Description
本発明は、小麦アルブミンを含有する固形状組成物に関する。
近年、食生活の変化等により、肥満やII型糖尿病(高血糖症)に代表される糖代謝異常疾患に罹患する患者が著しく増加している。
通常、食後、特に糖質を含む食事を摂取した後は、血糖値が上昇することにより膵臓のβ細胞からインスリンが分泌される。インスリンは筋肉、肝臓、脂肪組織等に作用し、細胞内への糖の取り込みを促進することで、食後の急激な血糖値上昇を抑制する。しかし、インスリン感受性の低下(インスリン抵抗性)により、食後の高血糖状態が続くと、膵臓はインスリンを多量に分泌して血糖の上昇を抑えようとする。そして、このような状態が長く続くと膵臓が疲弊し、膵β細胞からのインスリンの分泌が低下し、最終的にインスリン作用機構が正常に機能しなくなり、II型糖尿病等になることが知られている。
通常、食後、特に糖質を含む食事を摂取した後は、血糖値が上昇することにより膵臓のβ細胞からインスリンが分泌される。インスリンは筋肉、肝臓、脂肪組織等に作用し、細胞内への糖の取り込みを促進することで、食後の急激な血糖値上昇を抑制する。しかし、インスリン感受性の低下(インスリン抵抗性)により、食後の高血糖状態が続くと、膵臓はインスリンを多量に分泌して血糖の上昇を抑えようとする。そして、このような状態が長く続くと膵臓が疲弊し、膵β細胞からのインスリンの分泌が低下し、最終的にインスリン作用機構が正常に機能しなくなり、II型糖尿病等になることが知られている。
インスリン抵抗性に伴う食後過血糖症状は、糖尿病でない健常人や糖尿病の境界型の方においても見られ、さらに、II型糖尿病以外にも肥満、高脂血症、動脈硬化等の原因や増悪因子となることが知られている。したがって、健康維持及びこれら諸症状・疾患の発症リスクの低下、予防の観点から、該食後過血糖症状を防止することは極めて重要である。
そこで近年、食後の急激な血糖上昇やインスリンの分泌を抑制できる物質の開発が多く行なわれている。その一つとして、アミラーゼ阻害物質があり、小麦由来のアミラーゼ阻害物質が糖尿病や肥満等の予防、治療に用いられている(非特許文献1)。

小麦の胚乳部には約10~15%のタンパク質が含まれ、タンパク質組成の約11%を占めるアルブミン(水可溶性タンパク質)は、α-アミラーゼ阻害活性を有し、食後血糖上昇抑制作用やインスリン抵抗性改善作用等の生理機能を有することが報告されている(非特許文献1、2)。なかでも、電気泳動の移動度が0.19の小麦アルブミンは、高いα-アミラーゼ阻害活性を有することから、多様な食品への応用が期待されている。
小麦アルブミンの生理機能を発現させるには、前記電気泳動の移動度が0.19の小麦アルブミン(以下0.19小麦アルブミンともいう)を、一食あたり125mg以上を一度に摂取するのが有効であると考えられ(非特許文献2)、これまでに、有効量の小麦アルブミンを配合した健康食品として、スープやハードカプセルが上市されている。また、特許文献1には、0.19小麦アルブミンを配合した錠剤についての開示がある。
小麦アルブミンの生理機能を発現させるには、前記電気泳動の移動度が0.19の小麦アルブミン(以下0.19小麦アルブミンともいう)を、一食あたり125mg以上を一度に摂取するのが有効であると考えられ(非特許文献2)、これまでに、有効量の小麦アルブミンを配合した健康食品として、スープやハードカプセルが上市されている。また、特許文献1には、0.19小麦アルブミンを配合した錠剤についての開示がある。

小麦アルブミンを無理なく長期間継続して摂取するには1回当たり少量で、手軽に摂取可能な形態である固形状組成物とすることが有利である。
しかしながら、本発明者らが検討したところ、一度少量摂取するだけで有効量を満たせるほどに高い濃度で小麦アルブミンを固形状組成物中に配合することは困難であることが判明した。すなわち、小麦アルブミンを高濃度化すると、当該小麦アルブミンに由来する異臭が感じられて喫食が困難であり、また、もどり臭が残ることが判明した。ここで、もどり臭とは、摂取時及び/又は摂取後の食道からのもどりの臭気である。
しかしながら、本発明者らが検討したところ、一度少量摂取するだけで有効量を満たせるほどに高い濃度で小麦アルブミンを固形状組成物中に配合することは困難であることが判明した。すなわち、小麦アルブミンを高濃度化すると、当該小麦アルブミンに由来する異臭が感じられて喫食が困難であり、また、もどり臭が残ることが判明した。ここで、もどり臭とは、摂取時及び/又は摂取後の食道からのもどりの臭気である。
したがって、本発明は、高濃度の小麦アルブミンを含みながらも、異臭やもどり臭が低減され、風味の良好な固形状組成物を提供することに関する。なお、前記特許文献1には、小麦アルブミンを含有する錠剤の異臭等の改善について何ら記載はない。
本発明者らは、上記課題を解決するため鋭意検討を重ねた結果、糖アルコール、とりわけ特定の糖アルコールを含有させることにより、小麦アルブミンを高濃度に含有するにもかかわらずその特有の異臭や、摂取時及び/又は摂取後のもどり臭が低減されて、良好な風味が感じられる固形状組成物とすることができることを見出した。
本発明によれば、小麦アルブミンを高濃度に含有しながらも、摂食時の小麦アルブミンに由来する異臭や、摂取時及び/又は摂取後のもどり臭が低減された、風味の良好な固形状組成物を提供することができる。
本発明の固形状組成物は、一度少量摂取するだけで、小麦アルブミンの生理効果発現に必要な量を摂取できるので、該小麦アルブミンによる効果を長期に亘って十分に期待できる。

本発明で用いる(A)小麦アルブミンは、小麦の胚乳部に由来するアルブミンファミリーに属する水可溶性タンパク質である。小麦アルブミンは、高いα-アミラーゼ阻害活性を有する点から、電気泳動の移動度が0.19の小麦アルブミンを含有することが好ましい。なお、ここでの電気泳動の移動度とは、試料をDavisの方法(Annals of the NewYork Academy of Science,121,404-427,1964)に従って、ポリアクリルアミドゲル電気泳動にかけた際の移動度をさす。
上記の小麦アルブミンは、小麦の胚乳部から抽出により得ることができる。小麦アルブミンの小麦からの抽出法としては、例えば、特開平9-172999号公報に記載のアミラーゼ阻害物質の調製法を用いることができる。
また、小麦アルブミンNA-1(日清ファルマ株式会社)等の市販品を用いてもよい。

本発明の固形状組成物中、(A)小麦アルブミンの含有量は、生理効果を有効に発現する摂取量の点、摂取形態として一度に少量の摂取で可能であるという点から、10質量%(以下、単に「%」とする)以上、更に20%以上、更に30%以上が好ましく、また、小麦アルブミン特有の異臭及びもどり臭を低減する点から、70%以下、更に60%以下、更に55%以下、更に50%以下であるのが好ましい。具体的には10~70%、更に20~70%、更に30~60%、更に30~55%、更に30~50%であるのが好ましい。

本発明の固形状組成物においては、(A)小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(A)]を0.2以上とすることが重要である。(A)小麦アルブミン1に対し、(B)糖アルコールの割合を0.2以上とすることで、小麦アルブミン特有の異臭やもどり臭を低減できる。
(A)小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(A)]は、上記と同様の点から、0.2以上、更に0.4以上、更に0.5以上、更に0.7以上が好ましく、また、生理効果を有効に発現する小麦アルブミンを含有させる点から、10以下、更に7以下、更に5以下、更に2以下、更に1.9以下であるのが好ましい。具体的には0.2~10、更に0.4~7、更に0.4~5、更に0.5~2、更に0.7~1.9であるのが好ましい。
(A)小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(A)]は、上記と同様の点から、0.2以上、更に0.4以上、更に0.5以上、更に0.7以上が好ましく、また、生理効果を有効に発現する小麦アルブミンを含有させる点から、10以下、更に7以下、更に5以下、更に2以下、更に1.9以下であるのが好ましい。具体的には0.2~10、更に0.4~7、更に0.4~5、更に0.5~2、更に0.7~1.9であるのが好ましい。
また本発明の固形状組成物中、(a)0.19小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(a)]は0.8以上が好ましい。(a)0.19小麦アルブミン1に対し、(B)糖アルコールの割合を0.8以上とすることで、小麦アルブミン特有の異臭やもどり臭を低減できる。
(a)0.19小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(a)]は、上記と同様の点から、0.8以上、更に1.5以上、更に2.5以上が好ましく、また、生理効果を有効に発現する0.19小麦アルブミンを含有させる点から、10以下、更に7.5以下であるのが好ましい。具体的には0.8~10、更に1.5~10、更に1.5~7.5、更に2.5~7.5であるのが好ましい。
(a)0.19小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(a)]は、上記と同様の点から、0.8以上、更に1.5以上、更に2.5以上が好ましく、また、生理効果を有効に発現する0.19小麦アルブミンを含有させる点から、10以下、更に7.5以下であるのが好ましい。具体的には0.8~10、更に1.5~10、更に1.5~7.5、更に2.5~7.5であるのが好ましい。
本発明の固形状組成物は、更に(C)アスパラギン酸又はその塩を含有するのが、小麦アルブミンに由来する異臭を低減すると共に、小麦アルブミンに由来する特有の異味を低減する点から好ましい。
本発明で用いるアスパラギン酸の塩としては、塩酸、臭化水素酸、硫酸、リン酸、硝酸等の無機酸との酸付加塩;酢酸、プロピオン酸、コハク酸、グリコール酸、乳酸、リンゴ酸、酒石酸、クエン酸、マレイン酸、フマル酸、メタンスルホン酸、ベンゼンスルホン酸、p-トルエンスルホン酸、アスコルビン酸等の有機酸との酸付加塩;ナトリウム、カリウム等のアルカリ金属、カルシウム等のアルカリ土類金属、アンモニウム等の無機塩基との塩;メチルアミン、ジエチルアミン、トリエチルアミン、エチレンジアミン等のアミン、モノエタノールアミン、ジエタノールアミン、トリエタノールアミン等のアルカノールアミン等の有機塩基との塩が挙げられる。なかでも、アルカリ金属塩が好ましい。
本発明の固形状組成物中、(C)アスパラギン酸又はその塩の含有量は、小麦アルブミンの異味・異臭の低減の点から、アスパラギン酸換算で、0.005%以上、更に0.008%以上、更に0.01%以上、更に0.012%以上が好ましく、また、風味のバランスの点から1%以下、更に0.8%以下、更に0.5%以下、更に0.1%以下、更に0.05%以下、更に0.025%以下、更に0.02%以下であるのが好ましい。具体的には0.005%~1%、更に0.008~0.8%、更に0.01~0.5%、更に0.01~0.1%、更に0.01~0.05%、更に0.01~0.02%であるのが好ましい。
本発明における(C)アスパラギン酸又はその塩の含有量には、配合されたアスパラギン酸又はその塩以外にも小麦アルブミン等の他の原料由来のものが含まれる。

本発明の固形状組成物においては、固形状組成物中の(a)0.19小麦アルブミンの含有量と固形状組成物中の(c)アスパラギン酸又はその塩のアスパラギン酸換算量の質量比[(a)/(c)]は、小麦アルブミン特有の異味・異臭を低減する点から1000以下、更に910以下、更に900以下、更に850以下、更に780以下、更に750以下であるのが好ましく、また、アスパラギン酸又はその塩の味を抑え、風味のバランスが良好となる点から10以上、更に15以上、更に50以上、更に100以上、更に200以上、更に400以上、更に500以上であるのが好ましい。具体的には、(a)0.19小麦アルブミンの含有量と(c)アスパラギン酸又はその塩のアスパラギン酸換算量の質量比[(a)/(c)]は、10~1000、更に15~910、更に50~900、更に100~850、更に200~780、更に500~750が好ましい。
また、本発明の固形状組成物は、更に(D)炭酸塩と(E)有機酸を含有するのが好ましい。小麦アルブミンに炭酸塩と有機酸とを組み合わせ、炭酸ガスを発生させることにより、小麦アルブミンを高濃度に含有するにもかかわらず、口内でのねとつき・付着が抑制され、また小麦アルブミンの特有の異味が低減されて、食感及び風味の良好な固形状組成物とすることができる。
本発明で用いる(D)炭酸塩としては、例えば、炭酸ナトリウム、炭酸水素ナトリウム、炭酸カリウム、炭酸水素カリウム、炭酸カルシウム、炭酸マグネシウム、セスキ炭酸ナトリウム等が挙げられ、これらは単独で又は2種以上を組み合わせて用いることができる。
本発明で用いる(D)炭酸塩としては、例えば、炭酸ナトリウム、炭酸水素ナトリウム、炭酸カリウム、炭酸水素カリウム、炭酸カルシウム、炭酸マグネシウム、セスキ炭酸ナトリウム等が挙げられ、これらは単独で又は2種以上を組み合わせて用いることができる。
本発明の固形状組成物中、(D)炭酸塩の含有量は、物性の点から2%以上、更に3%以上、更に10%以上が好ましく、また、風味の点から20%以下、更に19.5%以下、更に14%以下であるのが好ましい。具体的には2~20%、更に3~19.5%、更に10~14%であるのが好ましい。
また、本発明で用いる(E)有機酸としては、可食性の酸を使用することができる。例えば、クエン酸、リン酸、コハク酸、アスコルビン酸、酢酸、グルコン酸、リンゴ酸、酒石酸、フマル酸、アジピン酸等の有機酸が挙げられ、これらは単独で又は2種以上を組み合わせて用いることができる。なかでも、摂食時の口内でのねとつき・付着が少ない点、発生する泡の食感が良好な点から、クエン酸又はリンゴ酸が好ましく、更にクエン酸が好ましい。
本発明の固形状組成物中、(E)有機酸の含有量は、物性の点から2%以上、更に2.5%以上、更に3%以上、更に8%以上が好ましく、また、風味の点から18%以下、更に15%以下、更に12%以下、更に11%以下であるのが好ましい。具体的には2~18%、更に2.5~15%、更に3~12%、更に8~11%であるのが好ましい。
本発明の固形状組成物においては、(A)小麦アルブミンと(D)炭酸塩の含有質量比[(A)/(D)]は、摂食時の口内でのねとつき・付着性を抑制でき、また、小麦アルブミン特有の異味を低減できる点から1.5以上、更に2.5以上、更に2.6以上、更に3.5以上が好ましく、また、風味の点から16.5以下、更に15.5以下、更に12以下、更に5以下であるのが好ましい。具体的には1.5~16.5、更に2.5~15.5、更に2.6~12、更に3.5~5であるのが好ましい。
(a)0.19小麦アルブミンと(D)炭酸塩の含有質量比[(a)/(D)]は、摂食時の口中のねとつき・付着性を抑制でき、また、小麦アルブミン特有の異味を低減できる点から、0.2以上、更に0.3以上、更に0.35以上が好ましく、また4.1以下、更に3.8以下、更に3以下であるのが好ましい。具体的には0.2~4.1、更に0.3~3.8、更に0.35~3であるのが好ましい。
(a)0.19小麦アルブミンと(D)炭酸塩の含有質量比[(a)/(D)]は、摂食時の口中のねとつき・付着性を抑制でき、また、小麦アルブミン特有の異味を低減できる点から、0.2以上、更に0.3以上、更に0.35以上が好ましく、また4.1以下、更に3.8以下、更に3以下であるのが好ましい。具体的には0.2~4.1、更に0.3~3.8、更に0.35~3であるのが好ましい。
更に、本発明の固形状組成物においては、(E)有機酸と(D)炭酸塩の当量比は、炭酸塩由来のえぐみや有機酸の酸味が突出せず、風味のバランスが良好となる点から0.7以上、更に0.8以上、更に0.85以上、更に0.9以上が好ましく、また1.9以下、更に1.8以下、更に1.2以下、更に1.1以下であるのが好ましい。具体的には0.8~1.8、更に0.85~1.2、更に0.9~1.1であるのが好ましい。
尚、本発明において、前記「当量比」とは、固形状組成物に含まれる(E)有機酸の当量を(D)炭酸塩の当量で除した値である。

本発明の固形状組成物には、上記成分の他に、本発明の効果を損なわない範囲において、ミネラル(例えば、カルシウム、マグネシウム、鉄、亜鉛、クロム、セレン、マンガン、モリブデン、銅、ヨウ素、リン、カリウム、ナトリウム)、ビタミン(例えば、ビタミンA、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンB12、ビタミンC、ビタミンE、葉酸及びそれらの塩、又はそれらのエステル)、甘味料(例えば、フルクトース、グルコース、ガラクトース、キシロース、タガトース等の単糖、ショ糖、乳糖、麦芽糖、トレハロース、イソマルトオリゴ糖、ガラクトオリゴ糖、フラクトオリゴ糖、乳果オリゴ糖、大豆オリゴ糖、イソマルツロース、カップリングシュガー等の少糖、炭素数6~12以外の糖アルコール、サッカリン、スクラロース、アセスルファムカリウム等の合成甘味料)、酸味料(例えば、クエン酸、リンゴ酸、酒石酸、乳酸、コハク酸、アジピン酸、グルコノデルタラクトン、グルコン酸、酢酸、フマル酸)、香料、着色料、保存料等が適宜配合されていてもよい。
本発明の固形状組成物の形態としては、例えば、室温(15~25℃)で固形状のものであれば特に限定されないが、例えば、カプセル剤、顆粒剤、散剤、錠剤、丸剤等が挙げられる。なかでも、1回あたり少量で摂取可能な点から、錠剤が好ましく、更に摂取が簡便な点から、チュアブル錠が好ましい。
本発明の固形状組成物が(D)炭酸塩と(E)有機酸を含有する場合、当該固形状組成物は口内又は水の存在下で炭酸ガスを発生するものである。

このような剤型の組成物を調製するには、必要に応じて、乳糖、デンプン類、結晶セルロース、蔗糖、軽質無水ケイ酸、リン酸水素カルシウム等の賦形剤;ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルセルロース、ゼラチン、アルファー化デンプン、ポリビニルピロリドン、ポリビニルアルコール、プルランメチルセルロース、硬化油等の結合剤;カルメロース、カルメロースカルシウム、クロスカルメロースナトリウム、クロスポピドン、トウモロコシデンプン、低置換度ヒドロキシプロピルセルロース等の崩壊剤;ステアリン酸カルシウム、ステアリン酸マグネシウム、フマル酸ステアリルナトリウム、タルク、二酸化ケイ素等の滑沢剤;ステビア、アスパルテーム等の嬌味剤;香料、増量剤、界面活性剤、分散剤、緩衝剤、保存剤、被膜剤、希釈剤等の担体を適宜組み合わせて用いることができる。
本発明の固形状組成物は、特に制限はなく常法に従い製造される。例えば、(A)小麦アルブミン、(B)炭素数6~12の糖アルコール及び必要に応じて添加される添加剤の混合物を調製後、圧縮成形することによって製造することができる。
例えば、錠剤を製造する場合、直接圧縮して成形(直接粉末圧縮法)しても、乾式造粒法、湿式造粒法等を用いて造粒してから圧縮して成形(顆粒圧縮法)しても良い。なかでも、工程の簡便性の点から、直接粉末圧縮法を用いて錠剤とするのが好ましい。
直接圧縮して成形して錠剤を製造する場合、打錠成形機としてはロータリー式打錠機や単発式打錠機等通常使用されるものを用いることができる。
また、造粒法より造粒してから錠剤とする場合、円筒造粒機、球形整粒機、ペレッター等を使用する押し出し造粒法;スピードミル、パワーミル等を使用する破砕造粒法;転動造粒法、攪拌造粒法、流動層造粒法等により造粒物を製造し、乾燥・整粒した後、得られた造粒物を前記打錠成形機で圧縮して錠剤を形成できる。造粒物の粒子径は、45μm~850μmとするのが好ましく、100μm~500μmとするのが更に好ましい。
錠剤の形状としては、円形錠もしくは楕円形、長円形、四角形等の面形を有する各種異形錠であってもよい。
また、打錠時の圧縮成型圧は、成形物の硬度維持、崩壊性等の点から、100~3000kg/cm2程度である。
例えば、錠剤を製造する場合、直接圧縮して成形(直接粉末圧縮法)しても、乾式造粒法、湿式造粒法等を用いて造粒してから圧縮して成形(顆粒圧縮法)しても良い。なかでも、工程の簡便性の点から、直接粉末圧縮法を用いて錠剤とするのが好ましい。
直接圧縮して成形して錠剤を製造する場合、打錠成形機としてはロータリー式打錠機や単発式打錠機等通常使用されるものを用いることができる。
また、造粒法より造粒してから錠剤とする場合、円筒造粒機、球形整粒機、ペレッター等を使用する押し出し造粒法;スピードミル、パワーミル等を使用する破砕造粒法;転動造粒法、攪拌造粒法、流動層造粒法等により造粒物を製造し、乾燥・整粒した後、得られた造粒物を前記打錠成形機で圧縮して錠剤を形成できる。造粒物の粒子径は、45μm~850μmとするのが好ましく、100μm~500μmとするのが更に好ましい。

また、本発明の錠剤の1錠当りの重量は0.1~2g、更に1~2gとするのが簡便性及び有効性の点で好ましい。
後記実施例に示すとおり、炭素数6~12の糖アルコールにより小麦アルブミンの異臭ともどり臭がマスキングされたことから、炭素数6~12の糖アルコールは、小麦アルブミンのマスキング剤として有用であり、小麦アルブミンをマスキングするために使用できる。糖アルコールのなかでもマルチトール、ソルビトール及びラクチトールから選ばれる少なくとも1種がより好ましく、更にマルチトール、ソルビトール、又はこれらの組み合わせが好ましく、更にマルチトールが好ましい。
上述した実施形態に関し、本発明はさらに以下の組成物、方法、或いは用途を開示する。

<8>固形状組成物中の(B)炭素数6~12の糖アルコールの含有量が、10質量%以上、好ましくは20質量%以上、より好ましくは30質量%以上、更に好ましくは35質量%以上であり、また90質量%以下、好ましくは65質量%以下、より好ましくは60質量%以下、更に好ましくは55質量%以下である前記<1>~<7>のいずれか1に記載の固形状組成物。
<9>(a)0.19小麦アルブミンと(B)炭素数6~12の糖アルコールの含有質量比[(B)/(a)]が、0.8以上であって、好ましくは1.5以上、より好ましくは2.5以上であり、また10以下、好ましくは7.5以下である前記<1>~<8>のいずれか1に記載の固形状組成物。
<10>更に(C)アスパラギン酸又はその塩を含有する前記<1>~<9>のいずれか1に記載の固形状組成物。
<11>固形状組成物中の(C)アスパラギン酸又はその塩の含有量が、アスパラギン酸換算で、0.005質量%以上、好ましくは0.008質量%以上、より好ましくは0.01質量%以上、更に好ましくは0.012質量%以上であり、また1質量%以下、好ましくは0.8質量%以下、より好ましくは0.5質量%以下、より好ましくは0.1質量%以下、更に好ましくは0.05質量%以下、更に好ましくは0.025質量%以下、更に好ましくは0.02質量%以下である前記<10>に記載の固形状組成物。
<12>固形状組成物中の(a)0.19小麦アルブミンの含有量と固形状組成物中の(c)アスパラギン酸又はその塩のアスパラギン酸換算量の質量比[(a)/(c)]が、1000以下、好ましくは910以下、より好ましくは、900以下、より好ましくは850以下、より好ましくは780以下、更に好ましくは750以下であり、また10以上、好ましくは15以上、より好ましくは50以上、より好ましくは100以上、より好ましくは200以上、更に好ましくは400以上、更に好ましくは500以上である前記<10>又は<11>に記載の固形状組成物。
<13>更に(D)炭酸塩と(E)有機酸を含有する前記<1>~<12>のいずれか1に記載の固形状組成物。
<14>(D)炭酸塩が、炭酸ナトリウム、炭酸水素ナトリウム、炭酸カリウム、炭酸水素カリウム、炭酸カルシウム、炭酸マグネシウム及びセスキ炭酸ナトリウムから選ばれる少なくとも1種である前記<13>に記載の固形状組成物。
<15>(E)有機酸が、クエン酸、リン酸、コハク酸、アスコルビン酸、酢酸、グルコン酸、リンゴ酸、酒石酸、フマル酸及びアジピン酸から選ばれる少なくとも1種であり、好ましくはクエン酸、リンゴ酸又はこれらの組み合わせであり、より好ましくはクエン酸である前記<13>又<14>に記載の固形状組成物。
<16>固形状組成物中の(D)炭酸塩の含有量が、2質量%以上、好ましくは3質量%以上、より好ましくは10質量%以上であり、また20質量%以下、好ましくは19.5質量%以下、より好ましくは14質量%以下である前記<13>~<15>のいずれか1に記載の固形状組成物。
<17>固形状組成物中の(E)有機酸の含有量が、2質量%以上、好ましくは2.5質量%以上、より好ましくは3質量%以上、更に好ましくは8質量%以上であり、また18質量%以下、好ましくは15質量%以下、より好ましくは12質量%以下、更に好ましくは11質量%以下である前記<13>~<16>のいずれか1に記載の固形状組成物。
<18>固形状組成物中の(A)小麦アルブミンと(D)炭酸塩の含有質量比[(A)/(D)]が、1.5以上、好ましくは2.5以上、より好ましくは2.6以上、更に好ましくは3.5以上であり、また16.5以下、好ましくは15.5以下、より好ましくは12以下、更に好ましくは5以下である前記<13>~<17>のいずれか1に記載の固形状組成物。
<19>固形状組成物中の(a)0.19小麦アルブミンと(D)炭酸塩の含有質量比[(a)/(D)]が、0.2以上、好ましくは0.3以上、より好ましくは0.35以上であり、また4.1以下、好ましくは3.8以下、より好ましくは3以下である前記<13>~<18>のいずれか1に記載の固形状組成物。
<20>(E)有機酸と(D)炭酸塩の当量比が、0.7以上、好ましくは0.8以上、より好ましくは0.85以上、更に好ましくは0.9以上であり、また1.9以下、好ましくは1.8以下、より好ましくは1.2以下、更に好ましくは1.1以下である前記<13>~<19>のいずれか1に記載の固形状組成物。
<21>チュアブル錠である前記<1>~<20>のいずれか1に記載の固形状組成物。
<22>炭素数6~12の糖アルコールを有効成分とする小麦アルブミンのマスキング剤。
<23>炭素数6~12の糖アルコールを小麦アルブミンのマスキング剤として使用する方法。
<24>炭素数6~12の糖アルコールの、小麦アルブミンのマスキングのための使用。

[原料]
小麦アルブミン:小麦アルブミンNA-1、日清ファルマ株式会社製 (0.19小麦アルブミン含有量25%)
小麦アルブミン:小麦アルブミンNA-1、日清ファルマ株式会社製 (0.19小麦アルブミン含有量25%)
[アスパラギン酸の分析]
固形状組成物50mgを蒸留水1gに溶解し、遠心分離装置により3000rpm、25℃、10分間の条件で遠心分離した。その上清500μLに5%トリクロロ酢酸500μLを加えた後、遠心分離装置により10000r/min、5℃、10分間の条件で遠心分離した。その上清500μLを0.02N塩酸で2~10倍に希釈したのち、0.2μmフィルターでろ過しサンプルとした。サンプルをアミノ酸分析計(日立L-8800)により測定した。
固形状組成物50mgを蒸留水1gに溶解し、遠心分離装置により3000rpm、25℃、10分間の条件で遠心分離した。その上清500μLに5%トリクロロ酢酸500μLを加えた後、遠心分離装置により10000r/min、5℃、10分間の条件で遠心分離した。その上清500μLを0.02N塩酸で2~10倍に希釈したのち、0.2μmフィルターでろ過しサンプルとした。サンプルをアミノ酸分析計(日立L-8800)により測定した。
[炭酸塩の分析]
固形状組成物中の炭酸塩の含有量の分析方法は以下の通りである。
固形状組成物を0.1~0.2g採取し、水10mLと50%りん酸2mLを加え密栓する。10分間超音波処理を行った後、1時間放置しヘッドスペースガスをガスクロマトグラフに供してCO2量を求め、発生したCO2量から算出する。

機種:GC-14B[島津製作所]
検出器:TCD
カラム:Chromosorb101,80~100mesh
ガラス管,φ3.2mm×2m
温度:カラム50℃,注入口及び検出器100℃
セル電流75mA
ガス圧力:ヘリウム(キャリヤーガス)100kPa
注入量:ヘッドスペースガス0.2mL
固形状組成物中の炭酸塩の含有量の分析方法は以下の通りである。
固形状組成物を0.1~0.2g採取し、水10mLと50%りん酸2mLを加え密栓する。10分間超音波処理を行った後、1時間放置しヘッドスペースガスをガスクロマトグラフに供してCO2量を求め、発生したCO2量から算出する。

検出器:TCD
カラム:Chromosorb101,80~100mesh
ガラス管,φ3.2mm×2m
温度:カラム50℃,注入口及び検出器100℃

[有機酸の分析]
固形状組成物中の有機酸の含有量の分析方法は以下の通りである。
固形状組成物を1g採取し5%過塩素酸20mLを加え、10分間振とうすることで抽出する。これを水で200mLに定容し10分間超音波処理を行なう。ろ過後高速液体クロマトグラフに供する。
<高速液体クロマトグラフ操作条件>
機種:LC-20AD[株式会社島津製作所]
検出器:紫外可視吸光光度計SPD-20AV[島津製作所]
カラム温度:40℃
移動相:3mmоl/L過塩素酸
反応液:0.2mmоl/Lブロムチモールブルー含有
15mmоl/Lりん酸水素二ナトリウム溶液
流量:移動相1.0mL/min、反応液1.4mL/min
測定波長:445nm
固形状組成物中の有機酸の含有量の分析方法は以下の通りである。

〔チュアブル錠の調製〕
実施例1~実施例13及び比較例1~比較例4
表1に記載の配合組成で各原料成分を混合した。次に単発式打錠機(RIKEN社製)を用いて、穴径13mmのリング状杵で、錠剤重量1000mgで打錠し、チュアブル錠を得た。チュアブル錠中の(a)0.19小麦アルブミン含有量は表1のとおりであった。
実施例1~実施例13及び比較例1~比較例4
表1に記載の配合組成で各原料成分を混合した。次に単発式打錠機(RIKEN社製)を用いて、穴径13mmのリング状杵で、錠剤重量1000mgで打錠し、チュアブル錠を得た。チュアブル錠中の(a)0.19小麦アルブミン含有量は表1のとおりであった。
上記で得た本発明品と比較品について官能評価を行なった。評価は、専門パネル3名で、摂食時の小麦アルブミン特有の異臭と、摂取時及び/又は摂取後に感じる食道からのもどり臭について、下記に示す判断基準に従って行い、その平均値をもって評点とした。結果を表1に示す。

5:異臭を感じない
4:異臭を殆ど感じない
3:異臭を僅かに感じる
2:異臭を強く感じる
1:異臭を非常に強く感じる
〔もどり臭〕
5:もどり臭を感じない

1:もどり臭を非常に強く感じる
〔小麦アルブミン特有の異臭〕
5:異臭を感じない

表1から明らかなように、本発明品は比較品と比べ、小麦アルブミン特有の異臭及びもどり臭の低減が確認された。また、炭素数6~12の糖アルコールを配合しなかった比較例4やその配合割合が少ない比較例1及び3、スクロースを配合した比較例2では、小麦アルブミン特有の異臭が強く、もどり臭が強かった。
〔チュアブル錠の調製〕
実施例14~実施例21
表2に記載の配合組成で各原料成分を混合した以外は実施例1と同様にチュアブル錠を得た。チュアブル錠中の(a)0.19小麦アルブミン含有量は表2のとおりであった。

上記で得た本発明品について官能評価を行なった。評価は、専門パネル3名で、上記に示す判断基準に従って「摂食時の小麦アルブミン特有の異臭」と、「摂取時及び/又は摂取後に感じる食道からのもどり臭」について行った。また、「小麦アルブミン特有の異味」、「添加したアミノ酸に由来する味」、「摂食時の口内付着性」、「泡の食感」、「風味のバランス」について下記に示す判断基準に従って評価した。いずれも平均値をもって評点とした。結果を表2に示す。

表2から明らかなように、更にアスパラギン酸塩を配合した実施例15~17及び実施例21は、小麦アルブミン特有の異味の低減も確認され、添加アミノ酸に由来する味も感じられず、良好な風味であった。また、更に炭酸塩及び有機酸を配合した実施例18~21は、摂食中の口内のねとつき・付着性も抑えられ、また、泡の食感が良く、風味のバランスも良好であった。
Claims (11)
- 次の成分(A)及び(B):
(A)小麦アルブミン、
(B)炭素数6~12の糖アルコール、
を含有し、成分(A)と成分(B)の含有質量比[(B)/(A)]が0.2以上である固形状組成物。 - (B)炭素数6~12の糖アルコールが、マルチトール、ソルビトール及びラクチトールから選ばれる少なくとも1種である請求項1記載の固形状組成物。
- (A)小麦アルブミンが、(a)0.19小麦アルブミンを10質量%以上60質量%以下含有する請求項1又は2記載の固形状組成物。
- 更に(C)アスパラギン酸又はその塩を含有し、固形状組成物中の(a)0.19小麦アルブミンの含有量と(c)アスパラギン酸又はその塩のアスパラギン酸換算量の質量比[(a)/(c)]が10以上1000以下である請求項1~3のいずれか1項記載の固形状組成物。
- 更に(D)炭酸塩と(E)有機酸を含有し、固形状組成物中の(A)小麦アルブミンと(D)炭酸塩の含有質量比[(A)/(D)]が1.5以上16.5以下であり、且つ(E)有機酸と(D)炭酸塩の当量比が0.7以上1.9以下である請求項1~4のいずれか1項記載の固形状組成物。
- チュアブル錠である請求項1~5のいずれか1項記載の固形状組成物。
- 次の成分(a)及び(B):
(a)0.19小麦アルブミン、
(B)炭素数6~12の糖アルコール、
を含有し、成分(a)と成分(B)の含有質量比[(B)/(a)]が0.8以上である固形状組成物。 - 炭素数6~12の糖アルコールを有効成分とする小麦アルブミンのマスキング剤。
- 炭素数6~12の糖アルコールを小麦アルブミンのマスキング剤として使用する方法。
- 小麦アルブミンのマスキング剤としての炭素数6~12の糖アルコールの使用。
- 小麦アルブミンのマスキング剤の製造のための炭素数6~12の糖アルコールの使用。
