Description
本申请要求于2024年03月25日提交中国专利局、申请号为202410346188.9、发明名称为“一种基于时间干涉的闭环多模态神经刺激系统及方法”的中国专利申请的优先权,其全部内容通过引用结合在本申请中。
本申请属于神经刺激技术领域,尤其是涉及一种基于时间干涉的闭环多模态神经刺激系统及方法。
大脑的活动主要表现为神经元放电,因此可以通过调节神经元的电位活动诱发短时间或长期的脑功能改变。人为的直接调控神经元活动,与神经和精神方面的疾病治疗有着非常大的相关性。在临床上也可用于脑神经疾病的治疗当中,有着巨大的潜力,并且也能有助于科学的研究人类大脑神经在感觉、运动及认知方面的潜在机制。
在一些情形下神经刺激方法主要有电刺激、磁刺激、光遗传刺激和超声刺激几个大类,其中电刺激是通过向神经系统传递电流来激活神经元,有植入侵入式电极的深部脑刺激和无创的经颅电刺激两大路线;磁刺激是通过在头皮上放置线圈产生磁场进而影响大脑活动;光遗传刺激是通过基因工程给细胞转染光敏蛋白,通过光线刺激这些蛋白引起神经元的兴奋;超声刺激是通过聚焦超声波刺激神经元。其中电刺激和光遗传刺激因其效果明显、可靠性高被广泛应用在脑功能的研究和神经相关疾病的治疗之中。从刺激方式的角度出发现有的主流刺激方法都有明显缺陷:深部脑刺激需要脑部植入电极来完成,这种侵入性操作可能导致感染、出血等潜在的并发症,增加了植入手术的复杂性和风险,同时患者也需要更长的恢复时间。此外电极植入方式因为植入深度的限制和电极定位的限制,在刺激的选择性和刺激的空间分辨率上都有较大的劣势;经颅电刺激虽然是无创刺激,但经颅电流只能提供阈下刺激,使用这种方式可以提高神经元的活性让其更加容易被激活但无法直接激活神经,并且经颅电流是没有指向性的被电流覆盖的区域都会受其影响,这两点特性极大地限制了经颅电刺激的应用;光遗传刺激刺激精度高、反应速度快,但因为需要进行外科手术植入刺激器并且涉及到转基因技术,因而在实际应用中也十分有限。
从系统结构角度出发,传统的刺激方式没有闭环调控系统,缺乏校正与优化手段,因使用者的个体差异较大,刺激方案与使用者的磨合周期长,不利于其应用。
本申请的目的是提供一种基于时间干涉的闭环多模态神经刺激系统及方法,解决上述技术存在的传统的刺激方式没有闭环调控系统,缺乏校正与优化手段,且刺激方案与使用者的磨合周期长,不利于应用的问题。
为实现上述目的,本申请提供一种基于时间干涉的闭环多模态神经刺激系统,包括:时间干涉刺激系统、脑电-功能性近红外采样系统和上位控制系统;时间干涉刺激系统利用两组电极产生的差拍电场对脑部指定区域进行精确刺激;脑电-功能性近红外采样系统是耦合脑电和功能性近红外的双模态采集器,包含信号提取和相关性分析两部分,通过整合EEG时间精度和fNIRS空间精度的统合脑信号数据来分析刺激效果和调整刺激方案;上位控制系统包括双模态融合模型计算、图卷积神经网络预测和刺激方案制定。
在一实施方式中,时间干涉刺激系统包括:通信控制模块、电源模块和时间干涉刺激模块;通信控制模块采用SoC芯片,内部融合处理器和无线通讯的功能,用于时间干涉刺激系统与上位控制系统之间的通信和刺激电流的控制;电源模块采用具有低噪音和低压降的稳压器外加屏蔽层的设计;时间干涉刺激模块由两路差拍电极组构成,由两个正弦波发生器产生两路相差10Hz-20Hz的2KHz交流信号,两路交流信号经由贴于头皮表面的电极组在颅内发生干涉,干涉产生的低频包络电流刺激指定区域的神经细胞。
在一实施方式中,脑电-功能性近红外采样系统包括:EEG脑电采集模块和fNIRS功能性近红外脑信号采集模块;EEG脑电采集模块使用表皮电极进行信号记录;在EEG脑电采集模块中时间干涉刺激产生的脑电信号经由预处理电路进行信号的滤波和去噪,然后通过模数转换器将信号转化为数字形式发送至通讯和控制子系统,脑电信号通过MCU进行去除伪影和去除基线漂移后上传至上位控制系统;fNIRS功能性近红外脑信号采集模块通过监测大脑血红蛋白和氧合血红蛋白在近红外光谱范围内的吸收变化来测量大脑特定区域的活动水平,fNIRS功能性近红外脑信号采集模块使用发光二极管作为光源产生650nm近红外光,光电二极管作为光信号探测器将经过脑组织的光信号转化为电信号进行分析,得到的电信号通过预处理进行去噪和滤波后再经由模数转换器将信号数字转化后发送至通信和控制子系统,MCU进行脑组织血氧计算获得大脑活动信号将活动信号上传至上位控制系统。
在一实施方式中,双模态融合模型计算为对EEG和fNIRS的信号数据进行特征提取并将两者组合成融合模型然后进行相关性分析。
在一实施方式中,图卷积神经网络预测是根据所期望的刺激效果预测刺激位置,并制定刺激方案,表示如下:

步骤1、确定刺激目标环节,确定计划刺激的位置和预期的四种脑波特征信号反应,包括δ、θ、α、β;其中δ的频率为0.5Hz-3.5Hz、θ的频率为4Hz-7Hz、α的频率为8Hz-13Hz、β的频率为14Hz-30Hz;
步骤2、设定刺激参数环节,若是初次刺激,此时尚无可利用的图卷积神经网络反馈结果,需要根据包络电场的产生特性计算两组电极的放置位置和电流频率,若不是初次刺激,则根据图卷积神经网络预测的结果确定电极的放置位置和频率参数设定;
步骤3、执行刺激环节,使用两组电极置于头皮产生有微小频率差距的经颅交流电,两组电流将在颅内特定区域激发低频包络电场以激活神经元;
步骤4、双模态数据采集和预处理环节,由EEG和fNIRS两套采集系统联合采集大脑信号,并将所采集信号进行预处理后,按对应采样时间打包;
步骤5、数据处理和相关性分析环节,上位控制系统将对两种模态的数据进行特征提取,进行相关性分析计算,得到时间成分特征和空间成分特征;
步骤6、判断刺激是否达到预期环节,通过分析采集到的被刺激者的四种脑波信号和生理上的反应,判断刺激是否达到预期目标;
步骤7、图卷积神经网络预测环节,利用先前刺激数据集和刺激中收集到的刺激数据集训练图卷积神经网络预测模型,以预期中大脑活动特征四种脑波为出发点逆向预测原始的被刺激点和被激活情况;
步骤8、刺激方案优化环节,利用图卷积神经网络预测的被刺激位置和被激活程度调整电极的放置位置和产生的包络电场频率。
在一实施方式中,步骤2中根据包络电场的产生特性计算两组电极的放置位置的表达式如下:

在一实施方式中,步骤4中双模态数据采集和预处理环节的过程如下:

S43、将数据汇总到数据采集系统MCU进行去伪影、去基线漂移处理;
S44、对处理过后的EEG和fNIRS数据按统一时钟进行数据打包,将数据包按时间排序发送至上位控制系统。
在一实施方式中,步骤5中数据处理和相关性分析环节的过程如下:
S51、对EEG和fNIRS数据进行数据特征提取;
S52、使用CCA算法对提取的特征进行相关性分析,得到时间成分特征和空间成分特征;
S53、利用时间成分特征和空间成分特征进行交叉验证,汇总出双模态融合数据。
因此,本申请采用上述一种基于时间干涉的闭环多模态神经刺激系统及方法,可以精准化、定制化的确定在不同个体条件下的实际刺激方案;此外可以在不进行电极植入手术的情况下进行深部神经刺激,大幅降低成本的同时,极大的提升了安全性;最后本申请设置的完整闭环系统使操作者可以高效的优化调整刺激方案,极大缩减了实际使用时的时间成本。

使本申请的上述目的、特征和优点能够更加明显易懂,下面结合附图和具体实施方式对本申请作进一步详细的说明。
请参阅图1到图5,一种基于时间干涉的闭环多模态神经刺激系统,包括:时间干涉刺激系统、脑电-功能性近红外采样系统和上位控制系统;时间干涉刺激系统利用两组电极产生的差拍电场对脑部指定区域进行精确频率的刺激;脑电-功能性近红外采样系统是耦合脑电和功能性近红外的双模态采集器,包含信号提取和相关性分析两部分,通过整合EEG时间精度和fNIRS空间精度的统合脑信号数据来分析刺激效果和调整刺激方案;上位控制系统包括双模态融合模型计算、图卷积神经网络预测和刺激方案制定。在本实施例中上位控制系统具体为上位机。
时间干涉刺激系统包括:通信控制模块、电源模块和时间干涉刺激模块;通信控制模块采用Nordic nRF52832系列的SoC芯片,其内部融合了STM32M4F级别的处理器和2.4GHz无线通讯的功能,负责时间干涉刺激系统与上位机控制系统之间的通信和刺激电流的控制;电源模块采用具有低噪音(纹波电压峰峰值优于5mV)和低压降(最小压差300mV以内)的稳压器外加屏蔽层的设计,以保证电极部分的精确高频信号不会受到噪音干扰;时间干涉刺激模块由两路差拍电极组构成,由两个正弦波发生器产生两路相差10Hz-20Hz的2KHz交流信号,两路交流信号将经由贴于头皮表面的电极组在颅内发生干涉,干涉产生的低频包络电流可以刺激所指定区域的神经细胞,该刺激方式可以直接刺激所制定区域神经细胞产生动作电位,同时不对制定刺激区域临近的脑区产生影响。
脑电-功能性近红外采样系统包括:EEG脑电采集模块和fNIRS功能性近红外脑信号采集模块;EEG脑电采集模块使用表皮电极进行信号记录,按照国际10-20系统或定制布局进行电极排布;在该模块中时间干涉刺激产生的脑电信号经由预处理电路进行信号的滤波和去噪,然后通过模数转换器(ADC)将信号转化为数字形式发送至通讯和控制子系统,通讯和控制子系统将转化为数字形式的脑电信号发送到MCU,转化为数字形式的脑电信号通过MCU进行去除伪影和去除基线漂移后上传至上位机。fNIRS功能性近红外脑信号采集模块通过监测大脑血红蛋白和氧合血红蛋白在近红外光谱范围内的吸收变化来测量大脑特定区域的活动水平,该模块使用发光二极管作为光源产生650nm近红外光,光电二极管作为光信号探测器将经过脑组织的光信号转化为电信号进行分析,得到的电信号通过预处理进行去噪和滤波后再经由模数转换器将信号数字化后发送至通信和控制子系统,通讯和控制子系统将数字化后的电信号发送到MCU,MCU根据数字化后的电信号进行脑组织血氧计算以获得大脑活动信号并将活动信号上传至上位机。在本申请中信号采集系统耦合了EEG和fNIRS两种方式对脑信号进行多模态采集,通过结合EEG的高时间分辨率(可以捕捉毫秒级别脑活动)和fNIRS的高空间分辨率(可以识别脑活动在不同深度的区域)同时获得在时间域和空间域上更详细的、全面的脑活动信息。同时该方式互补性的结合了EEG对神经电活动的敏感性和fNIRS对脑血流、脑血氧变化的敏感性,提供了种类上更全面的脑活动信息。EEG和fNIRS将被集成在一个硬件系统上使用同一标准时钟以解决两套采集方式因采样率不同产生的定时精度问题。该方案将使由两种方法采集到的脑活动数据的时间误差小于1.5ms,从而使从两个模块采集到的数据可以被精确定位到一个时间节点上。为后续对刺激效果的分析提供有效、全面的脑活动信息。
双模态融合模型计算为对EEG和fNIRS的信号数据进行特征提取并将两者组合成融合模型然后进行相关性分析,如图3,步骤如下:
EEG的特征提取主要在于提取目标刺激相关响应,将目标刺激的ERP(事件相关电位)组合成数据矩阵,再将数据矩阵与血液动力学响应函数卷积,进而获得可以进行后续相关性分析的EEG数据特征,构成特征矩阵XEEG;fNIRS的特征提取通过将神经刺激后40s内的血氧数据分时采样,按时间排序构建数据矩阵,得到与EEG数据特征对应的fNIRS数据特征,构成特征矩阵XfNIRS。
融合模型的构建是通过CCA(典型关联分析)算法计算得到两个特征数据集的最大相关矩阵AEEG和AfNIRS,进而通过最小二乘法拟合出EEG数据所代表的时间成分和fNIRS数据所代表的空间成分CEEG和CfNIRS。在该算法中可以构建如下模型公式:

在该式中Ai∈KT×D,i为模态数量,T是特征的采样时间点数量,Vi是特征矩阵的特征变量,D是特征矩阵的最小秩。

图卷积神经网络预测是根据所期望的刺激效果预测刺激位置,并制定刺激方案,图结构的数据通常包括节点和边,其中节点表示实体、边表示它们之间的关系。在神经刺激模型中,不同的脑区可以被构建成节点,而脑区与脑区间不同的连接状态可被构建成边,进行节点级别的预测便是对刺激位置的预测。表示如下:

因此,本申请采用上述一种基于时间干涉的闭环多模态神经刺激系统及方法,使用时间干涉刺激技术对脑神经进行精确的无创刺激;利用EEG耦合fNIRS的双模态脑信号采集方案获取全面的、精确的大脑活动数据;构建EEG和fNIRS的数据特征融合模型并利用图神经网络预测模型指导刺激方案的制定,可在使用中不断优化其效果。
以上实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
