Abstract

Description

아토피 피부염 유사 3차원 피부조직 모델 및 이를 이용한 아토피 피부염 치료제의 스크리닝 방법
본 발명은 아토피 피부염 유사 3차원 피부조직 모델 및 이를 이용한 아토피 피부염 치료제의 스크리닝 방법에 관한 것으로, 더욱 상세하게는 TSLP(Thymic stromal lymphopoietin) 또는 환경인자인 톨루엔의 처리를 통해 필라그린(Filaggrin)의 발현이 감소하고, 항균펩타이드인 β-디펜신(β-defensin) 또는 서라이어신(Psoriasin)의 발현이 추가로 감소된 아토피 피부염 유사 소견을 보이는 3차원 피부조직 모델을 제조하고, 이에 아토피 피부염 치료제 후보물질을 처리한 후, 필라그린의 발현을 증가시키고, 항균펩타이드인 β-디펜신 또는 서라이어신의 발현을 추가로 증가시키는 후보물질을 치료제로 선정하는 것을 특징으로 하는 아토피 피부염 치료제의 스크리닝 방법에 관한 것이다.
3차원 피부 모델은 사람 피부의 표피를 이루는 주요 세포인 각질세포(keratinocytes)를 기체-액체 경계면(air-liquid interphase)에서 분화유도시킴으로써 3차원적으로 사람의 표피를 재구성한 인공피부 모델(RhE, Reconstituted human skin equivalent model)이다. 이를 통해 피부의 일차 자극 대체시험 등 많은 연구가 가능하다. 이미 EPISKINTM, EpiDermTM, SkinEthicTM 등과 같은 인공피부 모델은 다양한 검증(validation) 연구를 통해 피부자극 대체법으로서의 유용성을 인증하여 OECD 가이드라인(guideline)에 등재되어 있다.
아토피 피부염은 심한 가려움증과 전체적인 건조피부를 동반하며 습진 병변이 특정 부위에 반복하여 재발하는 만성 염증성 피부질환으로, 유전적인 소인을 나타내고 음식물 알레르기, 알레르기 천식, 알레르기 비염 등 알레르기 질환과 동반하는 알레르기 행진을 하는 특징을 가지고 있다(Akiharu Kubo et al., J Clin Invest. 122(2):440-447, 2012; Hywel CW et al., N Engl J Med, 352:2314-2324, 2005). 오늘날 아토피 피부염의 병인은 크게 피부장벽과 연관된 병인과 비정상적인 면역반응과 연관된 병인으로 나누어지는데, 필라그린 유전자의 돌연변이가 아토피 피부염 환자의 40% 정도에서 나타나며 심한 증상을 나타내거나 성인까지 지속되는 경우에는 더 흔하게 나타난다고 보고된 이후에는 선천적으로 손상된 피부장벽을 가진 사람에서 외부의 알레르겐이 더 쉽게 각질층을 통과하여 몸에 감작을 일으킴으로써 아토피 피부염을 유발시킨다.
한편, 피부장벽을 구성하는 핵심 단백질은 필라그린(Filaggrin), 로리크린(Loricrin), 인볼루크린(Involucrin) 등으로 각질층의 형성에 중요한 역할을 하는 것으로 알려져 있다.
그 중, 필라그린은 과립층에서는 케라토하이알린 과립(Keratohyalin granule)을 구성하는 단백질인 프로필라그린 형태로 존재하다가, 각질형성세포의 최종분화 과정에서 필라그린으로 분해되어서 각질세포막을 형성할 때에 케라틴 필라멘트를 응집하여 각질세포의 단단하고 편평한 구조를 만들어 피부장벽에서 벽돌의 역할을 수행한다. 이후 각질층 상부로 올라가면서 필라그린은 아미노산으로 분해되고, 함습에 매우 중요한 자유아미노산과 PCA(pyrrolidone carboxylic acid)로 변화한다. 필라그린의 분해산물이 피부를 촉촉하게 유지하는데 가장 중요한 역할을 수행하는 것으로 보고된 바 있다(McGrath JA et al., Trends in Molecular Medicine, 14:20-27, 2008; Brown SJ et al., Journal of Investigative Dermatology, 132:751-762, 2012).
필라그린은 분해과정을 통하여 시트룰린(citrulline), PCA, trans-UCA를 생성하며 이들은 각질층 수화, 각질층 pH의 정상화, 투과장벽, 각질층의 견고함(integrity/cohesion), 항균 및 항염작용을 수행한다.
필라그린 유전자(FLG)의 돌연변이로 발생하는 보통 어린선(Ichthyosis vulgaris)은 매우 심한 피부건조를 나타낸다. 즉, 상기 필라그린 유전자의 돌연변이는 필라그린 단백질의 생성을 감소시키고, 자연보습인자의 생성을 감소시켜서 건조피부를 유발하게 된다. 아토피 피부염 환자의 비병변 피부에서 표피 분화 단백질 중 많은 비중을 차지하는 로리크린(Loricrin)이나 인볼루크린(Involucrin) 등은 차이가 없는데 유독 필라그린의 발현만이 감소되었다는 보고가 있으며 이를 통해서 필라그린이 아토피 피부염 병인으로 대두되어 활발히 연구되고 있다.
발견 초기와 달리 필라그린 유전자의 LOF 돌연변이(Loss of function mutation)를 가지지 않은 아토피 피부염 환자들이 보고됨에 따라 유전적 요인이 아닌 다른 외부 환경적 요인에 의한 필라그린의 감소 현상에 대한 연구들이 수행되었다.
아토피 피부염 환자들에서 증가되어 있는 것으로 보고된 바 있는 Th2 사이토카인에 의한 필라그린의 발현 감소 현상이 몇몇 논문들에 의해 보고된 바 있다(Kim BE et al., Journal of Investigative Dermatology, 131:1272-1279, 2011; Howell MD et al., J Allergy Clin Immunol,120:150-5, 2007).
아토피 피부염 환자의 각질형성세포에는 고농도의 TSLP(Thymic stromal lymphopoietin)가 존재한다. 비정상적 신호전달의 최상위 단백질이 바로 TSLP 싸이토카인(cytokine)이다. 알러젠이 몸에 침투되면 TSLP 등의 물질이 수지상 세포를 자극하여 반응을 하게 되는데, 이 과정에서 아토피 피부염의 증상이 나타나는 것이다. TSLP는 CD11c+ 수지상세포를 자극해서 TARC(Thymus and activation-regulated chemokine)와 MDC 생성을 증가시킨다. 이러한 사이토카인의 농도는 아토피 피부염의 증상과 밀접한 연관성을 가지고 있다(Holgate ST et al., Trends in Immunology, 28:248-251, 2007).
항균펩타이드(anti-microbial peptide)의 감소는 아토피 피부염 환자의 급성 병변에서 뚜렷하게 관찰된다. 항균펩타이드는 인체의 선천면역 체계를 구성하는 중요한 요소이며 양이온을 띤 펩타이드로서 상피조직에서 생성되어 바이러스, 세균, 진균 등 모든 균에 대하여 광범위한 항균작용을 나타내는 1차 방어선을 구축하고, 세포면역과 획득면역을 활성화시키고, 생체 손상 후 상처회복 기능에 관여한다. 이러한 항균펩타이드의 저하는 아토피 피부염에서 증가되어 있는 Th-2 싸이토카인에 의한 억제 때문이며, 각질형성세포에 IL-4, IL-13을 투여할때 β-디펜신(β-defensin)과 카텔리시딘(cathelicidin)의 증가가 억제됨이 보고되었다.
이뿐만 아니라 휘발성 유기화합물(Volatile Organic Compounds: VOCs)은 증기압이 높아 대기 중으로 쉽게 증발되는 액체 또는 기체 상 유기 화합물로써 대기 중에서 광화학 반응을 일으켜 오존 등 광화학 산화성 물질을 생성시켜 광화학 스모그를 유발하는 발암성 물질로써 최근 알러지성 질환과 관련된 연구가 활발히 진행되고 있다.
본 발명에서는 휘발성 유기화합물(Volatile Organic Compounds: VOCs) 중 환경 위해성 물질인 톨루엔과 아토피 피부염 환자의 병변부에서 과발현되어 있는 싸이토카인인 TSLP에 의한 아토피 피부염 발생 기전을 3차원 피부 모델을 이용하여 제시하고자 한다.
본 발명자들은 아토피 피부염 유사 3차원 피부조직 모델 연구에 기반을 둔 아토피 피부염 치료제의 스크리닝 방법에 이용할 수 있는 동물 대체 모델을 개발하고자 예의 노력한 결과, TSLP 처리에 따른 아토피 피부병변에서 중요한 소견인 필라그린 및 항균펩타이드 유전자의 발현량 감소 및 톨루엔 처리에 따른 필라그린 유전자의 발현량 감소를 3차원 피부조직 모델에서 확인하고, 본 발명을 완성하게 되었다.
발명의 요약
본 발명의 목적은 아토피 피부염 유사 3차원 피부조직 모델 및 이를 이용한 아토피 피부염 치료제의 스크리닝 방법을 제공하는 데 있다.
상기 목적을 달성하기 위하여, 본 발명은 (a) 3차원 피부조직 모델에 TSLP(Thymic stromal lymphopoietin) 또는 톨루엔(Toluene)을 처리하는 단계; (b) 상기 처리된 3차원 피부조직 모델에서 필라그린(Filaggrin) 및 항균펩타이드의 발현을 확인하는 단계; 및 (c) 대조군에 비해 필라그린(Filaggrin)의 발현이 감소된 3차원 피부조직 모델을 아토피 피부염 유사 3차원 피부조직 모델로 수득하는 단계를 포함하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법을 제공한다.
본 발명은 또한, 상기 방법으로 제조되고, 대조군에 비해 필라그린(Filaggrin)의 발현이 감소된 아토피 피부염 유사 3차원 피부조직 모델을 제공한다.
본 발명은 또한, (a) 상기 아토피 피부염 유사 3차원 피부조직 모델에 아토피 피부염 치료제 후보물질을 처리한 다음, 상기 피부조직 모델의 세포에서 필라그린(Filaggrin) 및 항균펩타이드의 발현을 확인하는 단계; 및 (b) 대조군에 비해 상기 필라그린의 발현을 증가시키는 후보물질을 치료제로 선정하는 단계를 포함하는 아토피 피부염 유사 3차원 피부조직 모델을 이용한 아토피 피부염 치료제의 스크리닝 방법을 제공한다.

발명의 상세한 설명 및 바람직한 구현예
본 발명에서는 TSLP(Thymic stromal lymphopoietin) 또는 톨루엔으로 처리된 아토피 피부염 유사 3차원 피부조직 모델을 통해서, TSLP(Thymic stromal lymphopoietin)를 처리한 경우 아토피 피부염 환자에서 발현량이 증가되어 있는 TSLP(Thymic stromal lymphopoietin) 의존적인 필라그린 단백질 및 항균펩타이드의 발현 감소를 확인하고, 톨루엔을 처리한 경우 필라그린 단백질이 감소되는 것을 확인함으로써, 아토피 피부염 치료제의 효능 평가 및 기전 연구 등에 활용할 수 있는 아토피 피부염 유사 3차원 피부조직 모델을 이용한 새로운 동물 대체 시험 방법을 개발하였다.
본 발명의 일 실시예에서는 PBS(Phosphate-Buffered Saline)를 처리한 대조군에 비하여 TSLP(Thymic stromal lymphopoietin)를 처리한 단층(monolayer)으로 배양된 각질세포 내 필라그린(Filaggrin)의 RNA 및 단백질의 발현량이 TSLP 농도 의존적으로 감소하는 것을 확인하였다.
본 발명의 다른 실시예에서는 PBS를 처리한 대조군에 비하여 톨루엔을 처리한 단층으로 배양된 각질세포 내 필라그린(Filaggrin)의 RNA 및 단백질의 발현량이 톨루엔 농도 의존적으로 감소하는 것을 확인하였다.
본 발명의 또 다른 실시예에서는 TSLP 처리에 따른 필라그린(Filaggrin)과 항균펩타이드 발현량의 감소가 3차원 피부 모델 내에서 일어나는 것을 확인하였고, 이는 단층으로 배양된 상기 각질세포에서의 필라그린 발현의 추이와 유사하였다.
본 발명의 또 다른 실시예에서는 톨루엔 처리에 따른 필라그린(Filaggrin) 발현량의 감소가 3차원 피부 모델 내에서 일어나는 것을 확인하였고, 이는 단층으로 배양된 상기 각질세포에서의 필라그린 발현의 추이와 유사하였다..

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1: TSLP 또는 톨루엔 처리에 따른 단층에서 배양된 각질세포에서의 필라그린(Filaggrin)의 발현 변화
1) TSLP 처리
TSLP(Thymic stromal lymphopoietin)에 의한 필라그린(Filaggrin)의 발현 변화를 확인하기 위하여 단층(monolayer)으로 배양된 각질세포에 TSLP를 농도별로 처리한 다음, 상기 세포로부터 RNA와 단백질을 추출하여 필라그린의 발현량의 변화를 확인하였다.
필라그린의 RNA 발현량의 변화를 확인하기 위하여, 사람 각질 세포주(HaCaT cell line; order no.300493, CLS, Eppelheim, Germany; German Cancer Research Center, Heidelberg, Germany)(Boukamp et al., J. Cell Biol. 106:761-771, 1988)를 배양하여 0, 1, 10, 100ng/ml의 TSLP를 16시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), TRIzol 시약을 첨가한 뒤 균질기(homogenizer: OMNI-inc, 미국)를 이용하여 잘게 갈아준 뒤 제조사의 방법에 따라 RNA를 분리하였다. 상기 분리된 RNA는 역전사효소(Super-bio, Korea)를 이용하여 각 조직의 cDNA를 제조하였다. 상기 제조된 cDNA를 주형으로 하여 필라그린(Filaggrin) 프라이머(Filaggrin-f(forward)[서열번호 1]: 5’-ggtctggacgttcagggtct-3’, Filaggrin-r(reverse)[서열번호 2]: 5’-ggatgtggtgtggctgtgat-3’)와 GAPDH 프라이머(GAPDH-f(forward)[서열번호 3]: 5’-AGCAATGCCTCCTGCACCACCAAC-3’, GAPDH-r(reverse)[서열번호 4]: 5’- CCGGAGGGGCCATCCACAGTCT-3’) 프라이머를 이용하여 변성 단계(95℃, 30초), 어닐링 단계(56℃, 30초) 및 연장 단계(72℃, 40초)를 1싸이클로 하여 35싸이클을 실시하여 PCR(Polymerase Chain Reaction)로 증폭하였다. 상기 증폭된 시료는 1% 아가로즈 젤에서 발현 양상을 관찰하였다(도 1A).

필라그린의 단백질 발현량의 변화를 확인하기 위하여, 사람 각질 세포주(HaCaT cell line)를 배양하여 0, 1, 10, 100ng/ml의 TSLP를 16시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), RIPA 버퍼(Elpis, Korea)를 세포에 처리하였고 얼음에 30분간 정치시킨 후 냉장 원심분리기에서 13000rpm, 30분간 원심분리하였고, 상층액만을 취하여 단백질 정량 후 50㎍의 단백질을 SDS-PAGE 젤에 로딩하였다. 그다음 로딩된 젤에 함유된 단백질을 PVDF에 트랜스퍼(transfer)한 후 5% BSA(Bovine Serum Albumin)로 1시간 동안 블로킹한 후 필라그린(Filaggrin) 항체(Santa Cruz, USA)를 1:1000으로 희석한 후 인큐베이션하였다. 그다음 HRP가 부착된 2차 항체 반응과 세척과정을 거친 후 ECL 용액을 이용하여 단백질의 밴드를 확인하였다(도 1B).
필라그린의 단백질 발현량의 변화를 세포 내에서 확인하기 위하여, 배양된 사람 각질 세포주(HaCaT cell line)에 cytoperm/cytofix(BD, USA)를 제공된 실험법에 따라 처리하여 세포를 고정하고 필라그린(Filaggrin) 항체 PBS(Phosphate-Buffered Saline)에 1:100으로 희석하여 4℃에서 1시간 동안 반응시킨 후 PBS로 세척한 뒤 Cy3가 부착된 2차 항체를 1:100으로 희석하여 4℃에서 1시간 동안 반응시킨 후 PBS로 세척한 다음, 형광현미경 하에서 필라그린의 발현을 확인하였다(도 1C).
그 결과, PBS를 처리한 대조군에 비하여 TSLP(Thymic stromal lymphopoietin)를 처리한 단층에서 배양된 각질세포 내 필라그린(Filaggrin)의 RNA 및 단백질의 발현량이 TSLP 농도 의존적으로 감소하는 것을 확인하였다.
2) 톨루엔(Toluene) 처리
톨루엔(Toluene) 처리에 따른 필라그린(Filaggrin)의 발현 변화를 확인하기 위하여 단층으로 배양된 각질세포에 톨루엔을 농도별로 처리한 후 RNA와 단백질 레벨에서의 필라그린의 발현량 변화를 확인하였다.
필라그린의 RNA 발현량의 변화를 확인하기 위하여, 사람 각질 세포주(HaCaT cell line)를 배양하여 0, 12.5, 25, 50nM의 톨루엔을 24시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), TRIzol 시약을 첨가한 뒤 균질기(homogenizer)를 이용하여 잘게 갈아준 뒤 제조사의 방법에 따라 RNA를 분리하였다. 상기 분리된 RNA는 역전사효소(Super-bio, Korea)를 이용하여 각 조직의 cDNA를 제조하였다. 제조된 cDNA를 주형으로 하여 필라그린(Filaggrin) 프라이머(Filaggrin-f(forward)[서열번호 1]: 5’-ggtctggacgttcagggtct-3’, Filaggrin-r(reverse)[서열번호 2]: 5’-ggatgtggtgtggctgtgat-3’)와 GAPDH 프라이머(GAPDH-f(forward)[서열번호 3]: 5’-AGCAATGCCTCCTGCACCACCAAC-3’, GAPDH-r(reverse)[서열번호 4]: 5’- CCGGAGGGGCCATCCACAGTCT-3’) 프라이머를 이용하여 변성단계(95℃, 30초), 어닐링 단계(56℃, 30초) 및 연장 단계(72℃, 40초)를 1싸이클로 하여 35싸이클을 실시하여 PCR로 증폭하였다. 증폭된 시료는 1% 아가로즈 젤에서 발현 양상을 관찰하였다(도 2A).
필라그린의 단백질 발현량의 변화를 확인하기 위하여, 사람 각질 세포주(HaCaT cell line)를 배양하여 0, 12.5, 25, 50nM의 톨루엔을 24시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), RIPA 버퍼(Elpis, Korea)를 상기 세포에 처리하여 얼음에 30분간 정치시킨 후 냉장 원심분리기에서 13000rpm, 30분간 원심분리하고, 상층액만을 취하여 단백질 정량 후 50㎍의 단백질을 SDS-PAGE 젤에 로딩하였다. 그다음 로딩된 젤에 함유된 단백질을 PVDF에 트랜스퍼한 후 5% BSA로 1시간 동안 블로킹한 후 필라그린(Filaggrin) 항체(Santa Cruz, USA)를 1:1000으로 희석한 후 인큐베이션하였다. HRP가 부착된 2차 항체 반응과 세척과정을 거친 후 ECL 용액을 이용하여 단백질의 밴드를 확인하였다(도 2B).
필라그린의 단백질 발현량의 변화를 세포 내에서 확인하기 위하여, 배양된 사람 각질 세포주(HaCaT cell line)에 cytoperm/cytofix(BD, USA)를 제공된 실험법에 따라 처리하여 세포를 고정하고 필라그린(Filaggrin) 항체 PBS에 1:100으로 희석하여 4℃에서 1시간 동안 반응시킨 후 PBS로 세척한 뒤 Cy3가 부착된 2차 항체를 1:100으로 희석하여 4℃에서 1시간 동안 반응시킨 후 PBS로 세척한 뒤 형광현미경 하에서 필라그린의 발현을 확인하였다(도 2C).
그 결과, PBS를 처리한 대조군에 비하여 톨루엔을 처리한 단층에서 배양된 각질세포 내 필라그린(Filaggrin)의 RNA 및 단백질의 발현량이 톨루엔 농도 의존적으로 감소하는 것을 확인하였다.
실시예 2: TSLP 또는 톨루엔 투여에 따른 3차원 피부 모델에서의 필라그린(Filaggrin)과 항균펩타이드의 발현 변화
3차원 피부 모델
실시예 2 및 3에서 사용된 3차원 피부 모델(The EpiDerm™ Skin Model, EPI-100, MatTek)은 신생아의 음경 포피 조직(Neonatal-foreskin tissue) 또는 성인 유방 조직 유래의 정상 인간 상피 각질세포(Normal Human Epithelial Keratinocytes: NHEK)를 구성 성분으로 하는 8∼15 세포층(cell layers)으로 이루어져 있으며, 이는 기저층(basal)-, 가시(spinous)-, 과립(granular)- 및 각질(cornified)-층을 가지는 생체 내(in vivo) 상피조직과 유사하다(Carlson et al., Curr Protoc Cell Biol., Chapter 19:Unit 19.9. doi:10.1002/0471143030.cb1909s41, 2008). 또한, 상기 3차원 피부 모델은 대사 및 세포분열 활성을 가지며, 성숙된 상피 특이적인 분화 마커인 프로-필라그린(pro-filaggrin), K1/K10 사이토케라틴 쌍(cytokeratin pair), 인볼루크린(involucrin) 및 1형 상피 트랜스글루타미나아제(type I epidermal transglutaminase)를 발현한다. 상기 3차원 피부 모델은 상피 성장인자(Epidermal growth factor), 인슐린, 하이드로코티손(hydrocortisone), 항생제 등을 함유하는 DMEM(Dulbecco's Modified Eagle's Medium) 배지에서 통상 3주까지 정상적 상피 형태(epidermal morphology)를 유지하는 가운데 배양가능하다.
상기 3차원 피부 모델을 MatTek사의 배양방법을 이용하여 배양한 다음, 하기와 같이 TSLP(Thymic stromal lymphopoietin) 또는 톨루엔을 투여(처리)하고, 필라그린(Filaggrin) 및 항균펩타이드의 발현 변화를 확인하였다.
1) TSLP 처리
실시예 1의 시험관 내(in vitro) 실험을 통해 확인한 TSLP(Thymic stromal lymphopoietin)에 의한 필라그린(Filaggrin)의 발현 감소 현상이 3차원 피부 모델에서도 일어나는 현상인지 확인하기 위하여 3차원 피부 모델에 TSLP를 처리한 후 필라그린의 발현 변화를 면역조직화학염색법으로 확인하였다.
필라그린의 단백질 발현량의 변화를 확인하기 위하여, 사람의 정상 상피 각질세포(Normal Human Epithelial Keratinocytes)로 구성된 3차원 피부 모델(The EpiDerm™ Skin Model, EPI-100, MatTek)을 MatTek사의 배양방법으로 배양하여 100ng/ml의 TSLP를 16시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), 3차원 피부 모델을 채취(TSLP가 처리된 3차원 피부 모델은 실시예 3의 아토피 피부염 치료제의 스크리닝에 이용)하여 4% 포름알데하이드(Sigma, USA)에 고정한 다음, 파라핀에 포매하여 슬라이드를 제작하였다. 조직 슬라이드를 자일렌을 이용하여 탈파린화 과정과 알코올을 이용하여 탈수화 과정을 거친 후 시트레이트 버퍼에 슬라이드를 담궈 전자레인지에서 10분간 돌린 후 실온에서 한 시간 동안 식혀주었다. 증류수로 상기 슬라이드를 세척하여 실온에서 5% 염소 혈청으로 한 시간 동안 반응시킨 후 항-필라그린(Filaggrin), 항-β-디펜신(β-defensin), 항-서라이어신(Psoriasin) 항체를 1:50으로 희석하여 인큐베이션하였다. HRP가 부착된 2차 항체를 반응시킨 후 DAB 키트를 이용하여 발색시켜 조직 내 필라그린(Filaggrin)의 발현 변화를 확인하였다.
그 결과, PBS를 투여한 대조군에 비하여 TSLP를 투여한 3차원 피부 모델 내 필라그린(Filaggrin)과 항균펩타인드인 β-디펜신(β-defensin) 및 서라이어신(Psoriasin)의 발현량이 TSLP 농도 의존적으로 감소하는 것을 확인하였고, 이는 단층으로 배양된 상기 각질세포에서의 필라그린 발현의 추이와 유사하였다.
2) 톨루엔(Toluene) 처리
실시예 1의 시험관 내(in vitro) 실험을 통해 확인한 톨루엔에 의한 필라그린(Filaggrin)의 발현 감소 현상이 3차원 피부 모델에서도 일어나는 현상인지 확인하기 위하여 3차원 피부 모델에 톨루엔을 처리한 후 필라그린의 발현 변화를 면역조직화학염색법으로 확인하였다.
필라그린의 단백질 발현량의 변화를 확인하기 위하여, 3차원 피부 모델(The EpiDerm™ Skin Model, EPI-100, MatTek)을 MatTek사의 배양방법으로 배양하여 50μM의 톨루엔을 24시간 동안 배양액에 처리한 다음(대조군은 PBS(Phosphate-Buffered Saline)로 처리), 3차원 피부 모델을 채취(톨루엔이 처리된 3차원 피부 모델은 실시예 3의 아토피 피부염 치료제의 스크리닝에 이용)하여 4% 포름알데하이드(Sigma, USA)에 고정한 다음, 파라핀에 포매하여 슬라이드를 제작하였다. 조직 슬라이드를 자일렌을 이용하여 탈파린화 과정과 알코올을 이용하여 탈수화 과정을 거친 후 시트레이트 버퍼에 슬라이드를 담궈 전자레인지에서 10분간 돌린 후 실온에서 한 시간 동안 식혀주었다. 증류수로 상기 슬라이드를 세척하고, 실온에서 5% 염소 혈청으로 한 시간 동안 반응시킨 후 필라그린(Filaggrin) 항체를 1:50으로 희석하여 인큐베이션하였다. 그 결과, PBS를 투여한 대조군에 비하여 톨루엔을 투여한 3차원 피부 모델 내 필라그린(Filaggrin)의 발현량이 톨루엔 농도 의존적으로 감소하는 것을 확인하였고, 이는 단층으로 배양된 상기 각질세포에서의 필라그린 발현의 추이와 유사하였다.
통계처리
실시예 1 및 실시예 2에서의 통계분석은 두 그룹 간 유의성 검사로 t-test 검정법을 이용하였고, 대조군과 비교하여 *P<0.05, **P<0.01, ***P<0.001인 경우 유효한 차이로 간주하였다. 모든 데이터는 평균(Mean)±측정표준오차(SEM)로 나타내었다.
실시예 3: 아토피 피부염 유사 3차원 피부조직 모델을 이용한 아토피 피부염 치료제의 스크리닝 방법
실시예 2와 동일한 방법으로 3차원 피부조직 모델(The EpiDerm™ Skin Model, EPI-100, MatTek)에 TSLP 또는 톨루엔을 투여하여 수득한 아토피 피부염 유사 3차원 피부조직 모델을 이용하여, 대조군은 PBS(Phosphate-Buffered Saline) 또는 DMSO(Dimethyl sulfoxide)를 처리하였고, 실험군은 아토피 피부염 치료 후보물질을 처리한 다음, 필라그린 및 항균펩타이드(β-디펜신(β-defensin) 또는 서라이어신(Psoriasin))의 발현을 역전사-중합효소 연쇄반응(Reverse Transcription-PCR), 웨스턴블랏 및/또는 면역조직화학염색법을 이용하여 분석하였다. 여기서, 아토피 피부염 유사 3차원 피부조직 모델에 처리한 아토피 피부염 치료 후보물질, PBS(Phosphate-Buffered Saline) 또는 DMSO(Dimethyl sulfoxide)는 당업계의 통상적인 방법으로 적절한 농도와 노출 시간에서 처리하였다(Thomas K. Petersen, Basic & Clinical Pharmacology & Toxicology, 99:104-115, 2006). 대조군에 비해 실험군의 필라그린 및 항균펩타이드 발현을 증가시키는 후보물질을 잠재적인 아토피 피부염 치료제로 평가하고 선정하였다.
본 발명의 TSLP 또는 톨루엔 처리에 따른 3차원 피부조직 모델은 아토피 피부염의 피부병변에서 중요한 소견인 필라그린과 항균펩타이드인 β-디펜신 또는 서라이어신의 유전자 발현 감소를 보여줌으로써 아토피 피부염 유사 피부조직 모델로서의 가치가 크며, 아토피 피부염 치료제의 효능 평가 및 기전 연구 등에 활용할 수 있는 새로운 동물 대체 시험 방법을 제공할 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

Claims (15)

  1. 다음 단계를 포함하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법:
    (a) 3차원 피부조직 모델에 TSLP(Thymic stromal lymphopoietin) 또는 톨루엔(Toluene)을 처리하는 단계;
    (b) 상기 처리된 3차원 피부조직 모델에서 필라그린(Filaggrin) 및 항균펩타이드의 발현을 확인하는 단계; 및
    (c) 대조군에 비해 필라그린(Filaggrin)의 발현이 감소된 3차원 피부조직 모델을 아토피 피부염 유사 3차원 피부조직 모델로 수득하는 단계.
  2. 제1항에 있어서, 상기 3차원 피부조직 모델을 구성하는 세포는 인간의 각질세포(keratinocytes)인 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법.
  3. 제1항에 있어서, 상기 항균펩타이드는 β-디펜신(β-defensin) 또는 서라이어신(Psoriasin)인 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법.
  4. 제1항에 있어서, 상기 필라그린(Filaggrin) 및 항균펩타이드의 발현 확인은 역전사-중합효소 연쇄반응(Reverse Transcription-PCR), 웨스턴블랏 또는 면역조직화학염색법을 이용하는 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법.
  5. 제1항에 있어서, 상기 (c) 단계는 대조군에 비해 필라그린(Filaggrin)의 발현이 감소하고, 항균펩타이드의 발현이 추가로 감소된 3차원 피부조직 모델을 아토피 피부염 유사 3차원 피부조직 모델로 수득하는 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델의 제조방법.
  6. 제1항 내지 제5항 중 어느 한 항의 방법으로 제조되고, 대조군에 비해 필라그린(Filaggrin)의 발현이 감소된 아토피 피부염 유사 3차원 피부조직 모델.
  7. 제6항에 있어서, 상기 3차원 피부조직 모델을 구성하는 세포는 인간의 각질세포(keratinocytes)인 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델.
  8. 제6항에 있어서, 대조군에 비해 필라그린(Filaggrin)의 발현이 감소하고, 항균펩타이드의 발현이 추가로 감소된 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델.
  9. 제8항에 있어서, 상기 항균펩타이드는 β-디펜신(β-defensin) 또는 서라이어신(Psoriasin)인 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델.
  10. 제6항 내지 제9항 중 어느 한 항에 있어서, 상기 필라그린(Filaggrin) 및 항균펩타이드의 발현 확인은 역전사-중합효소 연쇄반응(Reverse Transcription-PCR), 웨스턴블랏 또는 면역조직화학염색법을 이용하는 것을 특징으로 하는 아토피 피부염 유사 3차원 피부조직 모델.
  11. 다음 단계를 포함하는 아토피 피부염 유사 3차원 피부조직 모델을 이용한 아토피 피부염 치료제의 스크리닝 방법:
    (a) 제1항 내지 제5항 중 어느 한 항의 방법으로 제조된 아토피 피부염 유사 3차원 피부조직 모델에 아토피 피부염 치료제 후보물질을 처리한 다음, 상기 피부조직 모델의 세포에서 필라그린(Filaggrin) 및 항균펩타이드의 발현을 확인하는 단계; 및
    (b) 대조군에 비해 상기 필라그린의 발현을 증가시키는 후보물질을 치료제로 선정하는 단계.
  12. 제11항에 있어서, 상기 3차원 피부조직 모델을 구성하는 세포는 인간의 각질세포(keratinocytes)인 것을 특징으로 하는 아토피 피부염 치료제의 스크리닝 방법.
  13. 제11항에 있어서, 상기 항균펩타이드는 β-디펜신(β-defensin) 또는 서라이어신(Psoriasin)인 것을 특징으로 하는 아토피 피부염 치료제의 스크리닝 방법.
  14. 제11항에 있어서, 상기 필라그린(Filaggrin) 및 항균펩타이드의 발현 확인은 역전사-중합효소 연쇄반응(Reverse Transcription-PCR), 웨스턴블랏 또는 면역조직화학염색법을 이용하는 것을 특징으로 하는 아토피 피부염 치료제의 스크리닝 방법.
  15. 제11항에 있어서, 상기 (b) 단계는 대조군에 비해 필라그린(Filaggrin)의 발현을 증가시키고, 항균펩타이드의 발현을 추가로 증가시키는 후보물질을 치료제로 선정하는 것을 특징으로 하는 아토피 피부염 치료제의 스크리닝 방법.
