本国際出願は、2022年10月20日に日本国特許庁に出願された日本国特許出願第2022-168359号に基づく優先権を主張するものであり、日本国特許出願第2022-168359号の全内容を本国際出願に参照により援用する。
本開示は、経口製剤及びその製造方法に関する。
ニコチンアミドモノヌクレオチド(NMN、β-NMN)は、ニコチンアミドアデニンジヌクレオチド(NAD+)の生合成における中間代謝産物である。最近の研究から、NMNは、長寿遺伝子を活性化し、抗老化作用を示すことが明らかとされてきている。
このようなNMNの特性に基づいて、近年、NMNを含有する製剤が開発されている。例えば、特許文献1には、NMNを含有するカプセル製剤が開示されている。
一方で、NMNは水中において不安定であるという問題があった。そのため、製剤の加工時において、NMNの分解等が生じ、高濃度のNMNを製剤中に含めることが困難である場合があった。また、それに伴い、十分な量のNMNを生体内へ吸収させることが困難である場合があった。

以下、本開示の例示的な実施形態について説明する。
1.経口製剤
本開示において、経口製剤は、有効成分として、リポソーム化されたニコチンアミドモノヌクレオチド(以下、NMN)を含む。

本開示において、「NMN」は、ニコチンアミドアデニンジヌクレオチド(NAD+)の生合成における中間代謝産物である。なお、NMNは、その薬理学的に許容される塩として存在してもよい。薬理学的に許容される塩とは、本開示のNMNが、無機又は有機の塩基又は酸と結合して形成した塩であって、医薬等として体内に投与することが許容可能な塩のことである。
本開示において、「リポソーム化」とは、目的成分を内包するリポソームを製造することをいい、リポソーム化において、目的成分とリポソームの膜材料となる任意の脂質(例えば、リン脂質、コレステロール類、脂肪酸等)とが混ぜ合わされ、目的成分を内包するリポソームが形成される。本開示において、リポソーム化は、公知の方法で行ってもよい。そのような公知の方法として、例えば、本開示のリポソーム化NMNは、少なくともNMN、レシチン、糖アルコールを混合して得られるリポソーム原料を撹拌混合することにより調製することができる。
本開示において、糖アルコールとしては、特に限定されるものではないが、例えば、マンニトール、ソルビトール、キシリトール、イソマルト、エリスリトール、還元麦芽糖、ラクチトール等が挙げられ、好ましくは、還元麦芽糖である。
また、リポソーム原料には、さらにリポソームの分野で配合され得る公知の成分を配合してもよい。
リポソーム化されたNMNは、いかなる大きさのものを含んでいてもよいが、平均粒径が1~1000μmであることが好ましく、平均粒径が10~100μmであることがさらに好ましい。なお、ここでいう「粒径」は、動的光散乱によって測定された粒子の直径を意味する。

経口製剤におけるリポソーム化されたNMNの含有量は、NMNとして、経口製剤100重量%あたり5~20重量%であってもよく、より好ましくは、7~20重量%であり、さらに好ましくは、9~20重量%であり、例えば、9~10重量%である。従来は、NMNの水中での不安定性等により、NMNを製剤中に高濃度で含有させることは困難であった。本開示において、NMNをリポソーム化することにより高濃度のNMNを含有する経口製剤を得ることができる。なお、NMNの含有量が20重量%を超えると、NMNを完全にリポソーム原料に溶解させることが困難であり、NMNのリポソーム化が困難になり得る。
また、本開示の経口製剤中にはNMN以外のその他の有効成分が含まれてもよい。そのような有効成分としては、例えば、長寿遺伝子を活性化させる成分が挙げられる。該成分は、公知の合成方法により人工的に合成されてもよいし、天然の植物から公知の抽出方法を用いて抽出された抽出物として用いられてもよい。
長寿遺伝子を活性化させる成分としては、特に限定されるものではないが、プテロスチルベン、レスベラトロール、ブラックジンジャー、脂肪酸トリプタミド、ウロリチン等が挙げられる。長寿遺伝子を活性化させる成分としては、好ましくは、レスベラトロールであり、更に好ましくは、メチル化レスベラトロールである。レスベラトロールのメチル化により、通常のレスベラトロールよりも、生体への吸収性及びバイオアベイラビリティを高めることができる。ここで、メチル化レスベラトロールは、インドキノキに多く含まれていることが知られている。したがって、本開示の経口製剤において、メチル化レスベラトロールを含有するインドキノキの抽出物を、長寿遺伝子を活性化させる成分として用いてもよい。
本開示の経口製剤において、その他の有効成分を0.001重量%~10重量%の範囲で含むことができ、好ましくは0.01重量%~10重量%、より好ましくは0.05重量%~5重量%、さらに好ましくは0.1重量%~1重量%の範囲で含むのがよい。
本開示の経口製剤は、上記NMN及び/又はその他の有効成分に加えて、他の成分、例えば、賦形剤、結合剤、崩壊剤、防腐剤、コーティング剤、分散剤、流動化剤、安定化剤、矯味剤、着色剤、及び香料等を含んでもよい。
本開示の経口製剤が投与される対象としては、特に限定されるものではないが、哺乳動物、例えばヒト及びチンパンジー等の霊長類、ラット、マウス、及びウサギ等の実験動物、ブタ、ウシ、ウマ、及びヒツジ等の家畜動物、並びにイヌ及びネコ等の愛玩動物が挙げられ、好ましくはヒトである。
本開示の経口製剤の投与回数、及び投与期間は、当業者であれば、対象の性別、体重、年齢、並びに症状等の種々の要因を考慮して適宜定めることができる。例えば、本開示の経口製剤の投与回数は、特に限定するものではないが、1日3回、1日2回、1日1回、2日に1回、3日に1回、1週間に1回、2週間に1回、1カ月に1回等であってもよい。また、投与期間は、特に限定するものではないが、1日、2日、3日、1週間、2週間、1カ月、半年、一年、又はそれ以上であってよい。
本開示の経口製剤は、化粧品、医薬部外品及び医薬品、又は食品として使用可能である。また、本開示の経口製剤は、例えば、対象における疾患又は状態を治療、改善及び/又は予防するために使用することができる。疾患又は状態としては、例えば、老化、疲労、体力低下、睡眠障害、神経疾患、肥満、糖尿病、機能障害、眼疾患、耳疾患、肌質低下等が挙げられる。
2.経口製剤の製造方法
本開示の経口製剤は、リポソーム化されたNMNを造粒することによって得ることができる。

具体的には、本開示の経口製剤の製造方法は、NMNと脂質と糖アルコールとを用いてリポソームを調製する工程と、調製したリポソームに結合液を加えて造粒する工程と、造粒した造粒物を乾燥する工程と、を含む。
本方法において用いられるNMN、糖アルコール、その他の有効成分としては、上述の「1.経口製剤」の項で述べたものを用いることができる。
以下、本開示の経口製剤の製造方法について、詳述する。

第一の工程として、NMNと脂質と糖アルコールとを用いてリポソームを調製する。リポソームの調製は、公知の方法で行ってもよい。例えば、本開示のリポソームは、少なくともNMN、レシチン、糖アルコールを混合して得られるリポソーム原料を撹拌混合することにより調製することができる。なお、粉末のリポソームを得るために、リポソームを含む溶液を公知の方法により乾燥(例えば、噴霧乾燥)させてもよい。また、リポソームの調製に際し、リポソームの分野で配合され得る公知の成分をリポソーム原料として配合してもよい。なお、本開示の経口製剤がNMN以外のその他の有効成分(例えば、長寿遺伝子を活性化させる成分)を含む場合には、調製したリポソームに対して、さらにその他の有効成分を添加してもよい。一例として、本開示の経口製剤の製造方法は、調製したリポソームに対して、メチル化レスベラトロールさらに加える工程を含んでもよい。
経口製剤におけるNMNと糖アルコールの配合割合としては、NMN:糖アルコールが1:2~1:20であってもよく、好ましくは1:3~1:15であり、より好ましくは1:4~1:10であり、さらに好ましくは、1:5~1:6である。
第二の工程として、調製したリポソームに結合液を加えて造粒を行う。
なお、調製したリポソームに対して、さらにその他の有効成分を添加する場合、調製したリポソームにその他の有効成分と結合液とを加える順番は、特に限定されるものではない。具体的には、調製したリポソームに対して、先にその他の有効成分を加えてもよいし、先に結合液を加えてもよい。また、調製したリポソームに対して、その他の有効成分と結合液とを同時に加えてもよい。
なお、調製したリポソームに対して、さらにその他の有効成分を添加する場合、調製したリポソームにその他の有効成分と結合液とを加える順番は、特に限定されるものではない。具体的には、調製したリポソームに対して、先にその他の有効成分を加えてもよいし、先に結合液を加えてもよい。また、調製したリポソームに対して、その他の有効成分と結合液とを同時に加えてもよい。
造粒方法は、特に限定されるものではなく、公知の方法を用いることができる。例えば、造粒方法としては、乾式造粒法、湿式造粒法等が挙げられる。乾式造粒法として、圧縮造粒法、ブリケット造粒法等が挙げられ、湿式造粒法としては、流動層造粒法、攪拌造粒法、転動造粒法、押出造粒法等が挙げられる。造粒方法としては、製造性等の点から湿式造粒法が好ましく、攪拌造粒法がさらに好ましい。
造粒条件は、任意の条件をとることができ、例えば、攪拌造粒法にて造粒を行う場合は、攪拌羽根(アジテーター)の回転速度100~500rpm(例えば、200~250rpm)、粉砕羽根(チョッパー)の回転速度500~3000rpm(例えば、1500~2500rpm)で攪拌を行うことができる。
結合液としては、例えば、水、無水エタノール、90~95%エタノールを用いてもよく、95%エタノールが好ましい。
結合液の添加量は、調製したリポソームを含む造粒に用いる材料の総量に対し、10~30重量%となる量であることが好ましく、より好ましくは13~25重量%であり、さらに好ましくは13~20重量%であり、さらに好ましくは15~20重量%である。

第三の工程として、造粒した造粒物を乾燥する。乾燥方法としては、特に限定されないが、箱式通気型式乾燥機等の乾燥機を使用して乾燥することができる。乾燥条件は、乾燥方法に応じて適宜選定できる。例えば、箱式通気型式乾燥機を用いて乾燥を行う場合は、乾燥温度40~45℃で乾燥させることができる。乾燥温度が高すぎると、経口製剤においてNMNが減衰し得る。乾燥温度40~45℃で乾燥させることにより、好適に本開示の経口製剤を製造できる。また、箱式通気型式乾燥機を用いて乾燥を行う場合は、乾燥時間5~8時間で乾燥させることができる。これにより、好適に本開示の経口製剤を製造できる。
なお、本開示の経口製剤の製造方法において、乾燥後の造粒物を、適宜篩等を選択して篩過することにより、所望の粒径となるように調整することができる。例えば、乾燥後の造粒物は、10~50メッシュの篩を通過したものであってもよく、好ましくは、15~25メッシュの篩を通過したものであってもよい。
なお、経口製剤の製造のために、乾燥後の造粒物に対して、本技術分野において公知の製造用剤を配合し、上述の攪拌造粒法における造粒条件と同様の条件にて撹拌混合してもよい。例えば、製造用剤として、ステアリン酸カルシウム、二酸化ケイ素等を配合してもよい。
[本開示の効果]
本開示の経口製剤は、有効成分として、リポソーム化されたNMNを含有する。また、本開示の経口製剤において、ニコチンアミドモノヌクレオチドの含有量は、経口製剤100重量%あたり9~20重量%であってもよい。これにより、NMNのリポソーム化によって水中におけるNMNの不安定性を軽減することができ、高濃度のNMNを製剤中に含めることができる。また、本開示の経口製剤は、リポソーム化NMNを含有することにより、NMNの高い生体内吸収性及び/又は組織浸透性を実現でき、製剤の特性を向上させることができる。
本開示の経口製剤は、有効成分として、リポソーム化されたNMNを含有する。また、本開示の経口製剤において、ニコチンアミドモノヌクレオチドの含有量は、経口製剤100重量%あたり9~20重量%であってもよい。これにより、NMNのリポソーム化によって水中におけるNMNの不安定性を軽減することができ、高濃度のNMNを製剤中に含めることができる。また、本開示の経口製剤は、リポソーム化NMNを含有することにより、NMNの高い生体内吸収性及び/又は組織浸透性を実現でき、製剤の特性を向上させることができる。
また、本開示の経口製剤は、メチル化レスベラトロールをさらに含んでもよい。これにより、本開示の経口製剤は、対象における疾患又は状態(例えば、抗老化)に対する効果を高めることができる。さらに、メチル化レスベラトロールは、通常のレスベラトロールよりも、生体内への吸収性及びバイオアベイラビリティが高く、製剤の特性を向上させることができる。
本開示の経口製剤の製造方法は、NMNと脂質と糖アルコールとを用いてリポソームを調製する工程と、調製したリポソームに結合液を加えて造粒する工程と、造粒した造粒物を乾燥する工程と、を含む。これにより、高濃度のNMNを含有し、高い生体内吸収性等、製剤の特性が向上した経口製剤を好適に製造できる。また、調製したリポソームを造粒することにより、服用性及び生体内での溶解性を向上させることができる。
また、本開示の経口製剤の製造方法において、NMNと糖アルコールとの配合割合は、1:5~1:6であってもよい。これにより、得られた経口製剤の生体内の吸収性が向上し得る。
また、本開示の経口製剤の製造方法において、調製したリポソームにメチル化レスベラトロールを加える工程をさらに含んでもよい。これにより、製剤の特性を向上させた経口製剤を製造できる。
[実施例]
以下に実施例を用いて本開示をより詳細に説明するが、これは本開示の範囲を限定するものではない。

以下に示すように、実施例1-7の経口製剤を製造した。
製剤100重量%あたり、9.09重量%のNMN、50重量%の還元麦芽糖、4重量%のレシチン(大豆由来)、0.1重量%のL-ヒスチジン、0.05重量%のL-ヒスチジン塩酸塩、33~37重量%デキストリンをリポソーム原料としてリポソームを調製した。造粒に用いる材料である、調製したリポソーム及び0.29重量%のメチル化レスベラトロール90%含有インドキノキ抽出物(総量50g)を高速攪拌造粒装置(ハイスピードミキサー、株式会社G-LABO製)に投入し、攪拌羽根(アジテーター)の回転速度250rpm、粉砕羽根(チョッパー)の回転速度2300rpmで5分間攪拌混合した。その後、所定の結合液を原料重量に対して所定の割合となるように添加し、攪拌羽根(アジテーター)の回転速度250rpm、粉砕羽根(チョッパー)の回転速度2300rpmで約15分間攪拌造粒した。箱式通気型式乾燥機(株式会社加藤理機製作所製)を用いて、所定の温度及び時間乾燥した後、場合により20メッシュパスにて篩過した。その後、高速攪拌造粒装置(ハイスピードミキサー、株式会社G-LABO製)を使用して、造粒物に1.5重量%のステアリン酸カルシウム、1重量%の二酸化ケイ素を投入し、攪拌羽根(アジテーター)の回転速度200rpm、粉砕羽根(チョッパー)の回転速度1500rpmで攪拌混合し、目的の経口製剤を得た。

なお、結合液の種類及び造粒に用いる材料に対しての添加量、乾燥温度及び時間、篩過の有無については、以下の表に示す通りとした。また、各実施例の条件による製剤の製造の可否及び製造した物の特徴について以下の表に示した。
上述の表(特に、実施例6)に示すように、造粒条件及び乾燥条件を調整することにより、本開示の所望の経口製剤を製造することができた。

以上、本開示の実施形態について説明したが、本開示は上述の実施形態に限定されることなく、種々変形して実施することができる。
上記実施形態における1つの構成要素が有する機能を複数の構成要素として分散させたり、複数の構成要素が有する機能を1つの構成要素に統合したりしてもよい。また、上記実施形態の構成の一部を省略してもよい。また、上記実施形態の構成の少なくとも一部を、他の上記実施形態の構成に対して付加、置換等してもよい。なお、請求の範囲に記載の文言から特定される技術思想に含まれるあらゆる態様が本開示の実施形態である。

Claims (6)
- 有効成分として、リポソーム化されたニコチンアミドモノヌクレオチドを含有する経口製剤。
- 前記経口製剤における前記ニコチンアミドモノヌクレオチドの含有量は、前記経口製剤100重量%あたり9~20重量%である、請求項1に記載の経口製剤。
- メチル化レスベラトロールをさらに含む、請求項1又は請求項2に記載の経口製剤。
- ニコチンアミドモノヌクレオチドと脂質と糖アルコールとを用いてリポソームを調製する工程と、
調製した前記リポソームに結合液を加えて造粒する工程と、
造粒した造粒物を乾燥する工程と、
を含む、経口製剤の製造方法。 - 前記経口製剤における前記ニコチンアミドモノヌクレオチドと前記糖アルコールとの配合割合は、1:5~1:6である、請求項4に記載の経口製剤の製造方法。
- 前記リポソームにメチル化レスベラトロールを加える工程をさらに含む、請求項4又は請求項5に記載の経口製剤の製造方法。
