Verfahren zur Herstellung von neuen Guanidinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Guanidinderivaten, und ihren Salzen mit anorganischen und organischen Säuren.

in welcher Ad den 1-Adamantylrest, X Sauerstoff, Schwefel, die Iminogruppe oder eine nie dere Alkyliminogruppe, alk einen nicht-geminal zweiwertigen, geraden oder ver zweigten aliphatischen Kohlenwasserstoffrest mit 2-6
Kohlenstoffatomen und Ri Wasserstoff oder einen niederen Alkylrest bedeutet, und R$, Rs und R4 Wasserstoff oder höchstens zwei dieser drei Symbole niedere Alkylreste bedeuten, sind bisher nicht bekannt geworden. Wie nun gefunden wurde, besitzen die Verbindungen der Formel I und ihre Salze mit anorganischen und organischen Säu- ren wertvolle pharmakologische Eigenschaften. Insbesondere zeigen sie sympathicolytische Wirkungen, die sich therapeutisch zur Behandlung der Hypertonie nutzen lassen. Ferner sind sie antiviral wirksam.
Die sympathicolytischen Eigenschaften der Verbin dungen der allgem inen Formel I lassen sich z. B. während und anschliessend an ihre intravenöse Infusion an mit Numal narkotisierten Katzen anhand der Reaktion des Blutdrucks auf in bestimmten Zeitintervallen wiederholte Injektionen von 2y/kg i. v. Noradrenalin bzw. von 1 mg'kg i. v. Tyramin nachweisen. Bei der Infusion von N-[ss-(Adamant-l-oxy)-äthyl]-guanidin (A) (12 mg/kg i. v. total innerhalb 2 Stunden) ist der pressorische Effekt des direkt wirkenden Amins, Noradrenalin, verstärkt. Der pressorische Effekt des indirekt, d. h. durch Verdrängung von Noradrenalin aus den Speichern der peripheren Synapsen, wirkenden Amins, Tyramin, ist dagegen während der Infusion vermindert.
Dies entspricht dem, durch die Abschirmung der Noradrenalin-Speicher erklärbaren, sog. Cocain-Effekt.
Bei der Infusion von N- [/ ?- (Adamant-l-yIamino)-äthyl]- guanidin (B) (Dosierung und Dauer wie bei Verbindung A) ist der pressorische Noradrenalin-Effekt ver stärkt, ebenso aber auch der Tyramin-Effekt. Die Maxima der Steigerungen sind phasenverschoben : Die Noradrenalin-Verstärkung erreicht ihr Maximum nach 150 Minuten mit dem Abklingen der Tyramin-Verstär kung, die nach 90 Minuten am ausgeprägtesten ist. Der Verlauf spricht für einen Cocain-Effekt, der von einer starken Noradrenalin-Freisetzung (Depletion of NA) nach Art der Reserpin-Wirkung überlagert ist.
Bei der Infusion von N-[ss-(Adamant-l-ylthio)-äthyl]-guanidin (C) ist im Gegensatz zu obigen Befunden sowohl der pressorische Effekt des direkt wirkenden Noradrenalins wie derjenige des indirekt wirkenden Tyramins vermindert. Die schwächste Reaktion auf beide Amine wird nach je 150 Minuten festgestellt.
Dieser Verlauf spricht für das Vorliegen einer direkten adrenolytischen Wirksamkeit an den Receptoren, die sich von den Wirkungen der Verbindungen A und B auf die Noradrenalin-Speicher deutlich unterscheiden lässt.
In den Verbindungen der Formel I ist eine niedere Alkyliminogruppe X, z. B. eine Methylamino-oder Athyliminogruppe. Als nicht-geminal zweiwertige Reste alk kommen insbesondere der Athylen-, Propylen-und Trimethylenrest, methylsubstituierte Trimethylenreste, der Tetramethylen-, Pentamethylen-oder Hexamethylenrest in Betracht. Die Symbole Ri bis R bedeuten als niedere Alkylreste insbesondere Methyl-oder Athylreste.
Zur Herstellung der Guanidinderivate der Formel I setzt man ein Amin oder ein Säureadditionssalz einer solchen Formzl II,

um und setzt gewünschtenfalls die Base aus dem als unmittelbares Reaktionsprodukt erhaltenen Salz frei und führt gewünschtenfalls die freie Base in ein Salz mit einer anderen anorganischen oder organischen Säure über.
Als Ausgangssteffe der Formel III können sich besonders S-Methyl-isothioharnstoff und N-Alkylderivate desselben in Form mineralsaurer, insbesondere schwefelsaurer Salze eignen. Ferner können z. B.
O-Methyl-isoharnstoff-sulfat und N-Alkylderivate desselben sowie Nitroso-guanidin in Frage kommen. Die Umsetzung kann beispielsweise in Wasser oder in einem wasserlöslichen organischen Lösungsmittel, z. B. einem niederen Alkanol, wie Methanol, Athanol, Propanol, Isopropanol, in einer ätherartigen Fliissigkeit, wie Dioxan, Tetrahydrofuran oder Diathylenglykoldi- methyläther, in einem Keton, wie Aceton oder Butanon, in einer niederen Alkansaure, wie Essigsaure oder in einem niederen aliphatischen Amid, wie Formamid oder Dimethylformamid,
oder in Mischungen solcher Lösungsmittel mit Wasser erfolgen. Die Reaktion kann vorzugsweise unter Erwarmen bis höchstens zur Siedetemperatur des eingesetzten Reaktionsmediums durchgeführt werden.
Die Herstellung von Ausgangsstoffen der Formel II kann, ausgehend von bekannten Adamantanderivaten, auf verschieden-n Wegen erfolgen. Beispielsweise kann zunächst 1-Brom-adamantan mit einem Alkylester einer niederen fl-Hydroxy-oder (X-Mercapto-alkansäure in Gegenwart eines säurebindenden Mittels, wie z. B.
Collidin, erhitzt oder ein Alkylester einer niederen Halogsnalkansäure oder Toluolsulfonyloxy-alkansäure, insbzsondere einer a-oder j-Bromalkansaure oder a oder ss-Toluolsulfonyloxy-alkansäure, mit einer Alkalimetallverbindung des 1-Adamantanols oder 1-Adamantanthiols umgesetzt werden. Die in beiden Fällen entstandenen (Adamant-1-oxy)-alkansäure-alkylester bzw. (Adamant-1-ylthio)-alkansäure-alkylester können in üblicher Weise in die entsprechenden Amide oder niederen N-Alkylamide übergeführt und letztere z. B. mittels Lithiumaluminiumhydrid in einem Ather oder ätherartigen Lösungsmittel, wie z. B. Diäthyläther bzw.
Tetrahydrofuran, zu Aminen der Formel II reduziert werden. Ferner kann man z. B. auch die Amide von niederen ss-(Adamant-l-oxy)-oder ss-(Adamant-l-yl- thio)-alkansäuren dem Hoffmann'schen Abbau unterwerfen oder die weiter oben erhaltenen Alkylester von niederen - (Adamant-1-oxy)- oder - (Adamant-1-yl- thio)-alkansäuren in die entsprechenden Azide überführen und letztere nach Curtius abbauen.
In beiden Fällen erhält man Verbindungen der Formel II mit einem gegebenenfalls durch eine niedere Alkylgruppe substi tuierten Athylenrest als alk und einem Wasserstoff- atom als R.
i. Zur Herstellung von Ausgangsstoffen der Formel II mit der Iminogruppe oder einer niederen Alkyliminogruppe als X kann beispielsweise zunächst das 1-Adamantanamin oder ein N-Alkyl-1-adamantan- amin mit einem Halogenid einer vorzugsweise in a oder/3-Stellung durch Halogen substituierten niedern Alkansäure acyliert und das erhaltene 1-Halogenalka- noylamino-adamantan entweder mit einem Alkalime tallazid oder mit Ammoniak oder einem niederen Al- kylamin umgesetzt werden.
Durch Reduktion der entstandenen 1-Azidoalkanoylamino-adamantane, 1-Ami- noalkanoylamino-adamantane, bzw. 1-Alkylaminoalkanoylamino-adamantane, deren Amidstickstoffatom gegebenenfalls einen niederen Alkylrest trägt, mittels Lithiumaluminiumhydrid kann man die gewünschten Aus gangsstoffe der allgemeinen Formel II erhalten.
Von den verschiedenen weitern Möglichkeiten zur Herstellung von Ausgangsstoffen der Formel II sei als weiteres Beispiel noch die Umsetzung von 1-Adaman- tanamin oder von Metallverbindungen des 1-Adaman- tanols oder 1-Adamantanthiols mit halogensubstituier- ten, niederen Alkansäurenitrilen, wie z. B. Chlor-oder Bromacetonitril, und anschliessende R duktion der erhaltenen Nitrile, z. B. mittels Lithiumaluminiumhy- drid, genannt.
Als Ausgangsstoffe der Formol IV eignen sich bei spielsweise Methyl-isothioharnstoffäther, Methyliso harnstoffdther oder auch die hier in der Isoform dargestellten, entsprechenden Thioharnstoffe bzw. Harnstoffe.
Solche können beispielsweise durch Umsetzung von Aminen der Formel II mit Thiocyansäurz oder einem niederen Alkylisocyanat erhalten werden. Harnstoffe der Formel IV können ferner entstehen, wenn der als Herstellungsverfahren für Amine der Formel II genannte Curtius-Abbau nur bis zur Stufe des Isocyanats geführt und letzteres mit Ammoniak oder einem niede- ren Alkylamin umgesetzt wird. Aus den Thioharnstoffen und Harnstoffen können durch Behandlung mit Alkylierungsmitteln in Gegenwart von Alkalihydroxyden in üblicher Weise die S-bzw. O-Alkyl-derivate erhalten werden.
Die Reaktionskomponente der Formel V kann in jedem Fall Ammoniak oder ein niederes Alkylamin und, falls R in der Reaktionskomponente der allgemeinen Formel IV durch Wasserstoff verkörpert ist, auch ein niederes Dialkylamin sein.
Die neuen Guanidinderivate sind starke Basen. Sie bilden mit anorganischen und und organischen Säuren einsäurige oder, falls X durch die Iminogruppe oder eine niedere Alkyliminogruppe verkörpert ist, auch zweisäurige Salze. Zur Salzbildung eignen sich z. B.
Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwe felsäure, Phosphorsäure, Methanolsulfonsäure, Äthandi sulfonsäure,,/3-Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Apfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure und Man delsäure.
Die neuen Guanidinderivate und ihre nicht-toxischen Salze können oral, rektal oder parenteral verabreicht werden. Unter nicht-toxischen Salzen sind Salze mit solchen S : uren zu verstehen, deren Anionen bei den in Frage kommenden Dosierungen pharmakologisch annehmbar sind, d. h. keine toxischen Wirkungen ausüben. Ferner ist es von Vorteil, wenn die zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind.
In den nachfolgenden Beispielen sind die Tempera- turen in Celsiusgraden angegeben.
Beispiel 1 a) 14, 5 g (69 mMol) Adamant-1-oxyessigsäure, vgl. belgische Patentschrift Nr. 644 741, werden eine Stunde mit 58 ml frisch destilliertem Thionylchlorid zum Rückfluss erhitzt. Nach dem Eindampfen wird der Rückstand mit 100 ml Benzol versetzt, letzteres abgedampft und das zurückbleibende, rohe Säurechlorid in 100 ml absolutem Dioxan gelöst. Diese Lösung wird bei 0 zu 200 ml gesättigter wässriger Ammoniaklö- sung zugetropft, wobei das entstandene Amid ausfällt.
Abfiltrieren, Lösen der Kristalle in 200 ml Methylenchlorid, Trocknen der Lösung über Magnesiumsulfat, Eindampfen und Umkristallisieren des Rückstandes aus Hexan ergibt a- (Adamant-l-oxy)-essigsäureamid als farblose Kristalle vom Smp. 120-123 .
Zu einer zum Rückfluss erhitzten Suspension von 12, 1 g (319 mMol) Lithiumaluminiumhydrid in 2000 ml Ather werden 33, 5 g (160 mMol) a- (Adamant-l-oxy)-essigsäureamid in 340 ml absolutem Tetrahydrofuran getropft. Das Gemisch wird hierauf 12 Stunden unter Rückfluss gekocht, dann abgekühlt, tropfenweise mit 100 ml Athylacetat und schliesslich bis zur sauren Reaktion mit 2-n. Schwefelsäure versetzt. Die wässrige Phase wird abgetrennt, zweimal mit je 500 ml Ather gewaschen und mit konz. Natronlauge und Eis unter 500 ml Ather alkalisch gestellt.
Die wässrige Phase wird zweimal mit je 500 ml Ather extrahiert, die ätherischen Lösungen werden vereinigt, mit Wasser gewaschen, über Kaliumcarbonat getrocknet und eingedampft, wobei A-(Adamant-l-oxy)-äthyl- anrn als Ol zurückbleibt. b) 7, 4 g (38 mMol) oF-(Adamant-l-oxy)-äthylamin und 10, 8 g (76 mMol) feingepulvertes Methylisothiuronium-hydrogensulfat werden in 150 ml Äthanol/Wasser 1 : 1 24 Stunden zum Rückfluss erhitzt. Nach dem Eindampfen auf 75 ml wird das Reaktionsgemisch filtriert, das Filtergut dreimal mit Athanol gewaschen und hierauf in 2-n. Natronlauge gelöst.
Durch dreimalige Extraktion mit je 100 ml Methylenchlorid, Waschen der vereinigten organischen Lösungen mit gesättigter Natri umchloridlösung, Trocknen über Kaliumcarbonat und Eindampfen isoliert man das N-[ss-(Adamant-l-oxy)- äthyl]-guanidin. Dieses wird in 40 ml Methanol gelöst und durch Zugabe von ätherischer Chlorwasserstofflö- sung in das Hydrochlorid übergeführt. Smp. 184-185 .
Beispiel 2 a) 9, 0 g (40 mMol) a- (Adamant-1-ylthio)-essigsaure, Smp. 68-70 , vgl. französische Patentschrift Nr. 1 350 317, werden in 40 ml destilliertem Thionylchlorid 30 Minuten zum Rückfluss erhitzt. Das über- schüssige Thionylchlorid wird unter Vakuum abgedampft, der Rückstand mit Benzol versetzt und das resultierende Gemisch wiederum eingedampft. Das zurückbleibende rohe Säurechlorid wird in 80 ml Dioxan , gelöst und die Lösung bei 0 in 200 ml gesättigte wäss- rige Ammoniak-Lösung eingetropft.
Nach Verdünnen mit Wasser, wird das ausgeschiedene rohe Amid abfiltriert, in 200 ml Methylenchlorid aufgenommen, die Lösung über Magnesiumchlorid getrocknet und eingedampft. Nach dem Trocknen erhält man das a- (Ada- mant-1-ylthio)-essigsäureamid vom Smp. 130-131 .
Eine Lösung von 6, 0 g (27 mMol) a- (Adamant1-ylthio)-essigsäureamid in 300 ml absolutem Äther wird unter starkem Rühren bei Rückflusstemperatur zu 3, 5 g (92Mol) Lithiumaluminiumhydrid in 100 ml absolutem Ather getropft. Nach 24 Stunden Rühren bei Rückflusstemperatur wird auf 0 abgekühlt. Hierauf werden 100 ml Athylacetat und danach 200 ml 2-n. Schwefelsäure zugetropft. Die organische Phase wird abgetrennt und die wässrige Phase zweimal mit je 100 ml Ather gewaschen. Die saure wässrige L¯sung wird darauf unter 300 ml Ather mit konzentrierter Natronlauge alkalisch gestellt, abgetrennt und noch dreimal je 100 ml Ather extrahiert.
Trocknen der vereinigten Extrakte über Kaliumcarbonat und Eindampfen liefert das ss-(Adamant-1-ylthio)-äthylamin als hellgelbes 01 (Struktur durch IR-und NMR-Spektren verifiziert). b) Nach einem zweiten Verfahren wird das vorste hendgenannte Amin wie folgt erhalten :
33, 8 g (0, 2 Mol) 1-Thio-adamantan werden in 500 ml Diäthylenglykoldimethyläther unter Stickstoff auf 150 erhitzt und danach portionenweise mit 36 ml Natriumamid Suspension (Gehalt 11, 7 g Natriumamid, 0, 3 Mol) in Toluol versetzt.
Nach fünfstündigem Erhitzen zum Rückfluss werden innerhalb 5 Minuten 72, 5 g (0, 4 Mol) ss-Brom-propionsäure-äthylester zugetropft und das Gemisch weitere 14 Stunden unter Rückfluss gekocht. Nach dem Abkühlen auf 20 werden 50 ml Athanol zugetropft, das Gemisch in 1000 ml Eiswasser gegossen und schliesslich dreimal mit je 1000 ml Äther extrahiert. Die vereinigten Ätherextrakte werden zweimal mit je 1500mol Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Die Destillation des Rückstandes ergibt den - (Adamant-1-ylthio)-pro- pionsäure-äthylester als helles 01, Kp. 160-165 /0, 7 Torr nD250 1, 5185.
Zur Verse-fun wird der Ester 4 Stunden mit einem Gemisch von 1000 ml Wasser zum Rückfluss erhitzt, die Lösung abgekühlt und eingedampft. Der Rückstand wird mit Aktivkohle und Hyflo (Diatomeenerde) in 500 ml Wasser zum Sieden erhitzt, die Suspension filtriert, das Filtrat abgekühlt und mit konz. Salzsäure angesäuert, wobei die/ ?- (Adamant1-ylthio)-propionsäure kristallin ausfällt. Sie wird abfiltriert, in Methylenchlorid aufgenommen, die L¯sung über Magnesiumsulfat getrocknet und eingedampft. Die zurückbleibende/'- (Adamant-l-yIthio) propionsäure wird durch Umkristallisieren aus Hexan gereinigt, Smp.
108-110', pkmus 6, 8.
2, 4 g (10 mMol) ss (Adamant-1-ylthio)-propion- sÏure werden 10 Minuten mit 12ml Thionylchlorid gekocht. Das bersch ssige Reagens wird abdestilliert, das zurückbleibinde rohe Siurechlorid mit Toluol versetzt und nach dem Abdestillieren des letzteren in 20 ml Aceton gel¯st und bei 10¯ zur L¯sung von 6,5 g (100 mMol) Natriumazid in 20ml Wasser gegeben.
Nach einst ndigem R hren wird das Gemisch mit 200 ml Eiswasser versetzt, mit Natriumcarbonatl¯sung alkalisch gestellt und zweimal mit je 200 ml ¯ther/ Benzol 1 : 1 extrahiert. Die vereinigten Extrakte werden mit 200 ml Eiswasser gewaschen, über Magnesiumsulfat getrocknet und bei 30 bis zu einem Volumen von 50 ml eingeengt. Diese L¯sung wird darauf zu 100 ml siedendem ¯thanol getropft und 2 Stunden unter Rückfluss erhitzt. Beim Eindampfen des Gemisches erhalt man 2, 9 g rohes Urethan, welches mit einer Mischung von 3 g Kaliumhydroxyd, 2, 5 ml Wasser und 50 ml Athylenglykol 3 Stunden unter Rückfluss gekocht wird. Das Reaktionsgemisch wird mit 200 nal Wasser verd nnt, mit 2-n. SalzsÏure sauer gestellt und dreimal mit je 100ml ¯ther/Benzol 1:1 extrahiert.
Die wässrige Phase wird unter Äther mit konz. Natronlauge alkalisch gestellt und nach Abtrennen des Äthers dr imal mit je 100 ml des gleichen Lösungsmittels extrahiert. Nach Trocknen der Ätherextrakte über Kaliumcarbonat und Eindampfen erhält man das/ ?- (Ada- mant-l-ylthio)-äthylamin als 01, identisch in IR-und NMR-Spektren sowie Dünnschichtchromatogramm mit dem unter a) beschriebenen Produkt. c) 7, 0 g (33 mMol), - (Adamant-l-ylthio)-äthyl- amin und 6,9 g (50 mMol) Methylisothiuronium-hydrogensulfat werden in 300ml ¯thanol/Wasser 1:1 wÏhrend 4 Stunden zum Rückfluss erhitzt. Dann wird das Gemisch abgekühlt, mit 300 ml Wasser und 3 g Aktivkohle versetzt, zum Sieden erhitzt und heiss filtriert.
Beim Abkühlen des Filtrats kristallisiert das Sulfat des N-[/S-(Adamant-l-ylthio)-äthyl]-guanidin aus. Es wird abfiltriert, in 100 ml Chloroform suspendiert und mit konz. Natronlauge versetzt. Die organische Phase wird abgetrennt, über Kaliumcarbonat getrocknet und eingedampft. Die zurückbleibende Base wird mit Salzsäure/ Isopropanol versetzt und das N- [- (Adamant-1-ylthio)- äthyl-guanidin-hydrochlorid durch Zusatz von Äther zur Kristallisation gebracht. Smp. 160-165 .
Beispiel 3 a) 11,3g (0,1 Mol) ChloressigsÏurechlorid werden bei 20¯ unter R hren in eine L¯sung von 15,1 g (0,1 Mol) 1-Adamantanamin und 15ml TriÏthylamin in 250 ml Methylenchlorid eingetropft. Nach vierstün dig@m R hren wird die rotgelbe L¯sung im Vakuum eingedampft, der so erhaltene R ckstand in 200ml Benzol gel¯st und mit 2-n. SalzsÏure, Wasser, Kaliumbicarbonatl¯sung und wiederum Wasser gewaschen.
Die organische Phase wird abgetrennt, tuber Magnesi umsulfat getrocknet und eingedampft. Durch Umkristallisieren des R ckstandes aus 100 ml ¯thanol/Was- ser 9:1 erhÏlt man das ChloressigsÏure-N (1-adamantyl) amid vom Smp. 119 121¯.
8, 5 g (37 mMol) Chloressigs*iur2-N- (l-adamantyl)- m'd, g--Ibst in 240 ml Aceton, werden bei 20 mit einer L¯sung von 25g (0,38 Mol) Natriumezid in
120ml Wasser versetzt und 20 Stunden bei 50¯ ger hrt. Dann wird das Aceton abdastilliert und die zu rückbleibende wassrige Lösung dreimal mit je 250 ml Äther extrahiert. Die vereinigten Atherextrakte werden über Magnesiumsulfat getrocknet und eingedampft.
Durch Umkristallisieren des Rückstandes aus 70 ml n-Hexan erhält man das Azidoessigsäure- (l-adamantyl)- amid vom Smp. 93-94 .
8, 0 g (34 mMol) Azidoessigsïure-(l-adamantyl)- amid werden in 160 ml absolutem Äther gelöst, die L¯sung unter kräftigem Rühren zu einer L¯sung von 6, 5 g (172 mMol) Lithiumaluminiumhydrid in 200 ml absolutem Äther getropft und das Ganze 16 Stunden zum Rückfluss erhitzt. Nach dem Abkühlen auf 0 werden unter Rühren zuerst ca. 100 ml Äthylacetat und darauf 200 ml 2-n. Schwefelsäure zugetropft. Die saure wässrige Phase wird vom Sither getrennt, noch zweimal mit je 100 ml Äther gewaschen und schliesslich über 300 ml Methylenchlorid mit konz. Natronlauge und Eis alkalisch gestellt.

Nach dem Abkühlen verdünnt man das Reaktionsgemisch mit 100 ml Äthanol, filtriert es und wäscht das Filtergut mit Äthanol. Die so erhaltenen Kristalle werden mit 60 ml konz. Natronlauge versetzt und mit 200ml Methylenchlorid während 30 Minuten ger hrt.
Die organische Phase wird abgetrennt und die wässrige Phase mit Methylenchlorid nachgewa schen. Die vereinigten Methylenchloridlösungen werden ber Kaliumcarbonat getrocknet und eingedampft.
Das zur ckbleibende N-[¯-(Adamant-1-ylamino)-Ïthyl]guanidin wird mittels ätherischer Chlorwasserstofflösung in das Dihydrochlorid übergeführt, dessen Schmelzpunkt oberhalb 270 liegt.

With the infusion of N- [/? - (Adamant-l-yIamino) -ethyl] - guanidine (B) (dosage and duration as with compound A) the pressor norepinephrine effect is strengthened, but also the tyramine effect . The maxima of the increases are out of phase: the norepinephrine increase reaches its maximum after 150 minutes with the decay of the tyramine increase, which is most pronounced after 90 minutes. The course suggests a cocaine effect, which is superimposed by a strong release of noradrenaline (depletion of NA) in the manner of the reserpine effect.
In the infusion of N- [ss- (adamant-l-ylthio) -ethyl] -guanidine (C), in contrast to the above findings, both the pressor effect of the directly acting noradrenaline and that of the indirectly acting tyramine are reduced. The weakest reaction to both amines is determined after 150 minutes each.

The new guanidine derivatives and their non-toxic salts can be administered orally, rectally or parenterally. Example 2 a) 9.0 g (40 mmol) of α- (adamant-1-ylthio) acetic acid, melting point 68-70, cf. French Patent No. 1,350,317 are refluxed for 30 minutes in 40 ml of distilled thionyl chloride. The excess thionyl chloride is evaporated off in vacuo, benzene is added to the residue and the resulting mixture is again evaporated. The remaining crude acid chloride is dissolved in 80 ml of dioxane, and the solution is added dropwise at 0 to 200 ml of saturated aqueous ammonia solution.
After dilution with water, the precipitated crude amide is filtered off, taken up in 200 ml of methylene chloride, the solution is dried over magnesium chloride and evaporated. After drying, the α- (adamant-1-ylthio) acetic acid amide of melting point 130-131 is obtained.
A solution of 6.0 g (27 mmol) of α- (adamant1-ylthio) acetic acid amide in 300 ml of absolute ether is added dropwise to 3.5 g (92 mol) of lithium aluminum hydride in 100 ml of absolute ether at reflux temperature with vigorous stirring.
