足できる程度には生じない。また、上記のとおり種々のニトリルヒドラターゼ変異体が報告されてきたが、これらも精製工程で用いられるような酸性条件下においては活性が大きく低下する。このように、酸性条件下での精製工程においてニトリル化合物からアミド化合物を合成する反応を触媒する酵素活性の良好なpH安定性を示すニトリルヒドラターゼ変異体に関する技術は、これまでに報告されていなかった。特にpH5.0以下といった比較的強い酸性条件下においては、タンパク質は失活する傾向がより高く、このようなpH条件下でも酵素活性が良好に保持されるニトリルヒドラターゼ変異体に関する技術は、これまでに報告されていなかった。

反応条件として、基質であるアクリロニトリルを2.5%(v/v)含む、50mMトリス塩酸水溶液(pH8.0)で、反応温度20℃で15分から60分反応させる。反応終了後、生成したアクリルアミドを定量する。アクリルアミド量はHPLCによって分析することができる。また、酸処理は、例えば、pH4.0近辺で30℃、30時間処理することで行うことができる。具体的には、実施例におけるpH安定性の評価の欄に記載したやり方で評価できる。

合成する方法、遺伝子をヒドロキシアミン処理する方法、遺伝子を保有する菌株を紫外線照射する方法、ニトロソグアニジンや亜硝酸などの化学薬剤で処理する方法、市販の突然変異導入キットを使用する方法などが挙げられる。本開示に係る核酸はDNAであってもRNAであってもよい。
本開示に係るベクターは、前記変異型ニトリルヒドラターゼのアミノ酸配列をコードするヌクレオチド配列で示される核酸を含むベクターであれば特に限定されず、公知のベクターに前記変異型ニトリルヒドラターゼを導入したものが例として挙げられる。また、前記ベクターはファージベクターであってもプラスミドベクターであってもかまわない。
本開示に係る発現ベクターは、前記変異型ニトリルヒドラターゼのアミノ酸配列をコードするヌクレオチド配列で示される核酸を含む発現ベクターであれば特に限定されるものではないが、形質転換効率や翻訳効率を向上させるなどの観点より、以下に示すような構成を示すプラスミドベクターやファージベクターであることがより好ましい。
発現ベクターは、前記変異型ニトリルヒドラターゼをコードするヌクレオチド配列を含み、前記宿主細胞を形質転換しうるものであれば特に限定されない。必要に応じて、該ヌクレオチド配列の他に、他の領域を構成するヌクレオチド配列(以下、単に「他の領域」とも言う。)を含んでいてもよい。 他の領域としては、例えば、前記形質転換体が、前記変異型ニトリルヒドラターゼを産生するために必要とする制御領域や、自律複製に必要な領域などが挙げられる。
また、前記形質転換体の選択を容易にするという観点より、選択マーカーとなりうる選択遺伝子をコードするヌクレオチド配列をさらに含んでいてもよい。
前記変異型ニトリルヒドラターゼを産生するために必要となる制御領域としては、プロモーター配列(転写を制御するオペレーター配列を含む。)、リボゾーム結合配列(SD配列)、転写終結配列等を挙げることができる。
酵母を宿主細胞とした場合、発現ベクターとしては、前記変異型ニトリルヒドラターゼをコードするヌクレオチド配列の他に、プロモーター配列を含んでいることが好ましい。プロモーター配列としては、酵母を宿主細胞とする形質転換体中で前記変異型ニトリルヒドラターゼを発現できるものであればいずれを用いてもよい。
なお、上記プロモーター配列の由来は宿主細胞となる酵母に限定されない。
サイトメガロウイルス(CMV)プロモーターなどのように、外来のプロモーターを用いてもよい。これらは用いる酵素の由来や種類によって適宜選択することができる。
体が前記変異型ニトリルヒドラターゼを産生した場合、細胞外に前記変異型ニトリルヒドラターゼを分泌することが可能となる。
分泌シグナルとしては、宿主細胞となる酵母から前記変異型ニトリルヒドラターゼを分泌できるものであれば、特に限定されない。分泌効率の観点より、αファクターシグナル配列、インベルターゼシグナル配列、酸ホスファターシグナル配列、グルコアミラーゼシグナル配列などを用いることが好ましい。
糸状菌を宿主細胞とした場合、発現ベクターとしては、前記変異型ニトリルヒドラターゼをコードするヌクレオチド配列の他に、プロモーター配列を含んでいることが好ましい。プロモーター配列としては、糸状菌を宿主細胞とする形質転換体中で前記変異型ニトリルヒドラターゼを発現できるものであればいずれを用いてもよい。

また、pUC18、pBR322,pUC100、pSL1180(ファルマシア・インク社製)、pFB6及びアスペルギルス(Aspergillus)pRAX、トリコデルマ(Trichoderma)pTEX等のような一般的に用いられる他の発現ベクターを用いることもできる。
大腸菌、枯草菌、放線菌などの原核生物を宿主細胞とする場合、発現ベクターは、前記変異型ニトリルヒドラターゼをコードするヌクレオチド配列の他に、プロモーター配列を含んでいることが好ましい。また、プロモーター配列の他にリボゾーム結合配列や転写終結配列等を含んでいてもよい。
また、tacプロモーターのように独自に改変又は設計されたプロモーター配列も利用できる。
前記リボゾーム結合配列としては、例えば、16SリボゾームRNAの3’末端領域に相補的な配列のうち、4ヌクレオチド以上連続したコンセンサス配列をDNA合成により作成した配列などが挙げられる。
転写終結配列は必ずしも必要ではないが、ρ因子非依存性のもの、例えばリポプロテインターミネーター、trpオペロンターミネーター等が利用できる。
これら制御領域の発現ベクター上での配列順序は、特に制限されるものではないが、転写効率を考慮すると5’末端側上流からプロモーター配列、リボゾーム結合配列、目的蛋
白質をコードする遺伝子、転写終結配列の順に並ぶことが望ましい。
また、2種類以上の宿主内での自律複製が可能な発現ベクターの例として、pHV14、TRp7、YEp7及びpBS7等を発現ベクターとして利用することができる。
本開示に係る形質転換体は、公知の方法により作製することができる。例えば、本開示に係る変異型ニトリルヒドラターゼをコードするヌクレオチド配列と、必要に応じて前記他の領域とを含む前記発現ベクターを構築し、該発現ベクターを所望の宿主細胞に形質転換する方法等が挙げられる。具体的には、Sambrook,J.,et.al.,”Molecular Cloning A Laboratory Manual, 3rd Edition”,Cold Spring Harbor Laboratory
Press,(2001)等に記載されている分子生物学、生物工学及び遺伝子工学の分野において公知の一般的な方法を利用することができる。
これにより、発現ベクターに組み込んだ前記変異型ニトリルヒドラターゼ由来のタンパク質の生産量を増加させることができる可能性がある。
本開示に係る変異型ニトリルヒドラターゼの製造方法は、前記形質転換体を培地中で培養すること、及び培養された形質転換体及び培地のうち少なくとも一方から、前記変異型ニトリルヒドラターゼを回収することを含む。
〔形質転換体の培養方法〕
前記発現ベクターで形質転換して得られた形質転換体の培養の条件は、形質転換前の宿主細胞の培養条件と同様であり、公知の条件を用いることができる。
培地としては炭素源、窒素源、無機物及びその他の栄養素を適量含有する培地ならば合成培地または天然培地のいずれでも使用可能である。培地に使用する成分としては、公知のものを用いることができる。例えば、肉エキス、酵母エキス、麦芽エキス、ペプトン、NZアミン及びジャガイモ等の有機栄養源、グルコース、マルトース、しょ糖、デンプン及び有機酸等の炭素源、硫酸アンモニウム、尿素及び塩化アンモニウム等の窒素源、リン酸塩、マグネシウム、カリウム及び鉄等の無機栄養源、ビタミン類を適宜組み合わせて使用できる。
なお、前記選択マーカーを含む発現ベクターにより形質転換した形質転換体の培養においては、例えば、前記選択マーカーが薬剤耐性である場合には、それに対応する薬剤を含む培地を使用し、前記選択マーカーが栄養要求性である場合には、それに対応する栄養素を含まない培地を使用する。培地のpHは、pH4~pH8の範囲で選べばよい。
培養は前記培地を含有する液体培地中で、前記形質転換体を振とう培養、通気攪拌培養
、連続培養、流加培養などの通常の培養方法を用いて行なうことができる。
培養条件は、前記形質転換体、培地、培養方法の種類により適宜選択すればよく、形質転換体が生育し、本開示に係る変異型ニトリルヒドラターゼを産生できる条件であれば特に制限はない。
培養温度は20℃~45℃、好ましくは24℃~37℃で好気的に培養を行う。
培養期間は1日間~7日間の範囲で目的の変異型ニトリルヒドラターゼ活性を有する蛋白質の含量が最大になるまで培養すればよい。
変異型ニトリルヒドラターゼ回収工程は、培養された形質転換体及び培養後の培地のうち少なくともいずれか一方から、前記変異型ニトリルヒドラターゼを回収する工程である。
本開示に係る前記変異型ニトリルヒドラターゼが形質転換した形質転換体外に分泌される場合は、該形質転換体の培養物を遠心分離、ろ過等を行うことで粗酵素液を容易に得ることができる。また、本開示に係る前記変異型ニトリルヒドラターゼが形質転換した形質転換体内に蓄積される場合は、培養した該形質転換体を遠心分離等の手段により回収し、回収した該形質転換体を緩衝液に懸濁し、リゾチーム処理、凍結融解、超音波破砕などの公知の方法に従い該形質転換体の細胞膜を破壊することにより、粗酵素液を回収すればよい。
回収されたニトリルヒドラターゼ活性を有する粗酵素液について、分離精製を必要とする場合は、例えば、硫酸アンモニウムなどによる塩析、アルコールなどによる有機溶媒沈殿法、透析及び限外ろ過などによる膜分離法、あるいはイオン交換体クロマトグラフィー、逆相高速クロマトグラフィー、アフィニティークロマトグラフィー、ゲルろ過クロマトグラフィーなどの公知のクロマト分離法を適宜組み合わせて行うことができる。
上述した接触させる温度は、好ましくは、前記変異型ニトリルヒドラターゼが失活しない温度範囲内であり、より好ましくは、0℃~60℃、より好ましくは15℃~35℃である。例えば、前記変異型ニトリルヒドラターゼを産生する形質転換体や細胞株を培養した培養液を、ニトリル化合物を含む水溶液中にそのまま添加してもよく、あるいは培養液を遠心処理して菌体を分離し、菌体をニトリル化合物を含む水溶液に添加してもよい。反
応時における水溶液のpHは7~9であることが好ましく、7.5~8.5であることがより好ましい。ニトリル化合物は水溶液中に、例えば0.25体積%~20.0体積%、より好ましくは2.0体積%~5.0体積%の濃度で存在してもよい。形質転換体の培養についての詳細は、変異型ニトリルヒドラターゼの製造方法の項で述べたとおりである。
30mLの試験管に10mLのLB液体培地を調製し、121℃における20分間のオートクレーブにより該液体培地を滅菌した。この液体培地に対し、終濃度が100μg/mLとなるようにアンピシリンを添加した後、上記の細胞株MT-10822を一白金耳植菌し、37℃で300rpmにて約20時間培養した。その後、得られた培養液1mLを適当な遠心チューブに分取した後、遠心分離(15000rpm×5分)により該菌体を分離した。
続いてアルカリSDS抽出法により、分離して得られた菌体よりプラスミドpPT-DB1を調製した。調製したプラスミドを大腸菌HB101のコンピテントセル(東洋紡績社製)に形質転換し、形質転換体(1)を得た。形質転換体(1)は、野生型シュードノカルディア・サーモフィラ由来ニトリルヒドラターゼであるニトリルヒドラターゼ(1)を産生する。
表14で示すような野生型シュードノカルディア・サーモフィラ由来ニトリルヒドラターゼのαサブユニットのN末端から92番目のアミノ酸残基であるAspをGluに置換した変異型ニトリルヒドラターゼを発現する形質転換体(2)を取得するために、宝酒造社製の「LA PCR in vitro mutagenesis Kit」(以後、変異導入キットと呼ぶ)を用いた部位特異的な変異導入を行った。野生型シュードノカルディア・サーモフィラ由来ニトリルヒドラターゼ発現プラスミドpPT-DB1を鋳型としてPCR反応を行った。
PCR反応No.1は、配列番号5のプライマー及びM13プライマーM4(配列番号12に配列を記載)を各々50pmol含む全量50μLの系(組成は変異導入キットに記載の条件による)で、熱変性(98℃)15秒、アニーリング(55℃)30秒、伸長反応(72℃)120秒の条件を25サイクル繰り返す事により行った。
PCR反応No.2は、MUT4プライマー(配列番号14に配列を記載)及びM13プライマーRV(配列番号13に配列を記載)を各々50pmol含む全量50μLの系(組成は変異導入キットに記載の条件による)で、PCR反応No.1と同様の操作により行った。
Microcon100(宝酒造社製)を用いてそれぞれのPCR反応終了液より過剰なプライマー及びdNTPを除去した後、TEを加えて各々50μLの溶液を調製した。該TE溶液を各0.5μLずつ含む全量47.5μLのアニーリング溶液(組成は変異導入キットに記載の条件による)を調製し、熱変性処理(98℃)を10分間行った後、37℃まで60分間かけて一定の速度で冷却を行い、続いて37℃で15分間保持することによってアニーリング処理を行った。
アニーリング処理液にTaKaRa LA Taqを0.5μL加えて72℃で3分間加熱処理を行い、ヘテロ2本鎖を完成させた。
これにM13プライマーM4(配列番号12に配列を記載)及びM13プライマーRV(配列番号13に配列を記載)を各々50pmol加えて全量を50μLとした後、熱変性(98℃)15秒、アニーリング(55℃)30秒、伸長反応(72℃)120秒の条件を25サイクル繰り返すことによるPCR反応No.3を行った。
続いて、アガロースゲルから約2KbのDNA断片のみを切り出し、該アガロース片(約0.1g)を細かく粉砕し1mLのTE溶液に懸濁後、55℃で1時間保温してアガロースを完全に融解させた。
この融解液に対してフェノール/クロロホルム抽出とエタノール沈澱を行って該DNA断片を精製し、最終的に10μLのTEに溶解した。
精製した約2kbの増幅DNA断片を制限酵素EcoRI及びHindIIIにより切断した後、この制限酵素処理液に対してフェノール/クロロホルム抽出とエタノール沈澱を行って該DNA断片を精製し、最終的に10μLのTEに溶解した。
同様に、EcoRI及びHindIIIによりニトリルヒドラターゼ発現プラスミドpPT-DB1を切断し、アガロースゲル電気泳動(シグマ社製タイプVII低融点アガロース使用;アガロース濃度0.7%)を行い、アガロースゲルから約2.7KbのDNA断片のみを切り出した。
切りだしたアガロース片(約0.1g)を細かく粉砕し1mLのTE溶液に懸濁後、55℃で1時間保温してアガロースを完全に融解させた。
この融解液に対してフェノール/クロロホルム抽出とエタノール沈澱を行って該DNA断片を精製し、最終的に10μLのTEに溶解した。
この様にして得られた約2kbと約2.7KbのDNA断片をDNAライゲーションキット(宝酒造社製)を用いて連結させた後、大腸菌HB101のコンピテントセル(東洋紡績社製)を形質転換し、形質転換体(2)を得た。また上記菌体からアルカリSDS抽出法によりプラスミドを調製し、DNAシーケンサーにてニトリルヒドラターゼ遺伝子部分のヌクレオチド配列を決定し、実施例1記載のプラスミドpPT-DB1に、αサブユニットのN末端から92番目のアミノ酸残基であるAspをGluとする変異を有していることを確認した。形質転換体(2)は、表14に記載の変異を有するニトリルヒドラターゼ(2)を産生する。
こうして得られた形質転換体(2)、及び、そのベースとなったpPT-DB1を含む形質転換体(1)を用いたアミド化合物の製造におけるpH安定性を以下の方法により比較した。これによりニトリルヒドラターゼ(2)のニトリルヒドラターゼ(1)と比較した相対的なpH安定性が評価できる。
試験管に40μg/mLの硫酸第二鉄・七水和物及び10μg/mLの塩化コバルト・二水和物を含む5mLのLB液体培地を調製し、121℃における20分間のオートクレーブにより滅菌した。
この培地に終濃度が100μg/mLとなるようにアンピシリンを添加した後、各形質転換体を一白金耳植菌し、37℃において200rpmにて約20時間其々培養した。
該培養終了液40μlを取り、740μLの54mMトリス塩酸水溶液(pH8.0)に懸濁し、これに20μLのアクリロニトリルを添加して20℃で緩やかに攪拌しながら15分から60分間反応させた。反応終了後、HPLCを用いて反応液の分析を行い、ニトリル化合物からアミド化合物を生成する酵素活性を求めた。反応及び分析は各形質転換体に対し3回以上行い、分注操作等でのデータのばらつきを補正した。
同様に、前記培養終了液1000μlを取り、遠心分離し回収した菌体に50mMクエン酸緩衝液(pH4.0)1000μlを加え、撹拌しながら30℃、30hr処理した(酸処理)。処理後、780μlを遠心分離し菌体を回収し、780μLの54mMトリス塩酸水溶液(pH8.0)に懸濁し、これに20μLのアクリロニトリルを添加して20℃で緩やかに攪拌しながら15分から60分間反応させた。反応終了後、HPLCを用いて反応液の分析を行い、ニトリル化合物からアミド化合物を生成する酵素活性を求めた。反応及び分析は各形質転換体に対し3回以上行い、分注操作等でのデータのばらつきを補正した。

50mMクエン酸緩衝液(pH4.0)で処理した菌体の活性と処理無しの菌体の酵素活性の比を取りpH安定性とした。形質転換体(2)のpH安定性は野生型(1)と比較し0.82倍であった。
表14で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(3)を取得するために、配列番号5のプライマーに代えて配列番号7のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(3)を得た。形質転換体(3)は、表14に記載の変異を有するニトリルヒドラターゼ(3)を産生する。
ニトリルヒドラターゼ(3)のpH安定性を上記「pH安定性の比較」に記載した方法で評価した。得られた結果を表14に示す。
表14で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(4)を取得するために、配列番号5のプライマーに代えて配列番号8のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(4)を得た。形質転換体(4)は、表14に記載の変異を有するニトリルヒドラターゼ(4)を産生する。
ニトリルヒドラターゼ(4)のpH安定性を上記「pH安定性の比較」に記載した方法で評価した。得られた結果を表14に示す。
表14で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(5)を取得するために、配列番号5のプライマーに代えて配列番号9のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(5)を得た。形質転換体(5)は、表14に記載の変異を有するニトリルヒドラターゼ(5)を産生する。

つ以上組み合わせても、ニトリルヒドラターゼのpH安定性の向上をもたらすことが分かる。また、その向上の程度は一段と高い傾向がある。
野生型シュードノカルディア・サーモフィラ由来ニトリルヒドラターゼ及び表18で示すアミノ酸残基置換を有するニトリルヒドラターゼの反応初速度を、「pH安定性の比較」に記載の酸処理前のアミド化合物生成の測定における反応初速度を測定することで調べた結果を表18に示す。反応初速度の値は、野生型シュードノカルディア・サーモフィラ由来ニトリルヒドラターゼにおける反応初速度の値に対する相対値で示す。
表19で示すようなアミノ酸置換位置で変異させたニトリルヒドラターゼ変異体を含むプラスミド(日本国特許第5551081号形質転換体番号92参照)を用いて、比較例2に記載の形質転換と同様の方法によって形質転換体(25)を得た。形質転換体(25)は、表19に記載の変異を有するニトリルヒドラターゼ(25)を産生する。
ニトリルヒドラターゼ(25)のpH安定性を上記「pH安定性の比較」に記載した方法で評価した。得られた結果を表19に示す。
表19で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(26)を取得するために、比較例3のプラスミドを鋳型として用い、配列番号5のプライマーに代えて配列番号8のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(26)を得た。形質転換体(26)は、表19に記載の変異を有するニトリルヒドラターゼ(26)を産生する。

表21で示すようなアミノ酸置換位置で変異させたニトリルヒドラターゼ変異体を含むプラスミド(日本国特許第5551081号形質転換体番号114参照)を用いて、比較例2に記載の形質転換と同様の方法によって形質転換体(27)を得た。形質転換体(27)は、表21に記載の変異を有するニトリルヒドラターゼ(27)を産生する。
ニトリルヒドラターゼ(27)のpH安定性を上記「pH安定性の比較」に記載した方法で評価した。得られた結果を表21に示す。
表21で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(28)を取得するために、比較例4のプラスミドを鋳型として用い、配列番号5のプライマーに代えて配列番号8のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(28)を得た。形質転換体(28)は、表21に記載の変異を有するニトリルヒドラターゼ(28)を産生する。

表23で示すようなアミノ酸置換位置で変異させたニトリルヒドラターゼ変異体を含むプラスミド(特許第5551081号形質転換体番号33参照)を用いて、比較例2に記載の形質転換と同様の方法によって形質転換体(29)を得た。形質転換体(29)は、表23に記載の変異を有するニトリルヒドラターゼ(29)を産生する。
ニトリルヒドラターゼ(29)のpH安定性を上記「pH安定性の比較」に記載した方法で評価した。得られた結果を表23に示す。
表23で示すようなアミノ酸残基置換を有する変異型ニトリルヒドラターゼを発現する形質転換体(30)を取得するために、比較例5のプラスミドを鋳型として用い、配列番号5のプライマーに代えて配列番号8のプライマーを用いること以外は比較例2と同様にしてプラスミドを得て、形質転換体(30)を得た。形質転換体(30)は、表23に記載の変異を有するニトリルヒドラターゼ(30)を産生する。

[48]上記[1]~[46]のうちいずれか1つのニトリルヒドラターゼ(B)におけるαサブユニットにおいて付加、置換、欠失、及び/又は挿入されたアミノ酸残基(前記(a)及び(b)の置換されるアミノ酸残基を除く)の総数が1~10であり、かつ、前記ニトリルヒドラターゼ(B)におけるβサブユニットにおいて、付加、置換、欠失、及び/又は挿入されたアミノ酸残基(前記(c)~(e)の置換されるアミノ酸残基を除く)の総数が1~10であるニトリルヒドラターゼ - 請求項1~請求項8のいずれか1項に記載の変異型ニトリルヒドラターゼをコードする核酸。
- 請求項9に記載の核酸を含むベクター。
- 発現ベクターである、請求項10に記載のベクター。
- 請求項11に記載の発現ベクターを含む形質転換体。
- 請求項12に記載の形質転換体を培地中で培養すること、並びに培養された形質転換体及び培地のうち少なくとも一方から、請求項1~請求項8のいずれか1項に記載の変異型ニトリルヒドラターゼを回収することを含む変異型ニトリルヒドラターゼの製造方法。
- 請求項13に記載の製造方法によって得られた変異型ニトリルヒドラターゼ。
- 請求項1~請求項8及び請求項14のうちいずれか1項に記載の変異型ニトリルヒドラターゼをニトリル化合物に接触させることを含む、アミド化合物の製造方法。
- さらに、pH3.5~pH6.5で、アミド化合物を含む溶液から不純物を除去することを含む、請求項15に記載のアミド化合物の製造方法。
- さらに、アミド化合物を活性炭により精製することを含む、請求項15または請求項16に記載のアミド化合物の製造方法。
