본 발명은 크립토코쿠스 네오포만스에 대한 뇌-혈관 장벽 통과 저해제 스크리닝 방법, 항진균제 스크리닝 방법 및 항진균용 조성물에 관한 것으로, 본 발명의 스크리닝 방법은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과를 조절하는 신규 유전자를 이용하므로, 뇌-혈관 장벽 통과 저해제, 항진균제를 효과적으로 스크리닝 할 수 있다.

Description

크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과 조절 신규 유전자 및 이의 용도
본 발명은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과를 조절하는 신규 유전자 및 이의 용도에 관한 것으로, 구체적으로는 크립토코쿠스 네오포만스에 대한 뇌-혈관 장벽 통과 저해제 스크리닝 방법, 항진균제 스크리닝 방법 및 항진균용 조성물에 관한 것이다.
크립토코쿠스 네오포만스는 다양한 육상 및 수중 환경에서 흔히 발견되는 자실 균류다. 특정 환경 조건에서 널리 퍼진 진균은 양성 또는 단성 생식을 통해 감염성 자낭포자를 생성할 수 있다. 인간 숙주가 이 포자를 흡입하면 하부 호흡기까지 도달할 수 있다. 이 생태에서 크립토코쿠스 네오포만스는 다당류 캡슐과 폴리페놀 멜라닌 색소와 같은 독특한 항식작용 인자를 사용하여 폐포 대식세포의 식작용을 저항한다. 또한 식작용 후에도 크립토코쿠스 네오포만스는 포식소의 성숙을 억제하고 대식세포 내에서 증식하며, 자주 비용해성 세포외배출을 통해 탈출한다. 이후 크립토코쿠스 네오포만스는 혈액으로 퍼져 나가 중추신경계를 감염시키고, 치명적인 뇌수막염을 일으킨다.
상기 진균은 다른 인간 병원성 진균들과 달리 독특한 신경 추적 운동을 하며 혈뇌장벽(BBB)을 통과하는 능력이 있다. 본 발명자들은 이전 연구를 통해 인체 뇌 모세혈관 내피세포(HBMECs)가 자란 트랜스웰 막을 이용한 체외 BBB 모델을 사용하여 여러 BBB 통과 인자를 식별하였다. 이러한 인자에는 이노시톨 수송체(Itr1a와 Itr3c), 히알루론산 합성효소(Cps1), 금속단백분해효소(Mpr1), 요소분해효소(Ure1), 포스포리파아제(Plb1), Fnx1, Rub1 등이 포함된다. 크립토코쿠스 네오포만스는 뇌의 높은 이노시톨 수준을 이용하여 Cps1의 발현을 유도하고, 이후 크립토코쿠스 세포 표면에 히알루론산을 생성한다. 크립토코쿠스의 히알루론산은 HBMECs의 CD44 글리코단백질에 의해 인식되어 크립토코쿠스의 횡행성 이동을 촉진한다. 그러나 최근에 크립토코쿠스 네오포만스의 전사 인자와 인산화 효소 돌연변이 라이브러리에 대한 체계적인 분석을 통해 BBB 통과와 뇌 실질 내 생존에 관여하는 광범위한 신호 구성 요소 네트워크가 밝혀졌다. 또한 새로 개발된 3차원 인간 신경혈관 유닛(hNVU) 칩을 사용하여 신경추적 운동과 BBB 침투에 관여하는 특정 전사 인자와 인산화 효소가 밝혀졌다. 이러한 발견은 크립토코쿠스 네오포만스의 BBB 통과에 대한 신호 네트워크와 조절 메커니즘이 이전에 예상했던 것보다 훨씬 복잡하다는 것을 시사한다.
이전 연구에 근거하여, 본 발명자들은 호메오박스 전사 인자인 Hob1에 주목하였다. HOB1의 결실은 크립토코쿠스 네오포만스의 부착과 BBB 침투에 심각한 결함을 초래하여 그 병원성을 현저히 약화시키고, hob1Δ 돌연변이에서 ITR1a, ITR3c, MPR1, FZC3, PDR802와 같은 다른 BBB 통과 인자들의 유도가 감소한다.
반면에, 크립토코쿠스 네오포만스의 형제종인 크립토코쿠스 가티아이는 주로 폐를 감염시키지만 뇌를 감염시키지는 않으며, BBB 통과와 병원성에는 Hob1이 기여하지 않는다. 이는 크립토코쿠스 네오포만스에서 BBB 통과와 병원성의 주된 조절자로서의 Hob1의 역할을 강조하며, 크립토코쿠스 가티아이에서는 이러한 역할이 나타나지 않는다. 그러나 크립토코쿠스 네오포만스에서 Hob1이 지배하는 하류 신호 네트워크와 BBB 침투를 조절하는 직접적인 메커니즘은 아직 잘 밝혀지지 않은 실정이다.
본 발명자들은 크립토코쿠스가 혈뇌장벽을 통과하는 메커니즘 연구를 통해 이 과정을 조절하는 호메오박스 전사 인자인 Hob1을 발견하였다(한국공개특허 제10-2017-0077066). 이어 본 발명에서는 전사체 분석을 통해 Hob1의 하류 네트워크를 탐색하였다. Hob1에 의존하는 100개의 단백질 중에서, 호스트 모방 조건에서 Hob1에 의해 현저히 유도되는 중요한 혈뇌장벽 통과 인자인 Hbf1을 발견하였으며, Hbf1의 결실은 크립토코쿠스 네오포만스의 혈뇌장벽 침투 능력을 현저히 저해함을 확인하여 본 발명을 완성하였다.
본 발명이 이루고자 하는 기술적 과제는 크립토코쿠스 네오포만스에 대한 항진균제 스크리닝 방법을 제공하는 것이다.
본 발명의 다른 목적은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과 저해제 스크리닝 방법을 제공하는 것이다.
또한, 본 발명이 이루고자 하는 기술적 과제는 크립토코쿠스 네오포만스에 대한 항진균용 조성물을 제공하는 것이다.
본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 기술적 과제로 제한되지 않으며, 언급되지 않은 또 다른 기술적 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
상기 기술적 과제를 달성하기 위하여, 본 발명의 일실시예는 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및 상기 크립토코쿠스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계를 포함하는 크립토코쿠스 네오포만스의 뇌-혈관 장벽(BBB) 통과 저해제를 스크리닝 하는 방법을 제공한다.
본 발명의 실시예에 있어서, 상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 뇌-혈관 장벽(BBB) 통과 저해제로 선별하는 단계를 더 포함할 수 있다.
상기 기술적 과제를 달성하기 위하여, 본 발명의 다른 실시예는 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및 상기 크립토코쿠스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계를 포함하는 크립토코쿠스 네오포만스에 대한 항진균제 스크리닝 방법을 제공한다.
본 발명의 실시예에 있어서, 상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 항진균제로 선별하는 단계를 더 포함일 수 있다.
상기 기술적 과제를 달성하기 위하여, 본 발명의 다른 실시예는 상기 방법에 따라 스크리닝된 저해제를 포함하는 항진균용 조성물을 제공한다.
상기 기술적 과제를 달성하기 위하여, 본 발명의 다른 실시예는 크립토코쿠스 네오포만스의 HBF1 유전자의 발현, 또는 이로부터 발현되는 단백질의 발현 또는 활성을 억제하는 물질을 포함하는, 크립토코쿠스 네오포만스에 대한 항진균용 조성물을 제공한다.
본 발명의 실시예에 있어서, 상기 물질은 핵산, 항체, 앱타머, 펩타이드, 단백질, 화합물 또는 천연물 일 수 있다.
본 발명의 실시예에 있어서, 상기 물질은 HBF1 유전자의 mRNA에 상보적인 서열을 가지는 안티센스, siRNA(small interfering RNA) 또는 shRNA(short hairpin RNA) 핵산을 포함할 수 있다.
본 발명은 크립토코쿠스 네오포만스에 대한 뇌-혈관 장벽 통과 저해제 스크리닝 방법, 항진균제 스크리닝 방법 및 항진균용 조성물에 관한 것으로, 본 발명의 스크리닝 방법은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과를 조절하는 신규 유전자를 이용하므로, 뇌-혈관 장벽 통과 저해제, 항진균제를 효과적으로 스크리닝 할 수 있다.
본 발명의 효과는 상기한 효과로 한정되는 것은 아니며, 본 발명의 설명 또는 청구범위에 기재된 발명의 구성으로부터 추론 가능한 모든 효과를 포함하는 것으로 이해되어야 한다.

이하, 본 개시내용을 실시예 및 실험예에 의하여 상세히 설명한다. 본 개시내용은 다양한 변환을 가할 수 있고 여러 가지 실시 예를 가질 수 있는 바, 이하 특정 실시 예들을 도면에 예시하고 상세한 설명에 상세하게 설명하고자 한다. 그러나, 이는 본 개시내용을 특정한 실시 형태에 대해 한정하려는 것이 아니며, 본 개시내용의 사상 및 기술 범위에 포함되는 모든 변환, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다. 본 개시내용을 설명함에 있어서 관련된 공지 기술에 대한 구체적인 설명이 본 개시내용의 요지를 흐릴 수 있다고 판단되는 경우 그 상세한 설명을 생략한다.
이하, 본 발명을 상세히 설명한다.
본 발명은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과 저해제 스크리닝 방법에 관한 것이다.
본 발명은 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및 상기 크립토코쿠스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계; 를 포함한다.
상기 크립토코쿠스 네오포만스 균주는 당 분야에 공지의 방법 및 조건으로 배양될 수 있다.
상기 크립토코쿠스 네오포만스에 후보물질을 접촉시키는 단계는 후보물질을 당 분야에 공지된 방법 및 조건으로 공지된 수단을 이용하여 크립토코쿠스 네오포만스에 처리하는 것일 수 있다. 목적하는 처리량 및 처리방법에 따라 적절히 처리될 수 있다.
상기 후보물질은 예를 들어, 핵산, 항체, 앱타머, 펩타이드, 단백질, 화합물 또는 천연물을 포함할 수 있으나 이에 한정되지 않는다.
상기 mRNA 발현 수준의 측정은 당 분야에 공지된 방법으로 수행될 수 있으며, 예를 들면 역전사 중합효소반응(RT-PCR), 경쟁적 역전사 중합효소 반응, 실시간 역전사 중합효소반응, RNase 보호 분석법(RPA), 노던 블롯팅, DNA 칩(Microarray) 또는 RNA sequencing 분석에 의해 수행될 수 있다.
상기 단백질의 발현 수준 측정은 당 분야에 공지된 방법으로 수행될 수 있으며, 예를 들면 웨스턴 블랏, ELISA(enzyme-linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 유세포분석(Fluorescence Activated Cell Sorter, FACS), 또는 단백질 칩(protein chip)으로 수행될 수 있다.
상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 뇌-혈관 장벽 통과 저해제로 선별하는 단계를 더 포함할 수 있다.
상기 HBF1 유전자는 표 2에 기재된 고유번호(CNAG_03759)로 확인되는 서열로 이루어진 것일 수 있다.
본 발명은 크립토코쿠스 네오포만스에 대한 항진균제 스크리닝 방법에 관한 것이다.
본 발명은 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및 상기 크립토코쿠스 네오포만스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계;를 포함한다.
크립토코쿠스 네오포만스의 배양, 후보물질, 발현 또는 활성 측정 방법 등에 관하여는 전술한 바와 같다.
상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 항진균제로 선별하는 단계를 더 포함할 수 있다.
상기 항진균제는 진균 감염증을 예방, 치료하는 약물로서, 크립토코쿠스 네오포만스는 뇌-혈관 장벽 통과하여 뇌수막염을 일으키는 진균인 바, 혈뇌장벽 침투 능력과 관련된 HBF1의 발현 또는 활성을 현저히 저해함으로서 항진균효과를 나타낼 수 있다.
본 발명은 크립토코쿠스 네오포만스에 대한 항진균용 조성물에 관한 것이다.
본 발명은 크립토코쿠스 네오포만스의 HBF1 유전자의 발현, 또는 이로부터 발현되는 단백질의 발현 또는 활성을 억제하는 물질을 포함한다.
상기 물질은 핵산, 항체, 앱타머, 펩타이드, 단백질, 화합물 또는 천연물일 수 있으나 이에 제한되지 않는다.
상기 물질은 HBF1 유전자의 mRNA에 상보적인 서열을 가지는 안티센스, siRNA(small interfering RNA) 또는 shRNA(short hairpin RNA) 핵산일 수 있으나, 이에 한정되지 않는다.
본 발명의 항진균용 조성물은 항진균제, 약제학적 조성물, 피부 세정용 조성물, 항균용 건축자재 및 생활용품 조성물 또는 화장품 조성물로서 활용될 수 있다.
상기 항진균제는 진균의 증식과 성장을 억제하여 진균 감염증을 예방, 치료하는 약물이다.
상기 약제학적 조성물은, 상용되는 무기 또는 유기의 담체를 가하여 고체, 반고체 또는 액상의 형태로 경구 투 여하거나, 비경구, 직장, 국소, 경피, 정맥 내, 근육 내, 복강 내, 피하 등으로 투여될 수 있다.
상기 경구 투여를 위한 제재로서는 정제, 환제, 과립제, 연·경 캡슐제, 산제, 세립제, 분제, 유탁제, 시럽제, 펠렛제, 음료 등을 들 수 있다. 또한, 상기 비경구 투여를 위한 제재로는 주사제, 점적제, 연고, 로션, 스프레이, 현탁제, 유제, 좌제 등을 들 수 있다.
본 발명의 유효성분을 제제화하기 위해서는 통상의 방법에 따라서 실시하면 용이하게 제제화할 수 있으며, 계면 활성제, 부형제, 착색료, 향신료, 보존료, 안정제, 완충제, 현탁제, 기타 상용하는 보조제를 적절하게 추가하여 사용할 수 있다.
또한, 상기 유효 성분의 투여량은 치료받을 대상의 연령, 성별, 체중과, 치료할 특정 질환 또는 병리 상태, 질환 또는 병리 상태의 심각도, 투여경로 및 처방자의 판단에 따라 달라질 것이다. 이러한 인자에 기초한 투여량 결정은 당업자의 수준 내에 있으며, 일반적으로 투여량은 0.001mg/kg/일 내지 대략 2000mg/kg/일의 범위일 수 있으나, 어떠한 방법으로도 본 발명의 범위를 한정하는 것이 아니다.
이하, 본 발명을 구체적으로 설명하기 위해 실시예를 들어 상세하게 설명하기로 한다.
<재료 및 방법>
1. 전사체 분석(RNA 시퀀싱)
크립토코쿠스 네오포만스(Cryptococcus neoformans)와 크립토코쿠스 가티아이(Cryptococcus gattii)의 야생형 및 hob1Δ 돌연변이는 처음에 30°C에서 하룻밤 동안 배양하였고, 이후 OD600이 0.8에 도달할 때까지 신선한 YPD 배지로 옮겨졌다. 세포는 PBS 완충액으로 세 번 세척한 후 10% FBS가 첨가된 RPMI 1640 배지에서 37°C 및 5% CO2 조건에서 3시간 동안 배양되었다. 배양 후, 세포는 수집되어 급속 냉동 및 동결 건조되었다. Total RNA는 Easy-BLUE 키트(17061, iNtRON, South Korea)를 사용하여 추출하였고, 추가적으로 RNeasy Mini Kit(74106, Qiagen, Germany)를 사용하여 정제하였다. 이 절차는 각 균주에 대해 세 번의 독립 배양에서 반복되었다.
RNA 시퀀싱을 위해 각 샘플에서 1 μg의 total RNA를 사용하여 TruSeq mRNA Library Kit(Illumina, USA)를 이용해 cDNA 라이브러리를 준비하였으며, 시퀀싱은 Illumina 플랫폼에서 수행되었다. 시퀀싱 후 처리 과정에는 Cutadapt v2.4와 Python 3.7.4를 사용하여 어댑터 및 저품질 시퀀스를 제거하는 작업이 포함되었다. 정제된 읽기 결과는 Hisat2 v2.2.1을 사용하여 크립토코쿠스 네오포만스 H99 참조 게놈에 정렬되었으며, Hisat 및 Bowtie2 알고리즘을 활용하였다. 주석 데이터는 NCBI FTP 서버에서 가져왔다. Hisat2는 "-p 30" 및 "--dta -1" 옵션과 기본값을 사용하여 실행되었다. 정렬된 읽기는 Samtools v0.1.19를 사용하여 변환("-Sb -@ 8") 및 정렬("-@ 20 -m 2000000000")의 기본 매개변수를 수정하여 관리되었다. 전사체 조립 및 풍부도 추정은 Stringtie v1.3.6을 사용하여 "-p 12" 설정으로 수행되었으며, FPKM 값에 의해 전사체 풍부도를 정량화하였다. 데이터 매트릭스는 R 패키지 "isoformswitchanalyzerR"을 사용하여 생성 및 분석되었다. 품질 관리는 DEBrowser를 사용하여 수행되었다. 차등 발현 분석은 DESeq2 v1.24를 사용하여 수행되었으며, 결과는 R v4.1.0의 Enhanced Volcano 패키지를 사용하여 시각화되었고, 유의성을 위해 두 배 변화 및 P-value 값이 0.05 미만인 임계값을 적용하였다.
2. 유전자 결실 돌연변이체 구축
HBF1의 돌연변이 균주는 크립토코쿠스 네오포만스 혈청형 A 균주 H99S의 유전적 배경 내에서 제작되었다. 유전자 교란을 위해, 노르소트리신 내성을 부여하는 카세트(노르소트리신 아세틸트랜스퍼레이스 유전자, NAT에 의해 암호화됨)는 기존의 중첩 PCR 또는 NAT 분할 마커/이중 접합(DJ) PCR 방법론을 사용하여 제작되었다. 이 카세트를 구축하는 첫 번째 PCR 단계에서는 HBF1 유전자의 5' 및 3' 플랭킹 시퀀스를 각각 L1/L2 및 R1/R2 프라이머 세트를 사용하여 증폭하였으며, H99S 게놈 DNA를 템플릿으로 사용하였다. 전체 길이의 NAT 마커는 pNAT-STM 플라스미드에서 M13Fe(M13 forward extended) 및 M13Re(M13 reverse extended) 프라이머 쌍을 사용하여 증폭되었으며, 이 플라스미드는 고유의 시그니처 태그 시퀀스를 포함한 NAT 유전자를 포함하고 있다. 분할 마커/DJ-PCR 접근법에서는 NAT 마커의 5' 및 3' 부분을 각각 M13Fe/SM2 및 M13Re/SM1 프라이머 조합을 사용하여 개별적으로 증폭하였다. 초기 증폭 후, 중첩 PCR 방법은 L1 및 R2 프라이머 쌍을 사용하여 초기 증폭된 제품을 템플릿으로 병합하여 완전한 유전자 교란 카세트를 합성하는 것을 포함하였다. 반면에, 분할 마커/DJ-PCR 전략은 초기 PCR 제품을 템플릿으로 사용하여 NAT 분할 유전자 교란 카세트의 5' 및 3' 부분을 각각 L1/SM2 및 R2/SM1 프라이머 조합을 사용하여 증폭하였다. 생물학적 변환 과정을 위해, H99S 균주는 효모 추출물-펩톤-덱스트로스(YPD) 배지에서 30°C에서 하룻밤 동안 배양한 후 원심 분리하여 수집하고, 5mL의 멸균 증류수에 재부유시켰다. 약 200μL의 이 세포 현탁액을 1M 소르비톨이 포함된 YPD 고체 배지에 고르게 분배하고, 30°C에서 3시간 동안 배양하였다. PCR로 생성된 교란 카세트는 600μg의 0.6μm 직경 금 마이크로캐리어 비드(Bio-Rad)에 흡착된 후, PDS-100 입자 전달 시스템(Bio-Rad)을 사용하여 세포에 전달되었다. 변환 후, 세포는 세포막 무결성 회복을 촉진하기 위해 30°C에서 4시간 동안 배양된 후 수집되어, 100μg/mL 노르소트리신이 포함된 YPD 고체 배지(YPD+NAT)에서 배양하여 안정적인 형질전환체를 선택하였다. 이후 이들을 YPD+NAT 배지에서 여러 차례 통과시켜 정제하였다. 초기 확인을 위해, NAT 양성 균주는 진단 PCR로 선별되었다. 정확한 유전자 결실을 보장하고 비표적 통합을 배제하기 위해, 각 돌연변이 균주에 대해 서던 블롯 분석이 수행되었다. 관련 없는 돌연변이 효과를 배제하기 위한 돌연변이 표현형의 추가 검증은 각 돌연변이마다 여러 독립적인 결실 균주를 생성하는 과정을 포함하였다.
3. HBF1 과발현, 보완 및 형광 단백질 태깅 균주의 구축
크립토코쿠스 네오포만스에서 HBF1 지속적 과발현 균주를 제작하기 위해, HBF1 유전자의 원래 프로모터를 히스톤 H3 프로모터로 대체하였다. 이는 여러 단계로 증폭된 상동 재조합 카세트를 구축하여 달성되었다. 초기 PCR 단계에서는 HBF1의 5'-플랭킹 영역과 5'-코딩 영역을 각각 L1/OEL2 및 OER1/PO 프라이머 쌍을 사용하여 증폭하였다. 동시에, NAT-H3 프로모터 영역은 표 1에 나열된 프라이머 쌍을 사용하여 증폭되었다.

두 번째 PCR 단계에서는 더블 조인트(DJ) PCR을 사용하여 PH3:HBF1 카세트의 5' 영역을 구축하였다. 이는 HBF1의 5'-플랭킹 영역과 NAT-H3 프로모터 영역을 템플릿으로 사용하여 표 1에 나열된 프라이머 쌍으로 증폭하였다. 마찬가지로, PH3:HBF1 카세트의 3' 영역은 HBF1의 3'-플랭킹 영역과 NAT-H3 프로모터 영역의 혼합 템플릿을 사용하여 DJ-PCR로 합성되어 또 다른 프라이머 쌍으로 증폭되었다. 조립된 PH3 :HBF1 카세트는 생물학적 변환을 통해 야생형 H99S 균주에 도입되었다. 안정적인 형질전환체는 노르소트리신(NAT)이 첨가된 효모 추출물-펩톤-덱스트로스(YPD) 배지에서 선택되었으며, SO/B79 프라이머 쌍을 사용한 진단 PCR로 올바른 삽입을 확인하였다. 카세트의 통합 및 적절한 방향은 L1 및 PO 프라이머로 PCR 증폭된 특정 프로브를 사용한 서던 블롯 분석을 통해 확인되었다.
비태그 및 mRuby3-태그 균주를 구축하기 위해, 종결 코돈을 제외하고 프로모터 영역을 포함한 HBF1의 개방 읽기 프레임(ORF)을 PCR 증폭하여 New England BioLabs(USA)의 Gibson Assembly Master Mix 키트를 사용하여 pNEO-mRuby3 벡터에 클로닝하였다. Dcm 메틸화를 피하기 위해, pNEO-mRuby3 플라스미드는 dam-/dcm- 유능한 대장균 세포(New England BioLabs, USA 제공)에 형질전환되었다. 형질전환 후 플라스미드를 추출하였다. pNEO_HBF1-mRuby3 플라스미드는 StuI 효소로 절단하여 선형화된 후 생물학적 변환을 통해 hbf1Δ(YSB10384) 균주에 도입되었다.
4. 비병원성 진균 사카로마이세스 세레비지에의 HBF1 발현 균주 구축
비병원성 진균 사카로마이세스 세레비지에에서 HBF1 지속적 과발현 균주를 제작하기 위해, HBF1 유전자의 cDNA를 크립토코쿠스 네오포만스 H99 균주의 cDNA로부터 프라이머 세트를 이용해 합성하였다. HBF1 유전자의 cDNA는 89 amino acid로 굉장히 짧기에, 원활한 클로닝을 위해 고발현 유전자인 TEF1 유전자의 프로모터를 합성하였다. 이는 여러 단계로 증폭된 상동 재조합 카세트를 구축하여 달성되었다. 초기 PCR 단계에서는 HBF1의 5'-플랭킹 영역과 5'-코딩 영역을 각각 L1/OEL2 및 OER1/PO 프라이머 쌍을 사용하여 증폭하였다. 동시에, TEF1 프로모터 영역 또한 해당하는 프라이머 쌍을 사용하여 증폭되었다. 두 번째 PCR 단계에서는 더블 조인트(DJ) PCR을 사용하여 PTEF :HBF1 카세트의 5' 영역을 구축하였다. 이는 HBF1의 5'-플랭킹 영역과 TEF1 프로모터 영역을 템플릿으로 사용하여 해당하는 프라이머 쌍으로 증폭하였다. 마찬가지로, PTEF :HBF1 카세트의 3' 영역은 HBF1의 3'-플랭킹 영역과 TEF1 프로모터 영역의 혼합 템플릿을 사용하여 DJ-PCR로 합성되어 또 다른 프라이머 쌍으로 증폭되었다.
조립된 PTEF :HBF1 카세트는 pLEU_TEF 플라스미드에 삽입되었다. 제한 효소 BamHI과 SacI 처리한 PLEU_TEF 플라스미드와 HBF1 cDNA를 깁슨 어셈블리 방법을 통해서 클로닝을 진행하였다. 비병원성 진균 사카로마이세스 세레비지에 BY4742 균주에 형질 전환하였으며, 다음 방법과 같은 절차로 형질 전환이 진행되었다. YPD 30'C에서 16시간 이상 배양한 BY4742 균주를 서브 컬쳐하여 OD600 값이 0.6이 될 때까지 키운 뒤, 세포를 수집하여 2.5ml 멸균 증류수에 희석하였다. 50% PEG 1ml, 1M LiOAc 180ul, SS-DNA 125ul가 포함된 PEG/LiOAc 혼합물을 제작한 뒤, 300㎕의 혼합물과 1㎍의 플라스미드를 섞어주었다. 위에서 배양한 균주 희석액 100㎕를 섞어 준 뒤 42℃에서 45분간 반응시켰다. 균주는 원심분리를 통해 수집한 후, 상층액을 버리고 100㎕의 0.9% NaCl 용액으로 희석하여 일부 희석액을 SD-Leu 배지에 도말하여 3~4일간 30℃에서 배양하였다. SD-Leu 배지에서 선택된 집락 형성군들을 대상으로, 프라이머 쌍을 이용한 진단 PCR로 올바른 균주임을 확인하였다.
5. HBF1 유전자 발현 확인을 위한 정량적 RT-PCR
류신 공벡터 형질 전환 균주와 PTEF_HBF1을 각각 EBM-2 배지에서 하룻밤 동안 배양한 후, OD600nm=0.8이 될 때까지 EBM-2 배지에서 재배양하였다. OD600nm=0.8에 도달한 후, 세포를 수집하여 PBS로 세척하고, 나머지 세포는 EBM-2 배지에서 120 rpm으로 흔들어주며 6시간, 12시간, 24시간 동안 샘플링하였다. 수집된 세포는 즉시 냉동하고 동결 건조하였다. 총 RNA는 위에서 설명한 방법에 따라 추출하였다. cDNA 합성을 위해, 총 RNA를 DEPC 처리된 물로 5 ㎍농도로 조절한 후, oligo(dT)-pdN6을 첨가하고 65℃에서 5분 동안 가열하였다. 그런 다음, RNAse 억제제(M007, Enzynomics, South Korea), dNTP, 그리고 reverse H minus Reverse transcriptase(EP0752, Thermo Scientific, USA)를 첨가하고 50℃에서 60분 동안 배양한 후, 85℃에서 10분 동안 열을 가하여 비활성화시켰다. 유전자 발현 수준은 액틴 유전자(ACT1)와 류신 유전자(LEU2) 발현 값을 사용하여 정규화하였으며, 류신 공벡터 형질 전환 균주에서의 기초 발현 수준에 대해 상대적으로 배수 차이(fold change)를 계산하였다.
6. 성장 및 화학 감수성 분석
크립토코쿠스 네오포만스 HBF1 돌연변이 균주의 성장 동태 및 화학 감수성을 평가하기 위해, 30°C에서 하룻밤 동안 배양된 균주를 10배씩 단계적으로 희석하였다(1:1에서 1:10,000까지). 이러한 희석액을 여러 농도의 화학 물질이 포함된 YPD 한천 배지에 점적으로 적용하였다. 화학 물질은 특정 종류의 스트레스를 유도하기 위해 사용되었다(삼투 스트레스를 위한 소르비톨; 양이온/염 스트레스를 위한 NaCl 및 KCl; 산화 스트레스를 위한 과산화수소(H2O2), tert-부틸 하이드로퍼옥사이드, 메나디온, 디아마이드; 중금속 스트레스를 위한 황산 카드뮴(CdSO4); 유전 독성 스트레스를 위한 메틸 메탄설포네이트 및 하이드록시우레아; 막 불안정을 위한 라우릴 황산 나트륨(SDS); 세포벽 불안정을 위한 칼코플루오르 화이트 및 콩고 레드; 소포체(ER) 및 환원 스트레스를 위한 튜니카마이신(TM) 및 디티오트레올(DTT); 그리고 항진균 감수성을 평가하기 위한 여러 항진균제(플루디옥시닐, 플루코나졸, 암포테리신 B, 플루시토신)).
이들 배지는 포도당을 포함하거나 포함하지 않는 조건에서 배양되어 다양한 영양 상태에서의 스트레스 반응을 테스트하였다. 처리 후, 배지는 30℃에서 배양되었고, 콜로니 성장을 모니터링하기 위해 2일차부터 5일차까지 매일 이미지를 촬영하였다. 추가적으로, 각 돌연변이 균주의 온도 내성을 확인하기 위해, 단계적으로 희석된 세포를 점적으로 적용한 YPD 배지를 25°C, 30°C, 37°C, 39°C의 네 가지 다른 온도에서 배양하였다. 성장은 2일차부터 4일차까지 촬영한 사진을 통해 기록되었다. 이러한 종합적인 실험 설계는 환경적 스트레스와 항진균제에 대한 돌연변이 균주의 강인성뿐만 아니라 다양한 온도에 대한 적응성을 자세히 평가할 수 있게 하였다.
7. 생체 외 독성 인자 생산 분석
캡슐 생산 효율을 측정하기 위해 각 돌연변이체를 30°C에서 배양한 후, Dulbecco's modified Eagle's (DME) 한천 배지에 점적하여 37°C에서 2일간 배양하였다. 이후 세포를 긁어내어 증류수로 세척하고, 10% 포르말린으로 고정한 후 다시 증류수로 세척하였다. 다른 배지에서의 캡슐 생산 효율을 측정하기 위해 각 배양액 5㎕를 Littman's 한천 배지와 FBS 한천 배지(10% 태아 소 혈청과 90% PBS)에 점적하여 37°C에서 2일간 배양한 후 긁어내어 증류수로 재현탁하였다. 재현탁된 세포는 India ink (BactiDrop; Remel, San Diego, CA, USA)로 염색하고, 차등 간섭 대비 현미경(DIC) (BX51, Olympus, Tokyo, Japan)으로 관찰했하였다. 캡슐 두께는 캡슐 직경에서 세포 직경(총 직경-세포 본체 직경)을 빼서 측정하였다. 캡슐 두께의 정량적 측정을 위해 H99S 균주 및 각 인산분해효소 돌연변이체에 대해 50개의 세포를 측정하였다. 이어서 멜라닌 생산 효율을 측정하기 위해, 야생형 균주를 포함한 돌연변이체를 YPD 배지에서 30°C에서 16시간 배양한 후 PBS로 세척하고, Niger seed 배지로 3μl씩 배양하였다. 배양된 세포는 37°C에서 배양한 후 1-3일 동안 사진 촬영하였다.
8. mRuby3-태그된 HBF1의 세포내 위치 시각화
Hbf1-mRuby3-태그 균주는 처음에 30°C에서 YPD 배지에서 하룻밤 배양하였다. 배양액은 YPD 배지에서 OD600이 0.8에 도달할 때까지 재배양하였다. 이후 배양액을 두 개의 25 ml 분주로 나누고 원심 분리한 후 인산 완충 식염수(PBS)로 세 번 세척하였다. 하나의 분주는 Hbf1의 기초 위치를 분석하기 위해 액체 질소 탱크에 보관하였고, 다른 하나는 10% FBS가 첨가된 RPMI 1640 배지와 동일한 부피로 재부유하였다. 이 샘플은 CO2 인큐베이터에서 37°C, 120 rpm으로 수평 진동하는 조건에서 배양하였다. 세포는 10, 30, 60, 180분에 수확되어, 실온에서 15분 동안 3.4% 자당을 포함한 4% 파라포름알데하이드 용액으로 고정되고, 이후 0.1 M KPO4와 1.2 M 소르비톨을 포함한 용액으로 세척하였다. 핵 염색을 위해, 세포는 10 ㎍/mL Hoechst 33342(Thermo Fisher, USA)로 30분 동안 어두운 곳에서 처리하였다. 최종적으로, 염색된 세포는 차등 간섭 대비(DIC) 및 형광 현미경(Nikon Eclipse, Japan)으로 관찰하였다
9. 시험관 내 BBB 통과 분석
BBB 통과 분석은 이전 연구(Lee KT et al. Nat Commun, 2020)에서 설명된 방법을 사용하여 수행하였다. 인간 뇌 미세혈관 내피 세포(HBMEC 라인 hCMEC/D3, Merck, USA)는 콜라겐-프리코팅된 플레이트(354236, Corning, USA)에서 80%의 융합에 도달할 때까지 배양하였다. 이후, 5 x 104 hCMEC/D3 세포를 8-μm 다공성 콜라겐 코팅 막(353097, BD Falcon)에 파종하고, EndoGROTM-MV Complete Media Kit(SCME004, Sigma-Aldrich, USA)에서 유지하였다. 24시간 후, 배지를 2.5% 인간 혈청(H4522, Sigma, USA)이 첨가된 신선한 EndoGRTM 배지로 교체하고, 세포를 추가로 4일 동안 배양하였다. 효모 접종 전날, 배지를 1.25% 인간 혈청이 포함된 EndoGROTM배지로 교체하였다. 세포 배양은 5% CO2, 37°C에서 진행되었다. 세포 접합의 무결성을 평가하기 위해 Epithelial Volt per Ohm Meter(EVOM2, World Precision Instruments, USA)를 사용하여 횡단 내피 전기 저항(TEER)을 측정하였다. 하룻밤 배양된 야생형 및 HBF1 돌연변이 효모 세포는 PBS로 세 번 세척하고, 5 x 105 세포를 100 μL PBS에 조정하여 hCMEC/D3-파종 막에 첨가하였다. 24시간 배양 후, 막을 통과한 효모 세포를 수집하였다. 각 돌연변이 균주의 BBB 통과 비율은 야생형과 비교하여 상대적 BBB 통과 비율을 결정하였다.
<결과>
1. 크립토코쿠스 네오포만스와 크립토코쿠스 가티아이의 hob1Δ돌연변이의 비교 전사체 분석
Hob1에 의해 조절되는 하류 네트워크를 분석하기 위해, 야생형(WT)과 hob1Δ돌연변이 균주에 대해 RNA 시퀀싱(RNA-seq) 기반 전사체 분석을 수행하였다(도1). 크립토코쿠스 네오포만스와 크립토코쿠스 가티아이에서 Hob1 삭제 후 나타나는 뚜렷한 표현형 변화를 인식하고, 이 두 종 간의 전사체 분석을 수행하였다. 또한 Hob1의 혈뇌장벽(BBB) 통과에서 중요한 역할을 확인하고, 숙주 모방 조건에서 전사체를 면밀히 조사하였다. 이를 위해 조직 배양 배지(RPMI)에 10% FBS를 첨가하고 37°C, 5% CO2조건에서 배양하는 조건을 사용하였다.
크립토코쿠스 네오포만스에서 hob1Δ 돌연변이는 기본 조건(YPD 배지, 30°C)에서 661개의 유전자가 상향 조절되고 817개의 유전자가 하향 조절되었다(WT 균주와 비교). 상향 조절된 유전자 집합은 주로 캡슐 생합성 유전자(CAS32, CAP4, CAP60 등)와 세포주기, 분열, 감수분열과 관련된 유전자(IRR1, CDC2801, CDC7 등)를 포함하였다. 반면, 하향 조절된 유전자들은 주로 유사분열 활성화 단백질 인산화효소 경로(MAPK 신호 경로)의 유전자(HSP12, PTP1, PTP2 등), 세포외 분비, 액틴 결합 유전자 카테고리에 속하였다. 특히, 일반적으로 MAPK 신호 경로와 관련된 ERG11은 역설적으로 상향 조절되었다. 이어 유전자 온톨로지 용어와 KEGG 경로를 통한 추가 분석은 신호 경로에서의 동적 변화를 강조했으며, RNA 분해 및 감수분열 경로의 유도와 대사 경로, 전분 및 자당 대사, 탄소 대사의 감소를 보여주었다. 반면에 크립토코쿠스 가티아이에서 hob1Δ 돌연변이의 전사체 변화는 상대적으로 미미했으며, 상향 조절된 3개와 하향 조절된 5개의 유전자만 식별되었다. 이 유전자들은 특정 GO 용어나 KEGG 경로에 속하지 않으며, 그 기능은 대체로 정의되지 않았다. 액틴 결합 유전자 카테고리는 HOB1 결손 변이체가 액틴 결합을 방해하는 약물인 latrunculin A가 포함된 약물 배지에서 생장할 때 그 생장도가 매우 증가하는 결과를 통해 간접적으로 영향을 확인할 수 있었다(도 2).
이어서 HMC 조건 하에서 뚜렷한 대조가 관찰되었다. 크립토코쿠스 네오포만스의 hob1Δ 돌연변이는 453개의 유전자(320개 유도, 133개 감소)가 중요하게 조절되어, 호스트와 유사한 조건에 적응하는 데 Hob1의 중요한 역할을 시사하였다. 이러한 적응은 크립토코쿠스 가티아이의 hob1Δ 돌연변이와 크게 대조되는데, 여기서는 단 7개의 유전자만이 중요하게 조절되어, 기본 및 HMC 조건 모두에서 두 종 사이의 Hob1의 중요성에 대한 차이를 강조하였다. 흥미롭게도 HMC 하에서 크립토코쿠스 네오포만스에서 유도된 유전자들은 기본 조건에서의 유전자들과 밀접하게 일치했으나, 하향 조절된 유전자의 수는 줄었다. 이 변화는 HMC가 부과하는 엄격한 생존 요구 사항, 즉 온도 상승 및 영양소 가용성 변화를 반영할 수 있다.
기본 조건과 HMC 조건간 비교 분석에서 크립토코쿠스 네오포만스에서 187개의 유전자가 두 조건 모두에서 일반적으로 상향 조절되었는데, 이는 염색질 리모델링, 탄수화물 대사 과정, 세포 분열과 관련된 경로를 강조한다(도 3). 이 공통성은 외부 환경에 관계없이 Hob1이 조절하는 핵심 기능을 나타낸다. 반대로, 서로 다른 조건에 따른 유전자 조절이 따로 존재함이 명확히 드러났으며, 그 결과 기본 조건과 HMC 조건에서 활성화된 서로 다른 유전자 세트가 환경적 도전에 대한 맞춤형 반응을 나타낸다고 해석할 수 있다. 하향 조절된 유전자의 개수의 변화는 이러한 조건 특이성을 더 강조하며, HMC 조건에서 기본 조건에 비해 하향 조절된 유전자 수가 현저하게 감소한 패턴을 보여준다. 이 패턴은 HMC가 대표하는 스트레스 조건 하에서 특히 대사 및 생합성 과정을 조절하는 Hob1의 복잡한 역할을 제안한다.
결론적으로, 우리의 자세한 비교 전사체 분석은 크립토코쿠스 네오포만스 내에서 Hob1 삭제가 유전자 발현을 조절하는 데 미치는 중대한 영향을 밝혀내며, 다양한 환경 조건에서 영향을 받는 광범위한 생물학적 과정을 강조한다. 크립토코쿠스 네오포만스와 크립토코쿠스 가티아이 간의 이러한 대조적인 반응은 호스트 환경 내에서 크립토코쿠스 네오포만스의 병원성과 적응 전략에 있어 Hob1의 핵심적 역할을 강조한다.
2. Hob1의 하류 타깃 후보 규명: 전사체 분석에 기반한 접근
이전 연구에서 크립토코쿠스 네오포만스와 크립토코쿠스 가티아이에서 Hob1의 기능이 다른 이유가 Hob1의 염기 서열에 있는 것이 아님을 밝혀냈다. 대신, Hob1의 하류 타깃 단백질들이 이 두 종에서 서로 다르게 작동할 수 있음을 시사하며, 종 특이적인 복잡한 조절 메커니즘의 상호작용이 있을 것으로 보였다. 이 가설을 더 깊이 탐구하기 위해, 우리는 기본 조건과 호스트 모방 조건(HMC) 모두에서 크립토코쿠스 네오포만스 hob1Δ 돌연변이의 전사체 분석을 종합적으로 수행하였다. 이 분석은 Hob1의 직접적 혹은 간접적 조절 하에 있는 것으로 추정되는 유전자 모음을 식별하는 데 중요한 역할을 했으며, 이를 표 2(Hob1 전사체 분석을 통한 잠재적 다운스트림 표적 예측 및 후보 유전자 목록)에 요약하였다.

하류 타깃들 중에서 CNAG_03759는 여러 가지 설득력 있는 이유로 특히 관심 있는 유전자로 부각되었다. 첫째, Hbf1은 기본 조건과 HMC 실험 설정 모두에서 Hob1이 결손되었을 때 가장 일관되게 발현이 감소된 유전자였다. 이 일관된 조절 패턴은 Hob1이 조절하는 경로 상에 Hbf1이 존재한다는 강력하고 특이적인 상호작용을 의미한다. 둘째, 다른 기능이 잘 문서화된 유전자들과는 달리, CNAG_03759는 그 역할이 아직 이해되지 않은 단백질을 암호화하고 있어, 추가 기능적 특성화를 위한 새로운 연구 대상이 된다.
CNAG_03759에 의해 암호화된 단백질은 그 짧은 길이(단 89 아미노산)와 분생 특이적 기능을 가진 도메인을 보유한 것이 특징이였다(도 4). 이 도메인은 유사한 단백질 크기와 도메인 구조를 가진 구조적 동위유전자인 CNAG_01056에도 존재한다. 그러나 CNAG_03759와는 대조적으로, CNAG_01056은 테스트된 조건 하에서 극도로 낮은 발현 수준 때문에 Hob1에 의해 조절되지 않는 것으로 보였다. 이러한 동위유전자들 사이의 차별적 발현은 Hob1의 선택적 조절 역할에 대한 흥미로운 통찰을 제공하였다.
크립토코쿠스 네오포만스는 다른 진균종에서 흔한 분생 생성 과정이 존재하지 않음에도 불구하고, CNAG_03759의 크립토코쿠스 네오포만스 병인성에서의 기능은 아직 추측에 불과했다. 그러나 이 단백질의 강력한 조절적 연관성과 독특한 단백질 구조는 우리로 하여금 이 단백질을 Hob1-의존적 혈뇌장벽 통과 인자(HBF1)로 지정하게 했으며, 구조적 동위유전자인 CNAG_01056은 HBF101으로 명명하였다. 본 연구진은 HBF1이 크립토코쿠스 네오포만스의 혈뇌장벽 통과를 촉진하는 데 중요한 역할을 할 수 있다고 가설을 세웠으며, 이는 뇌수막염 발달의 핵심 단계이다. 이는 크립토코쿠스 네오포만스가 중추신경계를 침범하는 분자 메커니즘을 이해하는 새로운 길을 열고 치료적 개입을 위한 잠재적 표적을 제안한다.
3. Hbf1의 크립토코쿠스 네오포만스의 산화 스트레스 반응에 관여
크립토코쿠스 네오포만스의 성장과 생존에 있어 Hbf1의 기능적 중요성을 더 잘 이해하기 위해 hbf1Δ 및 PH3 :HBF1 돌연변이 균주를 개발하였다. hbf1Δ 돌연변이 균주에서는 HBF1 유전자가 완전히 제거되고 항진균제 저항성 마커로 대체되었으며, 이는 유전자 결실의 표현형 결과를 관찰하여 유전자 기능을 평가하는 일반적인 전략이다. 반면, PH3 :HBF1 돌연변이 균주에서는 HBF1의 원래 프로모터를 고활성 H3 프로모터로 교체하여 HBF1 유전자의 과발현을 유도하였고, 이를 통해 유전자 발현 증가가 균류 생리학에 미치는 영향을 확인하고자 하였다(도 5).
산화 및 소포체(ER) 스트레스 유발 물질, 즉 과산화수소(H2O2)와 디티오트레올(DTT)을 사용한 다양한 시험관 내 표현형 분석을 통해 흥미로운 저항 패턴을 관찰하였다(도 6). 강력한 산화제인 H2O2는 세포 시스템에 심각한 산화 스트레스를 가하며, DTT는 산화 단백질 접합에 필요한 환경을 교란시켜 ER 스트레스를 유발한다. 두 물질 모두 hbf1Δ 균주가 약간의 저항성 증가를 나타냈다. 이는 Hbf1의 존재가 산화 조건에 대한 취약성을 높이고, 따라서 Hbf1 제거가 균류를 이러한 스트레스로부터 강화할 수 있음을 의미한다.
추가적으로, Hbf1 과발현이 크립토코쿠스 네오포만스의 중요한 보호 기제인 멜라닌 형성에 미치는 영향을 조사하였다. 멜라닌은 균 세포를 UV와 강한 이온화 방사선으로부터 보호할 뿐만 아니라 산화 손상을 완화하는 데도 중요한 역할을 한다. 우리의 결과는 Hbf1이 과발현될 때 멜라닌 생산이 크게 감소함을 나타냈다 (도 7). 멜라닌이 크립토코쿠스 네오포만스의 병원성에 중요한 역할을 한다는 점을 고려할 때, 이 감소는 균이 숙주 생물의 감염 동안 직면하는 적대적인 환경 조건에서 생존할 능력을 떨어뜨릴 수 있다.
예상치 못하게도, Hbf1 결실이 산화 스트레스에 대한 저항성을 증가시키는 반면, 과발현은 멜라닌을 효과적으로 활용하여 이러한 손상으로부터 보호할 수 없게 하였다. 이 모순적인 결과는 산화 스트레스 관리에서 Hbf1의 복잡한 조절 역할을 강조하며, Hbf1이 크립토코쿠스 네오포만스의 산화 스트레스 하에서의 생존뿐만 아니라 병원성에도 필수적일 수 있음을 시사한다. 이러한 발견은 Hbf1의 생물학적 기능에 대한 중요한 통찰력을 제공하며, 그 발현 또는 기능을 조절하여 크립토코쿠스 감염을 억제하기 위한 치료 전략의 목표로서의 잠재력을 부각시킨다.
4. Hbf1의 세포 내 위치 분석
크립토코쿠스 네오포만스에서 Hbf1의 세포 내 위치를 조사하기 위해, Hbf1 단백질의 C-말단에 mRuby3 형광 태그가 결합된 hbf1Δ::HBF1-mRuby3 리포터 균주를 제작하였다. 이 유전적 변형은 융합 단백질이 스트레스 조건에서 생물학적 활성을 유지하는지 평가하였다(도 8). 이 균주는 산화 스트레스와 소포체(ER) 스트레스에 대한 내성이 야생형 수준으로 회복되는 것을 보여주어, Hbf1-mRuby 융합 단백질이 완전한 기능을 유지하고 있음을 확인하였다.
이후, 표준 조건과 HBF1 발현을 강하게 유도하는 숙주 유사 조건(HMC)에서 Hbf1의 세포 내 위치를 밝히는 데 주력하였다. 표준, 처리되지 않은 조건에서는 Hbf1이 세포질 전반에 약한 형광 강도로 분포하였다. 그러나 HMC 조건에서는 Hbf1이 특정 세포 내 위치로 재배치되어, 숙주 유사 조건에서 특정 부위로 국한된다는 것을 나타냈다. 이 표적 재배치는 Hbf1이 스트레스 반응이나 세포 신호 전달 경로와 관련된 특수 기능을 수행할 가능성을 나타낸다.
형광 현미경을 사용한 추가 분석에서는 HMC 조건에서 처리되지 않은 세포에 비해 형광 신호의 밝기가 증가하는 것을 확인하였다(도9). 이 형광 강도의 증가는 Hbf1 발현의 상향 조절을 확인시켜 줄 뿐만 아니라, Hbf1이 숙주 유사 조건에 대한 곰팡이 세포의 반응에 더 적극적으로 관여하고 있음을 나타낸다. 또한, HMC 처리 후 30분 이내에 형광 증가가 최고조에 달하는 타이밍은 Hbf1이 초기 반응 단백질임을 시사하며, 이는 Hob1-의존적 전사 메커니즘에 의해 조절될 가능성이 있다. 이러한 시간적 발현 및 위치 패턴은 Hbf1이 환경 스트레스에 대한 즉각적인 세포 반응에서 중요한 역할을 한다는 것을 나타낸다.
이러한 관찰은 크립토코쿠스 네오포만스 내에서 Hbf1의 복잡한 조절과 중요한 기능적 역할, 특히 숙주 조건을 모방한 환경적 도전에 대한 반응을 강조한다. 이 확장된 분석은 Hbf1의 세포 행동과 곰팡이 생물학에 대한 중요한 통찰력을 제공하여, 크립토코쿠스 네오포만스가 적대적인 숙주 환경에서 적응하고 생존하는 분자 메커니즘을 이해하는데 기여한다.
5. Hbf1변이체의 크립토코쿠스 네오포만스의 혈액뇌장벽(BBB) 통과 분석
본 연구에서는 Hbf1이 BBB를 통과하는 데 필수적인지 조사하였다. 이는 전사체 분석에서 Hbf1이 중요한 하위 표적으로 확인된 것에 기인한다. Vu et al. 이 처음 개발한 시험관 내 BBB 모델은 transwell 막에 배양된 인간 뇌 미세혈관 내피 세포(HBMEC)를 사용하며, 혈액과 뇌 구획 사이의 물리적 분리를 모방한다. 이 모델은 이전 연구들에서 mpr1Δ 돌연변이, 비병원성 사카로마이세스 세레비지에(S288C 균주), 병원성 크립토코쿠스 네오포만스(H99 균주)를 사용하여 검증되었다. 우리는 Hbf1이 BBB 침투를 촉진할 수 있는지 평가하기 위해 유사한 transwell 기반 시스템을 채택하였다.
그 결과, 야생형(WT) 크립토코쿠스 네오포만스는 배양 후 24시간 이내에 BBB를 성공적으로 통과하였다. 이에 비해 hbf1Δ 돌연변이는 BBB를 통과하는 능력이 현저히 감소하여, 이 과정에서 Hbf1의 잠재적 역할을 강조하였다. 이어 Hbf1이 BBB 투과성에 직접적인 영향을 미치는지 확인하기 위해, 우리는 Hbf1 보완 균주(hbf1Δ::HBF1)와 Hbf1 유전자가 과발현된 돌연변이 균주(PH3 :HBF1)를 평가하였다. 보완 균주는 BBB 통과 능력을 야생형 수준으로 회복시켰으나, 과발현 돌연변이 균주는 야생형 수준을 초과하지 않았다(도 10). 이러한 결과는 Hbf1이 BBB 통과에 필수적이지만, 과발현이 BBB 투과성을 비례적으로 증가시키지는 않음을 나타낸다. RNA-seq 전사체 분석에서 HMC 조건에서 HBF1이 4.55배 상향 조절됨을 확인하여, 이러한 조건에서의 기능적 영향을 관찰하였다. 이 상관관계는 Hbf1이 인간 BBB를 통과하는 데 중요한 역할을 한다는 것을 강조한다.
본 발명은 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과를 조절하는 신규 유전자 및 이의 용도에 관한 것으로, 구체적으로는 크립토코쿠스 네오포만스에 대한 뇌-혈관 장벽 통과 저해제 스크리닝 방법, 항진균제 스크리닝 방법 및 항진균용 조성물로 사용할 수 있다.

Claims (8)

  1. 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및
    상기 크립토코쿠스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계;를 포함하는 크립토코쿠스 네오포만스의 뇌-혈관 장벽(BBB) 통과 저해제를 스크리닝 하는 방법.
  2. 청구항 1에 있어서, 상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 뇌-혈관 장벽 통과 저해제로 선별하는 단계를 더 포함하는, 크립토코쿠스 네오포만스의 뇌-혈관 장벽 통과 저해제를 스크리닝 하는 방법.
  3. 크립토코쿠스 네오포만스(C. neoformans)에 후보물질을 접촉시키는 단계; 및
    상기 크립토코쿠스의 HBF1 유전자의 mRNA 또는 그 단백질의 발현 수준을 측정하거나, 또는 그 단백질의 활성을 측정하는 단계;를 포함하는 크립토코쿠스 네오포만스에 대한 항진균제 스크리닝 방법.
  4. 청구항 3에 있어서, 상기 측정된 발현수준 또는 활성이 후보물질과 접촉하지 않은 대조군 대비 감소하는 경우, 상기 후보물질을 항진균제로 선별하는 단계를 더 포함하는, 크립토코쿠스 네오포만스에 대한 항진균제 스크리닝 방법.
  5. 청구항 1의 방법에 따라 스크리닝된 저해제를 포함하는 항진균용 조성물.
  6. 크립토코쿠스 네오포만스의 HBF1 유전자의 발현, 또는 이로부터 발현되는 단백질의 발현 또는 활성을 억제하는 물질을 포함하는, 크립토코쿠스 네오포만스에 대한 항진균용 조성물.
  7. 청구항 6에 있어서, 상기 물질은 핵산, 항체, 앱타머, 펩타이드, 단백질, 화합물 또는 천연물인, 크립토코쿠스 네오포만스에 대한 항진균용 조성물.
  8. 청구항 6에 있어서, 상기 물질은 HBF1 유전자의 mRNA에 상보적인 서열을 가지는 안티센스, siRNA(small interfering RNA) 또는 shRNA(short hairpin RNA) 핵산을 포함하는 크립토코쿠스 네오포만스에 대한 항진균용 조성물.
