Abstract
La présente invention concerne une nouvelle cible de Mycobacterium tuberculosis et son utilisation dans le criblage de médicaments contre la tuberculose, plus précisément concerne une asparaginase recombinante de Mycobacterium tuberculosis, un inhibiteur de ladite protéine, ainsi qu'une méthode de criblage pour sélectionner ledit inhibiteur. La présente invention propose également une souche mutante de Mycobacterium tuberculosis.

La présente invention concerne une nouvelle cible de Mycobacterium tuberculosis et son utilisation dans le criblage de médicaments contre la tuberculose, plus précisément concerne une asparaginase recombinante de Mycobacterium tuberculosis, un inhibiteur de ladite protéine, ainsi qu'une méthode de criblage pour sélectionner ledit inhibiteur. La présente invention propose également une souche mutante de Mycobacterium tuberculosis.
La tuberculose, maladie infectieuse causée principalement par Mycobacterium tuberculosis, est responsable de près de 1,5 millions de décès chaque année dans le monde et on estime qu'un tiers de la population mondiale est infecté. Le traitement actuel consiste en une quadrithérapie (isoniazide, rifampicine, pyrazinamide, ethambutol) pendant deux mois, suivi d'une bithérapie (isoniazide, rifampicine) pendant 4 mois. Ce traitement est donc long et contraignant. Par ailleurs, il n'est adapté qu'au traitement des tuberculoses dues à des souches sensibles aux antibiotiques. L'émergence de souches multi-résistantes et extrêmement résistantes a obligé les autorités de santé publique à implémenter un traitement renforcé incluant des antibiotiques de seconde intention (aminoglycosides, floroquinolones etc).
Cette lourdeur du traitement et l'émergence de souches résistantes obligent au développement de nouveaux antibiotiques contre la tuberculose, dirigés contre de nouvelles cibles.
Les cibles connues des molécules antituberculeuses sont nombreuses, par exemple les ribosomes, notamment les ARN polymérases (Kim et al., 2011, Nucleic Acid Research, vol. 39, No.6), ainsi que des enzymes impliquées dans la synthèse de composés de l'enveloppe des bactéries ou la croissance des bactéries, tels que la protéine ESX-3 qui est responsable de l'acquisition de fer et de zinc par des bactéries (Serafmi et al, 2009, J. Bacteriol, 191, 6340- 6344), ou la protéine CarD qui contrôle la transcription d'ARNr (Stallings et al., 2009, Cell, 138, 146-159).
Cependant, les antibiotiques visant ces cibles exigent des polythérapies très longues (6-8 mois dans le meilleur des cas).
II est nécessaire de trouver de nouvelles cibles thérapeutiques car des souches résistantes aux antibiotiques couramment utilisés apparaissent (mutées sur les gènes cibles de ces antibiotiques).
Par conséquent, il est nécessaire et urgent d'identifier de nouvelles cibles antituberculeuses chez M. tuberculosis et de développer des antibiotiques agissant spécifiquement sur ces cibles, seuls ou en combinaison avec les antibiotiques existants.
Une asparaginase est capable d'hydrolyser l'asparagine en aspartate et ammonium, ce qui permet aux bactéries d'utiliser l'asparagine comme source d'azote et de carbone.
Bien que la séquence de l'enzyme Ans A chez Mycobacterium tuberculosis soit connue (AnsA/Rvl538c (TubercuList), Malen et al, 2010, BMC Microbiol. 10: 132) et qu'elle soit annotée comme étant une asparaginase, aucun travaux scientifique antérieur permet de montrer la présence effective d'une asparaginase chez Mycobacterium tuberculosis et l'activité enzymatique de la protéine prédictive Rvl538c.
La présente invention repose sur la constatation pour la première fois faite par les Inventeurs que l'enzyme AnsA de Mycobacterium tuberculosis est requise pour la colonisation du poumon par ces bactéries et que l'inactivation de cette enzyme permet d'atténuer ces bactéries. Par ailleurs, AnsA n'a pas d'homologue chez l'homme, ce qui permet à ce gène ainsi que la protéine codée par ce gène de devenir une cible spécifique pour développer de nouveaux antibiotiques.
La présente invention a pour objet une souche mutante de Mycobacterium tuberculosis dans laquelle l'activité de l'asparaginase est absente ou réduite.
La présente invention concerne une souche mutante de Mycobacterium tuberculosis dont la virulence est inférieure à celle de la souche sauvage correspondante, le gène ansA représenté par la séquence SEQ ID NO : 2 étant inactivé ou l'expression de la protéine codée par le gène ansA représenté par la séquence SEQ ID NO : 2 étant inactivée dans ladite souche mutante.
On entend par « une souche mutante de Mycobacterium tuberculosis dont la virulence est inférieure à celle de la souche sauvage » une souche induisant une charge bactérienne pulmonaire significativement plus faible que celle induite par la souche sauvage chez la souris après 3 semaines d'infection.
Une valeur est considérée comme étant significative lorsque la valeur P est inférieure à 0.05 selon le test statistique de Student (test t).
On entend par l'expression « le gène ansA... étant inactivé » le fait qu'une mutation, une délétion ou une insertion d'un ou plusieurs nucléotides est présente à l'intérieur du gène
ansA représenté par SEQ ID NO : 2 et qu'elle inhibe la transcription de l'ARN messager dudit gène.
L'inactivation du gène ans A peut être mise en œuvre par toutes les méthodes de mutagenèse connues dans l'art antérieur, notamment par l'insertion d'un transposon, par la recombinaison homologue ou par approche utilisant des phages
Dans ce mode de réalisation, le gène ansA est interrompu, modifié ou supprimé et la transcription de l'ARN messager de ans A n'a pas lieu ou ne peut pas aboutir.
Dans un autre mode de réalisation, la protéine AnsA n'est pas exprimée chez Mycobacterium tuberculosis, alors que le gène ansA est intact dans le génome de ladite bactérie.
On entend par « l'expression de la protéine... étant inactivée » le fait que l'ARN messager du gène ansA est inactivé ou déstabilisé, par une régulation post-transcriptionnelle de l'ARN messager, ou qu'une protéine tronquée ou non fonctionnelle est produite ou que la protéine correctement traduite est déstabilisée par une régulation post-traductionnelle, par exemple la protéolyse.
Une régulation post-transcriptionnelle peut être effectuée par la technique telle que siARN, miARN ou ARN antisens.
Le gène de séquence SEQ ID NO : 2 code l'asparaginase naturelle de Mycobacterium tuberculosis représentée par la séquence SEQ ID NO : 3.
En effet, contre toute attente, Ans A n'est pas une protéine essentielle à là vie de la bactérie, elle participe seulement à sa virulence. Par conséquent, l'inactivation de cette protéine permet de diminuer le nombre de mutants résistants pouvant apparaître (pression de sélection moins forte) et faciliter l'élimination du pathogène par le système immunitaire.
Selon un mode de réalisation particulier, la virulence de la souche mutante de Mycobacterium tuberculosis est déterminée par infection de souris par voie intranasale et mesure de la charge bactérienne pulmonaire à différents temps après infection.
La charge bactérienne pulmonaire est mesurée par étalement sur milieu gélosé.
Dans un mode de réalisation particulier d'une souche mutante de Mycobacterium tuberculosis selon l'invention, le gène ansA représenté par la séquence SEQ ID NO : 2 est supprimé du génome de ladite souche mutante.
La suppression du gène peut être obtenue, à titre d'exemple, par la recombinaison homologue.
Dans un mode de réalisation particulier de l'invention, ladite souche mutante de Mycobacterium tuberculosis est la souche déposée à la CNCM le 21 mars 2013 sous le numéro : CNCM 1-4724.
Dans un autre mode de réalisation particulier d'une souche mutante de Mycobacterium tuberculosis selon l'invention, le gène ans A représenté par la séquence SEQ ID NO : 2 est présent dans le génome de la dite souche mutante, mais inactivé.
Cette inactivation peut être due à une modification ou interruption du gène, par à titre d'exemple un transposon ou une mutagenèse dirigée par PCR.
La présente invention a également pour objectif de fournir une composition immunogène comprenant une souche mutante de Mycobacterium tuberculosis susmentionnée.
La présente invention a aussi pour objet une composition pharmaceutique comprenant à titre de substance active, une souche mutante de Mycobacterium tuberculosis susmentionnée, en association avec un véhicule pharmaceutiquement acceptable.
Ladite composition pharmaceutique peut être utilisée dans le traitement ou la prévention de la tuberculose, notamment la tuberculose liée à l'infection de Mycobacterium tuberculosis.
Dans un mode de réalisation avantageux, ladite composition pharmaceutique est une composition vaccinale contre la tuberculose.
Dans un mode de réalisation avantageux, ladite composition pharmaceutique ou vaccinale comprend une souche mutant déposée le 21 mars 2013 sous le numéro CNCM I- 4724.
Ladite composition pharmaceutique ou vaccinale peut être une solution à pH neutre, notamment de pH 7.0 à 8.0, notamment de pH 7.4 à 7.6.
Ladite composition pharmaceutique ou vaccinale peut être en poudre lyophilisée. La solution est préparée juste avant l'administration.
Ladite composition pharmaceutique ou vaccinale convient d'être administrée à une dose permettant de provoquer une réponse immunitaire protective contre la tuberculose sans développement de ladite maladie.
Dans un mode de réalisation particulier, la dose administrable de ladite composition pharmaceutique ou vaccinale est une dose unitaire de 10 e5 à 10 e6 CFUs selon l'âge de la personne à vacciner.
La présente invention a également pour objet un kit contenant une composition pharmaceutique susmentionnée et un antibiotique connu dans le traitement de la tuberculose.
Un kit selon la présente invention comprend :
une composition pharmaceutique ou vaccinale susmentionnée, et
- un antibiotique choisi parmi ethambutol, isoniazide, pyrazinamiden rifampicine, acide p-aminosalicylique, amikacine, kanamycine, capreomycine, ciprofloxacirie, levofloxacine, moxifloxacine, ethionamide, rifabutine,
comme produit de combinaison pour une utilisation simultanée, séparée ou étalée dans le temps pour la prévention et/ou le traitement de la tuberculose, notamment la tuberculose liée à l'infection de Mycobacterium tuberculosis .
Dans un mode de réalisation particulier, un kit selon la présente invention comprend : une composition pharmaceutique ou vaccinale comprenant la souche mutant déposée à la CNCM sous le numéro CNCM 1-4724 le 21 mars 2013, et un antibiotique choisi parmi ethambutol, isoniazide, pyrazinamiden rifampicine, acide p-aminosalicylique, amikacine, kanamycine, capreomycine, ciprofloxacine, levofloxacine, moxifloxacine, ethionamide, rifabutine
comme produit de combinaison pour une utilisation simultanée, séparée ou étalée dans le temps, pour la prévention et/ou le traitement de la tuberculose, notamment la tuberculose liée à l'infection de Mycobacterium tuberculosis.
La présente invention a également pour objectif de mettre à disposition une asparaginase recombinante de Mycobacterium tuberculosis.
Plus particulièrement, la présente invention concerne une protéine recombinante représentée par la séquence SEQ ID NO : 1.
Ladite protéine recombinante comporte un tag His6 permettant la purification de ladite protéine par une colonne de purification, telle qu'une colonne Ni-NTA, et son suivi par micoscopie en utilisant un anticorps anti-HIS6.
Un autre objectif de la présente invention est de fournir une méthode de criblage d'un inhibiteur de l' asparaginase de M. tuberculosis représentée par la séquence SEQ ID NO : 1.
La protéine recombinante de Γ asparaginase de la présente invention, notamment la protéine recombinante de la séquence SEQ ID NO : 1, permet de mettre en œuvre la méthode de criblage d'inhibiteur de cette protéine dans des essais à haut débit.
La méthode de criblage de la présente invention comprend :
(i) la mise en contact de la protéine recombinante de séquence SEQ ID NO : 1 avec l'asparagine dans une solution réactionnelle en présence d'une molécule issue d'une banque de molécules chimiques,
(ii) la comparaison de l'activité de ladite protéine recombinante en présence de la susdite molécule avec celle de ladite protéine recombinante en absence de la susdite molécule,
(iii) l'identification d'un inhibiteur de ladite protéine recombinante lorsque
l'activité de ladite protéine recombinante en présence de la susdite molécule est inférieure à celle en absence de la susdite molécule.
La solution réactionnelle utilisée dans ladite méthode peut être le réactif Nessler.
On entend par « inhibiteur de l'asparaginase » une molécule biologique ou chimique permettant d'inhiber l'hydrolyse de l'asparaginase en aspartate.
Dans un mode de réalisation particulier, un tel inhibiteur biologique peut être un anticorps polyclonal ou monoclonal dirigé contre la protéine recombinante représentée par la séquence SEQ ID NO : 1 , un siARN, un miARN ou un oligonucléotide antisens inhibant la transcription du gène codant ladite protéine.
Un anticorps polyclonal ou monoclonal peut être obtenu par les méthodes de production d'anticorps connues dans l'art antérieur.
Un anticorps polyclonal peut être obtenu par immunisation d'un lapin ou d'une chèvre par la protéine de séquence SEQ ID NO : 1 purifiée. La dose d'immunisation convenable peut être déterminée par les expériences routines. La protéine de séquence SEQ ID NO : 1 peut être administrée avec un adjuvant d'immunisation, tel que l'adjuvant de Freund, pour optimiser la réponse immunitaire.
Un anticorps monoclonal peut être obtenu par la construction d'un hybridome, qui est la fusion du lymphocyte sécréteur d'un anticorps dirigé contre la protéine de séquence SEQ ID NO : 1 ayant servi à l'immunisation de l'animal avec une cellule de myélome.
Dans un mode de réalisation plus particulier, ledit inhibiteur selon l'invention est un anticorps et dirigé contre l'asparaginase de Mycobacterhim tubercidosis.
Dans un autre mode de réalisation, un inhibiteur chimique peut être une molécule issue du criblage dans une banque de molécules.
Un autre objet de la présente invention est un produit contenant un inhibiteur de la protéine de la séquence SEQ ID NO : 1 ou l'asparaginase de Mycobacterium taberculosis et un antibiotique connu pour le traitement de la tuberculose comme produit de combinaison.
Plus particulièrement, ledit kit de l'invention comprend :
- un inhibiteur décrit dans la présente invention, et
un antibiotique choisi parmi ethambutol, isoniazide, pyrazinamiden rifampicine, acide p-aminosalicylique, amikacine, kanamycine, capreomycine, ciprofloxacine, levofloxacine, moxifloxacine, ethionamide, rifabutine,
comme produit de combinaison pour une utilisation simultanée, séparée ou étalée dans le temps pour la prévention et/ou le traitement de la tuberculose, notamment la tuberculose liée à l'infection de Mycobacterium tuberculosis.
Les figures et les exemples ci-après illustrent à titre d'exemple la présente invention et ne limitent en aucun cas la portée de la présente invention.

Figure 2 : la figure 2 illustre l'expression de la protéine de séquence SEQ ID NO : 1 (l'asparaginase de Mycobacterium tuberculosis) chez Mycobacterium smegmatis (non pathogène) via le plasmide pVV16. L'expression de l'asparaginase recombinante est détectée par le test « western blot ». Le plasmide pVV16 vide est utilisé en tant que contrôle négatif. « F40o » correspond à la fraction AnsA purifiée. « L » correspond au lysat bactérien total.
Figure 3 : les figures 3 A et 3B illustrent l'activité de l'asparaginase recombinante dans les fractions purifiées F40o.
La figure 3A montre la disparition du NADPH en présence de l'asparagine et l'asparaginase recombinante (la ligne comportant les losanges). Un échantillon contenant le plasmide pVV16 et l'asparagine (la ligne comportant les points), un échantillon contenant seulement l'asparaginase recombinante (la ligne comportant les carrés) et un échantillon contenant seulement le plasmide pVV16 (la ligne comportant les triangles) sont respectivement utilisés en tant que contrôle.
La figure 3B confirme que l'asparaginase recombinante ne possède pas l'activité glutaminase (la ligne comportant les points). Un échantillon contenant l'asparaginase
recombinante et 50nM de l'asparagine (la ligne comportant les losanges) et un échantillon contenant l'asparaginase et 50nM de glutamine (la ligne comportant les triangles) sont respectivement utilisés en tant que contrôle.
Figure 4 : la figure 4 montre que le mutant de M. tuberculosis inactivé dans le gène ansA (ansA Ko) ne se développe pas in vitro en présence d'asparagine comme seule source d'azote, alors que la souche sauvage (wt) se multiplie. La croissance bactérienne est quantifiée par mesure de la turbidité (unités Me Farland) au cours du temps (en jours)
Figure 5 : la figure 5 montre qu'à 21 jours et 42 jours après l'infection le mutant de M. tuberculosis inactivé dans le gène codant AnsA (colonne en noir) est présent en quantité (mesurée en CFUs) 30 fois moindre dans les poumons de souris infectées, par rapport à la souche sauvage (colonne en blanc) ou la souche mutante « complémentée » dans laquelle une copie du gène codant AnsA a été ré-introduite (colonne en gris).
Exemples
Matériels et méthodes
Souches bactériennes :
Mycobacterhim smegmatis (souche MC2 155) et Mycobacterhim tuberculosis (H37Rv) sont cultivés en milieu liquide 7H9 (Middlebrook) + ADC 10% (albumine, dextrose, catalase, Middlebrook) supplémenté avec du Tween 80 à 0,05% final ou solide sur milieu 7H11 (Middlebrook) + OADC 10% (acide oléique, albumine, dextrose, catalase, Middlebrook) à 37°C. Quand ceci est nécessaire, de la kanamycine (50 μg/mL) de la zeomycine (25 μg/mL) ou de la streptomycine (25 μξ/raL) sont additionnés. Les tests de croissance (Fig4) ont été effectués en triplicat en milieu Sauton modifié dans lequel le citrate d'ammonium a été omis et où l'asparagine et le glutamate ont été additionnés à une concentration finale de 5mM + Tween 80 à 0,05%> final. La croissance bactérienne a été suivie via la lecture des Me Farland au cours du temps. La construction du mutant ansA KO s'est faite par des méthodes classiques de recombinaison génétiques permettant la détermination de l'essentialité de gènes mycobactériens (ref: voir la partie « supp material » dans Duckworth et al, 2011 Chem Biol. 23;18(11): 1432-41).
Extraction des protéines et purification d'AnsA :
Produits utilisés: tampon de lyse (50mM NaH2P04, 300mM NaCl, lOmM imidazole pH8) ; tampon de lavage (50mM NaH2P04, 300mM NaCl, 20mM imidazole pH8) ; tampon d'élution (50mM NaH2P04, 300mM NaCl, 400 mM imidazole pH8).
A densité optique supérieure ou à égale 1,5, 15mL de culture sont centrifugés (2952g ; 10 min), les culots bactériens sont remis en suspension avec lmL de tampon de lyse, puis les parois sont cassées par lyse mécanique avec des billes de verre (0,1 -0,25mm) pendant 10 minutes (appareil Bead Beater « Retscher, BioBrock scientific »). Les lysats sont récupérés après centrifugation (14000rpm ; 5 min).
Puis 600μΙ, des lysats sont déposés sur colonne Ni-NTA (Ni-NTA Spin column QIAGEN) équilibrée au préalable avec όθθμΐ, de tampon de lyse (890g ; 2 min), les colonnes sont centrifugées (270g ; 5 min) et les fractions F0 sont obtenues. Puis les colonnes sont lavées 2 fois avec 600μΙ, de tampon de lavage (890g ; 2min) et sont enfin éluées avec 300μΙ, de tampon d'élution et après centrifugation (890g ; 2 min) les fractions F400 sont obtenues. Détection de l'expression de AsnA par Western blot :
Les différents échantillons sont dénaturés après ajout d'un agent contenant du sodium dodecylsulfate et du dithiothreitol (NuPAGE, LDS Sample Buffer 4X et Reducing Agent 10X) SDS) et chauffés 5min à 95°C. Les extraits sont déposés sur un gel SDS-PAGE à 12% d'acrylamide. Les protéines sont séparées par migration à 200 mV avant d'être transférées sur une membrane de nitrocellulose. Après saturation 1H en PBS/Tween 20 (0.1%)/lait (5%), la membrane est incubée pendant la nuit à 4°C avec l'anticorps un primaire dirigé contre le Tag His6 (produit chez le lapin, eBioscience). Après 3 lavages de 5 minutes dans du PBS/Tween 20 (0.1%), la membrane est incubée pendant lh avec l'anticorps secondaire (Produit chez l'âne, GE Healthcare) dirigé contre l'anticorps primaire et couplé à la HRP (Horseradish Peroxidase). Après 3 nouveaux lavages, sont ajoutés pendant 5 minutes le substrat chimioluminescent (luminol) ainsi que le substrat de la peroxydase (peroxyde d'hydrogène), le produit de la réaction catalysée par la HRP est impressionné sur un film photographique permettant la détection de la protéine (AnsA-His6= 34kDa). L'activité de l'asparaginase est déterminée par le couplage de deux réactions ci- dessous.

La disparition du NADHP est déterminée par mesure optique.
L'essai enzymatique est réalisé dès la fin de la purification, chaque expérience de purification permet de faire 3 essais enzymatiques. La quantité de NADHP est mesurée par le kit Megazyme K-ASNAM (Glutamate déshydrogénase, NADPH, 2-oxoglutarate, tampon acétate de sodium = « solution A »). La solution d'Asn est à 150mM. Dans une cuve en Quartz de 1cm sont ajoutés 72μΙ, de solution A, 77μί de substrat (Asn et/ou eau de manière à avoir la concentration finale souhaitée) et 81 μί de l'échantillon à tester F400). Une cinétique enzymatique est réalisée et l'activité est donnée par la mesure de la diminution de l'absorbance à 340nm correspondant à la disparition du NADPH au cours du temps avec le spectrophotomètre << Safas monaco me2 » ; logiciel « SAFAS SP 2000 » permettant de faire des mesures à des intervalles de temps de 10 secondes.
Procédé de screening :
Afin de procéder au screening d'inhibiteurs de la protéine AnsA de Mycobacterium tuberculosis, la miniaturisation d'essais d"activité asparaginase est procédée. Pour cela, la protéine AnsA est produite chez Escherichia Coli. Les fractions purifiées de la protéine AnsA sont placées en plaque 96 puits. Après l'addition de l'asparagine 50 mM, l'activité asparaginase est mesurée en point final par spectrocolorimétrie (520 nm) après addition de réactif de Nessler, qui permet de former un complexe coloré avec les ions ammonium libérés. Des molécules provenant de chimiothèques sont ensuite ajoutées dans le milieu réactionnel afin d'identifier celles permettant d'inhiber l'activité asparaginase de AnsA.
Infection de souris :
Des souris C57BL/6 de 6 à 8 semaines ont été anesthésiées avec un coktail de kétamine (100 mg/kg ; Merial) et de xylasine (15 mg/kg ; Bayer) et ont été infectées par voie intranasale avec 500 CFUs (colonies formant unités) des différentes souches testées avec 25 μΐ de DPBS/0.01% Tween 80. A jour 21 et jour 42 post-infection, 5 souris par souches testées sont sacrifiées et les poumons et rates des animaux sont récupérés, broyés et étalés sur milieu 7H11 pour dénombrement bactérien.
Résultats
Le western blot montre que Γ asparaginase recombinante de la séquence SEQ ID NO : lest bien exprimée chez Mycobacterhim smegmatis (figure 2).
Cette asparaginase recombinante présente une activité d'asparaginase permettant d'hydrolyser l'asparagine (figure 3A). Cette même protéine n'a pas d'activité de glutaminase (figure 3B).
Le mutant AnsA KO de M. tuberciilosis ne pousse pas en présence d'asparagine comme seule source d'azote (figure 4).
Les résultats de la figure 5 montrent qu'AnsA est requise pour la colonisation du poumon par M. tuberculosis, et font de cette enzyme une cible de choix pour développer de nouveaux antibiotiques.
Claims

Souche mutante de Mycobacteriiim t berculosis dont la virulence est inférieure à celle de la souche sauvage correspondante, le gène ansA représenté par la séquence SEQ ID NO : 2 ou l'expression de la protéine codée par le gène ansA représenté par la séquence SEQ ID NO : 2 étant inactivée dans ladite souche mutante.
2. Souche mutante de Mycobacterhim tuberculosis selon la revendication 1, dans
laquelle le gène ansA représenté par la séquence SEQ ID NO : 2 est supprimé du génome de ladite souche mutante.
3. Souche mutante de Mycobacterhim tuberculosis selon la revendication 1, dans
laquelle le gène ansA représenté par la séquence SEQ ID NO : 2 est inactivé du génome de ladite souche mutante.
4. Souche mutante de Mycobacterhim tuberculosis selon l'une des revendications 1 à 3, ladite souche est la souche CNCM 1-4724 déposée le 21 mars 2013.
5. Composition immunogène comprenant une souche mutante selon l'une des

7. Composition pharmaceutique, notamment vaccinale, selon la revendication 5 ou 6, convenant pour l'administration d'une dose unitaire de 10 e5 à 10 e6 CFUs.

une composition selon l'une des revendications 5-7,
un antibiotique choisi parmi ethambutol, isoniazide, pyrazinamiden rifampicine, acide p-aminosalicylique, amikacine, kanamycine, capreomycine, ciprofloxacine, levofloxacine, moxifloxacine, ethionamide, rifabutine,
comme produit de combinaison pour une utilisation simultanée, séparée ou étalée dans le temps pour la prévention et/ou le traitement de la tuberculose, notamment la tuberculose liée à l'infection de Mycobacterium tuberculosis.
9. Protéine recombinante représentée par la séquence SEQ ID NO : 1.
10. Méthode de criblage d'un inhibiteur de la protéine recombinante représentée par la séquence SEQ ID NO : 1 , comprenant : un criblage colorimétrique ou fluorimétrique
(iv) la mise en contact de ladite protéine recombinante avec l'asparagine dans une solution réactionnelle en présence d'une molécule issue d'une banque de molécules chimiques,
(v) la comparaison de l'activité de ladite protéine recombinante en présence de la susdite molécule avec celle de ladite protéine recombinante en absence de la susdite molécule,
(vi) l'identification d'un inhibiteur de ladite protéine recombinante lorsque
l'activité de ladite protéine recombinante en présence de la susdite molécule est inférieure à celle en absence de la susdite molécule.
11. Inhibiteur de la protéine recombinante représentée par la séquence SEQ ID NO : 1 , ledit inhibiteur étant choisi parmi :
- un anticorps polyclonal ou monoclonal dirigé contre la protéine recombinante représentée par la séquence SEQ ID NO : 1, en particulier un anticorps dirigé contre l'asparaginase de Mycobacterium tuberculosis, ou
- un inhibiteur issu du criblage selon la revendication 10.
12. Produit contenant :
- un inhibiteur selon la revendication 11 ,
- un antibiotique choisi parmi ethambutol, isoniazide, pyrazinamiden rifampicine, acide p-aminosalicylique, amikacine, kanamycine, capreomycine, ciprofloxacine, levofloxacine, moxifloxacine, ethionamide, rifabutine,
