Abstract
神経細胞を培養する培養区画を有する培養チャンバと、前記培養チャンバを支持する計測基板と、を備え、前記培養チャンバは、厚さ方向に貫通し平面視で閉環状に囲まれた前記培養区画と、前記培養チャンバの裏面に設けられ、前記培養区画から前記培養チャンバの側面まで伸びる溝部と、を有し、前記計測基板は、基材と、前記基材の上面に設けられた熱刺激素子と、前記上面に設けられた計測部と、を有し、前記計測部は、計測電極と、前記計測電極に接続された配線と、を有し、前記基材の上面と前記溝部で囲まれた空間は、前記神経細胞から延びる軸索が導入される微小通路であり、前記熱刺激素子及び前記計測電極は、少なくとも一部が前記微小通路に露出している熱応答性評価デバイス。
Description
本発明は、熱応答性評価デバイスに関する。
従来、生体が感覚、痛み、炎症といった刺激に対してどのように応答するかを実験モデルを用いて評価することで、感覚障害や疼痛に対する効果的な治療方法が検討されている。近年では実験動物削減の観点やヒトとの種差への懸念から、従来採用されていた小動物を用いた実験モデルに代えて、ヒトiPS細胞をはじめとした幹細胞から得られる実験モデル(培養実験モデル)が注目されている。
上述のような培養実験モデルにおいては、刺激に対する応答性が保存されている神経細胞を用いることが多い。例えば、神経細胞の電気的な活動を計測可能な基板上で感覚神経細胞を培養し、ヒーターを用いて基板周囲を温めることで刺激を印加して、熱刺激に対する応答性を評価する実験モデルが提案されている(例えば、非特許文献1参照)。

上記非特許文献1に記載の技術では、ヒーターで培養基板の周囲を温めて熱刺激を加えているため、用いる神経細胞の全体が熱刺激を受ける。そのため、神経細胞に対して局所的に刺激を加えることが困難であり、神経細胞のどこで活動電位が生じているかを把握することができない。
本発明はこのような事情に鑑みてなされたものであって、培養した神経細胞に対し、局所的な熱刺激を加え応答を評価可能な熱応答性評価デバイスを提供することを目的とする。
上記の課題を解決するため、本発明の一態様は、神経細胞を培養する培養区画を有する培養チャンバと、前記培養チャンバを支持する計測基板と、を備え、前記培養チャンバは、厚さ方向に貫通し平面視で閉環状に囲まれた前記培養区画と、前記培養チャンバの裏面に設けられ、前記培養区画から前記培養チャンバの側面まで伸びる溝部と、を有し、前記計測基板は、基材と、前記基材の上面に設けられた熱刺激素子と、前記上面に設けられた計測部と、を有し、前記計測部は、計測電極と、前記計測電極に接続された配線と、を有し、前記基材の上面と前記溝部で囲まれた空間は、前記神経細胞から延びる軸索が導入される微小通路であり、前記熱刺激素子及び前記計測電極は、少なくとも一部が前記微小通路に露出している熱応答性評価デバイスを提供する。
本発明によれば、培養した神経細胞に対し、局所的な熱刺激を加え応答を評価可能な熱応答性評価デバイスを提供することができる。
以下、図1~図5を参照しながら、本実施形態に係る熱応答性評価デバイスについて説明する。なお、以下の全ての図面においては、図面を見やすくするため、各構成要素の寸法や比率などは適宜異ならせてある。
以下の説明においては、xyz直交座標系を設定し、このxyz直交座標系を参照しつつ各部材の位置関係について説明する。ここでは、水平面内の所定方向をx軸方向、水平面内においてx軸方向と直交する方向をy軸方向、x軸方向及びy軸方向のそれぞれと直交する方向(すなわち鉛直方向)をz軸方向とする。
また、「上」とは鉛直方向上方である+z方向とし、「下」とは鉛直方向下方である-z方向とする。
以下の説明において、「平面視」とは、対象物を鉛直方向上方(+z側)から下向き(-z向き)に見ることを指す。
《熱応答性評価デバイス》
図1は、本実施形態の熱応答性評価デバイス1の概略斜視図である。熱応答性評価デバイス1は、培養チャンバ10と、計測基板20とを有する。熱応答性評価デバイス1は、さらに培養リング30、光源40、制御部50を有していてもよい。

熱応答性評価デバイス1は、神経細胞の熱応答性を評価するために用いられる。すなわち、熱応答性評価デバイス1は、培養チャンバ10において神経細胞を培養し、培養した神経細胞の軸索に対して熱刺激を加え、加えた熱刺激に対する反応を細胞が発する活動電位に基づいて評価することができる。
[培養チャンバ]
図2は、培養チャンバ10を示す概略斜視図である。図1,2に示すように、培養チャンバ10は、平面視矩形の部材であり、培養区画11と複数の溝部12とを有する。培養チャンバ10は、計測基板20の上面21aに配置されている。

培養区画11は、培養チャンバ10の上面10aから下面(裏面)10bに向けて厚さ方向(Z方向)に貫通し、平面視で閉環状に囲まれた貫通孔である。培養区画11の内部空間は、神経細胞を培養する空間として用いられる。
図1,2では、培養区画11は、平面視でX方向及びY方向にそれぞれ平行な辺を有する矩形であることとして示している。培養区画11の一辺は、平面視において5mm以上20mm以下であると好ましい。
溝部12は、培養チャンバ10の裏面10bに設けられ、培養区画11から培養チャンバの側面10xまで伸びる。溝部12の一端は、側面10xに達し、他端は培養区画11の内側面11xに達している。
溝部12は、平面視においてX方向に直線状に延び、Y方向に複数(図では4本)並列に設けられている。溝部12は一部が曲がっていてもよい。また、培養チャンバ10は溝部12を1本のみ有してもよい。
溝部12の高さH及び幅W1は、1μm以上10μm以下であると好ましい。溝部12の内部には、培養区画11で培養された神経細胞の軸索が導入される。溝部12の各大きさが上記範囲であることにより、溝部12の内部には神経細胞が通過できず、軸索は導入可能となる。
溝部12の長さLは500μm以上5000μm以下であると好ましい。これにより、溝部12の内部で軸索が十分伸長可能である。
培養チャンバ10の材料は特に制限はないが、形成が容易であることから樹脂材料であることが好ましく、硬化性樹脂であることがさらに好ましい。培養チャンバ10の材料としては、硬化性のシリコーン樹脂(シリコーンゴム)が好ましい。培養チャンバ10をシリコーン樹脂製とすることで、培養チャンバ10を計測基板20の上面21aに密着させ固定させやすい。
隣り合う溝部12同士の離間距離W2は100μm以上であると好ましい。これにより、培養チャンバ10と計測基板20との接触面積を十分に確保し、両者を密着固定させやすい。
培養チャンバ10は、フォトリソグラフィ技術を用いて培養チャンバ10の鋳型を形成した後、硬化性樹脂を用いて鋳型形状を転写することにより製造することができる。
[計測基板]
図3は、計測基板20を示す平面図である。図1,3に示すように、計測基板20は、基材21と、熱刺激素子22と、計測部23とを有する。熱刺激素子22及び計測部23は、基材21の上面21aに設けられている。基材21の上面21aは、計測基板20の上面である。

基材21(計測基板20)は、上面21aで培養チャンバ10を培養チャンバ10の裏面10bから支持する。基材21の上面21aと溝部12で囲まれた空間は、神経細胞から延びる軸索が導入される微小通路MPである。複数の溝部12は、それぞれ微小通路MPを形成している。
基材21は、平面視矩形の板材である。基材21は、光透過性を有していなくてもよいが、後述するレーザー光の波長の光に対し透過性を有すると好ましい。基材21は、上面21aが絶縁性である。また基材21の上面21aは伝熱性も低い方が好ましい。
基材21の材料は、形成材料としては、発明の効果を損なわない範囲において、有機材料又は無機材料を選択することができる。無機材料としてはガラスが好ましい。
基材21の形成材料である有機材料としては、高分子材料や、エラストマーを挙げることができる。高分子材料としては、例えば、ポリ塩化ビニル、ポリスチレン、ABS樹脂、ポリ乳酸のような熱可塑性樹脂、ポリイミドやフェノール樹脂のような熱硬化性樹脂を挙げることができる。エラストマーとしては、ポリシリコーンや合成ゴムなどを挙げることができる。
熱刺激素子22は、光を吸収して発熱する性質を有する。図3では、平面視矩形の熱刺激素子22が複数(図では12個)示されている。
複数の熱刺激素子22は、3つの熱刺激素子22がX方向に並ぶ素子列22Aを形成し、形成される4つの素子列22AがY方向に並ぶことで、3行×4列のマトリクス状に配列している。複数の熱刺激素子22は、上面21aにおいてそれぞれ独立している。
熱刺激素子22の材料は、光を吸収し発熱する材料であり、細胞毒性を持たない材料であれば限定しない。熱刺激素子22の材料としては、上記性質を示すのであれば特に制限はなく、例えば、光熱変換材料として用いられる、グラフェンや金ナノ粒子、黒リンなどが挙げられる。
熱刺激素子22は、1辺10μm以上200μm以下の範囲であることが好ましい。熱刺激素子22の数や、熱刺激素子22同士の間隔は特に制限はなく、発明の目的に応じて適宜設定することができる。
計測部23は、軸索で生じる活動電位を検出する。計測部23は、計測電極231と、計測電極231に接続された配線232と、を有する。また、計測部23は、絶縁層233と、参照電極239とを有する。
図3では、複数(図では16個)の計測部23が示されている。複数の計測部23は、上面21aにおいてそれぞれ独立している。
計測電極231は、平面視矩形を呈し、基材21の平面視中央に配列している。計測電極231の大きさは、1辺10μm以上200μm以下であると好ましい。複数の計測電極231は、素子列22Aに含まれる3つの熱刺激素子22の間、及び素子列22AのX方向の両側にそれぞれ配置され、4行×4列のマトリクス状に配列している。
すなわち、計測電極231と熱刺激素子22とは、微小通路に沿って交互に配列している。
熱刺激素子22及び計測電極231は、平面視で溝部12と重なり、少なくとも一部が微小通路MPに露出している。また、素子列22Aを構成する複数の熱刺激素子22及び複数の計測電極231は、微小通路MPに沿って配列している。
さらに、複数の熱刺激素子22及び複数の計測電極231は、隣り合う微小通路MPにまたがることなく、微小通路MPごとに独立して配列している。
配線232は、一端が計測電極231に接続し、他端が基材21の上面21aの+X方向の端部21bにまで伸びている。複数の配線232の他端は、等間隔で配列している。
端部21bにおいて配列する配線232の両側には、参照電極239が設けられている。後述の制御部50では、参照電極239と計測電極231との電位差を常時計測している。参照電極239は、参照電極239と計測電極231との電位差から、神経細胞が発火した際の細胞外での電位変化を検出するために用いられる。
参照電極239の端部は、計測部23との間の電位差を検出可能であれば、目的に応じて適宜設定することができる。
計測電極231の材料は、細胞毒性を持たない導体であれば特に制限はないが、細胞観察の観点から光透過性を有すると好ましい。計測部23及び参照電極239の材料は、例えば、グラフェンや酸化インジウムスズ(Indium Tin Oxide:ITO)などが挙げられる。
配線232及び参照電極239の材料は、細胞毒性を持たない導体であれば特に制限はないが、細胞観察の観点から光透過性を有すると好ましい。配線232及び参照電極239の材料は、計測電極231と同じであってもよく、異なっていてもよい。
計測部23及び参照電極239の寸法(幅、厚さ、長さ)については、電気回路の設計思想において、検出精度を向上させるために通常採用する構成を適宜採用することができる。
絶縁層233は、基材21の端部21bにおいて、配線232間及び配線232と参照電極239との間に設けられている。絶縁層233は、端部21bから-X方向に延びている。
絶縁層233の材料は、電気回路の絶縁層として通常用いられ、細胞毒性を持たない公知の材料を採用することができる。
計測基板20において、絶縁層233が設けられている一定の領域は、他の機器との電気的接続を行うコネクションパッド235である。
このような計測部23は、公知のフォトリソグラフィ技術を用いて適宜形成することができる。
[培養リング]
図1に示すように、培養リング30は、計測基板20の上面21aに設けられる筒状の部材である。熱応答性評価デバイス1において、培養リング30の内側には、培養チャンバ10が配置されている。

培養リング30の内側と計測基板20の上面21aとで囲まれる空間30Sは、神経細胞を培養する際の培養液が貯留される貯留部として機能する。
[光源]
図1に示すように、光源40は、レーザー光LBを射出するレーザー光源である。光源40は、スポット径が例えば100μmのレーザー光LBを射出する。光源40は、計測基板20の下面(-Z側の面)から熱刺激素子22にレーザー光LBを照射すると好ましい。

レーザー光LBは、熱刺激素子22が吸収可能な波長の光である。例えば、レーザー光LBは、波長785nmの赤色レーザー光とすることができる。レーザー光LBが照射された熱刺激素子22は、レーザー光LBを吸収し発熱する。
光源40は、複数の熱刺激素子22のうち、特定の熱刺激素子22にレーザー光LBを照射可能とするため、光源40を移動させ位置を調整する位置調整部材を有していてもよい。また、レーザー光LBの照射位置を変更する光路変更部材を有していてもよい。光路変更部材としては、公知のミラーや導光光学系を採用できる。
[制御部]
図4は、制御部50を説明するブロック図である。制御部50は、計測基板20及び光源40に接続されている。制御部50は、光源40の動作を制御する。また、制御部50は、計測部23で検出した軸索の活動電位に基づいて、神経細胞の熱応答性を検出する。

制御部50は、計算部51と、電流パルス生成部52と、受信部53と、記憶部54とを有する。制御部50は、入力装置59を有していてもよい。
計算部51は、光源40に照射させるレーザー光LBの強度、パルス幅を設定し、レーザー光LBの照射条件を計算して求める。また、計算部51は、計測部23を介して検出される電気信号ESをA/D変換し、電気信号ESに基づいて神経細胞の活動電位を計算して求める。
電流パルス生成部52は、計算部51で生成したレーザー光LBの照射条件に基づいて、光源40に入力する電流パルスを生成する。生成された電流パルスは、光源40に入力され、光源40からレーザー光LBが射出される。
受信部53は、計測部23が接続させ、計測部23介して伝達される電気信号を受信する。受信部53は、受信した電気信号の増幅や、電気信号に含まれるノイズの除去等の加工を行ってもよい。
記憶部54は、計算部51における各計算結果を一時的に記憶する揮発性のメモリと、規定値としてレーザー光LBの照射条件を複数記憶する不揮発性の記憶装置との両方を含むとよい。記憶部54は、不揮発性の記憶領域に、レーザー光LBのスポット径、熱刺激素子22の大きさ、熱刺激素子22の光熱変換効率等の情報を記憶していてもよい。計算部51は、レーザー光LBの照射条件を計算する際に、これらの情報を記憶部54から引き出し、熱刺激素子22で生じさせる熱量及び熱の発生時間の設定値と、に基づいて、レーザー光の照射条件を計算することができる。
入力装置59は、例えば、レーザー光LBのスポット径、熱刺激素子22の大きさ、熱刺激素子22の光熱変換効率等の情報を制御部50に入力するために用いる。入力装置59としては、キーボード、マウス等の入力デバイスを採用することができる。
[動作説明]
図5は、熱応答性評価デバイス1の動作を説明する説明図である。図5に示すように、培養区画11において神経細胞Cを培養し、微小通路MP内に軸索Aが伸びた状態とする。この状態の熱応答性評価デバイス1において、不図示の光源から熱刺激素子22にレーザー光LBを照射する。
図5は、熱応答性評価デバイス1の動作を説明する説明図である。図5に示すように、培養区画11において神経細胞Cを培養し、微小通路MP内に軸索Aが伸びた状態とする。この状態の熱応答性評価デバイス1において、不図示の光源から熱刺激素子22にレーザー光LBを照射する。
熱刺激素子22においては、材料の性質に応じてレーザー光LBが熱へと変換され、熱刺激素子22と重なる軸索Aの一部分(図5中、符号αで示す)を加熱する。
熱刺激素子22で生じる熱量は、レーザー光LBのレーザー強度(パルス振幅)、パルス幅、スポット径、熱刺激素子22の大きさ(体積)、熱刺激素子22の光熱変換効率等の条件に応じて概算することができる。これらの条件のうち、レーザースポット径、熱刺激素子22の大きさ、光熱変換効率を固定値(定数)とすることで、レーザー光LBのレーザー強度及びパルス幅により発熱量を制御できる。
熱刺激を受けた軸索Aの一部分αでは、刺激に応じて活動電位を生じる。生じた活動電位は、電気信号ESとして軸索Aを伝達する。電気信号ESが計測電極231に達すると、計測部23では電気信号ESを検出し、配線232を介して制御部50に電気信号ESを伝達する。
制御部50では、受信した電気信号ES(活動電位)に基づいて、神経細胞の熱応答を評価する。
制御部50では、光源40からレーザー光LBを照射させた時間と、電気信号ESの検出の時間とを同期させておき、レーザー光LBを照射した時間から電気信号ESを受信するまでの経過時間を算出する。算出した経過時間と、熱刺激素子22と計測電極231との離間距離とから熱刺激を受けた神経細胞Cの応答速度を求めることができる。
制御部50には、微小通路MPに沿って配列した複数の計測電極231から、順に電気信号ESが伝達される。これにより、熱刺激により発生した活動電位が、神経細胞C内を伝達する速度を評価できる。
また、計測電極231と熱刺激素子22との間隔を等間隔としておくことで、複数の計測電極231のそれぞれで活動電位が検出された時間から、活動電位が軸索A内を伝達する速度を求めやすい。同構成によれば、軸索Aの位置と応答速度との関係も評価しやすい。
さらに、レーザー光LBを繰り返し照射することにより、熱刺激を複数回繰り返したときの脱感作について評価することが可能となる。
以上のような熱応答性評価デバイス1では、熱刺激素子22は1辺10μm以上200μm以下というような微小な部材であり、離散的に形成されている。加熱時には熱刺激素子22にレーザー光LBを照射して発熱させるため、神経細胞Cの軸索Aに対してμmスケールの局所的な刺激を与え、熱刺激に対する軸索Aの応答性を評価できる。
また、熱刺激素子22は1辺10μm以上200μm以下というような微小な部材であり、蓄熱しにくい。そのため、レーザー光LBの照射をやめると、すぐに放冷し加熱が継続されないと考えられる。これにより、熱応答性評価デバイス1では、神経細胞Cの軸索Aに対してミリ秒スケールの短期的な刺激を与え、熱刺激に対する軸索Aの応答性を評価できる。
さらに、例えば電気配線に通電させることによる抵抗発熱を利用して軸索Aに熱刺激を加える構成とすると、計測基板上の配線が混み合い、構成が複雑になることが想定される。このような構成の熱応答性評価デバイスでは、配線間が近接し、漏れ電流が生じやすくなり、制御が困難となりやすい。対して、上述の熱応答性評価デバイス1では、熱刺激素子22を用いて加熱することにより、計測基板上の配線が少なく、構成が簡素になる。
そして、レーザー光により軸索Aを直接加熱する方法を想定した場合、レーザー光が軸索Aの周囲の培養液に吸収され減衰するため、短時間のうちに適切な加熱が困難となる。これに対し、上述の熱応答性評価デバイス1では、熱刺激素子22を用いて加熱することにより、短期的な刺激を与えることができる。
これらにより、以上のような構成の熱応答性評価デバイス1によれば、培養した神経細胞に対し、局所的な熱刺激を加え応答を評価可能な熱応答性評価デバイスを提供することができる。これにより、培養実験系において、身体内と同様な熱受容からの信号伝達の過程を再現することができ、細胞レベルでの熱受容の研究を進めることが可能になる。
例えば、1)熱刺激から活動電位発生までの応答時間の遅れ、2)熱刺激の応答閾値に対する伝播特性の関係性、3)高頻度刺激に対する脱感作現象などが細胞レベルで可視化される。また、タンパク質や遺伝子の発現といった細胞の情報と対応させてこれらの現象を解析することで、熱受容に関する理解が飛躍的に進む。
なお、本実施形態においては、レーザー光LBの照射位置を制御するために、光源40が位置調整部材や光路変更部材を有していてもよいこととしたが、これに限らない。例えば、光源40の位置を固定するとともに、培養チャンバ10を備えた計測基板20を載置する載置台としてXYステージを採用し、光源40と計測基板20上の熱刺激素子22との相対位置を変更可能としてもよい。
以上、添付図面を参照しながら本発明に係る好適な実施の形態例について説明したが、本発明は係る例に限定されない。上述した例において示した各構成部材の諸形状や組み合わせ等は一例であって、本発明の主旨から逸脱しない範囲において設計、仕様等に基づき種々変更可能である。
以下に本発明を実施例により説明するが、本発明はこれらの実施例に限定されるものではない。
[熱応答性評価デバイスの作製]
培養チャンバは、流体デバイスに広く用いられるシリコーンゴムであるPolydimethylsiloxane(PDMS)により作製した。フォトリソグラフィ技術により作製した微小通路および培養区画の鋳型に、架橋前のPDMSを流し込み、熱硬化することで所望の構造の培養チャンバを得た。

計測基板は、フォトリソグラフィ技術を用いて、ITO膜付きガラス基板上のITO膜をエッチングし、配線及び参照電極を形成した。さらに、インクジェット用のグラフェン分散液を塗布した後に、フォトリソグラフィにより作製した保護膜を介して酸素プラズマによりエッチングすることで、計測電極および熱刺激素子を形成した。
さらにpoly(chloro-p-xylylene)(パリレンC)を蒸着した後、計測電極、配線、熱刺激素子に該当する箇所のパリレンCを酸素プラズマ処理して除去することで絶縁層を形成した。
細胞の接着性を上げる目的で、表面にPolyethyleneimineによる表面処理を行い、計測基板を得た。
顕微鏡下で位置を確認し、計測電極及びび熱刺激素子が微小通路に並ぶように、培養チャンバを計測基板上に設置し、熱応答性評価デバイスを得た。
図6は、作製した熱応答性評価デバイスの位相差顕微鏡像を示す。微小通路MPの高さは10μmとした。微小通路MPに沿って、計測電極231と熱刺激素子22が配列していることが確認できた。
計測電極231は、1辺50μmの正方形とし、微小通路MPに沿って隣り合う計測電極231の間隔は450μmとした。計測電極231は、隣り合う計測電極231の間の中央に配置し、計測電極231と熱刺激素子22との間隔が等間隔であることとした。
熱刺激素子22は、1辺100μmの正方形とした。
[熱刺激応答性の評価]
作製した熱刺激応答性デバイスの培養区画において、ラット胎児の後根神経節から採取した感覚神経細胞を培養したところ、1週間程度で微小通路MP内への軸索の伸長が確認された。熱応答性の評価実験は培養2~3週間後に行った。

レーザー光源から波長785nm、スポット径100μmのレーザー光を射出させ、熱刺激素子22に照射した。レーザー光は、出力200mW、パルス幅300μmとした。
図7は、代表的な神経細胞の応答を示す説明図であり、活動電位の検出結果を示す図である。
神経細胞Cの細胞体に最も近い熱刺激素子22にレーザー光を照射したところ(T0)、熱刺激素子22の両側の計測電極231A,231Bで検出したシグナルSA,SBにおいて、1ms程度後(T1=1ms)に活動電位を示すピークが計測された。図7中、「*」は、活動電位を示すピークを示す。熱刺激を加えることにより、軸索Aでは1ms以内には活動電位が発生したことを示す結果を得た。
また、100μmスケールで並べた電極から活動電位の伝播が計測されたことから、局所的な刺激に伴う刺激応答が評価できることが確認できた。
さらに、微小通路に沿って配列する複数の計測電極231では、熱刺激素子22から遠ざかるに従って、計測電極231C,231D,231E,231F,231G,231Hで検出したシグナルSC~SHの順に、遅れて活動電位が計測された。
これらにより、作製した熱応答性評価デバイスでは、マイクロメートルスケールで局所的、かつミリ秒スケールの短期的な熱刺激に対する感覚神経細胞の応答を評価できることが確認でき、本発明が有用であることが確認できた。

- 神経細胞を培養する培養区画を有する培養チャンバと、
前記培養チャンバを支持する計測基板と、を備え、
前記培養チャンバは、厚さ方向に貫通し平面視で閉環状に囲まれた前記培養区画と、
前記培養チャンバの裏面に設けられ、前記培養区画から前記培養チャンバの側面まで伸びる溝部と、を有し、
前記計測基板は、基材と、
前記基材の上面に設けられた熱刺激素子と、
前記上面に設けられた計測部と、を有し、
前記計測部は、計測電極と、前記計測電極に接続された配線と、を有し、
前記基材の上面と前記溝部で囲まれた空間は、前記神経細胞から延びる軸索が導入される微小通路であり、
前記熱刺激素子及び前記計測電極は、少なくとも一部が前記微小通路に露出している熱応答性評価デバイス。 - 前記計測基板は、複数の前記熱刺激素子と、複数の前記計測部とを有し、
複数の前記熱刺激素子及び複数の前記計測電極は、前記微小通路に沿って配列している請求項1に記載の熱応答性評価デバイス。 - 前記培養チャンバは、複数の前記溝部を有し、
複数の前記溝部は、それぞれ前記基材の上面との間に前記微小通路を形成し、
複数の前記熱刺激素子及び複数の前記計測電極は、前記微小通路ごとに独立して配列している請求項2に記載の熱応答性評価デバイス。 - 複数の前記熱刺激素子及び複数の前記計測部は、前記微小通路に沿って等間隔に交互に配列している請求項2又は3に記載の熱応答性評価デバイス。
- 前記熱刺激素子にレーザー光を照射する光源を有する請求項1から4のいずれか1項に記載の熱応答性評価デバイス。
- 前記計測基板及び前記光源に接続された制御部を有し、
前記制御部は、前記光源を制御すると共に、前記計測部を介して伝達される活動電位に基づいて前記神経細胞の熱応答を評価検出する請求項5に記載の熱応答性評価デバイス。 - 前記制御部は、前記レーザー光を照射した時間から前記活動電位を検出するまでの経過時間を計測し、前記神経細胞の熱刺激に対する応答速度を評価する請求項6に記載の熱応答性評価デバイス。
