また、従来のバイオセンサの構成は、基板と、この基板上に形成された電極層と、この電極層上に形成されたメディエータ層と、このメディエータ層上に形成された酵素層と、この酵素層上に形成された保護膜と、を備えた構成となっていた(例えば、特許文献3参照)。
そこで本発明は、センサ挿入装置を再使用する際に、一度使用したセンサ供給ユニットを、誤って再使用する動作を禁止することで、利便性を向上することを目的とするものである。
例えば、血糖値センサの中でも、間質液中のグルコースを測定するタイプ(例えばCGM:持続血糖計タイプ)があるが、間質液中のグルコース濃度は、血液中の濃度よりも低いので、センサには測定感度の向上が求められることになる。
本発明者は、このように測定感度が低い原因を鋭意検討する中で、酵素層に重要な要因があることを見出した。
この目的を達成するために、本発明のバイオセンサは、基板と、この基板上に形成された電極層と、この電極層上に形成されたメディエータ層と、このメディエータ層上に形成された酵素層と、この酵素層上に形成された保護膜と、を備え、酵素層はポリアクリル酸ナトリウムを含む構成とし、これにより所期の目的を達成するものである。
(発明の効果)
本発明のセンサ挿入装置によれば、センサ挿入装置を再使用する際に、一度使用したセンサ供給ユニットを、誤って再使用する動作を禁止することで、利便性を向上させることができる。
<持続血糖測定(CGM)装置の構成>
以下、本発明の一実施形態を、この持続血糖測定(CGM)システムの一例として、グルコースを測定するグルコースセンサに適用したものを、添付図面を用いて説明する。
持続血糖測定(CGM)システムは、糖尿病の患者を対象として、持続的に血糖値を測定する。
図1に、本実施形態における持続血糖測定(CGM)システムのセンサ装着装置1を示す。
持続血糖測定(CGM)システムのセンサ装着装置1は、糖尿病患者の上腕部2の皮下にセンサ4(図3参照)を留置し、皮下の組織間質液中のグルコース濃度を連続的に測定する。
図2に示すように、センサ装着装置1は、略円形状のトランスミッタ7によって、測定装置3と無線で送受信できるように構成されている。
センサ装着装置1では、トランスミッタ7がセンサからの電流値をメモリに記憶させ、メモリに記憶された値を無線で測定装置3に送信する。なお、送信する値は電流値でも、グルコース濃度に換算した後の値でもよい。測定装置3では、読み取った値からグルコース濃度を算出して、その結果を時間情報とともに表示するとともに、時間情報とグルコース濃度を測定装置3内のメモリに記憶する。
さらに、この血糖値の変動情報を用いて、患者に投与するインスリンの量とタイミングを算出することもできる。インスリンポンプは、センサ装着装置1(および/または、測定装置3と)に無線接続された状態で、患者の血糖値をリアルタイムにモニタリングしながら適切なインスリン量を投与す。これにより、人工膵臓の機能を実現できるので、理想的な血糖値のコントロールが可能となる。
センサ4のセンサ本体18は、糖尿病患者の皮下に挿入しやすくするために、先端形状は針状または棒状となっており、その長さは約1cmである。センサ4の先端部分は、被験物質であるグルコースを透過吸収し、このグルコースを酵素と反応させて測定物質を生成する保護膜および酵素層、メディエータ層などで構成される反応層によって覆われている。そして、反応層の下方には、グルコースを電気化学的に測定する電極が設けられている。この先端部分を皮下に刺して、間質液に留置することで、皮下組織の間質液のグルコースの濃度を観察(および/または、測定・検出)することができる。
<センサ装着装置の構成>
図4に、センサ装着装置1の分解斜視図を示す。
<ベースユニットの構成>
ベースユニット5は、人体に装着される人体装着体である。そして、センサユニット6は、人体内に留置されるセンサ本体を含む。トランスミッタ7は、センサユニット6から入力された信号から生体情報を算出して記憶し、そして測定装置3に送信する機能を持つ。センサ装着装置1は、ベースユニット5と、センサユニット6と、トランスミッタ7と、が着脱可能に構成されており、人体に対してセンサ4を装着する装置である。
そして、ベースユニット5は、例えば、エラストマー樹脂等の柔軟性のある材質で形成されている。これにより、ベースユニット5の人体に対する装着性を向上させ、センサユニット6やトランスミッタ7を把持しやすくすることができる。

装着孔9は、略円形状のトランスミッタ7の円の中心位置から離れた位置に設けられている。装着孔9に、センサユニット6の頭部円周縁に設けられた凸部10が嵌合するように装着される。センサユニット6の凸部10は、エラストマー樹脂やシリコーンゴム等の柔軟な材料で形成されているので、伸縮性が高い。このため、センサユニット6の凸部10が装着孔9に嵌合した状態では、装着孔9の内壁とセンサユニット6の凸部10とが密着した状態で嵌合する。この結果、ベースユニット5の下面から、例えば、患者の汗等の水分が、トランスミッタ7の内部に侵入することを防止することができる。
再び、図4に戻る。図4に示すように、ベースユニット5には、ベースユニット5の嵌合部11(図5(a)参照)が嵌合する凹状の嵌合部12が設けられている。よって、ベースユニット5に対してトランスミッタ7を装着する際の位置合わせが容易となる。
<ベースユニット、センサユニット、トランスミッタの接続構成>
また、図4に示すように、ベースユニット5の上面に設けられた嵌合部12には、センサユニット6の取付位置を示す矢印13aが設けられている。
同様に、トランスミッタ7の上面にも、センサユニット6の位置を示すように矢印14aが設けられている。これにより、ベースユニット5の矢印13aの向きに合うようにトランスミッタ7の矢印14aの向きを合わせることによって、トランスミッタ7をベースユニット5に対して、容易に位置決めして装着することができる。
図6(a)に示すB枠は、トランスミッタ7の装着孔9と、センサユニット6の凸部10が嵌合している部分であって、図6(b)はそのB枠の拡大図である。

次に、センサユニット6の構成について説明する。

図7(a)、図7(b)に示すように、センサユニット6は、センサ本体18と、センサ本体18を支持するセンサ支持体16aと、を備えている。
センサ本体18は、第2容器16と、第1容器15の側面の開口から端部が挿入されており、他方の棒状の端部を図7中の下方に伸ばした状態になっている。
第1容器15の上面には、導電性の3端子19a、19b、19cが設けられている。
第1容器15は、例えば、シリコーンゴムのような可とう性の弾性力を有する材料で形成されている。その側面には、糸リブ20が形成されている。

本実施形態の接続形態は、センサ本体18自体も、その接続部分も折れ曲げる必要がないので、機械的、電気的に安定した接続状態を実現できる。
図12(a)、図12(b)に示すように、センサユニット6は、使用者に対して、容器29に内装された状態で供給される。容器29の内部には、センサユニット6を含んだセンサ供給ユニット30が内装されている。センサ供給ユニット30は、滅菌処理された状態で内装されている。容器29の上部開口は、シール(図示せず)で封止されている。

図13に示すように、支持体31は、上方には支持体31を把持するための把持部34が設けられ、下方には針35を突出させるための開口が設けられている。ここで、支持体31自体が支持する構成には、針35および針35の開口内にセットされたセンサ4が含まれるだけで、トランスミッタ7は含まれていない。このため、支持体31の開口自体を小さくすることができる。よって、針35が見えにくくなるので、針35による穿刺の恐怖感が低減するとともに、安全性を高めることができる。

<センサユニット挿入装置の保持動作>
センサユニット挿入装置44は、図18(b)に示すように、センサ供給ユニット30を内装した容器29に対して、上方(容器29の開放端側)から挿入される。そして、センサ供給ユニット30は、センサユニット挿入装置44の先端部分に把持された後、図18(c)に示すように、上方に持ち上げられる。

図19(b)に示す状態においては、まだ、センサユニット挿入装置44の挿入部49の端面は、支持体31の円錐台形状の台座31aの台座上面に達していない。よって、開放爪43a、43bは、十分に開放された状態ではない。
図19(c)は、センサ供給ユニット把持部48がさらに下方に押し込まれることで、センサユニット挿入装置44の挿入部49の端面が、センサ供給ユニット30の円錐台形状の台座31aの台座上面に達した状態である。

このように、センサユニット挿入装置44は、本体ケース45と、本体ケース45内に設けられ前端に把持されたセンサ4を人体内に挿入する方向にスライド移動可能な状態で設けられた針スライダ52と、針スライダ52の後端に当接して針スライダ52をセンサ挿入方向に押し込む穿刺ノブ46と、を備えている。そして、センサユニット挿入装置44には、穿刺ノブ46のセンサ挿入開始位置から、穿刺方向前方へ動き出し始めるまでに負荷(抵抗力)をかける負荷機構(プランジャ56a,56b)が設けられている。
図25(a)~図25(c)に、穿刺動作直後に行われる針35の抜去動作を示す。
穿刺動作については上述したが、穿刺動作の後は、センサ4を皮下に留置した状態で、針35のみを素早く抜去することが望ましい。
図25(b)、図25(c)では、穿刺ノブ46の前端55側において、廃棄スライダ53の後端が当接した部分を開放する開放機構によって、針スライダ52を一気に穿刺方向後方へスライド移動させて、針35が抜去される状態を示したものである。
この開放機構および後述する復帰機構については、本実施形態では2つの実施形態を採用している。
図26(a)~図26(c)に示す穿刺ノブ46の開放機構は、針スライダ52の後端部分を挿入する挿入孔60の開口を開閉自在にスライド移動する窓体(開放機構)61と、挿入孔60の開口を閉じる方向に窓体61を付勢する弾性体(復帰機構)62と、廃棄スライダ53の穿刺方向後端と摺接して窓体61を開く方向にスライド移動させるハンドル(開放機構)63とを有する。
なお、本実施形態のセンサユニット挿入装置44に含まれる開放機構および復帰機構は、図26(a)~図26(c)に示す構成に限定されるものではない。
図27(a)~図27(f)に示す穿刺ノブ46の開放機構は、針スライダ52の後端(第2端)が挿入される挿入孔60の開口を開閉自在に回動する窓体(開放機構)65と、窓体65を挿入孔60の開放側を塞ぐ(閉じる)方向に付勢する弾性体(復帰機構)66と、廃棄スライダ53の端部と摺接して窓体(開放機構)65を開く方向に回動させるハンドル67とを有する。
図27(c)において、穿刺ノブ46が前方に押し込まれ、穿刺方向前方に針スライダ52がスライド移動した状態では、穿刺ノブ46の前端の開放機構のハンドル67が廃棄スライダ53の後端に当接する。
図28(a)~図28(e)に、センサユニット挿入装置44の廃棄動作および初期化動作を示す。
図28(b)では、廃棄ノブ47が穿刺方向前方に押されることで、針スライダ52および廃棄スライダ53の両方が同時に押し下げられて、センサ供給ユニット30の排出が始まる。
図28(c)では、廃棄ノブ47の穿刺方向前方への押し出しが完了した状態であるが、センサ供給ユニット30の針ユニット33の把持部34が針スライダ52の前端に嵌合した状態であるので、センサ供給ユニット30が落下することはない。
図28(e)では、穿刺ノブ46を穿刺方向後方に戻し、初期状態に戻る。このとき、開放機構もリセットされる。
センサユニット挿入装置44は、本体ケース45と、本体ケース45の穿刺方向前方に設けられたセンサ供給ユニット把持部48と、を有している。
そして、センサユニット挿入装置44には、本体ケース45内に設けられ前端で挿入されたセンサ供給ユニット30を把持しセンサ供給ユニット30内のセンサ4を人体内に挿入する方向にスライド移動する針スライダ52と、針スライダ52に対してセンサ挿入方向とは逆方向にバネ68によって付勢され前端に挿入されたセンサ供給ユニット30に当接する廃棄スライダ53と、が設けられている。
すなわち、廃棄スライダ53が固定された状態で、穿刺ノブ46が前方に押し込まれて、針スライダ52が穿刺方向前方にスライド移動すると、針スライダ52と廃棄スライダ53との穿刺方向における相対位置が変化する。このとき、針スライダ52と廃棄スライダ53とに連結されたバネ68が伸びて付勢力が蓄えられる。
本実施形態のセンサユニット挿入装置44では、使用者の手動操作によって人体の皮下へ針35とセンサ4とを挿入する穿刺動作の後、この穿刺方向後方に向かうバネ68の付勢力によって、センサ4を皮下に残したまま針35だけが抜去される。
そして、センサ4が皮下に挿入された後、センサ供給ユニット30をセンサユニット挿入装置44の先端から排出(廃棄)するための廃棄ノブ47が、本体ケース45の中央付近に設けられている。
本実施形態の容器29は、上部に開口を有しており、センサ供給ユニット30を収納するための容器であって、容器29の内底面に設けられたセンサ供給ユニット30を着脱自在に係合する係合部としての係合爪42a,42bと、係合爪42a,42bの上方に設けられた係合爪42a,42bを開放する開放部としての開放爪43a,43bと、開放爪43a,43bと係合爪42a,42bを内底面に対して支持する脚部41a,41bと、を備えている。
本実施形態のセンサ供給ユニット30は、センサ本体18と、センサ本体18を支持するセンサ支持体16aと、センサ本体18を着脱自在に挿入する針35と、針35を固定して支持するとともにセンサ支持体16aを着脱自在に支持する針支持部としての把持部34と、針支持部としての把持部34をスライド移動可能な状態で支持する支持体31とを備えている。支持体31は、支持体31を取り出す際に把持する把持部としての頭部31bが設けられている。
本実施形態のバイオセンサは、生体情報を検知し電気信号に変換するセンサ本体18と、センサ本体18を支持するとともにセンサ本体18からの電気信号を入力するセンサ支持体16aと、を備えている。センサ本体18は、生体内に挿入する測定部としての第2の端部24と、測定部に固定して接続された基板状の接点部25とを有している。センサ支持体16aは、センサ本体18の接点部25を挿入する第1容器15を有している。第1容器15内には、センサ本体18の接点部25を挟持するコネクタ21が設けられている。
本実施形態のバイオセンサは、生体情報を検知し電気信号に変換するセンサ本体18と、センサ本体18を支持するとともにセンサ本体18からの電気信号を入力するセンサ支持体16aと、を備えている。センサ本体18は、生体内に挿入する測定部としての第2の端部24と、測定部と接続された基板状の接点部25とを有している。センサ支持体16aは、センサ本体18の接点部25を挿入する円筒形の第1容器15と、第1容器15内に設けられたセンサ本体18の接点部25を挟時するコネクタ21と、第1容器15を覆う円筒形の形状の第2容器16と、を有している。
本実施形態のセンサ装着体は、人体に装着されるベースユニット5と、人体内に留置されるセンサ本体18を含むセンサユニット6と、センサユニット6から入力された信号から生体情報を算出して記憶するトランスミッタ7と、が着脱可能な状態で組み合わされて構成されている。
本実施形態のセンサユニット挿入装置44は、センサ4を支持した状態のセンサ供給ユニット30を容器29から取り出して、センサ供給ユニット30が支持するセンサ4を人体内に挿入する。そして、センサユニット挿入装置44は、本体ケース45と、本体ケース45の下方に設けられたセンサ供給ユニット把持部48と、を備えている。センサ供給ユニット把持部48には、下方に開口を有するセンサ供給ユニットの挿入部49が設けられている。
本実施形態のセンサユニット挿入装置44は、本体ケース45と、本体ケース45内に設けられ前端に把持されたセンサ4を人体内に挿入する方向にスライド移動可能な状態で設けられた針スライダ52と、バネ68によってセンサ挿入方向と逆方向に針スライダ52を付勢する廃棄スライダ53と、針スライダ52の後端に当接して針スライダ52をセンサ挿入方向に押し込む穿刺ノブ46と、を備えている。そして、センサユニット挿入装置44は、針スライダ52がセンサ4を人体内に挿入する際に、所定の距離分、穿刺方向における前方にスライド移動すると、針スライダ52の後端の当接部分を開放する開放機構が設けられている。
本実施形態のセンサユニット挿入装置44は、本体ケース45と、本体ケース45の前方に設けられたセンサ供給ユニット把持部48と、本体ケース45内に設けられ前端で挿入されたセンサ供給ユニット30を把持しセンサ供給ユニット30内のセンサ4を人体内に挿入する方向にスライド移動する針スライダ52と、バネ68によってセンサ挿入方向と逆方向に針スライダ52を付勢し前端で挿入されたセンサ供給ユニット30に当接する廃棄スライダ53と、針スライダ52の後端に当接して針スライダ52をセンサ挿入方向に押し込む穿刺ノブ46と、を備えている。センサ供給ユニット30がセンサ供給ユニット把持部48に挿入された状態では、廃棄スライダ53が穿刺方向後方へスライド移動する。
本実施形態のセンサ供給ユニット30は、センサ本体18と、センサ本体18を支持するセンサ支持体16aと、センサ本体18を着脱可能に挿入する針35と、針35を固定して支持するとともにセンサ支持体16aを着脱自在に支持する針支持部としての把持部34と、針支持部としての把持部34をスライド移動可能な状態で支持する支持体31とを備えている。支持体31は、上方には支持体31を把持する把持部としての頭部31bが設けられており、下方には針35を突出するための開口が設けられている。
本実施形態のセンサ供給ユニット30は、センサ本体18と、センサ本体18を支持するセンサ支持体16aと、センサ本体18を着脱自在に挿入する針35と、針35を固定して支持するとともにセンサ支持体16aを着脱自在に支持する針支持部としての把持部34と、針支持部としての把持部34をスライド移動可能な状態で支持する支持体31とを備えている。
<センサ供給ユニットの再利用防止の仕組み>
ここでは、図30(a)~図30(e)を用いて、センサユニット挿入装置44の利便性を向上させるための仕組みとして、センサ供給ユニット30の再利用を防止するための構成について説明する。
図30(a)、図30(b)に、センサユニット挿入装置44の穿刺動作を示す。
図30(a)、図30(b)では、使用者によって穿刺ノブ46が前方に押し込まれて穿刺方向前方へスライド移動することで、穿刺ノブ46と穿刺方向後方(穿刺側とは反対側)の端部において当接する針スライダ52が前方(穿刺方向前方)にスライド移動する。その結果、図30(b)に示すように、センサ供給ユニット30の針ユニット33が、支持体31の開口端を滑りながら前方にスライド移動する。
図30(b)は、針35による穿刺が完了した状態である。この状態では、針35が最大限、穿刺方向前方に突出している。
図30(c)に、針35の抜去動作を示す。
図30(a)、図30(b)では、穿刺ノブ46の前端55が、針スライダ52の後端(第2端)に当接して、針スライダ52がセンサ挿入方向に押し込まれる状態である。
図30(b)では、穿刺ノブ46の前端55(図25(a)等参照)が、廃棄スライダ53の後端に当接した状態である。この当接状態は、針スライダ52がセンサ4の挿入方向において所定の長さをスライド移動したことを示すものである。
これにより、図30(d)に示すように、穿刺ノブ46を図中上方の限界位置まで引き上げても、針スライダ52の後端は、穿刺ノブ46の挿入孔60内において保持されたままとなる。よって、この状態では、図30(e)に示すように、穿刺ノブ46は、針スライダ52の後端に当接して針スライダ52を穿刺方向前方に押し込むことができない。
開放機構は、針スライダ52の後端を挿入する挿入孔60の蓋としての窓体61と、廃棄スライダ53の後端に当接すると蓋を開状態へ移行させるハンドル63と、を備えている。挿入孔60の蓋となる窓体61は、針スライダ52の後端部分が挿入孔60から抜けた場合には、挿入孔60を閉じる方向に付勢される。一方、針スライダ52の後端部分が挿入孔60に挿入されている場合には、蓋としての窓体61は針スライダ52の外周面に接触して閉じることができず開いたままになる。よって、穿刺ノブ46は、針スライダ52の後端に当接して針スライダ52を前方にスライド移動させて押し込むことができない。
次に、穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1を、穿刺ノブ46の挿入孔60に挿入される針スライダ52の長さL2よりも短くするための構成について説明する。
図31(a)は、センサユニット挿入装置44の前端部分がセンサ供給ユニット30に差し込まれる直前の状態を示している。
図31(b)は、センサユニット挿入装置44の前端部分に、センサ供給ユニット30が装着された状態を示している。
次に、図31(b)に示すように、センサ供給ユニット30の針ユニット33の把持部34が針スライダ52の前端に当たり、針スライダ52を押し上げ始める。
この状態では、センサユニット挿入装置44のセンサ供給ユニット把持部48(図29等参照)は、支持体31の頭部31bを把持し、針ユニット33の係合羽33a、33bの羽部分を外側から押さえている。これにより、上述したように、係止解除された針35が前方にスライド移動可能な状態、つまり穿刺可能な状態となる。
一方、廃棄スライダ53は、センサ供給ユニット30の頭部31bの上面に当接している。そして、廃棄スライダ53の後端は、穿刺ノブ46の前端55には当たらない。よって、廃棄スライダ53は、センサ供給ユニット30がセンサユニット挿入装置44へ押し込まれた分だけ穿刺方向後方へスライド移動する。
すなわち、センサ供給ユニット30の装着時には、廃棄スライダ53が穿刺方向後方へスライド移動する長さは、針スライダ52が穿刺方向後方へスライド移動する長さよりも長い。
その結果、穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1よりも、穿刺ノブ46の挿入孔60に挿入される針スライダ52の長さL2の方が長くなる。
本実施形態のセンサユニット挿入装置44は、以上のように、本体ケース45と、針スライダ52と、廃棄スライダ53と、バネ68と、穿刺ノブ46と、開放機構(窓体61、ハンドル63)とを備えている。針スライダ52は、第1端側において針35とセンサ4とを把持し、センサ挿入方向においてスライド移動可能な状態で本体ケース45内に設けられている。廃棄スライダ53は、針スライダ52に対して平行にスライド移動可能な状態で本体ケース45内に設けられている。穿刺ノブ46は、針スライダ52の後端部分を挿入可能な挿入孔60を有する。開放機構を構成する窓体61とハンドル63とは、針スライダ52をセンサ挿入方向に所定の長さスライド移動させる際には挿入孔60を閉状態とし、針35による穿刺が完了すると針スライダ52の第2端と穿刺ノブ46との当接部分を開放して針スライダ52の第2端が挿入孔60内へ挿入された状態とする。
よって、使用者によって、一度、穿刺動作が行われた使用済みの針35が再利用されることを防止することができる。この結果、使用者の安全性を向上させて、使い勝手を向上させることができる。
このように、穿刺ノブ46を上方の限界位置まで引き上げても、針スライダ52の後端は、穿刺ノブ46の挿入孔60から出ない。よって、この状態では、図30(e)に示すように、穿刺ノブ46は、針スライダ52の後端に当接して前方にスライド移動させて押し込むことができない。
そして、本実施形態のセンサユニット挿入装置44は、図31(a)、図31(b)に示すように、センサ供給ユニット30装着時に、廃棄スライダ53が穿刺方向後方へスライド移動する長さL4は、センサ供給ユニット30装着時に、針スライダ52が穿刺方向後方へスライド移動する長さよりも長い。
このため、穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1よりも、穿刺ノブ46の挿入孔60に挿入される針スライダ52の長さL2の方が長くなる。
<センサユニット挿入装置の再利用の仕組み>
本実施形態のセンサユニット挿入装置44は、以下で説明する復帰機構(弾性体62(または弾性体66)を用いて、穿刺動作後に開放機構によって再利用不能な状態となっている状態から、新たなセンサ供給ユニット30を装填して再利用(穿刺動作)可能な状態へ復帰させる。
図32(a)~図32(e)に、センサ供給ユニット30を、センサユニット挿入装置44から取り外す工程を示す。
図32(c)は、上述したように、針35の抜去動作を示している。
図32(d)は、廃棄ノブ47が穿刺方向前方に押されることで、廃棄ノブ47が針スライダ52の外縁の係合部70と廃棄スライダ53の外縁の係合部71とに係合して両方を同時に押し下げて、センサ供給ユニット30の排出が始まった状態を示している。
図32(e)は、センサ供給ユニット30を、使用者が手でつまんで引き抜き、取り外し、廃棄が完了した状態を示している。
そして、上述したように、穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1と、穿刺ノブ46の挿入孔60に挿入される針スライダ52の長さL2との差分の長さをL3とする。
これにより、センサ供給ユニット30をセンサユニット挿入装置44から取り外した後に、穿刺ノブ46を長さL1分、穿刺方向後方に戻すことで、挿入孔60の蓋の開閉部(窓体61または窓体65)は、針スライダ52の後端部分が挿入孔60から抜けると同時に蓋を閉じた状態となる。
図33(a)~図33(c)に、センサ供給ユニット30を、センサユニット挿入装置44から取り外す際に、センサユニット挿入装置44を初期化する工程を示す。
図33(a)は、図32(e)と同じ状態であり、廃棄ノブ47の穿刺方向前方への押し出しが完了した状態において、センサ供給ユニット30を、使用者が手でつまんで引き抜いて取り外し、センサ供給ユニット30の取り外しが完了した状態である。
すなわち、このような構成とすることで、センサユニット挿入装置44の前端部分に、新しくセンサ供給ユニット30を装着して、皮下にセンサ4を挿入可能な状態としている。
本実施形態でのセンサユニット挿入装置44は、本体ケース45と、本体ケース45内に設けられ、前端にセンサ4を把持し、センサ4を人体内に挿入する方向においてスライド移動可能な状態で設けられた針スライダ52と、を備えている。そして、センサユニット挿入装置44は、針スライダ52に対して、センサ4の挿入方向(穿刺方向前方)とは逆方向に廃棄スライダ53を付勢するバネ68をさらに備えている。
これにより、センサ4を人体へ挿入する穿刺動作の後、廃棄ノブ47によって針スライダ52を穿刺方向前方にスライド移動させることで、穿刺ノブ46が穿刺ノブ46の挿入孔60から針スライダ52の後端(第2端)が引き出される方向に移動する。そして、穿刺ノブ46が、穿刺方向後方(図33(b)中の上方)にスライド移動されることで、針スライダ52の後端部分が挿入孔60から抜けた状態となる。
ここで、図30(a)および図30(b)に示すように、穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1は、穿刺ノブ46の挿入孔60に挿入される針スライダ52の長さL2よりも短い。そして、図32(d)に示す廃棄ノブ47が針スライダ52をセンサ挿入方向にスライド移動させる長さL5は、挿入孔60に挿入される針スライダ52の長さL2と穿刺ノブ46がセンサの挿入方向にスライド移動する長さL1の差分の長さL3(図30(d)参照)よりも長い。
本実施形態のセンサユニット挿入装置44では、廃棄ノブ47が針スライダ52をセンサ挿入方向にスライド移動させる長さL5が、長さL3よりも長い場合には、針スライダ52の後端部分が挿入孔60から抜かれた状態となる。
その結果、穿刺ノブ46が針スライダ52の後端部分に当接して再び穿刺動作を実施可能な初期状態に戻すことができる。つまり、復帰機構によって、穿刺ノブ46を穿刺方向前方へスライド移動させて針スライダ52を押し込んで穿刺可能な初期状態に復帰させることができる。よって、未使用の針35を含むセンサ供給ユニット30を再度装填すれば、センサユニット挿入装置44を再利用可能とすることができる。
本発明の他の実施形態に係るバイオセンサの構成について、図36(a)~図47を用いて説明すれば以下の通りである。
なお、上記実施形態と共通の構成については、同じ符号を付し、その詳細な説明は省略する。
図36(a)にセンサ本体18の上面図、図36(b)に下面図を示す。
センサ本体18は、人体に挿入するセンサ挿入部80と、このセンサ挿入部80から送られてくる電気信号を接続する端子を設けた、センサ接続部81により構成される。
センサ挿入部80は、保護膜82で覆われている。そして、センサ接続部81には、上面には参照電極接続端子83と、作用電極接続端子84が設けられ、下面には、対向電極接続端子85が設けられている。そして、これら端子以外の部分は、非導電性の絶縁膜であるレジスト膜86で覆われている。
<センサの製造工程>
図37(a)~図37(d)、図38(a)~図38(d)、図39(a)~図39(d)にセンサ本体18の製造工程を示す。
図37(c)、図37(d)に示す工程では、下面についても同様に、基板87上に金電極88を形成する。
図38(b)に示す工程では、参照電極形成領域91、作用電極92、参照電極接続端子83、および作用電極接続端子84の領域以外に、レジスト膜86をスクリーン印刷法などにより形成する。
図38(d)に示す工程は、下面について、対向電極94および対向電極接続端子85の領域以外にレジスト膜86を形成する。
図39(a)に示す工程では、作用電極92の表面に、メディエータ溶液を0.3uLを塗布し、図39(b)に示すメディエータ層95を形成する。
より具体的には、25mg/mLグルコースオキシダーゼ(from Aspergillus niger:シグマアルドリッチ社製)、0.001%ポリアクリル酸ナトリウム(和光純薬社製)を調整し、酵素溶液として0.3uLをメディエータ層95の上に固定する。
図39(d)に示す工程では、ポリ―4―ビニルピリジン溶液中に、酵素層96を形成した基板87をディップして、保護膜82を被膜する。
より具体的には、保護膜82として、4%ポリ―4―ビニルピリジン(エタノール溶媒)(シグマアルドリッチ社製)にセンサをディップし乾燥する。この処理を6回行って、6回被膜したものを保護膜82とする。この際、図40(a)から図40(b)に示す工程において、下面の保護膜82も同時に形成される。
<センサの構成>
図41(a)にセンサ本体18の上面図を示す。本図面では、説明を容易にするために、保護膜82を点線で示している。このA枠部分のC-C‘の縦断面について示したのが図41(b)である。
図42に示すように、A-A‘断面では、中央には樹脂材料であるPET(ポリエチレンテレフタレート)で形成された基板87が配置されている。そして、基板87の上方には、電極層として金電極で形成された作用電極92と、メディエータ層95と、ポリアクリル酸ナトリウムを添加した酵素層96と、保護膜82と、が順次形成されている。
図41(b)のB-B‘断面における横断面を、図43に示す。
図43に示すように、B-B‘断面では、中央には樹脂材料であるPET(ポリエチレンテレフタレート)で形成された基板87が配置されている。そして、基板87の上方には、電極層として金電極で形成された作用電極92と、同じく電極層として銀/塩化銀電極で形成された参照電極93およびレジスト膜86と、保護膜82と、が順次形成されている。
B-B‘断面の基板87の下方では、金電極で形成された対向電極94と、保護膜82と、が順次形成されている。
なお、酵素層96の酵素は、本実施形態ではグルコースオキシダーゼを用いているが、その他にもグルコースデヒドロゲナーゼを選択することもできる。
<センサ測定原理について>
図44にセンサの測定原理を示す。
図44は、図41(b)のA-A‘断面での作用電極92の上方を拡大した図である。
酵素層96は、酸化酵素または脱水素酵素を含む。具体的には、グルコース酸化酵素としてグルコースオキシダーゼ、グルコース脱水素酵素としてはグルコースデヒドロゲナーゼである。本実施形態では、グルコースオキシダーゼを用いる。
酵素層96は、メディエータ層95に接しており、電極の表面にメディエータ層95が設けられている。
このような構成においてのグルコースの測定原理について、以下で説明する。
そして、還元されたルテニウム錯体(2価)が、作用電極92での金電極表面にて酸化される際の電子を電流値として測定する。
これにより、基質であるグルコースと反応した酵素(グルコースオキシダーゼ)によって、酵素層96とメディエータ層95との界面で電子受容体であるルテニウム錯体(3価)が還元され還元体のルテニウム錯体(2価)に即座に作用することができる。
<実験結果1:酵素層にポリアクリル酸ナトリウムを添加>
本実施形態のバイオセンサの測定感度の向上性能を確認するために、グルコース濃度を変化させて、センサに流れる応答電流の変化を調べた結果を、図45に示す。
応答電流の測定には、バッチ式アンペロメトリー法を用いた。
比較例としては、酵素層に無添加のセンサを用いて、同様の実験を行った。
測定電流の電流値(nA)をY軸に、グルコース濃度(mg/dL)をX軸としてプロットした場合に、ポリアクリル酸ナトリウムを0.001%添加したセンサにおいてグルコースセンサの応答性が約4倍向上した結果が得られている。
以下、実験2、実験3においては、酵素層の添加物として、トレハロース、グルコマンナンを用いた。それらの添加物においても、ポリアクリル酸ナトリウムを添加した場合と同様にバイオセンサの感度が向上し、より正確な濃度測定を行うことができることを立証した。
<実験結果2:酵素層にトレハロースを添加>
本実施形態のバイオセンサの測定感度の向上性能を確認するために、グルコース濃度を変化させて、センサに流れる応答電流の変化を調べた結果を、図46に示す。
実験では、酵素層として、酵素にトレハロースを添加し固定された作用電極と、参照電極となる銀/塩化銀電極と、対向電極とを生理食塩水中に浸漬し、印加電位1.1V(vs 銀/塩化銀)の条件で生理食塩水中に含まれるグルコース濃度を測定し、グルコース濃度と応答電流との関係を評価した。
グルコースを100mg/dL、200mg/dL、300mg/dL、400mg/dL、500mg/dLとなるようにリン酸緩衝溶液中に添加を行い添加10分後の電流値を測定値とした。
比較例としては、酵素層に無添加のセンサを用いて、同様の実験を行った。
以上のことより、酵素層にトレハロースを添加することで、バイオセンサの感度が向上し、より正確な濃度測定を行うことができることが立証された。
<実験結果3:酵素層にグルコマンナンを添加>
本実施形態のバイオセンサの測定感度の向上性能を確認するために、グルコース濃度を変化させて、センサに流れる応答電流の変化を調べた結果を、図47に示す。
応答電流の測定には、バッチ式アンペロメトリー法を用いた。
比較例としては、酵素層に無添加のセンサを用いて、同様の実験を行った。
測定電流の電流値(nA)をY軸に、グルコース濃度(mg/dL)をX軸としてプロットした場合に、グルコマンナンを0.001%添加したセンサにおいてグルコースセンサの応答性が約1.6倍向上した結果が得られている。

- 第1端側において人体に穿刺を行う針を用いてセンサを人体内へ挿入するセンサ挿入装置であって、
本体ケースと、
前記第1端側において前記針と前記センサとを把持するとともに、前記センサの挿入方向においてスライド移動可能な状態で前記本体ケース内に設けられた針スライダと、
前記針スライダに対して平行にスライド移動可能な状態で前記本体ケース内に設けられた廃棄スライダと、
前記針スライダと前記廃棄スライダとの間に設けられており、前記センサの挿入方向へ移動した前記針スライダを前記センサの挿入方向とは反対の方向に付勢する付勢部材と、
前記針スライダの前記第1端とは反対側の第2端側に当接した状態で前記針スライダを前記センサの挿入方向にスライド移動させて所定の穿刺位置まで押し込むとともに、前記針スライダの後端部分を挿入可能な挿入孔を有する穿刺ノブと、
