Die vorliegende Erfindung betrifft feste, oral applizierbare, (2-{[(1S)-1-(3-Chlorphenyl)-2-fluorethyl]amino}-7-methoxy-1,3-benzoxazol-5-yl)[(2S,5S)-5-(2-hydroxyethyl)-2-methyl-morpholin-4-yl]methanon (I) in amorpher Form enthaltende pharmazeutische Darreichungsformen mit schneller Wirkstofffreisetzung sowie Verfahren zu ihrer Herstellung, ihre Anwendung als Arzneimittel, ihre Verwendung zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen sowie ihre Verwendung zur Herstellung eines Arzneimittels zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen.

Feste, oral applizierbare pharmazeutische Darreichungsformen mit schneller Wirkstofffreisetzung
Die vorliegende Erfindung betrifft feste, oral applizierbare, (2-{ [(llS)-l-(3-Chlorphenyl)-2- fluorethyl]amino }-7-methoxy-l,3-benzoxazol-5-yl)[(2lS,,5lS)-5-(2-hydroxyethyl)-2-methyl- morpholin-4-yl]methanon (I) in amorpher Form enthaltende pharmazeutische Darreichungsformen mit schneller Wirkstofffreisetzung sowie Verfahren zu ihrer Herstellung, ihre Anwendung als Arzneimittel, ihre Verwendung zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen sowie ihre Verwendung zur Herstellung eines Arzneimittels zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen. (2- { [( 1 S)- 1 -(3-Chlorphenyl)-2-fluorethyl] amino } -7-methoxy- 1 ,3-benzoxazol-5-yl) [(2S,5S)-5-(2- hydroxyethyl)-2-methyl-morpholin-4-yl]methanon mit der folgenden Formel (I)
ist ein niedermolekularer, oral applizierbarer Inhibitor des Blutgerinnungsfaktors IIa, der zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung verschiedener thromboembolischer Erkrankungen eingesetzt werden kann. Wenn im Folgenden vom Wirkstoff (I) die Rede ist, so ist dabei die amorphe Form von (2-{ [(llS')-l-(3-Chlorphenyl)-2-fluorethyl]amino}-7-methoxy-l,3- benzoxazol-5-yl)[(2lS',5lS')-5-(2-hydroxyethyl)-2-methyl-morpholin-4-yl]methanon (I) sowie die jeweiligen Hydrate und/oder Solvate mit umfasst.
Bei der Formulierungsentwicklung sind die physikalisch-chemischen und biologischen Eigenschaften des Wirkstoffes (I) zu berücksichtigen. Der Wirkstoff (I) liegt in amorpher Form vor. Er zeigt eine geringe Wasserlöslichkeit (ca. 230 mg/L; 25 °C). In organischen Lösemitteln wird eine höhere Löslichkeit beobachtet (z.B. in Ethanol > lOOOmg/100 mL, in Aceton > 3000 mg/mL). Der Wirkstoff (I) ist hygroskopisch.
Ziel der Entwicklung war es daher, eine geeignete Formulierung für den Wirkstoff (I) in amorpher Form zu identifizieren, wobei die Formulierung eine gute Freisetzungsrate und eine gute Bio Verfügbarkeit des Wirkstoffes (I) aufweist.
Überraschenderweise wurde nun gefunden, dass pharmazeutische Darreichungsformen die nach der Lösemethode hergestellt werden, den Wirkstoff (I) in amorpher Form enthalten und eine gute Freisetzungsrate und eine gute Bioverfügbarkeit des Wirkstoffes (I) aufweisen.
Gegenstand der vorliegenden Erfindung sind feste, oral applizierbare pharmazeutische Dar- reichungsformen mit schneller Wirkstofffreisetzung, enthaltend (2-{ [(llS)-l-(3-Chlorphenyl)-2- fluorethyl]amino }-7-methoxy-l,3-benzoxazol-5-yl)[(2lS,,5lS)-5-(2-hydroxyethyl)-2-methyl- morpholin-4-yl]methanon (I), dadurch gekennzeichnet, dass
(a) sie Wirkstoff (I) in amorpher Form enthalten und
(b) dass 80% des Wirkstoffes (I) über einen Zeitraum von maximal 2 Stunden gemäß USP- Freisetzungsmethode mit Apparatur 2 (Paddle; 50-100 UpM) freigesetzt werden.
Die freizusetzende Menge von 80% des Wirkstoffes (I) bezieht sich auf die in der Darreichungsform enthaltene Gesamtmenge an Wirkstoff (I).
In einer bevorzugten Ausführungsform der vorliegenden Erfindung wird durch das Vorliegen des Wirkstoffes (I) in amorpher Form eine schnelle Freisetzungsrate des Wirkstoffes (I) erreicht. Vorzugsweise enthalten die erfindungsgemäßen Darreichungsformen Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff (I) von > 2% bezogen auf die Gesamtmasse der Formulierung, bevorzugt in einer Konzentration von 5 bis 50% bezogen auf die Gesamtmasse der Formulierung, besonders bevorzugt in einer Konzentration von 5 bis 30% bezogen auf die Gesamtmasse der Formulierung. Zur Herstellung der erfindungsgemäßen pharmazeutischen Darreichungsformen kann die amorphe Form von (2- { [( IS)- 1 -(3-Chlorphenyl)-2-fluorethyl]amino } -7-methoxy- 1 ,3-benzoxazol-5- yl)[(2lS,,5lS)-5-(2-hydroxyethyl)-2-methyl-morpholin-4-yl]methanon (I) sowie die jeweiligen Hydrate, Solvate und Co-Kristallisate eingesetzt werden.
Zur Amorphisierung von Wirkstoffen und zur Erzeugung bzw. zur Stabilisierung dieser Wirkstoffformen in Formulierungen sind verschiedene pharmazeutisch geeignete Herstellmethoden möglich. Häufig werden die Lösemethode, das Schmelz verfahren oder eine Kombination dieser beiden Verfahren eingesetzt [Chiou, W.L.; Riegelman, S., „Pharmaceutical Applications of Solid Dispersion Systems", Journal of Pharmaceutical Sciences 60 (1971), 1281- 1302; Ford, J. L.,„The Current Status of Solid Dispersions", Pharm. Acta Helv. 61 (1986), 69-88; Rasenack, N.,„Poorly Water-soluble Drugs for Oral Delivery - A Challenge for Pharmaceutical Development, Part ΠΙ: Drug delivery Systems containing the drug molecularly dispersed / Aspects
on in vitro and in vivo characterization", Pharmazeutische Industrie 67, Nr. 5 (2005), 583-591]. Diese Herstellverfahren resultieren in einem als "feste Dispersion" bezeichneten Zustand des Wirkstoffs in der pharmazeutischen Darreichungsform. Im Folgenden werden die unterschiedlichen Arten„fester Dispersionen" wie feste Lösungen, glasartige Lösungen, glasartige Suspensionen, amorphe Fällungen in einem kristallinen Träger, Eutektika oder Monotektika, Wirkstoffkomplexe und Kombinationen der selbigen auf den Begriff„feste Dispersion" bezogen.
Neben Substanzen aus dem Polymerbereich ist auch für eine Reihe niedermolekularer, normalerweise kristallin vorliegender Substanzen ein amorpher Zustand beobachtet worden. So neigen auch manche Wirkstoffe beim raschen Abkühlen ihrer Schmelze oder beim Trocknen dazu, glasartig zu erstarren. Ebenfalls kann starke mechanische Beanspruchung, wie sie z.B. beim Vermählen auftritt, zu Amorphisierung führen. Dieser amorphe Zustand ist als nicht stabil bzw. nur metastabil zu bezeichnen. Daher kann es im Verlaufe weiterer Verarbeitung zu Rekristallisationserscheinungen kommen. Die Eigenschaften, wie z.B. mechanisches Verhalten, Hygroskopizität und Löslichkeit des amorphen Stoffes unterscheiden sich erheblich von denen der kristallinen Substanz. Verständlicherweise kann dies zu erheblichen Veränderungen der Produkteigenschaften einer pharmazeutischen Zubereitung führen. Zur Gewährleistung einer einheitlichen und gleichbleibenden Qualität des pharmazeutischen Produktes, auch über den langen Zeitraum des Inverkehrbringens und vorrätig Haltens hinaus, ist der amorphe Zustand des Wirkstoffs in der pharmazeutischen Zubereitung zu stabilisieren. Hier finden die gleichen pharmazeutischen Herstellmethoden Anwendung wie zur Amorphisierung ursprünglich kristalliner Wirkstoffe. Häufig werden die Lösemethode, das Schmelzverfahren oder eine Kombination dieser beiden Verfahren eingesetzt [Chiou, W.L.; Riegelman, S., „Pharmaceutical Applications of Solid Dispersion Systems", Journal of Pharmaceutical Sciences

Wirkstoffe können auch gleichzeitig amorphe und kristalline Bereiche aufweisen. Hier wäre ebenfalls der einheitlich amorphe Zustand mittels der beschriebenen Verfahren und aus den aufgeführten Qualitätssicherungsgründen zu stabilisieren.
Wenn vom Wirkstoff (I) in amorpher Form die Rede ist, so ist damit gemeint, dass der Wirkstoff (I) zu > 95% in amorpher Form vorliegt.
Eine pharmazeutische Darreichungsform der vorliegenden Erfindung besteht aus einer festen Dispersion, die den Wirkstoff (I) sowie eine pharmazeutisch akzeptable Matrix und gegebenenfalls ein Trägermaterial enthält.
Der Begriff "Matrix" oder "Matrixbildner", wie in der vorliegenden Erfindung benutzt, bezieht sich sowohl auf polymere als auch nicht-polymere Hilfsstoffe und Kombinationen aus diesen, geeignet den Wirkstoff (I) zu lösen oder zu dispergieren.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung besteht die Matrix aus einem pharmazeutisch akzeptablen Polymer, wie beispielsweise Polyvinylpyrrolidon,
Vinylpyrrolidon/Vinylacetat copolymer, Crospovidon, Polyalkylenglycole (z.B.
Polyethylenglycol), Polyethylenoxid, Poloxamer, Hydroxyalkylcellulosen (z.B.
Hydroxypropylcellulose), Hydroxyalkylmethylcellulosen (z.B. Hydroxypropylmethyl cellulose),
Carboxymethylcellulose, Natriumcarboxymethylcellulose, Ethylcellulose, Cellulosesuccinate (z.B. Celluloseacetatesuccinate und Hydroxypropylmethylcelluloseacetatsuccinat), Cellulosephthalate
(z.B. Celluloseacetatphthalat und Hydroxypropylmethylcellulosephthalat), Polymethacrylate (z.B.
Eudragit® Typen), Polyhydroxyalkylacrylate, Polyhydroxyalkylmethacrylate, Polyacrylate,
Polyvinylalkohol, Polyvinylacetat, Vinylalkohol/Vinylacetat copolymer, Xanthangummi,
Galactomannane, Carrageenan, Chitosan, Chitin, Alginsäure und deren Salze, Polylactide, Dextrine, Stärke und Stärkederivate, Proteine und Kombinationen aus den genannten pharmazeutisch akzeptablen Polymeren.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung stammt mindestens eine der als Matrixbildner in der festen Dispersion eingesetzten Substanzen aus der Gruppe: Polyvinylpyrrolidon, Copovidon, Hydroxypropylcellulose, Hydroxypropylmethylcellulose, Polyethylenglycol und Polyethylenoxid.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung dient Polyvinylpyrrolidon als Matrixbildner.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung sind der Wirkstoff (I) und Polyvinylpyrrolidon als Matrixbildner im Masseverhältnis 1 :0,5 bis 1 :20, besonders bevorzugt 1 :2 bis 1 :5 enthalten.
Weiterer Gegenstand der vorliegenden Erfindung ist eine pharmazeutische Darreichungsform bestehend aus einer festen Dispersion, bei der die Matrix einen Zucker und/oder Zuckeralkohol und/oder Cyclodextrin, wie z.B. Sucrose, Lactose, Fructose, Maltose, Raffinose, Sorbitol, Lactitol, Mannitol, Maltitol, Erythritol, Threitol, Adonitol, Arabitol, Xylitol, Dulcitol, Inositol, Trehalose, Isomalt, Inulin, Maltodextrin, ß -Cyclodextrin, Hydroxypropyl-ß -cyclodextrin, Sulfobutylethercyclodextrin, oder Kombinationen der selbigen enthält.
Für die Matrixbildung der festen Dispersion können weitere geeignete Hilfsstoffe ergänzt werden, dies sind beispielsweise Alkohole, organische Säuren, organische Basen, Salze, Aminosäuren, Peptide, Phospholipide, Lipide (z.B. Mono-, Di- und Triglyceride), Fettsäuren, Fettalkohole, Wachse, Fettsäureester, Polyoxyethylensorbitanfettsäureester, Polyglycolisierte Glyceride, Sucroseester, Harnstoff und Kombinationen der selbigen.
Die feste Dispersion, bestehend aus mindestens dem Wirkstoff (I) und einer Matrix, kann einige zusätzliche, pharmazeutisch akzeptable Inhaltsstoffe enthalten, wie z.B. Trägermaterialien, oberflächenaktive Substanzen, Füllstoffe, Zerfallshilfsmittel, Adsorbantien, Rekristallisationsinhibitoren, Weichmacher, Entschäumer, Antioxidatien, Antiklebemittel, Schmelzmittel, pH-Wert modifizierende Substanzen, Schmier- und/oder Gleitmittel.
Ein Trägermaterial im Sinn der vorliegenden Erfindung ist ein Hilfsstoff, welcher mit einer Mischung, bestehend aus mindestens der Matrix und dem Wirkstoff (I), während der Herstellung der festen Dispersion, z.B. durch Schmelzextrusion, Schmelzlackierung, Sprühkondensation, Erstarren, Lösemittelverdampfungsprozesse/Lösemethode (z.B. Beschichtung, Lackierung, Granulation) beladen wird und somit ein wesentlicher Bestandteil der festen Dispersion wird.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung enthält die feste Dispersion als Trägermaterial beispielsweise Croscarmellose Natrium, Natriumstärkeglycolat, Crospovidon, niedrig-substituierte Hydroxypropylcellulose (L-HPC), Stärke, mikrokristalline Cellulose oder eine Kombination der selbigen. Diese Inhaltsstoffe können auch als Zerfallshilfsmittel eingesetzt werden.
Das Trägermaterial bzw. die Kombination von Trägermaterialien kann dabei in einer Konzentration von 5 bis 90% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung, vorzugsweise von 10 bis 60% bezogen auf die Gesamtmasse der pharmazeutischen Zusammensetzung enthalten sein.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung enthält die feste Dispersion Polyvinylpyrrolidon als Matrix und Croscarmellose Natrium als Trägermaterial.
Γη einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung enthält die feste Dispersion Polyvinylpyrrolidon als Matrix, Croscarmellose Natrium und mikrokristalline Cellulose als Trägermaterial.
Die feste Dispersion nach vorliegender Erfindung wird mittels dem Fachmann bekannter Herstellprozesse von festen Dispersionen wie Schmelz/Sinter Technologie, Schmelzextrusion, Schmelzcoating, Sprühkondensation, Erstarren, Lösemittelverdampfungsprozesse/Lösemethode (zB. Gefriertrocknung, Sprühtrocknung, Vakuumtrocknung, Beschichten von Pulvern, Granulen oder Pellets und Wirbelschichtgranulation), Copräzipitation, Prozesse mit überkritischen Flüssigkeiten und elektrostatischer Zentrifugation gefertigt. Die Lösemethode ist der bevorzugte Herstellprozess zur Fertigung der festen Dispersion im Sinne der vorliegenden Erfindungen.
Der Wirkstoff (I) wird über die Lösemethode in dessen amorphen Zustand stabilisiert.
Ein für die Herstellung der festen Dispersion durch Lösemethoden, wie z.B. Sprühtrocknung, Beschichtung und Wirbelschichtgranulation geeignetes Lösemittel kann jeglicher Stoff sein, in dem sich der Wirkstoff (I) löst. Bevorzugte Lösungsmittel sind Alkohole wie z.B. Methanol, Ethanol, n-Propanol, Isopropanol und Butanol, Ketone wie z.B. Aceton, Methylethylketon und Methylisobutylketon, Ester wie z.B. Ethylacetat und Propylacetat, sowie verschiedene weitere Lösungsmittel wie Acetonitril, Methylenchlorid, Chloroform, Hexan, Toluol, Tetrahydrofuran, zyklische Ether und 1,1,1-Trichlorethan. Schwerflüchtige Lösungsmittel wie Dimethylacetamid oder Dimethylsulfoxid können ebenfalls eingesetzt werden. Mischungen dieser Lösungsmittel, z.B. 20% Ethanol und 80% Aceton, können genauso eingesetzt werden wie Mischungen mit Wasser, sofern der Wirkstoff (I) eine ausreichende Löslichkeit in der Mischung zeigt und gegebenenfalls der Matrixbildner eine ausreichende Löslichkeit in der Mischung zeigt, um den Prozess ausführen zu können. In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist das Lösungsmittel zur Herstellung einer festen Dispersion eine Mischung aus zwei oder mehreren Lösungsmitteln ausgewählt aus der Gruppe bestehend aus Methanol, Ethanol, n-Propanol, Isopropanol und Aceton.
In einer besonders bevorzugten Ausführungsform der vorliegenden Erfindung wird eine Mischung aus Ethanol und Aceton als Lösungsmittel für die Lösemethode zur Herstellung einer festen Dispersion eingesetzt.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist die feste Dispersion homogen, das heißt die Wirkstoffverteilung in der Matrix und im gegebenenfalls vorhandenen
Trägermaterial ist homogen. Unter homogen wird im Rahmen der vorliegenden Erfindung die gleichmäßige Verteilung des Wirkstoffes (I), der Matrix und des gegebenenfalls vorhandenen Trägermaterials in der festen Dispersion verstanden.
Für den Wirkstoff (I) in amorpher Form ist die Herstellung der festen Dispersion durch die Lösemethode, bei der ein Wirkstoff und gegebenenfalls eingesetzter Matrixbildner wie beispielsweise Polyvinylpyrrolidon gelöst und dann weiterverarbeitet werden, besonders gut geeignet, da der Wirkstoff (I) eine ausreichende Löslichkeit in pharmazeutisch geeigneten organischen Lösemitteln wie beispielsweise Aceton oder Ethanol aufweist.
Der Wirkstoff (I) liegt in der nach der Lösemethode hergestellten festen Dispersion vorzugsweise in einer Konzentration von 2 bis 30% bezogen auf die Gesamtmasse der gelösten Komponenten vor, besonders bevorzugt in einer Konzentration von 2 bis 20% bezogen auf die Gesamtmasse der gelösten Komponenten, insbesondere in einer Konzentration von 5 bis 15% bezogen auf die Gesamtmasse der gelösten Komponenten.
Die durch die Lösemethode gewonnene den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion kann unterschiedlich weiterverarbeitet werden:
Es kann beispielsweise zerkleinert werden und als Pulver oder Granulat verabreicht werden, gegebenenfalls nach Abfüllung als Sachet oder in Kapseln. Dabei können übliche pharmazeutische Hilfsstoffe zugesetzt werden wie beispielsweise Füllstoffe, Fließregulierungsmittel, Adsorbentien, Netzmittel, Aromen und Farbstoffe. Weiterhin kann die den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion zu Tablettenformulierungen weiterverarbeitet werden. Dazu kann es geschnitten, gemahlen und mit üblichen Tablettierhilfsstoffen wie Füllstoffen und Trockenbindemitteln (beispielsweise Cellulosepulver, mikrokristalline Cellulose, verkieselte mikrokristalline Cellulose, Dicalciumphosphat, Tricalciumphosphat, Magnesiumtrisilikat, Mannitol, Maltitol, Sorbitol, Xylitol, Laktose, Dextrose, Maltose, Saccharose, Glucose, Fructose oder Maltodextrine), Zerfallsförderern/Sprengmittel (beispielsweise Carboxymethylcellulose, Croscarmellose (quervernetzte Carboxymethylcellulose), Crospovidone (quervernetztes Polyvinylpyrrolidon), L- HPC (niedrigsubstituierte Hydroxypropylcellulose), Natriumcarboxy-methylstärke, Natriumglykolat der Kartoffelstärke, partiell hydrolysierte Stärke, Weizenstärke, Maisstärke, Reisstärke oder Kartoffelstärke), Schmier-, Gleit- und Fließregulierungsmitteln wie Fumarsäure, Staearinsäure, Magnesiumstearat, Calciumstearat, Natriumstearylfumarat, höhermolekulare Fettalkohole, Polyethylenglykole, Stärke (Weizen-, Reis,- Mais- oder Kartoffelstärke), Talkum, hochdisperses (kolloidales) Siliciumdioxid, Magnesiumoxid, Magnesiumcarbonat oder
Calciumsilikat, Adsorbentien, Aromen und Farbstoffen gemischt, zu Tabletten gepresst und diese gegebenenfalls anschließend noch lackiert werden.
Bevorzugt sind Tabletten, insbesondere den Wirkstoff (I) schnell freisetzende Tabletten.
Geeignete Materialien für einen Lichtschutz- und/oder Farblack sind beispielsweise Polymere wie Polyvinylalkohol, Hydroxypropylcellulose und/oder Hydroxypropylmethylcellulose, gegebenenfalls in Kombination mit geeigneten Weichmachern wie beispielsweise Polyethylenglykol oder Polypropylenglykol und Pigmenten wie beispielsweise Titandioxid oder Eisenoxiden, wie Eisenoxid gelb oder Eisenoxid rot. Der Lack kann weiterhin Talkum und/oder Lecithin enthalten.
Der Lack kann beispielsweise die folgende Zusammensetzung haben: Polyvinylalkohol, Talkum, Polyethylenglykol, Lecithin, Eisenoxid gelb, Eisenoxid rot und Titandioxid.
Weiterhin kann der Lack beispielsweise die folgende Zusammensetzung haben:
1,25 mg Hydroxypropylmethylcellulose, 0,25 mg Talkum, 0,05 mg Eisenoxid gelb und 0,95 mg Titandioxid oder 1,50 mg Hydroxypropylmethylcellulose, 0,50 mg Polyethylenglykol und 0,50 mg Titandioxid.
Bei den Tabletten handelt es sich vorzugsweise um schnell zerfallende Tabletten mit einer Zerfallszeit von maximal 30 Minuten.
Weiterer Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung der erfindungsgemäßen Tablettenformulierung, wobei mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird, welche dann gegebenenfalls gemahlen, mit weiteren dem Fachmann bekannten pharmazeutischen Hilfsstoffen gemischt und in Kapseln oder als Sachet abgefüllt werden oder mit den üblichen Tablettierhilfsstoffen gemischt und anschließend vorzugsweise mittels Direkttablettierung zu Tabletten gepresst wird, die abschließend mit einem Lack überzogen werden können. Die feste Dispersion kann vor der Weiterverarbeitung gegebenenfalls gemahlen, anderweitig zerkleinert, gesiebt oder gesichtet werden.
Weiterer Gegenstand der vorliegenden Erfindung ist die Verwendung von (2-{ [(lS)-l-(3- Chlorphenyl)-2-fluorethyl] amino } -7-methoxy- 1 ,3-benzoxazol-5-yl) [(2S,5S)-5-(2-hydroxyefhyl)-2-
methyl-morpholin-4-yl]methanon (I) zur Herstellung einer erfindungsgemäßen festen, oral applizierbaren pharmazeutischen Darreichungsform mit schneller Wirkstofffreisetzung.
Weiterer Gegenstand der vorliegenden Erfindung sind Arzneimittel, enthaltend eine erfindungsgemäße feste, oral applizierbare, Wirkstoff (I) in amorpher Form enthaltende pharmazeutische Darreichungsform mit schneller Wirkstofffreisetzung.
Weiterer Gegenstand der vorliegenden Erfindung ist die Verwendung einer erfindungsgemäßen festen, oral applizierbaren, Wirkstoff (I) in amorpher Form enthaltende pharmazeutische Darreichungsform mit schneller Wirkstofffreisetzung zur Herstellung eines Arzneimittels zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen, insbesondere von arteriellen und/oder venösen thromboembolischen Erkrankungen.
Weiterer Gegenstand der vorliegenden Erfindung ist die Verwendung der erfindungsgemäßen festen, oral applizierbaren pharmazeutischen Darreichungsform mit schneller Wirkstofffreisetzung, enthaltend amorphen Wirkstoff (I) zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen, insbesondere von arteriellen und/oder venösen thromboembolischen Erkrankungen wie Myocardinfarkt, Angina Pectoris (eingeschlossen instabile Angina), Reokklusionen und Restenosen nach einer Angioplastie oder aortokoronarem Bypass, Schlaganfall, transitorische ischämische Attacken, periphere arterielle Verschlusskrankheiten, Lungenembolien oder tiefen Venenthrombosen. Ganz besonders eigenen sich die erfindungsgemäßen Verbindungen für die Behandlung und/oder Prophylaxe von akutem Koronarsyndrom (ACS), venösen Thromboembolien, venösen Thrombosen, insbesondere in tiefen Beinvenen und Nierenvenen, Lungenembolien, Schlaganfall und/oder Thromboseprophylaxe im Rahmen von chirurgischen Eingriffen, insbesondere im Rahmen von chirurgischen Eingriffen bei Patienten, die eine Krebserkrankung haben.
Weiterer Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen, insbesondere von arteriellen und/oder venösen thromboembolischen Erkrankungen wie Myocardinfarkt, Angina Pectoris (eingeschlossen instabile Angina), Reokklusionen und Restenosen nach einer Angioplastie oder aortokoronarem Bypass, Schlaganfall, transitorische ischämische Attacken, periphere arterielle Verschlusskrankheiten, Lungenembolien oder tiefen Venenthrombosen. Weiterer Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Behandlung und/oder Prophylaxe von akutem Koronarsyndrom (ACS), venösen Thromboembolien, venösen Thrombosen, insbesondere in tiefen Beinvenen und Nierenvenen, Lungenembolien, Schlaganfall und/oder Thromboseprophylaxe im Rahmen von chirurgischen Eingriffen, insbesondere im
Rahmen von chirurgischen Eingriffen bei Patienten, die eine Krebserkrankung haben durch Verabreichung einer erfindungsgemäßen festen, oral applizierbaren, Wirkstoff (I) in amorpher Form enthaltende pharmazeutischen Darreichungsform mit schneller Wirkstofffreisetzung.
Weiterer Gegenstand der vorliegenden Erfindung sind die erfindungsgemäßen festen, oral applizierbaren pharmazeutischen Darreichungsformen mit schneller Wirkstofffreisetzung, enthaltend amorphen Wirkstoff (I) zur Verwendung in einem Verfahren zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen, insbesondere von arteriellen und/oder venösen thromboembolischen Erkrankungen wie Myocardinfarkt, Angina Pectoris (eingeschlossen instabile Angina), Reokklusionen und Restenosen nach einer Angioplastie oder aortokoronarem Bypass, Schlaganfall, transitorische ischämische Attacken, periphere arterielle Verschlusskrankheiten, Lungenembolien oder tiefen Venenthrombosen.
Weiterer Gegenstand der vorliegenden Erfindung sind die erfindungsgemäßen festen, oral applizierbaren pharmazeutischen Darreichungsformen mit schneller Wirkstofffreisetzung, enthaltend amorphen Wirkstoff (I) zur Verwendung in einem Verfahren zur Behandlung und/oder Prophylaxe von akutem Koronarsyndrom (ACS), venösen Thromboembolien, venösen Thrombosen, insbesondere in tiefen Beinvenen und Nierenvenen, Lungenembolien, Schlaganfall und/oder Thromboseprophylaxe im Rahmen von chirurgischen Eingriffen, insbesondere im Rahmen von chirurgischen Eingriffen bei Patienten, die eine Krebserkrankung haben.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung einer festen, oral applizierbaren pharmazeutischen Darreichungsform enthaltend Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff (I) von > 2% bezogen auf die Gesamtmasse der Formulierung, dadurch gekennzeichnet, dass mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung einer festen, oral applizierbaren pharmazeutischen Darreichungsform enthaltend Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff (I) von > 2% bezogen auf die Gesamtmasse der Formulierung, dadurch gekennzeichnet, dass
(a) zunächst mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird, welche dann gegebenenfalls gemahlen (b) und dann, gegebenenfalls unter Zusatz pharmazeutisch geeigneter üblicher Tablettierhilfsstoffe, in die pharmazeutische Darreichungsform überführt wird.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung einer festen, oral applizierbaren pharmazeutischen Darreichungsform enthaltend Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff (I) von > 2% bezogen auf die Gesamtmasse der Formulierung, dadurch gekennzeichnet, dass (a) zunächst mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird, welche dann gegebenenfalls gemahlen
(b) und dann, gegebenenfalls unter Zusatz pharmazeutisch geeigneter üblicher Tablettierhilfsstoffe, in die pharmazeutische Darreichungsform überführt wird, und wobei aus der pharmazeutischen Darreichungsform betreffend 6 Prüfkörper nach 2 Stunden mindestens 80% Wirkstoff (I) pro Prüfkörper gemäß der Freisetzungsmethode des Europäischen Arzneibuchs mit Apparatur 2 (Paddle) in das Freisetzungsmedium abgegeben werden.

(b) und dann, gegebenenfalls unter Zusatz pharmazeutisch geeigneter üblicher Tablettierhilfsstoffe, in die pharmazeutische Darreichungsform überführt wird. Gegenstand der vorliegenden Erfindung ist eine feste, oral applizierbare pharmazeutische Darreichungsformen mit schneller Wirkstofffreisetzung, enthaltend Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff (I) von > 2% bezogen auf die Gesamtmasse der Formulierung, dadurch gekennzeichnet, dass sie hergestellt wird indem
(a) zunächst mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird, welche dann gegebenenfalls gemahlen
(b) und dann, gegebenenfalls unter Zusatz pharmazeutisch geeigneter üblicher Tablettierhilfsstoffe, in die pharmazeutische Darreichungsform überführt wird, und wobei aus der pharmazeutischen Darreichungsform betreffend 6 Prüfkörper nach 2 Stunden mindestens 80% Wirkstoff (I) pro Prüfkörper gemäß der Freisetzungsmethode des Europäischen Arzneibuchs mit Apparatur 2 (Paddle) in das Freisetzungsmedium abgegeben werden.
Die Erfindung wird nachstehend durch bevorzugte Ausführungsbeispiele näher erläutert, auf welche sie jedoch nicht eingeschränkt ist. Soweit nicht anders angegeben, beziehen sich nachstehend alle Mengenangaben auf Gewichtsprozente.
Experimenteller Teil
1. Freisetzungsmethode
Gemäß Europäischem Arzneibuch, 6. Ausgabe, Grundwerk 2008 wird die Arzneiform mit Apparatur 2 (Paddle) geprüft. Die Umdrehungsgeschwindigkeit des Rührers liegt bei 50-100 UpM (Umdrehungen pro Minute) in 900 ml Phosphatpuffer pH 6,8. Das Freisetzungskriterium ist dann erfüllt, wenn alle 6 Prüfkörper nach einem Untersuchungszeitraum von 2 Stunden mindestens 80% Wirkstoff (I) in das Freisetzungsmedium abgegeben haben. Diese Methode findet für schnell freisetzende Tabletten Verwendung, bei denen die Tablettendosis < 100 mg beträgt, um sink- Bedingungen im Freisetzungsmedium zu gewährleisten. Unter Sink-Bedingungen wird das 3-fache Freisetzungsmedien- Volumen verstanden, welches benötigt würde, um mit der in der Tablette enthaltenen Wirkstoffmenge eine gesättigte Lösung herzustellen.
Beispielformulierung 1
Ein Aceton-Ethanol Gemisch (80:20) enthaltend Wirkstoff (I) in amorpher Form und Polyvinylpyrrolidon im Verhältnis 1:3 resultierend in Granulat (feste Dispersion) folgender Zusammensetzung:
Wirkstoff (I) 25 mg
Polyvinylpyrrolidon 75 mg
Mikrokristalline Cellulose 50 mg
Croscarmellose Natrium 50 mg
Das erhaltene Granulat wird anschließend zu einer Tablette folgender Zusammensetzung weiterverarbeitet :
Granulat 200 mg
Croscarmellose Natrium 23,2 mg
Magnesiumstearat 1,8 mg
Die erhaltene Tablette kann anschließend mit einem Lack überzogen werden:
Lack 10 mg Herstellung: Wirkstoff (I) und Polyvinylpyrrolidon werden im organischen Lösungsmittelgemisch bestehend aus Aceton und Ethanol (80:20) gelöst. Die so hergestellte Lösung wird als
Granulierflüssigkeit im Rahmen einer Wirbelschichtgranulation auf die Vorlage aus mikrokristalliner Cellulose und Croscarmellose Natrium aufgesprüht. Nach Trocknung und Siebung (0.8 mm Maschenweite) des entstandenen Granulates werden nacheinander Croscarmellose Natrium und Magnesiumstearat zugegeben und jeweils gemischt. Die so erhaltene pressfertige Mischung wird zu Tabletten mit 12 mm Durchmesser (12x6wr5+2) und einer Bruchfestigkeit von 25 bis 80 N verpresst. Die anschließende Lackierung der Tabletten erfolgt mit Titandioxid und Eisenoxid, die in einer wässrigen Lösung aus Hydroxypropylmethylcellulose und Talkum suspendiert sind.
Beispielformulierung 2 Ein Aceton-Ethanol Gemisch (80:20) enthaltend Wirkstoff (I) in amorpher Form und Polyvinylpyrrolidon im Verhältnis 1:3 resultierend in Granulat (feste Dispersion) folgender Zusammensetzung:
Wirkstoff (I) 25 mg
Polyvinylpyrrolidon 75 mg
Mikrokristalline Cellulose 50 mg

Das erhaltene Granulat wird anschließend zu einer Tablette folgender Zusammensetzung weiterverarbeitet :

Hochdisperses (kolloidales) Siliciumdioxid 1,3 mg
Magnesiumstearat 2,1 mg Die erhaltene Tablette kann anschließend mit einem Lack überzogen werden:
Lack 10 mg
Herstellung: Wirkstoff (I) und Polyvinylpyrrolidon werden im organischen Lösungsmittelgemisch bestehend aus Aceton und Ethanol (80:20) gelöst. Croscarmellose Natrium 35,1 mg
Hochdisperses (kolloidales) Siliciumdioxid 1,9 mg
Magnesiumstearat 3,0 mg
Die erhaltene Tablette kann anschließend mit einem Lack überzogen werden:
Lack 10 mg
Herstellung: Wirkstoff (I) und Polyvinylpyrrolidon werden im organischen Lösungsmittelgemisch bestehend aus Aceton und Ethanol (80:20) gelöst. Die so hergestellte Lösung wird als Granulierflüssigkeit im Rahmen einer Wirbelschichtgranulation auf die Vorlage aus mikrokristalliner Cellulose und Croscarmellose Natrium aufgesprüht. Nach Trocknung und Siebung (0.8 mm Maschenweite) des entstandenen Granulates werden nacheinander Croscarmellose Natrium, hochdisperses (kolloidales) Siliciumdioxid und Magnesiumstearat zugegeben und jeweils gemischt. Die so erhaltene pressfertige Mischung wird zu Tabletten mit 14 mm Durchmesser (14x7wr6+2) und einer Bruchfestigkeit von 25 bis 80 N verpresst. Die anschließende Lackierung der Tabletten erfolgt zum Beispiel mit Titandioxid und Eisenoxid, die in einer wässrigen Lösung
aus Hydroxypropylmethylcellulose und Talkum suspendiert sind.
Beispielformulierung 4
Ein Aceton-Ethanol Gemisch (80:20) enthaltend Wirkstoff (I) in amorpher Form und Polyvinylpyrrolidon im Verhältnis 1:3 resultierend in Granulat (feste Dispersion) folgender Zusammensetzung:
Wirkstoff (I) 5 mg
Polyvinylpyrrolidon 15 mg
Mikrokristalline Cellulose 30 mg
Croscarmellose Natrium 15 mg
Das erhaltene Granulat wird anschließend zu einer Tablette folgender Zusammensetzung weiterverarbeitet :

Mikrokristalline Cellulose 10,225 mg
Croscarmellose Natrium 8,5 mg
Hochdisperses (kolloidales) Sihciumdioxid 0,425 mg
Magnesiumstearat 0,85 mg Die erhaltene Tablette kann anschließend mit einem Lack überzogen werden:

Herstellung: Wirkstoff (I) und Polyvinylpyrrolidon werden im organischen Lösungsmittelgemisch bestehend aus Aceton und Ethanol (80:20) gelöst. Die so hergestellte Lösung wird als Granulierflüssigkeit im Rahmen einer Wirbelschichtgranulation auf die Vorlage aus mikrokri- stalliner Cellulose und Croscarmellose Natrium aufgesprüht. Nach Trocknung und Siebung (0.8 mm Maschenweite) des entstandenen Granulates werden nacheinander mikrokristalline Cellulose, Croscarmellose Natrium, hochdisperses (kolloidales) Sihciumdioxid und Magnesiumstearat zugegeben und jeweils gemischt. Die so erhaltene pressfertige Mischung wird zu Tabletten mit 6 mm Durchmesser und einer Bruchfestigkeit von 50 bis 120 N verpresst. Die anschließende Lackierung der Tabletten erfolgt mit Titandioxid und Eisenoxid gelb, wobei die Pigmente zuvor mit Talkum in einer wässrigen Lösung aus Hydroxypropylmethylcellulose suspendiert werden.
Beispielformulierung 5
Ein Aceton-Ethanol Gemisch (80:20) enthaltend Wirkstoff (I) in amorpher Form und Polyvinylpyrrolidon im Verhältnis 1:3 resultierend in Granulat (feste Dispersion) folgender Zusammensetzung: Wirkstoff (I) 5 mg
Polyvinylpyrrolidon 15 mg
Mikrokristalline Cellulose 25 mg
Croscarmellose Natrium 30 mg Das erhaltene Granulat wird anschließend zu einer Tablette folgender Zusammensetzung weiterverarbeitet :
Granulat 75 mg
Croscarmellose Natrium 8,875 mg
Hochdisperses (kolloidales) Sihciumdioxid 0,425 mg
Magnesiumstearat 0,70 mg
Die erhaltene Tablette kann anschließend mit einem Lack überzogen werden:
Lack 2,5 mg
Herstellung: Wirkstoff (I) und Polyvinylpyrrolidon werden im organischen Lösungsmittelgemisch bestehend aus Aceton und Ethanol (80:20) gelöst. Die so hergestellte Lösung wird als Granulierflüssigkeit im Rahmen einer Wirbelschichtgranulation auf die Vorlage aus mikrokristalliner Cellulose und Croscarmellose Natrium aufgesprüht. Die anschließende Lackierung der Tabletten erfolgt mit Titandioxid und Eisenoxid gelb, wobei die Pigmente zuvor mit Talkum in einer wässrigen Lösung aus Hydroxypropylmethylcellulose suspendiert werden.
Claims
1. Feste, oral applizierbare pharmazeutische Darreichungsform mit schneller Wirkstofffreisetzung, enthaltend (2- { [( 1 S)- 1 -(3-Chlorphenyl)-2-fluorethyl] amino } -7- methoxy-l,3-benzoxazol-5-yl)[(2lS',5lS')-5-(2-hydroxyethyl)-2-methyl-morpholin-4- yl]methanon (I), dadurch gekennzeichnet, dass
(a) sie Wirkstoff (I) in amorpher Form enthält und
(b) dass 80% des Wirkstoffes (I) über einen Zeitraum von maximal 2 Stunden gemäß USP-Freisetzungsmethode mit Apparatur 2 (Paddle; 50-100 UpM) freigesetzt werden.
2. Pharmazeutische Darreichungsform gemäß Anspruch 1, dadurch gekennzeichnet, dass der Wirkstoff (I) über eine Lösemethode in dessen amorphen Zustand stabilisiert wird.
3. Pharmazeutische Darreichungsform gemäß Anspruch 2, dadurch gekennzeichnet, dass Matrixbildner zugesetzt werden und der Wirkstoff (I) in der nach der Lösemethode vorliegenden festen Dispersion in einer Konzentration von 2 bis 30 % bezogen auf die Gesamtmasse der gelösten Komponenten vorliegt.
4. Pharmazeutische Darreichungsform gemäß Anspruch 3, dadurch gekennzeichnet, dass die Matrixbildner sowohl polymere als auch nicht-polymere Hilfsstoffe und Kombinationen aus diesen sind, geeignet den Wirkstoff (I) zu lösen oder zu dispergieren.
5. Pharmazeutische Darreichungsform gemäß Anspruch 3 oder 4, dadurch gekennzeichnet, dass Polyvinylpyrrolidon als Matrixbildner eingesetzt wird.
6. Pharmazeutische Darreichungsform gemäß einem der Ansprüche 3 bis 5, dadurch gekennzeichnet, dass die feste Dispersion als Trägermaterial Croscarmellose Natrium, Natriumstärkeglykolat, Crospovidon, niedrig-substituierte Hydroxypropylcellulose, Stärke, mikrokristalline Cellulose oder eine Kombination derselben enthält.
7. Pharmazeutische Darreichungsform gemäß einem der Ansprüche 3 bis 6, dadurch gekennzeichnet, dass die feste Dispersion homogen ist.
8. Pharmazeutische Darreichungsform gemäß einem der Ansprüche 1 bis 7 in Form eines Pulvers, Granulates, Sachets, einer Kapsel oder einer Tablette.
9. Verfahren zur Herstellung einer festen, oral applizierbaren pharmazeutischen Darreichungsform enthaltend Wirkstoff (I) in amorpher Form in einer Menge an Wirkstoff
(I) von > 2% bezogen auf die Gesamtmasse der Formulierung, dadurch gekennzeichnet, dass mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird.
Verfahren gemäß Anspruch 9, dadurch gekennzeichnet, dass
(a) zunächst mit Hilfe der Lösemethode eine den Wirkstoff (I) in amorpher Form enthaltende feste Dispersion hergestellt wird, welche dann gegebenenfalls gemahlen
(b) und dann, gegebenenfalls unter Zusatz pharmazeutisch geeigneter üblicher Tablettierhilfsstoffe, in die pharmazeutische Darreichungsform überführt wird.
Verfahren gemäß Anspruch 9 oder 10, dadurch gekennzeichnet, dass das Lösungsmittel zur Herstellung der festen Dispersion eine Mischung aus zwei oder mehreren Lösemitteln ausgewählt aus der Gruppe bestehend aus Methanol, Ethanol, n-Propanol, Isopropanol und Aceton ist.
Pharmazeutische Darreichungsform hergestellt gemäß einem der Ansprüche 9 bis 11.
Arzneimittel, enthaltend eine pharmazeutische Darreichungsform gemäß einem der Ansprüche 1 bis 8 oder 12.
Verwendung einer pharmazeutischen Darreichungsform gemäß einem der Ansprüche 1 bis 8 oder 12, zur Herstellung eines Arzneimittels zur Prophylaxe, Sekundärprophylaxe und/oder Behandlung von Erkrankungen.
Pharmazeutische Darreichungsform gemäß einem der Ansprüche 1 bis 8 oder 12 zur Verwendung in einem Verfahren zur Behandlung und/oder Prophylaxe von akutem Koronarsyndrom (ACS), venösen Thromboembolien, venösen Thrombosen, insbesondere in tiefen Beinvenen und Nierenvenen, Lungenembolien, Schlaganfall und/oder Thromboseprophylaxe im Rahmen von chirurgischen Eingriffen, insbesondere im Rahmen von chirurgischen Eingriffen bei Patienten, die eine Krebserkrankung haben.
