-1,1-dioxide and 0.071 g (0.71 mmol) of 2-aminothiazole in 30 ml of abs.
Dissolved xylene and heated to reflux for t / 2 hours. After cooling, the crystals are filtered off and recrystallized from glacial acetic acid; 3,4-Dihydro-2-methyl-3-oxo-4- (2thiazolyl-carbamoyl) -2H-thieno [2,3-e] 1,2-thiazine-1,1-dioxide of mp 236 is obtained. 238 "(dec.).

Using a procedure analogous to that in Example 2, starting from 3,4-dihydro-4-methoxycarbonyl-2-methyl-3-oxo 2H-thieno [2,3-e] -1,2-thiazine-1,1-dioxide and 4-bromoaniline, 4-bromo-3-hydroxy-2-methyl-2H-thieno [2,3-e3-1,2-thiazine-4-carboxanilide-1,1-dioxide, melting point 178179 C (dec. ).

Using a procedure analogous to that in Example 4, but using 4-chloroaniline, 4'-chloro-3hydroxy-2-methyl-2H-thieno [2,3-e] -1,2-thiazine-4-carboxanilide is obtained. 1.1 dioxide of melting point 172-173 "C.
