特に組織標本に関しては、標本の形態を観察するための形態観察染色として、ヘマトキシリンおよびエオジンの2つの色素を用いるヘマトキシリン・エオジン染色(HE染色)が標準的に用いられている(非特許文献1、特許文献1~2)。
ヘマトキシリン染色により細胞核・石灰部・軟骨組織・細菌・粘液が青藍色~淡青色に染色され、エオジン染色により細胞質・間質・各種線維・赤血球・角化細胞が赤~濃赤色に染色される。病理医は、染色された組織標本の顕微鏡画像の中で、細胞の核の大きさや形の変化、組織としてのパターンの変化などの形態学的な情報、染色情報、をもとに診断を行っている。
なお、この他の形態観察染色としては、例えば細胞診用いられるパパニコロウ染色(Pap染色)等がある。
免疫染色には、例えば、酵素を用いた色素染色法(DAB染色)が用いられる。DAB染色は、色素で染色されるように修飾された抗体を用いて観察対象となる抗原を染色して観察することで抗原量を測るものである。あるいは蛍光標識法が用いられることもある。蛍光標識法は、蛍光色素が修飾された抗体を用いて対象となる抗原を染色して観察することで抗原量を測るものである。
しかしながら、DAB染色のような酵素標識による染色はHE染色と色が近いため、HE染色による染色と酵素標識による染色の識別がしにくく、同時観察が困難という課題がある。加えて、DAB染色は染色濃度が温度・時間などの環境条件により大きく左右されるため、染色濃度から実際の抗体等の量を見積もることが難しいという課題がある。
蛍光法はDAB染色と比べて定量性に優れるという特徴がある(非特許文献1)。
しかしながら、蛍光標識を用いて病理診断と形態観察を同時に行う場合、組織染色に用いた染色剤の蛍光の影響を受け易いという課題がある。これに対して例えば、可視光の影響を受けない、赤外励起・発光の蛍光色素を使うことが挙げられる(特許文献4)。例えば、AlexaFluor647(Molecular Probes社)やCY5(GE社)のような赤外発光色素がある。
したがって、本発明の主な目的は、形態観察染色と免疫染色とを同時におこなう場合において、赤外・励起発光の蛍光色素を使用しなくても、蛍光標識体を用いて形態観察染色と免疫染色とを識別することができる生体物質検出方法を提供することにある。
病理切片から生体物質を特異的に検出する生体物質検出方法において、
蛍光標識体を用いて前記病理切片を免疫染色する工程と、
形態観察のための染色試薬を用いて前記病理切片を形態観察染色する工程と、
染色後の前記病理切片に励起光を照射して蛍光発光させ、前記病理切片から前記生体物質を検出する工程とを、有しており、

病理切片から生体物質を特異的に検出する生体物質検出方法において、
蛍光標識体を用いて前記病理切片を免疫染色する工程と、
形態観察のための染色試薬を用いて前記病理切片を形態観察染色する工程と、
染色後の前記病理切片に励起光を照射して蛍光発光させ、前記病理切片から前記生体物質を検出する工程とを、有しており、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、蛍光色素が有機物または無機物の粒子に内包され、かつ、励起波長が前記染色試薬の励起波長域とは異なる領域に存在する蛍光色素内包粒子を使用する生体物質検出方法が提供される。
病理切片から生体物質を特異的に検出する生体物質検出方法において、

前記病理切片を免疫染色する工程では、前記蛍光標識体として、蛍光体または半導体で組成された蛍光ナノ粒子が有機物または無機物の粒子に内包され、かつ、励起波長が前記染色試薬の励起波長域とは異なる領域に存在する蛍光ナノ粒子内包粒子を使用する生体物質検出方法が提供される。
本発明の好ましい実施形態にかかる生体物質検出方法は、病理切片から生体物質を特異的に検出する方法であり、基本的には(1)蛍光標識体を用いて病理切片を免疫染色する工程と、(2)形態観察のための染色試薬を用いて病理切片を形態観察染色する工程と、(3)染色後の病理切片に励起光を照射して蛍光発光させ、その病理切片から生体物質を検出する工程とを、有している。
特に(1)の病理切片を免疫染色する工程では、蛍光標識体として、励起波長が形態観察のための染色試薬の励起波長域とは異なる領域に存在する特殊な蛍光ナノ粒子、蛍光色素内包粒子または蛍光ナノ粒子内包粒子を使用するようになっている。
蛍光標識体の特性や種類、免疫染色、形態観察染色などの詳細は下記のとおりである。
なお、(1)の免疫染色する工程と(2)の形態観察染色する工程とでは、どちらの工程の処理が先に実行されてもよく、その順序(先後)は不問である。
HE染色に用いられるエオジンは顕微鏡観察条件によっては蛍光を放つ。エオジン吸収波長は多くの蛍光標識体の励起波長と重複するため、染色に用いたエオジンの発光が蛍光標識体の観察を妨害するという課題があった。エオジンの蛍光スペクトル(励起波長520nm)と励起スペクトル(蛍光波長540nm)を図1に示す。励起スペクトルより、エオジンは350nm未満の波長域および450nmを超えかつ550nm未満の波長域で効率よく励起されることがわかる。
従って、本実施形態にかかる蛍光標識体は、この波長域を回避した350~450nmの波長域かまたは550nm以上の長波長域で励起される蛍光標識体である必要がある。
また、本実施形態にかかる蛍光標識体の発光波長は、エオジンの吸収・発光の影響や組織自家蛍光との兼ね合いから550nm以上の長波長側であって、蛍光顕微鏡観察時に目視確認できる必要があるため、700nm以下の短波長側であることが好ましい。特に視感度の観点から、本実施形態にかかる蛍光標識体の発光波長は、好ましくは590~650nm(590nm以上で650nm以下)であり、より好ましくは590~630nm(590nm以上で630nm以下)である。
蛍光標識体の輝度が高い方が、ノイズであるエオジンの蛍光や細胞自家蛍光に対する信号値の比の観点から好ましい。従って、本発明の蛍光標識体としては、蛍光色素と比して輝度が高い、(A)蛍光ナノ粒子や(B)蛍光色素内包粒子、(C)蛍光ナノ粒子内包粒子が好適に用いられる。
本発明で用いられる蛍光ナノ粒子とは、粒子サイズが1~500nmのものであり、好ましくは10~200nmである。
蛍光ナノ粒子は半導体または蛍光体から構成される。
半導体は例えばII-VI族半導体であるZnSe、ZnTe、CdSe、CdTe、PbS、PbSe、PbTe等やII-VI族半導体であるAlAs、AlSb、GaP、GaAs、GaSb、InP、InAs、InSb等を用いることができ、毒性の観点から、GaPやInPを好適に用いることができる。
蛍光体は、例えば母体にY2O3やYVO4、ZnO、ZnS等を用い、発光中心にEuやNd等を用いることができる。
蛍光ナノ粒子の粒子サイズ、母体組成、不純物量を調整することで観察に適した励起波長とする。励起波長はエオジンの吸収波長と重複しないよう、350~450nmの波長域かまたは550nm以上の長波長域で励起される蛍光ナノ粒子である必要がある。
本発明で用いられる蛍光色素内包粒子とは、有機物または無機物でできた粒子に対し、蛍光色素が内包されてなるものである。
内包用の粒子は、例えば、ポリスチレン、ポリアミド、ポリ乳酸、ポリアクリロニトリル、ポリグリシジルメタクリレート、ポリメラミン、ポリウレア、ポリベンゾグアナミン、ポリフラン、ポリキシレン、フェノール樹脂、多糖、シリカ等であって、安定に蛍光体を内包できるものである。粒子原料であるモノマーに色素分子を結合させて粒子を合成する方法、粒子に色素を吸着させて導入する方法等、粒子への色素の導入はいかなる方法を用いても構わない。粒子サイズは10~500nmのものであり、好ましくは50~200nmである。
内包される蛍光色素は、エオジンの吸収波長と重複しないよう、350~450nmの波長域かまたは550nm以上の長波長域で励起する必要がある。
内包される蛍光色素としては、例えば、ローダミン系色素分子、Alexa Fluor(インビトロジェン社製)系色素分子、BODIPY(インビトロジェン社製)系色素分子、Texas Red系色素分子、スクアリリウム系色素分子、シアニン系色素分子、ローダミン系色素分子、オキサジン系色素分子、芳香環系色素分子、カルボピロニン系色素分子等を挙げることができる。

本発明で用いられる蛍光ナノ粒子内包粒子とは、有機物または無機物でできた粒子に対し、上記(A)で説明した蛍光ナノ粒子が内包されてなるものである。
粒子原料であるモノマーに蛍光ナノ粒子を結合させて粒子を合成する方法、粒子に蛍光ナノ粒子を吸着させて導入する方法等、粒子への蛍光ナノ粒子の導入はいかなる方法を用いても構わない。
内包される蛍光ナノ粒子の粒子サイズ、母体組成、不純物量を調整することで観察に適した励起波長とする。
励起波長はエオジンの吸収波長と重複しないよう、350~450nmの波長域かまたは550nm以上の長波長域で励起される蛍光ナノ粒子である必要がある。例えば前述の蛍光ナノ粒子の項に記載の蛍光体である。

免疫組織染色の方法としては蛍光染色法が好適に用いられる。
蛍光染色法は抗原部を蛍光標識体で染色する方法である。蛍光染色法以外の方法として例えば酵素を用いた色素染色法(DAB染色)があるが、これは感度の点で蛍光染色法に劣る。
染色の際には、蛍光標識体と1次抗体を直接結合した標識を作製し、抗原を染色する方法(1次抗体法)、蛍光標識体と2次抗体を直接結合した標識を作製し、抗原に1次抗体を結合したものを染色する方法(2次抗体法)、蛍光標識体とビオチンを直接結合した標識を作製し、抗原に1次抗体とアビジンあるいはストレプトアビジン修飾した2次抗体を結合したものを染色する方法(ビオチン-アビジン法またはサンドイッチ法)等を用いることができる。
染色に用いる1次抗体はいかなるものでも構わない。免疫組織染色を行ないたい対象によって変わる。例えばHER2抗原の染色を行なう場合には、抗HER2抗体を用いる。また、2次抗体は如何なるものを用いても構わない。1次抗体によって変わり、例えば抗マウス・ラビット・牛・ヤギ・羊・犬・チキンが挙げられる。
蛍光標識体と抗体やビオチンの結合は既存の如何なる方法を用いても構わない。例えば、アミンとカルボン酸の反応によるアミド化、マレイミドとチオールの反応によるスルフィド化、アルデヒドとアミンの反応によるイミン化、エポキシとアミンの反応によるアミノ化等を用いることができる。
なお、上記では組織染色について示したが、これに限定するものではなく、細胞染色にも使用可能である。
(サンプル1:蛍光ナノ粒子)
末端がアミノ基となっているPEG修飾CdSe/ZnS蛍光ナノ粒子(インビトロジェン社Qdot655)を抗体結合用の蛍光ナノ粒子として準備した。
一方、抗ヒトER抗体を1Mジチオスレイトール(DTT)で還元処理を行い、ゲルろ過カラムにより過剰のDTTを除去することによりシリカ粒子に結合可能な還元化抗体溶液を得た。
上記の抗体結合用の蛍光ナノ粒子と還元化抗体とを、EDTAを2mM含有したPBS中で混合し、1時間反応させた。10mMメルカプトエタノールを添加し、反応を停止させた。得られた溶液を遠心フィルターで濃縮後、精製用ゲルろ過カラムを用いて未反応抗体等を除去し、抗ヒトER抗体結合・蛍光ナノ粒子を得た。
サンプル1と同一の抗ヒトER抗体結合・蛍光ナノ粒子を作製した。
なお、後述の観察において、サンプル1に対しては励起波長375nm、サンプル2に対しては励起波長575nmで観察を行なった。サンプル1,2に対しては、同一の蛍光ナノ粒子を使用し、励起波長だけを変えて観察した。

それ以外はサンプル1と同様にして、抗ヒトER抗体結合・蛍光ナノ粒子を作製した。
蛍光色素CY5-SE(ロシュ社製)9.9mgと3-アミノプロピルトリメトキシシラン(3-aminopropyltrimetoxysilane、信越シリコーン社製、KBM903)3μLとをDMF中で混合し、オルガノアルコキシシラン化合物を得た。得られたオルガノアルコキシシラン化合物0.6mlを、48mlのエタノール、0.6mlのTEOS(テトラエトキシシラン)、2mlの水、2mlの28%アンモニア水と3時間混合した。上記工程で作製した混合液を10000Gで20分遠心分離を行い、上澄みを除去した。エタノールを加え、沈降物を分散させ、再度遠心分離を行った。同様の手順でエタノールと純水による洗浄を2回ずつ行った。得られたテトラメチルローダミン内包・シリカナノ粒子のSEM観察を行ったところ、平均粒径は104nm、変動係数は12%であった。
得られた蛍光体内包シリカナノ粒子を、EDTA(エチレンジアミン四酢酸)を2mM含有したPBS(リン酸緩衝液生理的食塩水)を用いて3nMに調整し、この溶液に最終濃度10mMとなるようSM(PEG)12(サーモサイエンティフィック社製、succinimidyl-[(N-maleomidopropionamid)-dodecaethyleneglycol]ester)を混合し、1時間反応させた。この混合液を10000Gで20分遠心分離を行い、上澄みを除去した後EDTAを2mM含有したPBSを加え、沈降物を分散させ、再度遠心分離を行った。同様の手順による洗浄を3回行うことで抗体結合用の蛍光色素内包粒子を得た。

蛍光色素としてTAMRA色素(PCC社製)を用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
蛍光色素としてTexasRed色素(シグマアルドリッチ社製)を用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
サンプル4の作製において、蛍光色素としてオキサジン170色素(シグマアルドリッチ社製)を用いるとともに、3-アミノプロピルトリメトキシシラン(3-aminopropyltrimetoxysilane、信越シリコーン社製、KBM903)に代えて3-グリシジルオキシプロピルトリメトキシシラン(3-Glycidyloxypropyltrimethoxysilane、TCI社製)を用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
オキサジン170(シグマアルドリッチ社製)2.5mgを水22.5mlに加えた後、ホットスターラ―上において60℃で20分間加熱し、ニカラックMX-035(日本カーバイド工業社製)1.5gを加え、さらに5分間加熱撹拌した。
その後、上記溶液にギ酸100μlを加え、60℃で20分間加熱攪拌し、室温放冷した。
冷却後、反応混合物を、遠心用チューブに入れて遠心分離機にセットし、12000rpmで20分間遠心分離し、その上澄みを除去した。
その後、上澄みを除去した反応混合物を、エタノールと水で洗浄した。
得られた粒子を、サンプル4と同様に、SM(PEG)12(サーモサイエンティフィック社製、succinimidyl-[(N-maleomidopropionamid)-dodecaethyleneglycol]ester)を用いて抗ヒトER抗体修飾を行ない、抗体結合用の蛍光色素内包粒子を得た。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
蛍光色素としてTexasRed色素(シグマアルドリッチ社製)を用いた。
内包用の粒子としてフェノール樹脂を用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
蛍光色素としてTexasRed色素(シグマアルドリッチ社製)を用いた。
内包用の粒子としてポリフランを用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。

それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。

それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
蛍光色素としてTexasRed色素(シグマアルドリッチ社製)を用いた。
内包用の粒子としてPS/アセトニトリル/GMA複合体を用いた。
それ以外はサンプル4と同様にして、抗ヒト抗体結合・蛍光色素内包粒子を合成した。
蛍光ナノ粒子としてCdSe/ZnSデカン分散液(インビトロジェン社Qdot655)10μLを準備し、テトラエトキシシラン40μLを混合した。これとは別に、エタノール4mL、14%アンモニア水1mLを混合し、室温下で撹拌した。ここに先ほどのCdSe/ZnSデカン分散液とテトラエトキシシランの混合液を加え、添加から12時間、撹拌を続け、蛍光ナノ粒子内包粒子を得た。反応液を10000Gで30分遠心分離を行ない、上澄みを除去した。これにエタノールを加えて再分散後、再度遠心分離を行ない、蛍光ナノ粒子内包粒子の洗浄を行なった。同様の手順でエタノールと純水による洗浄を1回ずつ行なった。得られた蛍光ナノ粒子内包粒子のSEM観察を行ったところ、平均粒径は120nm、変動係数は12%であった。

サンプル6と同一の抗ヒトER抗体結合・蛍光ナノ粒子内包粒子を作製した。
なお、後述の観察において、サンプル6に対しては励起波長375nm、サンプル7に対しては励起波長575nmで観察を行なった。サンプル6,7に対しては、同一の蛍光ナノ粒子内包粒子を使用し、励起波長だけを変えて観察した。

それ以外はサンプル6と同様にして、抗ヒト抗体結合・蛍光ナノ粒子内包粒子を作製した。

それ以外はサンプル6と同様にして、抗ヒトER抗体結合・蛍光ナノ粒子内包粒子を作製した。
サンプル1と同様の方法で、抗ヒトER抗体にCY5色素(インビトロジェン社製)を結合したものを作製した。
サンプル1と同様の方法で、抗ヒトER抗体にTAMRA色素(PCC社製)を結合したものを作製した。
サンプル1と同様の方法で、抗ヒトER抗体にFITC色素(PCC社製)を結合したものを作製した。
サンプル1と同一の抗ヒトER抗体結合・蛍光ナノ粒子を作製した。
なお、サンプル14に対しては、後述の観察時の励起波長を300nmに変更している。
サンプル1と同一の抗ヒトER抗体結合・蛍光ナノ粒子を作製した。
なお、サンプル15に対しては、後述の観察時の励起波長を500nmに変更している。

それ以外はサンプル1と同様にして、抗ヒトER抗体結合・蛍光ナノ粒子を作製した。
蛍光色素としてFITC色素(PCC社製)を用いた。
それ以外はサンプル4と同様にして、抗ヒトER抗体結合・蛍光色素内包粒子を合成した。
サンプル6と同一の抗ヒトER抗体結合・蛍光ナノ粒子内包粒子を作製した。
なお、サンプル18に対しては、後述の観察において励起波長300nmで観察を行なった。
サンプル6と同一の抗ヒトER抗体結合・蛍光ナノ粒子内包粒子を作製した。
なお、サンプル19に対しては、後述の観察において励起波長500nmで観察を行なった。
蛍光ナノ粒子としてCdSe/ZnSデカン分散液(インビトロジェン社Qdot565)を用いた。
それ以外はサンプル6と同様にして、抗ヒトER抗体結合・蛍光ナノ粒子内包粒子を作製した。
サンプル1~9,11~20を用いて、ヒト乳房組織の免疫染色と形態観察染色(HE染色)とを行なった。
染色切片はコスモバイオ社製の組織アレイスライド(CB-A712)を用いた。組織アレイスライドを脱パラフィン処理後、水に置換洗浄、10mMクエン酸緩衝液(pH6.0)中で15分間オートクレーブ処理することで、抗原の不活化処理を行った。抗原の不活化処理後の組織アレイスライドはPBS緩衝液を用いて洗浄後、湿潤箱中で1時間1%BSA含有PBS緩衝液を用いてブロッキング処理を行った。ブロッキング処理後、1%BSA含有PBS緩衝液で0.05nMに希釈した各サンプル1~9,11~20を組織切片と3時間反応させた。各サンプル1~9,11~20と反応後、組織アレイスライドを、PBS緩衝液を用いて洗浄した。
免疫染色した切片をマイヤーヘマトキシリン液で5分間染色してヘマトキシリン染色を行なった後、流水水洗(約45℃)を3分間行なった。次に、1%エオジン液で5分間染色してエオジン染色を行なった後、純エタノールに5分間つける操作を4回行ない、洗浄・脱水を行なった。続いてキシレンに5分間つける操作を4回行ない、透徹を行なった。最後に、封入剤エンテランニュー(Merck社製)を用いて封入し観察用サンプルスライドとした。
励起波長(nm)・蛍光波長(nm)は光学フィルターにより設定した(表1~表4では、光学フィルターの励起波長・蛍光波長の中心値を記している。)。顕微鏡画像取得時の露光条件は、各励起波長において、焦点付近でのトータルの照射エネルギーが50Jとなるようにした。
Image-Jを用いて取得画像より各画素の輝度を算出し、蛍光標識体で染色した部位(標識部)の平均輝度(標識部輝度)を算出した。当該平均輝度は信号値(S)に対応する。輝度は「0」を黒(一番暗い)と、「255」を白(一番明るい)としている。同時に、蛍光標識された細胞近傍の、蛍光標識されておらず且つエオジン染色された部位(エオジン染色部)についても平均輝度(エオジン染色部輝度)を算出した。当該平均輝度はノイズ値(N)に対応する。
標識部輝度とエオジン染色部輝度との比をS/N比として、S/N比が1.5以上であれば、標識部とエオジン染色部との判別が容易であったことから、S/N比1.5を判断値とした。
顕微鏡画像取得時には目視による蛍光観察も行ない、目視視認性の評価を行なった。
判別性(S/N比評価)については、S/N比が1.5以上で標識部とエオジン染色部の判別が容易となるので、1.5以上の場合を「○(適)」と、1.5未満の場合を「×(不適)」とした。
蛍光標識発光色視認性(目視視認性)については、目視による蛍光観察で見易いものを「○」と、見えにくいものを「△」と、全く見えないものを「×」とした。
Claims (10)
- 病理切片から生体物質を特異的に検出する生体物質検出方法において、
蛍光標識体を用いて前記病理切片を免疫染色する工程と、
形態観察のための染色試薬を用いて前記病理切片を形態観察染色する工程と、
染色後の前記病理切片に励起光を照射して蛍光発光させ、前記病理切片から前記生体物質を検出する工程とを、有しており、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、蛍光体または半導体で組成され、かつ、励起波長が前記染色試薬の励起波長域とは異なる領域に存在する蛍光ナノ粒子を使用する生体物質検出方法。 - 病理切片から生体物質を特異的に検出する生体物質検出方法において、
蛍光標識体を用いて前記病理切片を免疫染色する工程と、
形態観察のための染色試薬を用いて前記病理切片を形態観察染色する工程と、
染色後の前記病理切片に励起光を照射して蛍光発光させ、前記病理切片から前記生体物質を検出する工程とを、有しており、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、蛍光色素が有機物または無機物の粒子に内包され、かつ、励起波長が前記染色試薬の励起波長域とは異なる領域に存在する蛍光色素内包粒子を使用する生体物質検出方法。 - 病理切片から生体物質を特異的に検出する生体物質検出方法において、
蛍光標識体を用いて前記病理切片を免疫染色する工程と、
形態観察のための染色試薬を用いて前記病理切片を形態観察染色する工程と、
染色後の前記病理切片に励起光を照射して蛍光発光させ、前記病理切片から前記生体物質を検出する工程とを、有しており、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、蛍光体または半導体で組成された蛍光ナノ粒子が有機物または無機物の粒子に内包され、かつ、励起波長が前記染色試薬の励起波長域とは異なる領域に存在する蛍光ナノ粒子内包粒子を使用する生体物質検出方法。 - 請求項1~3のいずれか一項に記載の生体物質検出方法において、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、励起波長が350~450nmの波長域かまたは550nm以上の長波長域に存在する蛍光標識体を使用し、
前記病理切片を形態観察染色する工程では、前記染色試薬として、励起波長域が350nm未満および450nmを超えかつ550nm未満のエオジンを使用する生体物質検出方法。 - 請求項4に記載の生体物質検出方法において、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、発光波長が550~700nmの波長域に存在する蛍光標識体を使用する生体物質検出方法。 - 請求項4に記載の生体物質検出方法において、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、発光波長が590~650nmの波長域に存在する蛍光標識体を使用する生体物質検出方法。 - 請求項4に記載の生体物質検出方法において、
前記病理切片を免疫染色する工程では、前記蛍光標識体として、発光波長が590~630nmの波長域に存在する蛍光標識体を使用する生体物質検出方法。 - 請求項2に記載の生体物質検出方法において、
前記蛍光色素内包粒子の内包用の粒子が、ポリスチレン、ポリアミド、ポリ乳酸、ポリアクリロニトリル、ポリグリシジルメタクリレート、ポリメラミン、ポリウレア、ポリベンゾグアナミン、ポリフラン、ポリキシレン、フェノール樹脂、多糖のうちの1種以上の物質から構成される生体物質検出方法。 - 請求項2に記載の生体物質検出方法において、
前記蛍光色素内包粒子中の蛍光色素が、ローダミン系色素分子、BODIPY系色素分子、Texas Red系色素分子、スクアリリウム系色素分子、シアニン系色素分子、ローダミン系色素分子、オキサジン系色素分子、カルボピロニン系色素分子のうちの1種以上の物質から構成される生体物質検出方法。 - 請求項3に記載の生体物質検出方法において、
前記蛍光ナノ粒子内包粒子中の蛍光ナノ粒子が、半導体または酸化物蛍光体から構成される生体物質検出方法。
