Abstract
본 발명은 아미노산을 유효성분으로 함유하는 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 이의 용도에 관한 것으로, 본 발명에 따른 안정성 증진용 조성물은 바이러스 백신에서 감염 역가의 상실없이 예방학적으로 유효한 바이러스 역가를 효과적으로 유지할 수 있고, 백신의 운반 및 보관이 용이해질 수 있으므로, 관련 분야에 유용하게 활용될 수 있을 것이다.
Description
본 발명은 아미노산을 유효성분으로 함유하는 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 이의 용도에 관한 것이다.
바이러스 또는 바이러스 벡터를 면역원(항원)으로 하는 백신의 경우, 제조과정 중에 발생하는 교반(agitation) 및 동결융해(freeze-thawing) 단계, 그리고 제조 후의 보관 온도 등으로 면역원의 안정성이 보장되지 않는 경우가 있다.
백신 안정화제는 동결건조 가공 또는 동결 건조 후 보관 기간 동안 백신의 안정성을 향상시키기 위해서 백신 제형에 첨가되는 화학적 화합물로, 당해 분야에 널리 공지되어 있다. 이러한 안정화제 성분들은 단독으로 또는 다수 성분의 배합으로 사용되며 백신의 제조과정 또는 보관 중에 면역원(예컨대, 바이러스)의 능력을 보존하는 작용을 하며, 동결 공정 동안에 면역원의 집결을 방지하는 기능을 한다.
약독화 바이러스 백신은 주로 세포성 면역작용에 의하여 그 효과를 나타내므로, 체액성 면역작용에 의존하는 사독 백신에 비하여 1회 접종으로도 장기간의 면역효과를 나타낸다. 백신에 사용되는 약독화한 바이러스는 열에 민감하여 고온에서는 쉽게 그 활성을 유지할 수 없다. 또한, 열 뿐만 아니라 다양한 물리적, 화학적 자극에 의해서도 항원성을 상실할 수 있기 때문에 백신의 효능을 유지하기 위해서는 바이러스의 역가를 유지하기 위한 다양한 방법이 고려되어야 한다.
광견병, 아프리카돼지열병 등 공익을 위한 미끼백신의 사용에 있어서, 미끼에 포함된 바이러스는 상온 중에 노출시켜 공급해야 되는 특성이 있어 온도에 의한 바이러스 역가 감소가 크게 우려되는 백신의 형태이다. 안정화제의 개발을 통해 온도에 민감한 바이러스 백신의 유효기간을 증진시킬 수 있다.
한편, 한국등록특허 제1679723호에는 '안정화제 및 1종 이상의 생 약독화 플라비바이러스를 포함하는 백신 조성물'이 개시되어 있고, 한국공개특허 제2018-0135912호에는 '개선된 제형을 이용한 알파비바이러스의 안정화 조성물 및 방법'이 개시되어 있으나, 본 발명의 '아미노산을 유효성분으로 함유하는 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 이의 용도'에 대해서는 기재된 바가 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명자들은 바이러스를 면역원으로 하는 동물용 백신에서, 동결융해 또는 제조 후 보존 시 바이러스의 불활성화와 역가 감소를 억제하기 위해, 다양한 아미노산의 조합을 포함하는 백신 조성물용 안정화제를 제공하고자 하였다.
본 발명자는 글루탐산을 비롯한 12종의 아미노산으로 이루어진 백신 조성물용 안정화제를 제조한 후 ssRNA 바이러스 및 dsDNA 바이러스를 대상으로 안정화제 사용에 따른 바이러스의 생존능을 확인한 결과, 본 발명에서 제조한 안정화제가 다양한 바이러스의 저온/상온 보관 안정성 및 동결융해 스트레스에 대한 안정성을 부여하는 것을 확인함으로써, 본 발명을 완성하였다.
상기 과제를 해결하기 위해, 본 발명은 글루탐산(glutamic acid), 메티오닌(methionine), 발린(valine), 글루타민(glutamine), 아스파라긴(asparagine), 히스티딘(histidine), 세린(serine), 알라닌(alanine), 글리신(glycine), 트레오닌(threonine), 프롤린(proline), 아르지닌(arginine) 및 아스파르트산(aspartic acid)을 유효성분으로 포함하는, 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물을 제공한다.
또한, 본 발명은 상기 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 1종 이상의 항원을 포함하는, 백신 조성물을 제공한다.
또한, 본 발명은 동물용 백신 항원을 제조하는 단계; 및 제조된 동물용 백신 항원에 본 발명의 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물을 혼합하는 단계;를 포함하는, 안정성이 증진된 백신의 제조 방법을 제공한다.
본 발명의 항원의 안정성 증진용 조성물은 교반 및 동결융해를 포함하는 백신 제조 공정과 보관 중에 항원(면역원)인 바이러스를 안정화하는 효과가 있으므로, 동물용 바이러스 백신에서 감염 역가의 상실없이 예방학적으로 유효한 바이러스 역가를 효과적으로 유지할 수 있고, 백신의 운반 및 보관이 용이해질 수 있으므로, 관련 분야에 유용하게 활용될 수 있을 것이다.
도 1은 각기 다른 아미노산 조성으로 이루어진 안정화제 조성물(표 1)과 혼합한 바이러스(Akabane virus K-9 strain)의 25℃ 조건에서 생존능을 확인한 결과이다. Control은 1X PBS 첨가군이고, CVS75가 본 발명의 청구대상 안정화제이다.

도 5는 25℃ 조건에서 본 발명의 안정화제(CVS)가 바이러스(Foot-and-mouth disease virus O1 Manisa strain)의 안정화에 미치는 영향을 분석한 결과이다. Control은 1X PBS 첨가군이고, Sucrose는 기존에 사용되던 5% 수크로스를 첨가한 비교대조군이다.
도 6은 4℃ 조건에서 본 발명의 안정화제(CVS)가 바이러스(recombinant Adeno virus KVCC-VR1500041)의 안정화에 미치는 영향을 분석한 결과이다. Control은 1X PBS 첨가군이고, Trehalose는 기존에 사용되던 5% 트레할로스를 첨가한 비교대조군이다.
도 7은 25℃ 조건에서 본 발명의 안정화제(CVS)가 바이러스(African swine fever virus Yc1-Fc PSV strain)의 안정화에 미치는 영향을 분석한 결과이다. Control은 1X PBS 첨가군이다.
도 8은 4℃ 또는 25℃ 조건에서 본 발명의 안정화제(CVS)가 바이러스(Aujeszky's disease virus YS400 strain)의 안정화에 미치는 영향을 분석한 결과이다. Control은 1X PBS 첨가군이다.
도 9는 반복된 동결융해 조건에서 본 발명의 안정화제(CVS)가 바이러스(Akabane virus) 안정화에 미치는 영향을 분석한 결과이다. Control은 1X PBS 첨가군이다.
본 발명의 목적을 달성하기 위하여, 본 발명은 글루탐산(glutamic acid), 메티오닌(methionine), 발린(valine), 글루타민(glutamine), 아스파라긴(asparagine), 히스티딘(histidine), 세린(serine), 알라닌(alanine), 글리신(glycine), 트레오닌(threonine), 프롤린(proline), 아르지닌(arginine) 및 아스파르트산(aspartic acid)을 유효성분으로 포함하는, 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물을 제공한다.
본 발명에 따른 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물은 상기 글루탐산 100 중량부에 대하여 메티오닌 40 내지 60 중량부, 발린 40 내지 60 중량부, 글루타민 40 내지 60 중량부, 아스파라긴 40 내지 60 중량부, 히스티딘 40 내지 60 중량부, 세린 40 내지 60 중량부, 알라닌 40 내지 60 중량부, 글리신 40 내지 60 중량부, 트레오닌 40 내지 60 중량부, 프롤린 40 내지 60 중량부, 아르지닌 40 내지 60 중량부 및 아스파르트산 40 내지 60 중량부로 이루어진 것일 수 있고, 보다 바람직하게는 글루탐산 100 중량부에 대하여 메티오닌 45 내지 55 중량부, 발린 45 내지 55 중량부, 글루타민 45 내지 55 중량부, 아스파라긴 45 내지 55 중량부, 히스티딘 45 내지 55 중량부, 세린 45 내지 55 중량부, 알라닌 45 내지 55 중량부, 글리신 45 내지 55 중량부, 트레오닌 45 내지 55 중량부, 프롤린 45 내지 55 중량부, 아르지닌 45 내지 55 중량부 및 아스파르트산 45 내지 55 중량부로 이루어진 것일 수 있으며, 가장 바람직하게는 글루탐산 100 중량부에 대하여 메티오닌 50 중량부, 발린 50 중량부, 글루타민 50 중량부, 아스파라긴 50 중량부, 히스티딘 50 중량부, 세린 50 중량부, 알라닌 50 중량부, 글리신 50 중량부, 트레오닌 50 중량부, 프롤린 50 중량부, 아르지닌 50 중량부 및 아스파르트산 50 중량부로 이루어진 것일 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 상기 아미노산은 D형 아미노산과 L형 아미노산을 모두 포함할 수 있고, 바람직하게는 L형 아미노산일 수 있으나, 이에 제한되지 않는다
본 명세서에 개시된 바람직한 성분 범위는 다른 특징들 중에서도 동결융해 스트레스에 대한 안정성 및 백신의 면역원인 항원을 안정화시키고, 상온에서 장기간 동안 열-불활성화에 대해 보호하는데 유리하게 사용될 수 있다.
또한, 본 발명에 따른 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물에 있어서, 상기 항원은 동물에서 질병을 유발하는 바이러스, 세균 또는 기생충의 전체 세포이거나 이들 유래의 재조합 단백질일 수 있으나, 이에 제한되지 않는다. 또한, 상기 바이러스, 세균 또는 기생충의 전체 세포는 살아있는 전체 세포 또는 또는 화학적/물리적 방법으로 불활성화시킨 세포를 모두 포함할 수 있다. 화학적 불활성화는 예를 들어 효소, 포름알데하이드, 베타-프로피오락톤, 에틸렌-이민 또는 그것의 유도체로 처리함으로써 수행될 수 있으며, 물리적 불활성화는 에너지가 충분한 방사능, 예를 들면 UV 광을 조사함으로써 수행될 수 있으나, 이에 제한되지 않는다.
본 발명에서 용어 "백신용 항원"은 적당한 세포와 접촉하여 유입됨에 따라 민감성 및/또는 면역 반응성 상태를 유도시키고, 생체 내(in vivo) 또는 시험관 내(in vitro)에서 이와 같이 감작된 대상체의 면역 세포 및/또는 항체와 입증 가능한 방식으로 반응하는 모든 물질을 지칭한다. 본 명세서에서 용어 "항원"은 "면역원"이라는 용어와 동일한 의미로 사용될 수 있으며, 바람직하게 숙주 면역 체계가 그 항원에 특이적인 분비성, 체액성 및/또는 세포성 면역 반응을 일으키도록 촉진할 수 있는 하나 또는 그 이상의 에피토프(epitope)를 포함하는 분자를 의미한다.
본 발명은 또한, 본 발명에 따른 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 1종 이상의 항원을 포함하는, 백신 조성물을 제공한다.
본 발명의 백신 조성물에 있어서, 상기 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물은 전술한 것과 같다.
본 발명에서 용어 "백신(vaccine)"은 감염성 질환(infectious disease)에 대한 면역 기능을 가진 약학적 조성물을 의미한다.
또한, 본 발명의 백신 조성물에 있어서, 상기 백신은 항원(antigen)이 바이러스 또는 바이러스 유래 재조합 단백질인 백신일 수 있으나, 이에 제한되지 않는다.
본 발명의 일 구현 예에 따른 백신 조성물에 있어서, 상기 항원은 바람직하게는 단일가닥 음성 RNA 외피보유 바이러스(ss(-)RNA enveloped virus), 단일가닥 양성 RNA 외피보유 바이러스(ss(+)RNA enveloped virus), 단일가닥 양성 RNA 비-외피보유 바이러스(ss(+)RNA non-enveloped virus), 이중가닥 DNA 외피보유 바이러스(dsDNA enveloped virus) 또는 이중가닥 DNA 비-외피보유 바이러스(dsDNA non-enveloped virus)일 수 있고, 보다 바람직하게는 오쏘분야바이러스 속(Orthobunyavirus) 또는 리사바이러스 속(Lyssavirus)의 단일가닥 음성 RNA 외피보유 바이러스; 플라비바이러스 속(Flavivirus)의 단일가닥 양성 RNA 외피보유 바이러스; 아프토바이러스 속(Aphthovirus)의 단일가닥 양성 RNA 비-외피보유 바이러스; 아데노바이러스과의 이중가닥 DNA 비-외피보유 바이러스; 또는 아스피바이러스 속(Asfivirus) 또는 바리셀로바이러스 속(Varicellovirus)의 이중가닥 DNA 외피보유 바이러스;일 수 있으며, 가장 바람직하게는 아카바네 바이러스(Akabane virus) 또는 광견병 바이러스(Rabies virus) 등의 단일가닥 음성 RNA 외피보유 바이러스; 일본뇌염 바이러스(Japanese encephalitis virus) 등의 단일가닥 양성 RNA 외피보유 바이러스; 구제역병 바이러스(Foot-and-mouth disease virus) 등의 단일가닥 양성 RNA 비-외피보유 바이러스; 아데노바이러스 등의 이중가닥 DNA 비-외피보유 바이러스; 또는 아프리카돼지열병 바이러스(African swine fever virus) 또는 오제스키병 바이러스(Aujeszky's disease virus) 등의 이중가닥 DNA 외피보유 바이러스;일 수 있으나, 이에 제한되지 않는다. 또한, 상기 바이러스는 생 바이러스, 생 약독화 바이러스 또는 불활성화 바이러스일 수 있으나, 이에 제한되지 않는다.
또한, 당해 분야의 통상적인 기술자는 안정성 증진용 조성물 대 항원의 부피 비의 변화를 청구된 발명을 실시하는데 적용시킬 수 있으며, 이는 백신 면역원인 항원의 역가 또는 활성에 근거하여 안정성 증진용 조성물 혼합 비율의 변화를 요할 것임을 자명하게 알 것이다. 따라서, 본 발명에서 백신을 생성시키기 위해서 1:1의 안정성 증진용 조성물/항원 혼합물로만 제한하는 것은 아니다.
또한, 본 발명에 따른 백신 조성물은 약학적으로 허용되는 담체 및/또는 보조제(adjuvant)를 추가로 포함할 수 있다. 또한, 상기 담체 및/또는 보조제 외에 본 발명의 백신 조성물은 부형제 및/또는 희석제를 추가로 더 포함할 수 있다.
본 명세서에서 용어 "약학적으로 허용가능한"이란, 생리학적으로 허용되고 대상 개체에게 투여될 때, 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 물질을 말한다. 상기 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
본 명세서에서 상기 "보조제(adjuvant)"는 백신 또는 약학적으로 활성 있는 성분들에 첨가되어 면역 반응을 증가시키거나 및/또는 영향을 주는 물질 또는 조성물을 의미한다. 대표적으로 면역원(immunogen)용 캐리어(carrier) 또는 보조 물질 및/또는 다른 약학적으로 활성 있는 물질 또는 조성물을 통상적으로 의미한다. 전형적으로, 용어 '보조제'는 넓은 개념에서 해석되어야 하며, 상기 보조제에 통합되거나 상기 보조제와 함께 투여된 항원의 면역원성(immunogenicity)을 증진시킬 수 있는 넓은 범위의 물질을 의미한다. 또한, 보조제는, 이에 제한되지 않고, 면역 강화제(immune potentiator), 항원 전달계 또는 이들의 조합으로 나뉘어질 수 있다. 당해 분야에서 특정 백신의 첨가제의 수는 다양하며, 제조공정과 백신의 용도에 따라 달라진다.
본 발명의 백신 조성물은 포유동물에 투여시, 활성 성분의 신속한 방출, 지속 또는 지연된 방출이 가능하도록 당업계에 공지된 방법을 사용하여 제형화될 수 있다. 제형은 액상 또는 동결건조 형태일 수 있으나, 이에 제한되지 않는다.
본 발명은 또한, 동물용 백신 항원을 제조하는 단계; 및 제조된 동물용 백신 항원에 본 발명의 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물을 혼합하는 단계;를 포함하는, 안정성이 증진된 백신의 제조 방법을 제공한다.
본 발명의 제조 방법에 있어서, 상기 안정성 증진용 조성물의 구성 및 항원의 종류는 전술한 것과 같다.
또한, 본 발명의 일 구현 예에 따른 제조 방법에 있어서, 상기 혼합은 안정성 증진용 조성물 : 항원의 비율이 1:1 내지 1:3의 부피비인 것일 수 있으나, 이에 제한되지 않는다.
이하, 본 발명을 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
제조예 1. 바이러스 백신용 안정화제의 제조
본 발명자는 한국등록특허 제1759694호의 내용을 참고하여, 바이러스를 면역원(항원)으로 하는 백신 조성물에서 항원(면역원)의 안정성을 높일 수 있는 안정화제(CAVAC Vaccine Stabilizer, CVS)를 개발하고자 하였다. 다양한 농도 조합으로 이루어진 안정화제 조성물을 하기 표 1의 조성으로 제조하였다. 하기 표 1의 안정화제는 증류수 1 L를 기준으로 제조한 후 pH를 6.5~7.0으로 조정한 후 필터로 여과하여 사용하였으며, 각각의 시약은 시그마-알드리치사에서 구매하였다.

제조예 2. 안정화제를 포함하는 바이러스 백신의 제조
본 발명에서는 아카바네 바이러스(Akabane virus) 또는 광견병 바이러스(Rabies virus) 등의 단일가닥 음성 RNA 외피보유 바이러스(ss(-)RNA enveloped virus); 일본뇌염 바이러스(Japanese encephalitis virus) 등의 단일가닥 양성 RNA 외피보유 바이러스(ss(+)RNA enveloped virus); 구제역병 바이러스(Foot-and-mouth disease virus) 등의 단일가닥 양성 RNA 비-외피보유 바이러스(ss(+)RNA non-enveloped virus); 아데노바이러스 등의 이중가닥 DNA 비-외피보유 바이러스(dsDNA non-enveloped virus); 또는 아프리카돼지열병 바이러스(African swine fever virus) 또는 오제스키병 바이러스(Aujeszky's disease virus) 등의 이중가닥 DNA 외피보유 바이러스(dsDNA enveloped virus)를 사용하였다.
상기 각 바이러스는 Vero 세포주에 감염시켜 바이러스 원액(bulk)을 수득하였으며, 상기 바이러스 bulk를 제조예 1에서 제조한 안정화제(CVS75)와 1:1의 부피비로 혼합하여 사용하였다.
실시예 1. 바이러스 안정화 효과가 우수한 최적 조성의 안정화제 선발
본 발명자는 표 1의 각 후보 안정화제 조성물의 효과를 검증하기 위해, 각각의 안정화제를 아카바네 바이러스(Akabane virus K-9 strain) 원액과 1:1로 혼합하여 25℃에서 바이러스의 생존능을 분석하였다. 안정화제와 혼합한 바이러스의 바이러스 생존능은 Tissue culture infection dose50 (TCID50) 방법을 통해 측정하였다. 구체적으로, 96-웰 세포 배양 플레이트에 Vero-k 세포를 1.2 X 104 cells/well의 농도로 분주 및 배양하여 단일세포층(monolayer)을 형성시킨다. 이 후 세포 배양배지를 제거하고, 5% FBS (fetal bovine serum)을 보충한 a-MEM 배지로 바이러스를 원액부터 10진 단계 희석한 뒤 상기 96-웰 세포 배양 플레이트에 분주하여 37℃, 5% CO2 조건에서 배양하면서 CPE (cytopathic effect) 양상을 확인하여 결과를 판정하고, Reed-Muench method로 역가(titer)를 계산하였다.
분석 결과, CVS75를 사용한 경우 30일까지 높은 수준의 바이러스 생존능을 나타내었으나, 대조군, CVS43 및 CVS472의 안정화제를 사용한 경우 시간이 경과함에 따라 바이러스 생존능이 현저하게 감소하기 시작하여 20일차에 모든 바이러스가 사멸하는 것을 확인하였다(도 1). 이를 통해 최종적으로 13종의 아미노산으로 이루어진 안정화제인 CVS75를 선발하여 이후 실험을 진행하였다.
실시예 2. 다양한 바이러스에 대한 안정화제의 안정화 효과 비교
본 발명자는 상기 실시예 1을 통해 선발된 안정화제 조성물인 CVS75(이하, CVS)의 다양한 바이러스에 대한 안정화 효과를 분석하였다.
2-1. 단일가닥 음성 RNA 외피보유 바이러스(ss(-)RNA enveloped virus)에 대한 안정화 효과
아카바네 바이러스(Akabane virus) K-9 strain은 국립수의과학검역원에서 분양받아 사용하였다. 아카바네 바이러스 bulk (106.7 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 4℃ 또는 25℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 그 결과, 도 2에 개시된 바와 같이 4℃ 조건에서는 PBS를 혼합한 대조군과 CVS 혼합군 간에 큰 차이가 확인되지 않았으나, 25℃ 조건에서는 대조군의 바이러스 생존능이 시간이 경과함에 따라 현저하게 감소되어 28일차에는 모두 사멸되는 것으로 확인된 반면, CVS 혼합군에서는 아카바네 바이러스의 생존능이 35일차에도 매우 높게 유지되는 것이 관찰되었다.
또한, 광견병 바이러스(Rabies virus) ERAGS strain은 국립수의과학검역원에서 분양받아 사용하였으며, 광견병 바이러스 bulk (107.5 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 4℃ 또는 25℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 또한, 종래 바이러스 안정화에 사용되던 5%(w/v) 트레할로스를 혼합한 비교대조군을 함께 분석하였다. 그 결과, 도 3에 개시된 바와 같이 광견병 바이러스의 경우 4℃ 및 25℃ 조건 모두에서 대조군 및 비교대조군(트레할로스 혼합군) 대비 본 발명의 안정화제를 혼합한 실험군에서 현저히 우수한 바이러스 생존능을 확인할 수 있었다.
2-2. 단일가닥 양성 RNA 외피보유 바이러스(ss(+)RNA enveloped virus)에 대한 안정화 효과
본 발명에서는 일본뇌염 바이러스(Japanese encephalitis virus) KV1899 strain을 사용하였다. 일본뇌염 바이러스 bulk (107.25 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 25℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 그 결과, 도 4에 개시된 바와 같이 4일째까지는 대조군과 CVS 혼합군간 바이러스 생존능에 큰 차이가 없었으나, 5일째부터 대조군의 바이러스는 생존능이 급격하게 떨어지는 것을 확인할 수 있었다.
2-3. 단일가닥 양성 RNA 비-외피보유 바이러스(ss(+)RNA non-enveloped virus)에 대한 안정화 효과
본 발명에서는 구제역병 바이러스(Foot-and-mouth disease virus) O1 Manisa strain을 사용하였다. 구제역병 바이러스 bulk (105.7 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 25℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 또한, 종래 바이러스 안정화에 사용되던 5%(w/v) 수크로스를 혼합한 비교대조군을 함께 분석하였다. 그 결과, 도 5에 개시된 바와 같이 대조군의 경우 시간경과에 따라 바이러스 생존능이 감소되어 4일째에 모두 사멸되는 것으로 확인되었으며, 비교군은 수크로스 혼합군은 7일차에 바이러스가 모두 사멸되는 것으로 관찰되었다. 그러나, 본 발명의 안정화제를 혼합한 실험군(CVS)은 7일차에도 높은 바이러스 생존능을 나타냄을 알 수 있었다.
2-4. 이중가닥 DNA 비-외피보유 바이러스(dsDNA non-enveloped virus)에 대한 안정화 효과
본 발명에는 재조합 아데노바이러스 (Korea Veterinary Culture Collection accession no. KVCC-VR1500041)를 사용하였다. 상기 아데노바이러스는 293A 세포에 배양한 후 세포변성 효과를 확인하고 바이러스 bulk를 수득하여 실험에 사용하였다. 아데노바이러스 bulk (109.0 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 4℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였으며, 종래 바이러스 안정화에 사용되던 5%(w/v) 트레할로스를 혼합한 비교대조군을 함께 분석하였다. 그 결과, 도 6에 개시된 바와 같이 20일째까지는 모든 군에서 바이러스 생존능에 큰 차이가 확인되지 않았으나, 20일째 이후 대조군 및 트레할로스 혼합군은 시간 경과에 따라 바이러스 생존능이 감소되는 것이 관찰되었고, CVS 혼합군은 30일째까지 높은 바이러스 생존능을 보여주었고, 30일 이후 바이러스 생존능이 감소되어, 대조군 및 트레할로스 혼합군 대비 우수한 바이러스 안정화 효과를 나타내었다.
2-5. 이중가닥 DNA 외피보유 바이러스(dsDNA enveloped virus)에 대한 안정화 효과
아프리카돼지열병 바이러스(African swine fever virus, ASFV) Yc1-Fc PSV strain을 사용하였으며, ASFV는 Cas-01 세포에 배양한 후 세포변성 효과를 확인하고 바이러스 bulk를 수득하여 실험에 사용하였다. ASFV bulk (106.1 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 25℃ 또는 37℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 그 결과, 도 7에 개시된 바와 같이 대조군은 25℃ 및 37℃ 조건 모두에서 시간 경과에 따라 바이러스 생존능이 감소되기 시작하여 55일째에 모든 바이러스가 사멸한 것으로 관찰되었으나, CVS 혼합군의 경우 25℃ 조건에서는 83일째까지 높은 바이러스 생존능을 보여주었으며, 37℃ 조건에서는 83일째에 모든 바이러스가 사멸한 것으로 확인되었다.
또한, Vero-k 세포에서 배양한 오제스키병 바이러스(Aujeszky's disease virus) YS400 strain을 실험에 사용하였으며, 오제스키병 바이러스 bulk (106.5 TCID50/㎖)를 CVS와 1:1의 부피비로 혼합한 후 4℃ 또는 25℃에서 보관하며 바이러스의 생존능을 TCID50 방법을 통해 측정하였다. 그 결과, 도 8에 개시된 바와 같이 4℃ 조건에서 CVS 혼합군은 55일째까지 바이러스 생존능의 감소가 확인되지 않았다. 또한, 25℃ 조건에서 대조군은 시간 경과에 따라 바이러스 생존능이 감소되어 55일째에 모두 바이러스가 사멸된 것을 관찰되었으나, CVS 혼합군은 대조군 대비 우수한 바이러스 생존능을 보여주었다.
이상의 결과를 통해 본 발명의 안정화제는 바이러스 백신의 제조 후 보관 중에 면역원인 바이러스를 안정화하는 효과가 우수하고, 바이러스 백신에서 감염 역가의 상실없이 예방학적으로 유효한 바이러스 역가를 효과적으로 유지하는게 유용하게 활용될 수 있음을 시사하였다.
실시예 3. 동결융해 스트레스에 따른 안정화제의 바이러스 안정성 영향 분석
본 발명에 따른 안정화제(CVS)가 백신 제조 공정 중에 수반되는 동결융해 단계에서 바이러스의 안정성을 향상시킬 수 있는지 확인하기 위해 실험을 실시하였다. 간단하게, 아카바네 바이러스(Akabane virus) bulk (106.7 TCID50/㎖)와 안정화제를 1:1의 부피비로 혼합하여 총 100 ㎖를 준비한 후, -74℃의 초저온냉동고(Deep freezer)에서 밤새 동결시킨 뒤 상온(25±2℃)에서 4시간 동안 융해시키는 과정을 1회(cycle)로 하여 각 횟수마다 융해된 시료에서 일부를 취하여 TCID50 방법으로 바이러스 역가를 측정하였다.
그 결과, 동결융해 과정이 4회 이상 반복되자 대조군(PBS 첨가)의 경우 바이러스의 생존능이 감소되기 시작하였으나, CVS를 첨가한 시험군은 동결융해 과정이 8회 반복된 조건에서도 바이러스의 생존능이 감소되지 않는 것을 확인할 수 있었다(도 9).
또한, CVS를 바이러스 bulk와 1:1 또는 1:3의 부피비로 혼합한 뒤 동결건조 제형을 제조해 본 결과, 본 발명의 안정화제가 동결건조 제형에 아무런 영향도 미치지 않음을 확인할 수 있었다. 이상의 결과를 통해 본 발명에 따른 안정화제(CVS)가 동결융해 스트레스 조건에서 백신의 면역원인 바이러스의 안정성을 크게 향상시킬 수 있음을 알 수 있었다.
Claims (8)
- 글루탐산(glutamic acid), 메티오닌(methionine), 발린(valine), 글루타민(glutamine), 아스파라긴(asparagine), 히스티딘(histidine), 세린(serine), 알라닌(alanine), 글리신(glycine), 트레오닌(threonine), 프롤린(proline), 아르지닌(arginine) 및 아스파르트산(aspartic acid)을 유효성분으로 포함하는, 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물.
- 제1항에 있어서, 상기 조성물은 글루탐산 100 중량부에 대하여 메티오닌 40 내지 60 중량부, 발린 40 내지 60 중량부, 글루타민 40 내지 60 중량부, 아스파라긴 40 내지 60 중량부, 히스티딘 40 내지 60 중량부, 세린 40 내지 60 중량부, 알라닌 40 내지 60 중량부, 글리신 40 내지 60 중량부, 트레오닌 40 내지 60 중량부, 프롤린 40 내지 60 중량부, 아르지닌 40 내지 60 중량부 및 아스파르트산 40 내지 60 중량부인 것을 특징으로 하는, 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물
- 제1항에 있어서, 상기 항원은 동물에서 질병을 유발하는 바이러스, 세균 또는 기생충의 전체 세포이거나 이들 유래의 재조합 단백질인 것을 특징으로 하는, 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물.
- 제1항에 따른 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물 및 1종 이상의 항원을 포함하는, 백신 조성물
- 제4항에 있어서, 상기 항원은 단일가닥 RNA 바이러스 또는 이중가닥 DNA 바이러스인 것을 특징으로 하는 백신 조성물.
- 제4항에 있어서, 상기 백신 조성물은 약학적으로 허용가능한 담체 또는 보조제(adjuvant)를 추가로 포함하는 것을 특징으로 하는 백신 조성물.
- 제4항에 있어서, 상기 백신 조성물은 액상 또는 동결건조 제형인 것을 특징으로 하는 백신 조성물.
- 동물용 백신 항원을 제조하는 단계; 및제조된 동물용 백신 항원에 제1항의 동물용 백신 또는 진단용 항원의 안정성 증진용 조성물을 혼합하는 단계;를 포함하는, 안정성이 증진된 백신의 제조 방법.
