Abstract

Methods for hydrolyzing lignocellulose are provided, comprising contacting the lignocellulose with at least one chemical treatment. Methods for pretreating a lignocellulosic material comprising contacting the material with at least one chemical are also provided. Methods for liberating a substance such as an enzyme, a pharmaceutical, or a nutraceutical from plant material are also provided. These methods are more efficient, more economical, and less toxic than current methods.

5 treated JignoceliuloRe, and conuiclmg Sflid treated lignoceiluiose with at leusl one enzyme capable of hydrolyzing lignoceilulose, wherein said chumieal is selected from the group consiaύng uf oxidizing agents, denaturants, detergents, organic solvents, bases, and combinations thereof, wherein said moderate conditions compritie: a) a temperature from about \QaC to about 9O0C; 10 b) Ά pr.eHsuτιs tew Ui an about 2 atm; and, c) Li pH between about pPJ 4.0 and about pi I 10.0.
2. The method of claim I , wherein said chemical comprises an oxidizing agent selected from the group consisting of hydro gen peroxide, urea hydrogen
] 5 peroxide, benzoyl peroxide, a superoxide, potassium superoxide, a hypochlorite, hypochloiυiiti acid, chlorine, nitric acid, a peroxyacid, perαxyacetic acid, a pereulfate, n purcarbonate, a permanganate, osmium tetraαxide, chromium oxide, and sodium dodcoylbenucnesulfonate.

8. The method of claim I1 wherein said moderate conditions comprise a mmperature of about 80ΩC.
9. The method of claim I , wherein said moderate conditions comprise a 5 pH of about pW 5.0.
10. The method of claim J , wherein said contact occurs for about 24 hours.
1 J . The method of claim 1 , wherein said enzyme comprises at least oπu ] Q enzyme selected from the group consisting of ϋellulase, xylanasc, ligπinnse, amylase, glucuronidase, protease, lipase, and glucuronidase.
J 2. The method of claim 1 , wherein said temperature is adjusted Io be optimal for said enzyme prior to enzyme addition. 15
13, The method of claim 1 , wherein said pH is adjusted to be optimal for said enzyme prior to enzyme addition.
14. The method of claim 1 , wherein said chemical is removed prior to 0 addition of said enzyme.
15, The method of claim 1 , further comprising removal of said chemical from said lreaiud lignoccllulose prior to additional treatment to obtain a recycled chemical. 5
16. The method of claim 1 , wherein contacting said lignocelluloae with at least one chemical occurs simultaneously with contacting said lignoceiiuloøe with at least one enzyme capable ofhydrolyziπg lignocellulose.
0 17. The method of claim 1 , further comprising the addition of at least one fermenting organism, wherein said method results in Ihe production of ut least απts fermentation-based product.

18. The method of claim 17, wherein said product is selected from the group consisting of lactic acid, a fuel, an organic acid, an jndusiriαl enzyme, a pharmaceutical, and an amino acid.
19 A method for prctreαl ing a lignocellulosic material cornpriying coniactiπy said material with at least one chemical under moderate conditions to generate ι\ treated lignoccllulose, wherein .said chemical is selected from the group consisting of oxidizing agents, denaturαnts- detergents., organic solvents bases, and a combination Ihereof, wherein said moderate conditions eompri-.e: a) a lempeiature from aboiit 100C to about 90αC; b) a pleasure lea., than about 2 utm; and, c) u pH between about pH 4.0 and about pTT 10.0.

A meihod for liberating a gubslance from plnni malerial, comprising contacting said plant material with at least one chemical under the following conditions: i]) a temperature from about 100C to about 9O0C; b) a pressure less than about 2 atm; and, c) a pH between abuui pH 4.0 and about pH 10.0, jo generate a iroated plant material, wherein said chemical is selected from ihe group consisting of oxidizing agents, denaUirants, detergents, organic solvents, bases, and a combination thereof
21 , The meUiod of claim 2O1 farther comprising contacting said treated plant materia] with at least one enzyme capable of hydrolyzing lignocclliilose.
22, The method of claim 2] , wherein said plant material comprises at least one enzyme capable of hydrolyzing ligπoeelUilose.

5 24. The method of claim 23, comprising incubating said plant material under conditions that allow expression of said enzyme capable of hydrojyzmg lignocelUiIoae prior to contacting said plant material with said chemical.

2S. A niυthod for hydrolyzing lignocellulose, comprising contacting said 5 Hgnoceiluiose with a bases at a pH of aboqi 9.0 to about 14.0 to generate a treated ligπocisllulose, and contacting said trented lignocelluloae with at leasi one enzyme capable of hydrolyuing lignocelHilosc.
29. The method of claim 2S5 wherein said base is sodium carbonate or 0 potassium hydroxide.
