Abstract

The present invention relates to a transparent liquid-type cosmetic composition containing an azelaic acid and a sodium base, and having a high azelaic acid content. A transparent liquid-type cosmetic composition according to the present invention not only has a high azelaic acid content due to including a sodium base, an amino acid, and a polyol that increase the solubility of azelaic acid, but is also optimized for troubled skin, provides a refreshing feeling, and has a skin moisture improvement effect.

본 발명은 아젤라산 및 소듐염기를 포함하는 투명 액상 타입의 아젤라산 고함량 화장료 조성물에 관한 것으로, 더욱 상세하게는, 아젤라산의 용해도를 증가시키는 소듐염기, 아미노산 및 폴리올을 포함하여 아젤라산을 고함량 함유하는 투명 액상 타입의 화장료 조성물에 관한 것이다.The present invention relates to a transparent liquid type cosmetic composition containing azelaic acid and a sodium base, and more particularly, to a transparent liquid type cosmetic composition containing azelaic acid in a high content, including a sodium base, an amino acid, and a polyol that increase the solubility of azelaic acid.

아젤라산 (Azelaic acid)은 여드름, 색소 침착, 탈모, 주름, 다한증, 염증성 피부 질환 및 감염성 피부 질환을 포함하는 광범위한 생리학적 만성 질환을 완화하기 위하여 사용되어 왔다.Azelaic acid has been used to alleviate a wide range of physiological and chronic conditions, including acne, pigmentation, hair loss, wrinkles, hyperhidrosis, inflammatory skin diseases, and infectious skin diseases.

현재 알려져 있는 아젤라산의 제형은 분산액 제형으로 분산액은 아젤라산을 용해되지 않은 상태로 전달하고, 용해되지 않은 아젤라산은 피부에 도포되었을 때 쉽게 흡수되지 않으며, 그 결과 효과를 나타내기 위해서는 과량의 아젤라산이 필요하다.Currently known formulations of azelaic acid are dispersion formulations, which deliver azelaic acid in an undissolved state. Undissolved azelaic acid is not readily absorbed when applied to the skin, and as a result, an excessive amount of azelaic acid is required to exhibit an effect.

따라서, 완전하게 용해된 아젤라산 조성물이 요구되며, 용해된 아젤라산은 용해되지 않은 형태보다 더 쉽게 흡수되기 때문에 피부를 덜 자극시키므로 피부 자극에 대한 부작용의 위험을 감소시킬 수 있다.Therefore, a fully dissolved azelaic acid composition is required, as dissolved azelaic acid is more readily absorbed than the undissolved form and is therefore less irritating to the skin, thus reducing the risk of adverse skin irritation effects.

아젤라산은 물, 화장용 오일 및 알코올에 다소 용해되긴 하지만, 이들 용매는 각각 심각한 한계를 가지고 있다. 물은 아젤라산을 극히 미량 용해시킬 수 있으므로 물과 아젤라산을 포함하는 용액은 최대 약 0.24 중량% (w/w)의 아젤라산만을 함유할 수 있으며, 이것은 효과를 나타내기에 충분하지 못하다. 그리고, 화장용 오일에도 아젤라산은 거의 또는 전혀 용해되지 않는다. 또한, 알코올은 좋은 용매이긴 하지만 국소용 조성물 중에 다량의 알코올, 예를 들어, 이소프로필 알코올이 존재하는 경우 피부를 건조시킬 수 있는 부작용을 나타내기 때문에 만족스러운 효과를 발휘하기 어렵다. 실제로, 일부의 알코올, 예를 들어, 에틸 알코올은 표준 온도 및 대기압 조건에서 아젤라산을 불안정하게 만드는 경향이 있어서, 그 결과 전체적으로 피부에 효과적인 화장료 조성물을 제조하기 힘든 실정이다.Although azelaic acid is somewhat soluble in water, cosmetic oils, and alcohols, each of these solvents has significant limitations. Water can only dissolve azelaic acid in very small amounts, so a solution containing water and azelaic acid can contain only about 0.24% by weight (w/w) of azelaic acid, which is insufficient to be effective. Furthermore, azelaic acid is also poorly or completely soluble in cosmetic oils. Furthermore, while alcohol is a good solvent, the presence of large amounts of isopropyl alcohol in topical compositions can negatively affect the skin, hindering its effectiveness. In fact, some alcohols, such as ethyl alcohol, tend to destabilize azelaic acid under standard temperature and atmospheric pressure conditions, making it difficult to formulate cosmetic compositions that are effective on the skin as a whole.

이에, 물, 사과 펙틴 및 해바라기유를 기초로 10 내지 20% 농도의 아젤라산을 함유하고 있는 에멀젼이 베로바 등에 의해 문헌에 발표된 바 있으며 (Berova, N., et al., in Hypoallergic Cosmetic Emulsion with Azelaic Acid for Prophylaxy and Treatment of Acne Vulgaris, Berova, N., Nkiolova, A., Kratchanov, Chr., and Popova, M., Journal of Applied Cosmetology, vol. 12, no. 3, p. 51 (1994)), 베로바 등은 그들이 만든 제형이 자극이 적은 것은 아젤라산 비히클에 비-천연 물질 대신 사과 펙틴 및 해바라기유와 같은 천연 성분을 사용하였기 때문이라고 판단하였다. 다만, 베로바 등에 의해 개시된 에멀젼은 아젤라산이 불완전하게 용해된 것이므로 제형 내의 아젤라산이 차지하는 중량 백분율은 화장료 조성물에 요구되는 것보다 훨씬 더 높다.Accordingly, an emulsion containing 10-20% azelaic acid in a concentration based on water, apple pectin and sunflower oil was published in the literature by Berova et al. (Berova, N., et al., in Hypoallergic Cosmetic Emulsion with Azelaic Acid for Prophylaxy and Treatment of Acne Vulgaris, Berova, N., Nkiolova, A., Kratchanov, Chr., and Popova, M., Journal of Applied Cosmetology, vol. 12, no. 3, p. 51 (1994)), and Berova et al. attributed the less irritating nature of their formulation to the use of natural ingredients such as apple pectin and sunflower oil instead of non-natural substances as the azelaic acid vehicle. However, since the emulsion disclosed by Veroba et al. has incompletely dissolved azelaic acid, the weight percentage of azelaic acid in the formulation is much higher than that required for a cosmetic composition.

확인 결과, 도 1, 도 2 및 상기 표 4에 제시된 바와 같이, 소듐염기 및 아미노산을 포함하는 실시예 1 내지 실시예 4의 화장료 조성물은 상온 (25℃) 이상의 온도뿐만 아니라, 저온 (4℃)의 온도 조건에서도 석출되는 물질이 없는 것을 확인하였으며, 비교예 1 내지 비교예 3 및 비교예 6 내지 비교예 8의 화장료 조성물은 고온 (45℃) 이상의 온도에서는 석출되는 물질이 없었으나, 상온 (25℃) 및 저온 (4℃)의 온도 조건에서는 석출되는 물질이 존재하고, 비교예 4 및 비교예 5의 화장료 조성물은 상온에서 재용해되어 다른 비교예 보다는 우수하고 화장료 조성물로 이용이 가능하였다.As a result of the verification, as shown in FIG. 1, FIG. 2 and Table 4 above, it was confirmed that the cosmetic compositions of Examples 1 to 4 including a sodium base and an amino acid did not have substances precipitated not only at a temperature higher than room temperature (25°C) but also at a low temperature (4°C), and the cosmetic compositions of Comparative Examples 1 to 3 and Comparative Examples 6 to 8 did not have substances precipitated at a temperature higher than high temperature (45°C), but substances precipitated were present at temperature conditions of room temperature (25°C) and low temperature (4°C), and the cosmetic compositions of Comparative Examples 4 and 5 were redissolved at room temperature and were superior to the other comparative examples and could be used as cosmetic compositions.

또한, 동일한 양의 아젤라산을 포함하는 실시예 1과 비교예 1 내지 3의 화장료 조성물을 비교하면, 화장료 조성물에 포함되는 염기가 소듐염기인지 여부에 따라 저온 (4℃) 및 상온 (25℃)의 온도 조건에서 석출 여부에 차이가 있으므로, 소듐염기가 아젤라산의 용해도를 증가시키는 효과가 있음을 확인하였다. 이 때, 비교예 2 및 비교예 3에서 아젤라산을 용해시키기 위해 계면활성제를 했음에도 실시예 1에서 용해도가 현저히 우수하였다. 비교예 및 실시예의 화장료 조성물은 pH를 5 내지 7로 조정하였기 때문에 이들 사이의 석출 여부의 차이는 pH에 따른 용해도 차이에서 기인하는 것이 아닌, 염기의 종류에 따라서 석출 여부가 결정된 것으로 판단된다. 더욱이, 용매의 종류만 상이하고 다른 구성성분 및 그 함량이 동일한 실시예 1과 비교예 6 내지 8의 화장료 조성물을 비교하면, 용매의 종류에 따라 저온 (4℃)에서 석출 여부에 차이가 있으므로, 용매로서 부틸렌글라이콜이나 부탄디올만을 사용한 경우와 비교해 프로필렌글라이콜을 사용하는 경우에 아젤라산의 용해도를 증가시키는 효과가 있음을 확인하였다 (표 4 참조). 또한, 도 6에 나타낸 바와 같이, 저온 (4℃)에서 실시예 1은 석출 현상이 발생하지 않았고, 비교예 6 내지 8은 석출된 물질이 하단에 가라앉아 있음을 확인할 수 있었다. 또한, 이러한 결과는 아미노산 포함 여부와 관계없이(예를 들어, 베타인이나 알지닌이 포함되지 않더라도) 동일한 결과였다.Moreover, when comparing the cosmetic compositions of Example 1 and Comparative Examples 6 to 8, which differ only in the type of solvent but have the same other components and contents, there was a difference in whether precipitation occurred at a low temperature (4°C) depending on the type of solvent, and thus it was confirmed that the use of propylene glycol had the effect of increasing the solubility of azelaic acid compared to the case where only butylene glycol or butanediol was used as a solvent (see Table 4). 사용감 평가를 위해, 24세 내지 45세 전문 패널 10명을 대상으로 실시예 1, 비교예 2 및 비교예 3의 화장료 조성물을 얼굴에 도포한 후, 끈적임 평가를 수행하였다.To evaluate the usability, the cosmetic compositions of Example 1, Comparative Example 2, and Comparative Example 3 were applied to the face of 10 professional panelists aged 24 to 45, and then a stickiness evaluation was performed.

수분활성도 평가를 위해, 33세 전문 패널 1명을 대상으로 시험 부위 (하박내측)를 비누로 세정하고 10분 동안 항온항습실 (22 ± 2℃, 40 ~ 60% RH)에서 대기한 후 실시예 1의 화장료 조성물을 도포하고 10분 뒤에 측정 기기 (Epsilon, BiOX)를 사용하여 피부수분도 (A.U., Arbituary Unit)를 측정해 수분활성도를 확인하였으며, 이를 도 3에 나타내었다.To evaluate water activity, one 33-year-old professional panelist washed the test area (inner side of the lower arm) with soap and waited for 10 minutes in a constant temperature and humidity room (22 ± 2℃, 40 to 60% RH). 염증성 병변 개선 평가를 위해, 24세 내지 35세 전문 패널 5명을 대상으로 실시예 1의 화장료 조성물을 사용하여 4주 동안 염증성 병변 개선 여부를 평가하였으며, 이를 도 4 및 도 5에 나타내었다.To evaluate the improvement of inflammatory lesions, the cosmetic composition of Example 1 was used on five expert panelists aged 24 to 35 years to evaluate whether inflammatory lesions improved for 4 weeks, and the results are shown in Figs. 4 and 5.

실시예 1의 화장료 조성물에 대한 항여드름 시험 효과를 확인하기 위해서 최소억제농도 평가(Minimum Inhibitory Concentration (MIC) assay) 이후 콜로니(colony)를 계수하여 평가하였다. 최소억제농도(MIC) 평가의 경우, 시료 20 μL에 여드름 원인균인 큐티박테리움 아크네스(Cutibacterium acnes, C. acnes) 배양액 180 μL를 분주하여 실험을 진행하였으며, 활성물질(즉, 아젤라산) 함량이 10 중량%인 시료를 처리하였을 때 최종 농도가 1.0 중량%로 희석되도록 하여 실험을 진행하였다. 음성 대조군에서는 시료로 아무런 처리를 하지 않았고, 양성 대조군에서는 시료로 항생제인 테트라사이클린염산염(tetracycline hydrochloride) 250 μg/mL를 사용하였고, 실험군에서는 시료로 실시예 1의 화장료 조성물을 사용하였다.In order to confirm the anti-acne test effect of the cosmetic composition of Example 1, the colonies were counted after the Minimum Inhibitory Concentration (MIC) assay. 실시예 1의 화장료 조성물에 대해서 19~59세의 여성 지원자 31명 패널을 대상으로 일차자극 평가를 진행하였다. 연구대상자의 등 부위에 시험물질을 IQ UltraTM chamber를 이용하여 24시간 동안 첩포한 후, 첩포 제거 30분 후 및 24시간 후 시점에서 PCPC Guideline (Personal Care Products Council, 미국화장품협회)의 판정 기준에 따라 피부 자극 정도를 평가하였다.A primary irritation evaluation was conducted on a panel of 31 female volunteers aged 19 to 59 years for the cosmetic composition of Example 1.
