Abstract
본 발명은 리피테그라스트의 신규한 결정형 및 이를 포함하는 약제학적 조성물을 제공한다. 본 발명에 따른 리피테그라스트 결정형은 용해도가 개선되고 열 및 수분에 대하여 안정성이 뛰어나므로 약학적 제제의 제조에 유용하게 사용될 수 있다.
Description
본 발명은 리피테그라스트 결정형 및 이를 포함하는 약제학적 조성물에 관한 것이다. 보다 구체적으로 본 발명은 용해도가 개선되고 열 및 수분에 대하여 안정성이 뛰어난 리피테그라스트 결정형 및 이를 포함하는 약제학적 조성물에 관한 것이다.
안구건조증이란 안구에서 눈물이 부족하거나 눈물이 지나치게 증발해 생기는 질환이다. 눈물층은 지방층, 수성층 및 점액층으로 구성된다. 이들은 각각 눈물 증발 방지, 수분공급 및 눈에서 눈물을 잘 머무르게 하는 역할을 한다. 이러한 눈물층에 외부자극이 가해지거나 기타요인에 의해 각 층의 성분 변화 등이 발생할 경우 안구건조증이 생긴다. 안구건조증이 생기면 눈에서 눈물이 마르거나 흐르지 않아 안구에서 이물감이 느껴지고 안구 표면이 쉽게 손상되어 자극을 느끼게 된다. 안구건조증을 앓고 있는 사람들은 이러한 이물감이나 자극으로 인해 눈이 쉽게 충혈되고 피로함을 느끼게 되며, 안구건조증이 심할 경우에는 두통이 발생되는 경우가 있다. 현대인들의 경우, 이러한 안구 건조증이 쉽게 유발될 수 있는 각종 디지털 기기 등의 노출로 인한 안구 건조증이 많이 발생하고 있다. 이 외에도 약물 등에 의한 부작용, 노화, 렌즈 착용 등의 이유로 안구 건조증의 발생률이 증가하는 추세이다.
이러한 안구 건조증 치료를 위해 리피테그라스트를 주성분으로 하는 인공눈물이 사용되고 있다.
하기 화학식 1의 리피테그라스트는 샤이어사에서 개발한 안구건조증 치료제인 자이드라 점안액의 유효성분으로서 2016년 7월에 미국 식품의약국의 승인을 받아 출시하였으며 현재 실온보관 1회용 점안제로 판매되고 있다.

이러한 리피테그라스트 및 이의 결정형은 미국특허 제8,367,701호 및 미국특허 제10,435,395호에 개시되어 있다.
그러나, 상기 리피테그라스트 및 이의 결정형은 물에 대한 용해도가 낮아 제제화가 용이하지 않으며 생체 이용률이 떨어지는 문제점이 있었다.
아울러, 열 및 수분에 대하여 안정성이 뛰어난 신규한 리피테그라스트 결정형에 대한 연구가 여전히 요구되고 있다.
본 발명의 목적은 용해도가 개선되고 열 및 수분에 대하여 안정성이 뛰어난 리피테그라스트 결정형을 제공하는 것이다.
본 발명의 다른 목적은 유효성분으로서 상기 리피테그라스트 결정형을 포함하는 약제학적 조성물을 제공하는 것이다.
본 발명의 일 실시형태는 X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 9.8±0.2, 10.9±0.2, 16.5±0.2, 18.0±0.2, 19.5±0.2, 20.2±0.2, 20.9±0.2, 21.8±0.2, 22.9±0.2, 23.7±0.2, 24.3±0.2, 24.4±0.2, 24.9±0.2, 26.0±0.2 및 29.7±0.2인 하기 화학식 1로 표시되는 리피테그라스트 결정형 S형에 관한 것이다.

본 발명의 일 실시형태에 따른 리피테그라스트 결정형 S형은 상기 X선 분말 회절피크 이외에 약한 강도로, 12.2±0.2, 15.4±0.2, 28.2±0.2, 30.6±0.2, 32.2±0.2, 34.2±0.2, 34.8±0.2, 35.3±0.2 및 38.9±0.2의 회절각 (2θ)에서 X선 분말 회절피크를 추가로 가질 수 있다. 이때, "약한 강도"란 I/I0(상대강도)가 10% 미만인 피크를 나타낸다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 S형은 시차주사열량 분석에서, 첫번째 흡열점이 57.0℃, 두번째 흡열점이 164.3℃의 값을 나타낸다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 S형은 리피테그라스트 무정형을 에탄올에 현탁시키고, 열을 가하여 교반하거나 상온에서 교반한 후, 여과함으로써 제조되거나, 리피테그라스트 결정형 Y형을 25℃ 상대습도 60% 조건에서 보관함으로써 제조될 수 있다. 상기 보관은 12시간 이상 수행될 수 있다.
또한, 본 발명의 일 실시형태에 따른 리피테그라스트 결정형 S형은 유기용매에 용해되어 있는 리피테그라스트로부터 제조될 수 있다.
구체적으로, 유기용매에 용해되어 있는 리피테그라스트에 에탄올을 가하여 공비하여 용매를 치환하고 열을 가하여 교반하거나 상온에서 교반한 후, 여과하여 감압 건조함으로써 제조될 수 있다. 이때, 리피테그라스트를 용해하는데 사용할 수 있는 유기용매로는 물; 펜탄, 석유 에테르, 헥산 등의 지방족 용매; 톨루엔, 자일렌 등의 방향족 용매; 다이클로로메테인 등의 할로겐화 용매; 부탄온, 아세톤 등의 지방족 케톤; 아세트산에틸, 아세트산이소프로필, 아세트산부틸 등의 지방족 에스테르; 디에틸 에테르, tert-부틸메틸 에테르(TBME), 테트라하이드로퓨란 등의 지방족 에테르; 방향족 에테르; 아세토니트릴 등의 니트릴계 용매; 알켄; 알킨; 알케닐 에스테르; 및 알키닐 에스테르를 예로 들 수 있다. 이들 유기용매 중에서 특히 다이클로로메테인, 아세트산에틸, 부탄온 및/또는 테트라하이드로퓨란이 바람직하고, 다이클로로메테인이 가장 바람직하다.
또한, 재결정 용매로서 사용하는 에탄올은 상기에서 언급한 유기용매와 혼합하여 사용할 수 있다. 유기용매/에탄올의 비율은 약 10/90, 20/80, 30/70, 40/60/, 50/50, 60/40, 70/30, 80/20, 또는 90/10이고, 모든 양은 "약"을 포함한다.

본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Y형은 상기 X선 분말 회절피크 이외에 약한 강도로, 11.9±0.2, 16.1±0.2, 22.4±0.2, 30.8±0.2, 31.5±0.2, 34.4±0.2, 35.4±0.2, 36.4±0.2, 36.5±0.2 및 39.7±0.2의 회절각 (2θ)에서 X선 분말 회절피크를 추가로 가질 수 있다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Y형은 시차주사열량 분석에서, 흡열점이 162.2의 값을 나타낸다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Y형은 리피테그라스트 무정형을 에탄올에 현탁시키고, 열을 가하여 교반하거나 상온에서 교반한 후, 여과하여 감압 건조함으로써 제조될 수 있다.
또한, 본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Y형은 리피테그라스트 에틸렌글라이콜 용매화물로부터 제조될 수 있다.
구체적으로, 리피테그라스트 결정형 Y형은 리피테그라스트 에틸렌글라이콜 용매화물을 현탁용매에 현탁시키고 상온에서 교반한 다음, 여과하여 감압 건조 또는 60 내지 80℃에서 오븐 건조함으로써 제조될 수 있다. 이때, 현탁 용매로는 물, 에탄올, 아세톤 등이 사용될 수 있고, 특히 물이 바람직하다.
또한, 본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Y형은 유기용매에 용해되어 있는 리피테그라스트로부터 제조될 수 있다.
구체적으로, 유기용매에 용해되어 있는 리피테그라스트에 에탄올을 가하여 공비하여 용매를 치환하고 열을 가하여 교반하거나 상온에서 교반한 후, 여과하여 감압 건조함으로써 제조될 수 있다. 이때, 리피테그라스트를 용해하는데 사용할 수 있는 유기용매로는 물; 펜탄, 석유 에테르, 헥산 등의 지방족 용매; 톨루엔, 자일렌 등의 방향족 용매; 다이클로로메테인 등의 할로겐화 용매; 부탄온, 아세톤 등의 지방족 케톤; 아세트산에틸, 아세트산이소프로필, 아세트산부틸 등의 지방족 에스테르; 디에틸 에테르, tert-부틸메틸 에테르(TBME), 테트라하이드로퓨란 등의 지방족 에테르; 방향족 에테르; 아세토니트릴 등의 니트릴계 용매; 알켄; 알킨; 알케닐 에스테르; 및 알키닐 에스테르를 예로 들 수 있다. 이들 유기용매 중에서 특히 다이클로로메테인, 아세트산에틸, 부탄온 및/또는 테트라하이드로퓨란이 바람직하고, 다이클로로메테인이 가장 바람직하다.
또한, 재결정 용매로서 사용하는 에탄올은 상기에서 언급한 유기용매와 혼합하여 사용할 수 있다. 유기용매/에탄올의 비율은 약 10/90, 20/80, 30/70, 40/60/, 50/50, 60/40, 70/30, 80/20, 또는 90/10이고, 모든 양은 "약"을 포함한다.
본 발명의 또 다른 실시형태는 X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 9.7±0.2, 12.6±0.2, 18.7±0.2, 20.8±0.2, 21.6±0.2, 23.4±0.2 및 24.1±0.2 인 리피테그라스트 에틸렌글라이콜 용매화물에 관한 것이다.
본 발명의 일 실시형태에 따른 리피테그라스트 에틸렌글라이콜 용매화물은 상기 X선 분말 회절피크 이외에 약한 강도로, 6.7±0.2, 8.6±0.2, 11.6±0.2, 13.9±0.2, 16.2±0.2 17.1±0.2, 17.9±0.2, 18.0±0.2, 19.9±0.2, 22.2±0.2, 22.7±0.2, 24.8±0.2, 25.5±0.2, 25.8±0.2, 26.2±0.2, 27.0±0.2, 28.1±0.2, 29.0±0.2, 34.1±0.2, 35.6±0.2, 36.3±0.2, 37.9±0.2 및 38.9±0.2의 회절각 (2θ)에서 X선 분말 회절피크를 추가로 가질 수 있다.
본 발명의 일 실시형태에 따른 리피테그라스트 에틸렌글라이콜 용매화물은 시차주사열량 분석에서, 흡열점이 144.5℃의 값을 나타낸다.
본 발명의 일 실시형태에 따른 리피테그라스트 에틸렌글라이콜 용매화물은 리피테그라스트 무정형을 에틸렌글라이콜에 현탁시키고, 열을 가하여 교반하거나 상온에서 교반한 후, 여과하여 감압 건조함으로써 제조될 수 있다.
본 발명의 또 다른 실시형태는 X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 7.5±0.2, 8.1±0.2, 8.7±0.2, 9.8±0.2, 10.2±0.2, 12.5±0.2, 17.5±0.2, 18.3±0.2, 18.6±0.2, 19.0±0.2, 20.3±0.2, 21.1±0.2, 21.5±0.2, 21.9±0.2, 22.2±0.2, 22.9±0.2, 23.8±0.2, 24.3±0.2, 24.8±0.2, 25.6±0.2, 26.4±0.2, 27.0±0.2, 28.5±0.2 및 34.8±0.2인 리피테그라스트 결정형 Z형에 관한 것이다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Z형은 시차주사열량 분석에서, 흡열점이 162.3℃의 값을 나타낸다.
본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Z형은 리피테그라스트 무정형을 현탁용매에 현탁시키고 열을 가하여 용해시키거나 상온에서 교반하여 이를 냉각한 후, 여과하여 감압 건조함으로써 제조될 수 있다. 이때, 현탁용매로는 에틸렌글라이콜다이메틸에터, 1,2-다이메톡시에테인, 프로피오나이트릴 등이 사용될 수 있고, 특히 에틸렌글라이콜다이메틸에터가 바람직하다.
또한, 본 발명의 일 실시형태에 따른 리피테그라스트 결정형 Z형은 리피테그라스트 에틸렌글라이콜 용매화물 또는 결정형 S형이나 Y형으로부터 제조될 수 있다.
구체적으로, 리피테그라스트 결정형 Z형은 리피테그라스트 에틸렌글라이콜 용매화물을 현탁용매에 현탁시키고 열을 가하여 용해시키거나 상온에서 교반하여 이를 냉각한 후, 여과하여 감압 건조함으로써 제조될 수 있다. 이때, 현탁 용매로는 에틸렌글라이콜다이메틸에터, 1,2-다이메톡시에테인, 프로피오나이트릴 등이 사용될 수 있고, 특히 에틸렌글라이콜다이메틸에터가 바람직하다.
본 발명의 또 다른 실시형태는 상기 리피테그라스트 결정형 S형, 상기 리피테그라스트 결정형 Y형, 상기 리피테그라스트 에틸렌글라이콜 용매화물 및 상기 리피테그라스트 결정형 Z형으로 구성된 군으로부터 선택되는 하나 이상의 결정형 또는 용매화물을 약제학적으로 허용되는 담체와 함께 포함하는 안구건조증을 치료하기 위한 약제학적 조성물에 관한 것이다.
본 발명의 약제학적 조성물은 상기 리피테그라스트 결정형 또는 용매화물 이외에 또 다른 생리활성물질을 함께 포함할 수 있다.
본 발명에 따른 약제학적 조성물은 경구적으로 투여될 수 있으며, 정제, 캡슐제, 과립제, 산제, 유탁액제, 현탁액제, 시럽제 등 여러 형태로 제형화될 수 있다. 상기 여러 형태의 약제학적 조성물은 부형제, 충진제, 증량제, 결합제, 붕해제(disintegrator), 활택제, 윤활제, 방부제, 항산화제, 등장제(isotonic agent), 완충제, 피막제, 감미제, 용해제, 기제(base), 분산제, 습윤제, 현탁제, 안정제, 착색제, 방향제 등 각 제형에 통상적으로 사용되는 약제학적으로 허용되는 담체(carrier)를 사용하여 공지 기술에 의해 제조될 수 있다.
상기 약제의 제조에 있어서 본 발명의 리피테그라스트 결정형 또는 용매화물의 함량은 약제의 형태에 따라 다르지만, 바람직하게는 10 내지 90 중량%의 농도, 보다 바람직하게는 30 내지 80 중량%의 농도이다.
본 발명의 약제학적 조성물의 투여량은 치료받는 사람을 포함한 포유동물의 종류, 투여경로, 체중, 성별, 나이, 질환의 정도, 의사의 판단 등에 따라 넓은 범위에서 다양하게 변화된다. 일반적으로 경구투여의 경우에는 60kg 정도의 일반 성인을 기준으로 활성성분의 투여량이 1일 약 1 내지 240mg, 바람직하게는 5 내지 180mg, 보다 바람직하게는 10 내지 120mg이 투여될 수 있다. 상술한 일일 투여량은 질환의 정도, 의사의 판단 등에 따라 한 번에 또는 나누어서 사용될 수 있다.
본 발명에 따른 리피테그라스트 결정형 S형, Y형, Z형 또는 에틸렌글라이콜 용매화물은 용해도가 개선되어 제제화시 보다 우수한 생체 이용율을 나타낼 수 있다. 아울러, 본 발명에 따른 리피테그라스트 결정형 S형, Y형, Z형 또는 에틸렌글라이콜 용매화물은 열 및 수분에 대하여 안정성이 뛰어난 성질을 가지므로, 이를 함유하는 제제에서 유연물질의 발생을 억제할 수 있을 뿐만 아니라, 흡습성이 낮아 원료 보관 및 제조 공정이 편리하고 안전하다.

이하, 실시예에 의해 본 발명을 보다 구체적으로 설명하고자 한다. 이들 실시예는 오직 본 발명을 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 국한되지 않는다는 것은 당업자에게 있어서 자명하다.
비교예 1: 리피테그라스트 무정형의 제조
(S)-메틸-2-(2-(벤조퓨란-6-카보닐)-5,7-디클로로-1,2,3,4-테트라히드로아이소퀴놀린-6-카복스아미도)-3-(3-(메틸술포닐)페닐)프로피오네이트 (1 g, 1.589 mmol)를 테트라하이드로퓨란 (5 mL, 5 v/w)에 현탁시키고, 0℃로 냉각하였다. 반응액에 1 M LiOH 수용액 (1.75 mL, 1.747 mmol)을 5℃ 이하를 유지하며 천천히 적가 투입 후, 0℃에서 2시간 동안 교반하였다. 반응 종결을 HPLC로 확인하고 반응액을 실온으로 승온하였다. 반응액을 농축하고 물 (5 mL, 5 v/w), NaHCO3 포화수용액 (1 mL, 1 v/w)과 아세트산에틸 (3 mL, 3 v/w)을 적가 투입 후 30분간 교반하며 세척하였다. 수층을 분리하여 아세트산에틸 (3 mL, 3 v/w)로 한 번 더 세척하고 유기층을 분리하여 제거하였다. 수층에 남아 있을 유기층 제거를 위해 농축을 진행하고, 빠르게 교반하며 수층에 37% HCl 수용액 (3 mL, 3 v/w)을 천천히 적가하며 산성화 및 고체화를 진행하였다. pH가 1 이하로 떨어지면 여과하고, 여과된 고체는 물 (2 mL, 2 v/w)로 세척하였다. 얻어진 고체를 12시간 이상 진공 건조하여 백색의 무정형 고체인 화학식 1의 화합물 (0.83 g, 1.351 mmol, 수율 85%, 순도 98.5%)을 얻었다.
실시예 1: 리피테그라스트 결정형 S형의 제조
상기 비교예 1에서 얻어진 무정형의 리피테그라스트 (1.0 g)를 에탄올 (10 mL)에 현탁시킨 후, 가열하여 교반하였다. 이를 실온으로 냉각하여 1일 이상 교반하였다. 생성된 고체를 여과한 후, 에탄올과 물로 각각 세척하여, 순도 99.8%의 리피테그라스트 (0.80 g)를 수득하였다. 얻어진 고체의 X선 분말 회절분석, 시차주사열량 분석 및 열중량 분석을 수행하여, 그 결과를 하기 표 1, 도 1, 도 5 및 도 9에 나타내었으며, 이를 결정형 S형이라 하였다.

실시예 2: 리피테그라스트 결정형 Y형의 제조

상기 실시예 2의 결정형 Y형의 리피테그라스트를 온도 25℃ 상대습도 60% 조건에서 12시간 이상 보관하여 리피테그라스트 결정형 S형을 수득하였다.
실시예 4: 리피테그라스트 결정형 Y형의 제조
실시예 1에서 얻어진 S형 결정형의 리피테그라스트를 70℃에서 1일 이상 진공 건조하여 결정형 Y형을 수득하였다
실시예 5: 리피테그라스트 에틸렌글라이콜 용매화물의 제조
비교예 1에서 얻어진 무정형의 리피테그라스트 (1.0 g)를 에틸렌글라이콜 (10 mL)에 녹인 후, 실온에서 24 시간 정도 교반하였다. 생성된 고체를 여과하고 에틸렌글라이콜 (3 mL, 3 v/w)로 세척하였다. 얻어진 고체는 70℃에서 12시간 이상 진공 건조하여 순도 99.7%의 리피테그라스트 에틸렌글라이콜 용매화물 (0.83 g)을 수득하였다. 얻어진 고체의 X선 분말 회절분석, 시차주사열량 분석 및 열중량 분석을 수행하여, 그 결과를 하기 표 3, 도 3, 도 7 및 도 11에 나타내었다.

실시예 6: 리피테그라스트 결정형 Y형의 제조
상기 실시예 5의 리피테그라스트 에틸렌글라이콜 용매화물 (1.0 g)을 물 (10 mL, 10 v/w)에 현탁시킨 후, 실온에서 12 시간 이상 교반하였다. 충분히 교반을 진행한 후 여과하고 물 (3 mL, 3 v/w)로 세척하였다. 얻어진 고체는 70℃에서 1일 이상 진공 건조하여 순도 99% 이상의 리피테그라스트 결정형 Y형 (0.92 g)을 수득하였다.
실시예 7: 리피테그라스트 결정형 Y형의 제조
상기 실시예 5의 리피테그라스트 에틸렌글라이콜 용매화물 (1.0 g)을 에탄올 (10 mL, 10 v/w)에 현탁시킨 후 실온에서 12 시간 이상 교반하였다. 충분히 교반을 진행한 후, 여과하고 에탄올 (3 mL, 3 v/w)로 세척하였다. 얻어진 고체는 70℃에서 1일 이상 진공 건조하여, 순도 99% 이상의 리피테그라스트 결정형 Y형 (0.89 g)을 수득하였다.
실시예 8: 리피테그라스트 결정형 Y형의 제조
상기 실시예 5의 리피테그라스트 에틸렌글라이콜 용매화물 (1.0 g)을 아세톤 (10 mL, 10 v/w)에 현탁시킨 후 실온에서 12 시간 이상 교반하였다. 충분히 교반을 진행한 후 여과하고 아세톤 (3 mL, 3 v/w)으로 세척하였다. 얻어진 고체는 70℃에서 1일 이상 진공 건조하여, 순도 99% 이상의 리피테그라스트 결정형 Y형 (0.88 g)을 수득하였다.
실시예 9: 리피테그라스트 결정형 Z형의 제조
비교예 1에서 얻어진 무정형의 리피테그라스트 (1.0 g)를 에틸렌글라이콜다이메틸에터 (8 mL, 8 v/w)에 현탁시킨 후, 열을 가하여 용해시키거나 상온에서 교반한 후, 이를 냉각하여 여과하고 에틸렌글라이콜다이메틸에터 (2 mL, 2 v/w)로 세척하였다. 얻어진 고체는 70℃에서 12시간 이상 진공 건조하였다. 건조된 고체(순도 99.7%)의 X선 분말 회절분석, 시차주사열량 분석 및 열중량 분석을 수행하여, 그 결과를 하기 표 4, 도 4, 도 8 및 도 12에 나타내었으며, 이를 결정형 Z형이라 하였다.

비교예 1에서 얻어진 무정형의 리피테그라스트 (1.0 g)를 1,2-다이메톡시에테인 (10 mL, 10 v/w)에 현탁시킨 후, 열을 가하여 용해시키거나 상온에서 교반한 후, 이를 냉각하여 여과하고 1,2-다이메톡시에테인 (2 mL, 2 v/w)으로 세척하였다. 얻어진 고체는 70℃에서 12시간 이상 진공 건조하여 순도 99.7%의 리피테그라스트 결정형 Z형 (0.77 g)을 수득하였다.
실시예 11: 리피테그라스트 결정형 Z형의 제조
비교예 1에서 얻어진 무정형의 리피테그라스트 (1.0 g)를 프로피오나이트릴 (8 mL, 8 v/w)에 현탁시킨 후, 열을 가하여 용해시키거나 상온에서 교반한 후, 이를 냉각하여 여과하고 프로피오나이트릴 (2 mL, 2 v/w)로 세척하였다. 얻어진 고체는 70℃에서 12시간 이상 진공 건조하여 순도 99.7%의 리피테그라스트 결정형 Z형 (0.64 g)을 수득하였다.
비교예 2: 리피테그라스트 결정형 A형의 제조
미국 등록 특허 제8,367,701호에 기재된 제조방법에 따라 리피테그라스트 결정형 A 형 (순도 99.6%)을 얻었다.
X선 분말 회절분석
X선 분말 회절분석 (XRD)은 분말 X선 회절기를 사용하여 3~40 °2θ의 범위에서 회절 패턴을 얻었다. 분말 X선 회절분석 조건은 다음과 같다.
- 기기: Bruker A26X1 D2 Phaser
- Time per step: 0.5 s
- 주사방식: Continuous PSD fast
- 검출기: Lynxeye (1D mode)
시차주사열량 분석
시차주사열량 분석 (DSC)은 TA instrument의 Q2000 model을 사용하여 수행하였다. 약 2~3 mg의 시료를 알루미늄 팬에 넣고, 구멍 뚫린 뚜껑으로 덮어 DSC 실험에 필요한 시료를 준비하였다. 정확한 무게를 기록한 후, 질소 하에 10℃/min의 속도로 25 ~ 250℃까지 가열하였다.
열중량 분석
열중량 분석 (TGA)은 TA instrument의 Q50 model을 사용하여 수행하였다. 약 10 mg의 시료를 백금 팬에 넣어, TGA 실험에 필요한 시료를 준비하였다. 질소 하에 10℃/min의 속도로 25 ~ 600℃까지 가열하였다.
실험예 1: 열 안정성 시험
실시예에서 수득한 리피테그라스트 결정형 Y형, S형을 각각 50 mg씩 갈색 바이알에 넣고 알루미늄 파우치로 2차 포장을 한 후, 40±2℃, 75±5% RH에서 보관하였다. 4주 후에 각각의 시료를 꺼내어 결정형의 안정성 평가를 위해 분말 X선 회절분석을 수행하고 유연물질의 발생과 증가 여부를 평가하기 위해 고성능 액체크로마토그래피로 분석하였다.
분말 X선 회절분석 및 고성능 액체크로마토그래피 분석 결과를 각각 하기 표 5 및 표 6에 나타냈다.

상기 표 5 및 표 6에서 보듯이, 결정형 Y형 및 S형은 40℃/75% RH에서 4 주간 성상의 변화 및 결정형의 변화가 발생하지 않았으며 유연물질 발생 또한 관찰되지 않았다.
따라서, 리피테그라스트 신규 결정형인 결정형 Y형 및 S형이 열과 수분에 안정함을 알 수 있다.
실험예 2: 흡습성 시험
실시예에서 수득한 리피테그라스트 결정형 Y형, S형 및 비교예의 결정형 A형을 각각 동적 증기 흡착 및 탈착 장치인 DVS (dynamic vapor absorption, VTI SGA-100, TA Instruments)를 이용하여 25℃의 등온 조건 및 상대습도 5~95% 범위에서 5% RH 간격으로 흡습과 탈습을 1회 반복하여 실험하였으며 그 결과를 표 7, 도 13 및 도 14에 나타냈다.

DVS 분석 결과, 결정형 Y형 및 S형이 적은 흡습성을 나타내는 것을 확인할 수 있었다. 특히, 결정형 S형은 EP 기준(25℃, 상대습도 80%)으로 결정형 A형보다 낮은 흡습성을 나타냈다.
따라서, 상기한 열 안정성 시험과 흡습성 시험을 통해 결정형 S형이 가장 안정함을 확인하였으며, S형은 수분에 대하여 안정성이 뛰어난 성질을 가지므로, 이를 함유하는 제제에서 수분에 대한 유연물질의 발생을 억제함으로써 리피테그라스트 제제의 안정성을 크게 개선할 수 있을 뿐만 아니라, 원료 보관 및 제조 공정이 편리하고 안전함을 알 수 있다.
실험예 3: 용해도 시험
실시예에서 수득한 리피테그라스트 결정형 Y형, S형 및 비교예의 결정형 A형을 각각 물에 용해시킨 후, 실온에서 충분히 교반하여 용해도 시험을 진행하였으며, 그 결과를 표 8 및 도 15에 나타냈다.

상기 표 8에서 보듯이, 결정형 Y형 및 S형은 결정형 A형에 비해 물에서의 용해도가 두배 이상임을 확인할 수 있다.
따라서, 결정형 Y형 및 S형이 난용성 약물인 리피테그라스트의 용해도가 개선된 결정형임을 알 수 있으며, 제제화시 결정형 A형에 비해 보다 우수한 생체 이용율을 나타낼 수 있음을 알 수 있다.
Claims (9)
- X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 9.8±0.2, 10.9±0.2, 16.5±0.2, 18.0±0.2, 19.5±0.2, 20.2±0.2, 20.9±0.2, 21.8±0.2, 22.9±0.2, 23.7±0.2, 24.3±0.2, 24.4±0.2, 24.9±0.2, 26.0±0.2 및 29.7±0.2인 리피테그라스트 결정형 S형.
- 제1항에 있어서, 시차주사열량 분석에서, 첫번째 흡열점은 57.0℃, 두번째 흡열점은 164.3℃의 값을 나타내는 리피테그라스트 결정형 S형.
- X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 10.2±0.2, 10.8±0.2, 16.6±0.2, 17.1±0.2, 18.1±0.2, 18.4±0.2, 18.9±0.2, 20.3±0.2, 20.5±0.2, 21.0±0.2, 21.6±0.2, 22.8±0.2, 23.6±0.2, 24.0±0.2, 24.9±0.2, 25.9±0.2, 26.5±0.2, 28.2±0.2 및 29.3±0.2인 리피테그라스트 결정형 Y형.
- 제3항에 있어서, 시차주사열량 분석에서, 흡열점은 162.2의 값을 나타내는 리피테그라스트 결정형 Y형.
- X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 9.7±0.2, 12.6±0.2, 18.7±0.2, 20.8±0.2, 21.6±0.2, 23.4±0.2 및 24.1±0.2인 리피테그라스트 에틸렌글라이콜 용매화물.
- 제5항에 있어서, 시차주사열량 분석에서, 흡열점은 144.5℃의 값을 나타내는 리피테그라스트 에틸렌글라이콜 용매화물.
- X-선 분말 회절분석에서 I/Io(I: 각 회절각에서의 피크의 강도, I0: 가장 큰 피크의 강도)가 10% 이상인 회절각 (2θ)의 값이 7.5±0.2, 8.1±0.2, 8.7±0.2, 9.8±0.2, 10.2±0.2, 12.5±0.2, 17.5±0.2, 18.3±0.2, 18.6±0.2, 19.0±0.2, 20.3±0.2, 21.1±0.2, 21.5±0.2, 21.9±0.2, 22.2±0.2, 22.9±0.2, 23.8±0.2, 24.3±0.2, 24.8±0.2, 25.6±0.2, 26.4±0.2, 27.0±0.2, 28.5±0.2 및 34.8±0.2인 리피테그라스트 결정형 Z형.
- 제7항에 있어서, 시차주사열량 분석에서, 흡열점은 162.3℃의 값을 나타내는 리피테그라스트 결정형 Z형.
- 제1항에 따른 리피테그라스트 결정형 S형, 제3항에 따른 리피테그라스트 결정형 Y형, 제5항에 따른 리피테그라스트 에틸렌글라이콜 용매화물 및 제7항에 따른 리피테그라스트 결정형 Z형으로 구성된 군으로부터 선택되는 하나 이상의 결정형 또는 용매화물을 약제학적으로 허용되는 담체와 함께 포함하는 안구건조증을 치료하기 위한 약제학적 조성물.
