Abstract
Description
このような深在性真菌症の治療を目的とした薬剤は、抗菌剤と比較してはるかに少なく、アムホテリシンB、フルシトシン、ミコナゾール、フルコナゾール、ホスフルコナゾール、イトラコナゾール、ボリコナゾールおよびミカファンギンなどにすぎない。
たとえば、アムホテリシンBは、殺真菌作用が非常に強いが、腎毒性などの副作用の問題があり、臨床使用には制約がある。フルシトシンは、耐性化などの問題がある。ミカファンギンは、クリプトコッカス属に活性を示さない。フルコナゾールおよびボリコナゾールなどのアゾールは、有効性と安全性の兼ね合いから、現在、最も多用されているが、真菌に対する殺真菌作用は、アムホテリシンBよりも劣る(非特許文献2、3)。
皮膚表面の角質層は、外界に存在する様々な化学物質が、体内に侵入するのを防ぐバリアとしての機能を持つ(非特許文献6)。この機能のため、多くの薬物の皮膚透過性は低く、薬物を皮膚表面に単に塗布するだけでは、薬理効果を発揮するのに充分な薬物量を体内に吸収させることは困難である(非特許文献7)。さらに、角質層は親油性の高い膜であるため、脂溶性の高い化合物(オクタノール/水分配係数が100~1000)のほうが膜透過しやすく、水溶性の高い化合物は膜透過しにくい傾向にある(非特許文献6、8)。この角質層が持つ高いバリア機能をいかにして克服するかが経皮吸収製剤の開発における重要な課題である(非特許文献9)。こうした困難な課題を克服するために、多くの経皮吸収促進方法が研究、報告されている。しかし、その経皮吸収促進効果は、薬物によって大きく異なり、過去の情報から新規化合物に適切な経皮吸収促進方法を見出すことは極めて困難である(非特許文献10)。
本発明の経皮吸収製剤の有効成分は、化合物Aまたはその塩である。化合物Aは、種々の酸性化合物と塩を形成する。本発明は、そのいずれをも使用することができる。
化合物Aの塩としては、たとえば、塩酸、臭化水素酸、硝酸、リン酸および硫酸などの鉱酸との塩;ギ酸、酢酸、クエン酸、シュウ酸、フマル酸、乳酸、マレイン酸、コハク酸、リンゴ酸、酒石酸、アスパラギン酸、トリクロロ酢酸、トリフルオロ酢酸、オレイン酸およびカプリル酸などのカルボン酸との塩;ならびにメタンスルホン酸、ベンゼンスルホン酸、p-トルエンスルホン酸、メシチレンスルホン酸およびナフタレンスルホン酸などのスルホン酸との塩が挙げられる。好ましくは、塩酸、乳酸、メタンスルホン酸および酢酸との塩が挙げられ、より好ましくは、塩酸との塩が挙げられる。これらは、水和物および溶媒和物も含む。
経皮吸収製剤中の有効成分の濃度は、たとえば、製剤全体に対し、0.01~40重量%であり、好ましくは、0.1~10重量%であり、より好ましくは、0.3~6重量%であり、さらに好ましくは、1~3重量%である。
極性基を有さない非芳香族モノテルペンとしては、たとえば、リモネン、メンタン、カンフェン、ピネン、ミルセン、オシメン、アロオシメン、テルピノレン、テルピネン、フェランドレンおよびカレンなどが挙げられ、好ましくは、リモネン、メンタン、カンフェン、ピネンおよびアロオシメンが挙げられる。
極性基を有する非芳香族モノテルペンとしては、たとえば、ヒドロキシル基、カルボニル基またはエーテル結合を有する非芳香族モノテルペンが挙げられる。
極性基を有する非芳香族モノテルペンとしては、たとえば、ゲラニオール、テルピネオール、メントール、ボルネオール、イソボルネオール、ネロール、カンフル、シトロネロール、フェンチルアルコール、カルベオール、カルボン、プレゴン、ピペリトン、メントン、ネオメントール、シトロネラールおよびシネオールなどが挙げられ、好ましくは、ゲラニオール、メントール、ボルネオール、カンフル、メントンおよびシネオールが挙げられる。
低級アルコールとしては、エタノール、プロパノールおよび2-プロパノールなどが挙げられ、好ましくは、エタノールおよび2-プロパノールが挙げられる。
多価アルコールとしては、エチレングリコール、プロピレングリコール、1,3-プロパンジオール、1,2-ブタンジオール、1,3-ブタンジオール、1,4-ブタンジオール、2,3-ブタンジオール、1,5-ペンタンジオール、グリセリン、ジプロピレングリコール、ジエチレングリコール、トリエチレングリコールおよびポリエチレングリコール400などが挙げられ、好ましくは、プロピレングリコールが挙げられる。
pH調整剤とは、この分野において通常pH調整に用いられる酸または塩基であり、たとえば、塩酸、リン酸、硫酸、メタンスルホン酸、クエン酸、グルコン酸、コハク酸、乳酸、酢酸、水酸化ナトリウム、水酸化カリウム、クエン酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、酢酸ナトリウム、ジイソプロパノールアミンおよびジエタノールアミンなどが挙げられる。
本発明の経皮吸収製剤のpH範囲は、好ましくは、3~10である。
貼付剤の基材としては、通常使用される基材原料を使用することができる。たとえば、アクリル系基材、天然ゴム系基材、シリコン系基材、ビニルアルコール系基材、ビニルエステル系基材、ビニルエーテル系基材などが挙げられる。さらに、ロジン、ロジン変性マレイン酸、水添ロジンエステルなどの粘着付与剤を用いてもよい。

マトリックス拡散制御方式の貼付剤は、支持体層、薬剤貯蔵層、粘着剤層および剥離性保護フィルムから構成される。製剤を構成する支持体層、薬剤貯蔵層、粘着剤層および剥離性保護フィルムには、通常用いられている基材を、特に限定されることなく広く使用することができる。マトリックス拡散制御方式の貼付剤は、本発明の範囲に含まれる有効成分および経皮吸収促進剤を含有する組成物を薬剤貯蔵層および/または粘着層に用いて製造できる。さらに、必要に応じて他の成分を用いてもよい。
膜透過制御方式の貼付剤は、支持体層、薬剤貯蔵層、薬物放出制御膜、粘着剤層および剥離性保護フィルムで構成される。膜透過制御方式の貼付剤は、本発明の範囲に含まれる有効成分および経皮吸収促進剤を含有する組成物を薬剤貯蔵層内部に封入することにより、製造できる。さらに、必要に応じて他の成分を用いてもよい。
本発明で使用されるすべての成分は、常法により、溶解、懸濁、乳化してから用いることもできる。

有効成分3部、1,8-シネオール2部、プロピレングリコール20部、2-プロパノール20部、エタノール30部、および精製水25部を混合し、製剤100部を得た。
有効成分3部、l-メントン2部、プロピレングリコール20部、2-プロパノール20部、エタノール30部、および精製水25部を混合し、製剤100部を得た。
有効成分3部、(±)-カンフル2部、プロピレングリコール20部、2-プロパノール20部、エタノール30部、および精製水25部を混合し、製剤100部を得た。
雄性ヘアレスラット(HWY/Slc、8週齢)摘出腹部皮膚を、真皮側がレセプター側になるようにリン酸緩衝生理食塩水で満たしたフランツ型拡散セル(有効拡散面積:0.636cm2)に装着し、セル外部に32~33℃の恒温水を循環させた。角質層側に各実施例および比較例の製剤0.5mLを適用し、3時間毎にレセプター液1.5mLをサンプリングした(マイクロエッテ、ハンソンリサーチ製、MODEL57-12A-S)。各時間におけるレセプター液中の化合物Aの濃度を高速液体クロマトグラフィで測定することにより、24時間累積透過量を求めた。
検出器:紫外吸光光度計
測定波長:260nm
カラム:Symmetry Shield RP8(ウォーターズ製)、3.5μm、4.6×100mm
プレカラム:Symmetry Shield RP8(ウォーターズ製)、3.5μm、3.9×20mm
カラム温度:40℃
移動相:メタンスルホン酸・トリエチルアミン溶液:水:メタノール(5:70:25)
メタンスルホン酸・トリエチルアミン溶液は、次の方法で調製した。
水300mLにメタンスルホン酸60mLおよびトリエチルアミン60mLを加えた後、水を加えて全量を600mLとした。この溶液に1mol/Lリン酸水素二カリウム水溶液を加え、pH3.5に調整した。
流速:0.6mL/分
実施例1、3~6、8~11、15、17、27、29、30および32~35の製剤は、比較例1の製剤、脂肪族ジカルボン酸ジエステルを含有する比較例2および3の製剤、芳香族ジカルボン酸ジエステルを含有する比較例4の製剤、芳香族カルボン酸を含有する比較例5の製剤、界面活性剤を含有する比較例6の製剤ならびに軟化剤を含有する比較例7の製剤よりも顕著に高い皮膚透過性を示した。
実施例12および13の製剤は、高い皮膚透過性を示した。また、ミリスチン酸イソプロピルの添加量が多いほど有効成分の透過量が増加した。
実施例2、7および14の製剤は、高い皮膚透過性を示した。実施例18の製剤は、比較例8の製剤よりも高い皮膚透過性を示し、実施例2の製剤と比較してさらに高い皮膚透過性を示した。実施例19の製剤は、比較例8の製剤よりも高い皮膚透過性を示し、実施例7の製剤と比較してさらに高い皮膚透過性を示した。実施例24の製剤は、比較例9の製剤よりも高い皮膚透過性を示し、実施例14の製剤と比較してさらに高い皮膚透過性を示した。実施例18、19および24の製剤は、経皮吸収促進剤、および、l-メントールまたはゲラニオールの併用による高い皮膚透過促進効果を示した。
実施例20および23の製剤は、経皮吸収促進剤およびl-メントールの併用による高い皮膚透過促進効果を示した。
実施例21および22の製剤は、高い皮膚透過性を示した。
また、実施例21の製剤は、40℃で6ヶ月間保存後、有効成分の残存率は99%であり、安定な製剤であった。
実施例16の製剤は、高い皮膚透過性を示した。実施例25の製剤は、比較例8の製剤よりも高い皮膚透過性を示し、実施例16の製剤と比較してさらに高い皮膚透過性を示した。また、実施例26の製剤は、比較例9の製剤よりも高い皮膚透過性を示し、実施例16の製剤と比較してさらに高い皮膚透過性を示した。実施例25および26の製剤は、経皮吸収促進剤および極性基を有する非芳香族モノテルペンの併用による高い皮膚透過促進効果を示した。
実施例36および37の製剤は、経皮吸収促進剤および極性基を有する非芳香族モノテルペンの併用による高い皮膚透過促進効果を示した。
Claims (9)
- 4-{3-[4-(3-{4-[アミノ(イミノ)メチル]フェノキシ}プロピル)-1-ピペリジニル]プロポキシ}ベンズアミジンまたはその塩、および、経皮吸収促進剤を含有することを特徴とする経皮吸収製剤。
- 経皮吸収促進剤が、高級アルコール、高級モノカルボン酸、高級モノカルボン酸エステル、芳香族モノテルペンおよび極性基を有さない非芳香族モノテルペンから選ばれる1種または2種以上である請求項1に記載の経皮吸収製剤。
- 高級アルコールが、炭素数8~18の飽和アルコールおよび炭素数8~18の不飽和アルコール;高級モノカルボン酸が、炭素数8~18の飽和脂肪酸および炭素数8~18の不飽和脂肪酸;高級モノカルボン酸エステルが、炭素数6~18のモノカルボン酸および炭素数1~6のアルコールの反応生成物である請求項2に記載の経皮吸収製剤。
- 高級アルコールが、オクタノール、デカノール、ラウリルアルコール、ミリスチルアルコールおよびオレイルアルコール;高級モノカルボン酸が、カプリル酸、カプリン酸、ラウリン酸、ミリスチン酸、オレイン酸、リノール酸およびリノレン酸;高級モノカルボン酸エステルが、カプロン酸エチル、ラウリン酸エチル、ミリスチン酸イソプロピルおよびパルミチン酸イソプロピル;芳香族モノテルペンが、シメンおよびチモール;極性基を有さない非芳香族モノテルペンが、リモネン、メンタン、カンフェン、ピネンおよびアロオシメンである請求項2に記載の経皮吸収製剤。
- さらに、極性基を有する非芳香族モノテルペンを1種または2種以上含有する請求項1から4のいずれか一項に記載の経皮吸収製剤。
- 極性基を有する非芳香族モノテルペンが、ゲラニオール、メントール、ボルネオール、カンフル、メントンおよびシネオールである請求項5に記載の経皮吸収製剤。
- 経皮吸収促進剤の各成分の濃度が、製剤全体に対し、0.1~40重量%である請求項2から4のいずれか一項に記載の経皮吸収製剤。
- 経皮吸収促進剤の各成分の濃度が、製剤全体に対し、0.01~20重量%である請求項5または6に記載の経皮吸収製剤。
- 極性基を有する非芳香族モノテルペンの各成分の濃度が、製剤全体に対し、0.05~20重量%である請求項5から8のいずれか一項に記載の経皮吸収製剤。
