(1)大量の抗インフルエンザウイルス薬(ノイラミニダーゼ阻害剤)の備蓄
(2)大量のプレパンデミックワクチンの備蓄
(3)パンデミックワクチンの生産体制の構築
本発明は、以上の現状を打開すべく、インフルエンザウイルスのパンデミック対策に有効な手段を提供することを目的とする。
主として以上の成果に基づき、以下の発明が提供される。
[1]被検者由来の生体サンプル中における、VH1-69遺伝子を用いた抗体の存否を指標として、A型インフルエンザウイルスに対する抵抗力を判定する方法。
[2]以下のステップ(1)~(3)を含む、[1]に記載の方法:
(1)インフルエンザワクチン接種後の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させるステップ;
(2)生成する免疫複合体を検出するステップ;
(3)ステップ(2)の検出結果に基づきA型インフルエンザウイルスに対する抵抗力の強さを判定するステップであって、検出された免疫複合体の量がA型インフルエンザウイルスに対する抵抗力の強さの指標となるステップ。
[3]以下のステップを更に含む、[2]に記載の方法:
(4)インフルエンザワクチン接種前の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させることにより生成する免疫複合体を検出し、該免疫複合体の量と、ステップ(2)で検出された免疫複合体の量を比較し、比較結果に基づき、抵抗力の強さを判定するステップ。
[4]生体サンプルが血液サンプルである、[2]又は[3]に記載の方法。
[5]A型インフルエンザウイルスが、H1N1型、H1N2型、H2N2型、H3N2型、H5N1型、H5N2型、H6N1型、H7N2型、H7N3型、H7N7型、H7N9型、H9N2型及びH9N1型からなる群より選択される一又は二以上のウイルスである、[1]~[4]のいずれか一項に記載の方法。
[6]A型インフルエンザウイルスが、H1N1型、H2N2型、H3N2型及びH5N1型からなる群より選択される一又は二以上のウイルスである、[1]~[4]のいずれか一項に記載の方法。

[8]VH1-69遺伝子を用いた抗体を特異的に認識する抗イディオタイプ抗体を含む、A型インフルエンザウイルスに対する抵抗力判定用キット。
本発明の第1の局面は、A型インフルエンザウイルスに対する抵抗力を判定する方法に関する。本発明によれば、A型インフルエンザウイルスに対する被検者の抵抗力を客観的な指標で判定することができる。本発明の方法は、将来起こり得るインフルエンザのパンデミック対策の手段として有効であり、その価値は極めて高い。本発明の方法では、VH1-69遺伝子を用いる抗体がA型インフルエンザウイルスに対して幅広く中和活性を示すという知見、及びVH1-69遺伝子を用いる抗体を産生するB細胞がメモリー細胞として残存し、A型インフルエンザに対する生体の防御に重要な役割を果たすという知見に基づき(詳細は後述の実施例を参照)、被検者由来の生体サンプル中における、VH1-69遺伝子を用いた抗体の存否を指標として、A型インフルエンザウイルスに対する被検者の抵抗力を判定する。
(1)インフルエンザワクチン接種後の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させるステップ
(2)生成する免疫複合体を検出するステップ
(3)ステップ(2)の検出結果に基づきA型インフルエンザウイルスに対する抵抗力の強さを判定するステップであって、検出された免疫複合体の量がA型インフルエンザウイルスに対する抵抗力の強さの指標となるステップ
<判定基準の例>
(a) 免疫複合体が検出されればA型インフルエンザウイルスに対する抵抗力がある
(b) 免疫複合体が検出されればA型インフルエンザウイルスに対する抵抗力が強い
(c) 免疫複合体の量が多いほどA型インフルエンザウイルスに対する抵抗力が強い
基準値よりも検出値が高いときに「A型インフルエンザウイルスに対する抵抗力がある」と判定し、基準値よりも検出値が低いときに「A型インフルエンザウイルスに対する抵抗力がない」と判定する。
(定性的判定の例2)
基準値よりも検出値が高いときに「A型インフルエンザウイルスに対する抵抗力が強い」と判定し、基準値よりも検出値が低いときに「A型インフルエンザウイルスに対する抵抗力が弱い」と判定する。
(定性的判定の例3)
検出できたとき(陽性の場合)に「A型インフルエンザウイルスに対する抵抗力がある」と判定し、検出できないとき(陰性の場合)に「A型インフルエンザウイルスに対する抵抗力がない」と判定する。
(定性的判定の例4)
検出できたとき(陽性の場合)に「A型インフルエンザウイルスに対する抵抗力が強い」と判定し、検出できないとき(陰性の場合)に「A型インフルエンザウイルスに対する抵抗力が弱い」と判定する。
以下に示すように検出値の範囲毎に抵抗力の強さを予め設定しておき、検出値から抵抗力を判定する。
測定値<a:抵抗力がない
a≦測定値<b:抵抗力が弱い
b≦測定値<c:抵抗力があるが十分ではない
c<測定値:抵抗力が強い
この例では4段階に区分したが、区分数は任意に設定できる。区分数の例は2~7、好ましくは3~6である。
(4)インフルエンザワクチン接種前の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させることにより生成する免疫複合体を検出し、該免疫複合体の量と、ステップ(2)で検出された免疫複合体の量を比較し、比較結果に基づき、抵抗力の強さを判定するステップ
(a) VH1-69遺伝子を用いた抗体(A型インフルエンザウイルスに対する中和抗体)を大量に保有する者
(b) VH1-69遺伝子を用いた抗体(A型インフルエンザウイルスに対する中和抗体)を少量保有する者
(c) VH1-69遺伝子を用いた抗体(A型インフルエンザウイルスに対する中和抗体)を保有しない者
(d) ワクチン接種後も、VH1-69遺伝子を用いた抗体(A型インフルエンザウイルスに対する中和抗体)が誘導されない者
グループ(a):基本的に対策は必要ない。
グループ(b):H5N1型インフルエンザワクチン接種による、標的中和抗体産生細胞の選択的増殖刺激

グループ(d):グループ1インフルエンザウイルス中和ヒト抗体を予防・治療用抗体として準備し、必要に応じて投与
本発明の第2の局面は本発明の判定方法に用いられるキットに関する。本発明のキットは、VH1-69遺伝子を用いた抗体を検出又は捕捉するため、VH1-69遺伝子を用いた抗体を特異的に認識する抗イディオタイプ抗体を主要構成要素として含む。抗イディオタイプ抗体はその用途に適するように必要に応じて標識化されている。抗体の標識化に使用可能な標識物質の例はフルオレセイン、ローダミン、テキサスレッド、オレゴングリーン等の蛍光色素、西洋ワサビペルオキシダーゼ、マイクロペルオキシダーゼ、アルカリ性ホスファターゼ、β-D-ガラクトシダーゼ等の酵素、ルミノール、アクリジン色素等の化学又は生物発光化合物、32P、131I、 125I等の放射性同位体、及びビオチンを挙げることができる。抗イディオタイプ抗体を認識する標識化2次抗体をキットに含めることもできる。この場合には、通常、無標識の抗イディオタイプが用いられることになる、抗イディオタイプ抗体が固相化されていてもよい。固相化に用いる不溶性支持体は特に限定されない。例えばポリスチレン樹脂、ポリカーボネート樹脂、シリコン樹脂、ナイロン樹脂等の樹脂や、ガラス等の水に不溶性の物質からなる不溶性支持体を用いることができる。不溶性支持体への抗体の担持は物理吸着又は化学吸着によって行うことができる。
(1)ウイルス
本検討では以下のインフルエンザウイルス株を使用した。A/H1N1pdm: A/California/7/2009pdm (Cal09), A/Suita/1/2009pdm (Sui09); A/H1N1: A/New Caledonia/20/1999 (NC99), A/Solomon Islands/3/2006 (SI06), A/Brisbane/59/2007(Bri07). A/H3N2: A/Panama/2007/1999. A/H5N1: A/Indonesia/5/2005/PR8-IBCDC-RG2. 括弧内は略称。
既報の方法(参考文献8)に従ってファージ抗体ライブラリーを構築した。概要を説明すると、まず、ワクチン接種の前後で、1947年生まれのドナーからアフェレーシスで単球(血液3Lに相当する量)を回収した。回収した単球は8.0 x 108個(ワクチン接種前)又は1.2 x 109個(ワクチン接種後)のBリンパ球を含んでいた。既報のファージディスプレイ方法(参考文献17)に従って、巨大なコンビナトリアル抗体ライブラリーを構築した。ライブラリーサイズは、ワクチン接種前ではH鎖が1.6 x 109 クローン、L鎖が2.0 x 109クローン、Fabが1.4 x 1010、ワクチン接種後ではH鎖が3.2 x 109 クローン、L鎖が1.3 x 109クローン、Fabが2.6 x 1010クローンであった。
既報の方法(参考文献17、18)に従い、ウイルス粒子に結合するファージを選択した。概要を説明すると、ホルマリン処理後のCal09株又はBri07株ウイルス粒子を使用してスクリーニングした。2回又は3回のパニングの後、E. coli (DH12S)細胞に溶出ファージを感染させ、100μg/mlアンピシリン及び0.2%グルコースを含むLBプレートに播種した。ファージミドを保有するE. coliのコロニーをピックアップし、100μg/mlアンピシリン、0.05%グルコース及び1 mM isopropyl-β-D-thiogalactopyranosideを含む2 x YT培地を使用し、30℃で終夜培養した。E. coliの生育に伴い、Fab-cp3型抗体が培地中に分泌される(参考文献19)。Fab-cp3分子を含む培養上清を、H1N1ウイルス(スクリーニングに使用したウイルス株)及びH3H2ウイルスを抗原としたELISAに供した。H1のみに結合性を示すクローンを選択し、更なる解析に使用した。
ホルマリン処理ウイルス粒子を96ウェルMaxisorp immunoplate (Nunc)にコートし、E. coliの培養上清に含まれるFab-cp3抗体を各ウェルに添加した。ウサギ抗cp3抗体(株式会社医学生物学研究所)とインキュベートした後、ウェルにペルオキシダーゼ結合ヤギ抗ウサギIgG (H + L鎖; 株式会社医学生物学研究所)を添加し、更にインキュベートした。その後、HRP基質 (OPD;和光純薬工業株式会社)を各ウェルに添加し、発色させた。H2SO4の添加によりペルオキシダーゼ反応を停止させた後、492nmの吸光度を測定した。
抗体クローンから単離したVH断片のヌクレオチド配列をGenomeLab Dye Terminator Cycle Sequencing Quick Start Kit (BECKMAN COULTER)及びCEQ2000 DNA Analysis System (BECKMAN COULTER)を使用して決定した。T7ETZ (5’-TAATACGACTCACTATAGGG-3’(配列番号129)をVHシークエンス用プライマーに使用した。
ウイルス中和活性を測定するために、シングルサイクル法(VN)又はマルチサイクル法(M-VN)を実施した。200又は500μg/mlのFab-pp型抗体又は2倍段階希釈した血清と等量の100 FFU インフルエンザウイルスを混合し、96ウェルプレートに用意したMDCK細胞に添加した。VN法では、混合物をインキュベートした後、血清フリーのMEMで洗浄し、0.4% BSAを含むMEMを使用して、37℃で15時間、培養した。M-VN法では、インキュベートの後、混合物を除くことなく、混合物と等量の0.4 % BSA、5μg/mlアセチル化トリプシン、0.5 %メチルセルロースを含むMEMを細胞に添加し、37℃で28時間、培養した。続いて、細胞をエタノールで固定化し、PAP(ペルオキシダーゼ及び抗ペルオキシダーゼ)複合体で染色した。フォーカス当たり1以上の細胞(VN法の場合)を含むフォーカス又は4以上の細胞(M-VN法の場合)を含むフォーカスの数を計数した。結果を、Fab-pp型抗体についてはフォーカス減少率(%)で表し、血清については50%フォーカス減少率を示す、血清の希釈倍率の逆数で表した。
既報の方法(参考文献20)に従いHI試験を行った。概要を説明すると、段階希釈した160μg/mlの精製Fab-pp型抗体又はドナー血清(PBSで希釈)を用意した。段階希釈したFab-pp型抗体又は血清をウェル当たり4 HAユニットのウイルスとプレインキュベートした。PBSで希釈した0.75 %のモルモット赤血球細胞を各ウェルに添加し、室温で30~60分インキュベートした。結果は、ヘマグルチニン凝集を阻害した、Fab-pp型抗体の最小濃度 (μg/ml)又は血清の最大希釈倍率の逆数で表した。
ウイルス株に対する結合活性の検出用抗体としてFab-pp型抗体又はC179を、競合物質としてFab-cp3型抗体を用いて、競合ELISAを実施した。使用前に培養上清内のFab-cp3分子を20倍濃縮した。ホルマリン不活化ウイルス粒子を96ウェルMaxisorp immunoplateにコートした。最適濃度にしたFab-pp型抗体(総量50μl)を50μlの20倍濃縮Fab-cp3型抗体と混合し、ウイルスでコートしたウェルに添加した。次に、ペルオキシダーゼ結合ウサギ抗ストレプトアビジン抗体を2次抗体として各ウェルに添加した。終濃度0.25μg/mlのC179を使用したときには、ペルオキシダーゼ結合ヤギ抗マウスIgG(H + L鎖; MBL)を2次抗体に使用した。続いて、HRP基質(OPD; 和光純薬株式会社)を各ウェルに添加し、発色させた。H2SO4の添加によりペルオキシダーゼ反応を停止させた後、492nmの吸光度を測定した。
5種類の1-69抗体のFab-pp型 (F081-007, F083-103, F083-115, F083-305, F083-311)を精製し、完全フロイントアジュバントとともにマウスの足底球に一回注入し、11日後にアジュバントなしでさらに一回注入した。2回目の注入の3日後、リンパ球を鼠径リンパ節から単離し、ミエローマ株化細胞P3-X63AG8.653と融合した。ハイブリドーマをクローニングした後、培養液を上述の1-69抗体とVH1-69遺伝子を使用していない様々な種類の抗体に対し、ELISAによりスクリーニングした。複数のVH1-69遺伝子を使用している抗体には結合するが、VH1-69遺伝子を使用していない抗体には結合しない抗体を選択した。K1-18抗体の単離は、株式会社モノクローナル抗体研究所(札幌, 日本)が実施した。
ヒトIgG ELISA Quantitation set (Bethyl Laboratories, Inc)を多少の修正を加えて使用した。VH1-69遺伝子を使用するIgGの検出用にK1-18抗体を、標準曲線を作成するためにアフィニティー精製ヒトIgGコート抗体(affinity purified Human IgG coating antibody)を96ウェルMaxisorp immuonoplateにコートした。血清及びヒト参照血清(Human reference serum;標準曲線用)を所定のウェルに添加した。HRP結合ヒトIgG検出抗体とインキュベート後、TMB基質を各ウェルに添加した。H2SO4を添加してペルオキシダーゼ反応を停止させ、450nmの吸光度を測定した。ヒト参照血清を用いて作成した標準曲線を利用してK1-18抗体に結合したIgGの濃度を算出した。
前述のウイルス中和試験を修正して使用した。概要を説明すると、RDEで処理した血清を血清フリー培地で希釈し(1:10又は1:20の希釈率)、等量のK1-18抗体(800又は1,600 μg/ml)と混合した。インキュベート後、2倍段階希釈し、等量のインフルエンザウイルス(100 FFU)と混合し、96ウェルプレートに用意したMDCK細胞に添加した。インキュベート後、細胞をエタノールで固定し、PAP(ペルオキシダーゼ及び抗ペルオキシダーゼ)複合体で染色した。50%フォーカス減少率を示した、血清の希釈倍率の逆数をウイルス中和活性とした。
(1)異なる特徴を持つ2タイプの中和抗体
S-IOVのワクチンを接種することで誘導された抗体のレパートリーを解析した。ドナーは1947年生まれで、子供の頃に数回インフルエンザに感染し(おそらくH1N1型ウイルスとH2N2型ウイルス)、1968年にも感染している(おそらくH3N2型ウイルス)。その後41年間、インフルエンザにはかかっておらず、ワクチンも接種していない。ワクチン接種と血液採取のスケジュールは図4に示した。ワクチン接種と血液採取は、2009年10月末から12月中ごろの間に行っており、その間S-IOVによる自然感染はなかった。ワクチン接種前後におけるBリンパ球からそれぞれ巨大ライブラリーを作製し、パンデミックH1N1ワクチン株(A/California/2009pdm)と季節性H1N1ワクチン株でスクリーニングを行った。2回または3回パニングを行った段階で120クローンをそれぞれ単離し、スクリーニングに使ったH1N1株に結合するクローンの解析をさらに行った。H1N1とH3N2両株に同じ強さで結合するクローンは抗NP抗体である可能性が高いため、解析からは除外した(参考文献8)。それぞれのスクリーニングで単離された240クローンのうち、抗HA抗体であると判断したクローンは次のようであった。スクリーニング1 (ワクチン接種後のBリンパ球より作製したライブラリーをパンデミック株でスクリーニング), 105クローン; スクリーニング2 (ワクチン接種前のBリンパ球より作製したライブラリーをパンデミック株でスクリーニング), 3クローン; スクリーニング3 (ワクチン接種後のBリンパ球より作製したライブラリーを季節性株でスクリーニング), 58クローン; スクリーニング1 (ワクチン接種前のBリンパ球より作製したライブラリーを季節性株でスクリーニング), 16クローン。これらクローン全てのVHの塩基配列を決定し、そのアミノ酸配列を比較した結果、182クローンは96種類のモノクローナル抗体からなることが判明した。さらにアミノ酸配列の類似性、特にCDR3領域の同一性から、96種類は63グループに分類できた。VHのアミノ酸配列を図5に示した。
実質的にタイプ1の抗体はすべてHI活性を示すので、それらのエピトープはシアル酸結合ポケットの周辺領域であると考えられる。これら抗体が認識するエピトープの相対的な位置を体系的に確かめるために、以前の研究で使った競合実験を行った(参考文献10)。ライブラリーのスクリーニング後は、それぞれの抗体はFab-cp3型であるが、それらは簡単にFab-pp型に変換できるようにベクター内で設計されている(参考文献11)。もし、クローンAが認識するエピトープがクローンBの認識するエピトープと重複しているならば、Fab-cp3型のクローンBが過剰量存在する時には、Fab-pp型のクローンAのHAに対する結合活性は大きく妨げられることになる。この原理を基にして、図1で示したタイプ1の抗体から選ばれた17クローンを使って、競合実験を行った。図2で示したように、17クローンのうち14クローンは、HAの結合に関しては互いによく競合した。F082-317、F082-254、F082-022の3クローンは、結合阻害の程度が低かった。この結果は、他のデータと一致する。F082-022はHI活性が非常に低い(タイター:160 μg/ml)。F082-254はパンデミックウイルスだけでなく季節性ウイルスにも結合し、両ウイルス株を中和する。さらに、高い変異導入率を示している(18%)。F082-317は、HAに対する結合活性は高いが、中和活性は相対的には低い。
タイプ2のクローンは32グループに分類されるが、3クローンを除いて全てVH1-69遺伝子を使っていた。さらに、これら抗体の大半がH1N1ウイルスだけでなくH5N1ウイルスも中和できた。このことから、いくつかのグループが示してきたように、C179抗体が認識するエピトープの近くにこれら抗体のエピトープがあると考えられた(参考文献12、13)。そこで、C179抗体のHAに対する結合が、これらクローンのFab-cp3型が過剰量存在することで妨げられるかどうかを検討した。図3に示したように、VH1-69遺伝子を使っていない3クローンを含むクローンの全てが、C179のHAへの結合を阻害した。このようにタイプ2のクローンは、すでに他グループが報告しているCR6261やF10のようなVH1-69遺伝子を使っているほかの抗体と同じかよく似た方法でHAの膜近傍の軸索(stem)に結合していると考えられる(参考文献12、13)。図3では、F081-268、F082-243、F082-237、F083-373の4クローンは他のクローンよりも阻害活性が低いことを示している。図1で示したデータで、F081-268はHA結合活性は強いが中和活性は弱いことを示している。他の3クローンは結合活性も中和活性も相対的に弱い。このように、タイプ2の抗体が認識するエピトープは、C179が認識するエピトープと正確には同じではないが、C179のエピトープと全く異なる場所をエピトープとするクローンはなかった。このことは、このエピトープがグループ1ウイルス間で共有されており、安定に保たれていることを示唆している。このエピトープの免疫原性がたとえ弱かったとしても、一度ヒトがこのエピトープを認識する抗体を産生するB細胞を獲得してしまえば、そのB細胞は体内でメモリー細胞として長期間残り、グループ1ウイルスに対し主要な働きをするだろう。興味深いことに、F083-115はVH1-69遺伝子を使っているが、H3ウイルスも中和できる。
3種類のウイルス(H1N1パンデミック株、季節性H1N1株とH5N1株)に対する2種類の生物活性、HI活性とウイルス中和活性を図4で示したように、異なった日にドナーから採取した8種類の血清サンプルに対して測定した。
以前に報告した研究において本発明者らの研究グループは、1968年から2004年の間に単離された12種類の異なるH3株に対する抗HA抗体の多量の単離を通して、3人のヒトの中和抗体レパートリーを解析した(参考文献8、10、14)。1960年と1944年に生まれた二人のドナーから単離された抗体の大半は、異なる株特異性を持つ3つのグループ(1968~1973の株特異的、1977~1993の株特異的、1997~2003の株特異的)に分類された。HA頭部に位置するA、B、C、D、Eといった5つのサイトが中和エピトープとして単離されているが、1977~1993年の株を中和するクローンの多くはサイトCに結合した。1974年に生まれた3番目のドナーから単離されたクローンは4タイプに分類できた。タイプ1は1973年株に強く結合し、株特異性も狭かった。タイプ2は1997年~2003年の株のHAに結合した。他の2タイプは幅広く株を中和する抗体であった。ひとつはH3N2全てを中和する抗体であり、もうひとつはH3だけでなくH1、H2、H5を含むグループ1のウイルスも中和する抗体であった。ウイルスを中和することのできる抗体を作る一連のB細胞が感染やワクチン接種によって免疫されることにより作られた後、更なる抗原刺激により、異なるコースをたどるだろう。あるB細胞は消えてなくなり、他のB細胞はメモリー細胞として残る。さらに、それらは長寿命のメモリー細胞と短寿命のメモリー細胞になるはずである。ほぼ毎年インフルエンザを経験しているヒトは、以前のウイルスから変化した新しいインフルエンザウイルスに感染する機会があるはずである。あるメモリー細胞は新しいウイルスを中和できる抗体を産生し、ある細胞は中和できない抗体を作る。それらは、抗原刺激があるかないかで選択されているはずである。この仮説に従えば、エピトープの安定性はメモリーB細胞の運命に大きな影響を与えていると考えられる。今回の研究では、グループ1ウイルスを中和することのできる1-69抗体を産生する細胞のみが、ドナーの体内にメモリー細胞として残っていたことを示していた。この実験では、若い頃にはインフルエンザを経験したが、その後全くかかっていないヒトを解析の対象とした。ドナーはインフルエンザワクチンをこれまで接種していないため、ドナーの体内で作られた抗体レパートリーは、生きているウイルス感染の影響のみで作られたと考えられる。ホルマリン処理されたウイルスで何回もワクチンを受けているヒトの体内で形成された抗体レパートリーは、この研究で示されたようにシンプルな傾向は示さないだろう。さらに、この研究では一人のヒトを対象としている。しかし、全てのヒトでグループ1ウイルスを幅広く中和することのできる1-69抗体を作ることができると思われる。なぜなら、VH1-69遺伝子のみが、VLドメインがなくても抗原結合サイトを形成して幅広く中和活性を示す抗体を作ることができ、さらに、VHのCDR3領域の、HAの軸索部分への結合への関与は限られているようである。タイプ2の抗体が病原性鳥インフルエンザ(HPAI)H5N1ウイルスによって将来引き起こされると考えられているパンデミックでの感染を防ぐのに十分に強力であるかどうかは更なる検証が必要であるが、タイプ2抗体を産生するクローンがメモリー細胞として存在していることは、その体内でパンデミックウイルスが増殖するのを防ぐのに役立つといえる。H5N1パンデミックへの対処法は、それぞれの個人の免疫プロフィールによって構築されたほうが良いと考えられる。VH1-69遺伝子を使っている抗HA抗体(1-69抗体)の存在は、インフルエンザウイルスを幅広く中和する抗体が体内に存在していることを示すのに役立つ指標となる。また、1-69抗体に対する抗イディオタイプ抗体は、インフルエンザウイルスを幅広く中和する抗体の検出に有用な試薬となるであろう。

Claims (8)
- 被検者由来の生体サンプル中における、VH1-69遺伝子を用いた抗体の存否を指標として、A型インフルエンザウイルスに対する抵抗力を判定する方法。
- 以下のステップ(1)~(3)を含む、請求項1に記載の方法:
(1)インフルエンザワクチン接種後の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させるステップ;
(2)生成する免疫複合体を検出するステップ;
(3)ステップ(2)の検出結果に基づきA型インフルエンザウイルスに対する抵抗力の強さを判定するステップであって、検出された免疫複合体の量がA型インフルエンザウイルスに対する抵抗力の強さの指標となるステップ。 - 以下のステップを更に含む、請求項2に記載の方法:
(4)インフルエンザワクチン接種前の被検者の生体サンプルと、VH1-69遺伝子を用いた抗体を認識する抗イディオタイプ抗体とを接触させることにより生成する免疫複合体を検出し、該免疫複合体の量と、ステップ(2)で検出された免疫複合体の量を比較し、比較結果に基づき、抵抗力の強さを判定するステップ。 - 生体サンプルが血液サンプルである、請求項2又は3に記載の方法。
- A型インフルエンザウイルスが、H1N1型、H1N2型、H2N2型、H3N2型、H5N1型、H5N2型、H6N1型、H7N2型、H7N3型、H7N7型、H7N9型、H9N2型及びH9N1型からなる群より選択される一又は二以上のウイルスである、請求項1~4のいずれか一項に記載の方法。
- A型インフルエンザウイルスが、H1N1型、H2N2型、H3N2型及びH5N1型からなる群より選択される一又は二以上のウイルスである、請求項1~4のいずれか一項に記載の方法。
- A型インフルエンザウイルスが、H1N1型、H1N2型、H2N2型、H5N1型及びH5N2型らなる群より選択される一又は二以上のウイルスである、請求項1~4のいずれか一項に記載の方法。
- VH1-69遺伝子を用いた抗体を特異的に認識する抗イディオタイプ抗体を含む、A型インフルエンザウイルスに対する抵抗力判定用キット。
