Description
본 발명은 매크로사이클릭 호스트 분자 기반의 코어-쉘 나노입자 및 이의 합성방법에 관한 것이다.
생물학적 샘플 및 기타 샘플로부터의 단일 분자에 대한 고감도의 정확한 검출은 의학 진단학, 병리학, 환경 샘플링, 화학적 분석 및 기타 많은 분야에서 널리 사용될 수 있으며, 이를 위하여 지난 수년간 생물-화학 분야에서는 소량의 합성물질 및 생체 분자의 대사, 분포 및 결합 등을 연구하는데 특정 물질로 표지된 나노입자 및 화학물질을 널리 사용해 왔다. 대표적으로는 방사성 동위원소를 이용한 방법, 유기 형광물질을 이용한 방법, 무기 물질인 양자점(Quantum dots)을 이용한 방법이 있다.
방사성 동위원소를 이용한 방법에서 방사성 표지물질(Radioactive isotope)로는 생체 내에서 널리 발견되는 1H, 12C, 31P, 32S, 127I 등의 방사성동위체인 3H, 14C, 32P, 35S, 125I 등이 널리 사용된다. 방사성 동위원소들은 비 방사성의 동위체와 화학적 성질이 거의 비슷하여 임의로 치환이 가능하고, 방출 에너지가 비교적 커서 소량의 검출도 가능하다는 장점이 있기 때문에 오랫동안 사용되어 왔다. 그러나, 인체에 해로운 방사선 때문에 다루기가 용이하지 않고, 일부 동위원소의 방사성은 방출에너지가 큰 대신 반감기가 짧아서 장시간의 보관이나 실험에 불편한 단점도 있다.
방사성 동위원소에 대한 대안으로 널리 사용되는 것은 유기 형광물질(Organic fluorescent dyes)이다. 형광물질들은 특정 파장에 의해서 활성이 되면 고유의 파장을 갖는 빛을 발광하게 된다. 특히, 검색법이 소형화됨에 따라, 방사성 물질 역시 검출 한계를 나타내어 검색에 오랜 시간이 요구된다. 이에 비해 형광물질의 경우 적절한 조건에서 분자당 수천 개의 광자를 방출할 수 있어 단일분자 수준의 검출까지도 이론적으로 가능하다. 그러나, 방사성 동위원소처럼 활성 리간드의 원소를 직접 치환할 수는 없고, 구조 활성관계를 통해 비교적 활성에 영향을 주지 않는 부분을 변형하여 형광물질을 연결해야 하는 제한이 있다. 또한, 이러한 형광 표지물질들은 시간이 지나면서 형광 세기가 약해지며(photobleaching), 활성을 시키는 빛의 파장이 매우 좁고 발광되는 빛의 파장이 매우 넓어 서로 다른 형광 물질 간에 간섭이 있는 단점을 가지고 있다. 또한, 사용할 수 있는 형광물질의 수가 극히 제한되어 있다.
또한, 반도체 나노 물질인 양자점은 CdSe, CdS, ZnS, ZnSe 등으로 구성되어 있으며 크기 및 종류에 따라서 각각 다른 색의 빛을 발광한다. 유기 형광물질에 비하여 넓은 활성 파장을 가지고 있으며 좁은 발광 파장을 나타내기 때문에 다른 색을 발광하는 가짓수가 유기 형광물질보다 많다. 따라서, 최근 들어 유기 형광물질의 단점들을 극복하기 위한 방법으로 양자점이 많이 사용되고 있다. 그러나, 독성이 강하고, 대량 생산이 힘들다는 단점이 있다. 또한, 이론적으로 그 가지 수는 다양하나 실제적으로 사용되고 있는 수는 극히 제한되어 있다.
이러한 문제점들을 해결하기 위한 방법으로 최근에는 라만 분광학 및/또는 표면 플라스몬 공명을 이용한 표지물질을 이용하고 있다. 그 중에서도, 표면 증강 라만 산란법(Surface Enhanced Raman Scattering, SERS)는 금, 은 등의 금속 나노구조의 거친(roughened) 표면에 분자가 흡착될 때 라만 산란의 세기가 106 ~ 108배 이상 급격히 증가되는 현상을 이용한 분광법이다. 빛을 유형 매질에 통과시키는 경우 어느 정도의 양은 고유 방향에서 벗어나는데, 이러한 현상은 라만 산란으로 알려져 있다. 산란된 광 중 일부가 광의 흡수 및 전자의 높은 에너지 준위로 여기함에 따라 고유의 자극된 광과 진동수가 상이하며, 라만 방출 스펙트럼의 파장은 샘플 내의 광 흡수 분자의 화학 조성 및 구조 특성을 나타내므로, 라만 분광법은 현재 아주 빠른 속도로 발전하고 있는 나노 기술과 결합하여 단 하나의 분자를 직접 측정할 수 있는 고감도의 기술로 발전가능하며, 특히 메디컬 센서로서 긴요하게 쓰일 수 있을 것으로 많은 기대를 받고 있다. 이 표면 증강 라만 분광법(SERS) 효과는 플라스몬 공명의 현상과 관련되며, 여기서 금속 나노입자는 금속 내 전도 전자의 집단 커플링으로 인해 입사 전자기 방사선에 응답하여 뚜렷한 광학적 공명을 나타내므로, 본질적으로 금, 은, 구리 및 다른 특정 금속의 나노입자들은 전자기 방사선의 집중화 효과를 향상시키는 소형 안테나로서 작용할 수 있다. 이러한 입자 부근에 위치한 분자는 라만 분광법 분석에 대해 훨씬 큰 감도를 나타낸다.
따라서, SERS 센서를 이용하여 다양한 질병과 관련된 유전자, 단백질(바이오 마커)의 조기 진단을 수행하려는 연구가 활발히 진행되고 있다. 라만 분광법은 다른 분석법(적외선 분광법) 방법과는 달리 여러 가지 장점을 가지고 있다. 적외선 분광법은 분자의 쌍극자 모멘트의 변화가 있는 분자의 경우 강한 신호를 얻을수 있는 반면, 라만 분광법은 분자의 유도 편극률 변화가 있는 비극성 분자의 경우에도 강한 신호를 얻을 수 있으므로, 거의 모든 유기 분자들이 고유의 라만 시프트(Raman Shift, cm-1)를 가지고 있다. 또한 물 분자에 의한 간섭의 영향을 받지 않으므로, 단백질, 유전자 등의 생체분자(biomolecules)의 검출에 더욱 적합하다. 하지만 낮은 신호 세기로 인하여 오랜 연구 기간에도 불구하고 실용화되는 수준에 이르지는 못하였다.
본 발명은 높은 수율로 균일한 나노갭을 포함하는 나노입자를 합성할 수 있는 방법 제공을 목적으로 한다.
또한, 본 발명은 초고감도의 센싱 및 이미징이 가능한 나노입자를 제공하는 것을 목적으로 한다.
본 발명의 일 실시예에 따르면, 코어 구조체; 및 상기 코어 구조체를 커버하며 상기 코어 구조체와 나노갭을 두고 이격되어 제공되는 쉘 구조체를 포함하고, 상기 코어 구조체의 표면에는 분자 내부에서 소수성을 나타내고 분자 외부에서 친수성을 나타내는 매크로사이클릭 호스트 분자 및 상기 매크로사이클릭 호스트 분자의 분자 내부에 삽입된 라만활성물질이 제공되고, 상기 매크로사이클릭 호스트 분자 및 상기 라만활성물질은 상기 나노갭을 채우는, 나노입자가 제공된다.
본 발명의 일 실시예에 따르면, 상기 매크로사이클릭 호스트 분자는 사이클로덱스트린(cyclodextrin), 쿠커비투릴(cucurbituril), 칼리싸레인(calixarene), 및 필라렌(pillararene) 등으로 이루어진 군에서 선택된 적어도 하나의 화합물 및 이의 유도체를 포함하는, 나노입자 가 제공된다.
본 발명의 일 실시예에 따르면, 상기 매크로사이클릭 호스트 분자와 상기 라만활성물질은 비공유 결합으로 결합되는, 나노입자가 제공된다.
본 발명의 일 실시예에 따르면, 상기 코어 구조체 및 상기 쉘 구조체는 금, 은, 및 구리로 이루어진 군에서 선택된 적어도 하나의 물질을 포함하는, 나노입자가 제공된다.
본 발명의 일 실시예에 따르면, 상기 나노갭의 크기는 0.1 nm 내지 10 nm인, 나노입자가 제공된다.
본 발명의 일 실시예에 따르면, 코어 구조체 표면에 매크로사이클릭 호스트 분자를 부착하여 개질 코어 구조체를 형성하는 제1 단계; 상기 개질 코어 구조체와 라만활성물질을 혼합하여 상기 매크로사이클릭 호스트 분자의 분자 내부에 상기 라만활성물질을 삽입하는 제2 단계; 및 상기 라만활성물질이 삽입된 상기 개질 코어 구조체 표면에 쉘 구조체를 합성하는 제3 단계를 포함하고, 상기 코어 구조체와 상기 쉘 구조체 사이에는 상기 매크로사이클릭 호스트 분자 및 상기 라만활성물질로 채워진 나노갭이 제공되는, 나노입자 합성 방법이 제공된다.
본 발명의 일 실시예에 따르면, 상기 제1 단계는 상기 코어 구조체와 상기 매크로사이클릭 호스트 분자를 혼합하고 인큐베이션하여 수행되고, 상기 매크로사이클릭 호스트 분자는 사이클로덱스트린(cyclodextrin), 쿠커비투릴(cucurbituril), 칼리싸레인(calixarene), 및 필라렌(pillararene) 등으로 이루어진 군에서 선택된 적어도 하나의 화합물 및 이의 유도체를 포함하는, 나노입자 합성 방법이 제공된다.
본 발명의 일 실시예에 따르면, 상기 제1 단계는 세틸트리메틸암모늄 브로마이드(CTAB, Cetyltrimethylammonium bromide) 캡핑된 코어 구조체를 제공하는 단계; 및 상기 세틸트리메틸암모늄 브로마이드 캡핑된 코어 구조체를 L-아스코르브산(AA) 및 HAuCl4와 반응시켜 세틸트리메틸암모늄 클로라이드(CTAC, Cetyltrimethylammonium chloride) 캡핑된 코어 구조체를 제공하는 단계를 더 포함하고, 상기 세틸트리메틸암모늄 클로라이드 캡핑된 코어 구조체 상에 매크로사이클릭 호스트 분자를 부착하여 개질 코어 구조체를 형성하는, 나노입자 합성 방법이 제공된다.
본 발명의 나노입자 합성 방법에 따르면 균일하면서도 고 수율로 내부갭을 형성할 수 있으며, 나노갭에 호스트-게스트 상호작용을 이용해 여러 가지 라만활성물질 도입이 가능하다.
본 발명의 나노입자는 강하면서도 좁은 SERS 향상 계수(EF, Enhancement Factor)분포를 나타내고, 이를 이용해 초고감도의 센싱 및 이미징이 가능하다. 나아가 서로 다르고 구분 가능한 SERS 신호를 이용해 여러 가지 검지물질을 동시에 검지 가능하다.

본 발명은 다양한 변경을 가할 수 있고 여러 가지 형태를 가질 수 있는 바, 특정 실시예들을 도면에 예시하고 본문에 상세하게 설명하고자 한다. 그러나, 이는 본 발명을 특정한 개시 형태에 대해 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변경, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다.
본 발명의 일 실시예에 따르면 코어-쉘 구조를 갖는 나노입자에 대하여, 코어 구조체 표면에 매크로사이클릭 호스트 분자가 제공되고 매크로사이클릭 호스트 분자 내에 라만활성물질이 비공유 결합으로 삽입됨으로써 안정적인 구조를 갖고 분광 성능이 우수한 나노입자가 제공될 수 있다.

도 1을 참고하면, 나노입자는 코어 구조체(100), 코어 구조체(100)를 커버하며 코어 구조체(100)와 나노갭을 두고 이격되어 제공되는 쉘 구조체(200), 코어 구조체(100)의 표면에는 분자 내부에서 소수성을 나타내고 분자 외부에서 친수성을 나타내는 매크로사이클릭 호스트 분자(310) 및 매크로사이클릭 호스트 분자(310)의 분자 내부에 삽입된 라만활성물질(320)이 제공되고, 매크로사이클릭 호스트 분자(310) 및 라만활성물질(320)은 상기 나노갭을 채우는 형태로 제공된다.
코어 구조체(100)는 나노입자의 중심부에 제공되며, 쉘 구조체(200), 매크로사이클릭 호스트 분자(310), 라만활성물질(320) 등이 제공되는 기재로 기능할 수 있다. 코어 구조체(100)는 구, 타원구, 다면체(예를 들어, 정육면체, 직육면체) 등의 형상을 가질 수 있다.
코어 구조체(100)는 약 1 nm 내지 약 100 nm의 평균 직경을 가질 수 있다. 코어 구조체(100)의 평균 직경은 코어 구조체(100)가 방향에 따라 다른 직경을 가질 때, 전체 직경의 평균을 의미할 수 있다.
코어 구조체(100)는 금, 은, 및 구리로 이루어진 군에서 선택된 적어도 하나의 물질을 포함할 수 있다. 경우에 따라 코어 구조체(100)는 금으로 구성될 수 있다. 금은 화학적으로 안정하기 때문에 물질 검지에 사용하기 적합하다. 또한, 이하에서 살펴볼 것과 같이 매크로사이클릭 호스트 분자(310)를 표면에 용이하게 부착할 수 있다는 장점을 갖는다.
코어 구조체(100)와 이격되어 쉘 구조체(200)가 제공된다.
쉘 구조체(200)는 코어 구조체(100)를 커버하는 형태로 제공된다. 쉘 구조체(200)는 코어 구조체(100)가 내부에 제공될 수 있도록 안쪽에 빈 공간을 포함할 수 있다. 쉘 구조체(200)의 내부 공간의 형태는 코어 구조체(100)의 형태와 대응될 수 있다. 예를 들어, 코어 구조체(100)가 구 형태를 갖는 경우, 쉘 구조체(200)의 내부 공간도 구 형태를 가질 수 있다. 이때, 쉘 구조체(200)는 코어 구조체(100)를 커버해야 하므로, 쉘 구조체(200) 내부 공간의 직경은 코어 구조체(100)보다 클 수 있다.
쉘 구조체(200)의 외주면은 쉘 구조체(200)의 내부 공간(내면)과 다른 형태를 가질 수 있다. 예를 들어, 쉘 구조체(200)의 내부 공간은 구 형태를 갖는 것에 비해, 외주면은 육면체, 팔면체 등 다면체 형태를 가질 수 있다. 쉘 구조체(200)의 내부 공간은 코어 구조체(100)의 형태에 대응되어 달라질 수 있는 것에 비하여, 쉘 구조체(200)의 외주면은 쉘 구조체(200)를 형성하기 위한 전구체 입자의 응집 양상 및 전구체 입자의 응집에 따라 형성된 나노브릿지의 병합에 따라 형태가 달라질 수 있다.
쉘 구조체(200)는 코어 구조체(100) 표면에 제공된 매크로사이클릭 호스트 분자(310) 및 라만활성물질(320)을 보호할 수 있다. 구체적으로, 매크로사이클릭 호스트 분자(310) 및 라만활성물질(320)은 코어 구조체(100)와 쉘 구조체(200) 사이에 제공되고, 쉘 구조체(200)는 안쪽에 제공된 매크로사이클릭 호스트 분자(310) 및 라만활성물질(320)이 나노입자 외부의 화합물과 반응하여 변성되는 것을 막을 수 있다. 따라서, 나노입자를 이용하여 보다 안정적으로 물질 검지를 수행할 수 있다.
쉘 구조체(200)는 금, 은, 및 구리로 이루어진 군에서 선택된 적어도 하나의 물질을 포함할 수 있다. 경우에 따라 쉘 구조체(200)는 금으로 구성될 수 있다. 금은 화학적으로 안정하기 때문에 물질 검지에 사용하기 적합하다.
쉘 구조체(200)와 코어 구조체(100) 사이에는 나노갭이 제공된다. 나노갭은 내부에서 증폭된 전자기장 집속/구속에 의해 라만, 형광, nonlinear optics, IR 흡수 등 여러 광학신호의 크기를 키운다. 나노갭에서 증폭되는 신호 중 라만신호는 분자의 진동 모드부터 유래된 비탄성 산란신호로서, 신호의 세기가 매우 약하지만, 상대적으로 작은 반치폭을 갖고 있기 때문에 형광 등의 넓은 피크를 갖는 신호보다 복합적 신호에 대한 구분이 용이하며 따라서 다중검지에 유용하다. 플라즈몬성 나노갭 구조물을 활용하면, 약한 라만 신호를 상당히 증폭 시킬 수 있고 이를 SERS라 한다.
그러나 이러한 나노갭 내부에서의 강한 전자기장 증폭/집속 때문에, 나노갭의 크기나 모양 그리고 분자의 위치 등의 작은 변화에도 광학신호는 민감하게 변한다. 따라서 1 nm 수준의 매우 작은 나노갭을 균일하며 재현성있고 고수율로 합성 할 수 있을 뿐 아니라, 원하는 광학활성물질을 나노갭내부에 위치시킬 수 있는 전략은 재현성 있는 광학신호 구현에 반드시 필요하다.
나노갭의 크기는 0.1 nm 내지 10 nm일 수 있다. 상술한 크기의 나노갭이 제공됨으로써 라만신호가 증폭될 수 있다. 나노갭에 의하여 완전히 접촉되지 않고 코어 구조체(100)와 쉘 구조체(200)가 구분될 수 있다. 일부 영역에서는 코어 구조체(100)와 쉘 구조체(200)가 나노브릿지에 의하여 접촉되어 있을 수 있다. 따라서, 본 발명에서 사용되는 "나노갭"이 반드시 코어 구조체(100)와 쉘 구조체(200) 사이를 완전하게 분리하는 공간을 의미하는 것은 아니다.
나노갭 내부에는 매크로사이클릭 호스트 분자(310) 및 라만활성물질(320)이 제공될 수 있다.
매크로사이클릭 호스트 분자(310)는 코어 구조체(100) 표면에 제공되며, 라만활성물질(320)을 나노갭 내에 고정한다. 매크로사이클릭 호스트 분자(310)는 고리 형태를 가지며, 분자 내부에서 소수성을 나타내고 분자 외부에서 친수성을 나타내는 물질일 수 있다. 따라서 매크로사이클릭 호스트 분자(310)의 고리 내부는 소수성을 나타내고, 고리 외부는 친수성을 나타낼 수 있다. 따라서, 소수성을 나타내는 물질은 매크로사이클릭 호스트 분자(310)의 고리 내부에 삽입된 후에는 소수성을 나타내는 고리 내부와 비-공유 결합(non-covalent bonding)을 형성하고, 친수성을 나타내는 고리 외부와 사이에 척력의 영향을 받을 수 있다. 이에 따라, 한번 매크로사이클릭 호스트 분자(310) 내부에 삽입된 이후에는 삽입된 물질이 쉽게 매크로사이클릭 호스트 분자(310) 밖으로 빠져나가지 않고, 안정적으로 고정될 수 있다. 이러한 원리를 이용하여 매크로사이클릭 호스트 분자(310) 내부에 라만활성물질(320)을 고정할 수 있다.
매크로사이클릭 호스트 분자(310)는 사이클로덱스트린(cyclodextrin), 쿠커비투릴(cucurbituril), 칼리싸레인(calixarene), 및 필라렌(pillararene) 등으로 이루어진 군에서 선택된 적어도 하나의 화합물 및 이의 유도체를 포함할 수 있다. 예를 들어, 매크로사이클릭 호스트 분자(310)는 모노-(6-메르캅토-6-디옥시)-β-사이클로덱스트린(Mono-(6-Mercapto-Deoxy)-β-Cyclodextrin)일 수 있다.
라만활성물질(320)은 분석물에 부착되었을 때 라만 검출 장치에 의한 분석물의 검출 및 측정을 용이하게 하는 물질을 의미한다. 라만 분광법에 사용될 수 있는 라만활성물질(320)은 유기 원자, 분자 또는 무기 원자, 분자 등을 포함한다. 구체적으로, 라만활성물질(320)의 예로서, FAM, Dabcyl, TRITC(테트라메틸 로다민-5-아이소티오시아네이트), MGITC(말라키트 그린 아이소티오시아네이트), XRITC(X-로다민-5-아이소티오시아네이트), DTDC(3,3-디에틸티아디카보시아닌 아이오다이드), TRIT(테트라메틸 로다민 아이소티올), NBD(7-니트로벤즈-2-1,3-다이아졸), 프탈산, 테레프탈산, 아이소프탈산, 파라-아미노벤조산, 에리트로신, 비오틴, 다이곡시게닌(digoxigenin), 5-카복시-4',5'-다이클로로-2',7'-다이메톡시, 플루오레세인, 5-카복시-2',4',5',7'-테트라클로로플루오레세인, 5-카복시플루오레세인, 5-카복시로다민, 6-카복시로다민, 6-카복시테트라메틸 아미노 프탈로시아닌, 아조메틴, 시아닌(Cy3, Cy3.5, Cy5), 크산틴, 석신일플루오레세인, 아미노아크리딘, 양자점, 탄소동소체, 시아나이드, 티올, 클로린, 브롬, 메틸, 인 또는 황 등이 있으나 이에 제한되지 않는다.
라만활성물질(320)은 뚜렷한 라만 스펙트럼을 나타내어야 하고 서로 다른 유형의 분석물과 특히 결합하거나 연관될 수 있는 물질일 수 있다. 바람직하게는 라만 분석 시 사용하는 여기 레이저 파장과 공명하여 더욱 높은 라만 신호 세기를 나타내는 분자들이 라만활성물질(320)로 사용될 수 있다.
라만활성물질(320)은 상술한 매크로사이클릭 호스트 분자(310) 내에 삽입되어 제공된다. 재현성있고 강한 SERS 신호를 위해선, 나노갭 구조물을 균일하게 합성하는 것뿐만 아니라 나노갭에 라만활성물질(320)을 위치시키는 전략 또한 중요하다. 추가적으로 다중 검지를 위해서는 여러 가지 서로 다른 라만활성물질(320)을 나노갭에 도입할 수 있어야 하고, 이들의 신호가 구분이 가능해야 한다.
본 발명의 일 실시예에 따르면, 코어 구조체(100) 표면에 매크로사이클릭 호스트 분자(310)가 제공되고, 매크로사이클릭 호스트 분자(310) 내부에 라만활성물질(320)이 삽입되어 제공된다. 매크로사이클릭 호스트 분자(310)가 균일하게 제공되기 때문에, 매크로사이클릭 호스트 분자(310)를 커버하는 쉘 구조체(200) 역시 균일한 나노갭을 포함하는 형태로 제공될 수 있다. 따라서, 라만 스펙트럼에 영향을 미치는 나노갭의 크기, 균일성, 라만활성물질(320) 제공 위치의 균일성이 모두 담보되는 바, 나노입자는 뚜렷한 라만 신호를 나타낼 수 있다.
이상에서는 본 발명의 일 실시예에 따른 나노입자의 구조 및 조성에 대하여 살펴보았다. 이하에서는 나노입자 합성 방법에 대하여 더 자세히 살펴보고자 한다.

도 2를 참고하면, 나노입자 합성 방법은 코어 구조체 표면에 매크로사이클릭 호스트 분자를 부착하여 개질 코어 구조체를 형성하는 제1 단계(S100), 개질 코어 구조체와 라만활성물질을 혼합하여 매크로사이클릭 호스트 분자의 분자 내부에 라만활성물질을 삽입하는 제2 단계(S200), 및 라만활성물질이 삽입된 개질 코어 구조체 표면에 쉘 구조체를 합성하는 제3 단계(S300)를 포함하고, 코어 구조체와 쉘 구조체 사이에는 매크로사이클릭 호스트 분자 및 라만활성물질로 채워진 나노갭이 제공된다.
먼저 제1 단계(S100)는 코어 구조체를 준비하고, 준비된 코어 구조체와 매크로사이클릭 호스트 분자를 혼합하고 인큐베이션시킴으로써 수행될 수 있다. 아울러, 제1 단계(S100)는 세틸트리메틸암모늄 브로마이드(CTAB, Cetyltrimethylammonium bromide) 캡핑된 코어 구조체를 제공하는 단계; 및 세틸트리메틸암모늄 브로마이드 캡핑된 코어 구조체를 L-아스코르브산(AA) 및 HAuCl4와 반응시켜 세틸트리메틸암모늄 클로라이드(CTAC, Cetyltrimethylammonium chloride) 캡핑된 코어 구조체를 제공하는 단계를 더 포함할 수 있다. 상술한 방법을 포함하여 제1 단계(S100)를 수행함으로써 코어 구조체 상에 균일하게 매크로사이클릭 호스트 분자를 결합시킬 수 있다. 이에 따라 이후 공정에서 나노갭이 균일하게 형성된 나노입자를 형성할 수 있다.
다음으로, 제2 단계(S200)는 개질 코어 구조체와 라만활성물질을 혼합하여 매크로사이클릭 호스트 분자의 분자 내부에 라만활성물질을 삽입하는 단계이다. 제2 단계(S200)에서 개질 코어 구조체 상의 매크로사이클릭 호스트 분자와 라만활성물질은 비공유 결합으로 결합한다.
종래 기술에 따르면, 코어 구조체에 라만활성물질을 제공하기 위해서는 올리고뉴클레오타이드와 같은 물질에 라만활성물질을 결합시킨 후, 라만활성물질과 결합한 올리고뉴클레오타이드를 코어 구조체에 결합시켰다. 그러나, 이러한 방법은 공유 결합에 의해 결합되어 있는 올리고뉴클레오타이드-라만활성물질 복합체를 하나하나 합성해야 하기 때문에 제조 방법이 무척 번거롭고 제조 수율도 낮다는 문제가 있었다.
종래기술에 따르면 또한, 올리고뉴클레오타이드와 라만활성물질을 반응시켜도 100% 수율로 올리고뉴클레오타이드-라만활성물질 복합체가 제조되는 것이 아니고, 반응 결과물로부터 올리고뉴클레오타이드-라만활성물질 복합체만 선택적으로 분리하는 것도 어렵기 때문에, 이후 올리고뉴클레오타이드-라만활성물질 복합체를 코어 구조체 표면에서 개질하는 과정에서 라만활성물질이 없는 올리고뉴클레오타이드가 다수 코어 구조체에 결합될 수 있다는 문제가 있었다. 이러한 불균일한 개질 또는 결합은 재현성있고 강한 SERS 신호를 만들어내는데 걸림돌이 되었다.
본 발명의 일 실시예에 따르면 매크로사이클릭 호스트 분자와 라만활성물질을 비공유 결합으로 반응기 내에서 쉽게 그리고 높은 효율로 결합시킬 수 있다. 이들은 종래 기술과 같이 공유 결합으로 하나씩 반응해서 생성되는 것이 아니기 때문에 상대적으로 쉽게 높은 수율로 매크로사이클릭 호스트 분자-라만활성물질 결합체가 제공될 수 있다. 또한, 제2 단계(S200)에 따르면 종래기술과 같이 라만활성물질이 결합된 올리고뉴클레오타이드를 코어 구조체에 부착하는 것이 아니라, 매크로사이클릭 호스트 분자를 코어 구조체에 부착한 후, 부착된 매크로사이클릭 호스트 분자에 라만활성물질을 삽입하는 것이기 때문에 보다 균일한 개질/라만활성물질 삽입이 가능하다.
다음으로, 제3 단계(S300)에서는 라만활성물질이 삽입된 개질 코어 구조체 표면에 쉘 구조체를 합성한다. 이때 쉘 구조체는 코어 구조체 표면에 균일하게 부착된 매크로사이클릭 호스트 분자 상에 형성되기 때문에 자연스럽게 코어 구조체와 쉘 구조체 사이의 나노갭이 균일하게 형성될 수 있다.
제3 단계(S300)에서 쉘 구조체는 전구체 분자의 응집으로 자라난 나노 브릿지들이 서로 병합되면서 형성될 수 있다.
이상에서는 본 발명의 일 실시예에 따른 나노입자 합성 방법에 대하여 살펴보았다. 이하에서는 실시예와 비교예 데이터를 이용하여 본 발명에 따른 나노입자의 우수한 효과에 대하여 더 자세히 살펴보고자 한다.
본 발명의 일 실시예에 따르면, 사이클로덱스트린(CD) 기반의 플라스모닉 나노갭 입자(CIPs)를 Au 코어 구조체 상의 Au 쉘 구조체 성장 동안에 사이클로덱스트린을 내부 나노갭 형성 촉진 중간층 분자로 사용하여, 설계하고 합성하였다. 이때 다양한 라만활성물질들이 게스트 분자로 사용되었고, 사이클로덱스트린은 이들에 대한 호스트 분자로 사용되었다. 합성된 나노입자는 입자 내부의 약 1 nm의 균일한 나노갭을 나타냈고, 합성 수율은 약 97 %였다. 나노입자는 좁은 분포에서 강력한 SERS EF(Enhancement Factor) 값으로 강력하고 안정적인 SERS 신호를 생성했다. 또한, Au 쉘 구조체 성장 전에 염료 용액을 추가함으로써 간단하게 10가지 다른 라만활성물질을 나노입자 내에 도입할 수 있었으며, 동일한 용액에서 10개의 고유한 SERS 스펙트럼을 표시할 수 있었는 바 다중검지에 대한 가능성을 확인했다.
실험예 1. Au 코어-쉘 나노갭 나노입자 합성
Au 코어-쉘 나노갭 나노입자를 합성하기 위하여, 다음과 같은 물질을 준비했다.
L-아스코르브산(AA), 염화금삼수화물(HAuCl4·3H2O), 크리스탈 바이올렛(CV), 로다민 B(RB), 메틸렌 블루(MB), 사프라닌 O(SO), 염기성 푹신(BF), 브릴리언트 블루 G(BBG), 나일 블루 A(NBA), 브로모 페놀 블루(BPB), L-글루타티온 환원체, 소듐 도데실설페이트(SDS), 세틸트리메틸암모늄 브로마이드(CTAB), N-(3-디메틸아미노 프로필)-N'-에틸카보디이미드 염산염(EDC), N-하이드록시설포숙 신이미드 소듐염(Sulfo-NHS) 및 2-(N-모르폴리노)에탄설폰산(MES)은 Sigma-Aldrich에서 구입했다. 에티듐 브로마이드(EtBr) 및 Pyronin Y(PYY)는 Thermo Fisher Scientific에서 구입했다. 모노-(6-메르캅토-6-디옥시)-β-사이클로덱스트린(Mono-(6-mercapto-6-deoxy)-β-cyclodextrin(CD))은 Zhiyuan Biotechnology에서 구입했다. 소듐 보로하이드라이드는 Alfa Aesar에서 구입했다. 세틸트리메틸암모늄 클로라이드(Cetyltrimethylammonium chloride (CTAC))는 Tokyo Chemical Industry (TCI)에서 구입했다. Cyclo (Arg-Gly-Asp-D-Phe-Lys) (cRGDyK, cRGD) 펩티드는 Peptides International, Inc. (Louisville, KY, USA)에서 구입했다. 카르복시 메틸-PEG-티올 (CM-PEG-SH, Mw≒5000)은 Laysan Bio, Inc. (Arab, AL, USA)에서 구입했다. N,N-다이메틸포르마이드(N,N-Dimethylformamide (DMF)) 및 다이메틸 설폭사이드(dimethyl sulfoxide (DMSO))는 Samchun Chemicals에서 구입했다. 탈 이온수 (DIW; Milli-Q,> 18.0 MΩ)는 모든 실험에 사용되었다. 모든 화학 물질은 추가 정제없이 사용되었다.
실험예 1-1. 50 nm Au 코어 구조체(AuNS) 합성
다음과 같은 절차에 따라 CTAC-캡핑된 50 nm 크기의 Au 코어 구조체(AuNS)를 합성했다. 먼저, CTAB-캡핑된 Au 클러스터가 준비되었다. 차가운 10 mM NaBH4의 용액 600 μL를 HAuCl4(0.25 mM) 및 CTAB(100 mM) 수용액 10 mL에 빠르게 첨가하고 교반했다(500 rpm). 혼합물에서 NaBH4의 완전한 분해를 보장하기 위해 혼합물을 27 ℃에서 2시간 동안 두었다. 그 후, CTAC (200 mM, 2 mL), L-아스코르브산(AA) (100 mM, 1.5 mL)를 포함하는 수용액에 HAuCl4 수용액(0.5 mM, 2 mL) 및 CTAB-캡핑된 Au 클러스터(50 μL)를 원샷 주입하여 CTAC 캡핑 ~ 10 nm AuNS를 합성했다. 반응은 27℃에서 15분간 계속되었다. 생성물을 15000 rpm에서 30 분 동안 원심 분리하여 수집 한 다음 1 mM CTAC 용액으로 2 회 세척하고 최종적으로 1 mL의 20 mM CTAC 수용액에 분산시켰다. 마지막으로 주사기 펌프를 이용하여 교반 하에 HAuCl4 수용액(0.5 mM, 2 mL)을 CTAC(100 mM, 2mL), AA(8 mM, 130 μL) 및 10 nm AuNS (6 μL)를 포함하는 수용액에 첨가함으로써 CTAC-캡핑된 50 nm AuNS를 합성하였다. 주입이 완료된 후 생성물을 27 ℃에서 10 분 동안 두었다. 생성물을 원심 분리로 수득한 후 SDS 용액 및 CTAC 용액을 이용하여 세척하였다.
실험예 1-2. 사이클로덱스트린 개질된 50 nm Au 코어 구조체(CD-AuNS) 합성
CD-AuNS를 준비하기 위해 50 nm AuNS의 표면에 모노-(6-메르캅토-6-디옥시)-β-사이클로덱스트린(CD) 분자를 부착했다. DMSO에 용해된 10 μL의 50 mM CD를 0.1% SDS 용액에서 1 mL의 100 pM AuNS와 혼합했다. 용액을 60 ℃에서 18 시간 동안 인큐베이션하였다. 생성물 용액을 원심 분리(7500 rpm, 3 분)로 수집하고 0.1% SDS 용액에 3회 분산시키고 마지막으로 추가 사용을 위해 0.01% SDS 용액에 분산시켰다.
실험예 1-3. 내부 나노갭을 포함하는 사이클로덱스트린 기반 나노입자 합성
사이클로덱스트린 기반 나노입자(CIP)는 라만활성물질 존재 하에 CD-AuNS에 Au 쉘 구조체를 형성하여 합성되었다. 이를 위하여 먼저 100 μL의 50 pM CD-AuNS 용액과 20 μL의 100 mM CTAC 용액을 혼합하였다. 다음으로, DMF에 용해된 2 μL의 10 mM 크리스탈 바이올렛(CV)을 첨가하면서 혼합하였다. 다음으로, 25 μL의 40 mM L-아스코르브산(AA) 용액과 25 μL의 5 mM HAuCl4를 혼합물에 순차적으로 첨가하였다.
생성된 혼합물을 실온에서 30 분 동안 인큐베이션했다. 생성물을 원심 분리(6000 rpm, 3분)로 수집한 후 1 mM CTAC 용액으로 4 회 세척하고 50 μL 1 mM CTAC 용액에 분산시켰다. 다음으로, 1 mM 글루타티온 용액 5 μL를 100 pM 나노입자 용액 50 μL에 첨가하여 표면에 흡착된 라만활성물질의 탈착을 유도했다. 모든 SERS 측정은 표면에 흡착된 라만활성물질의 영향을 배제하기 위해 글루타티온 처리 후 수행되었다. 다른 라만활성물질을 포함하는 나노입자들 역시 라만활성물질 용액의 부피와 용매만 달리하며 위와 동일하게 합성하였다. 각 나노입자들 합성에 사용된 라만활성물질 용액의 부피와 용매는 아래의 표 1과 같다.

이하는 실험예 1-1 내지 1-3과 관련하여, 합성된 나노입자 및 중간체의 구조와 물성을 분석한 결과이다.도 3은 본 발명의 일 실시예에 따른 매크로사이클릭 호스트 분자 기반 나노입자 합성과 분석 결과를 나타낸 그래프 및 이미지이다. 도 4는 50 nm 크기 코어 구조체의 TEM 이미지로, 축척은 100 nm이다. 도 5는 나노입자의 HAADF-STEM이미지로, 축척은 200 nm 이다.
도 3의 (a)는 내부 나노갭에 다양한 라만활성물질을 포함하는 나노입자의 합성을 모식적으로 나타낸 것이다. 도 3의 (b)는 아스코르브 산(AA) 및 HAuCl4를 첨가하고 각각 5 초, 20 초, 및 60 초가 지난 후의 중간체 구조를 나타낸 것으로, 각각의 구조의 확대된 TEM 이미지를 보여준다. 도 3의 (c)는 중간체 및 나노입자의 UV-vis 스펙트럼을 나타낸 것이고, 도 3의 (d)는 나노입자의 나노갭, 쉘 구조체, 및 코어 구조체의 크기 분포를 나타낸 것이다.
도 3의 (a)와 도 4를 참고하면, 앞서 살펴본 것과 같이 모노-(6-메르캅토-6-디옥시)-β-사이클로덱스트린로 개질된 Au 코어 구조체 상에 Au 쉘 구조체를 형성하는 합성 방법은 코어 구조체, 세틸트리메틸암모늄 클로라이드(CTAC) 및 크리스탈 바이올렛(CV)을 포함하는 용액 상에 L-아스코르브 산(AA)을 도입함으로써 개시되었다. 이로부터 생성된 물질은 CV로 코딩된 나노입자(CV-CIPs)이다.
도 3의 (b)를 참고하면, 투과전자현미경(TEM) 이미지에서 볼 수 있듯이 코어 표면에서 작은 Au 버딩(budding) 구조가 성장하고 서로 합쳐져 연속적인 쉘 구조체를 형성하는데, 동시에 쉘 구조체 안쪽에서 내부갭이 형성된다.
도 3의 (c)를 참고하면, 반응이 진행됨에 따라 구조가 변형되는 것을 UV-vis 스펙트럼을 통해 확인할 수 있다.
또한 도 3의 (d)와 도 5를 참고하면, 고각 환형 암시야 주사 투과 전자 현미경(HAAF-STEM) 이미지로부터 97 % 이상의 구조체에서 약 1 nm의 균일한 내부 나노갭이 형성되는 것을 관측할 수 있다.
도 6은 실시예와 비교예 나노입자의 HAADF-STEM이미지이다. 도 7은 코어 구조체로부터 합성된 비교예의 나노입자로, 메르캅토프로판올을 이용하여 5 초, 20 초, 60 초에서 반응을 중단한 중간체의 TEM 이미지이다. 도 8은 코어 구조체와 매크로사이클릭 호스트 분자의 비율에 따른 쉘 구조체 형성 결과를 분석한 이미지 및 그래프이다. 도 9는 코어 구조체 상에 제공된 매크로사이클릭 호스트 분자를 정량화한 것이다.
도 6의 (a)는 사이클로덱스트린 없이 라만활성물질만을 사용하여 합성한 나노입자(왼쪽), 라만활성물질 없이 사이클로덱스트린을 이용하여 합성한 나노입자(가운데), 사이클로덱스트린 및 라만활성물질을 이용하여 합성한 나노입자(오른쪽)의 이미지이다. 가운데와 오른쪽 이미지에 대하여, 사이클로덱스트린(CDs)/AuNSs의 비율은 5500으로 고정되었다. 도 6의 (b)는 사이클로덱스트린(CDs)/AuNSs의 비율을 다르게 하며(약 4200(왼쪽), 약 5500(가운데), 약 6700(오른쪽)), 나노입자를 합성한 결과이다. 각각의 이미지는 확대된 것이며 축척은 100 nm이다.
도 6의 (a)의 왼쪽 이미지와 도 7을 참고하면, 사이클로덱스트린 개질 없이 코어 구조체를 사용하였을 때 내부갭이 형성되지 않았음을 확인할 수 있다. 또한 도 6의 (a)의 가운데 이미지를 참고하면, 라만활성물질 없이 나노입자를 합성하였을 때 내부 나노갭이 뚜렷하게 관찰되는데, 이는 코어 구조체 상의 사이클로덱스트린이 나노갭 형성에 영향을 미침을 시사한다. 도 6의 (a)의 오른쪽 이미지를 참고하면, 사이클로덱스트린과 라만활성물질을 모두 사용했을 때도 나노입자에서 내부 나노갭이 관찰할 수 있다.
도 6의 (b)와 도 8을 참고하면, 사이클로덱스트린(CDs)과 코어 구조체(AuNS)의 농도 비율을 달리하면서 CD의 수가 다른 CD-AuNSs를 수득할 수 있음을 알 수 있다. CDs/AuNSs가 증가함에 따라 코어의 버딩 구조의 수가 감소하였다. 이는 CD 개질된 코어 구조체 표면이 버딩 구조의 형성을 억제함을 의미한다(도 8의 (a), (b), (c)).
CDs/AuNSs 비율의 변화는 나노입자의 형태에도 영향을 미쳤다. CDs/AuNSs가 약 4200으로 낮을 때 코어 구조체와 쉘 구조체 사이에 점점 더 큰 브릿징 구조가 관찰되었다. CDs/AuNSs의 비율이 약 6700으로 높을 때에는 많은 입자들에서 코어 구조체 주변에 쉘 구조체가 형성되지 않았으며, 낮은 구조적 균일성을 보였다(도 6의 (b) 참고).
CDs/AuNSs가 약 5500일 때, 97 % 이상의 입자들에 대하여 내부 나노갭이 분명하고 균일하게 형성되었으며, 완전한 형태의 쉘 구조체가 형성되었다.
다음으로, 도 9를 참고하면, 50 nm 크기의 코어 구조체(AuNS)에서 사이클로덱스트린(CD) 분자를 정량화하였다. 각 코어당 제공된 사이클로덱스트린의 숫자는 티올 선택적 염색 기반 형광 분석법으로 정량화되었다.
코어 구조체(AuNS) 표면에 부착된 사이클로덱스트린 분자의 수를 정량화하기 위해 55 μL의 100 mM NaBH4 용액을 55 μL의 150 pM 사이클로덱스트린 개질 코어 구조체(CD-AuNS) 용액에 첨가하였다. 혼합물은 실온에서 5 분 동안 인큐베이션된 다음 흡착된 티올레이트 CD 분자의 환원적 탈착 반응이 수행됐다. 유리된 티올 CD 분자는 원심분리 후 상청액(supernatant solution)에 수집되었다. 다음으로, 수집된 용액에서 NaBH4가 분해되도록 용액을 실온에서 하루동안 인큐베이션했다. 다음으로, 30 μL의 유리된 티올 CD 분자 용액을 5분 동안 동량의 티올 선택적 염료 용액과 혼합했다. 혼합물로부터 발생되는 형광 신호는 여기 파장 380 nm와 방출 파장 510 nm를 사용하여 획득하였다. 코어 구조체 표면의 CD 분자 수는 다양한 농도의 CD 용액을 사용하여 동일한 절차로 얻은 표준 농도 곡선을 통해 획득한 것이다.
도 10은 라만활성물질과 결합된 나노입자의 SERS 결과 그래프이다. 도 11은 나노입자의 SERS 강도에 대한 GSH의 영향을 분석한 이미지 및 그래프이다. 도 10과 도 11은 CV 염료를 포함하는 나노입자를 이용하여 SERS 신호를 측정한 결과이다.
도 10의 (a)는 파장 의존 SERS 스펙트럼으로, 여기 파장은 633 nm (3 mW), 785 nm (6 mW), 514 nm (5 mW)이며 획득 시간은 10 초이다. 빨간색과 파란색 선은 각각 1171 cm-1과 1618 cm-1에서 두 개의 서로 다른 피크를 나타낸다. 도 10의 (b)는 사이클로덱스트린으로 개질된 코어 구조체 없이 합성된 나노입자의 두 가지 다른 피크에서의 SERS 강도를 비교한 것이다. 도 10의 (c)는 두 개의 서로 다른 피크에서 나노입자 농도 의존적 SERS 강도 변화를 나타낸 것이고, 도 10의 (d)는 3시간 동안 연속으로 레이저(633 nm, 3 mW)에 노출했을 때, 시간 종속 SERS 스펙트럼을 나타낸 것이다.
도 10의 (a)를 참고하면, 최적 여기 파장을 조사하기 위해 514 nm, 633 nm 및 785 nm의 입사 여기 소스를 사용하였으며, 이중 633 nm 레이저는 가장 강력하고 가장 독특한 SERS 신호를 생성했다. 나노입자의 소광 최대값(extinction maximum)과 CV의 최대 흡수율은 633 nm 레이저를 사용했을 때 다른 파장의 레이저보다 더 나은 스펙트럼 중첩을 나타냈다. 따라서, SERS 신호를 수집하기 위해 633 nm 레이저 소스를 선택하였다.
SERS 측정 전에 글루타티온을 나노입자 용액에 추가하여 합성된 나노입자의 표면에서 염료 탈착을 유도했다. 나노입자 용액의 SERS 스펙트럼은 글루타티온 처리에 거의 영향받지 않았으며, 이는 나노갭 내부의 염료(라만활성물질)가 SERS 신호를 나타내고, 쉘 구조체에 의해 잘 보호되고 있음을 의미한다(도 11 참고).
도 10의 (b)를 참고하면, 사이클로덱스트린 없이 형성된 갭이 없는 Au-Au 코어-쉘 나노입자와 비교하였을 때, 1171 cm-1과 1618 cm-1에서 SERS 강도가 각각 약 28배, 약 32배 증가한 것을 확인할 수 있었다.
도 10의 (c)를 참고하면, 나노입자 농도와 SRES 강도 사이에 선형 관계가 얻어졌고 펨토 몰 농도가 검출되었다. 중요한 것은 3시간 동안 연속 레이저 노출에서도 눈에 띠는 신호 변화 없이 나노입자 용액에서 안정적인 SERS 신호를 얻었다는 것이며(도 10의 (d)), 이는 나노입자에서 안정적이고 정량적인 SERS 신호를 얻을 수 있음을 의미한다.
도 12는 AFM 관련 라만 분광 분석 결과와 SERS 향상 계수(EF) 분포를 나타낸 것이다.
도 12의 (a)는 개별 입자에서 SERS 스펙트럼을 수집하기 위한 나노입자의 AFM 이미지(왼쪽)와 레일리(Rayleigh) 산란 이미지(오른쪽)이다. 도 12의 (b)는 도 12의 (a)의 빨간색 선을 따라 나노입자를 가로지르는 높이 프로파일이다. 도 12의 (c)는 단일 CV-나노입자의 대표적인 SERS 스펙트럼이다. 빨간색과 파란색 선은 각각 1171 cm-1과 1618 cm-1에서의 두 개의 서로 다른 피크를 나타낸다. 도 12의 (d)는 두 지문 피크에 대한 SERS EF 분포이다.
AFM (Atomic Force Microscopy) 관련 라만 분광법을 사용하여 나노입자에 대한 단일 입자 SERS 분석을 수행했다. AFM 관련 라만 분광 분석을 위해 시료는 커버 글래스 상에서 0.1 mM CTAC 용액에 2.5 pM 나노입자를 드롭-캐스팅하여 준비되었으며, 실온에서 3분동안 인큐베이션됐다. 커버 글래스에 남아있는 용액은 공기 펌프를 사용하여 날려버렸다. 도립 광학 현미경(IX73, Olympus)과 오일 침지 현미경 대물 렌즈 (100 Х, NA = 1.4, Olympus)가 장착 된 AFM 관련 라만 현미경 (Ntegra, NT-MDT)을 사용하여 단일 입자 수준에서 나노입자로부터 SERS 스펙트럼을 얻었다.
AFM 팁 위치와 레이저 초점은 이전 문헌 절차를 기반으로 매칭되었다. 일반적으로 레이저 빔은 나노입자가 배치된 커버 글래스의 상부 표면에 집중되었다. 다음으로, AFM 팁의 실리콘(520 cm-1)의 라만 신호가 CCD(Peltier 냉각 -70 ℃)에 의해 검출되는 동안 AFM 헤드의 압전 x, y 튜브 스캐너를 사용하여 초점 주변에서 AFM 팁을 스캔하였다.
가장 높은 라만 신호 강도는 레이저 초점에서 관찰되었다. 팁 매칭 절차 후, 개별 나노입자의 레일리 산란은 633 nm 레이저로 획득하고 광전자 증배관으로 감지했다. 개별 나노입자의 SERS 스펙트럼은 광전자 증배관 이미지에서 레일리 산란이 발생하는 위치에서 얻었다. SERS 스펙트럼은 획득 시간이 2 초인 633 nm (30 μW)를 사용하여 얻었다.
도 12의 (a), (b), (c)를 참고하면, Rayleigh 산란 및 입자의 해당 AFM 이미지는 획득한 SERS 스펙트럼이 단일 나노입자로부터 발생되었음을 확인할 수 있다.
또한, 도 12의 (d)를 참고하면, 133 개의 입자에 대해 1171 cm-1 및 1618 cm-1에서의 피크 강도를 사용하여 단일 나노입자의 SERS 향상 계수(EF) 분포를 계산하였다. 측정된 입자의 평균 EF 값은 3.0 x 109이며, 107 - 108의 SERS EF는 단일 분자 검출에도 충분히 강할 수 있다. EF는 1171 cm-1 및 1618 cm-1 피크 각각에 대해 9.5 x 108에서 1.2 x 1010까지 및 8.2 x 108에서 1.5 x 1010까지 1-2차로 분포되었다. 1171 cm-1에 대한 EF는 측정된 나노입자의 약 95%에 대해 9.5 x 108에서 9.5 x 109 범위의 1차 범위 내에서 매우 좁게 분포되어 있어 나노입자가 매우 정량적인 SERS 프로브임을 나타냈다.
도 13은 라만활성물질과 결합된 나노입자의 SERS 기반 이미징 능력을 나타낸 분석한 것이다. 도 14는 다양한 종류의 라만활성물질과 결합된 나노입자의 HAADF-STEM 이미지로, 축척은 100 nm이다. 도 15는 다양한 종류의 라만활성물질과 결합된 나노입자의 SERS 기반 이미지이다. 도 16은 HeLa 세포의 이미지로 SERS 기반 이미지와 중첩된 것이다.
사이클로덱스트린(CD)은 호스트-게스트 상호 작용을 통해 다양한 게스트 분자와 포접 복합체를 형성할 수 있으며, 이에 따라 다중 감지 적용을 위한 다양한 라만활성물질을 포함할 수 있다. 사이클로덱스트린과 함께 포접 복합체를 나노입자 내에서 형성 할 수 있는 10 가지 다른 라만활성물질을 도입했다.
SERS에 기반한 다중 세포 이미징은 다음과 같이 수행되었다. 먼저 SERS 기반 다중 세포 이미징을 수행하기 전에 SERS 기반 이미징에서 10개의 지문 SERS 피크 (표 2)사이의 스펙트럼 중첩을 조사했다.

스펙트럼 중첩이 거의없는 라만활성물질을 선택하기 위해 10 개의 서로 다른 라만활성물질로 코딩된 나노입자를 커버 글래스의 10개의 서로 다른 위치에 떨어 뜨려 건조했다. 다음으로 나노입자가 밀집된 10개의 커피 링의 각 링 영역에 대한 SERS 기반 이미징을 수행했다. 10가지 색상으로 코딩된 10개의 지문 SERS 피크 채널을 통해 10가지 다른 SERS 이미지를 시각화했다 (표 2 및 도 15 참고). 이로부터 10종의 라만활성물질 중 지문 SERS 피크의 스펙트럼 중첩이 거의없는 6 종의 라만활성물질(CV, BF, BBG, NBA, RB, SO)을 선택했다. 마지막으로, HeLa 세포의 SERS 기반 다중 이미징은 6개의 라만활성물질 코딩된 나노입자를 사용하여 수행되었다. 모든 SERS 측정은 20x 대물 렌즈 (NA = 0.40, Leica)를 통해 633 nm 여기 레이저 (3 mW)를 사용하는 라만 현미경 (Renishaw)를 사용하여 수행되었고, 각 픽셀 (5 μm Х 5 μm) 당 획득 시간은 1 초였다. 6 개의 지문 SERS 피크의 통합 신호 강도는 6 가지 색상으로 코딩되었다(도 16 참고). 병합 된 SERS 이미지도 표시되었다(도 13의 (c)).도 13의 (a)와 도 14를 참고하면, 내부 나노갭은 10 종의 서로 다른 라만활성물질에 대해 모두 분명하게 형성되었다. 이때 쉘 구조체 형성은 게스트 라만활성물질의 종류에 영향을 받았다. 이는 라만활성물질이 나노갭 구조를 조정하는데 사용될 수 있음을 나타낸다.
도 13의 (b)를 참고하면, 단일 여기 소스 (633 nm)를 사용하여 10 개의 라만활성물질의 지문 피크를 감지하고 서로 구별한 것을 확인할 수 있다. 여기에서 6 개의 라만활성물질(CV, BF, BBG, NBA, RB, SO)을 선택했으며, 도 15를 참고하면, 이들은 10종의 라만활성물질들 사이에서 거의 스펙트럼 중첩을 보여주지 않았다.
나노입자의 다중화 기능을 조사하기 위해 6 개의 라만활성물질로 코딩된 나노입자를 사용하여 HeLa 세포의 SERS 기반 다중화 이미징을 수행했다. 나노입자의 표면은 cyclo(Arg-Gly-Asp-D-Phe-Lys) 펩타이드로 기능화되었으며, 이는 종양 내피에 의해 과발현되는 인테그린 αvβ3에 대한 특이적 결합 친화력을 가지고 있어 HeLa 세포에서 효율적인 축적을 유도한다. 각 웰의 세포는 6종의 나노입자 중 1개로 단일 라벨링되었고 모든 웰은 라벨링 후 결합되었다.
도 13의 (c)와 도 16을 참고하면, 6종의 서로 다른 라만활성물질로 코딩된 나노입자로 라벨링된 6가지 세포 각각은 공간적으로 최소한으로 겹치는 6개의 서로 다른 SERS 지문 피크의 채널을 통해 개별적으로 식별 될 수 있었다. 따라서, 단일 여기 소스를 이용한 나노입자의 다중화 기능을 확인할 수 있었다.
도 13의 (d)를 참고하면, 다른 세포의 SERS 스펙트럼 또한 분석되었고, 각각의 지문 SERS 피크는 스펙트럼 중첩이 거의 없었다. 형광 스펙트럼을 이용할 경우 넓고 단순한 스펙트럼 특성 때문에 이러한 다중화 기능 및 분해능을 달성하기 어렵다. 따라서, 형광을 사용하여 사용 가능한 색의 숫자를 확장하기 위해서는 특별한 도구 또는 분석 방법이 필요하다. 또한, 단일 레이저 소스를 사용할 경우, 여기된 서로 다른 형광체 사이의 스펙트럼 중첩이 크기 때문에 단일 여기 소스를 사용하여 형광 신호로 다중화 된 이미지를 얻는 것은 매우 어렵다.
본 발명에 따른 나노입자는 SERS를 이용하여 다중 검출 및 이미징이 가능함을 나타내었다. 라만활성물질은 단일 파장 여기를 사용하여 스펙트럼 겹침이 거의 없는 여러 개의 날카로운 피크를 나타내기 때문이다. 코어 구조체 상의 사이클로덱스트린을 이용한 합성 전략은 복잡한 컨쥬게이션 및 정제 절차 없이 호스트-게스트 작용을 이용해 다양한 라만활성물질을 나노갭 내에 정량적으로 위치시킬 수 있음을 보여줬다. 또한 위치된 라만활성물질은 쉘 구조체에 의해 안정적으로 보호됨을 확인하였다.
이상에서는 본 발명의 바람직한 실시예를 참조하여 설명하였지만, 해당 기술 분야의 숙련된 당업자 또는 해당 기술 분야에 통상의 지식을 갖는 자라면, 후술될 특허청구범위에 기재된 본 발명의 사상 및 기술 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.
따라서, 본 발명의 기술적 범위는 명세서의 상세한 설명에 기재된 내용으로 한정되는 것이 아니라 특허청구범위에 의해 정하여져야만 할 것이다.
Claims (8)
- 코어 구조체; 및상기 코어 구조체를 커버하며 상기 코어 구조체와 나노갭을 두고 이격되어 제공되는 쉘 구조체를 포함하고,상기 코어 구조체의 표면에는 분자 내부에서 소수성을 나타내고 분자 외부에서 친수성을 나타내는 매크로사이클릭 호스트 분자 및 상기 매크로사이클릭 호스트 분자의 분자 내부에 삽입된 라만활성물질이 제공되고,상기 매크로사이클릭 호스트 분자 및 상기 라만활성물질은 상기 나노갭을 채우는, 나노입자.
- 제1항에 있어서,상기 매크로사이클릭 호스트 분자는 사이클로덱스트린(cyclodextrin), 쿠커비투릴(cucurbituril), 칼리싸레인(calixarene), 및 필라렌(pillararene)으로 이루어진 군에서 선택된 적어도 하나의 화합물 및 이의 유도체를 포함하는, 나노입자.
- 제1항에 있어서,상기 매크로사이클릭 호스트 분자와 상기 라만활성물질은 비공유 결합으로 결합되는, 나노입자.
- 제1항에 있어서,상기 코어 구조체 및 상기 쉘 구조체는 금, 은, 및 구리로 이루어진 군에서 선택된 적어도 하나의 물질을 포함하는, 나노입자.
- 제1항에 있어서,상기 나노갭의 크기는 0.1 nm 내지 10 nm인, 나노입자.
- 코어 구조체 표면에 매크로사이클릭 호스트 분자를 부착하여 개질 코어 구조체를 형성하는 제1 단계;상기 개질 코어 구조체와 라만활성물질을 혼합하여 상기 매크로사이클릭 호스트 분자의 분자 내부에 상기 라만활성물질을 삽입하는 제2 단계; 및상기 라만활성물질이 삽입된 상기 개질 코어 구조체 표면에 쉘 구조체를 합성하는 제3 단계를 포함하고,상기 코어 구조체와 상기 쉘 구조체 사이에는 상기 매크로사이클릭 호스트 분자 및 상기 라만활성물질로 채워진 나노갭이 제공되는, 나노입자 합성 방법.
- 제6항에 있어서,상기 제1 단계는 상기 코어 구조체와 상기 매크로사이클릭 호스트 분자를 혼합하고 인큐베이션하여 수행되고,상기 매크로사이클릭 호스트 분자는 사이클로덱스트린(cyclodextrin), 쿠커비투릴(cucurbituril), 칼리싸레인(calixarene), 및 필라렌(pillararene)으로 이루어진 군에서 선택된 적어도 하나의 화합물 및 이의 유도체를 포함하는, 나노입자 합성 방법.
- 제6항에 있어서,상기 제1 단계는 세틸트리메틸암모늄 브로마이드(CTAB, Cetyltrimethylammonium bromide) 캡핑된 코어 구조체를 제공하는 단계; 및상기 세틸트리메틸암모늄 브로마이드 캡핑된 코어 구조체를 L-아스코르브산 및 HAuCl4와 반응시켜 세틸트리메틸암모늄 클로라이드(CTAC, Cetyltrimethylammonium chloride) 캡핑된 코어 구조체를 제공하는 단계를 더 포함하고,상기 세틸트리메틸암모늄 클로라이드 캡핑된 코어 구조체 상에 매크로사이클릭 호스트 분자를 부착하여 개질 코어 구조체를 형성하는, 나노입자 합성 방법.
