Abstract

La présente invention concerne un processus de réaction de polymérase en chaîne (PCR) et un cycleur thermique. Dans le procédé, des échantillons biologiques sont contenus dans un porte-échantillon ayant une pluralité d'espaces d'échantillon ayant chacun des extrémités supérieure et inférieure et les échantillons sont séquentiellement chauffés et refroidis. Le cycleur thermique selon l'invention comprend des moyens de transfert thermique pour le chauffage et le refroidissement automatiques des échantillons dans le porte-échantillon, des moyens de fermeture pouvant être chauffés au-dessus des extrémités supérieures des espaces d'échantillon pour prévenir la condensation de vapeur d'échantillon pendant le processus, et des moyens pour ajuster un paramètre de processus, de préférence la température des moyens de fermeture pouvant être chauffés, en fonction d'au moins un autre desdits paramètres de processus. L'invention contribue à diminuer le nombre d'essais PCR défaillants, en particulier en raison de changements de volume d'échantillon.
