Abstract
본 발명은 5-히드록시말톨(5-hydroxymaltol)을 포함하는 항염증용 조성물 및 염증성 질환 예방, 개선 또는 치료용 조성물에 관한 것으로, 본 발명에 따르면, 5-히드록시말톨은 NO 생성, iNOS 발현, ROS 축적을 억제하고, MAPK, NF-κB 및 Nrf-2/HO-1 신호 전달을 조절하여 항염증제로 작용할 수 있는 효과가 있으므로, 이를 염증성 질환 예방, 개선 또는 치료 용도로 유용하게 활용할 수 있다.
Description
본 발명은 5-히드록시말톨(5-hydroxymaltol)을 포함하는 항염증용 조성물 및 염증성 질환 예방, 개선 또는 치료용 조성물에 관한 것이다.
선천적 또는 비특이적인 면역 반응은 체내 해로운 물질과 유기체의 침입에 대한 첫 번째 장벽 역할을 담당하며, 대식세포와 염증성 생체분자를 수반한다. 염증(inflammation)은 모든 인간 조직에서 발생하며 외부 병원체 및 내부 손상에 대한 보호 효과를 나타내는 인간에게 이로운 반응이다. 그러나 과도한 염증은 류마티스 관절염, 죽상 동맥 경화증 및 당뇨병과 같은 만성 질환을 유발한다. 대식세포는 전신 조직에서 발생하여 이물질과 죽은 세포를 탐식할 뿐만 아니라 다양한 감염 신호에 반응한다. 대식세포는 감염에 대한 1차 숙주 방어 역할을 하는 톨-유사(toll-like) 수용체를 가지고 있다. 지질다당류(LPS)는 그람음성균의 세포벽을 구성하는 주요 성분으로 선천면역계에서 대식세포의 강력한 세포 신호를 유도한다. LPS는 톨유사 수용체 4(toll-like receptor 4, TLR 4)에 결합하여 대식세포를 활성화하고, 핵인자 κB(nuclear factor κB, NF-κB) 경로와 3개의 미토겐 활성화 단백질 키나제(mitogen-activated protein kinase, MAPK) 경로, 즉, 세포외 신호 조절 키나제(extracellular signal-regulated kinases, ERK), c-Jun N-말단 키나제(c-Jun N-terminal kinases, JNK)/스트레스 활성화 단백질 키나제(stress-activated protein kinases) 및 p38 MAPK 등을 포함한 여러 세포 내 신호 전달 경로를 유도한다. 대식세포는 LPS 처리시 활성화되어 염증 매개체, 사이토카인 및 활성산소종(reactive oxygen species, ROS)의 과도한 생성을 유발한다. NO 합성효소에 의해 유도된 산화질소(nitric oxide, NO)는 전염증성 매개체로서, 산화 스트레스와 염증을 유발한다. LPS 처리된 대식세포에서는 종양괴사인자-α(tumor necrosis factor-α, TNF-α) 및 인터루킨-1β(interleukin-1β, IL-1β)가 과발현되어 염증성 질환의 발병기전을 촉진시킨다. 헴 옥시게나제 1(heme oxygenase 1, HO-1)은 항염증 및 철 항상성에 중요한 역할을 한다. HO-1의 발현은 염증과 산화 손상에 대한 적응성(후천성) 세포 반응으로 여겨져 왔으며, 염증 및 산화 스트레스에 대한 세포 보호에 중요한 역할을 하는 핵 인자 적혈구계 2 관련 인자 2(nuclear factor erythroid 2-related factor 2, Nrf2)에 의해 매개되는 것으로 알려져 있다.
한편, 비트(Beta vulgaris)는 근공채, 홍채두, 화염채라고도 불리며, 지중해 연안의 남부 유럽과 북아프리카가 원산지이다. 비교적 재배가 쉽고 식물체 전체를 사용할 수 있어 외국에서는 집에서 손쉽게 재배하는 인기작물이다. 국내에서는 주로 경기도 이천, 강원도 평창 및 제주도에서 생산되고 있으며, 붉은색 비트 뿌리는 우리나라 강화 순무와 비슷하다. 비트의 8%는 염소로 구성되어 있는데, 이 염소 성분은 간 정화 작용을 하고 골격 형성 및 유아 발육에 효과가 있다. 또한, 철분과 비타민이 다량 함유되어 있어 적혈구 생성을 돕고, 혈액을 맑게 하여 혈액순환이 원활하도록 도와주어 혈액순환장애를 개선시켜주는 효과가 있으며, 월경불순이나 갱년기 여성에게 좋은 것으로 알려져 있다. 또한, 비트는 위 손상을 막아주고 위 점막을 보호해 줄 뿐만 아니라 노화, 빈혈, 스트레스, 시력 회복, 세포복제기능, 해독작용 등에도 효과가 있다고 알려져 있다.
본 발명자들은 비트 발효물을 제조하여 이로부터 항산화 활성을 나타내는 화합물을 분리 및 정제하였으며, 상기 정제된 화합물에 대하여 NF-κB, MAPK 및 Nrf-2 신호를 조절하여 항염증제로 적용할 수 있는 효과가 있음을 확인하여 본 발명을 완성하였다.
본 발명의 목적은 염증성 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
또한, 본 발명의 목적은 염증성 질환의 예방 또는 개선용 식품 조성물 및 화장료 조성물을 제공하는 것이다.
아울러, 본 발명의 목적은 항염증용 조성물을 제공하는 것이다.
상기 과제를 해결하기 위하여, 본 발명은 5-히드록시말톨(5-hydroxymaltol) 또는 약학적으로 허용 가능한 그의 염 또는 그의 용매화물을 유효성분으로 포함하는 염증성 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 5-히드록시말톨(5-hydroxymaltol) 또는 식품학적으로 허용 가능한 그의 염 또는 그의 용매화물을 유효성분으로 포함하는 염증성 질환의 예방 또는 개선용 식품 조성물을 제공한다.
또한, 본 발명은 5-히드록시말톨(5-hydroxymaltol) 또는 화장품학적으로 허용 가능한 그의 염 또는 그의 용매화물을 유효성분으로 포함하는 염증성 질환의 예방 또는 개선용 화장료 조성물을 제공한다.
아울러, 본 발명은 5-히드록시말톨(5-hydroxymaltol) 또는 그의 염 또는 그의 용매화물을 유효성분으로 포함하는 항염증용 조성물을 제공한다.
마지막으로, 본 발명은 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 염증성 질환 치료방법에서, 상기 염증성 질환은 상기 염증성 질환은 관절염, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 요도염, 방광염, 화상 염증, 피부염, 알레르기, 아토피, 치주염, 치은염, 중이염, 인후염, 관절염, 류마티스 관절염, 건염, 건초염, 퇴행성 신경 염증 및 급성 내지 만성 염증 질환으로 이루어진 군에서 선택되는 어느 하나 이상인 것인, 방법을 제공한다.
본 발명에 따르면, 본 발명의 비트 발효물로부터 분리된 5-히드록시말톨(5-hydroxymaltol)은 대식세포에서 NO 생성, iNOS 발현, ROS 축적, MAPK 신호 전달 활성화 및 NF-κB 신호 전달 활성화를 억제할 뿐만 아니라, Nrf-2/HO-1 신호 전달을 상향조절하는 효과를 나타내므로, 이를 항염증제 및 염증성 질환에 대한 예방, 개선 또는 치료 용도로 유용하게 활용할 수 있다.
도 1은 락토바실러스 람노서스 GG 균주 발효에 의해 제조된 비트 발효물로부터 유효 화합물(5-히드록시말톨)의 분리 및 정제 과정을 나타낸 모식도이다.

본 발명의 5-히드록시말톨(5-hydroxymaltol)은 합성에 의해 제조되는 것일 수도 있으며, 상업적으로 판매하는 제품을 구매하여 사용하는 것도 가능하나, 바람직하게는, 비트 발효물로부터 추출, 분리 및 정제되어 수득되는 것일 수 있다.
일 구현예에서, 본 발명의 비트 발효물은, (1) 7×105 CFU/㎖ 농도의 유산균을 함유하는 배지를 준비하는 단계; 및 (2) 비트 분말을 상기 배지에 2%(w/v) 농도로 혼합하고 25-35℃에서 3-7일 동안 배양하여 발효시킨 후 원심분리하여 상등액을 수득하는 단계;를 포함하는 방법으로 제조되는 것일 수 있다. 상기 배지는 MRS 배지 일 수 있으나, 이에 한정되지 않으며, 상기 배지 외에 유산균 배양에 사용될 수 있는 다양한 상업 배지가 사용될 수 있다. 상기 유산균은 락토바실러스 람노서스(Lactobacillus rhamnosus)일 수 있고, 더 바람직하게는 락토바실러스 람노서스(Lactobacillus rhamnosus) GG일 수 있다.

일 구현예에서, 본 발명의 조성물은 산화질소(NO) 생성 또는 분비를 억제 및 ROS 생성 또는 축적을 억제할 수 있으며, iNOS의 단백질 또는 유전자의 발현 또는 생성을 억제할 수 있다.
일 구현예에서, 본 발명의 조성물은 전염증성 사이토카인의 발현 또는 생성을 억제할 수 있으며, 특히, TNF-α 및 IL-1β의 단백질 또는 유전자의 발현 또는 생성을 억제할 수 있다.
일 구현예에서, 본 발명의 조성물은 NF-κB 신호 전달 경로를 억제할 수 있으며, NF-κB의 활성화 및 핵 국소화를 억제할 수 있고, NF-κB p65의 단백질 또는 유전자의 발현 또는 생성을 억제 및 IκB-α의 단백질 또는 유전자의 발현 또는 생성을 증가시킬 수 있다.

일 구현예에서, 본 발명의 조성물은 5-히드록시말톨을 0.01 내지 5,000 μM의 농도로 포함할 수 있으며, 바람직하게는, 0.1 내지 1,500μM, 보다 바람직하게는 50 내지 1,500μM의 농도로 포함할 수 있고, 50 내지 1,100μM의 농도로 포함하는 것이 더욱 바람직하다.
본 발명에서, 용어 "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 염증성 질환의 발생, 확산 및 재발을 억제 또는 지연시키는 모든 행위를 의미하고, "치료"란 본 발명의 조성물의 투여로 염증성 질환의 증세를 호전시키거나 이롭게 변경하는 모든 행위를 의미한다. 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자라면, 대한의학협회 등에서 제시된 자료를 참조하여 본원의 조성물이 효과가 있는 질환의 정확한 기준을 알고, 개선, 향상 및 치료된 정도를 판단할 수 있을 것이다.
본 발명에서 유효성분과 결합하여 사용된 "치료학적으로 유효한 양"이란 용어는 염증성 질환을 예방 또는 치료하는데 유효한 양을 의미하며, 본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적부위, 환자의 상태 등에 따라 달라질 수 있다. 따라서, 인체에 사용 시 투여량은 안전성 및 효율성을 함께 고려하여 적정량으로 결정되어야 한다. 동물실험을 통해 결정한 유효량으로부터 인간에 사용되는 양을 추정하는 것도 가능하다. 유효한 양의 결정시 고려할 이러한 사항은, 예를 들면 Hardman and Limbird, eds., Goodman and Gilman's The Pharmacological Basis of Therapeutics, 10th ed.(2001), Pergamon Press; 및 E.W. 본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서 사용되는 용어, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 염증성 질환의 종류, 염증성 질환의 발병 원인, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여, 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물은 생물학적 제제에 통상적으로 사용되는 담체, 희석제, 부형제 또는 둘 이상의 이들의 조합을 포함할 수 있다. 본 발명에서 사용되는 용어, "약학적으로 허용가능한"이란 상기 조성물에 노출되는 세포나 인간에게 독성이 없는 특성을 나타내는 것을 의미한다. 상기 담체는 조성물을 생체 내 전달에 적합한 것이면 특별히 제한되지 않으며, 예를 들면, Merck Index, 13th ed., Merck & Co. 일 구현예에서, 상기 약학적 조성물은 경구형 제형, 외용제, 좌제, 멸균 주사용액 및 분무제를 포함하는 군으로부터 선택되는 하나 이상의 제형일 수 있으며, 경구형 또는 주사 제형이 더욱 바람직하다.
본 발명에서 사용되는 용어, "투여"란, 임의의 적절한 방법으로 개체 또는 환자에게 소정의 물질을 제공하는 것을 의미하며, 목적하는 방법에 따라 비 경구 투여(예를 들어 정맥 내, 피하, 복강 내 또는 국소에 주사 제형으로 적용)하거나 경구 투여할 수 있으며, 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설률 및 질환의 중증도 등에 따라 그 범위가 다양하다. 본 발명의 조성물의 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 통상적으로 사용되는 단순 희석제인 물, 액체 파라핀 이외에 다양한 부형제, 예컨대 습윤제, 감미제, 방향제, 보존제 등이 함께 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조 제제, 좌제 등이 포함된다. 본 발명의 약학적 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수도 있다. 바람직한 투여방식 및 제제는 정맥 주사제, 피하 주사제, 피내주사제, 근육 주사제, 점적 주사제 등이다. 주사제는 생리식염액, 링겔액 등의 수성 용제, 식물유, 고급 지방산 에스테르(예, 올레인산에칠 등), 알코올 류(예, 에탄올, 벤질알코올, 프로필렌글리콜, 글리세린 등) 등의 비수성 용제 등을 이용하여 제조할 수 있고, 변질 방지를 위한 안정화제(예, 아스코르빈산, 아황산수소나트륨, 피로아황산나트륨, BHA, 토코페롤, EDTA 등), 유화제, pH 조절을 위한 완충제, 미생물 발육을 저지하기 위한 보존제(예, 질산페닐수은, 치메로살, 염화벤잘코늄, 페놀, 크레솔, 벤질알코올 등) 등의 약학적 담체를 포함할 수 있다.
본 발명에서 사용되는 용어, "개체"란, 상기 염증성 질환이 발병하였거나 발병할 수 있는 인간을 포함한 원숭이, 소, 말, 양, 돼지, 닭, 칠면조, 메추라기, 고양이, 개, 마우스, 쥐, 토끼 또는 기니아 피그를 포함한 모든 동물을 의미하고, 본 발명의 약학적 조성물을 개체에게 투여함으로써 상기 질환들을 효과적으로 예방 또는 치료할 수 있다. 본 발명의 약학적 조성물은 기존의 치료제와 병행하여 투여될 수 있다.
본 발명의 약학적 조성물은 약제학적으로 허용 가능한 첨가제를 더 포함할 수 있으며, 이때 약제학적으로 허용 가능한 첨가제로는 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 사용될 수 있다. 본 발명에 따른 약제학적으로 허용 가능한 첨가제는 상기 조성물에 대해 0.1 중량부 내지 90 중량부 포함되는 것이 바람직하나, 이에 한정되는 것은 아니다.
본 발명의 약학적 조성물은 또한 상기 5-히드록시말톨을 유효성분으로 포함하는 외용제의 제형으로 제공할 수 있다. 본 발명의 염증성 질환 예방 또는 치료용 약학적 조성물을 피부외용제로 사용하는 경우, 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제 (foaming agent), 방향제, 계면활성제, 물, 이온형 유화제, 비이온형 유화제, 충전제, 금속이온봉쇄제, 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 활성제, 친유성 활성제 또는 지질 소낭 등 피부 외용제에 통상적으로 사용되는 임의의 다른 성분과 같은 피부 과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 또한 상기 성분들은 피부 과학 분야에서 일반적으로 사용되는 양으로 도입될 수 있다.
본 발명의 염증성 질환 예방 또는 치료용 약학적 조성물이 피부 외용제로 제공될 경우, 이에 제한되는 것은 아니나, 연고, 패취, 겔, 크림 또는 분무제 등의 제형일 수 있다.

상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨,소르비톨, 에리트리톨 등의 당알콜이다. 상술한 향미제는 천연 향미제 (타우마틴), 스테비아 추출물 (예를 들어 레바우디오시드 A, 글리시르히진 등) 및 합성 향미제 (사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 본 발명의 식품 조성물은 상기 약학적 조성물과 동일한 방식으로 제제화되어 기능성 식품으로 이용하거나, 각종 식품에 첨가할 수 있다. 본 발명의 조성물을 첨가할 수 있는 식품으로는 예를 들어, 음료류, 육류, 초코렛, 식품류, 과자류, 피자, 라면, 기타 면류, 껌류, 사탕류, 아이스크림류, 알코올 음료류, 비타민 복합제 및 건강보조식품류 등이 있다.
또한, 상기 식품 조성물은 유효성분인 추출물 외에 여러 가지 영양제, 비타민, 광물 (전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제 (치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그밖에 본 발명의 식품 조성물은 천연 과일 쥬스 및 과일 쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다.
본 발명의 기능성 식품 조성물은, 정제, 캅셀, 분말, 과립, 액상, 환 등의 형태로 제조 및 가공될 수 있다. 본 발명에서 '건강기능성식품 조성물'이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 말하며, 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다. 본 발명의 건강기능성식품은 통상의 식품 첨가물을 포함할 수 있으며, 식품 첨가물로서의 적합 여부는 다른 규정이 없는 한, 식품의약품안전청에 승인된 식품 첨가물 공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다. 상기 '식품 첨가물 공전'에 수재된 품목으로는 예를 들어, 케톤류, 글리신, 구연산칼슘, 니코틴산, 계피산 등의 화학적 합성물; 감색소, 감초추출물, 결정셀룰로오스, 고량색소, 구아검 등의 천연첨가물; L-글루타민산나트륨 제제, 면류첨가알칼리제, 보존료 제제, 타르색소제제 등의 혼합제제류 등을 들 수 있다. 예를 들어, 정제 형태의 건강기능성식품은 본 발명의 유효성분을 부형제, 결합제, 붕해제 및 다른 첨가제와 혼합한 혼합물을 통상의 방법으로 과립화한 다음, 활택제 등을 넣어 압축성형하거나, 상기 혼합물을 직접 압축 성형할 수 있다. 또한 상기 정제 형태의 건강기능성식품은 필요에 따라 교미제 등을 함유할 수도 있다. 캅셀 형태의 건강기능성식품 중 경질 캅셀제는 통상의 경질 캅셀에 본 발명의 유효성분을 부형제 등의 첨가제와 혼합한 혼합물을 충진하여 제조할 수 있으며, 연질 캅셀제는 본 발명의 유효성분을 부형제 등의 첨가제와 혼합한 혼합물을 젤라틴과 같은 캅셀기제에 충진하여 제조할 수 있다. 상기 연질 캅셀제는 필요에 따라 글리세린 또는 소르비톨 등의 가소제, 착색제, 보존제 등을 함유할 수 있다. 환 형태의 건강기능성식품은 본 발명의 유효성분과 부형제, 결합제, 붕해제 등을 혼합한 혼합물을 기존에 공지된 방법으로 성형하여 조제할 수 있으며, 필요에 따라 백당이나 다른 제피제로 제피할 수 있으며, 또는 전분, 탈크와 같은 물질로 표면을 코팅할 수도 있다. 과립 형태의 건강기능성식품은 본 발명의 유효성분의 부형제, 결합제, 붕해제 등을 혼합한 혼합물을 기존에 공지된 방법으로 입상으로 제조할 수 있으며, 필요에 따라 착향제, 교미제 등을 함유할 수 있다.
본 발명에 따른 건강기능식품 조성물은 당업계에 공지된 통상적인 방법에 따라 다양한 형태로 제조할 수 있다. 일반 식품으로는 이에 한정되지 않지만 음료 (알콜성 음료 포함), 과실 및 그의 가공식품 (예: 과일통조림, 병조림, 잼, 마아말레이드 등), 어류, 육류 및 그 가공식품 (예: 햄, 소시지 콘비이프 등), 빵류 및 면류 (예: 우동, 메밀국수, 라면, 스파게이트, 마카로니 등), 과즙, 각종 드링크, 쿠키, 엿, 유제품(예: 버터, 치이즈 등), 식용식물 유지, 마아가린, 식물성 단백질, 레토르트 식품, 냉동식품, 각종 조미료 (예: 된장, 간장, 소스 등) 등에 본 발명의 5-히드록시말톨을 첨가하여 제조할 수 있다. 또한, 영양보조제로는 이에 한정되지 않지만 캡슐, 타블렛, 환 등에 본 발명의 5-히드록시말톨을 첨가하여 제조할 수 있다. 또한, 건강기능식품으로는 이에 한정되지 않지만 예를 들면, 본 발명의 5-히드록시말톨 자체를 차, 쥬스 및 드링크의 형태로 제조하여 음용 (건강음료)할 수 있도록 액상화, 과립화, 캡슐화 및 분말화하여 섭취할 수 있다. 또한, 본 발명의 5-히드록시말톨을 식품 첨가제의 형태로 사용하기 위해서는 분말 또는 농축액 형태로 제조하여 사용할 수 있다. 또한, 본 발명의 5-히드록시말톨와 염증성 질환 예방 또는 개선 효과가 있다고 알려진 공지의 활성 성분과 함께 혼합하여 조성물의 형태로 제조할 수 있다.
본 발명의 5-히드록시말톨을 건강음료로 이용하는 경우, 상기 건강음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 슈크로스와 같은 디사카라이드; 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당알콜일 수 있다. 감미제는 타우마틴, 스테비아 추출물과 같은 천연 감미제; 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 mL 당 일반적으로 약 0.01 ~ 0.04 g, 바람직하게는 약 0.02 ~ 0.03 g 이다.
또한, 본 발명의 5-히드록시말톨은 염증성 질환 예방 또는 개선용 식품 조성물의 유효성분으로 함유될 수 있는데, 그 양은 염증성 질환 예방 또는 개선 작용을 달성하기에 유효한 양으로 특별히 한정되는 것은 아니나, 전체 조성물 총 중량에 대하여 0.01 내지 100 중량%인 것이 바람직하다. 본 발명의 건강기능식품 조성물은 5-히드록시말톨와 함께 염증성 질환에 효과가 있는 것으로 알려진 다른 활성 성분과 함께 혼합하여 제조될 수 있다.
상기 외에 본 발명의 건강기능식품은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산, 펙트산의 염, 알긴산, 알긴산의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 건강기능식품은 천연 과일주스, 과일주스 음료, 또는 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부당 0.01 ~ 0.1 중량부의 범위에서 선택되는 것이 일반적이다.

화장료 조성물의 외형은 화장품학 또는 피부과학적으로 허용 가능한 매질 또는 기제를 함유한다. 이는 국소적용에 적합한 모든 제형으로, 예를 들면, 용액, 겔, 고체, 반죽 무수 생성물, 수상에 유상을 분산시켜 얻은 에멀젼, 현탁액, 마이크로에멀젼, 마이크로캡슐, 미세과립구 또는, 이온형 (리포좀) 및 비이온형의 소낭 분산제의 형태로, 또는 크림, 스킨, 로션, 파우더, 연고, 스프레이 또는 콘실 스틱의 형태로 제공될 수 있다. 이들 조성물은 당해 분야의 통상적인 방법에 따라 제조될 수 있다. 본 발명에 따른 조성물은 또한 포말 (foam)의 형태로 또는 압축된 추진제를 더 함유한 에어로졸 조성물의 형태로도 사용될 수 있다.
본 발명의 일 실시예에 따른 상기 화장료 조성물은 그 제형에 있어서 특별히 한정되는 바가 없으며, 예를 들면, 유연화장수, 수렴화장수, 영양화장수, 영양크림, 마사지크림, 에센스, 아이크림, 아이에센스, 클렌징크림, 클렌징폼, 클렌징워터, 팩, 파우더, 바디로션, 바디크림, 바디오일 및 바디에센스 등의 화장품으로 제형화될 수 있다.
본 발명의 화장료 조성물의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물섬유, 식물섬유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 화장료 조성물의 제형이 용액 또는 유탁액의 경우에는 담체 성분으로서 용매, 용매화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 화장료 조성물의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 계면-활성제 함유 클린징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 리놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
본 발명의 화장료 조성물은 스킨, 로션, 크림, 에센스, 팩, 파운데이션, 색조화장품, 선크림, 투웨이케이크, 페이스파우더, 콤팩트, 메이크업베이스, 스킨커버, 아이쉐도우, 립스틱, 립글로스, 립픽스, 아이브로우 펜슬, 화장수 등의 화장품 및 샴푸, 비누 등의 세정제에 적용될 수 있다.
본 발명의 일 실시예에 따른 화장료 조성물에는 상기 5-히드록시말톨 이외에 기능성 첨가물 및 일반적인 화장료 조성물에 포함되는 성분이 추가로 포함될 수 있다. 상기 기능성 첨가물로는 수용성 비타민, 유용성 비타민, 고분자 펩티드, 고분자 다당, 스핑고 지질 및 해초 엑기스로 이루어진 군에서 선택된 성분을 포함할 수 있다.
본 발명의 화장료 조성물에는 또한, 상기 기능성 첨가물과 더불어 필요에 따라 일반적인 화장료 조성물에 포함되는 성분을 배합해도 된다. 이외에 포함되는 배합 성분으로서는 유지 성분, 보습제, 에몰리엔트제, 계면 활성제, 유기 및 무기 안료, 유기 분체, 자외선 흡수제, 방부제, 살균제, 산화 방지제, 식물 추출물, pH 조정제, 알콜, 색소, 향료, 혈행 촉진제, 냉감제, 제한 (制汗)제, 정제수 등을 들 수 있다.
일 측면에서, 본 발명은 상기 화학식 1로 표시되는 5-히드록시말톨(5-hydroxymaltol) 또는 약학적으로 허용 가능한 그의 염 또는 그의 용매화물을 유효성분으로 포함하는 항염증용 조성물에 관한 것이다.
일 구현예에서, 상기 항염증용 조성물은 약학적 조성물, 화장료 조성물 및 식품 조성물로 이루어진 군으로부터 선택되는 어느 하나 이상인 것일 수 있다.
본 발명에서 "항염증"이란, "염증 억제 또는 개선"과 혼용될 수 있으며, 염증 반응이 완화되는 모든 작용을 의미할 수 있다.
일 측면에서, 본 발명은 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 염증성 질환 치료방법에서, 상기 염증성 질환은 상기 염증성 질환은 관절염, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 요도염, 방광염, 화상 염증, 피부염, 알레르기, 아토피, 치주염, 치은염, 중이염, 인후염, 관절염, 류마티스 관절염, 건염, 건초염, 퇴행성 신경 염증 및 급성 내지 만성 염증 질환으로 이루어진 군에서 선택되는 어느 하나 이상인 것인, 방법에 관한 것이다.
하기의 실시예를 통하여 본 발명을 보다 상세하게 설명한다. 그러나 하기 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐 이에 의해 본 발명이 한정되는 것은 아니다.
실시예 1. 비트 발효물로부터 5-히드록시말톨의 분리 및 정제
1-1. 비트 발효물의 제조
제주산 비트를 입수하여 증류수로 세척하고 블렌더(신일, 서울, 한국)로 분쇄하여 비트 분말을 준비하였다. 발효를 위한 균주, 락토바실러스 람노서스 GG(Lactobacillus rhamnosus GG, KCTC 3237)는 한국생명공학연구원 생물자원센터(KCTC, Seoul, Korea)에서 구입하여 사용하였다. 보관용 스톡(stock cultures)은 20%(v/v) 글리세롤이 포함된 MRS(Man, Rogosa, and Sharpe) 배지(Difco, Detroit, USA)에서 -80℃에서 보관하였다.
MRS 배지에 7×105 CFU/㎖ 농도의 락토바실러스 람노서스 GG 균주를 접종하여 180rpm, 30℃의 조건에서 1일 동안 사전배양하였다. 상기 비트 분말(1kg)을 락토바실러스 람노서스 GG 균주가 함유된 MRS 배지(50L)와 함께 180rpm, 30℃의 조건에서 5일 동안 배양하고, 발효된 배지를 원심분리하여 상등액, 즉, 비트 발효물(50L)을 수득하였다.
비트 발효물로부터 유효 화합물을 분리하기 위한 분획 및 정제 과정을 도 1에 나타내었으며, 구체적인 방법은 하기에 기재된 바와 같다.
한편, 상기 발효 전의 비트(0 day) 및 발효 후의 비트(5 day)에 대하여 HPLC 분석을 실시한 결과, 약 7분에서 비발효 비트의 HPLC 스펙트럼에서는 보이지 않던 피크가 비트 발효물의 HPLC 스펙트럼에서 나타나, 발효에 의해 새로운 물질이 생성되었음을 확인하였다(도 2A).
1-2. 비트 발효물의 EA 추출물 제조
상기 방법으로 수득된 50L의 비트 발효물을 50L의 에틸 아세테이트(EA)를 혼합하여 추출함으로써, 비트 발효물의 EA 추출물을 수득하였다. EA 추출물을 농축한 후, 메탄올로 가용화시켜 하기 실시예에서 사용하였다.
1-3. ODS 크로마토그래피를 이용한 1차 분획
메탄올로 가용화된 EA 추출물을 RP-18 수동 컬럼(ODS C-18 오픈 컬럼, 10×14cm)에 로딩하고, ODS(octadecyl silica) 크로마토그래피(chromatography)를 실시하여 0% 메탄올(물), 10% 메탄올 및 20% 메탄올로 분획화하였다. 상기 3개의 1차 분획물에 대하여 DPPH(diphenyl-picrylhydrazyl) 소거 활성 분석을 사용하여 항산화 활성을 조사한 결과, 0% 메탄올 분획물이 항산화 효과를 보여 2차 분획 대상 시료로 사용하였다.
1-4. 실리카겔 크로마토그래피를 이용한 2차 분획
상기 0% 메탄올 분획물을 농축한 다음, 실리카겔 60(silica gel 60, 0.035-0.2 mm) 레진(MERCK, Darmstadt, Germany)을 충진한 컬럼(30×450mm)에 로딩하고 클로로포름:메탄올(10:1, v/v)을 용출용매로 사용하여 실리카겔 크로마토그래피를 통해 4개의 분획물(1, 2, 3 및 4번 분획물)을 수득하였다. 4개의 분획물에 대하여 항산화 활성을 조사한 결과, 2번 분획물이 항산화 효과를 보였고, 이를 농축하여 3차 분획 대상 시료로 사용하였다.
1-5. 세파덱스 G-10을 이용한 3차 분획
상기 2번 분획물을 세파덱스 G-10(Sephadex G-10TM, 40-120 μm) 분말 (Cytiva, Marlborough, MA, USA) 겔을 충진한 컬럼(3 × 45cm)에 로딩하고 물을 용출용매로 사용하여 세파덱스 겔 크로마토그래피를 수행하였다. 이로부터 2개의 분획(2-1번 분획물 및 2-2번 분획물)을 분리하고, 항산화 활성을 조사한 결과, 두 번째 분획(2-2번 분획물)이 항산화 효과를 보였으며, 이를 농축하여 4차 분획 대상 시료로 사용하였다.
1-6. ODS C18 prep TLC를 이용한 4차 분획
상기 2-2번 분획물을 ODS C-18이 코팅된 분취용 TLC 유리 플레이트(Silica gel 60 RP-18 F254 plate, 10×20 cm; Sigma, Burlington, IA, USA)에 점적하고 TLC 챔버에서 메탄올:물(1:1, v/v)을 전개용매로 이용하여 전개시켜 밴드 형태로 물질이 분리되도록 하였다. 전개가 완료된 후 유리 플레이트를 챔버에서 꺼내고 UV 검출기(UV254nm)에서 형광을 분석한 결과, 물질이 2개의 밴드(1번 물질 및 2번 물질)로 분획되었음을 확인하였다(도 3). 분획된 밴드 부분을 포함하는 각 겔을 칼을 사용하여 유리 플레이트에서 긁어서 분리하고, 50mL 원뿔형 튜브에 각각 수집하였다. 이어서, 분리된 물질이 함유된 각 겔에 메탄올 20mL를 첨가하고, 혼합물을 와동(vortex)시키고 원심분리하여 1번 물질 및 2번 물질을 정제하였다. 각 물질의 항산화 활성을 조사한 결과, 1번 물질이 우수한 항산화 효과를 보여, 해당 물질을 최종 분리 및 정제하기 위해 HPLC를 수행하였다.
1-7. Prep HPLC를 이용한 유효 화합물 정제
상기 ODS C18 prep TLC를 통해 얻어진 1번 물질을 분취용 고성능 액체 크로마토그래피(preparatory high-performance liquid chromatography; prep HPLC)에 주입하였다. HPLC 분석은 Shimadzu HPLC(Shimadzu, Tokyo, Japan)를 사용하여 수행하였다. HPLC 분리는 RP-18(10×250mm) 컬럼을 사용하여 수행하였으며, 분리된 물질은 0.2μm 필터를 통해 체질하였다. 이때, 주입 부피는 0.5 mL, 유속은 2.5 mL/min, 220 및 254 nm에서 컬럼 온도는 실온이었으며, 이동상은 물(용매 A)과 아세토니트릴(용매 B)로 구성되었고, 구배 용출의 경우, 용매 B는 초기에 0%로 설정되었고, 20분에 5%로 증가 및 30분에 100%로 증가되었다. 최종 분리 및 정제된 유효 화합물의 피크는 36분에 표시되었다(도 2B).
1-8. 유효 화합물의 구조 분석
최종적으로 분리 및 정제된 유효 화합물의 구조를 분석하기 위해 NMR(nuclear magnetic resonance) 분석을 통해 구조를 동정하고, ESI-Mass spectrometer로 분자량을 측정하여 분자량이 143g/mol임을 확인하였다. 이를 통해, 본 발명의 비트 발효물에서 정제된 물질이 5-히드록시말톨(5-hydoxymaltol)인 것으로 확인되었다(도 4A).
1-9. 5-히드록시말톨의 항산화 효과 및 대식세포 증식에 대한 영향 분석
5-히드록시말톨의 항산화 효과를 DPPH 소거활성 분석을 통해 조사하였다. 먼저, 96-웰 플레이트의 각 웰에 5-히드록시말톨(0, 100, 200, 400, 500μM) 또는 10mM N-아세틸-L-시스테인(NAC)을 분주하고, 200μM DPPH 용액(4mM 스톡, 메탄올)을 혼합하여 30분 동안 배양하였다. OD517에서의 흡광도를 VersaMax ELISA(효소 결합 면역흡착 분석) 판독기(Molecular Devices, San Jose, CA, USA)를 사용하여 측정하였고, DPPH 소거 활성은 하기 수학식 1을 이용하여 계산하였다:

(여기서 Sample은 시료의 흡광도, Control은 DPPH만 처리된 대조군의 흡광도, Blank는 DPPH를 비처리한 시료의 흡광도이다).
DPPH 소거 활성 분석 결과, 5-히드록시말톨은 농도 의존적으로 강력한 항산화 효과를 보였으며, 특히, 500μM의 고농도에서 42.5%로 증가하여 양성 대조군인 NAC와 유사한 항산화 효과를 나타냈다(도 4B).
또한, 대식세포주 RAW 264.7의 세포 증식에 대한 5-히드록시말톨이 미치는 영향을 확인하기 위하여, RAW 264.7 대식세포를 한국세포주은행(서울, 한국)에서 입수하여 MTS 분석을 수행하였다. 대식세포는 10% 소태아혈청(Hyclone, Logan, UT, USA) 및 1% 페니실린/스트렙토마이신(Hyclone)이 보충된 고-포도당 DMEM(Dulbecco’s modified Eagle’s medium)에서 5% CO2 대기 및 37℃의 조건하에 배양되었다. RAW 264.7 세포(3×105 cells/mL)를 96웰 플레이트에 분주하고 1일 동안 배양한 다음, 5-히드록시말톨 농도를 증가시키면서(0, 50, 100, 200, 400, 500, 750 및 1000μM) 1일 동안 처리하였다. 제조사의 프로토콜에 따라 CellTiter 96 AQueous One Solution(Promega, Madison, WI, USA)을 사용하여, 상기 5-히드록시말톨을 처리하여 배양한 배양액과 1:5(용액:배양액, v/v)로 혼합한 후, 100μL의 혼합물을 96웰 플레이트에 넣고 37℃에서 2시간 동안 반응시켰다. 흡광도는 OD490에서 VersaMax ELISA 판독기를 사용하여 측정하였으며, 이로부터 세포 생존율을 조사하였다.
RAW 264.7 대식세포를 여러 농도의 5-히드록시말톨(최대 1000μM)과 함께 배양한 결과, 5-히드록시말톨을 1000 μM로 고농도로 처리하여도 세포 증식에 어떠한 영향도 미치지 않았다(도 4C).
실시예 2. 5-히드록시말톨의 NO 생성 및 iNOS 발현 억제 효과
2-1. NO 생성 억제 효과 분석
산화질소(NO)는 LPS로 자극된 대식세포에서 생산되는 생물학적 매개체이다. RAW 264.7 세포에서 LPS(lipopolysaccharide from E. coli O111:B4; InvivoGen, San Diego, CA, USA) 로 유도된 NO 생성에 대한 5-히드록시말톨의 억제 효과를 평가하기 위해, LPS의 유무에 관계없이 5-히드록시말톨(200, 500 및 1000μM)로 RAW 264.7 대식세포에 24시간 동안 처리하여 NO 분석(NO assay)을 수행하였다.
구체적으로, RAW 264.7 세포(3×105 cells/mL)를 6-웰 플레이트에서 배양하고 1일 동안 인큐베이션하였다. 세포를 LPS 처리 없이 5-히드록시말톨(0 및 500μM)을 처리하거나 LPS(1μg/mL)를 처리하기 전 다양한 농도의 5-히드록시말톨(0, 200, 500 및 1000μM)을 24시간 동안 처리하였다. NO Plus Detection kit(iNtRON Biotechnology, 경기도, 한국)를 사용하여 상층 배양 배지 내의 NO 생성량을 측정하였다. 96-웰 플레이트의 각 웰에 100 μL의 배양 배지 또는 아질산염 표준물질을 분주하고, 50 μL의 N1 완충액(완충액 중 설파닐아미드)을 첨가하여 예비 반응을 유도하였다. 그 다음 96-웰 플레이트를 20분 동안 인큐베이션하고 혼합물을 50 μL의 N2 완충액(완충액 중 나프틸-에틸렌디아민)과 반응시켰다. 혼합물을 10분 동안 인큐베이션한 후, VersaMax ELISA 판독기를 사용하여 OD560 nm에서의 흡광도 값으로 NO 생성량을 측정하였다.
그 결과, LPS 자극은 NO 농도의 51배 증가를 유도한 반면, 5-히드록시말톨 200, 500 및 1000μM 처리 세포는 LPS 처리 대조군과 비교하여 각각 34%, 41% 및 54%로 감소를 보였다(도 5A). 따라서, LPS에 의해 NO 생성이 유도되며, 5-히드록시말톨이 LPS에 의해 유도된 NO 생성을 감소시키는 것으로 나타났다.
2-2. iNOS 단백질 발현 억제 효과 분석
웨스턴 블롯(Western blot) 분석을 이용하여 유도성 NO 합성효소(inducible NO synthase, iNOS)의 단백질 수준을 평가하였다. 5-히드록시말톨을 처리하거나 비처리한 대식세포에 LPS(1㎍/mL)를 24시간 동안 처리한 다음 단백질 용해물을 분리하여 웨스턴 블롯 분석을 수행하였다.
또한, 5-히드록시말톨을 처리하거나 비처리한 다음 LPS(1μg/mL)로 1일 동안 자극시킨 RAW 264.7 대식세포로부터, 총 RNA(total RNA)를 분리하고, 역전사 정량적 중합효소 연쇄 반응(RT-qPCR)을 통해 iNOS의 mRNA 수준을 조사하였다. 제조업체의 프로토콜에 따라 TaKaRa MiniBEST RNA Extraction Kit(TaKaRa, Kyoto, Japan)를 사용하여 총 RNA를 정제하였고, RNA-direct™ SYBR® Green Realtime qPCR Master Mix(TOYOBO, Osaka, Japan)를 사용하여 RT-qPCR을 수행하였다. RT-qPCR 혼합물에는 10μL의 RNA-direct SYBR Green Realtime qPCR 마스터 믹스, 1μL의 50mM Mn(OAc)2, 2μL의 템플릿 RNA(100ng/μL), 2μL의 특이적 프라이머-F(10ng/μL), 2μL의 특이적 프라이머-R(10ng/μL) 및 3μL의 뉴클레아제 프리 H2O를 함유하도록 하였다. 상기 RT-PCR에 사용된 iNOS에 대한 특이적 프라이머는 바이오니아(대전, 대한민국)에서 구입하였다.
그 결과, 도 5B에 나타난 바와 같이, LPS로 자극된 RAW 264.7 세포는 iNOS 단백질 발현이 증가된 반면, 5-hydroxylmaltol은 이를 고도로 억제했다. 또한, RAW 264.7 세포에서의 iNOS의 전사 수준을 분석한 결과, LPS는 iNOS 전사체 수준을 증가시킨 반면, 5-hydroxylmaltol은 증가된 iNOS 전사체 수준을 감소시켰다(도 5B). LPS 처리는 iNOS의 메신저 RNA( messenger RNA, mRNA) 및 단백질 수준을 각각 21배 및 14배 증가시킨 반면, 5-히드록시말톨(1000μM)은 iNOS의 mRNA 및 단백질 수준을 LPS 처리 대조군 대비 각각 46% 및 61% 감소시켰다.
따라서, 5-히드록시말톨이 iNOS의 하향 조절을 통해 NO 생성을 억제할 수 있음을 확인하였다.
실시예 3. 5-히드록시말톨의 TNF-α 및 IL-1β 생산 및 발현 억제 효과
LPS로 활성화된 대식세포는 전염증성 사이토카인과 성장 인자를 생성한다. ELISA(enzyme-linked immunosorbent assay) 및 RT-qPCR 분석을 이용하여 LPS로 자극된 RAW 264.7 세포에서 TNF-α 및 IL-1β의 증가된 단백질 및 전사체 수준에 대한 5-히드록시말톨의 효과를 평가하였다.
먼저, ELISA 분석을 위하여, RAW 264.7 대식세포를 6웰 플레이트에서 배양하였다. 부착된 세포를 24시간 동안 다양한 농도의 5-히드록시말톨(0, 200, 500 및 1000μM)로 전처리하고 24시간 동안 LPS(1μg/mL)로 자극하였다. TNF-α 및 IL-1β의 측정은 상청액에서 평가하였다. 제조사의 프로토콜에 따라 IL-1β Mouse ELISA Kit(Invitrogen, Carlsbad, CA, USA)를 이용하여 IL-1β의 양을 측정 및 정량화하였고, ELISA MAX™ kit(BioLegend, 샌디에고, 캘리포니아, 미국)을 이용하여 TNF-α의 양을 측정 및 정량화하였다.
한편, RT-qPCR 분석을 위하여, RAW 264.7 세포를 5-히드록시말톨(1000μM)이 포함된 6웰 플레이트에서 배양하고 LPS(1μg/mL)로 1일 동안 처리한 다음, 제조업체의 프로토콜에 따라 TaKaRa MiniBEST RNA Extraction Kit(TaKaRa, Kyoto, Japan)를 사용하여 총 RNA를 정제하였다. 실시예 2-2와 동일한 방법으로 RT-qPCR 분석을 수행하였다. 상기 RT-PCR에 사용된 TNF-α 및 IL-1β에 대한 특이적 프라이머는 바이오니아(대전, 대한민국)에서 구입하였다.
분석 결과, RAW 264.7 세포에서 LPS 처리는 IL-1β의 mRNA 및 단백질 수준을 각각 101배 및 1.6배 증가시킨 반면, 5-히드록시말톨(1000μM)은 LPS 처리 대조군과 비교하여 각각 61% 및 23% 하향 조절하였다(도 6A). LPS 처리는 TNF-α의 mRNA 및 단백질 수준을 무처리 대조군 대비 각각 5.9배 및 11.9배 증가시킨 반면, 5-히드록시말톨(1000μM)은 LPS 처리 대조군과 비교하여 TNF-α의 mRNA 및 단백질 수준을 각각 42% 및 43% 하향 조절하였다(도 6B). 이러한 결과는, 5-히드록시말톨이 IL-1β 및 TNF-α 유전자의 하향 조절을 통해 IL-1β 및 TNF-α 사이토카인 생성을 억제한다는 것을 의미한다.
또한, LPS 처리는 RAW 264.7 세포에서 TNF-α 및 IL-1β의 생성을 유의하게 증가시킨 반면, 5-히드록시말톨 처리는 농도 의존적 방식으로 LPS-자극된 TNF-α 및 IL-1β 생성을 감소시켰고, LPS 처리는 TNF-α 및 IL-1β 유전자의 전사체 수준을 증가시킨 반면, 5-히드록시말톨 전처리는 LPS로 자극된 TNF-α 및 IL-1β 유전자 발현을 하향 조절하였다(도 6A, 6B).
이러한 결과로부터, 5-히드록시말톨이 LPS로 자극된 사이토카인 생성을 억제함으로써 RAW 264.7 세포의 면역 반응을 감소시키는 것을 확인하였다.
실시예 4. 5-히드록시말톨의 LPS-자극 NF-κB 활성화 억제 효과
NF-κB의 핵 전위는 대식세포에서 염증 관련 유전자의 발현을 증가시키는 것으로 알려져 있다. 이에, 5-히드록시말톨이 NF-κB의 핵 전위를 억제할 수 있을지에 대하여 면역 블롯 분석(immuno blot analysis)을 수행하였다.
RAW 264.7 대식세포를 1시간 동안 5-히드록시말톨로 전처리하고 30분 동안 LPS(1μg/mL)로 자극했다. 대식세포 세포를 수거하고 방사성면역침전 분석 완충액(Thermo Fisher Scientific, Waltham, MA, USA)을 사용하여 용해시켰다. 각 핵 및 세포질 분획 샘플을 소듐 도데실 설페이트-폴리아크릴아미드 겔 전기영동으로 분리하고, Immobilin-FL 폴리비닐리덴 플루오라이드(PVDF) 멤브레인(Millipore, Burlington, MA, USA)으로 전기 이동(electro-transferred)시켰다. Odyssey® 블로킹 완충액(LICOR, Lincoln, NE, USA)으로 1시간 동안 차단한 후, 블롯을 4℃에서 1차 항체와 함께 밤새 인큐베이션하였다. 블롯을 세척한 후, 0.2% Tween-20/0.01% 나트륨 도데실 설페이트를 함유하는 Odyssey 블로킹 완충액을 사용하여, 희석된 IRDye 680- 및 IRDye 800-표지된 이차 항체와 함께 60분 동안 인큐베이션하였다. 단백질 밴드는 Odyssey CLx 이미징 머신(LI-COR)으로 검출하였다. p65에 대한 1차 항체는 Cell Signaling Technology(Beverly, MA, USA)에서 입수하였고, 라민 B(lamin B) 및 β-액틴(β-actin)의 항체는 Santa Cruz Biotechnology, Inc(Dallas, TX, USA)에서 구입하여 사용하였다. 라민 B 및 β-액틴은 핵 및 세포질 분획에 대한 내부 대조군으로 사용하여, 상대적 발현량을 정량화하였다.
LPS 처리된 세포에서 NF-κB의 핵 전위는 대조군에 비해 8.6배 증가한 반면, 5-히드록시말톨은 LPS 처리된 대조군에 비해 핵 p65 단백질을 73% 하향 조절하였다. 또한, NF-κB의 조절 인자인 세포질(cytosolic) 억제제 κB(inhibitor κB, IκB)를 조사한 결과, 5-히드록시말톨 전처리는 LPS 처리 세포에서 IκB-α의 하향 조절을 차단하여, LPS 처리된 대조군과 비교하여 세포질 IκB가 2배 증가하였다. 이러한 결과는 LPS 처리가 IκB-α의 하향 조절과 함께 NF-κB(p65)의 핵 전위를 유도하는 반면, 5-히드록시말톨 전처리는 LPS 처리 세포에서 NF-κB(p65)의 핵 전위를 억제하고 IκB-α의 하향 조절을 차단할 수 있음을 나타낸다(도 7).
따라서, 5-히드록시말톨이 RAW 264.7 대식세포에서 LPS로 유도된 NF-κB 활성화의 억제제로 작용할 수 있음을 확인하였다.
실시예 5. 5-히드록시말톨의 LPS-자극 MAPK 활성화 억제 효과
MAPK는 세포 기능, 증식, 유전자 발현, 세포 생존 및 세포 사멸을 조절하는 것으로 알려져 있다. 5-히드록시말톨이 RAW 264.7 세포에서 LPS로 자극된 MAPK의 인산화 및 활성화에 미치는 영향을 면역 블롯 분석을 사용하여 평가하였다.
RAW 264.7 세포를 1시간 동안 1000μM의 5-히드록시말톨과 함께 배양한 후 LPS(1μg/mL)로 30분 동안 처리하였다. 총 단백질 추출물은 소듐 도데실 설페이트-폴리아크릴아미드 겔 전기영동을 사용하여 분리하고, 상기 실시예 4와 동일한 방법으로 면역 블롯 분석을 수행하였다. pp38, JNK, pJNK, ERK, 및 pERK에 대한 1차 항체는 Cell Signaling Technology(Beverly, MA, USA)에서 입수하였고, p38 및 β-액틴의 항체는 Santa Cruz Biotechnology, Inc(Dallas, TX, USA)에서 구입하여 사용하였다. β-액틴을 전체 단백질에 대한 내부 대조군으로 사용하여, 상대적 발현량을 정량화하였다.
LPS 처리가 대조군과 비교하여 ERK, p38 및 JNK의 인산화 수준을 각각 4.8배, 3배 및 1.9배로 유도한 반면, 5-히드록시말톨은 LPS 처리 대조군과 비교하여 ERK, p38 및 JNK의 인산화 수준을 각각 69%, 73% 및 59% 하향 조절하는 것으로 나타났다(도 8).
따라서, 5-히드록시말톨이 ERK, p38 및 JNK의 LPS로 유도된 인산화를 억제하여, MAPK 신호를 억제함으로써 LPS 유도 RAW 264.7 세포의 염증 반응을 억제할 수 있음을 확인하였다.
실시예 6. 5-히드록시말톨의 LPS-유도 ROS 생산 억제 효과
LPS로 자극된 대식세포는 활성산소종(reactive oxygen species, ROS) 축적과 산화 스트레스를 유도한다. 이에, LPS로 유도된 RAW 264.7 세포에서 세포 ROS 축적에 대한 5-히드록시말톨의 효과를 CellROX Green Dye를 사용하여 분석하였다.
ROS는 제조업체의 프로토콜에 따라 CellROX Green 프로브(Life Technologies, Carlsbad, CA, USA)를 사용하여 측정되었다. RAW 264.7 세포(2×106 셀/10 mL)를 플레이트에서 배양하고 1일 동안 인큐베이션 하였다. 대식세포를 5-히드록시말톨(1000 μM) 또는 NAC(10 mM)로 1시간 동안 전처리하고 LPS(1μg/mL)로 30분간 처리하였다. 그 다음 CellROX Green Dye를 세포에 처리하고, 세포를 37℃에서 10분 동안 배양한 후, 배지를 버리고 세포를 1×인산완충식염수로 세척하였다. 염색된 대식세포는 형광현미경(Lionheart, Biotek, VT, USA)을 사용하여 10× 배율에서 시각화하였다. 형광 신호 수준은 대조군(무처리군)을 기준으로 상대적 수준으로 정량화하였다.
도 9는 LPS 처리가 CellROX Green의 세포 밀도를 증가시키는 반면, 5-히드록시말톨과 N-acetyl cysteine(NAC)은 LPS에 의해 유발된 세포의 ROS 함량 증가, 즉, ROS 생산을 감소시켰음을 보여준다. 형광 강도는 무처리 대조군에 비해 LPS 처리 세포에서 13.6배 증가하고, 5-히드록시말톨 처리 세포에서 LPS 처리 대조군에 비해 87% 감소하였다.
따라서, 5-히드록시말톨이 RAW 264.7 세포에서 강력한 ROS 소거 효과를 나타냄을 확인하였다.
실시예 7. 5-히드록시말톨의 HO-1 및 Nrf2의 단백질 발현 증가 효과
Nrf2 및 HO-1 신호 전달 경로는 동물 모델에서 산화 스트레스를 감소시키는 항산화 시스템이다. 본 발명자들은 5-히드록시말톨이 Nrf2 및 HO-1 신호 전달 경로에 관여하는지 여부를 조사하였다.
먼저, RAW 264.7 대식세포를 1000μM의 5-히드록시말톨과 함께 배양하고, 총 단백질을 10% 나트륨 도데실 설페이트-폴리아크릴아미드 겔 전기영동으로 분리한 다음 Nrf2 및 HO-1 항체를 사용하여 웨스턴 블롯을 통해 단백질 발현 수준을 분석하였다. β-액틴은 내부 대조군으로 사용되었으며, 이로부터 상대적 발현량을 정량화하였다.
또한, RAW 264.7 대식세포를 500 μM의 5-히드록시말톨로 12시간 동안 처리한 다음, 소듐 도데실 설페이트-폴리아크릴아미드 겔 전기영동을 사용하여 핵 및 세포질 분획을 분리한 후, Nrf2 항체를 사용하여 상기 실시예 4와 동일한 방법으로 면역 블롯 분석을 수행하였다. 라민 B 및 β-액틴을 핵 및 세포질 분획에 대한 내부 대조군으로 사용하여, 상대적 발현 수준을 정량화하였다.
Nrf2 및 HO-1에 대한 1차 항체는 Cell Signaling Technology(Beverly, MA, USA)에서 입수하였고, 라민 B 및 β-액틴의 항체는 Santa Cruz Biotechnology, Inc(Dallas, TX, USA)에서 구입하여 사용하였다.
웨스턴 블롯 분석 결과, 5-히드록시말톨 처리는 HO-1과 Nrf2의 단백질 발현을 12시간까지 각각 3.9배와 3.8배 증가시켰고, 5-히드록시말톨 처리가 시간 의존적 방식으로 Nrf2와 HO-1의 단백질 수준을 증가시키는 것으로 나타났다(도 10A). RAW 264.7 세포에서 Nrf2 단백질에 대한 세포질 및 핵 분획을 면역 블롯 분석한 결과, 5-히드록시말톨의 처리에 의해 핵 Nrf2 단백질 수준이 1.9배까지 유의하게 증가되는 것으로 나타났다(도 10B).
따라서, 5-히드록시말톨이 Nrf2/HO-1 신호 전달 경로를 증가시키는 효과가 있음을 확인하였다.
종합적으로, 본 발명의 상기 실시예들을 통하여, 본 발명의 비트 발효물로부터 분리된 5-히드록시말톨이 NO 생성 및 iNOS 발현을 억제하며, ROS 축적, MAPK 신호 전달 활성화 및 NF-κB 핵 전위를 억제할 뿐만 아니라, Nrf-2/HO-1 신호 전달을 상향 조절시킴으로써 RAW 264.7 세포에 항염증 효과를 나타내므로(도 11), 이를 통해 5-히드록시말톨이 항염증 효과를 발휘하여 염증성 질환을 치료하는데 유용하게 활용될 수 있음을 확인하였다.

- 제1항에 있어서, 산화질소(NO) 생성 또는 분비를 억제 및 ROS 생성 또는 축적을 억제하는, 약학적 조성물.
- 제1항에 있어서, 전염증성 사이토카인의 발현 또는 생성을 억제하는, 약학적 조성물.
- 제1항에 있어서, NF-κB 신호 전달 경로를 억제하는, 약학적 조성물.
- 제1항에 있어서, MAPK 신호 전달 경로를 억제하는, 약학적 조성물.
- 제1항에 있어서, Nrf-2/HO-1 신호 전달을 증가시키는, 약학적 조성물.
- 제1항에 있어서, 상기 염증성 질환은 상기 염증성 질환은 관절염, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 요도염, 방광염, 화상 염증, 피부염, 알레르기, 아토피, 치주염, 치은염, 중이염, 인후염, 관절염, 류마티스 관절염, 건염, 건초염, 퇴행성 신경 염증 및 급성 내지 만성 염증 질환으로 이루어진 군에서 선택되는 어느 하나 이상인 것인, 약학적 조성물.
- 제1항의 약학적 조성물을 개체에 투여하는 단계를 포함하는 염증성 질환 치료방법에서,상기 염증성 질환은 상기 염증성 질환은 관절염, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 요도염, 방광염, 화상 염증, 피부염, 알레르기, 아토피, 치주염, 치은염, 중이염, 인후염, 관절염, 류마티스 관절염, 건염, 건초염, 퇴행성 신경 염증 및 급성 내지 만성 염증 질환으로 이루어진 군에서 선택되는 어느 하나 이상인 것인, 방법.
