Abstract
본 발명은 실리콘 고분자 화합물, 이의 제조방법, 이를 포함하는 경피 전달용 조성물 및 이를 포함하는 항암용 조성물에 관한 것으로, 고리형 실란을 단량체로 활용하고, 개시제의 종류, 단량체 대비 개시제의 비율, 중합 온도 등의 중합 조건을 조절함으로써, 합성된 실리콘 고분자 화합물의 분자량, 점도, 접촉각, 함수율, 약물 봉입률 및 방출 거동을 조절할 수 있는 바, 약물 및 다양한 활성 물질을 탑재한 경피 전달 시스템으로 활용할 수 있다. 또한, 본 발명의 실리콘 고분자 화합물에 항암제를 봉입함으로써, 항암제의 효과를 증가시킬 수 있는 바, 항암 용도로도 활용될 수 있다.
Description
본 발명은 실리콘 고분자 화합물, 이를 포함하는 경피 전달 시스템 및 이를 포함하는 항암용 조성물에 관한 것이다.
약물 전달 시스템(Drug Delivery System, DDS)이란 기존 의약품의 부작용을 최소화하고 효능을 극대화하기 위하여 필요한 양의 약물을 효율적으로 전달할 수 있도록 제형을 설계하여 약물치료를 최적화하는 기술을 총칭한다. 상기 DDS는 전달경로, 약물의 종류 및 전달 기술의 형태에 따라 경구형, 주사형, 폐흡입형, 경피형, 점막투여형, 삽입형 등으로 분류될 수 있다.
현재까지 경구 전달을 기반으로 하는 약물 전달 시스템은 다양한 제형, 통증 없는 투여 용이성, 고형 제제에 대한 실행 가능성, 자가 투여 및 환자 순응도를 비롯한 고유한 이점으로 인해 제약 성분을 투여하는 데 가장 선호되는 방법으로 남아 있으나, 이러한 장점에도 불구하고 경구 약물 전달 시스템은 위장관에서의 약물 안정성, 간 초회 통과 대사, 낮은 수용성, 생리학적 장벽에 의한 약물 흡수 제한과 같은 몇 가지 제한 사항이 있다. 예를 들어, 위에서 효소적 생화학적 반응이나 산에 의한 가수분해에 의해 약물이 분해될 가능성이 있고, 장액에서 약물의 용해도 문제와 장막을 통한 약물의 투과성은 약물 흡수의 속도를 제한시켜 약물의 낮은 생체 이용률을 유발할 수 있다.
상술한 경구 전달을 기반으로 하는 DDS의 단점을 극복하기 위해 경피를 통한 약물 전달이 새로운 방안으로 제안되었다. 경피 약물 전달 시스템(Transdermal Drug Delivery System; TDDS)은 피부를 약물 투여 부위로 사용하는 방식으로써 통증 없이 덜 침습적이며, 초회 통과 대사 회피, 적용 및 투여 용이성, 전문 인력이 필요 없고 투여 빈도를 줄일 수 있는 장점이 있다. 또한 TDDS는 친수성 및 소수성의 다양한 약물 전달에 사용될 수 있기 때문에 많은 연구자들의 주목을 받고 있다. 현재까지 TDDS를 통해 약물 전달 효율을 개선하기 위한 몇 가지 전략이 있으며, 크게 4 가지 영역으로 분류할 수 있다.
첫 번째로 겔(gel) 형태의 약물 제형이다. 이러한 제형은 정교한 시스템이나 플랫폼이 필요 없다는 장점이 있다. 주로 고분자(polymer)인 겔화제(gelating agent)와 약물이 3 차원 네트워크를 이루어 분자의 이동성을 감소시켜 약물을 봉입한다. 겔 제형은 수동 확산을 이용하여 약물을 피부로 흡수시키기 때문에 봉입된 약물은 충분한 소수성을 갖는 저 분자량(약 600 Da 미만)을 가져야 하며 저용량 투여에 효과적이어야 한다.
여러 종류의 겔화제 중 구조 내에 높은 비율의 글루코사민을 함유하는 천연 중합체인 키토산은 무독성 및 분해에 대한 민감성으로 인해 젤 형태의 약물 제형에 많이 사용되고 있다. 상기 키토산은 약산성 조건에서 양전하를 띠게 되고 음전하를 띤 세포막을 탈분극시켜 세포막 전위를 감소시킬 수 있으며, 이를 통해 활성 성분 또는 약물의 흡수를 유도할 수 있다는 장점이 있으나, 키토산의 아미노기의 부분적인 양성자화로 인해 생리학적 pH에서의 용해 단백질 분해 효소의 존재 하에서 봉입된 약물의 분해를 유발할 수도 있다는 단점이 있다.
두 번째는 나노/마이크로 크기의 리포솜 또는 입자이다. 이 입자는 극성 머리와 비극성 꼬리로 구성된 독특한 구조로 인해 친수성 및 소수성 약물의 캡슐화에 사용할 수 있다. 상기 입자는 피부 각질층이 지질 이중층과 융합하여 피부로 흡수되고 크기가 작을수록 비표면적이 높아 피부와의 접촉점이 많아서 약물이 피부에 침투하는데 유리하다.
N-아세틸-글루코사민과 글루쿠론산으로 구성된 음이온성 다당류인 히알루론산은 보습 특성과 생체 적합성으로 인해 리포솜 입자에 많이 사용되고 있다. 상기 히알루론산 기반의 나노리포좀은 약물 및 생체분자와의 정전기적, 입체적, 소수성 상호작용으로 인해 입자안정성이 높고 리포좀 구조의 유연성으로 인해 각질층 투과성이 우수한 장점이 있으나, 피부 표면의 증가된 수화작용은 더 많은 수화된 표피층 내에서 약물의 체류를 촉진하여 진피층으로의 약물 전달이 제한될 수 있다는 단점이 있다.
세 번째는 경피 패치형으로, 상기 경피 패치는 피부를 통해 특정 용량의 약물을 혈류로 전달하거나 피부에 퍼뜨리기 위해 피부에 부착하는 약용 접착 패치로써, 약물 방출을 위한 지지체 및 플랫폼을 제공하는 불활성 고분자, 매트릭스에 용해되거나 분산된 약물, 약물 저장소에 우수한 결합을 제공하는 유연한 지지막이 포함된다.
폴리(디메틸실록산)(poly(dimethylsiloxane), PDMS)은 불활성 및 무독성 특성으로 인해 저장소, 매트릭스 및 약물-접착제 층을 포함한 경피 패치 제형에 가장 널리 사용되고 있다. 그러나 상기 PDMS는 강한 소수성으로 인해 봉입된 약물과 강하게 결합하여 약물의 방출을 제한할 수 있다는 단점이 있다.
마지막으로 마이크로니들(microneedle)은 미크론 크기의 바늘을 이용하여 50 μm 내지 900 μm 깊이의 피부 각질층을 관통하여 약물을 전달하는 방법으로, 작은 바늘 크기로 인해 환자의 고통이 적고 약물을 타겟 깊이에 정확하게 전달할 수 있다는 장점이 있다. 마이크로니들 제조에 사용되는 고분자는 수용성, 생체적합성, 피부 삽입에 대한 기계적 강도가 높아야 한다.
폴리락타이드 글리콜라이드(Poly(lactide-co-glycolide, PLGA)는 기계적 강도, 높은 로딩 효율, 생분해성 및 생체 적합성으로 인해 미세 바늘 재료로 많이 사용된다. 상기 PLGA는 분해산물이 수용성이고 무독성이며, 고분자의 분해 속도는 사슬 길이(분자량) 및 락타이드/글리콜라이드 비율에 따라 조절이 가능하다는 장점이 있지만 PLGA 기반 마이크로니들은 삽입된 약물이 방출되도록 며칠 동안 피부에 삽입되어 있어야 한다는 단점이 있다.
상술한 바와 같이 종래 TDDS에 이용되는 고분자들은 각각의 단점들이 존재하는 바, 상기 고분자들의 장점을 결합하고, 각각의 단점의 단점을 대체할 수 있는 새로운 형태의 고분자에 대한 개발이 필요한 실정이다.

본 발명의 실리콘 고분자 화합물은 동일한 단량체를 활용하여, 개시제의 종류, 단량체 대비 개시제의 비율, 중합 온도 등의 중합 조건을 조절함으로써, 합성된 실리콘 고분자 화합물의 분자량, 점도, 접촉각, 함수율, 약물 봉입률 및 약물 방출 거동을 조절할 수 있다는 특성으로 인해 약물 및 다양한 물질을 탑재한 전달시스템으로 활용할 수 있다는 장점이 있다.
나아가, 본 발명의 실리콘 고분자 화합물은 독성이 없는 우수한 생체적합성을 보이고 고분자에 약물 모델을 봉입하여 경피 흡수 경로를 통한 약물전달의 효율을 확인하였을 때, 대조군 대비 높은 흡수율 및 침투율을 확인한 바, 새로운 경피 흡수용 약물전달시스템의 전달체로써 활용될 수 있다.
더 나아가, 본 발명의 실리콘 고분자 화합물은 종양 모델에서도 독성 및 용혈성을 나타내지 않아 우수한 생체적합성을 보이고 항암제를 봉입하여 경피 흡수 경로를 통한 암 치료 효과를 확인하였을 때, 대조군 및 항암제 단독 적용 대비 높은 체중 감소 억제 및 종양 크기/부피 감소 효과를 확인한 바, 암 치료를 위한 제제로써 활용될 수 있다.
본 발명의 효과는 상기한 효과로 한정되는 것은 아니며, 본 발명의 상세한 설명 또는 특허청구 범위에 기재된 발명의 구성으로부터 추론 가능한 모든 효과를 포함하는 것으로 이해되어야 한다.

본 발명자들은 종래 경피 전달 시스템(transdermal delivery system, TDDS)에 이용되는 고분자들의 단점을 대체하고, 장점을 결합할 수 있는 새로운 형태의 고분자를 합성하기 위하여, 고리형 실록산 구조의 단량체의 개환 중합을 통해 PDMS(polydimethylsiloxane)과 유사한 구조의 실리콘 고분자 화합물인 폴리(디메틸실릴에틸렌디메틸실록산)(poly(dimethylsilylethylene-dimethylsiloxane), PDDS) 시리즈를 개발하였다.
본 발명의 PDDS 시리즈는 PDMS와 유사한 구조를 가짐으로써, PDMS의 높은 생체 적합성, 소수성 및 높은 점도 특성을 가지는 동시에, 말단에 키토산 및 히알루론산에 다량 포함하고 있는 히드록시기(-OH)를 도입하여 친수성 및 소수성 특성을 모두 포함하는 것을 특징으로 한다.
본 발명의 개발된 PDDS 시리즈의 화학적, 기계적 성질(분자량, 점도, 접촉각 등)은 산성 개시제의 종류, 단량체와 개시제의 비율, 중합 온도 등에 따라서 다양하게 조절되고, 이러한 특성들로 인해 PDDS 시리즈마다 약물 캡슐화 효율 및 약물 방출 거동이 달라지는 것을 확인하였다. 또한, 본 발명의 일 실시예의 실리콘 고분자가 마우스에서 낮은 독성과 우수한 생체 적합성을 보일 뿐만 아니라 피부 흡수 효율을 확인하였을 때에도 대조군에 비해서 오랜 시간동안 피부로 흡수가 되고, 피부의 진피층을 통과하여 피하조직까지도 약물을 전달할 수 있는 것을 확인함으로써, 본 발명을 완성하였다.

본 발명의 일 실시예에서, 상기 개시제는 유기산, 유기 술폰산, 무기산 등이 포함될 수 있으며, 구체적으로 트리플루오로아세트산(trifluoroacetic acid), 인산(Phosphoric acid), 푸마르산(Fumaric acid), 초산(Acetic acid), 탄소수 1 내지 12을 갖는 알킬 술폰산, 황산(sulfonic acid), 염산(hydrochloric acid), 인산(phosphoric acid), 카르복실산(carboxylic acid), 설폰산(sulfonic acid) 및 시트르산(Citric acid)으로 이루어지는 군에서 선택된 1종 이상, 예를 들면, 초산(Acetic acid), 트리플루오로아세트산(trifluoroacetic acid), 시트르산(Citric acid) 또는 염산(hydrochloric acid) 중 어느 하나, 예를 들면, 초산(Acetic acid), 시트르산(Citric acid) 또는 염산(hydrochloric acid) 중 어느 하나, 바람직하게는 시트르산(Citric acid) 또는 염산(hydrochloric acid) 중 어느 하나일 수 있다.

본 발명의 일 실시예에서, 상기 실리콘 고분자 화합물은 상기 개환 중합하는 단계에서, 개시제의 종류, 개시제와 단량체의 몰비 및 중합 온도 중 어느 하나 이상이 조절되어, 제조되는 실리콘 고분자 화합물의 물성이 조절될 수 있다.
이때, 상기 제조되는 실리콘 고분자 화합물의 물성은 분자량, 점도, 접촉각 및 함수율 중 어느 하나 이상일 수 있다.
본 발명의 일 실시예에서, 본 발명의 제조방법에 이용될 수 있는 개시제의 종류는 상술한 바와 같고, 상기 개시제의 종류에 따라 제조되는 실리콘 고분자 화합물의 분자량, 점도 및/또는 함수율이 조절될 수 있다.
구체적으로, 상기 개시제의 산성도(acidity)가 강할 수록, 상기 제조되는 실리콘 고분자 화합물의 분자량 및 점도가 모두 증가할 수 있고, 상기 개시제의 산성도가 약할 수록 함수율이 증가할 수 있다.
본 발명의 일 실시예에서, 상기 개시제는 상기 단량체 기준 0.01 내지 1.5 몰비로 첨가될 수 있으나, 상기 개시제의 첨가비율은 상술한 개시제의 종류에 따라 상이해질 수 있다.
예를 들면, 상기 개시제가 시트르산인 경우, 상기 개시제는 상기 단량체 기준 0.01 내지 1.0 몰비, 예를 들면, 0.01 내지 0.5 몰비, 예를 들면, 0.01 내지 0.3 몰비, 예를 들면, 0.01 내지 0.2 몰비로 포함될 수 있다.
또 다른 예를 들면, 상기 개시제가 트리플루오로아세트산인 경우, 상기 개시제는 상기 단량체 기준 0.01 내지 1.5 몰비, 예를 들면, 0.01 내지 1.4 몰비, 예를 들면, 0.02 내지 1.3 몰비로 포함될 수 있다.

본 발명의 일 실시예에서, 상기 개시제와 단량체의 몰비에 따라 제조되는 실리콘 고분자 화합물의 분자량 및/또는 점도가 조절될 수 있다.
구체적으로, 상기 단량체 대비 개시제의 비율이 감소할수록 상기 제조되는 실리콘 고분자 화합물의 분자량 및 점도가 모두 증가할 수 있다.
본 발명의 일 실시예에서, 상기 중합하는 단계의 시간 및 온도 등의 중합 조건은 합성하고자 하는 실리콘 고분자 화합물의 물성이 따라 설정될 수 있고, 예를 들면, 10 ℃내지 100 ℃예를 들면, 10 ℃내지 90 ℃예를 들면, 10 ℃내지 80 ℃예를 들면, 20 ℃내지 80 ℃예를 들면, 25 ℃내지 80 ℃의 온도에서, 10 시간 내지 100 시간, 예를 들면, 10 시간 내지 80 시간, 예를 들면, 10 시간 내지 60 시간, 10 시간 내지 50 시간, 예를 들면, 20 시간 내지 50 시간 동안 수행될 수 있다.
본 발명의 일 실시예에서, 상기 중합 온도 조건에 따라 제조되는 실리콘 고분자 화합물의 분자량 및/또는 점도가 조절될 수 있다.
구체적으로, 상기 중합 온도가 높아질 수록 상기 제조되는 실리콘 고분자 화합물의 분자량 및 점도가 모두 증가할 수 있다.
본 발명의 일 실시예에서, 상기 제조되는 실리콘 고분자 화합물의 접촉각의 경우, 상술한 개시제의 종류, 단량체 대비 개시제의 비율 및/또는 중합되는 온도 조건 등에 따라 조절될 수 있다.
구체적으로, 제조되는 실리콘 고분자 화합물이 상기 화학식 2로 표시되는 화합물인 경우, 단량체 대비 개시제의 비율이 감소할 수록, 중합 온도가 높아질 수록 접촉각이 증가할 수 있고, 제조되는 실리콘 고분자 화합물이 상기 화학식 3 또는 화학식 4로 표시되는 화합물인 경우, 단량체 대비 개시제의 비율이 감소할 수록 접촉각이 증가할 수 있다.
본 발명의 실리콘 고분자 화합물이 개시제의 종류, 개시제와 단량체의 몰비 및 중합 온도 중 어느 하나 이상이 조절되어, 제조되는 실리콘 고분자 화합물의 물성이 조절될 수 있음은 상술한 바와 같다.
구체적인 실시예에서, 본 발명의 실리콘 고분자 화합물은 소수성 모델 약물, 친수성 모델 약물, 친수성 항암제, 소수성 항암제, 펩티드, 항생제 등의 다양한 특성을 가지는 약물을 탑재할 수 있게 되는데, 이때, 봉입되는 활성 물질(약물)의 특성, 상기 약물의 봉입률 및/또는 봉입된 약물의 방출 거동 또한, 상기 개시제의 종류, 개시제와 단량체의 몰비 및 중합 온도 중 어느 하나 이상에 따라 조절되어 제조되는 실리콘 고분자 화합물의 물성에 따라 상이해질 수 있다.
따라서, 용도, 용법 및 약물 특성에 따라, 본 발명의 실리콘 고분자 화합물을 설계하여 제조할 수 있게 된다.
본 발명의 일 실시예에서, 상기 제조방법을 이용하여 제조되는 실리콘 고분자 화합물의 분자량은 1,000 g/mol 내지 100,000 g/mol일 수 있으나, 상기 실리콘 고분자 화합물의 분자량은 상술한 개시제의 종류에 따라 상이해질 수 있다.
예를 들면, 상기 개시제가 시트르산인 경우, 상기 실리콘 고분자 화합물의 분자량은 1,000 g/mol 내지 100,000 g/mol, 예를 들면, 1,000 g/mol 내지 10,000 g/mol, 예를 들면, 1,000 g/mol 내지 8,000 g/mol, 예를 들면, 1,000 g/mol 내지 6,000 g/mol일 수 있으나, 이에 제한되는 것은 아니다.
또 다른 예를 들면, 상기 개시제가 트리플루오로아세트산인 경우, 상기 실리콘 고분자 화합물의 분자량은 1,000 g/mol 내지 100,000 g/mol, 예를 들면, 5,000 g/mol 내지 100,000 g/mol, 예를 들면, 5,000 g/mol 내지 80,000 g/mol, 예를 들면, 9,000 g/mol 내지 8,000 g/mol일 수 있으나, 이에 제한되는 것은 아니다.

본 발명의 일 실시예에서, 상기 제조방법을 이용하여 제조되는 실리콘 고분자 화합물의 점도는 60 cP 내지 120,000 cP일 수 있으나, 상기 실리콘 고분자 화합물의 점도는 상술한 개시제의 종류에 따라 상이해질 수 있다.
예를 들면, 상기 개시제가 시트르산인 경우, 상기 실리콘 고분자 화합물의 점도는 60 cP 내지 120,000 cP, 예를 들면, 60 cP 내지 10,000 cP, 예를 들면, 60 cP 내지 1,000 cP, 예를 들면, 60 cP 내지 600 cP일 수 있으나, 이에 제한되는 것은 아니다.
또 다른 예를 들면, 상기 개시제가 트리플루오로아세트산인 경우, 상기 실리콘 고분자 화합물의 점도는 60 cP 내지 120,000 cP, 예를 들면, 100 cP 내지 100,000 cP, 예를 들면, 500 cP 내지 100,000 cP, 예를 들면, 600 cP 내지 96,000 cP 일 수 있으나, 이에 제한되는 것은 아니다.

예를 들면, 상기 개시제가 시트르산인 경우, 상기 실리콘 고분자 화합물의 물 접촉각은 49° 이상, 예를 들면, 49° 내지 100°, 예를 들면, 49° 내지 90°, 예를 들면, 49° 내지 85°일 수 있으나, 이에 제한되는 것은 아니다.

본 발명의 일 실시예에서, 상기 경피 전달용 조성물은, 경피 전달이 필요한 활성 물질, 예를 들어 경피 흡수율의 증가가 필요한 경피로 투여되는 제제를 추가로 포함할 수 있고, 상기 경피 흡수는, 표피(epidermis), 진피(dermis), 및 피하조직(hypodermis)에서 흡수되는 것일 수 있다.
상기 활성 물질은 본 발명의 실리콘 고분자 화합물에 탑재되어 경피 흡수율이 증가될 수 있으며, 구체적으로 상기 고분자 화합물의 사슬구조에 상기 활성 물질이 탑재되어 경피 흡수율이 증가될 수 있다.
상기 '탑재'는 '담지', '봉입'등의 용어로 혼용하여 표기할 수 있으며, 상기 고분자 화합물과 상기 활성 물질이 혼합되어 물리적 혹은 화학적으로 결합되는 것을 의미할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 실리콘 고분자 화합물은 활성 물질의 탑재율, 즉, 봉입률이 높은 것일 수 있으며, 구체적으로 상기 활성 물질은 10 % 내지 99 %, 예를 들면, 10 % 내지 90 %의 봉입률로 탑재된 것일 수 있다.
본 발명의 일 실시예에서, 상기 실리콘 고분자 화합물이 상기 활성 물질을 탑재하여 경피 투여할 경우, 예를 들면, 상기 활성 물질은 피부 도포 후 3시간 이상, 6시간 이상, 12시간 이상, 24시간 이상, 또는 48시간 이상 흡수가 지속되는 것일 수 있다. 이 때, 상기 활성 물질의 흡수 지속 시간의 상한 값이 특정되지 않더라도, 활성 물질의 피부 흡수 지속력을 향상시키는 본원발명의 고분자 화합물의 특징을 통상의 기술자가 명확하게 이해할 수 있을 것이나, 예를 들어 240 시간 이하, 200 시간 이하, 168 시간 이하, 150 시간 이하, 120 시간 이하, 100 시간 이하, 또는 72 시간 이하일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 활성 물질은, 경피 흡수율의 개선 또는 증가가 필요한 경피로 투여되는 제제라면 제한 없이 포함될 수 있고, 예를 들면, 약학 조성물, 화장료 조성물, 진단 시약 조성물, 또는 영상화제일 수 있다.
또한, 상기 활성 물질은 소수성 활성 물질, 친수성 활성 물질, 펩티드 및 항생제 중 어느 하나 이상일 수 있고, 상기 실리콘 고분자 화합물에 탑재되는 활성 물질의 종류 또한, 상기 실리콘 고분자 화합물의 물성에 따라 설계하여 결정될 수 있다.

본 발명의 일 실시예에서, 상기 항암용 조성물은 항암제가 상기 실리콘 고분자 화합물에 탑재된 것일 수 있다. 상기 항암제는 본 발명의 실리콘 고분자 화합물에 탑재되어 경피 흡수율이 증가될 수 있으며, 구체적으로 상기 고분자 화합물의 사슬구조에 상기 항암제가 탑재되어 경피 흡수율이 증가될 수 있다. 상기 경피 흡수는, 표피(epidermis), 진피(dermis), 및 피하조직(hypodermis)에서 흡수되는 것일 수 있다. 상기 탑재는 상기 고분자 화합물과 상기 항암제가 혼합되어 물리적 혹은 화학적으로 결합되는 것을 의미할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에서, 상기 실리콘 고분자 화합물은 항암제의 탑재율, 즉, 봉입률이 높은 것일 수 있으며, 구체적으로 상기 항암제는 10 % 내지 99 %, 예를 들면, 10 % 내지 90 %의 봉입률로 탑재된 것일 수 있다.
본 발명의 일 실시예에서, 상기 항암용 조성물은 항암제가 상기 실리콘 고분자 화합물에 탑재되어, 항암제의 독성 및 용혈성을 나타내지 않으면서도 항암제의 항암 효과를 증대시키는 효과를 나타내는 것일 수 있다. 구체적으로, 암 또는 종양의 크기 및 부피를 감소시키는 암 치료 효과가 증가될 수 있다. 상기 암 치료 효과는 암에 의한 체중 감소를 억제하는 효과를 포함하는 것일 수 있다.
본 발명의 일 실시예에서, 상기 항암용 조성물에 포함되는 실리콘 고분자 화합물은 항암 보조제로 적용하여 사용하는 것일 수 있다. 상기 항암 보조제는 항암제와 병용하여 환자에게 투여됨에 따라 항암제의 항암 치료 효과를 증대시키고, 항암제의 부작용을 억제하거나 개선시키기 위하여 사용되는 것을 의미할 수 있다.
본 발명의 일 실시예에서, 상기 항암용 조성물은 예를 들면, 경피로 투여되는 경피 투여제 또는 경피 투여 제제일 수 있으며, 암 치료를 위해 경피로 투여되는 제제라면 제한 없이 제제화 될 수 있고, 약학 조성물, 피부외용 조성물 또는 화장료 조성물일 수 있다.
본 발명의 일 실시예에서, 상기 암은 피부암, 갑상선암, 위암, 대장암, 폐암, 간암, 유방암, 전립선암, 담낭암, 담도암, 췌장암, 구강암, 식도암, 방광암, 대장암 및 자궁경부암으로 이루어진 군에서 선택된 어느 하나인 것일 수 있다. 상기 피부암은 흑색종, 편평상피세포암, 기저세포암 등을 포함하는 것일 수 있다.
본 발명의 일 실시예에서, 상기 암의 예방 또는 치료 방법은 상기 항암용 조성물을 치료적 유효량으로 개체에 투여하는 것을 포함한다. 특정 개체에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 개체의 연령, 체중, 일반건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다. 따라서 본 발명의 목적에 적합한 조성물의 유효량은 전술한 사항을 고려하여 결정하는 것이 바람직하다.
본 발명의 일 실시예에서, 상기 암의 예방 또는 치료 방법은 상기 항암용 조성물을 치료가 필요한 개체에게 투여하는 것일 수 있고, 상기 치료가 필요한 개체는 환자를 의미하는 것일 수 있고, 상기 개체는 임의의 포유동물에 적용가능하며, 상기 포유동물은 인간 및 영장류뿐만 아니라, 소, 돼지, 양, 말, 개 및 고양이 등의 가축을 포함한다.
이하, 본 발명을 하기의 실시예에 의하여 더욱 상세하게 설명한다. 단, 하기의 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기의 실시예에 의해 한정되는 것은 아니다.
실시예
실시예 1. 실리콘 고분자(PDDS) 시리즈의 합성
단량체(monomer)로 2,2,5,5-테트라메틸-2,5-다이실라-1-옥사시클로펜탄(2,2,5,5-Tetramethyl-2,5-disila-1-oxacyclopentane) 및 개시제(initiator)로 초산(Acetic acid, AA), 시트르산(Citric acid, CA), 트리플루오로아세트산(Trifluoroacetic acid, TFA) 또는 염산(hydrochloric acid, HCl)의 중합 개환 반응을 통하여, 실리콘 기반 고분자 (폴리(디메틸실릴에틸렌-디메틸실록산), poly(dimethylsilylethylene-dimethylsiloxane); 이하, PDDS) 시리즈를 합성하였다.
상기 PDDS 시리즈의 개시제의 종류, 단량체 및 개시제의 비율 및/또는 중합 온도 조건을 달리하여 합성을 수행하였고, 반응 조건을 정리하여 하기의 표 1에 도시하였다:

도 1은 본 발명의 실리콘 고분자 시리즈 중 C2 화합물의 합성 메커니즘의 모식도를 나타낸 것이다. 상기 표 1 및 도 1을 참조하여, 상기 C2 화합물의 합성을 구체적으로 살펴보면, 2,2,5,5-테트라메틸-2,5-디실라-1-옥사사이클로펜탄(200 μL, 1.07 mmol)을 2 M 시트르산(40 μL, 용매: 에탄올) 4 mL 유리 바이알에 넣은 후 1,000 rpm의 속도로 교반하면서 80 ℃에서 48 시간 동안 반응시킨 후, 소량의 물을 추가하여 반응을 종결시킨 후 에탄올(1 mL)로 3 회 세척하여 반응하지 않은 2,2,5,5-테트라메틸-2,5-디실라-1-옥사시클로펜탄과 시트르산을 제거하여 정제하고, n-헥산(200 μL)에 녹인 후 80 ℃ 오븐에서 밤새 건조시켜 C2 화합물을 수득하였다.
상기 C2 화합물의 합성방법에서, 개시제의 종류, 단량체 및 개시제의 비율 및/또는 중합 온도 조건을 상기 표 1에 개시된 조건에 따라 상이하게 하여, A1 내지 A6, C1, C3 내지 C6, T1 내지 T6 및 H1 내지 H6 화합물을 각각 합성하여 수득하였다.
실험예
실험예 1. 특성 분석
(1) 화학적 구조 분석
상기 실시예 1에서 합성한 PDDS 시리즈 화합물을 푸리에 변환 적외선 분광학(Fourier-transform infrared spectroscopy, FT-IR)과 핵자기공명(nuclear magnetic resonance, NMR)을 이용하여 화학적인 구조를 분석하고, 분석 결과를 도 2 내지 도 5에 도시하였다.
한편, 초산(AA)을 개시제로 사용하여 제조된 A 시리즈 화합물의 경우 고분자 형성이 잘 이루어지 않아 FT-IR 및 NMR 데이터가 선명하게 얻어지지 않았고, 이에, 하기 실험예에서는 A 시리즈를 제외한 C, T 및 H시리즈 화합물에 대하여 구체적인 특성 및 효과를 확인하였다.
- FT-IR 분석
도 2 내지 도 3은 본 발명의 일 실시예에 따른 PDDS 시리즈 작용기를 푸리에 변환 적외선 분광학으로 분석한 결과이며 Thermo Fisher 사의 Attenuated total reflection Fourier transform infrared (FT-IR) spectroscopy를 사용하여서 분석하였다.
상기 도 2 및 도 3을 참조하면, 단량체의 경우 2,950 cm-1 내지 2,850 cm-1에서 알칸(alkane)의 ν(C-H)피크, 1,450 cm-1와 1,385 cm-1 내지 1,380 cm-1에서 각각 메틸기(methyl group) 및 디메틸기(dimethyl group)의 δ(C-H)피크 및 1,065 cm-1에서 ν피크(Si-O)를 확인함으로써(ν=stretching, δ=bending), 단량체 구조에 가지고 있는 실란 작용기와(Si-O-Si) 실리콘카바이드(Si-C) 작용기를 확인할 수 있었고, PDDS 시리즈의 경우에도 1,065 cm-1에서 ν의 강도만 증가하였을 뿐 단량체와 동일한 작용기를 가지고 있는 것을 확인하여 중합시 단량체 구조의 변화없이 고분자가 형성된 것을 확인할 수 있었다.
- NMR 분석
도 4 및 도 5는 PDDS 시리즈의 핵자기공명(NMR) 분석 결과이다. 구체적으로, 단량체와 중합체를 CDCl3 (δ7.26 ppm) 용액에 용해시켜서 분석 시료를 제작하고, 핵자기공명 분광기 FT-NMR 500 MHz (JEOL사 제품)를 이용하여 25 ℃의 온도에서 1H NMR을 측정하였다. 측정 횟수는 16 회였고, CDCl3의 피크를 7.26 ppm으로 정하여서 기준점을 잡았다.
도 4 및 도 5를 참조하면, 단량체의 핵자기공명 분석결과 0.313 ppm에서 Si-CH3에 있는 수소원자(proton, 1H)와 0.745 ppm에서 Si-CH2에 있는 수소원자를 찾을 수 있었고, PDDS 시리즈를 분석한 결과 수소원자의 위치가 각각 0.020 ppm과 0.414 ppm으로 움직이는 것을 확인할 수 있었다. 이는 단량체의 고리 구조가 열리면서 분자간의 상호작용이 달라지게 되었기 때문인 것을 예상할 수 있었다.
(2) 분자량 및 점도 분석
겔 침투 크로마토그래피(gel permeation chromatography, GPC)와 회전형 레오미터(Rheometer)를 이용하여 실시예 1에서 합성한 PDDS 시리즈의 분자량과 점도를 측정하여 각각의 측정결과를 도 6 내지 도 9에 도시하였고, 단량체 및 PDDS 시리즈 중 C2, T2, H2을 유리 바이알(vial)에 넣은 후 정방향과 역방향에서의 이미지를 측정하여 도 10 내지 도 12에 도시하였다.
도 6 내지 도 12를 참조하면, GPC 결과에서, 개시제의 산도에 따라 PDDS의 분자량이 증가함을 확인할 수 있었다. 구체적으로, C2는 4,274 g/mol의 분자량(Polydisperisty index; PDI: 2.07)과 86.6 % 전환율, T2는 30,929 g/mol의 분자량(PDI: 1.75)과 96.9 % 전환율, H2는 42,650 g/mol의 분자량(PDI: 1.60)과 96.6 % 전환율을 가지는 것을 확인할 수 있었다.
PDDS 시리즈의 분자량은 단량체 대 개시제의 비율이 증가함에 따라 증가하고 높은 반응 온도(80 ℃)에서 약간 감소하였지만 모든 중합 조건에서 PDDS 시리즈는 1.40 < Mw/Mn < 2.07의 좁은 분자량 분포 범위와 높은 전환율(>85 %)을 보였고, 모든 PDDS 시리즈는 단량체에 비해 점도가 증가했고 분자량 결과와 유사한 강산성 개시제, 높은 단량체 대 개시제 비율 및 낮은 중합 온도에서 더 높은 점도를 나타내는 것을 확인할 수 있었다.
(3) 접촉각과 함수율 분석
도 13 및 도 14는 단량체 및 실시예 1의 PDDS 시리즈의 접촉각을 분석한 결과이다.
구체적으로, 단량체 및 PDDS 시리즈 화합물 각각을 100 μL를 피펫을 이용해서 Marienfeld사의 슬라이드 글라스 위에 도포한 후, 슬라이드 글라스를 좌우로 기울여 가면서 단량체 및 PDDS 시리즈와 슬라이드 글라스 위에 균일하게 코팅 되도록 하였다. 흄후드(fuem hood)안에서 약 1 시간 정도 코팅 후, 다시 단량체와 PDDS 시리즈를 각각 100 μL 떨어트리고, 위의 과정을 2 회 더 반복하여 슬라이드 글라스의 빈 부분이 없도록 코팅을 진행하였다. 그 후 Kruss사의 접촉각 측정기를 이용하여 코팅된 슬라이드 글라스 표면에 증류수를 떨어뜨려 그 접촉각을 분석하였다.
도 13 및 도 14를 참조하면, 접촉각 측정 결과 단량체의 접촉각은 69.0 ± 1.2° 였으나 중합이 진행된 후 접촉각이 증가한 것을 확인할 수 있었다.
접촉각은 개시제의 산성도가 강할수록, 단량체 대비 개시제의 비율이 줄어들수록, 중합온도가 높아질수록 접촉각이 증가하는 경향을 보였지만, 특이하게 시트르산을 사용하여 25 ℃에서 합성한 고분자(C4 내지 C6)들은 단량체에 비해서 더 친수성을 나타내는 것을 확인하였다. (C4: 65.6 ± 3.9°, C5: 56.5 ± 2.7°, C6: 49.2 ± 0.1°)
도 15는 PDDS 시리즈의 도 13 및 도 14의 접촉각을 통한 친수성/소수성에 따른 함수율을 측정한 그래프이다. 구체적으로 PDDS 시리즈 각각 100 μL를 Eppendorf사의 1.5 mL tube에 넣고 동결건조기에서 24 시간동안 건조시킨 후 무게를 측정하였다. 그 후 물 100 μL를 각 tube에 넣고 2 시간 동안 Sientific Industries사의 볼텍스 믹서를 통해 섞어준 후 1 시간정도 상온에 방치하여서 물과 PDDS층이 분리되게 하였다. 남아있는 물층을 제거한 후 다시 무게를 측정하여 함수율을 측정하였다.
도 15를 참조하면, 함수율은 접촉각 결과와 비슷하게 소수성이 강한 PDDS 시리즈(T 시리즈, H 시리즈)의 경우 10 % 미만의 작은 함수율을 보였으나 C 시리즈의 경우 중합 조건에 따라 76 % 이상을 보이는 것을 확인할 수 있었다.
상기 실험예 1의 PDDS 시리즈 각각 화합물의 특성을 분석한 결과를 하기의 표 2 및 표 3에 정리하였다:

실험예 2. 약물 봉입률 및 약물 방출 거동 분석
- 약물 봉입률
상기 실시예 1에서 합성한 PDDS 시리즈의 다양한 약물 봉입률을 확인하기 위하여, 소수성 약물 모델로 oil red O 및 친수성 약물 모델로 메틸렌 블루를 봉입시키고, 봉입 효율 및 인산완충 생리 식염수(Phosphate-buffered saline, PBS)상에서의 방출 거동을 분석하여 관련 도면을 도 16 내지 도 20에 도시하였다.
약물 모델을 탑재시키기 위하여, PDDS 시리즈 100 μL를 4 mL 바이알(vial)에 넣은 후, oil red O 스톡(stock) (1 mg/mL ethanol) 혹은 methylene blue 스톡 (1 mg/mL ehtanol) 1 mL을 넣어주었다. 교반기를 활용하여서 상온 (25 ℃)에서 24 시간 동안 교반을 시켜서 고분자와 약물을 섞어주어 약물의 봉입을 유도하였다. 24 시간이 지난 후 봉입되지 않은 약물 제거하기 위하여, 에탄올 1 mL을 첨가하여 10 분 동안 교반시킨 뒤, 상층액을 제거하는 과정을 3 회 반복하였다.
약물의 봉입 효율을 측정하기 위하여 각각의 세척 과정으로부터 상층액을 수집하였고, Agilent 사의 UV/Vis spectrophotometer를 사용하여서 약물 모델의 흡수 스펙트럼 측정을 통한 표준 곡선(standard curve)과 비교하여 봉입 효율을 분석하였다.
표준 곡선을 구하기 위하여 2 μg/mL 내지 125 μg/mL 농도의 oil red O를 에탄올에 녹인 용액과 2 μg/mL 내지 31 μg/mL 농도의 methylene blue를 에탄올에 녹인 용액을 제조한 뒤 농도에 대한 흡광도를 분석하여 도 16에 도시하였다.
구체적으로, 도 16의 (a)는 소수성 약물인 oil red O의 농도에 따른 흡광도를 측정하여 표준곡선을 구한 그래프이고, (b)는 친수성 약물인 methylene blue의 농도에 따른 흡광도를 측정하여 표준곡선을 구한 그래프이다.
상기 도 16의 각각 약물 모델의 표준곡선을 참조하고, 실시예 1의 PDDS 시리즈 화합물의 최대 피크(peak) 값을 측정하여 각 약물의 표준 곡선을 계산하였다.
실시예 1의 PDDS 시리즈의 oil red O 및 methylene blue 봉입률을 하기의 표 4에 개시하였고, PDDS 시리즈 중 C2, T2 및 H2 화합물의 oil red O의 봉입률(%)을 측정한 그래프 및 methylene blue의 봉입률(%)을 측정한 그래프를 도 17(각각 도 17의 (a) 및 (b))에 도시하였다:

상기 표 4 및 도 17을 참조하면, 본 발명의 PDDS 시리즈는 개시제의 종류, 단량체 대비 개시제의 비율, 중합 온도에 따라 각각 약물 모델의 봉입률이 달라지는 것을 확인할 수 있었고, 중합조건에 따라 oil red O의 경우 넣어준 양에 대비하여 최대 79 % 이상의 봉입 효율을 보였고(PDDS 시리즈 중 T4), methylene blue의 경우 넣어준 양에 대비하여 최대 87 % 이상의 봉입 효율을 확인(PDDS 시리즈 중 C5)하여, 약물의 봉입을 성공적으로 유도함과 동시에 봉입효율을 조절할 수 있음을 확인할 수 있었다. 도 18의 (a)는 단량체, PDDS 시리즈 중 C2, T2 및 H2 화합물에 oil red O가 봉입된 제형의 이미지이고, (b)는 단량체, PDDS 시리즈 중 C2, T2 및 H2 화합물에 methylene blue가 봉입된 제형의 이미지이고, (c)는 PDDS 시리즈 중 C2 화합물에 소수성 약물인 oil red O, 친수성 항암제인 doxorubicin, 소수성 항암제인 SN-38, CGKRK 서열의 펩티드, 항생제인 ciprofloxacin을 봉입한 제형 및 C2 단독의 UV(365 nm) 빛을 조사했을 때의 이미지이다.
상기 도 18의 (a) 및 (b)를 참조하면, 약물을 탑재한 후의 PDDS 시리즈에서 눈에 띄는 점도 감소는 확인할 수 없었고, 도 18의 (c)를 참조하면, 본 발명 실리콘 고분자 화합물 C2에 소수성 약물 모델(oil red O), 친수성 항암제(doxorubicin), 소수성 항암제(SN-38), 펩티드(CGKRK peptide), 항생제(ciprofloxacin)를 봉입하였을 때에도 성공적으로 봉입이 되는 것을 확인할 수 있었다.
도 19 및 도 20은 각각 PDDS 시리즈 안에 봉입된 oil red O(도 19)와 methylene blue(도 20)의 인산완충생리식염수(Phosphate-buffred saline, PBS)에서의 방출곡선을 측정한 그래프이다.
구체적으로, oil red O 및 methylene blue이 각각 봉입된 PDDS 시리즈 100 μL을 PBS 1 mL에 분산시킨 후 37℃에서 배양하였다. 일정한 시간이 지난 후, PDDS 시리즈와 상 분리된 PBS층을 수득하여 이를 흡수 스펙트럼으로 측정하여서 표준 곡선과 비교하여 방출된 oil red O와 methylene blue의 양을 정량 하였다.
도 19 및 도 20을 참조하면, PDDS 시리즈에 따라 봉입된 약물의 방출거동을 조절할 수 있는 것을 확인할 수 있었고, oil red O를 탑재한 T 시리즈와 H 시리즈의 경우 9 일이 지난 시점에서도 봉입된 약물의 10 % 정도만이 방출된 반면 C 시리즈의 경우 70 %가 방출된 것을 알 수 있었다. methylene blue의 경우 대부분의 시리즈에서 3 일이 지난 시점에서 거의 대부분의 약물이 방출되는 것을 알 수 있었다.
실험예 3. 생체적합성 확인(생체 내(in vivo) 독성 확인)
PDDS 시리즈의 생체적합성을 확인하기 위하여 쥐를 이용하여 면역독성을 확인하였다.
구체적으로, 음성 대조군으로 인산완충생리식염수(phosphate-buffered saline, PBS), 양성 대조군으로 면역독성을 유발하는 유게놀(eugenol, Eug) 및 PDDS 시리즈 중 C2 화합물 각각을 25 μL씩을 쥐의 귀 뒷부분에 3 일간 도포 하고, 6 일이 지난 후의 쥐의 체중 변화, 급식량과 음수량, 귀림프절(auricular lymph node)의 무게, 림프절 세포 수, 자극지수, 혈액 내 히스타민(histamine)과 면역글로불린 E(immunoglobulin E)의 농도를 측정하였다.
도 21은 PBS, C2 화합물 및 eugenol을 6 일간 처리한 후 쥐의 체중 변화(a), 급식량(b) 및 급수량(c)을 측정한 그래프이다.
도 21을 참조하면, PBS를 처리한 그룹에서는 실험 전과 종료 후 쥐의 체중변화가 없었고, C2를 처리한 그룹에서는 약 0.5 g 체중이 증가하였고, eugenol을 처리한 그룹에서는 약 0.5 g의 체중감소가 보이는 것을 확인할 수 있었다.
급식량과 음수량 또한 C2를 처리한 그룹은 PBS 그룹과 비슷한 양의 사료와 물을 먹었지만 eugenol을 처리한 그룹에서는 더 적은 양의 사료와 물을 먹은 것을 확인할 수 있었다.
도 22는 PBS, C2 화합물 및 eugenol을 6 일간 처리한 후 쥐의 림프절(lymph node) 무게를 측정한 그래프이다.
도 22를 참조하면, C2 화합물을 처리한 그룹에서는 림프절의 무게가 PBS를 처리한 그룹과 차이가 없었으나, eugenol을 처리한 그룹은 2 배 이상 무게가 증가한 것을 확인할 수 있었다.
도 23은 PBS, C2 화합물 및 eugenol을 6 일간 처리한 후 브로모데옥시유리딘(Bromodeoxyuridine) 분석법을 통해 쥐의 림프절의 세포양(BrdU 농도)과 자극치수를 측정한 그래프이고, 도 24는 쥐의 혈액 내 histamine과 immunoglobulinE의 양을 측정한 그래프이다.
도 23을 참조하면, 쥐의 적출한 귀 림프절의 세포숫자를 BrdU assay를 통하여 측정하고, 이를 토대로 자극지수를 계산한 결과, 림프절의 무게와 마찬가지로 eugenol을 처리한 그룹은 림프절의 세포수와 자극지수가 통계적으로 유의미하게 증가하였으나, C2 화합물을 처리한 그룹은 PBS 처리 그룹과 동일한 세포수와 자극지수를 보이는 것을 확인할 수 있었다.
도 24를 참조하면, 쥐의 혈액을 채취하여 혈액내 히스타민과 면역글로불린 E의 농도를 측정한 결과, C2 화합물을 처리한 그룹은 PBS 그룹과 비슷한 히스타민 농도를 보였고, 오히려 면역글로불린 E의 농도가 감소한 반면, eugenol을 처리한 그룹은 히스타민과 면역글로불린 E의 농도 둘다 PBS 그룹에 비해 증가한 것을 확인할 수 있었다.
상기 실험예 3을 통하여, C2 화합물의 처리가 쥐의 체중 감소, 급식/급수량에 영향을 미치지 않을 뿐만 아니라, 림프절의 자극 혹은 체내 면역반응을 유도하지 않으므로, 생체적합성이 우수하다는 것을 확인할 수 있었다.
실험예 4. 피부 침투 효율 확인
실시예 1에서 합성한 PDDS 시리즈의 피부 흡수 효율을 확인하기 위하여 모델 약물인 oil red O가 탑재된 C2 화합물과 oil red O를 쥐의 피부에 도포 후 24 시간과 48 시간이 지난 후 피부 조직을 절편하여 oil red O의 위치를 추적한 결과를 도 5에 도시하였다.
구체적으로, oil red O가 탑재된 C2 화합물(C2; 50μL) 및 oil red O(OR; 100μL, 스톡 용액: PBS(1% 에탄올) 중 24μM)을 각각 100 μL를 피부에 도포한 후 24 시간과 48 시간 경과 후, 피부조직을 절편하고, 형광현미경을 이용하여서 oil red O의 형광을 추적하고(여기:559 nm. 검출: 636±20nm), 이를 Image-J 프로그램을 통해, 정량하여 플롯팅하였다.
도 25는 oil red O 단독 물질 및 C2 화합물에 oil red O가 봉입된 제형(C2)처리 24 시간 후(a-c) 및 48시간 후(d-f)의 마우스 피부의 시편 단면 CLSM 이미지(a, d) 및 피부조직 내 oil red O의 흡수율을 확인한 그래프(b, c, e, f)이다. 여기서, (b, c) 및 (e, f)는 각각 (a) 및 (d)의 이미지에서의 노란색 점선을 따라 Oil red O 또는 oil red O가 탑재된 C2 화합물(C2)의 형광 강도 플롯을 나타낸 것이다.
도 25를 참조하면, oil red O를 단독으로 처리한 그룹은 24 시간 및 48 시간 경과 후에도 피하조직으로 거의 흡수가 되지 않았으나, oil red O가 탑재된 C2 화합물을 처리한 그룹은 24 시간 경과 후 표피층(epidermis)을 지나 진피층(dermis)에서 가장 높은 형광 intensity를 보였고, 피하조직(hypodermis)에서도 형광이 측정된 것을 확인할 수 있었다.
48 시간 경과 후에는, 봉입된 oil red O가 더 깊이 침투하여 진피층과 피하조직에서 강한 형광을 나타내는 것을 확인할 수 있었다.
실험예 5. 항암 효과 확인
실시예 1에서 합성한 PDDS 시리즈의 항암 효과를 확인하기 위하여 흑색종 이종이식 마우스에서 C2 화합물의 항암 효과를 평가하였다.
(1) C2 화합물 단독 투여의 항암 효과
먼저, C2 화합물 단독(약물 미봉입)의 항암 효과 분석을 위해, B16F10(마우스 뮤린 흑색종 세포주) 세포를 0일째에 마우스(6주령 암컷 Balb/c 누드)의 좌측 또는 우측 대퇴부(피하)에 이식하여 흑색종 종양을 유도한 다음, PBS 또는 C2 화합물을 16일 동안 매일 100μL씩 국소 적용하면서, 2~3일마다 체중 및 종양 부피 변화를 측정하였다. 또한, 용혈 분석을 위해 이소플루란(isoflurane)으로 마취 된 마우스의 심장에서 혈액 샘플을 얻은 다음, 차가운 1×PBS(1mL, 3회)로 4℃(3000rpm, 3분)에서 원심분리하여 적혈구를 수집하였다. 정제된 적혈구에 100μL의 PBS 및 C2 화합물을 각각 8%(v/v) 처리하고 37℃에서 1시간 동안 추가로 배양하였다. 그 다음 혼합물을 4℃(3000rpm, 3분)에서 원심분리를 사용하여 수집하였으며, 상층액의 UV/vis 흡수 피크(492 nm)를 25℃에서 측정하였다.
도 26을 참조하면, C2 화합물을 단독으로 처리한 마우스의 체중 및 종양 부피 변화는 PBS를 처리한 마우스와 유사한 결과를 나타내었으나, 독성 및 용혈성은 나타내지 않아 C2 화합물의 생체적합성이 우수함을 다시 확인할 수 있었다.
(2) 항암제를 봉입한 C2 화합물의 항암 효과
다음으로, 실험예 2와 동일한 방법을 이용하여 항암제(7-Ethyl-10-hydroxycamptothecin; SN-38)를 담지한 C2 화합물 제제(캡슐화 효율=봉입율=14.7%)를 제조하여, 이의 항암 효과를 확인하였다. 구체적으로, B16F10(마우스 뮤린 흑색종 세포주) 세포를 0일째에 마우스(6주령 암컷 Balb/c 누드)의 좌측 또는 우측 대퇴부(피하)에 이식하여 흑색종 종양을 유도한 다음, 10일이 지난 후(종양 크기=~200 mm3), 흑색종 모델 마우스를 3그룹으로 나누고(그룹당 5마리), 각 그룹에 PBS, SN-38(3mM) 또는 C2+SN-38(SN-38 항암제 봉입된 C2 화합물) 100μL를 10, 12, 13, 14일에 국소 도포를 통해 투여하였다(도 27(a)). 16일 동안 흑색종 모델 마우스에서의 독성 및 치료 효과를 확인하기 위해 2~3일마다 체중과 종양 부피 변화를 측정하였다. 실험 종료 시점(15일째)에 각 마우스의 몸통을 디지털 카메라로 종양 형태 및 크기를 촬영하고, 실험 종료 후(16일째) 마우스로부터 종양을 적출하였다. 적출된 종양 조직(harvested tumor)은 디지털 카메라로 촬영하고, 크기 및 부피를 측정하였다. 또한, 용혈 분석을 위해 마우스 심장으로부터 얻어진 혈액 샘플로부터 정제된 적혈구에 100μL의 PBS(음성 대조군), SN-38(3mM), C2+SN-38 및 Triton-X(0.1%(v/v), 양성 대조군)을 각각 8%(v/v) 처리하고, 37℃에서 1시간 동안 추가로 배양한 다음, 혼합물을 4℃(3000rpm, 3분)에서 원심분리하여 최종 수득된 상층액의 UV/vis 흡수 피크(492 nm)를 25℃에서 측정하였다.
도 27(a-e)을 참조하면, PBS 및 SN-38 처리된 마우스의 체중은 15일 동안 유의한 체중 변화가 없는 반면, C2+SN-38 처리된 마우스는 10일부터 체중 증가를 나타내었고, C2+SN-38을 처리한 마우스는 PBS 및 SN-38 단독 처리에 비해 실험기간(16일) 동안 종양 부피가 유의하게 감소되는 것으로 나타났다. 적출된 종양 조직의 부피를 측정한 결과, SN-38은 종양 성장을 약간만 감소시킨 반면, C2+SN-38 처리시 종양 크기가 극적으로 감소되었고, C2+SN-38 처리 그룹의 평균 종양 부피는 281.2±102.1 mm3인 반면 PBS 및 SN-38 처리 그룹의 평균 종양 부피는 각각 1845±647.1 mm3, 1689±498.6 mm3였다.
이러한 결과로부터 SN-38 항암제 봉입된 C2 화합물(C2+SN-38)이 암의 진행을 유의하게 감소시키고 흑색종 이종이식 마우스를 회복시킬 수 있음을 확인하였으며, SN-38 항암제 봉입된 C2 화합물은 항암제 단독 투여시 보다 깊은 피부 침투 능력을 통해 항암제의 치료 효능을 증가시킬 수 있음을 알 수 있었다.
또한, SN-38 항암제 봉입된 C2 화합물의 투여 안전성을 확인하기 위해 용혈성을 분석한 결과, 도 27(f)에 나타난 바와 같이, SN-38 단독 처리의 경우 심각한 용혈성을 나타내었지만, C2+SN-38은 적혈구에서 <0.1%의 무시할만한 용혈율을 나타내었다. 따라서, C2 화합물이 항암제 단독 투여에 비해 비해 봉입된 항암제의 혈액 독성을 감소시킬 뿐만 아니라, 항암제 약물 봉입(로딩) 후에도 C2 화합물 자체의 높은 생체 적합성을 유지할 수 있음을 확인하였다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특히 청구범위에 나타나 있으며, 그와 동등한 범위내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (25)
- 제 1 항에 있어서,상기 실리콘 고분자 화합물의 분자량은 1,000 g/mol 내지 100,000 g/mol인 것을 특징으로 하는 실리콘 고분자 화합물.
- 제 1 항에 있어서,상기 고분자 화합물의 점도는 60 cP 내지 120,000 cP인 것을 특징으로 하는 실리콘 고분자 화합물.
- 제 1 항에 있어서,상기 고분자 화합물의 물 접촉각은 49° 이상인 것을 특징으로 하는 실리콘 고분자 화합물.
- 개시제를 이용하여 하기 화학식 A로 표시되는 단량체를 개환 중합하는 단계를 포함하는, 하기 화학식 1로 표시되는 실리콘 고분자 화합물의 제조 방법:[화학식 A]상기 식에서,m은 1 내지 3의 정수이고,R1 내지 R4는 각각 독립적으로 알킬(alkyl), 비닐(vinyl), 아세틸(acetyl), 아로마틱(aromatic), 아지드(azide), 히드록시(hydroxy) 및 알킬실록시(alkylsiloxy) 중 어느 하나이고,R5는 수소 또는 알킬(alkyl)이다.[화학식 1]상기 식에서,m은 1 내지 3의 정수이고,n은 10 내지 505의 정수이고,R1 내지 R4는 각각 독립적으로 알킬(alkyl), 비닐(vinyl), 아세틸(acetyl), 아로마틱(aromatic), 아지드(azide), 히드록시(hydroxy) 및 알킬실록시(alkylsiloxy) 중 어느 하나이고,R5는 수소 또는 알킬(alkyl)이고,
- 제 6 항에 있어서,상기 개환 중합하는 단계에서,개시제의 종류, 개시제와 단량체의 몰비 및 중합 온도 중 어느 하나 이상이 조절되어, 제조되는 실리콘 고분자 화합물의 물성이 조절되는 것을 특징으로 하는 실리콘 고분자 화합물의 제조방법.
- 제 8 항에 있어서,상기 조절되는 실리콘 고분자 화합물의 물성은 분자량, 점도, 접촉각 및 함수율 중 어느 하나 이상을 포함하는 것을 특징으로 하는 실리콘 고분자 화합물의 제조방법.
- 제 6 항에 있어서,상기 개시제는 트리플루오로아세트산(trifluoroacetic acid), 인산(Phosphoric acid), 푸마르산(Fumaric acid), 초산(Acetic acid), 황산(sulfonic acid), 염산(hydrochloric acid), 인산(phosphoric acid), 카르복실산(carboxylic acid), 설폰산(sulfonic acid) 및 시트르산(Citric acid)으로 이루어지는 군에서 선택된 1종 이상인 것을 특징으로 하는 실리콘 고분자 화합물의 제조방법.
- 제 6 항에 있어서,상기 개시제는 상기 단량체 기준 0.01 내지 1.5 몰비로 첨가되는 것을 특징으로 하는 제조방법.
- 제 6 항에 있어서,상기 중합하는 단계는 10 ℃ 내지 100 ℃에서 10 시간 내지 100 시간 동안 수행되는 것을 특징으로 하는 제조방법.
- 제 13 항에 있어서,상기 경피 전달은 표피(epidermis), 진피(dermis) 및 피하조직(hypodermis)에 전달되는 것을 특징으로 하는 경피 전달용 조성물.
- 제 13 항에 있어서,상기 경피 전달용 조성물은 소수성 활성 물질, 친수성 활성 물질, 펩티드 및 항생제 중 어느 하나 이상의 활성 물질이 상기 실리콘 고분자 화합물에 탑재된 것을 특징으로 하는 경피 전달용 조성물.
- 제 16 항에 있어서,상기 활성 물질은 10 % 내지 99 %의 봉입률로 탑재된 것을 특징으로 하는 경피 전달용 조성물.
- 제 13 항의 경피 전달용 조성물 및 활성 물질을 국소적으로 적용하는 것을 포함하는 활성 물질의 경피 전달 방법.
- 제 18 항에 있어서,상기 활성 물질은 소수성 활성 물질, 친수성 활성 물질, 펩티드 및 항생제 중 어느 하나 이상인 것을 특징으로 하는 활성 물질의 경피 전달 방법.
- 제 20 항에 있어서,상기 항암용 조성물은 SN-38, 이리노테칸, 독소루비신, 파크리탁셀, 에피루비신, 젬시타빈, 실로리무스, 에토포사이드, 빈블라스틴, 빈카알칼로이드, 도세탁셀, 시스플라틴, 글리벡, 아드리아마이신, 시클로포스파미드, 테니포사이드, 5-플로오로우라실, 캠토세신, 타목시펜, 아나스테로졸, 플록슈리딘, 류프로리드, 플로타미드, 졸레드로네이트, 빈크리스틴, 스트렙토조토신, 카보플라틴, 이포스파마이드, 토포테칸, 벨로테칸, 이리노테칸, 비노렐빈, 히도록시우레아, 발루비신, 메소트렉세이트, 메클로레타민, 클로람부실, 부술판, 독시플루리딘, 프레드니손, 테스토스테론, 미토산트론, 도세탁셀, 비노렐빈 및 프레드니솔론으로 이루어진 군에서 선택된 어느 하나 이상의 항암제를 더 포함하는 것을 특징으로 하는 항암용 조성물.
- 제 20 항에 있어서,상기 항암용 조성물은 항암제가 상기 실리콘 고분자 화합물에 탑재된 것을 특징으로 하는 항암용 조성물.
- 제 20 항에 있어서,상기 암은 갑상선암, 위암, 대장암, 폐암, 간암, 유방암, 전립선암, 담낭암, 담도암, 췌장암, 구강암, 식도암, 방광암, 대장암 및 자궁경부암으로 이루어진 군에서 선택된 어느 하나인 것을 특징으로 하는 항암용 조성물.
- 제 20 항의 항암용 조성물을 국소적으로 적용하는 것을 포함하는 암의 예방 또는 치료 방법.
