Abstract
본 발명은 양극성 장애 치료제의 스크리닝 방법에 관한 것으로서, 보다 구체적으로, 본 발명은 시료에 양극성 장애 치료제 후보 물질을 투여하고, 시료의 이온 채널의 활성을 측정한 다음, 상기 후보 물질이 TREK1 채널만을 활성화시키는 경우, 양극성 장애 치료제로 판정하는 단계를 포함하는 양극성 장애 치료제의 스크리닝 방법에 관한 것이다. 본 발명의 스크리닝 방법을 이용하면, 양극성 장애에 대한 치료효과를 나타낼 수 있는 물질을 용이하게 검출할 수 있으므로, 양극성 장애의 치료 또는 예방에 널리 활용될 수 있을 것이다.
Description
본 발명은 양극성 장애 치료제의 스크리닝 방법에 관한 것으로서, 보다 구체적으로, 본 발명은 시료에 양극성 장애 치료제 후보 물질을 투여하고, 시료의 이온 채널의 활성을 측정한 다음, 상기 후보 물질이 TREK1 채널만을 활성화시키는 경우, 양극성 장애 치료제로 판정하는 단계를 포함하는 양극성 장애 치료제의 스크리닝 방법에 관한 것이다.
조울병으로도 알려진 양극성 스펙트럼 장애는 우울증 증상이 적어도 하나의 조증, 경조증 또는 혼합 삽화(episode)와 조합된 기분 장애이다. 조증 삽화는 비정상적이면서 지속적으로 매우 기분이 좋거나, 팽창된 또는 짜증스러운 기분이라는 뚜렷한 기간을 특징으로 한다. 혼합 삽화는 조증 및 주요 우울 삽화의 기준 모두가 충족되는 적어도 한 주의 지속 기간을 특징으로 한다. 조증 삽화와 유사하게, 경조증 삽화는 비정상적이고도 지속적으로 매우 기분이 좋거나, 팽창된 또는 짜증스러운 기분이 존재하는 뚜렷한 기간을 특징으로 한다.
그러나, 조증 삽화와는 반대로, 경조증 삽화는 사회적 또는 직업적 기능에 현저히 손상을 일으키거나 입원을 요할 만큼 심각하지는 않으며, 어떠한 정신병적 특징도 없다. 양극성 우울증 삽화의 증상은 주요 우울증 삽화를 특징으로 하는 증상과 상이하지 않다. 이는 또한 많은 양극성 환자가 초기에 주요 우울증을 겪는 것으로 진단되는 이유이다. 앞서 언급한 바와 같이, 조증 또는 혼합 또는 경조증 삽화의 발생으로 양극성 진단을 내리게 하나, 주요 우울증 진단과는 별개이다.
양극성 스펙트럼 장애는, 제Ⅰ형 양극성 장애, 제Ⅱ형 양극성 장애, 순환성 장애 및 달리 명시하지 않은 양극성 장애로 세분화될 수 있다. 제Ⅰ형 양극성 장애는 하나 이상의 조증 또는 혼합 삽화 발생을 특징으로 하는데, 종종 개체들은 하나 이상의 주요 우울증 삽화를 지닌다. 제Ⅱ형 양극성 장애는 적어도 하나의 경조증 삽화를 수반하는 하나 이상의 주요 우울증 삽화 발생을 특징으로 한다. 제Ⅰ형 양극성 장애 및 제Ⅱ형 양극성 장애의 진행성으로 인해, 환자들은 매번 새로운 삽화와 함께 증상의 재발이라는 위험이 증가되고, 더불어 아직 치료를 받지 않았다면, 후속 삽화의 지속 기간 및 중증도가 증가하여 점점 커지는 위험을 겪는다. 이러한 이유로, 제Ⅰ형 양극성 장애 및 제Ⅱ형 양극성 장애 환자 모두는 결국 1 년에 적어도 4 가지의 삽화를 겪는 급속 순환성 환자로 분류될 수 있다. 순환성 장애는 양극성 장애의 하부 그룹으로, 여기서 기분 교란은 만성의 변동적인 기분 교란을 특징으로 하며, 이는 경조증의 오랜 기간 및 우울증 증상 기간을 포함한다. 달리 명시하지 않은 양극성 장애는 상기에서 언급된 임의의 특정 양극성 장애의 기준을 충족하지 않는 양극성 특징이 있는 장애 카테고리로 지칭된다.
양극성 스펙트럼 장애는 생명을 위협하는 병태인데, 양극성 장애로 진단받은 환자들이 일반 집단보다 15 배나 더 높은 자살 위험성을 가지는 것으로 추정되기 때문이다. 현재, 환자가 각각 조증 또는 우울증 삽화로 재발하는 경우에는, 양극성 환자를 기분 안정제(주로 리튬 또는 항간질약)로 유지시키고, 항조증제(리튬 또는 항정신병제) 또는 항우울증제(삼환식 항우울증제 또는 선택적 세로토닌 재흡수 억제제)를 추가함으로써 양극성 스펙트럼 장애를 치료하고 있다. 따라서, 오직 하나의 치료제로 이들 장애의 세가지 중요한 요소 모두를 효과적으로 치료하고자하는 요구에 부합하기 위해 양극성 스펙트럼 장애용 신규한 치료법을 개발시키는 것이 필요하다. 이러한 신규 제제는 신속히 작용이 개시되어 조증 증상을 완화시키고(항조증 활성), 신속히 작용이 개시되어 우울증 증상을 완화시키며(항우울증 활성), 우울증 증상뿐 아니라 조증 증상의 재발을 방지해야 한다(기분 안정화 활성).
최근 10여년 간 분자생물학 기법의 발전으로 인해 상당수의 포타슘(potassium, kalium; 칼륨) 통로 구조가 밝혀졌고, 현재에는 3차원 구조까지도 밝혀지고 있다. 이들 포타슘 통로는 포타슘에 선택적인 투과성을 보이는 이온통로를 형성하는 소수성의 서브유닛으로 구성되어 있으며, 경우에 따라 필수 서브유닛(accessory subunits)이 같이 결합되어 있기도 한 단백질 복합체이다. 현재까지 포유동물에서만 60가지 이상의 기공-형성 서브유닛(pore-forming subunits)이 알려져 있으며, 포타슘 통로는 막위상(membrane topology)에 의해 크게 3종류로 구분된다. 첫 번째는 6개의 막전위 절편(transmembrane segments; TM)과 하나의 기공-도메인을 가지고 있는 그룹이며, 두 번째는 2개의 TM과 하나의 기공-도메인을 가지고 있는 그룹이다. 또 다른 하나는 4개의 TM과 2개의 기공-도메인(4TM/2P)을 가지고 있다.
2-기공 도메인 K+(K2P) 채널 패밀리의 멤버인 TREK1 및 TREK2는 항우울제(fluoxetine, norfluoxetine, 및 paroxetine) 및 항정신병제(fluphenazine, chlorpromazine, haloperidol, flupenthixol, loxapine, 및 pimozide)에 의해 농도-의존적 및 가역적으로 저해된다. 따라서, 우울증의 병리생리학과 관련된 이온 채널인 TREK1 채널은 우울증 치료를 위한 잠재적인 표적으로써 제시되어져 왔으며, 다양한 세포에서 TREK1 및 TREK2는 생물리학적 및 약리학적 특성이 유사하다고 알려져 있다. 그러나, TREK1에 비해 TREK2에 관한 연구는 미흡한 상황이며, TREK 채널과 기분 안정제의 연관성에 대해서는 보고된 바가 없다.
본 발명자들은 양극성 장애와 TREKs 간의 연관성을 확인하기 위하여 예의 노력한 결과, 양극성 장애를 치료하기 위하여 통상적으로 사용되는 약물인 lithium chloride(LiCl), valproate, 및 carbamazepine과 같은 기분 안정제를 TREKs이 형질감염된 세포에 처리하였을 때, TREK1 채널은 조절되고, TREK2 채널은 조절되지 않는다는 사실을 확인하였으며, TREK1 채널이 우울증 뿐만 아니라, 양극성 장애의 치료를 위한 잠재적인 치료적 표적으로써 작용할 수 있음을 확인함으로써 본 발명을 완성하였다.
본 발명의 주된 목적은 (a) 시료에 양극성 장애 치료제 후보 물질을 투여하는 단계; (b) 상기 시료에서 TREK1 채널 및 TREK2 채널의 활성을 검출하는 단계; 및 (c) TREK1 채널만을 활성화시키는 경우, 상기 후보물질이 양극성 장애 치료활성을 나타내는 것으로 판정하는 단계를 포함하는 양극성 장애 치료제의 스크리닝 방법을 제공하는 것이다.
본 발명의 스크리닝 방법을 이용하면, 양극성 장애에 대한 치료효과를 나타낼 수 있는 물질을 용이하게 검출할 수 있으므로, 양극성 장애의 치료 또는 예방에 널리 활용될 수 있을 것이다.
도 1a 및 1b는 TREK1 및 TREK2 전류에 영향을 미치는 항우울제 및 항정신병제의 효과를 나타낸 그래프이다. 도 1a는 fluoxetine, paroxetine 및 chlorpromazine에 의한 TREK1 및 TREK2 전류의 저해를 나타내는 그래프이며, 도 1b의 A는 TREK1 및 TREK2 전류에 영향을 미치는 fluoxetine(TREK1, n=10; TREK2, n=6), paroxetine(TREK1, n=10; TREK2, n=7), 및 chlorpromazine(TREK1, n=4; TREK2, n=16)의 효과, B는 TREK1(n=8) 및 TREK2(n=4) 전류에 영향을 미치는 fluoxetine의 농도-의존적 효과를 나타내는 그래프이다.

상기 목적을 달성하기 위한 하나의 양태로서, 본 발명은 시료에 양극성 장애 치료제 후보 물질을 투여하고, 이온채널의 활성을 측정하는 단계를 포함하는 양극성 장애 치료제의 스크리닝 방법을 제공한다.
구체적으로, 본 발명에 따른 스크리닝 방법은
(a) 시료에 양극성 장애 치료제 후보 물질을 투여하는 단계;
(b) 상기 시료에서 TREK1 채널 및 TREK2 채널의 활성을 검출하는 단계; 및
(c) TREK1 채널만을 활성화시키는 경우, 상기 후보물질이 양극성 장애 치료활성을 나타내는 것으로 판정하는 단계를 포함한다.
본 발명에서 용어, "시료"는 시험, 검사, 분석 따위에 사용되는 물질이나 생물을 의미하며, 검체 또는 시험감으로도 불리운다. 바람직하게 상기 시료는 세포막에 이온채널이 구비된 세포를 의미하며, 더욱 바람직하게는 TREK1 채널 및 TREK2 채널이 구비된 신경세포를 의미한다.
본 발명에서 용어, "후보 물질"은 조울증으로도 알려진 양극성 스펙트럼 장애 관련 질환의 예방 또는 치료가 가능할 것으로 여겨지는 모든 물질을 의미하며, TREK1을 표적으로 함으로써 상기 질환을 치료할 수 있는 능력을 측정하는 대상을 말한다. 예를 들어, 단백질, 올리고펩티드, 소형 유기 분자, 다당류, 폴리뉴클레오티드 및 광범위한 화합물 등의 임의 분자를 포함한다. 이러한 후보 물질은 또한 천연 물질 뿐만 아니라 합성 물질을 모두 포함한다.
본 발명에서 용어, "양극성 장애"는 진행성 정신과 질환으로, 기분 변화의 반복 에피소드를 특징으로 한다. 당해 에피소드는 조증, 경조증(조증 형태보다 덜 중증), 우울증 또는 조증과 우울증의 결합 증상을 나타낼 수 있다. 제Ⅰ형 양극성 장애는 보다 조증의 또는 혼합된 기분 증상을 특징으로 하는 반면, 제Ⅱ형 양극성 장애는 1차 우울증 에피소드 및 자발적 경조증 에피소드로 구별된다. 양극성 우울증은 주요 우울증 증상을 특징으로 하는 증상과 상이하지 않다. 급성 조증은 명랑하거나 짜증스런 기분 및 적어도 3개 내지 4개의 전형적 조증 징후 및 증상(예를 들어, 비정상적으로 명랑하거나 짜증스런 기분, 과대 또는 과장된 자존심, 수면 감소, 비정상적 사고, 산만)과 관련이 있다.
경조증은 조증 에피소드와 관련된 장애는 없지만, 일정 기간의 온화한 기분 상승, 날카로와진 긍정적 사고 및 증가된 에너지 및 활성 수준과 관련이 있다. 조증은 비정상적이면서 지속적으로 매우 기분이 좋거나, 팽창된 또는 짜증스러운 기분이라는 뚜렷한 기간을 특징으로 한다. 혼합 조증은 조증 및 주요 우울 삽화의 기준 모두가 충족되는 적어도 한 주의 지속 기간을 특징으로 한다. 짧은 주기 순환은 조증과 우울증의 순환 주기가 매우 짧은 주기로서 정의된다. 급속한 순환은 우울증과 조증/흥분의 교대로서 정의된다. 순환성 기분 장애는 하위증후군의 우울증 및 경조증 에피소드의 잦고 짧은 순환으로 특징 지워지는 약해진 양극성 장애이다. 양극성 장애 질환의 재발은 전기생리학적/신경생리학적 자극으로 야기되는 것으로 가정하였다. 기분 안정제가 양극성 장애 치료에 사용되며, 기존의 기분 안정제는 항-자극 효과를 나타낸다. 항-자극 효과를 나타내는 항경련제 및 항간질제는 급속한 순환 양극성 장애를 포함하는 양극성 장애 치료 및 급성 조증의 치료 및 예방에서 중요한 대안제이고, 보조제이다.
칼륨 채널은 선택적으로 K+ 이온을 세포막 통과를 촉진하는 막 단백질로서, 세포막 전위차 결정 및 막 저항성 조절을 통한 세포의 흥분도 조절에 중요한 역할을 담당한다. 인체에는 다양한 종류의 K+ 채널이 존재하며 이들은 여러 기전에 의해 활성화된다.
본 발명에서 용어, "TREK1" 및 "TREK2"는 2-기공 도메인 K+(K2P) 채널 패밀리의 멤버로서, 항우울제(fluoxetine, norfluoxetine, 및 paroxetine) 및 항정신병제(fluphenazine, chlorpromazine, haloperidol, flupenthixol, loxapine, 및 pimozide)에 의해 농도-의존적 및 가역적으로 저해된다. 다양한 세포에서 TREK1 및 TREK2는 생물리학적 및 약리학적 특성이 유사하다고 알려져 있으나, TREK1에 비해 TREK2에 관한 연구는 미흡한 상황이며, 아직까지 TREK 채널과 기분 안정제의 연관성에 대하여 보고된 바가 없다.
본 발명의 일 실시예에서, HT-22 세포에 항우울제, 기분 안정제를 처리한 결과, TREK1의 활성이 항우울제에 억제(도 1a 및 1b)되는 반면, 기분 안정제에 의해서는 TREK1이 활성화(도 2a 및 2b)되었으며, TREK2의 활성은 오직 항우울제에 의해서만 억제(도 1a 및 1b)되는 것을 확인하였다. 따라서, 상기 결과로부터 TREK1이 양극성 장애의 치료적 타겟(target)이 될 수 있음을 확인하였다.
본 발명에서 용어, "스크리닝"은 항생물질, 효소 등 특정한 화학물질에 대하여 감수성 또는 활성이 있는 등 특정한 성질을 갖고 있는 것을 찾아내는 일을 의미한다.
본 발명의 스크리닝 방법에서, TREK1과 양극성 장애 치료제 후보 물질 사이의 반응 확인은 단백질-단백질간 또는 단백질-화합물간의 반응 여부를 확인하는데 통상 사용되는 방법들을 사용할 수 있다. 예를 들어, TREK1과 양극성 장애 치료제 후보 물질을 반응시킨 후 활성을 측정하는 방법, 효모 이중 혼성법(yeast two-hybrid), TREK1에 결합하는 파지 디스플레이 펩티드 클론의 검색, 천연물 및 화학물질 라이브러리 등을 이용한 HTS(high throughput screening), 드럭 히트 HTS 또는 세포 기반 스크리닝(cell-based screening) 등을 이용하는 스크리닝 법 등을 사용할 수 있으나, 이러한 방법에 제한되는 것은 아니다.
또한, TREK1을 암호화하는 유전자와 양극성 장애 치료제 후보 물질 사이의 반응 확인은 DNA-DNA, DNA-RNA, DNA-단백질 간의 반응 여부를 확인하는데 사용되는 통상적인 방법들을 사용할 수 있다. 예를 들어 in vitro 상에서 상기 유전자와 양극성 장애 치료제 후보 물질 사이의 결합 여부를 확인하기 위한 혼성화 시험, 포유류 세포와 양극성 장애 치료제 후보 물질을 반응시킨 후 노던 분석(Northern blotting)을 통한 상기 유전자의 발현률 측정 방법, 또는 상기 유전자에 리포터 유전자를 연결시켜 세포 내로 도입한 후 양극성 장애 치료제 후보 물질과 반응시키고 리포터 단백질(reporter protein)의 발현률 측정하는 방법 등을 사용할 수 있으나, 이러한 방법에 제한되는 것은 아니다.
본 발명의 스크리닝 방법에서, 양극성 장애 치료제 후보 물질은 통상적인 선정방식에 따라 양극성 장애의 예방 또는 치료의 가능성을 지닌 것으로 추정되거나 또는 무작위적으로 선정된 개별적인 핵산, 단백질, 기타 추출물 또는 천연물, 화합물 등이 될 수 있다.
이러한 스크리닝 방법에 의하여 얻어진 물질은 이후의 양극성 장애의 예방 또는 치료제 개발 과정에서 선도물질(leading compound)로서 작용하게 되며, 선도 물질이 TREK1의 활성화 물질 또는 이를 암호화하는 유전자에 대한 효과를 나타낼 수 있도록 그 구조를 변형시키고 최적화함으로써, 새로운 양극성 장애 예방 또는 치료제를 개발할 수 있다. 이러한 물질은 TREK1을 활성화시키는 물질, 바람직하게는 TREK2의 활성화에는 영향을 미치지 않는 물질, 또는 이를 암호화하는 유전자에 대해 부분적 또는 완전한 활성 효과를 나타내게 되므로 양극성 장애 관련 질환들을 예방 또는 치료할 수 있다.
바람직하게, TREK1 채널 및 TREK2 채널의 활성 측정은 패치클램프 방법에 의해 수행되는 것이 바람직하다.
본 발명에서 용어, "패치클램프 방법(patch clamp method)"은 팁의 직경이 1 내지 3 ㎛ 정도인 마이크로파이펫을 세포막 등의 막 표면에 접촉하여 미세한 이온 커런트(current)를 측정하는 방법으로서, 이온 채널의 기능을 분석하는데 통상적으로 사용되는 방법이다. 상기 패치클램프 방법은 현미경 하에서 마이크로-매니퓰레이터(micro-manipulator)에 의해 구동되는 유리 피펫(glass pipette)을 사용하여 세포막(cell membrane) 패치를 흡입하여 1 GΩ 정도의 고저항 밀봉(sealing)을 형성한 후, 이온 채널을 통한 세포막 패치나 전체 세포의 세포막에서의 전류를 측정한다.
이하, 본 발명을 실시예에 의하여 더욱 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다.
실시예 1: 세포 배양 및 형질감염
HEK-193 세포를 35 ㎜ 배양 용기에 2 × 105 세포 밀도로 접종한 후, 10% 소태아혈청(FBS; Invitrogen사, Grand Island, NY, USA) 및 50 U/㎖ 페니실린 및 스트렙토마이신(Invitrogen사)이 포함된 Dulbecco's modified Eagle's medium(DMEM)에서 24시간 동안 배양하였다. HEK-293 세포는 LipofectAMINE(Invitrogen사) 및 Opti-MEM® I Reduced Serum Medium(Invitrogen사)를 이용하여 인간 TREK1 DNA(진뱅크 수탁번호 제BC069462호) 또는 인간 TREK2 DNA(진뱅크 수탁번호 제AF385400호)가 포함된 pcDNA3.1 및 pcDNA3.1/녹색 형광 단백질(GFP)로 공동-형질감염(co-transfection) 시켰다.
전기생리학적 실험을 위하여, 형질감염된 세포를 세포 부착이 용이하도록 poly-L-lysine이 코팅된 35 ㎜ 배양 용기 내의 12 ㎜ 현미경 커버글라스에 부착시켰고, 37℃, 5% CO2, 95% 습도 조건에서 배양하였다. 수은 램프 광원이 탑재된 형광현미경을 이용하여 GFP 형광을 발현하는 세포를 탐지하였으며, 형질감염 1 내지 3일 후 세포를 사용하였다. 모든 실험은 경상대학교의 연구 윤리위원회의 동의하에 수행하였다.
실시예 2: 전기생리학적 연구
패치클램프 증폭기(Axopatch 200, Axon Instruments사, Union City, CA, USA)를 이용하여 전기생리학적 레코딩(recording)을 수행하였다. 모든 세포 전류는 과도 용량성(capacitive transient)을 캔슬링(canceling) 한 후에 레코딩 하였다. 단일-채널 및 전체-세포 전류는 pCLAMP 프로그램(버전 9, Axon)을 이용해 분석하였다. 파이펫 및 바스 솔루션(bath solution)은 150 mM KCl, 1 mM MgCl2, 5 mM EGTA, 및 10 mM HEPES(pH 7.3)를 포함하고, 전체-세포 전류의 레코딩을 위한 바스 솔루션은 135 mM NaCl, 5 mM KCl, 1 mM CaCl2, 1 mM MgCl2, 5 mM glucose, 및 10 mM HEPES(pH 7.3)을 포함한다. HCl 또는 KOH를 이용하여 pH를 조정하였다. 농도-의존적 저해 및 활성화 각각에 대한 IC50 및 EC50 값을 얻기 위하여, 데이터를 평균한 후, 표준 시그모이드 함수에 대입하였다.
실시예 3: 웨스턴 블롯 분석
마우스 해마 HT-22 신경세포(Dr. David R. Schubert, Salk Institute, San Diego, CA, USA로부터 수령)를 37℃ 온도, 10% FBS, 2 mM L-glutamate, 0.24% HEPES, 0.375% sodium bicarbonate, 100 U/㎖ 페니실린, 및 100 ㎍/㎖ 스트렙토마이신이 포함된 DMEM에서 배양하였다. TREK 단백질의 발현 변화를 확인하기 위하여, 세포에 fluoxetine(30 μΜ), paroxetine(30 μΜ), LiCl(1 mM), 또는 carbamazepine(100 μΜ)을 24시간 동안 처리하였다.
24시간 경과 후, HT-22 세포를 수득하여 차가운 PBS로 세 번 세척하였고, lysis buffer(RIPA buffer, Cell signaling technology사, Danvers, MA, USA; 20 mM Tris-HCl(pH 7.5), 150 mM NaCl/1 mM Na2EDTA, 1 mM EGTA, 1% NP-40, 1% sodium deoxycholate, 2.5 mM sodium pyrophosphate, 1 mM β-glycerophosphate, 1 mM Na3VO4, 및 1 ㎍/㎖ leupeptin)로 용해시켰고, 4℃에서 30분 동안 배양하였으며, 4℃에서 13,000 rpm(16,609 × g, Micro 17TR, Hanil사, Korea)으로 30분 동안 원심분리 하였다. 상기 과정을 통해 원심분리된 상층액을 수득하여 Bradford protein assay(Bio-Rad, Hercules, CA, USA)를 실시하였고, 총 단백질에 대하여 정량화 하였다. 총 단백질 샘플(25 ㎍/레인)은 10% SDS-폴리아크릴 아미드겔을 이용하여 전기영동(10% SDS-PAGE) 하였고, semi-dry blotter(Bio-Rad사)를 이용하여 buffer solution(TBS; 25 mM Tris-base, 190 mM glycine, 및 20% methanol)에서 polyvinylidene fluoride(PVDF) 멤브레인(0.45 ㎛, Millipore사, Bedford, MA, USA)으로 이동시켰다.
5% 탈지분유 및 0.05% Tween 20이 포함된 TBS를 이용하여 상기 멤브레인을 1시간 동안 차단한 후, 멤브레인에 항-TREK1(Alomone Labs사, Jerusalem, Israel/ Chemicon, Temecula, CA, USA) 및 항-TREK2(Alomone Labs사) 폴리클로날 항체를 1:1000으로 희석하여 반응시킨 후, 4℃에서 하룻밤 동안 면역블로팅 하였다. Horseradish peroxidase(HRP)-접합된 염소 항-토끼 항체(1:3000; assay designs, Ann Arbor사, MI, USA)로 실온에서 1시간 동안 반응시킨 후, 화학발광(ECL Plus kit; ELPIS사, Taejeon, Korea) 반응을 이용하여 항원 및 항체의 결합을 측정하였다.
실시예 4: 화학물질
전기생리학적 연구를 위하여, fluoxetine, paroxetine, citalopram, chlorpromazine, lamotrigine, LiCl, gabapentin, valproate, 및 carbamazepine를 포함하는 약제(pharmacological agents)를 최저 농도부터 처리하였다. fluoxetine(10 mM), citalopram(10 mM), chlorpromazine(100 mM), gabapentin(100 mM), valproate(1,000 mM), 및 LiCl(1,000 mM)의 스탁 솔루션(stock solution)은 D.W.로 제조하였고, carbamazepine(100 mM)은 에탄올로 제조하였고, lamotrigine(100 mM) 및 paroxetin(20 mM)는 DMSO로 제조하였으며, 바스 솔루션으로 희석하여 워킹 농도(working concentrate)로 조정하였다. 에탄올 또는 DMSO를 용매로 사용하였고, 당량농도를 함유하는 용액은 콘트롤로서 사용하였다. carbamazepin, lamotrigine, 및 paroxetine에 대한 에탄올 및 DMSO의 최종 농도는 0.1%였다. 모든 화학물질은 Sigma Chemical Co.사(St Louis, MO, USA)에서 구입하였다.
실시예 5: 데이터 분석 및 통계
형광 이미지 분석장치인 LAS-4000(Fujifilm corp사, Tokyo, Japan)를 이용하여 웨스턴 블롯 이미지를 캡쳐하였다. 웨스턴 블롯을 통해 전기영동된 밴드는 GS-800 calibrated densitometer(Bio-Rad사)에 연결된 시그마 겔 이미지 분석 소프트웨어(버전 1.0, Jandel Scientific, CA, USA) 또는 퀀티티 원 소프트웨어(버전 4.6.3)를 이용하여 정량화 하였다. 알파-튜불린(α-tubulin) 농도에 대하여 정상화하여 단백질의 상대적 수준을 계산하였다. 실험 결과의 통계학적 분석은 Student's t-test를 이용하여 처리구간의 유의성을 검정하였다(p < 0.05).
결과
1. TREK1 및 TREK2 전류에 영향을 미치는 항우울제 및 항정신병제의 효과
TREKs에 영향을 미치는 항우울제 및 항정신병제의 효과를 확인하기 위하여, TREK1 또는 TREK2로 형질감염된 세포를 포함하는 바스 솔루션에 fluoxetine, paroxetine, 및 chlorpromazine(항정신병제)을 처리하였다. TREK1 전류는 10 μΜ fluoxetine, 20 μΜ paroxetine, 및 30 μΜ chlorpromazine에 의해 각각 28 ± 11, 40 ± 9, 및 52 ± 10% 감소되었고(p < 0.05, 도 1a 및 도 1b의 A), TREK2 전류는 fluoxetine(10 μΜ), paroxetine(20 μΜ), 및 chlorpromazine(100 μΜ)에 의해 각각 55 ± 4, 33 ± 7, 및 57 ± 5%로 감소하였으며(n=5, p < 0.05, 도 1a 및 도 1b의 A), 이러한 저해는 농도-의존적 및 가역적이었다. fluoxetine에 의한 TREK1 및 TREK2 전류의 억제에 해당하는 IC50 값은 40 μΜ(TREK1은 37.9 ± 7.7 μΜ, TREK2는 28.7 ±7.6 μΜ) 이내였다(도 1b의 B).
상기 결과로부터, 항우울제 및 항정신병제는 TREKs의 활성을 저해한다는 것을 확인하였다.
2. TREK1 및 TREK2 전류에 미치는 기분 안정제의 효과
기분 안정제가 TREKs에 미치는 영향을 확인하기 위하여, 바스 솔루션에 LiCl, gabapentin, valproate, 및 carbamazepine을 처리하였다. TREK1의 전류는 LiCl(1 mM), gabapentin(100 μΜ), valproate(100 μΜ), 및 carbamazepine(100 μΜ)에 의해 각각 31 ± 14%, 25 ± 11%, 28 ± 12%, 및 72 ± 12% 활성화 되었지만(n=5, p < 0.05) (도 2a 및 도 2b의 A), 상기 약물들에 의해 TREK2 전류가 활성화되지는 않았다(도 2a 및 도 2b의 A). TREK1의 활성화는 LiCl, gabapentin, valproate, 및 carbamazepine에 농도-의존적 및 가역적이었다. TREK1 전류는 carbamazepine의 농도에 의존적으로 활성화 되었고, 이의 EC50 값은 78.9 ± 18.5 μΜ이었지만, 1 mM 이상의 LiCl 농도에 대해서는 추가적인 효과가 나타나지 않았다(도 2b의 B). 다른 약물들에 비해 carbamazepine는 TREK1에 보다 민감하게 작용하였다(도 2a 및 도 2b의 A).
또한, TREK1 및 TREK2 채널 활성은 LiCl 및 valproate와 구조적으로 유사한 화합물을 이용하여 확인하였다. RbCl 및 2-en-valproate(valproate의 대사체)는 TREK1 및 TREK2 채널 활성에 대하여 LiCl 및 valproate와 유사한 효과를 나타내었다. TREK1은 RbCl(1 mM) 및 2-en-valproate(100 μΜ)에 의해 각각 27 ± 13% 및 39 ± 17%로 활성화 되었다(도 3의 A 및 도 3의 C). LiCl 및 valproate와 유사하게 RbCl 및 2-en-valproate은 TREK2 활성에 어떠한 영향도 미치지 않았다(도 3의 B 및 도 3의 C). 또한, 인사이드 아웃(inside-out) 및 아웃사이드 아웃(outside-out) 패치 환경(patch configurations)에서 레코딩 하였을 때, 기분 안정제는 TREK1 전류를 증가시켰다.
상기 결과로부터, 기분 안정제는 특이적으로 TREK1 활성에 영향을 미친다는 것을 확인하였다.
3. 항우울제 및 기분 안정제에 의한 TREK 단백질 발현 변화
항우울제 및 기분 안정제가 TREKs의 발현 변화에 미치는 영향을 확인하기 위하여, HT-22 세포에 LiCl, carbamazepine, paroxetine, 또는 fluoxetine을 처리하였다. 기분 안정제 중, LiCl은 양극성 장애를 치료하기 위해 통상적으로 사용되는 약물이고, carbamazepine은 TREK1에 대하여 가장 효과적이기 때문에, LiCl 및 carbamazepine을 선택하였다. 서루 다른 두 회사로부터 구입한 항-TREK1 항체를 사용하여, ~50 kDa에 해당하는 두 개의 밴드를 확인하였다. 알파-튜불린을 이용하여 TREK 단백질의 발현 수준을 정상화 하였다. TREK1 발현 수준을 나타내는 두 개 밴드의 강도는 LiCl 또는 carbamazepine에 의해 유의하게 증가하였다(LiCl 강도: 1.4± 0.2(상위) 대 1.8 ± 0.4(하위) 및 carbamazepine 강도: 1.4 ± 0.3(상위) 대 1.5 ± 0.4(하위), 도 4의 A). 대조적으로, HT-22 세포에 paroxetine 및 fluoxetine을 처리하였을 때, 상위 밴드 및 하위 밴드 간에 서로 다른 양상을 나타내었다; 상위 밴드는 하향-조절되거나 변하지 않았던 반면(paroxetine: 0.9 ± 0.3 및 fluoxetine: 1.2 ± 0.5), 하위 밴드는 유의하게 상향-조절 되었다(paroxetine: 8.3 ± 3.0 및 fluoxetine: 9.0 ± 3.7, n=6, p < 0.05, 도 4의 B). 항-TREK2 항체를 사용하여 LiCl, carbamazepine, paroxetine, 및 fluoxetine을 처리한 HT-22 세포에서 ~60 kDa에 해당하는 밴드를 확인하였다(도 4의 A). LiCl 및 carbamazepine은 TREK2 발현 수준에 영향을 미치지 않았다. TREK2 발현은 paroxetine 및 fluoxetine에 의해 유의하게 증가하였다(paroxetine 강도: 1.3 ± 0.2 및 fluoxetine 강도: 1.6 ± 0.4). 또한, HT-22 세포에서 TREK2에 비해 TREK1의 발현 수준이 더 높았으며(도 4의 A), 음성 대조군으로 사용된 형질감염되지 않은 COS-7 세포에서는 어떠한 밴드도 나타나지 않았다.
Claims (6)
- (a) 시료에 양극성 장애 치료제 후보 물질을 투여하는 단계;(b) 상기 시료에서 TREK1 채널 및 TREK2 채널의 활성을 검출하는 단계; 및(c) TREK1 채널만을 활성화시키는 경우, 상기 후보물질이 양극성 장애 치료활성을 나타내는 것으로 판정하는 단계를 포함하는 양극성 장애 치료제의 스크리닝 방법.
- 제1항에 있어서,상기 시료는 세포막에 TREK1 채널 및 TREK2 채널이 구비된 세포를 포함하는 것인 양극성 장애 치료제의 스크리닝 방법.
- 제2항에 있어서,상기 세포는 신경세포인 것인 양극성 장애 치료제의 스크리닝 방법.
- 제1항에 있어서,상기 양극성 장애는 제Ⅰ형 양극성 장애, 제Ⅱ형 양극성 장애, 순환성 기분 장애, 급속한 순환, 짧은 주기 순환, 양극성 우울증, 급성 조증, 조증, 혼합 조증, 경조증 및 양극성 장애와 관련한 에피소드로 구성된 군으로부터 선택되는 것인 양극성 장애 치료제의 스크리닝 방법.
- 제1항에 있어서,상기 TREK1 채널 및 TREK2 채널의 활성 검출은 패치클램프 방법에 의해 수행되는 것인 양극성 장애 치료제의 스크리닝 방법.
- 제1항에 있어서,상기 (b) 단계에서, 상기 TREK2 채널의 활성은 증감이 없는 것인 양극성 장애 치료제의 스크리닝 방법.
