Abstract
薬理効果を発揮する金属サレン錯体の分子構造を明確し、係る分子構造を備えた金属サレン錯体及びその誘導体を提供する。金属サレン錯体又は当該金属サレン錯体の誘導体の複数分子がそれぞれの金属原子の部分で水を介して多量体化されていることを特徴とする金属サレン錯体化合物である。
Description
本発明は、自己磁性を有する金属サレン錯体化合物に関するものである。
一般に薬剤は生体内に投与され患部に到達し、その患部局所において薬理効果を発揮することで治療効果を引き起こすが、薬剤が患部以外の組織(つまり正常組織)に到達しても治療にはならない。
したがって、いかにして効率的に患部に薬剤を誘導するかが重要である。薬剤を患部に誘導する技術はドラッグ・デリバリと呼ばれ、近年研究開発が盛んに行われている分野である。
このドラッグ・デリバリには少なくとも二つの利点がある。一つは患部組織において十分に高い薬剤濃度が得られることである。薬理効果は患部における薬剤濃度が一定以上でないと生じず、低い濃度では治療効果が期待できない。二つめは薬剤を患部組織のみに誘導して、正常組織への副作用を抑制することができる。
このようなドラッグ・デリバリが最も効果を発揮するのが抗がん剤によるがん治療である。抗がん剤は細胞分裂の活発ながん細胞の細胞増殖を抑制するものが大半であるため、正常組織においても細胞分裂の活発な組織、例えば骨髄あるいは毛根、消化管粘膜などの細胞増殖を抑制する。
このため抗がん剤の投与を受けたがん患者には貧血、抜け毛、嘔吐などの副作用が発生する。これら副作用は患者にとって大きな負担となるため、投薬量を制限しなければならず、抗がん剤の薬理効果を十分に得ることが出来ないという問題がある。
この抗悪性腫瘍薬の中で、アルキル系抗悪性腫瘍薬は、核酸蛋白などにアルキル基(―CH2-CH2-)を結合させる能力をもつ抗がん剤の総称である。DNAをアルキル化してDNA複製を阻害し、細胞死をもたらす。この作用は細胞周期に無関係に働きG0期の細胞にもおよび、増殖が盛んな細胞に対する作用が強く、骨髄、消化管粘膜、生殖細胞、毛根などに障害を与えやすい。
また、代謝拮抗系抗悪性腫瘍薬は、核酸や蛋白合成過程の代謝物と類似の構造をもつ化合物であり、核酸合成を阻害するなどして細胞を障害し、分裂期の細胞に特異的に作用する。
また、抗腫瘍性抗生物質は、微生物によって産生される化学物質であり、DNA合成抑制、DNA鎖切断などの作用をもち抗腫瘍活性を示す。
また、微小管阻害薬は、細胞分裂の際に紡錘体を形成したり、細胞内小器官の配置や物質輸送など、細胞の正常機能の維持に重要な役割を果たしている微小管に直接作用することで抗腫瘍効果を示す。微小管阻害剤は細胞分裂が盛んな細胞や神経細胞などに作用を及ぼす。
また、白金製剤は、DNA鎖または鎖間結合あるいはDNA蛋白結合を作ってDNA合成を阻害する。シスプラチンが代表的薬剤であるが腎障害が強く、多量の補液が必要とされる。
また、ホルモン類似薬系抗悪性腫瘍薬は、ホルモン依存性の腫瘍に対して有効である。男性ホルモン依存性の前立腺がんに対して女性ホルモンを投与したり抗男性ホルモン剤を投与したりする。
また、分子標的薬は、それぞれの悪性腫瘍に特異的な分子生物学的特徴に対応する分子を標的とした治療法である。
また、トポイソメラーゼ阻害薬は、DNAに一時的に切れ目を入れてDNA鎖のからまり数を変える酵素である。トポイソメラーゼIは、環状DNAの一方の鎖に切れ目を入れ、もう一方の鎖を通過させた後、切れ目を閉じる酵素であり、トポイソメラーゼ阻害薬IIは環状DNAの2本鎖両方を一時的に切断し、その間を別の2本鎖DNAを通過させ、再び切れ目をつなぎ直す酵素である。
さらに、非特異的免疫賦活薬は、免疫系を活性することによってがん細胞の増殖を抑制する。
ドラッグ・デリバリの利点は、局所麻酔剤でも存在する。局所麻酔剤は、痔疾患、口内炎、歯周病、虫歯、抜歯、あるいは手術等による粘膜や皮膚等の居所的なかゆみや疼痛を処理ずるために用いられている。代表的な居所麻酔剤として、リドカイン(商品名:キシロカイン)が知られているが、リドカインは即効性に優れているものの、抗不整脈作用を有する。また、脊椎麻酔を行う際、脊髄液の中に麻酔薬であるリドカインを注入すると脊髄液中を拡散し、最悪の場合は頸部の脊髄に達することで呼吸機能が停止し重篤副作用をもたらす懸念がある。
ドラッグ・デリバリの具体的な手法としては、例えば、担体(キャリア)を用いたものがある。これは患部に集中しやすい担体に薬剤を載せて、薬剤を患部まで運ばせようというものである。
担体として有力視されているのが磁性体であり、薬剤に磁性体である担体を付着させ、磁場によって患部に集積される方法が提案されている。(例えば、特許文献1参照)。
しかしながら、磁性体担体をキャリアとして使用する場合、経口投与が困難なこと、担体分子が一般に巨大であること、担体と薬剤分子との間の結合強度、親和性に技術的な問題があることがわかり、そもそも実用化が困難であった。
そこで、本発明者は、有機化合物の基本骨格に対して、正又は負のスピン電荷密度付与する側鎖が結合され、全体として外部磁場に対して磁気共有誘導される範囲の適性を持ち、人体や動物に適用された際に、体外からの磁場によって局所的に磁場が与えられている領域で保持され、元来保有している医薬効果を前記領域において発揮するようにした局所治療薬を提案した。この提案には、このような薬剤として鉄サレン錯体が記載されている。(例えば、特許文献2参照)。
また、有機磁性体の総説が記載されている文献(例えば、非特許文献1参照)や、シスプラチンに含まれる白金を他の元素で置き換えることが記載されている文献(例えば、非特許文献2参照)も発行されている。

しかしながら、特許文献1及び2に記載された方法や化合物は、薬理効果を発揮する金属サレン錯体の分子構造が必ずしも明確ではなかった。また、非特許文献1及び2には、薬そのものが磁性になることについては言及されていない。
本発明は、このような事情に鑑みなされたものであり、薬理効果を発揮する金属サレン錯体の分子構造を明確にし、係る分子構造を備えると共に、それ自身に磁性を持たせた金属サレン錯体及びその誘導体を提供することを目的とするものである。
前記目的を達成するために、本発明は、金属サレン錯体又は当該金属サレン錯体の誘導体の複数分子がそれぞれの金属原子の部分で水を介して多量体化されていることを特徴とする金属サレン錯体化合物を提供するものである。
前記金属サレン錯体化合物は、金属サレン錯体又は当該金属サレン錯体の2量体を含むことができる。
また、前記金属サレン錯体化合物は、多量体化された金属サレン錯体又は当該金属サレン錯体の誘導体が自己磁性を有することもできる。
本発明の好適な形態は、下記化学式(I)で示される自己磁性金属サレン錯体及びその誘導体である。

本発明によれば、薬理効果を発揮する金属サレン錯体及びその誘導体の構造が明らかとなった。
(実施例1)
本発明の金属サレン錯体の製造を次のように行った。
本発明の金属サレン錯体の製造を次のように行った。
4-ニトロフェノール(4-nitrophenol);25g、0.18mol、ヘキサメチレンテトラミン(hexamethylenetetramine);25g、0.18mol、ポリりん酸(Polyphosphoric acid);200mlの混合物を1時間100℃で攪拌した。その後、その混合物を500mlの酢酸エチルと、1lの水の中に入れ、完全に溶解するまで攪拌した。さらにその溶液に400mlの酢酸エチルを追加で加えたところ、その溶液は2つの相に分離した。そのうちの水の相を取り除き、残りの化合物を塩性溶剤で2回洗浄し、無水MgSO4で乾燥させた結果、化合物2(compound 2)が17g(収率57%)合成できた。
化合物2(compound 2);17g、0.10mol、無水酢酸(acetic anhydride);200ml、H2SO4;少々、を室温で1時間攪拌させた。得られた溶液を2lの氷水中に入れて0.5時間混ぜ、加水分解を行った。次に、得られた溶液をフィルターにかけ、大気中で乾燥させたところ白い粉末状のものが得られた。酢酸エチルを含む溶液を使ってその粉末を再結晶化させたところ、24g(収率76%)の化合物3(compound 3)の白い結晶を得ることができた。
化合物3(compound 3);24g、77mmolと、メタノール;500mlに10%のパラジウムを担持したカーボン;2.4gの混合物を一晩、1.5気圧の水素還元雰囲気で還元した。終了後、フィルターでろ過したところ茶色油状の化合物4(compound 4)、21gが合成できた。
無水ジクロメタン(DCM);200mlに化合物4(compound 4);21g、75mmol、二炭酸ジ-tert-ブチル(di(tert-butyl) dicarbonate);18g、82mmolを窒素雰囲気で一晩攪拌した。得られた溶液を真空中で蒸発させた後、メタノール;100mlで溶解させた。その後、水酸化ナトリウム;15g、374mmolと水;50mlを加え、5時間還流させた。その後冷却し、フィルターでろ過し、水で洗浄後、真空中て乾燥させたところ茶色化合物がえられた。得られた化合物は、シリカジェルを使ったフラッシュクロマトグラフィーを2回行うことで、10g(収率58%)の化合物6(compound 6)が得られた。
無水エタノール;400mlの中に化合物6(compound 6);10g、42mmolを入れ、加熱しながら還流させ、無水エタノール;20mlにエチレンジアミン;1.3g、21mmolを0.5時間攪拌しながら数滴加えた。そして、その混合溶液を氷の容器に入れて冷却し15分間かき混ぜた。その後、200mlのエタノールで洗浄し、フィルターをかけ、真空で乾燥させたところ、8.5g(収率82%)の化合物7(compound 7)が合成できた。
通常のメタノール(昭和化学製メタノール、純度99.5%以上);50ml中に化合物7(compound 7);8.2g、16mmol、トリエチルアミン(triethylamine);22ml、160mmolを入れ、10mlのメタノールの中に、Feサレンの場合は、FeCl3(・4H2O);2.7g、16mmol、Mnサレンの場合は、MnCl3・4H2O;2.7g、16mmol、Crサレンの場合は、CrCl3・4H2O;2.7g、16mmolを加えた溶液を窒素雰囲気下で混合した。次いで、室温窒素雰囲気で1時間混合したところ茶色の化合物が得られた。その後、真空中或いはマグネシウムを使う等して十分に水を乾燥或いはマグネシウムに吸着除去させた。得られた化合物は、400mlのジクロロメタンで希釈し、塩性溶液で2回洗浄し、Na2SO4で乾燥させ、真空中で乾燥させたところ水分子を含む2量体の金属サレン錯体化合物が得られた。
得られた化合物は、ジエチルエーテルとパラフィンの溶液中で再結晶させ高速液化クロマトグラフィーで測定したところ純度95%以上の水分子を含む2量体の金属サレン錯体を得た。得られた水分子付2量体は下記の化学構造式の通りである。
得られた水分子付2量体の元素分析をしたところ、C;57.73%、H;4.42%、Fe;17.2%、N;8.49%、O;12.16%となり、計算値と実験値の違いはすべて絶対誤差の±0.4%以内に入っている。
次に、水分子混入の存在を明らかにするためTG-Mass分析を行った。TG-Mass分析の結果を図1~図3に示す。図1に示すTGA曲線によると、試料の質量は、100℃までは室温に対して100%、200℃になると99.9%、300℃になると97.4%になることが分かる。また、図2及び図3に示す質量分析の結果から、水分子のほとんどが除去されていることが分かる。また、300℃での質量の減少は2.6%である。これは、Feサレン分子2個に対して水分子1個(質量2.7%)相当が、結晶に取り込まれていることを示唆している。

(実施例5)
Feサレン錯体について、37℃(310K)における磁場-磁化曲線を図7に示す。また、図8は、Mnサレン錯体、Crサレン錯体及びFeサレン錯体の磁場-磁化曲線を示す図である。図8から、Feサレン錯体は、Crサレン錯体と比較して、磁化が多くなり、Mnサレン錯体は、Feサレン錯体と比較して磁場が30000Oe(3T)以上で磁化が多くなることが分かる。したがって、Feサレン錯体は、磁場が10000Oe(1T)未満の際に最も磁化が大きく、ネオジウム永久磁石等を用いる磁場誘導ドラッグ・デリバリ・システムに適している。しかしながら、磁場が30000Oe(1T)以上の場合は、Mnサレン錯体の磁化が一番大きく、超伝導磁石を用いる磁場誘導ドラッグ・デリバリ・システムに最適となる。
Feサレン錯体について、37℃(310K)における磁場-磁化曲線を図7に示す。また、図8は、Mnサレン錯体、Crサレン錯体及びFeサレン錯体の磁場-磁化曲線を示す図である。図8から、Feサレン錯体は、Crサレン錯体と比較して、磁化が多くなり、Mnサレン錯体は、Feサレン錯体と比較して磁場が30000Oe(3T)以上で磁化が多くなることが分かる。したがって、Feサレン錯体は、磁場が10000Oe(1T)未満の際に最も磁化が大きく、ネオジウム永久磁石等を用いる磁場誘導ドラッグ・デリバリ・システムに適している。しかしながら、磁場が30000Oe(1T)以上の場合は、Mnサレン錯体の磁化が一番大きく、超伝導磁石を用いる磁場誘導ドラッグ・デリバリ・システムに最適となる。
(実施例6)
ラットL6細胞が30%のコンフルエントの状態の時に、既述の方法によって得られたFeサレン錯体、Mnサレン錯体、Crサレン錯体、Coサレン錯体のそれぞれについて、金属サレン錯体の粉末を磁石に引き寄せられるのが目視できる程度の量を培地に散布して48時間後に培地の状態を写真撮影した。なお、図9は、ラットL6細胞の培地がある角型フラスコに棒磁石を接触させた状態を示している。
ラットL6細胞が30%のコンフルエントの状態の時に、既述の方法によって得られたFeサレン錯体、Mnサレン錯体、Crサレン錯体、Coサレン錯体のそれぞれについて、金属サレン錯体の粉末を磁石に引き寄せられるのが目視できる程度の量を培地に散布して48時間後に培地の状態を写真撮影した。なお、図9は、ラットL6細胞の培地がある角型フラスコに棒磁石を接触させた状態を示している。
次いで、48時間後角型フラスコ底面の一端から他端までを撮影し、細胞数を算出した結果を図10に示す。図10において磁石から近位とは、角型フラスコ底面における磁石端面の投影面積内を示し、磁石から遠位とは、角型フラスコ底面において磁石端面と反対側にある領域を示す。
図10に示すように、磁石から近位ではMnサレン錯体が引き寄せられてFeサレン錯体の濃度が増し、Feサレン錯体のDNA抑制作用によって細胞数が遠位よりも極端に低いことが分かる。この結果、本発明による、磁性を持った薬剤と、磁気発生手段とを備えたシステムによって、個体の目的とする患部や組織に薬剤を集中して存在させることが可能となることがわかる。
次に、誘導装置を用いた誘導例について説明する。この誘導装置は、図11に示すように、重力方向に互いに向き合う一対の磁石230及び232がスタンド234とクランプ235とによって支持されており、磁石230と磁石232との間には金属板236が配置されている。一対の磁石230と磁石232との間に金属板236、特に鉄板を配置することにより、局所的に一様で強力な磁界を作り出すことができる。
この誘導装置は、磁石の代わりに電磁石を用いて発生磁力を可変にすることができる。また、XYZ方向に一対の磁力発生手段を移動できるようにして、テーブル上の固体の目的とする位置に磁力発生手段を移動させることができる。この磁界の領域に固体の組織を置くことにより、この組織に薬剤を集中させることができる。
より具体的には、例えば、体重約30グラムのマウスに既述の金属錯体(薬剤濃度5mg/ml(15mmol))を静注して開腹し、右の腎臓を前記一対の磁石の間に来るようにマウスを鉄板の上に置く。なお、使用した磁石は、信越化学工業株式会社製
品番:N50(ネオジウム系永久磁石)、残留磁束密度;1.39~1.44Tである。このとき、右側の腎臓に与えられた磁場は、約0.3Tであり、左側の腎臓に与えられる磁場は、その約1/10である。

左の腎臓及び磁界を適用しない腎臓(control)と共に、マウスの右腎に磁界を加えて10分後、MRIでSNRをT1モード及びT2モードで測定した。その結果、図12に示すように、磁界を加えた右腎(RT)が左腎(LT)及びコントロールに比較して薬剤を組織内に留め置くことができることが確認された。
図13に、マウスにおけるメラノーマ成長に対するFeサレン錯体の効果を示す。メラノーマは、培養メラノーマ細胞(クローンM3メラノーマ細胞)の局所的移植によって、マウス尾腱においてインビボ(in vivo)に形成されたことが分かる。なお、図13(1)は、Feサレン錯体の代わりに、塩水を注入した塩水グループ(saline)、図13(2)は、磁場を適用せずにFeサレン錯体を注入したグループ(SC)、図13(3)は、磁場を適用しつつ(n=7~10)Feサレン錯体を注入したグループ(SC+Mag)の効果を示す写真である。
Feサレン錯体を尾腱の静脈から静脈投与(50mg/kg)し、市販の棒磁石(630mT、円筒状ネオジウム磁石;長さ150mm、直径20mm)を用いて、局所的に磁場を印加した。棒磁石の適用は、Feサレン錯体を10~14日注入した直後に、メラノーマサイトに3時間穏やかに接触させることにより行った。
棒磁石の適用は、磁場強度が、メラノーマ延長が予想される部位に最大強度となるように、150mm以下のマウス尾腱に対して2週間の成長期間行った。Feサレン錯体の初回注入の12日後に、メラノーマ染色された部位を評価することによって、メラノーマの延長を評価した。
図14に示すように、Feサレン錯体の代わりに、塩水を注入した塩水グループ(saline)では、メラノーマ拡張は最大であった(100±17.2%)。一方、磁場を適用せずにFeサレン錯体を注入したSCグループでは、メラノーマ拡張は緩やかに減少した(63.68±16.3%)。これに対して、磁場を適用しつつ(n=7~10)Feサレン錯体を注入したSC+Magグループでは、ほとんどのメラノーマが消失した(9.05±3.42%)。
図15に示すように、ヒストロジカル試験を、組織部の腫瘍増殖マーカーであるanti-Ki-67抗体及びanti-Cyclin D1抗体を用いて、ヘマトキシリン-エオジン染色及び免疫組織染色により行った。その結果、Feサレン錯体を注入した場合(SC)においてメラノーマの腫瘍拡張が減少し、さらにFeサレン錯体に磁場の適用が組み合わされた場合にはほとんどが消失することが分かった。
また、薬剤(Feサレン錯体;9.25mmol)に磁場強度;200Oe、周波数;50kHz~200kHzの交流磁場を印加したところ、薬剤の温度が2℃~10℃上昇した(図16)。これは、体内投与時の温度に換算すると39℃~47℃に相当し、がん細胞を殺傷することが可能な温度域であることが確認された。なお、図16(1)は、薬剤に交流磁場をかけたときの、時間に関する温度の変化であり、図16(2)は、周波数を固定して、磁場のみを変化させたときの最大温度であり、図16(3)は、磁場を固定して、周波数のみを変化させたときの最大温度である。
(実施例7)
金属サレン錯体に結合する化合物の電子の移動は第一原理計算で求めることができる。このコンピュータシミュレーションを実現するシステムは、コンピュータとしての公知のハードウエア資源を備えるものであって、メモリとCPUなどの演算回路を備える演算装置と、演算結果を出力する表示手段とを備えている。
金属サレン錯体に結合する化合物の電子の移動は第一原理計算で求めることができる。このコンピュータシミュレーションを実現するシステムは、コンピュータとしての公知のハードウエア資源を備えるものであって、メモリとCPUなどの演算回路を備える演算装置と、演算結果を出力する表示手段とを備えている。
メモリは、既存の有機化合物または3次元構造を特定するデータと、コンピュータシミュレーションを実現するソフトウエア・プログラムとを備えている。このソフトウエア・プログラムは、各化合物の側鎖を追加・変更・削除し、所定の側鎖間で架橋し、記述のスピン電荷密度の高い領域を計算し、構造全体としてのスピン電荷密度を決定可能なものである。このプログラムとして、例えば、市販品(Dmo13、アクセルリス社製)を利用することができる。
ユーザは化合物について、側鎖を追加する位置を入力し、または側鎖を変更し、あるいは削除するものを選択し、さらに、架橋を形成すべき箇所をメモリの支援プログラムを利用して演算装置に指定する。演算装置はこの入力値を受けて、スピン電荷密度を演算してその結果を表示画面に出力する。また、ユーザが既存の化合物の構造データをコンピュータシステムに追加することによって、既知の化合物についてのスピン電荷密度を得ることができる。

- 金属サレン錯体又は当該金属サレン錯体の誘導体の複数分子がそれぞれの金属原子の部分で水を介して多量体化されていることを特徴とする金属サレン錯体化合物。
