本発明の抗癌剤に用いるゴボウシ抽出物の製造方法では、原料とするゴボウシを抽出に適した大きさに切裁する。原料となる生薬は、植物の様々な部位や鉱物、動物など種々の大きさ、形状、固さがあり、その特質に応じた切裁が必要となる。
ゴボウシは、当業者に公知の任意の手段を使用して切裁することができる。たとえば、市販の切裁機を使用することができる。
0.4U/g未満の場合は、加水分解が不十分となり、アルクチゲニンの重量比が下がり、所望のゴボウシ抽出物を効率的に得られなくなる。
抽出工程は、生薬抽出物粉末製造工程中で、品質上最も重要な工程である。この抽出工程により、生薬抽出物粉末の品質が決まる。本発明のゴボウシ抽出物の製造方法では、ゴボウシ抽出物を抽出するために、酵素変換工程と有機溶媒による抽出工程の2段階に分けて抽出を行う。
酵素変換工程は、本発明の抗癌剤に用いるゴボウシ抽出物の製造方法において重要な工程である。酵素変換工程は、ゴボウシに内在する酵素であるβ-グルコシダーゼにより、ゴボウシに含まれているアルクチインをアルクチゲニンに酵素変換する工程である。
有機溶媒による抽出工程は、任意の適切な有機溶媒を使用して、ゴボウシからアルクチゲニンおよびアルクチインを抽出する工程である。すなわち、上記の酵素変換工程によりアルクチゲニンが高含量となった状態で、適切な溶媒を添加して、ゴボウシ抽出物を抽出する工程である。たとえば、ゴボウシ抽出物に適切な溶媒を添加して、適切な時間加熱攪拌してゴボウシ抽出物を抽出する。また、加熱攪拌以外にも、加熱還流、ドリップ式抽出、浸漬式抽出または加圧式抽出法などの当業者に公知の任意の抽出法を使用して、ゴボウシ抽出物を抽出することができる。
固液分離工程は、抽出の終わったゴボウシを抽出液から分離する工程である。固液分離は、当業者に公知の任意の方法を使用して行うことができる。固液分離法には、たとえば濾過法、沈降法および遠心分離法などがある。工業的には、遠心分離法が望ましい。
濃縮工程は、乾燥に先立ちゴボウシ抽出液から溶媒を除去する工程である。ゴボウシ抽出液からの溶媒の除去は、当業者に公知の任意の方法を使用して行うことができる。
上記工程によって得られたゴボウシ抽出物を粉末状に仕上げる工程である。乾燥は、当業者に公知の任意の方法を使用して行うことができる。たとえば、乾燥法として、凍結乾燥および噴霧乾燥などが知られているが、実験室レベルであれば前者、量産レベルであれば後者を用いるのが一般的である。
ゴボウシの酵素活性および酵素変換条件(温度と時間)が、アルクチゲニン/アルクチイン(重量比)に及ぼす影響、すなわち両者の因果関係を検証した。
産地やロットが異なるゴボウシをウイレー氏粉砕機により粉砕し、このゴボウシ粉砕品0.1gを10mLの水で希釈し、試料溶液とした。
p-ニトロフェニル-β-D-グルコピラノシド水溶液0.5mLを加えて、反応混液を調製し、37℃で約5分予備加熱を行った。

酵素活性が0.12、0.27、0.40U/g(試料1~3)である切裁したゴボウシ1gに水7mLを加えて、酵素反応温度15℃、20℃の温度条件でそれぞれの反応温度での反応時間を30分に設定し、反応後エタノールを加え80℃で抽出を行い、得られた抽出物のアルクチゲニン及びアルクチインを定量し、アルクチゲニン/アルクチイン重量比を求めた。結果を表1の比較例1~2、実施例1に示す。
酵素活性が4.03U/g(試料5)である切裁したゴボウシ1gに水7mLを加えて、酵素反応温度30℃、40℃、50℃、60℃の温度条件でそれぞれの反応温度での反応時間を15分、30分(30℃と60℃のみ)に設定し、反応後エタノールで抽出を行い、得られた抽出物のアルクチゲニン及びアルクチインを定量し、アルクチゲニン/アルクチイン重量比を求めた。
酵素活性が1.42U/g(試料4)である切裁したゴボウシ1gに水を7mL加えて、酵素反応温度25℃の温度条件で反応時間を10分、30分に設定し、反応後エタノールで抽出を行い、得られた抽出物のアルクチゲニン及びアルクチインを定量し、アルクチゲニン/アルクチイン重量比を求めた。
以上のことから酵素活性1.42U/gであっても所望の結果を得ることができた。
ゴボウシ(酵素活性8.23U/g)を切裁し、9.5mmの篩を全通するものをさらに0.85mmの篩に通し、75%が残ることを確認した。このゴボウシ細切80kgを29~33℃に保温した水560Lに加えて30分間攪拌した。次いで、エタノール265Lを加えて85℃に昇温し、さらに60分間加熱還流した。この溶液を遠心分離し、ゴボウシ抽出液を得た。この操作を2回繰り返して得られた抽出液を合わせて、減圧濃縮し、抽出物固形分に対してデキストリン20%を加えて、噴霧乾燥した。アルクチゲニンおよびアルクチイン含量は、それぞれ6.2%および7.1%であり、アルクチゲニン/アルクチイン(重量比)=0.89のゴボウシ抽出物粉末(デキストリン20%含有)が得られた。

ゴボウシ(酵素活性7.82U/g)を切裁し、9.5mmの篩を全通するものをさらに0.85mmの篩に通し、75%が残ることを確認した。このゴボウシ細切80kgを30~32℃に保温した水560Lに加えて40分間攪拌した後、60分後にエタノール258Lを加えて85℃に昇温し、さらに30分間加熱還流した。この液を遠心分離し、ゴボウシ抽出液を得た。この操作を2回繰り返して得られた抽出液を合わせて、減圧濃縮し、抽出物固形分に対してデキストリン20%を加えて、噴霧乾燥した。アルクチゲニンおよびアルクチイン含量は、それぞれ6.2%および6.7%であり、アルクチゲニン/アルクチイン(重量比)=0.93のゴボウシ抽出物粉末(デキストリン20%含有)が得られた。
ゴボウシ(酵素活性7.82U/g)を切裁し、9.5mmの篩を全通するものをさらに0.85mmの篩に通し、75%が残ることを確認した。このゴボウシ細切80kgを30~32℃に保温した水560Lに加えて30分間攪拌した後、エタノール253Lを加えて85℃に昇温し、さらに40分間加熱還流した。この液を遠心分離し、得られた抽出液を得た。この操作を2回繰り返して得られた抽出液を合わせて、減圧濃縮し、抽出物固形分に対してデキストリン25%を加えて、噴霧乾燥した。アルクチゲニンおよびアルクチイン含量は、それぞれ6.4%および7.2%であり、アルクチゲニン/アルクチイン(重量比)=0.89のゴボウシ抽出物粉末(デキストリン25%含有)が得られた。

「日局」製剤総則、顆粒剤の項に準じて顆粒剤を製造する。すなわち上表に記載の(1)~(3)までの成分をとり、顆粒状に製した。これを1.5gずつアルミラミネートフィルムに充填し、1包あたりゴボウシ抽出物粉末を0.5g含有する顆粒剤を得た。

「日局」製剤総則、顆粒剤の項に準じて顆粒剤を製造する。すなわち上表に記載の(1)~(3)までの成分をとり、顆粒状に製した。これを3.0gずつアルミラミネートフィルムに充填し、1包あたりゴボウシ抽出物粉末を2g含有する顆粒剤を得た。

「日局」製剤総則、錠剤の項に準じて錠剤を製する。すなわち上表に記載の(1)~(6)成分をとり、錠剤を得た。
実施例10の顆粒剤を用いて膵臓癌に対する効果を試験した。
前治療としてゲムシタビン療法を行い不応答であった膵臓癌患者3名を対象として、実施例10の顆粒剤3g(ゴボウシ抽出物粉末1gを含有する)を、1日1回朝食後、連日経口投与した。すなわち、本試験例において患者に投与した1日当たりのアルクチゲニンは59.4mgであり、アルクチインは68.5mgであった。
ゴボウシ細切200gを水(22℃)1Lに加えて1時間攪拌した後、エタノール0.45Lを加えて更に1時間加熱還流した。ガーゼ4枚(金網100mesh)でろ過し、30%エタノール0.5Lで洗浄し、合せた抽出液(1.5L)を凍結乾燥した。このようにして得た、生薬の刻みを冷浸抽出したゴボウシエキスは、アルクチイン及びアルクチゲニン含量がそれぞれ、13.3%及び11.4%で、アルクチゲニン/アルクチイン(重量比)=0.86であった。
(試験方法)
腫瘍モデル動物は、ドナーとなるヌードマウス(BALB-cAJnu/nu;日本クレア)の背皮下にヒトすい臓がん細胞株CAPAN-1もしくは、PSN-1を播種し、得られたドナーマウスの腫瘍塊をレシピエントマウスの背皮下に移植することによって作製した。アルクチゲニン(AG)、アルクチイン(A)およびゴボウシエキス(実施例12)は、DMSOに10mg/mlの濃度で溶解したものを生理食塩水で希釈し、マウス1匹あたり50μgを1週間に5回、胃内に経口投与した。抗腫瘍性は、背皮下の腫瘍塊のサイズを経時的に計測することによって評価した。
前治療としてゲムシタビン療法を行い不応答であった膵臓癌患者15名を対象として、用量制限毒性の発現頻度を調査するためアルクチゲニン含有顆粒剤の第I相試験を行った。この15名には、所定の用量のアルクチゲニン含有顆粒剤を1日1回朝食後、連日経口投与した。この15名のうち3名には、1回あたり、実施例10の顆粒剤3g(ゴボウシ抽出物粉末1g、すなわちアルクチゲニン59.4mgおよびアルクチイン68.5mgを含有する)を投与した。15名のうち別の3名には、1回あたり、実施例10の顆粒剤7.5g(ゴボウシ抽出物粉末2.5g、すなわちアルクチゲニン148.5mgおよびアルクチイン171.25mgを含有する)を投与した。さらに別の9名には、1回あたり、実施例10の顆粒剤12g(ゴボウシ抽出物粉末4g、すなわちアルクチゲニン237.6mgおよびアルクチイン274mgを含有する)を投与した。
次に、癌に対する様々な治療抵抗性や再発・転移巣の出現に深く関与すると考えられる癌幹細胞様集団(CSCs : Cancer Stem-like cells)に対するアルクチゲニンの効果を調べた。
グルコース含有培地は、4.75gダルベッコ変法イーグル培地2(日水製薬)を水に溶解し、12.5ml 1M HEPES pH7.4(DOJINDO、342-01375)を加えて滅菌後、18.5ml 10% NAHCO3、10ml L-glutamine(SIGMA)、5ml Anti-Anti(Life technologies)、5ml MEM NON-ESSENTIAL AMINO ACID SOLUTION(SIGMA)、56℃で30分温浴にて非動化させた50ml FETAL BOVINE SERUM(biowest)を加え、最終的に500mlにして作製した。
膵臓癌細胞PANC-1(ATCC No. CRL-1469)をグルコース含有培養培地に播種して一晩インキュベートした後、グルコース含有培地、グルコース阻害培地、3μMアルクチゲニンを含むグルコース含有培地および3μMアルクチゲニンを含むグルコース阻害培地のそれぞれにおいて24時間培養した。細胞を回収した後、定法にしたがってPI染色(死細胞染色)および癌幹細胞マーカー染色を行い、フローサイトメトリー(FACS)にて解析を行った。マーカーとしては、膵臓癌の幹細胞マーカーとして報告のあるCD44、CD24およびESA(CD326)の3重陽性、またはCD44陽性、c-Met強陽性の2重陽性を用いた。
アルクチゲニンと膵臓癌に対する既存の化学療法剤の一つであるシスプラチンの膵臓がん幹細胞に対する殺傷効果の比較を行った。

膵臓癌細胞Capan-1(ATCC No. HTB-79)をグルコース含有培養培地に播種して一晩インキュベートした後、グルコース含有培地、グルコース阻害培地、4μMアルクチゲニンを含むグルコース含有培地および4μMアルクチゲニンを含むグルコース阻害培地、または7μMシスプラチン(CDDP:和光純薬工業)のそれぞれにおいて24時間培養した。細胞を回収した後、定法にしたがってPI染色(死細胞染色)および癌幹細胞マーカー染色を行い、フローサイトメトリー(FACS)にて解析を行ったマーカーとしては、膵臓癌の幹細胞マーカーとして報告のあるCD44、CD24およびESA(CD326)の3重陽性を用いた。
Claims (7)
- アルクチゲニンを1日あたりの投与量が100mg以上となるように含有する、抗癌剤。
- さらにアルクチインを1日あたりの投与量が100mg以上となるように含有する、請求項1に記載の抗癌剤。
- アルクチゲニンおよびアルクチインを、アルクチゲニン/アルクチイン=0.7~1.3の重量比にて含有する、請求項2に記載の抗癌剤。
- 前記アルクチゲニンおよび前記アルクチインがゴボウシ由来である、請求項2または3に記載の抗癌剤。
- ゲムシタビンによる治療に不応答である膵臓癌を治療するための請求項1~4のいずれか1項に記載の抗癌剤。
- アルクチゲニンを含有する、抗癌幹細胞剤。
- アルクチゲニンを1日あたりの投与量が100mg以上となるように含有する、請求項6に記載の抗癌幹細胞剤。
