[方法]
HTRF法(Homogeneous Time Resolved Fluorescence)に基づくハイスループットスクリーニング

上記と同様のHTRFアッセイにより、ジスルフィラムによるフロント-CCR2相互作用及びフロント-CCR5相互作用の阻害能を調べた。CCR5 pro-Cペプチドとして、CCR5のC末端領域内の16残基EKFRNYLLVFFQKHIA(配列番号4)を用いた。コントロールとして、フロントとCCR2、CCR5との間の相互作用と同様のへリックスペプチド-タンパク質相互作用ではあるがこれらとは無関係なp53-MDM2相互作用(MDM-2特異的阻害剤であるNutlinにより阻害される)を用いた。

フロントタンパク質の第500番~第656番アミノ酸の領域からなる断片と、16残基のCCR2 pro-C領域断片(配列番号3)とを調製し、両者の結合状態及びフロントタンパク質とジスルフィラムとの結合状態をNMR解析により調べた。
SPR実験により、ジスルフィラムがCCR2ではなくフロントタンパク質に直接結合していることが明らかとなった(図2a)。NMR解析では、フロント由来のシグナルがジスルフィラム添加により影響され、フロントとジスルフィラムが直接的に相互作用していることが示唆された(図2b)。また、ジスルフィラムの代謝産物であるジメチルジチオカルバメートもジスルフィラムと同じフロント上の結合部位に結合することが示唆された(データ省略)。
[方法]
腫瘍モデル
皮下での腫瘍の増殖を評価するため、LLC(ルイス肺癌)細胞(5×105)を50μLのPBSに懸濁してマウスの右側腹部に皮下注射した。週に2回、ノギスを用いて腫瘍サイズを測定した。腫瘍体積は下記の式により算出した。
腫瘍体積=(腫瘍短径)2×腫瘍長径/2
ジスルフィラムを経口で与えたマウス群では、コントロール群と比較して腫瘍体積が有意に減少していた(図3)。ジスルフィラムによりフロントを阻害することで、生体内に移植した肺癌細胞の増殖が抑制されることが明らかとなった。
[方法]
肺転移モデルとして、1×106個のメラノーマ細胞B16F10を200μLのPBSに懸濁し、野生型C57BL/6マウスの尾静脈より投与した。阻害剤の投与は、腫瘍投与の1日前、30分前、及び1日後に行なった。コントロールとして、DMSO及びフッ化ピリミジン系の抗がん剤5-FUを投与した。腫瘍投与後9日目にマウスを安楽死させ、左心室からPBSを灌流させた後に肺を分離した。左葉中の目視可能な肺転移をカウントした。
腫瘍投与後9日目における転移結節の写真、及び転移の計数結果のグラフを図4に示す。ジスルフィラムは移植されたB16F10細胞による肺転移の形成を有意に低減した。
[方法]
細胞傷害性及び増殖アッセイ
阻害剤の細胞傷害性は、LDH Cytotoxicity Detection Kit(TaKaRa)を製造者の指示書に従い用いて調べた。簡潔には、腫瘍細胞を薬剤(ジスルフィラム、5FU)の存在下又は非存在下で72時間培養後、培養上清を回収し、損傷細胞から放出された乳酸脱水素酵素の濃度を調べた。細胞増殖の測定では、細胞を阻害剤と共にプレートウェル中で48時間インキュベートし、最後の30分の間にWST-1(同仁化学研究所)を培養液に添加、EnVision(PerkinElmer)を用いて各ウェルの650nm参照波長に対する450nmでの吸光度を測定し、フォルマザンレベルを検出した。
近年、ある種の腫瘍細胞に対しジスルフィラムが直接腫瘍細胞を殺傷する活性があることが報告されている(Chen, D., et al., Cancer Res 66, 10425-10433, (2006)、及びChiba, T. et al., PLoS One 9, e84807, (2014))。ジスルフィラムの抗腫瘍活性が直接的な腫瘍細胞の殺傷によるものか否かを調べるため、細胞傷害性を有する抗がん剤である5FUとジスルフィラムの抗がん作用及び細胞傷害活性を比較した。その結果、ジスルフィラムは、5FUがLLC腫瘍細胞を完全に殺傷する濃度と同濃度において、細胞傷害活性を示さなかった(図5a)。5FUは、B16F10メラノーマ細胞に対しても細胞傷害活性及び増殖阻害作用を示したが、ジスルフィラムは同濃度においても腫瘍細胞に対して何らの作用も示さなかった(図5b)。また、図4に示される通り、5FUは腫瘍殺傷活性があるにもかかわらずB16F10腫瘍の転移を低減しなかったが、その一方、ジスルフィラムはそれと同じ用量で腫瘍の転移を効果的に減少させた。これらの結果は、ジスルフィラムが示すB16F10メラノーマ細胞及びLLC細胞に対する抗腫瘍効果は細胞傷害活性によるものではなく、宿主の腫瘍微小環境に対する作用によるものであることを示している。
[方法]
フロント-gfp-ノックインマウスの作出
常法により、マウスのゲノム上のフロントプロモーターの下流にGFP遺伝子を組み込み、フロント-gfpノックインマウスを作出した。
2 mLの4%チオグリコレートを腹腔内投与して腹膜炎を誘導したマウスより、腹膜の浸潤細胞を採取した。2%ウシ胎仔血清を添加したPBSにて細胞を洗浄、再懸濁し、70μmのストレイナーで濾過した。抗マウスCD16/32抗体(BD biosciences)でインキュベーションすることでFcレセプターをブロッキングした後、蛍光標識抗体で細胞を染色した。抗マウスCD11b-Pacific Blue、抗マウスLy6C-APC-Cy7、抗マウスLy6G-Alexa Fluor 700、抗マウスCD4-FITC及び抗マウスB220-PE-Cy7はBiolegendより購入した。抗マウスCD8-Pacific BlueはBD biosciencesより購入した。
フロント-GFPノックインマウス由来の細胞のフローサイトメトリー解析によると、腹膜炎モデルの炎症部位にリクルートされた免疫細胞の中でも特にマクロファージにおいてフロントが高発現していた(図6)。CCL2で動員された細胞集団ではフロント高発現の単球/マクロファージが濃縮されており、その受容体であるCCR2を発現している細胞においてフロントが高発現していることが示された(データ省略)。
腫瘍微小環境におけるフロントの役割を調べるため、cre/loxPシステムを用いてフロントをノックアウトしたマウスを作出した。フロントは完全欠損で胎生致死となるため、タモキシフェン依存的に組み換え反応が誘導されるシステムを用いてコンディショナルなノックアウトを行なった。
5×105個のB16メラノーマ細胞をFNTflox/floxマウス及びFNT-cKOマウスの右側腹部に移植し、その後週2回、腫瘍サイズをノギスで測定し、腫瘍体積を算出した。その結果、非ノックアウト(FNTflox/flox)マウスに比べてFNT-cKOマウスでは腫瘍の増殖が有意に抑制され、また生存率も向上することが明らかとなった(図7a, b)。
1×106個のB16メラノーマ細胞をFNTflox/floxマウス及びFNT-cKOマウスに静脈内投与し、投与8日目に肺の転移結節の数及びサイズを目視で確認した。その結果、転移結節の数もサイズもFNT-cKOマウスで有意に低下することが明らかとなった(図8a, b)。
[方法]
in vivo走化性アッセイ
FNTflox/floxマウス及びFNT-cKOマウスに4%チオグリコレート培地(Difco社)2mLを腹腔内投与することで腹膜炎を誘導させた。腹腔中に5mLの氷冷PBSを注入して穏やかにマッサージすることで、腹膜の浸潤細胞を収集した。収集した細胞集団を、0.1%FBSを含むPBSで洗浄し、フローサイトメトリー解析に付して細胞数及び細胞ポピュレーションを調べた。
結果を図9に示す。フロント欠損マウスではマクロファージ数が低下しており、腹膜炎症部位へのマクロファージの浸潤が低下することが確認された(図9上段)。好中球数はフロント欠損マウスと非欠損マウスの間で差異がなかった(図9下段)。
[方法]
in vivo走化性アッセイ
野生型マウスに上記8と同様の処理を行ない、腹膜炎を誘導し、腹膜の浸潤細胞を収集した。収集した細胞集団を洗浄後、フローサイトメトリー解析により細胞数及び細胞ポピュレーションを調べた。ジスルフィラムの評価のため、DMSOに溶解し2% Tween 80含有生理食塩水で希釈した、図示した濃度のジスルフィラムをチオグリコレート投与の1日前と30分前にマウスに投与した。
フロント阻害剤であるジスルフィラムを野生型マウスに投与した場合でも、フロント欠損マウスと同様に、腹膜炎症部位へのマクロファージの浸潤は低下し、好中球の浸潤は影響されなかった(図10)。
[方法]
in vitro走化性アッセイ
ヒト白血球細胞株THP-1を計数してBoydenバッファー(0.1% BSA含有RPMI培地)中に再懸濁し、走化性アッセイ前にジスルフィラムと共にプレインキュベートした。ポリカーボネートフィルターのポアサイズが5-μmである96ウェルのChemoTXケモタキシスチャンバー(Neuro probe)を用いて細胞遊走活性を測定した。プレートの下部チャンバー中にケモカインをアプライし、上部チャンバー上に細胞をアプライした。37℃、5%CO2で90分間インキュベートした後、フィルターを除去し、Cell Counting Kit F(同仁化学研究所)を用いて下部チャンバー中の遊走細胞数をカウントした。
細胞毒性を示さない濃度(データ省略)でのジスルフィラム処理により、CCL2が媒介する細胞遊走が阻害された(図11)。
[方法]
フローサイトメトリー
上記4に記載した野生型マウスの肺転移モデル、又は上記7(2)に記載したFNT-cKOマウス及びFNTflox/floxマウスの肺転移モデルの肺細胞を、右下葉よりコラゲナーゼ及びDNaseで消化させて得た。2%ウシ胎仔血清を添加したPBSにて細胞を洗浄、再懸濁し、70μmのストレイナーで濾過した。抗マウスCD16/32抗体(BD biosciences)でインキュベーションすることでFcレセプターをブロッキングした後、蛍光標識抗体で細胞を染色した。抗マウスCD11b-Brilliant Violet 510、抗マウスLy6C-APC-Cy7、抗マウスLy6G-Alexa Fluor 700はBiolegendより購入した。染色した細胞は、Galliosフローサイトメーター(Beckman coulter)を用いて解析した。
マウスをPBSで灌流し、左肺を分離した。気管からOptimal Cutting Temperatureコンパウンド(OCT)(サクラファインテック)を注入し、OCT中に包埋して液体窒素で凍結させた。8μm厚の新鮮凍結切片を調製し、4%パラホルムアルデヒド-PBSで固定した。0.05% Tween 20-PBSで洗浄後、切片をBlocking One試薬(ナカライテスク)でブロッキングし、抗マウスF4/80抗体(BioLegend)及びAlexa Fluor 594 抗ラットIgG (Life technologies)で順次染色した。SP5共焦点顕微鏡(Leica Microsystems)により蛍光像を取得した。
フロント阻害剤であるジスルフィラムを野生型マウスに投与すると、溶媒投与マウスと比較して、腫瘍部位へのマクロファージの集積が低下していた(図12)。フロント欠損マウスにおいても、非欠損マウスと比較してがん転移結節へのマクロファージの集積が有意に低下していた(データ省略)。
[方法]
抗がん剤とジスルフィラム(DSF)の併用の一例として、免疫チェックポイント分子をターゲットとする抗がん剤である抗PD-1抗体とDSFの併用効果をLLC担癌モデル及びB16担癌モデルで検討した。B16担癌モデルは、5×105個のB16メラノーマ細胞を野生型C57BL/6マウスの右側腹部に移植して作製した。LLC担癌モデルの作製は上記3.と同様に行なった。DSF添加飼料は上記3.と同様とした。
LLC担癌モデルでの結果を図13に示す。LLC担癌モデルにおいて、抗PD-1抗体とDSFは単独でもがん細胞の増生を抑制したが、併用により相加効果が見られ、抗PD-1抗体のがん増生抑制効果はDSFの併用により相加的に増強することが確認された。
[方法]
抗がん剤とDSFの併用のさらなる一例として、高い細胞傷害活性を有する抗CD4抗体とDSFの併用効果をLLC担癌モデル及びB16担癌モデルで検討した。LLC担癌モデル及びB16担癌モデルは上記12.と同様に作製した。DSF添加飼料は上記12.と同様とした。腫瘍細胞の移植後、週に2回、腫瘍サイズをノギスで測定し、腫瘍体積を算出した。
腫瘍体積を経時的に測定した結果を図15に示す。LLC担癌モデル(左)、B16担癌モデル(右)のいずれでも、抗CD4抗体によるがん増生抑制効果がDSFの併用により有意に増強することが確認された。
Claims (21)
- 下記(1)~(3)のいずれかを有効成分として含有する、マクロファージ抑制剤。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - マクロファージの遊走又は組織浸潤の抑制に用いられる請求項1記載の剤。
- 炎症性疾患の炎症病変部又はがん病変部におけるマクロファージの浸潤の抑制に用いられる、請求項2記載の剤。
- 下記(1)~(3)のいずれかを有効成分として含有する、がん微小環境又は炎症性微小環境の構成細胞の制御剤。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - 下記(1)~(3)のいずれかを有効成分として含有する、CCR2B又はCCR5とフロントタンパク質との間の相互作用の阻害剤。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - 1種以上の抗がん剤と組み合わせて用いられる、請求項1ないし5のいずれか1項に記載の剤。
- 前記抗がん剤が抗体又はその抗原結合性断片である、請求項6記載の剤。
- 前記抗がん剤が、抑制性の免疫チェックポイント分子に対するアンタゴニスト、共刺激性の免疫チェックポイント分子に対するアゴニスト、細胞傷害活性を有する抗CD4抗体、並びに細胞毒成分を結合させた抗CD4抗体及びその抗原結合性断片から選択される少なくとも1種である、請求項6記載の剤。
- 抑制性の免疫チェックポイント受容体に対するアンタゴニスト性抗体及びその抗原結合性断片、抑制性の免疫チェックポイントリガンドに対する抗体及びその抗原結合性断片、共刺激性の免疫チェックポイント受容体に対するアゴニスト性抗体及びその抗原結合性断片、細胞傷害活性を有する抗CD4抗体、並びに細胞毒成分を結合させた抗CD4抗体及びその抗原結合性断片から選択される少なくとも1種である、請求項8記載の剤。
- 前記抗がん剤が、アンタゴニスト性抗PD-1抗体、及び細胞傷害活性を有する抗CD4抗体から選択される少なくとも1種である、請求項9記載の剤。
- 下記(1)~(3)のいずれかを有効成分として含有する、抗がん剤の抗がん活性の増強剤。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - フロントタンパク質の第564番~第600番アミノ酸の領域を含むフロントタンパク質断片と、配列番号6に示すCCR2Bのアミノ酸配列中の第312番~第323番アミノ酸の領域を含むCCR2B断片又は配列番号8に示すCCR5のアミノ酸配列中の第304番~第315番アミノ酸の領域を含むCCR5断片を、ジスルフィラム誘導体のライブラリーとインキュベートする工程、及び
フロントタンパク質断片とCCR2B断片又はCCR5断片との間の結合を阻害する活性がジスルフィラムよりも高いジスルフィラム誘導体を選択する工程
を含む、フロントタンパク質とCCR2B又はCCR5との間の相互作用阻害能が向上したジスルフィラム誘導体を同定する方法。 - ジスルフィラム誘導体のアルデヒドデヒドロゲナーゼ阻害活性を測定し、該阻害活性がジスルフィラムよりも低いジスルフィラム誘導体を選択する工程をさらに含む、請求項12記載の方法。
- 請求項12又は13記載の方法により、フロントタンパク質とCCR2B又はCCR5との間の相互作用阻害能が向上したジスルフィラム誘導体を同定する工程、及び
同定されたジスルフィラム誘導体を製造する工程
を含む、フロントタンパク質とCCR2B又はCCR5との間の相互作用の阻害剤の製造方法。 - フロントタンパク質の第564番~第600番アミノ酸の領域を含むフロントタンパク質断片と、配列番号6に示すCCR2Bのアミノ酸配列中の第312番~第323番アミノ酸の領域を含むCCR2B断片又は配列番号8に示すCCR5のアミノ酸配列中の第304番~第315番アミノ酸の領域を含むCCR5断片を、フロント阻害剤候補化合物の誘導体のライブラリーとインキュベートする工程、及び
フロントタンパク質断片とCCR2B断片又はCCR5断片との間の結合を阻害する活性がもとの候補化合物よりも高い誘導体を選択する工程
を含む、フロントタンパク質とCCR2B又はCCR5との間の相互作用阻害能が向上した化合物を同定する方法。 - フロントタンパク質の原子座標の少なくとも一部を用いて、M564、T565、I568、A569、M575、L578、L600からなる群より選択される少なくとも1つを含むアミノ酸残基で構成されるフロントタンパク質の結合ポケット構造をインシリコで構築し、化合物ライブラリーに対して前記結合ポケット構造との結合の強さを計算し、フロントタンパク質との安定な複合体を形成する化合物を選択することを含む、フロントタンパク質とCCR2B又はCCR5との間の相互作用を阻害する物質を同定する方法。
- フロントタンパク質とCCR2B又はCCR5とが結合する結合ポケット構造に対し、候補化合物をインシリコで結合させ、結合の強さを計算することを含む、フロントタンパク質とCCR2B又はCCR5との間の相互作用を阻害する物質の設計方法。
- それを必要とする対象に対し、下記(1)~(3)のいずれかの有効量を投与することを含む、マクロファージを抑制する方法。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - それを必要とする対象に対し、下記(1)~(3)のいずれかの有効量を投与することを含む、がん微小環境又は炎症性微小環境の構成細胞を抑制する方法。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - それを必要とする対象に対し、下記(1)~(3)のいずれかの有効量を投与することを含む、CCR2B又はCCR5とフロントタンパク質との間の相互作用を阻害する方法。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
(2) (1)の薬剤的に許容される塩
(3) (1)若しくは(2)の溶媒和物 - それを必要とする対象に対し、下記(1)~(3)のいずれかの有効量を投与することを含む、抗がん剤の抗がん活性を増強する方法。
(1) ジスルフィラム、ジエチルジチオカルバメート、若しくはジエチルジチオカルバメートの金属錯体
