Abstract

일 양상은 올살켄(olsalkene), 이의 제조방법 및 이를 포함하는 혈소판 감소증 예방 또는 치료 조성물 및 이의 용도에 관한 것이다. 일 양상에 따르면, 올살켄이 체내에서 아조레덕타제(AzoR)에 의해 절단되지 않고 빌리버딘 환원효소 B(BLVRB)에 결합하여 빌리버딘(biliverdin, BV)이 빌리루빈(bilirubin, BR)으로 환원을 억제하는 것을 확인했다. 또한, 빌리버딘 환원효소 B의 활성 억제를 통해 혈소판 생성을 촉진하는 것을 확인했다. 따라서, 상기 올살켄은 빌리버딘 환원효소 B의 활성을 억제하여 혈소판 생성을 촉진하는 역할을 통해 혈소판 감소증 치료제, 건강기능식품 등으로 유용하게 활용될 수 있다.

Description

신규 올살라진 유도체 화합물 및 이를 유효성분으로 포함하는 혈소판 감소증 예방 또는 치료용 조성물
본 발명의 일 예는 올살켄(Olsalkene, OSK), 이의 입체 이성질체 또는 이의 약학적으로 허용 가능한 염, 이의 제조방법, 이를 유효성분으로 포함하는, 혈소판 감소증의 예방 또는 치료용 조성물, 및 상기 조성물의 용도에 관한 것이다.
혈소판은 다능성 조혈 줄기세포에서 분화되는 거대핵세포(megakaryocyte, MK)에서 유래한다. 혈소판은 혈액 지혈과 혈전증뿐만 아니라 염증, 신생 혈관 형성, 선천성 면역, 적응 면역 반응, 종양 전이 등 다양한 생물학적 과정에서 중추적인 역할을 한다. 따라서 혈소판 수를 조절하고 특정 혈소판 반응을 제어하는 것은 혈소판 관련 질환을 치료하는 신약의 핵심 목표이다. 혈소판 조절 장애는 혈소판 감소증을 포함한 다양한 출혈 장애를 유발할 수 있다. 혈소판 감소증에 대한 일반적인 치료는 혈소판 생성 유도 또는 혈소판 수혈을 통해 혈소판 수를 증가시키는 것이다. 반대로 혈소판 증가증은 혈소판 감소증과는 대조적으로 혈소판이 혈액 내에 과도하게 많은 상태를 말한다. 원발성(본태성) 및 이차성(반응성) 혈소판증 집단을 대규모 혈소판 전사체 시퀀싱을 통해 분석한 결과, 빌리버딘 환원효소 B(biliverdin reductase B, BLVRB)가 활성 산소 종(ROS)을 조절하여 MK 분화를 통해 혈소판 생산을 조절하는 것으로 밝혀졌다.
헴(heme) 분해 경로에서 BLVRB는 헴 산소화효소(s)-1(유도성 HMOX1) 및 -2(구성성 HMOX2)의 하류에 있는 NAD(P)H를 활용하여 빌리버딘(biliverdin, BV)-IXβ를 빌리루빈(bilirubin, BR)-IXβ로 환원한다. 이 과정의 산물인 BR은 강력한 항산화제 역할을 하며 세포 보호 효과를 나타낸다. 다만, 고농도는 독성을 유발할 수 있다. 따라서 BLVRB의 제어를 받는 BV/BR 산화 환원 주기는 활성 산소 종(ROS)을 조절하는 데 중요한 역할을 한다. BLVRB의 기능상실 돌연변이(BLVRBS111L)를 발현하는 유도 만능 줄기세포(iPSC)를 사용한 연구에 따르면 변형된 CD34+/BLVRBS111L 조혈 줄기세포(HSC)에서 MK 콜로니 형성(CFU-MK)으로 측정한 결과 ROS 축적 및 증식이 많이 증가한 것으로 나타났다. 반면 야생형 BLVRB는 이러한 효과를 나타내지 않았다. 상기 기능 상실 돌연변이체(BLVRBS111L)는 MK 분화를 유도하고, 결과적으로 ROS 축적을 통해 혈소판 생성을 촉진한다. 따라서 BLVRB 활성을 억제하여 항산화제 BR을 효과적으로 제거하면 헴 분해 경로 내에서 BLVRB의 고유한 산화 환원 조절을 활용하여 혈소판 생성을 증가시키는 새로운 전략을 도모할 수 있다.
BLVRB 활성을 억제하기 위해 크산텐 염료와 아크리딘 함유 화합물이 개발되었다. 이러한 화합물은 천연 BLVRB 기질인 플라빈 모노뉴클레오티드(FMN)와 구조가 유사하다. 크산틴 염료 중에서도 에리스로신 B(erythrosin B)와 플록신 B(phloxine B)가 가장 강력한 억제제로 부상했다. 분자 모델링 및 X-선 결정학 연구에 따르면 에리스로신 B와 플록신 B는 BLVRB의 활성 부위에 결합하는 것으로 밝혀졌다. 또한, BLVRB 활성 부위 내의 수소 결합 네트워크가 밝혀졌고 촉매 활성에서 S111 잔기의 중요한 역할이 강조되었다. 흥미롭게도 선형 위버-버크 플롯 분석 결과, 이러한 억제제는 천연 기질인 FMN만큼 깊게 결합하지는 않으나 BLVRB의 결합 포켓과 유사하게 결합하지만 비경쟁적인 방식으로 작용하는 것으로 나타났다. 에리스로신 B와 플록신 B는 모두 식품 착색제로 활용되어 왔으며, 플록신 B는 다양한 그람 양성 박테리아에 대한 항균 활성도 나타냈다. 그러나 에리스로신 B를 만성적으로 투여하면 쥐의 갑상선 종양 촉진과 관련이 있는 것으로 나타났다. 또한, NMR과 동적광산란(DLS) 실험을 통해 에리스로신 B와 플록신 B가 모두 BLVRB의 다중화를 유도하여 치료 효과를 떨어뜨린다는 사실도 확인했다. 따라서 혈소판 장애를 치료하기 위해 BLVRB를 표적으로 하는 신약 후보 물질이 필요하다.

본 발명자들은 올살라진(olsalazine)의 생화학적, 생물물리학적 특성을 유지한 유도체인 올살켄(olsakene)을 개발하였다. 본 발명자들은 올살켄이 기존 올살라진과 달리 체내에서 아조레덕타제(AzoR)에 의해 절단되지 않고 빌리버딘 환원효소 B(BLVRB)에 결합하여 빌리버딘(biliverdin, BV)이 빌리루빈(bilirubin, BR)으로 환원을 억제하는 것을 확인했다. 또한, 빌리버딘 환원효소 B의 활성 억제를 통해 혈소판 생성을 촉진하는 것을 확인했다.
이를 통해 올살켄이 빌리버딘 환원효소 B의 활성을 억제하여 혈소판 생성을 촉진하는 역할을 통해 혈소판 감소증 치료제, 건강기능식품 등으로 유용하게 활용될 수 있다.

상기 용어 "패취제"란 피부에 부착하여 지속적으로 약효를 나타내도록 한 제제를 말한다. 패취제는 먹는 약으로 인한 위장 장애, 간 장애의 부작용의 위험이 낮고, 경구 투여가 곤란한 환자에게도 투여가 가능하며, 간 대사를 거치지 않고 피부를 통해 바로 혈관으로 흡수되므로 경구제에 비해 저용량 투여로 부작용 없이 동일한 약효를 나타낼 수 있다는 장점이 있다.
본 발명의 일 구현예로, 상기 제제는 주사제형, 주입제형, 분무제형, 액상제형 또는 패취제형일 수 있다.
또한, 본 발명은, 올살켄(Olsalkene) 및 이의 약학적으로 허용 가능한 염을 유효성분으로 포함하는, 혈소판 감소증의 예방 또는 개선용 건강기능식품 조성물을 제공한다.

상기 담체, 부형제, 희석제 및 첨가제의 구체적인 예로는 락토즈, 덱스트로즈, 슈크로즈, 솔비톨, 만니톨, 에리스리톨, 전분, 아카시아 고무, 인산칼슘, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 미세결정성 셀룰로즈, 폴리비닐피롤리돈, 셀룰로즈, 폴리비닐피롤리돈, 메틸셀룰로즈, 물, 설탕시럽, 메틸셀룰로즈, 메틸 하이드록시 벤조에이트, 프로필하이드록시 벤조에이트, 활석, 스테아트산 마그네슘 및 미네랄 오일로 이루어진 군으로부터 선택되는 하나 이상일 수 있다.
상기 건강기능식품은 상기 유효성분을 함유하는 것 외에 특별한 제한 없이 다른 성분들을 필수 성분으로서 함유할 수 있다. 예를 들어, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당 알코올일 수 있다. 상술한 것 이외의 향미제로서 천연 향미제 (타우마틴, 스테비아 추출물 (예를 들어, 레바우디오시드 A, 글리시르히진 등)) 및 합성 향미제 (사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.

상기 건강기능식품은 종래에 알려져 있는 혈소판 감소증의 예방 또는 개선용 건강기능식품 또는 새롭게 개발되는 혈소판 감소증의 예방 또는 개선용 건강기능식품과 혼합되어 제공될 수 있다. 상기 건강기능식품이 혈소판 감소증의 예방 또는 개선용 건강기능식품을 더 포함하는 경우, 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양이 혼합되는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.

실시예
실험재료 및 방법
1. 화학 물질
올살라진, 플록신-B, FMN, NADH, NAD+, NADPH, NADP+, 메틸하이드로퀴논(2-MHQ)을 포함한 모든 화학물질은 달리 명시되지 않는 한 Sigma Aldrich (USA)에서 구입했다. 15N-암모늄 클로라이드, 13C-글루코스 및 2H-디메틸설폭사이드(d6 -DMSO)는 Cambridge Isotope Laboratories에서 구입했다.
2. 올살켄(OSK)의 화학적 합성
모든 시약과 출발 물질, 순도 99.9% 이상의 무수 유기 용매는 Sigma Aldrich, TCI에서 조달하여 사용했다. 박층 크로마토그래피(TLC)는 실리카겔 60 F254를 사용하여 Merck 알루미늄 시트에서 수행했으며 자외선과 포스포몰리브산 및 KMnO4 로 염색하여 시각화했다. 화합물의 정제를 위해 Merck 실리카겔 60(230-400 메쉬)에서 컬럼 크로마토그래피를 수행했다. 1H NMR 스펙트럼은 Bruker DRX-400 분광기에서 기록했다. 화학적 이동(δ)은 내부 표준을 기준으로 측정한 백만 분의 1(ppm)로 보고하고 결합 상수(J)는 헤르츠(Hz)로 표시했다. 제품 질량은 Shimadzu(MALDI-TOF) 질량 분석기로 측정했다.
[올살켄의 합성]
하룻밤 동안 자력 교반기로 질소 기체 환경에서 보관된 100ml 두 목 둥근 바닥 플라스크에 담긴 건조 DMF 용매에 5-포밀-2-하이드록시벤조산 (1g, 0.0060 몰), 요오드화 메틸 (1.49 ml, 0.0240 몰) 및 건조된 K2CO3(2.91 g, 0.0210 몰)를 첨가하고 TLC 분석을 통해 반응 진행을 모니터링했다. 완료 후, 조덩어리를 물(3×25ml)로 담금질하고 디클로로메탄으로 추출한 후 컬럼 크로마토그래피법으로 정제하여 메틸 5-포밀-2-메톡시벤조산염(950mg)을 얻었다. 이와는 별도로, 0℃의 THF에서 Zn 분말 (1.01 g, 0.0224 몰)을 취하고 TiCl4 (2.12 g, 0.0112 몰)을 30분 동안 첨가하고 용액을 실온에서 60℃로 2시간 동안 연속적으로 가열하고 다시 반응 혼합물, 메틸 5-포밀-2-methoxybenzoate (500 mg, 0.0028 몰)를 THF에 한 방울씩 첨가하고 4시간 동안 환류시킨 후 K2CO3로 추가로 담금질하고 DCM 제공 (E)-디메틸 5,5'-(에텐-1,2-디일) 비스(2-메톡시벤조산염) (150 mg)으로 추출했다.
-78℃의 DCM에서 (E)-디메틸 5,5'-(에텐-1,2-디일)비스(2-메톡시벤조산염) (100 mg, 0.0003 몰) 용액에 BBr3 (240 μL, 0.0014)를 10ml 둥근 바닥 플라스크에 넣고 5시간 동안 실온에서 교반했다. 그런 다음 담금질을 위해 1M HCl 1ml를 첨가하고 15분 동안 잘 저어 준 다음 용매를 감압으로 제거하고 조덩어리를 에틸 아세테이트에 용해시키고 물과 소금물로 두 번 세척했다. 생성된 혼합물을 95% 에탄올에 용해시킨 다음 수산화 칼륨 용액을 첨가하여 환류 조건에서 3시간 동안 교반했다. 침전이 발생한 후, 반응 혼합물을 다시 1M HCl과 염수 용액으로 산성화하여 추출하고 에틸 아세테이트 용매에서 재결정화했다. 마지막으로 (E)-5,5'-(에텐-1,2-디일)비스(2-하이드록시벤조산)(60mg, 72%)의 무색 결정 고체를 얻었고 1H NMR 및 질량 스펙트럼 분석을 통해 추가로 확인했다. 1H NMR (CDCl3, 400MHz) δ6.96 (d, 2H, J = 8.64 Hz), 7.12 (s, 2H), 7.79 (d, 2H, J = 8.56 Hz), 7.96 (s, 2H); C16H12O6 : 300.27, 각각 322.5044 (M+Na) 발견됨에 대한 MALDI-TOF m/z calcd.
3. AzoR의 발현 및 부분 정제
대장균 DH5α 세포를 LB 배지에서 배양하여 37℃에서 AzoR 단백질을 얻었다. 다양한 친수성 퀴논은 AzoR 단백질을 암호화하는 acpD 유전자의 mRNA 수준을 향상시키기 때문에 600nm(OD600)에서 세포의 광학 밀도가 0.8에 도달했을 때 0.5mM 메틸하이드로퀴논(2-MHQ)을 배양액에 첨가했다. 세포를 3~4시간 후 원심분리기로 수확하고, AzoR 단백질을 농축하기 위해 Hitrap-Q HP 컬럼(Cytiva)을 사용한 음이온 교환 컬럼 크로마토그래피를 수행하여 컬럼을 완충액(pH 8.0, 25mM Tris-HCl)으로 사전 보정한 다음 1.0M NaCl 구배를 적용하여 단백질을 용출시켰다. 용출 분획의 AzoR 활성은 0.1mM 메나디온과 0.1mM NADH를 사용한 효소 활성 분석으로 측정하였으며, 340nm에서 흡수가 감소하는 것을 이전에 보고된 방법과 같이 측정했다. 메나디온의 0.1mM 스톡 용액을 100% DMSO에 준비했다. 용출 분획은 분석 전에 100배 희석했다. 마지막으로, 가장 높은 활성의 용출 분획을 인산염 기반 식염수(PBS) 완충액에 1000배 희석한 후 올살라진의 절단 분석에 사용했으며, 여기에 1% DMSO를 추가로 첨가하여 0.1mM 메나디온을 용해시켰다.
4. BLVRB의 발현 및 정제
pET-21b 단백질 발현 벡터를 사용하여 대장균 BL21 DE3 균주에서 N-말단 His6-태그 BLVRB를 발현했다. 형질 전환된 대장균 세포를 37℃에서 LB 및 M9 배지에서 배양하여 각각 비표지 및 동위원소 표지(15N 또는 13C/15N) BLVRB 단백질을 발현시켰다. 단백질 발현을 유도하기 위해 배양액이 OD600 0.8에 도달하면 0.5mM IPTG를 첨가한 후 3~4시간 후 세포를 채취하여 완충액(pH 8.0, 25mM Tris-HCl, 500mM NaCl, 10mM β에서 초음파 처리하여 세포를 용해시켰다. BLVRB 단백질은 먼저 HisTrap HP 컬럼(Cytiva)을 사용하여 His-tag 친화성 컬럼 크로마토그래피로 정제했다. His-tag를 제거하기 위해 단백질 용출물을 트롬빈(단백질 mg당 ~5단위)을 첨가한 후 4℃에서 하룻밤 동안 완충액(pH 8.0, 50mM Tris-HCl, 1mM 디티오트레이톨)에서 투석했다. 트롬빈이 pH 8.0에서 HiTrap-Q HP 컬럼에 결합하였으므로 완충액(pH 8.0, 25mM Tris-HCl 및 1mM 디티오트레이톨)으로 이온 교환 컬럼 크로마토그래피를 수행한 다음 1.0M NaCl 구배로 용출을 수행하여 각각 ~150 및 ~300mM NaCl에서 BLVRB와 트롬빈을 용출시켰다. 완전히 절단된 BLVRB에서 His6-태그가 부착된 BLVRB를 분리하기 위해 HisTrap HP 컬럼을 HiTrap-Q 컬럼의 옆에 나란히 부착했다. BLVRB 단백질은 비완충용액(50mM NaCl 및 1mM 디티오트레이톨)과 함께 HiLoad 16/600 Superdex 75(Cytiva)를 사용하여 크기 배제 크로마토그래피(SEC)로 추가 정제했다. 단백질 분획은 10kDa MWCO(Millipore)의 초원심 필터를 사용하여 농축한 다음 사용하기 전에 -70℃에서 보관했다.
정제된 BLVRB를 완충액(pH 7.5, 25mM Tris-HCl, 10mM β메르캡토에탄올 및 6.0M 요소)에서 변성시킨 다음 HisTrap HP 컬럼에 적용했다. 완충액(pH 5.0, 25mM Na-아세테이트, 10mM β메르캅토에탄올 및 6.0M 요소)으로 세척한 후 추가 1.0M NaCl 구배로 용출시켰다. 완충액(pH 5.0, 20mM Na-아세테이트, 10mM β메르캅토에탄올 및 6.0M 요소)으로 2배 희석한 후, HiTrap-SP 컬럼(Cytiva)을 사용하여 양이온 교환 컬럼 크로마토그래피로 apo-BLVRB를 추가로 정제했다. 단백질 용출은 6.0 M 요소의 존재 하에서 1.0 M NaCl 구배로 수행되었다. 단백질 용출물은 완충액(pH 6.5, 50mM Bis-tris, 150mM NaCl 및 1mM DTT)에서 투석하여 재접합했다. 원심분리기로 응집물(~20%)을 제거한 후, 완충액(pH 6.5, 10mM Bis-tris, 50mM NaCl, 1mM DTT)과 함께 HiLoad 16/600 Superdex 75를 사용하여 SEC로 정제했다. apo-BLVRB 용액을 농축한 다음 -70℃에서 보관했다. 냉동 샘플을 해동하면 apo-BLVRB가 약 10% 침전되지만, 구조의 유지 여부는 1H-15N HSQC 스펙트럼을 통해 확인했다.
5. BLVRB:OSK 복합체의 결정화 및 구조 결정
1mM NADPH+를 BLVRB 단백질 용액에 첨가한 다음, HiLoad 16/600 Superdex 75를 사용하여 완충액(pH 8.0, 20mM Tris-HCl, 150mM NaCl 및 1mM DTT)에서 Size exclusion chromatography로 추가 정제했다. 단백질 분획은 10kDa MWCO의 초원심 필터를 사용하여 농축했다. 단백질 상등액(14.8 mg/ml)을 적분한 다음 사용하기 전에 -70℃에서 보관했다.
BLVRB 결정은 18℃에서 행잉 드롭 방법 증기 확산법을 사용하여 완충액(pH 6.5, 0.1 M 비스-트리스, 1.9~2.0 M 황산암모늄)에서 성장시켰다. OSK 분자를 담그기 위해 결정은 1mM OSK와 0.2mM NADP+를 포함한 동일한 결정화 완충액에서 18℃, 1일 동안 배양했다. OSK 스톡 용액(100mM)은 100% DMSO로 제조되었기 때문에 1% DMSO가 침지 용액에 추가로 존재했다. BLVRB:OSK 복합체의 결정은 20~25% 글리세롤을 동결 보호제로 사용한 액체 질소에서 루프 및 급속 냉동되었다.
X-선 회절 데이터는 포항 가속기 연구소의 빔라인 5C에서 수집되었다. 회절 데이터는 HKL-2000 프로그램을 사용하여 처리한 후, 이전에 보고된 BLVRB 좌표(PDB 코드, 7ERA)를 사용하여 분자 치환(MR)을 통해 구조를 풀었다. 구조 모델은 Coot 프로그램을 사용한 모델 구축과 PHENIX 소프트웨어 패키지를 사용한 세분화를 번갈아 가며 개선했다. 구조 시각화는 Chimera 프로그램을 사용하여 수행했다. BLVRB와 복합된 OSK 및 OSA에 대한 상호 작용 네트워크의 개략도는 LIGPLOT을 사용하여 생성되었다.
6. 효소 활성 분석.
효소 동역학은 SpectraMax Gemini EM Microplate Reader (Molecular Devices)로 340nm에서 NADPH의 흡광도를 사용하여 모니터링했다. 효소 반응은 다양한 농도의 억제제(OSK 및 OSA)가 있는 상태에서 0.5% DMSO가 포함된 PBS 버퍼에 0.5 μM BLVRB, 100 μM FMN 및 100 μM NADPH를 넣어 수행했다. 반응의 초기 속도는 5분 동안 수집한 데이터로부터 추정하고 마이클리스-멘텐 방정식에 맞췄다. 억제 모델과 억제 상수는 비선형 최소 제곱 회귀 피팅의 사내 Python 스크립트를 사용하여 결정했다.
7. 등온 적정 열량 측정(ITC)
ITC(Isothermal Titration Calorimetry) 실험은 25℃의 완충액(pH 6.5, 50mM Bis-tris, 50mM NaCl, 0.1mM TCEP)에서 Microcal Auto-iTC200(Malvern Instruments)을 사용하여 수행했다. 열량 측정 셀(200 μl)과 주사기(40 μl)에 각각 0.09 mM BLVRB와 1.1mM 약물(OSK 또는 OSA)을 로드했다. ITC 데이터는 동일한 조건에서 측정되었지만 BLVRB가 없는 상태에서 측정된 블랭크 주사의 열을 뺀 후 분석되었다. 데이터 처리는 제조사에서 제공한 Origin 소프트웨어를 사용하여 수행되었으며, 피팅은 단일 사이트 결합 모델로 수행되었다.
8. NMR 실험
모든 NMR 실험은 25℃에서 Bruker 800MHz 분광기를 사용하여 수행했다. 0.2mM 15N-표지 BLVRB 또는 apo-BLVRB를 완충액(pH 6.5, 50mM Bis-tris, 50mM NaCl, 5% D2O)에서 제조하고 약물 분자의 유무에 따라 1H-15N 이종 핵 단일 양자 일관성(HSQC) 스펙트럼을 기록했다. 효소 반응을 모니터링하기 위한 1차원(1D)1 H 스펙트럼은 PBS 버퍼에서 수행했다. 화학적 이동 섭동(CSP) 데이터는 (Δ2 + [(Δ2의 제곱근 공식을 사용하여 1H 및 15N의 CS로부터의 상대적 효과를 정규화하여 제시했다. 확산 주문 분광법(DOSY) 스펙트럼은 80밀리초 확산 지연, 2초 이완 지연, 32개의 기울기 강도 증분으로 ledbpgppr2s를 사용하여 측정했다.
실험결과
1. 안정성 향상을 위해 디아제닐을 알켄 결합으로 치환하여 아조레덕타제 절단 방지 확인
본 발명자들은 부분적으로 정제된 대장균(E. coli) AzoR 단백질을 사용하여 OSA가 5-아미노살리실산(5-ASA)으로 절단되는 것을 테스트했다(도 1). 세포 배양에 메틸하이드로퀴논(2-MHQ)을 첨가하여 AzoR의 발현을 향상시켰다.
그 결과, AzoR과 NADPH를 함께 OSA를 배양하면 OSA의 디아제닐 결합이 절단되고 OSA의 원래 용도인 장 질환의 치료 효과를 담당하는 활성 화합물인 5-ASA가 형성된다(도 2). 또한, AzoR 촉매의 산물인 5-ASA와 BLVRB의 결합이 현저히 감소한다(도 3).
이를 위해 본 발명자들은 트랜스 구성을 유지하면서 디아제닐 결합을 알켄 결합으로 대체하는 새로운 합성 방식을 개발했다.
그 결과, 올살켄(OSK)이 생성되었으며, 이 올살켄은 그대로 유지되고 AzoR에 의해 절단되지 않은 것을 확인했다(도 4).
2. BLVRB과 OSK 결합의 열역학적 분석
일정한 온도에서 BLVRB-OSK 복합체가 형성되는 동안 발생하는 열을 직접 측정하는 등온 적정 열량계(ITC)를 사용하여 ΔH 및 ΔS와 같은 열역학적 양을 특성화하였다.
그 결과, OSK의 해리 상수(KD)는 140nm로 밝혀졌으며(표 1, 도 5), 이는 OSK가 BLVRB에 대한 강력하고 효율적인 결합체 역할을 한다는 것을 나타냈다. 이전에 확인된 크산텐 기반 억제제인 플록신 B와 비교 분석한 결과, OSK가 더 긴밀한 결합 친화력을 보이는 것으로 나타났다. 또한, ITC 데이터에서 관찰된 엔탈픽 기여도에서 알 수 있듯이 OSK는 잔틴 기반 억제제에 비해 BLVRB와 더 구체적인 상호작용을 나타냈다(표 1). OSK의 결합은 주로 엔탈피 변화에 의해 주도되며, 이는 BLVRB와 화합물 간의 특정 상호작용을 반영한다. OSA와 OSK 사이의 열역학적 값의 차이는 디아제닐 결합을 알켄 결합으로 치환함으로써 설명할 수 있다. 요약하면, ITC의 연구 결과는 BLVRB와 OSK 사이에 특정한 상호작용이 존재한다는 것을 뒷받침한다.

단백질 기반 NMR 분광법은 저분자 및 단백질과 관련된 상호작용을 포함한 분자 간 상호작용을 식별하는 데 매우 효과적인 방법으로 각광받고 있다. 또한 단백질 기반 NMR 방법은 복합체 내에서 저분자의 위치를 최적화하는 데 중요한 결합 부위에 대한 귀중한 통찰력을 제공한다. 일반적으로 여기에는 1H-15N HSQC 스펙트럼의 화학적 이동 변화를 모니터링하는 것이 포함된다. 아미드 공명을 할당하는 것은 HSQC 스펙트럼을 해석하는 데 필수적이며, 이는 백본 공명 할당의 95%를 제공하는 HNCA 및 HN(CO)CA 스펙트럼에 의해 수행된다.
이에 본 발명자들은 결합 시 화학적 이동 변화를 모니터링하기 위해 OSK를 점진적으로 증가된 양으로 적정하여 15N-표지된 BLVRB를 활용한 1H-15N HSQC 실험을 수행했다. 이전 연구를 통해, BLVRB의 FMN 결합 부위는 OSA 결합에 활용되고, 그 중 BLVRB의 촉매 메커니즘에서 Ser111이 주요 잔기임을 참고했다.
이에 HSQC 스펙트럼에서 아미드 화학적 이동 변화를 모니터링한 결과, OSK가 BLVRB에 결합하면 FMN 결합 부위의 아미드 공명에서 변화를 유도하는 것을 관찰했다(도 6A). 또한, 화학적 이동 차이(CSP, chemical shift perturbation)을 분석하고(도 6B) OSK를 BLVRB 표면 구조(도 6C)에 매핑하여 OSK의 BLVRB 표면 결합 효과를 검증하는 데 성공했다(도 6D). 이 분석을 통해 FMN 결합 부위, 특히 FMN의 크산틴 고리가 OSK의 결합 부위임을 명확히 확인했다.
4. BLVRB-OSK 복합체의 X-선 결정 구조 분석
본 발명자들은 올살켄이 BLVRB과의 결합을 상세히 분석하기 위해 BLVRB-OSK 복합체의 결정을 통해 복합체의 구조를 분석하였다.
그 결과, NADP+와 OSK에 결합된 BLVRB의 결정 구조는 1.7Å의 해상도로 결정되었다. 결정은 공간 그룹 P21212에 속하며, 각 결정 비대칭 단위는 두 개의 BLVRB 분자를 포함한다.
OSK와 복합체를 이룬 BLVRB의 전체 구조는 이전에 결정된 구조와 매우 유사하다. 예를 들어, 복합체의 백본 RMSD는 BLVRB-NADP+-FMN(PDB ID: 1HE4) 및 BLVRB-NADP+-OSA(PDB ID: 7ERA) 복합체와 비교하여 각각 1.06 및 0.17 Å로, OSK와 복합된 BLVRB와 기본 기판 및 OSA와 복합된 BLVRB 간의 구조적 유사성이 높다는 것을 나타냈다(도 7A).
구체적으로, OSK의 결합 부위는 NADP+ 보조 인자의 니코틴아마이드 모이티와 밀접하게 연관되어 있으며, FMN의 잔틴 고리는 플록신 B 및 에리스로신과 같은 잔틴 기반 억제제와 유사한 방향을 채택하고 있다(도 7B). OSK 결합의 주요 원동력은 NADP+ 의 니코틴아마이드 모이티와 OSK 사이의 링 스택 상호작용과 잔기 S111, F113, W116, L125, V128, P152 및 H153과 관련된 소수성 상호작용이다(도 7C). 또한, 양전하를 띠는 잔기 R78, K120, R124, R170, K178에 의해 유리한 상호작용이 촉진된다. 특히, OSK는 W116의 측쇄(FMN 복합체에서 관찰된 바와 같이)를 약 2.5Å 정도 결합 포켓 쪽으로 이동시키고, R78의 측쇄는 OSK와 수소 결합을 형성하여 약물 화합물의 결합을 잠재적으로 향상시킨다. 또한, R78의 배향은 약물 화합물과 BLVRB 사이의 소수성 상호작용을 강화할 수 있다.
5. 올살켄을 사용하여 BLVRB 활성 저해 확인
억제제 OSK 및 OSA를 사용하여 NADPH가 있을 때 BLVRB의 효소 동역학을 조사하였다. BLVRB의 OSK 또는 OSA에 의한 억제 메커니즘을 도 8A에 나타냈다. (이때, E는 BLVRB, I는 OSK 또는 OSA를 의미한다.)
그 결과, 선형 위버-버크 플롯은 경쟁적(Kic) 및 비경쟁적(Kiu) 억제 상수를 특징으로 하는 혼합 억제 패턴을 보여주었다(도 8B 내지 8C). OSK의 선형 위버-버크 플롯은 4.80 ± 0.16 μM/min의 최대 속도(Vm)와 61.73 ± 5.28 μM의 마이클리스 상수(Km)를 보여주었다. 559.95 ± 100.98 nM의 경쟁 억제 상수(Kic)는 OSK가 BLVRB의 활성 부위에 결합하기 위해 기질(FMN)과 경쟁한다는 것을 시사한다. 한편, 606.14 ± 61.32 nM의 비경쟁 억제 상수(Kiu)는 OSK가 NADP+ 및 NADPH 복합체 BLVRB에 모두 결합하여 그 형태를 변경하고 효소 활성을 억제한다는 것을 나타낸다(도 8B). OSA의 경우, 4.71 ± 0.09 μM/min의 Vm 및 61.57 ± 2.75 μM의 Km로 유사한 동역학이 관찰된다. 경쟁적(Kic = 551.83 ± 38.65 nM) 및 비경쟁적(Kiu = 787.53 ± 55.17 nM) 억제 상수는 OSA도 활성 부위 결합을 위해 기질과 경쟁하며 NADP+ 및 NADPH 복합체 BLVRB와 상호 작용한다는 것을 시사한다(도 8C).
전반적으로 OSK와 OSA에서 관찰된 혼합 억제 패턴은 이러한 억제제가 NADP+ 및 NADPH 복합체 BLVRB 모두에 결합하여 BLVRB 활성에 영향을 미칠 수 있음을 시사한다. 또한, OSK와 OSA는 모두 잔틴 기반 억제제인 플록신-B와 구별되는 아포-BLVRB에 결합할 수 있으며, 이는 OSK의 더 나은 억제 활성을 의미한다(도 9).
이러한 실험결과들은, 본 발명의 올살켄이 BLVRB의 활성을 억제하여 혈소판 감소증 예방 및 치료에 유용하게 이용될 수 있음을 의미하는 것이다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해되어야 한다.

Claims (12)

  1. 하기 화학식 1로 표시되는 올살켄(Olsalkene), 이의 입체 이성질체 또는 이의 약학적으로 허용가능한 염.

    5-포밀-2-하이드록시벤조산(5-formyl-2-hydroxybenzoic acid)을 요오드화 메틸과 반응시켜 중간체를 제조하는 단계;
    상기 중간체와 TiCl4를 반응시켜 하기 화학식 2로 표시되는 화합물을 제조하는 단계; 및
    하기 화학식 2로 표시되는 화합물과 BBr3을 반응시키는 단계.
