Abstract
本発明は、動物の分化細胞を大量かつ簡便に作製する手法、及び細胞の分化を調節する手法を提供する。 本発明の分化細胞の作製方法は、以下の工程:(a)血管内皮由来細胞を増殖因子を含む培地中で培養する工程、及び(b)培養した細胞の培地から増殖因子を除き、又は上記の増殖因子活性を阻害した状態で、さらに細胞を培養する工程、を含み、他の種の細胞への分化能力を有する細胞及び/又は他の種の細胞に分化した細胞を得ることを特徴とする。

本発明は、 血管内皮由来細胞から各種細胞への分化能力を有する細胞又は各種 細胞に分化した細胞を作製する方法、 及び該書方法により作製された分化細胞に関 する。 また本発明は、 血管内皮由来細胞を各種細胞への分化能力を有する細胞又 は各種細胞に分化した細胞に分化させる方法、 及びその分化を調節する方法に関 する。
背景技術 組織補填等の再生医療や遺伝子治療など、 先端的医療におけるヒト分化細胞の 必要性は極めて高い。 また、 ヒトの生体分子生産に用いうる分化細胞の必要性も 高い。 しかし、 このような必要性を満たす細胞を供給するには、 原理的及ぴ現実 的な困難性がある。 すなわち、 必要とされる細胞が生存中の他人又は死亡直後の 他人から供与される場合、 あるいは、 必要とされる細胞として他人の細胞から作 出された胎生幹細胞を用いる場合には、 倫理的な問題、 細胞の数が不足する問題 、 免疫的に拒絶される問題などの複数の困難性が存在する。 そのため、 上述した ような問題を解決し、 必要なヒト分化細胞を作製する方法を開発することが、 当 技術分野において大きな課題となっている。 上述したような分化細胞を得る方法に関して、 現在までにいくつかの報告があ
る。 例えば、 ゥシ生体由来の血管内皮細胞が平滑筋細胞に分化することが報告さ れている(Frid, M.G. et al., Circulation Research ( USA) , 2002年,第 90卷, p . 1189-1196 )。 しかしながら、 この系においてはごく少量 (約 0 . 0 1 ~ 0 . 0 3 % ) しか 分化細胞を得ることはできず、 実際的な使用には不十分である。
また、 マウス胚性幹細胞 (E S細胞) から、 血管内皮細胞と平滑筋細胞に分化 する能力を有する幹細胞が分離されている (Yamashita, J. et al., Nature ( UK), 2000 年, 第 408卷, p . 92-96) が、 E S細胞をヒトの医療に応用するには制約が多く、 現時点で現実的ではない。
さらに、 ゥシ大動脈由来血管内皮細胞が、 トランスフォーミング増殖因子/? ( T G F—/? ) ファミ リー、 特に T G F— /? 1の刺激によって平滑筋類似の細胞に 分化したことが報告されている (Aiciniegas, E. et al., Journal of cell science ( UK), 1992 年, 第 103卷, p . 521-529)。 しかし、 この系においては平滑筋細胞のごく初期 の分化段階の細胞のみが得られているにすぎない。 従って、 完全に分化した平滑 筋細胞を得るには、 まだ不十分なものであり、 ヒ トの再生医療には利用できない と考えられる。
従って、 動物、 特にヒ トの分化細胞を、 大量かつ簡便に作製する手法が望まれ ていた。 発明の開示
本発明は、 動物の分化細胞を大量かつ簡便に作製する手法、 及び細胞の分化を 調節する手法に関する。
本発明者らは、 上記課題を解決すべく鋭意研究を重ねた結果、 初代ヒ ト臍帯静
脈血管内皮由来細胞をあらかじめ増殖因子の 1種である F G Fとゥシ胎児血清で 刺激した状態で単層培養して増殖させ、 次いで F G Fを除きゥシ胎児血清のみで 刺激した状態で培養を続けた結果、 細胞の形態が著しく変化することに着目し、 この細胞の形態変化と細胞分化との関連性について明らかにするため、 各種細胞 分化マーカーの発現を解析したところ、 平滑筋細胞、 骨芽細胞及び脂肪細胞に特 異的に認められる遺伝子発現が上昇することを見出した。 従って本発明者は、 血 管内皮由来細胞を増殖因子添加培地中で培養した後に、 増殖因子不含培地中で培 養することにより、 各種細胞に分化可能な細胞を作製することができるという知 見を得、 本発明を完成するに至った。
すなわち本発明は、 分化細胞の作製方法であって、 以下の工程:
( a ) 血管内皮由来細胞を増殖因子を含む培地中で培養する工程、 及び
( b ) 培養した細胞の培地から増殖因子を除き、 又は上記の増殖因子活性を阻害 した状態で、 さらに細胞を培養する工程、
を含み、 他の種の細胞への分化能力を有する細胞及び/"又は他の種の細胞に分化 した細胞を得ることを特徴とする、 上記作製方法である。
上記作製方法において、 血管内皮由来細胞としては、 臍帯静脈血管内皮由来細 胞を用いることが好ましい。 また血管内皮由来細胞は、 哺乳動物、 特にヒ トから 採取されたものであることが好ましい。 , また上記作製方法において、 増殖因子は繊維芽細胞増殖因子 (F G F ) フアミ リーに属する因子とすることができる。 この繊維芽細胞増殖因子としては、 酸性 繊維芽細胞増殖因子 (F G F— 1 ) が挙げられるが、 それに限定されるものでは ない。
さらに上記作製方法において、 他の種の細胞としては、 平滑筋細胞、 骨芽細胞 及び脂肪細胞が挙げられる。
また本発明は、 上記作製方法により得られる分化細胞である。
本発明はまた、 上記作製方法に従って培養した細胞を、 さらに増殖因子添加培 地中で培養し、 血管内皮細胞に再分化させることを特徴とする、 血管内皮細胞の 作製方法である。
さらに本発明は、 血管内皮由来細胞を分化させる方法であって、 以下の工程:
(a) 血管内皮由来細胞を増殖因子を含む培地中で培養する工程、 及び
(b) 培養した細胞の培地から増殖因子を除き、 または上記の増殖因子活性を阻 害した状態で、 さらに細胞を培養する工程、
を含み、 他の種の細胞への分化能力を有する細胞及び/又は他の種の細胞に分化 した細胞を得ることを特徴とする、 上記分化方法である。
上記分化方法において、 増殖因子が繊維芽細胞增殖因子 (FGF) ファミ リー に属する因子とすることができる。 この繊維芽細胞増殖因子としては酸性繊維芽 細胞増殖因子 (FGF— 1) が挙げられる。
上記分化方法において、 他の種の細胞としては、 平滑筋細胞、 骨芽細胞及び脂 肪細胞が挙げられるが、 それに限定されるものではない。
また上記分化方法の工程 (b) において、 增殖因子不含培地に別の因子を添加 することにより、 細胞の分化を増強させる又は抑制することができる。 ここで例 えば別の因子としてトランスフォーミング増殖因子/? (TGF—^) ファミ リ一 に属する因子又は MAPキナーゼキナーゼ (MEK) 阻害物質を用いた場合には 、 細胞の分化が増強される。 トランスフォーミング増殖因子 としてはァクチビ
ン Aが挙げられる。 また MAPキナーゼキナ一ゼ (ME K) 阻害物質としては P D 9 8 0 5 9が挙げられる。 さらに例えば別の因子としてトランスフォーミング 増殖因子/? (T G F - ?) 阻害物質を用いた場合には、 細胞の分化が抑制される 。 トランスフォーミング增殖因子/? ( T G F - J3 ) 阻害物質としてはァクチビン 結合夕ンパク質及び s m a d 7夕ンパク質が挙げられる。 図面の簡単な説明

発明を実施するための最良の形態
以下、 本発明の実施の形態について詳細に説明する。
本発明は、 分化細胞の作製方法及び血管内皮由来細胞を分化させる方法に係る ものであり、 血管内皮由来細胞を増殖因子を含む培地中で培養し、 その後、 培養 した細胞を増殖因子を含まない培地中で培養することにより、 又は増殖因子活性 を阻害した状態でさらに培養することにより、 分化細胞 (すなわち、 他の種の細 胞への分化能力を有する細胞及びノ又は他の種の細胞に分化した細胞) を得るこ とを特徴とするものである。
1 . 血管内皮由来細胞の調製
本発明においては血管内皮由来細胞を培養し、 分化細胞を得る。 本発明におい て 「血管内皮由来細胞」 とは、 血管の内壁を酵素処理又は物理的処理することに より剥離する細胞を指し、 例えば、 静脈内皮由来細胞 (臍帯静脈内皮由来細胞等 ) などが挙げられる。
上述した細胞の供与源は、 動物であれば特に限定されるものではなく、 例えば 哺乳動物、 特にヒトが好ましい。 上述した細胞の調製方法は、 当技術分野で周知 の細胞単離法に従えばよく、 特に限定されるものではない。
例えば臍帯静脈内皮由来細胞は、 出生時に入手可能となる臍帯の静脈内皮をト リプシン液処理する.ことにより、 液内に剥がれ落ちる細胞の集団中に含.ま.れる.も のとして採取することができる。
2 . 細胞培養
上記 「 1 . 血管内皮由来細胞の調製」 に記載のように得られた細胞は、 最初に 増殖因子存在下にて培養する。 ここで増殖因子存在下とは、 物質的に解明された
増殖因子を培養液に添加すること、 及び増殖因子活性を有する血清などを添加す ることを指す。 本発明において 「増殖因子」 とは、 細胞 '組織 ·臓器 '個体など の数 ·重量 ·体積などを増加させる作用を有するタンパク質を指し、 限定するも のではないが、 繊維芽細胞増殖因子 (FGF)、 上皮増殖因子 (E GF)、 トラ ンスフォーミング增殖因子 (T GF) (例えば T GF—/?等) などが挙げられる 。 本発明においては、 増殖因子として: FGFを用いることが好ましく、 この FG Fとしては、 例えば酸性繊維芽細胞増殖因子 (F GF— 1又は a F GF)、 塩基 性繊維芽細胞増殖因子 (FGF— 2又は b F GF)、 角質細胞増殖因子 (FGF — 7又は KGF) などが知られている。 特に F G F— 1又は F G F— 2を用いる ことが好ましい。
細胞培養に使用する培地としては、 血管内皮由来細胞を培養するために一般的 に使用されている培地であれば特に限定されるものではなく、 例えば、 M l 99 培地 ( S i gma製)、 D MEM培地 ( S i gma製)、 MCD B培地 ( S i g ma製)、 MCDB 15 1培地 ( S i gma製) などが挙げられる。
培地に添加する増殖因子濃度は、 使用する増殖因子により異なり、 当業者であ れば適宜設定することが可能である。 例えば F GFを用いる場合には、 F GF濃 度は、 約 1 0~3 ng/ml、 好ましくは約 5 n gZm 1とする。
培地には、 血管内皮由来細胞を培養する.ために適切なその他の成分を添加して もよい。 例えば、 ゥシ胎児血清、 へパリン、 カナマイシン (抗生物質) などを添 加することが当技術分野で公知である。 .
培養条件は、 血管内皮由来細胞を培養するための当業者に公知の条件を採用す ることができ、 例えば、 約 37°Cにて、 約 1 0〜40日間、 好ましくは 1 4〜 3
5日間にわたり細胞を培養する。
好ましい実施形態においては、 血管内皮由来細胞を、 F G F— 1 ( 5 n g / m 1 ) 及びへパリン ( 1 0 g / m 1 ) 又は F G F— 2 ( 5 n g / m 1 ) を含むゥ シ胎児血清 ( 1 5 % ) 添加 M 1 9 9培地で、 3 7 °Cにて細胞密度が subconfluent の状態になるまで単層培養する。 增殖因子存在下にて、 血管内皮由来細胞は血管 内皮細胞の性質を有したまま増殖をく り返す。
続いて、 上述のように増殖因子含有培地中で培養した細胞を、 增殖因子を除去 した培地中又は増殖因子活性を阻害した状態で培養する。 増殖因子活性の阻害は 、 増殖因子とその受容体との結合阻害剤、 増殖因子又はその受容体に結合する中 和抗体、 増殖因子の特異的吸収剤、 増殖因子の細胞内シグナル伝達の阻害剤など を用いて行うことができるが、 これ以外の方法によってもよい。 このような増殖 因子活性の阻害は当業者に周知であり、 上述した結合阻害剤、 中和抗体、 特異的 吸収剤及び細胞内シグナル伝達阻害剤なども当業者であれば適宜選択することが できる。 培養期間は、 約 4日〜 3週間、 好ましくは 2〜 3週間である。 またその 他の培養条件は、 上記培養条件と同様である。
本発明者は、 上述したように培養した血管内皮由来細胞は、 増殖因子を除去し た培地中又は増殖因子活性を阻害した状態で長期間培養した後、 平滑筋細胞、 骨 芽細胞及び脂肪-細胞に特異的なマーカー遺伝子を発現する細胞に変化する..ことを 確認した (実施例 2 ~ 4、 8及び 9参照)。 従って本発明により、 血管内皮由来 細胞を分化させて、 他の種の細胞への分化能力を有する細胞及び/又は他の種の 細胞に分化した細胞を得ることができる。
3 . 分化細胞
以上のように、 本発明の培養技術により、 血管内皮由来細胞を分化させて分化 細胞を作製することができる。 本発明において 「分化細胞」 とは、 他の種の細胞 への分化能力を有する細胞、 及び他の種の細胞に分化した細胞を指す。 ここで 「 他の種の細胞」 とは、 血管内皮細胞以外の細胞を指し、 限定するものではないが 、 平滑筋細胞、 骨芽細胞及び脂肪細胞などが挙げられる。 また 「他の種の細胞へ の分化能力を有する細胞」 とは、 作製した細胞が、 他の種の細胞としての性質 · 機能を有しているが、 なおも分化前の血管内皮由来細胞の性質 ·機能を有してい るものを指す。 一方 「他の種の細胞に分化した細胞」 とは、 作製した細胞が、 他 の種の細胞としての性質 ·機能のみを有し、 分化前の血管内皮由来細胞の性質 - 機能は保持していないものを指す。
本発明に従って F G F不含培地において 3週間培養した細胞は、 ェンドセリン - 1 ( E T— 1 ) の刺激により収縮することから、 平滑筋細胞としての機能を有 していると考えられた (実施例 6参照)。 またこの細胞は、 平滑筋細胞マーカー 及び血管内皮細胞マーカーの両者を発現することから (実施例 2及び 3参照)、 これらの細胞は、 平滑筋細胞への分化能力を有する細胞といえる。
血管内皮由来細胞 (例えばヒト臍帯静脈血管内皮由来細胞) を、 増殖因子 (好 ましくは F G F ) 添加培地中で培養した場合、 その増殖能が非常に高いことから 、 本発明に従って、 他の種の細胞への分化能力を有する細胞及び他の種の細胞に 分化した細胞を簡便かつ大量に作製することが可能となる。 従って、 その分化細 胞を用いることにより、 再生医療などにおいて有用な細胞を大量に得ることがで きる。 また本発明において最も好ましい血管内皮由来細胞である臍帯静脈血管内
皮由来細胞は、 液体窒素中に長期保存可能であることから、 出生時に臍帯静脈血 管内皮由来細胞を採取しておけば、 将来的に何らかの病的原因により各種細胞 ( 例えば平滑筋細胞、 骨芽細胞及び脂肪細胞) の再生が必要になった際に保存細胞 を取り出して、 増殖及び分化させて使用することができる。 その結果、 自己から 取り出した細胞を分化させた後に体内に戻すことができるため、 免疫学的な問題 が少ないことが期待される。
4 . 分化の調節
( 1 ) 血管内皮細胞への再分化
本発明により分化した細胞を、 再度增殖因子添加培地中で培養することにより
、 血管内皮細胞に再分化させることができる。 この際、 用いる増殖因子は、 F G F— 1又は F G F— 2を用いることができるが、 それ以外の増殖因子であっても 、 血管内皮細胞への再分化を誘導可能な活性を有するものであれば用いることが できる。
このように一度他の種の細胞に分化した細胞を血管内皮細胞に再分化させるこ とにより達成される効果 ·利点としては以下のものがある :
•再生医療などの目的のために最初に十分な量として準備した分化細胞よりも実 際には多くの分化細胞が必要であると判断された場合には、 分化細胞を上記再分 化操作に.より血管内皮細胞に戻してから必要な量が得られるまで培養 ··.増殖させ 、 その後、 再度分化細胞に誘導して用いることが可能である。 この場合、 分化細 胞を増殖させるよりも、 血管内皮細胞を増殖させたほうが増殖率がよく、 また培 養も簡便である。
•動脈硬化などの生体の疾病として、 血管内皮細胞が平滑筋細胞などの他の種の
細胞に分化して不利益な性質を有することになる可能性が考えられるが、 本発明 に従って他の種の細胞の性質を有する細胞を血管内皮細胞に再分化させることが できれば、 疾病の治療 (改善) に役立つものと期待される。
(2 ) 分化の増強及び抑制
本発明に従って増殖因子不含培地中で培養する工程において、 他の種々の因子 を添加することにより、 細胞の分化を增強又は抑制することが可能である。 例えば、 他の因子としてトランスフォーミング増殖因子 (T G F -/?) に属 する因子又は MAPキナーゼキナーゼ (MEK) 阻害物質を使用した場合には、 血管内皮由来細胞から他の種の細胞を誘導可能な細胞への分化を増強することが できる。 TGF— /?ファミ リーに属する因子としては、 限定するものではないが 、 ァクチビン、 骨形成因子 (BMP) などが挙げられる。 本発明においては、 ァ クチビン、 特にァクチビン Aを用いることが好ましい。
MEK阻害物質としては、 限定するものではないが、 PD 98059、 U 0 1 26などが挙げられる。
また例えば、 他の因子として T GF— 阻害物質を使用した場合には、 分化を 抑制することができる。 T GF— /?阻害物質としては、 その阻害作用機構に限定 されることはなく、 例えばァクチビン結合タンパク質、 Smad 7タンパク質、 ァクチビン中和抗体などが挙げられる。..,本発明においては、 ァクチビン結合タン パク質として特にフォリス夕チン、 及び S mad 7タンパク質を用いることが好 ましい。
上述したように、 他の種の細胞への分化を調節することにより達成される効果 '利点としては以下のものがある :
•再生医療などの目的で、 分化した細胞の性質と分化前の細胞の性質の両者を有 するような中等度の分化細胞が必要であると判断されるような場合には、 本発明 に従って分化を調節する技術が有効であると考えられる。
•動脈硬化などの生体の疾病として、 血管内皮細胞が平滑筋細胞などの他の種の 細胞に分化して不利益な性質を有することになる可能性が考えられるが、 本発明 に従って他の種の細胞への分化を調節することができれば、 疾病の予防 · 治療 ( 改善) に役立つものと期待される。 本発明を以下の実施例により具体的に説明する。 但し、 本発明はこれらの実施 例によりその技術的範囲が限定されるものではない。
〔実施例 1〕 ヒト臍帯静脈血管内皮由来細胞の培養
ヒト臍帯静脈血管内皮由来細胞 (HUVE— D C) は、 既に報告されている H UVE C単離手法により単離した (Imamura, Τ·, and Mitsui, Y. (1987) Exp. Cell Res. 172, 92- 100)。 すなわち、 ヒト臍帯静脈血管内壁を洗浄後、 0 . 2 5 %トリプシン 含有リン酸緩衝溶液を注入し、 室温にて 1 5分静置することにより内皮細胞を細 胞壁より剥離させ回収した。
上記の通り単離した細胞を I型コラーゲンで被覆したプラスチック製培養ディ ヅシュ中に接種し、 1,5 %ゥシ胎児血清 (FB S ; F i l t r o n) を添加し、.— 5 n g/m 1 F G F - 1及び 1 0 n g/ 1へパリン ( S i gma) を添加し た又は添加しない M 1 9 9培地 ( S i gma) 中で培養した。
全実施例における細胞分化実験では、 いずれも一度 F GF添加培地で細胞を培 養し、 subconfluentの状態まで増殖させた後に F G F無添加培地にて培養した。
ここで使用した FGF— 1は遗伝子組換え手法により作製したものであり、 こ の'組換え F GF— 1は、 大腸菌 B L 2 1 (DE 3) pLy s S細胞において p E T— 3 C系 (Imamura, T. et al., ( 1990) Science 249, 1567-1570) を利用して発現させ た。 また得られた F G F— 1は、 へパリン一セファロ一スァフィニティークロマ トグラフィ一により均質に精製することにより調製した (Imamura, T. et al. ( 1990) Science 249, 1567-1570)。 へパリン (S i gma製) の添加は、 F G F— 1活性の 最適化のために行い、 細胞の分化に影響はなかった。 得られた細胞を以下の試験 において使用した。
〔実施例 2〕 免疫細胞化学試験
本実施例においては、 実施例 1において培養した細胞を用いて免疫細胞化学試 験を行った。
実施例 1において培養した細胞を、 血管内皮細胞マーカーであるフォンウィル ブランド因子 ( vWF) に対するポリクローナル抗体 (D a k o製)、 及び平滑 筋細胞マーカーである力ルポニンに対するモノクローナル抗体 (クローン h C P ) ( S i gma製) を一次抗体として用いて二重標識し、 その後、 それそれテキ サスレヅ ドコンジユゲートロバ抗ゥサギ I g G (Ame r s h a m製) 及び F I T Cコンジュゲートャギ抗マウス I (Amer s ham製) を用いて二次標 '-. 識した。 続いて、 共焦点レーザー走査顕微鏡 (Carl Zeiss, Jena, .Germany) を用いて シグナルを検出した。
結果を第 1図 a及び bに示す。 第 1図 aは、 F G F添加 ( + ) および F G F除 去後 3週目 (-) のヒト臍帯静脈内皮由来細胞の顕微鏡像を示す。 FGF除去後 3週目には平らで大きな細胞が出現していることがわかる。 また第 1図 bは、 F
GF添加(+ )および FGF除去後 3週目 (―) のヒ ト臍帯静脈内皮由来細胞を、 それそれ血管内皮細胞マーカーである抗 vWF抗体 (赤)、 及び平滑筋細胞マー カーである抗塩基性力ルポニン抗体 (緑) で同時染色した像を示す。 FGF除去 後 3週目において、両者の平滑筋細胞マーカーを発現している細胞が確認できた。 〔実施例 3〕 平滑筋細胞マーカー及び血管内皮細胞マーカーの発現確認 (RT— P C R/サザンプロッ ト分析)
本実施例においては、 実施例 1において培養した細胞を用いて、 平滑筋細胞マ 一力一の発現を、 RT— P CR/サザンプロッ ト法を利用して調べた。
半定量的 R T— P サザンプロヅ ト分析を、 既に記載されている通りに行 つた (Ozawa, K. et al. (1997) Brain Res. Protocol 1, 211-216)。 簡単に説明すると、 細 胞から I s o gen (Nippon Gene) を用いて総 R N Aを単離した後、 l〃g:のサ ンプルを、 M— MLV逆転写酵素 (GIBCO BRL) を製造業者の説明書に従って用 いて逆転写した。 4 %の逆転写混合物を用いて、 試験遺伝子の c DNA断片を、 その直線状部分を AmpliTaq Gold (PE Applied Biosynthesis) を用いて P CRにより 増幅した。 使用した平滑筋細胞マーカー及び血管内皮細胞マーカーに特異的なプ ライマーを表 1に示す。 表中、 血管内皮細胞マーカ一である 「 vWF」 はフォン ウィルブラン ド因子、 「P.E CAM— 1」 は血小板/血管内皮細胞固着分子を表 し、 平滑筋細胞マーカーである に S M.2 2 α」 はァクチン結合タンパク質、 「Ρ GE S」 はプロスタグランジン E合成酵素、 「力ルポニン」 はァクチン結合タン パク質を表す。 「GAPDH」 は、 内部標準として用いた。 また、 プライマー配 列の項目の 「F」 及び 「R」 はそれそれフォワードプライマー及びリバースプラ イマ一を表す。 サイクルの項目に記載の数値は、 当該プライマーセッ トを用いて
行う P C Rサイクル数を表し、'また断片の項目に記載の数値は、 P CRにより得 られる断片長を表す。

続いて、 P C R産物のァリコートを 1. 0 %ァガロースゲル上で分離させ、 そ れを Hy b o n d N +膜 ( Amersham-Pharmacia) にトランスファ一した。 続いて この膜を、 ジゴキシゲニン標識 dUT P (DIG-ffigh Prime, Roche) により標識した 対応する c DNAプローブとハイブリダィズさせた。 N I H I ma g eソフト ウェア (version 1.62) を用いてシグナルを可視化、 統合、 及ぴ定量した。
結果を第 2図 a ~ dに示す。 第 2図 a及び" bは、 各平滑筋細胞マーカー (a ) 及び各血管内皮細胞マーカー (b ) の発現を示す。 またこの結果をグラフ化した ものがそれぞれ第 2図 c及び dである。 第 2図 a及び cより、 0 ?除去後 1 ~ 2週経過後の細胞においては、 平滑筋細胞マーカーの発現が経時的に上昇してい ることがわかる。
〔実施例 4〕 ヒト臍帯静脈内皮由来細胞の細胞クローンを用いた試験
本実施例においては、 ヒト臍帯静脈内皮由来細胞の細胞クローンを培養し、 平 滑筋細胞に分化する能力について試験した。
ヒト臍帯静脈内皮由来細胞の細胞クローン C L 1及び C L 1 4は、 実施例 1 と 同様に F G F添加培地で培養した後、 F G F無添加培地で 2週間培養した。 その 後、 実施例 2と同様に血管内皮細胞マーカーに対する抗体と平滑筋細胞マーカー に対する抗体とを用いて同時染色した。 また、 実施例 3と同様に平滑筋細胞マー カーである力ルポニン遺伝子の発現を調べた。
その結果を第 3図 a ~ cに示す。 第 3図 aは、 血管内皮細胞マーカーに対する 抗体と平滑筋細胞マーカーに対する抗体とを用いた染色結果であり、 この写真か らヽ C L 1 4細胞においては平滑筋細胞マーカーのみが確認された。 また第 3図 b及び cは、 力ルポニン遺伝子の発現を経時的に調べた結果を示し、 C L 1細胞 及び C L 1 4細胞の両者において.力ルポニンの有意な発現増大が確認されたが、 細胞クローンごとに、 平滑筋細胞分化能力に差があることが判明した。
〔実施例 5〕 内皮細胞への再分化
本実施例においては、 実施例 2及び 3において確認した、 分化した平滑筋細胞 から再度内皮細胞を分化させることを確認した。
実施例 1と同様の条件下にて、 ヒト臍帯静脈内皮由来細胞を F G F無添加培地 で 4日間培養した後、 さらに F G F添加培地又は F G F無添加培地において 4日 間培養した。 その後、 実施例 3と同様の手法にて細胞から mRNAを抽出し、 R T— P CR/サザンブロヅ ト法で平滑筋細胞特異的マーカー遺伝子 (力ルポニン 及び SM 22 ) の発現を調べた。
結果を第 4図に示す。 第 4図において、 各平滑筋細胞のマーカーの発現は、 F G F除去後 (―) に経時的 (4日、 8日) に増加したが、 F GF除去後 4日目に F GFを再度添加しさらに 4日間培養したもの (+ ) については、 各マーカー遺 伝子の発現は F G F除去前のレベル近くにまで減少していた。 すなわち、 FGF 除去により平滑筋細胞に分化した細胞が、 再度 FGFを添加することによって、 再ぴ内皮細胞に分化したことがわかった。
〔実施例 6〕 平滑筋細胞の収縮
本実施例においては、 FGF除去培養したヒ ト臍帯静脈内皮由来細胞の収縮能 を試験した。
最初に、 F G F— 1 +へパリンの存在下又は不在下にて、 I型コラーゲンで被 覆したプラスチック製培養ディッシュ上でヒト臍帯静脈内皮由来細胞を 3週間培 養した後、 その細胞を 1 0mM 8—プロモー c GMP (C ALB I O CHEM 製) と共に.37.°Cにて 20分間インキュベートして弛緩させ、 24ゥエルプレー ト上でコラーゲンゲル中に包理した。 この際、 ゲル中の 8—ブロモ一 c GMPの 最終濃度は 1 mMであった ( 0. 75 m lゲル/ゥエル中、 2. 4 X 1 0 5細胞 コラーゲンゲルは、 Cellmatrix type I-P ( 0. 3 %ブ夕 I型コラーゲン ; Nitta-gelatin
Corp., Osaka, Japan) を製造業者の指示に従って用いて調製した。
簡単に説明すると、 1 0 XM 1 9 9培地、 1 XM 1 9 9培地中の細胞懸濁液、 重炭酸バッファー、 ゥシ胎児血清 (F B S)、 及び Cellmatrix type I-P溶液を、 1 : 1 : 1 : 1 . 5 : 5. 5の比率で混合し、 ゥエルに注入して、 重合させた。 得られる SMC封入ゲルを、 0. 5 m lの 1 5 % F B S含有 M 1 9 9で被覆し た。 ゲルがゥエルの底から分離する 1 7時間の時点までゲルをインキュベートし た後、 エンドセリン (E T— 1 ; CAL B I O C H EM製) ( 2 5 0 nM) を 3 0分毎に培養物に添加した。 定量に閧しては、 各ゲル片を 5 0 0 1の培養液を 含む 1. 5 m 1マイクロチューブに移し、 遠心し、 その体積を測定した。
その計測結果を第 5図に示す。 第 5図は、 F G F添加 (F) 又は無添加 (D ) で培養した細胞の E T - 1刺激による収縮率をグラフとして示している。 F G F 除去後 3週目のヒ ト臍帯静脈内皮由来細胞は、 E T— 1刺激により、 約 2 ひ%の ゲル収縮を引き起こしている。 図中、 星印はスチューデントの T試験にて: p値< 0. 0 5で有意差があることを示す。 この結果から、 F GF除去後 3週目のヒト 臍帯静脈内皮由来細胞の中には、 平滑筋細胞の特徴を有する細胞が含まれている ことがわかる。
〔実施例 7〕 分化の調節
本実施例においては、 ヒ ト臍帯静脈内皮由来細胞の平滑筋細胞への分化の調節 について試験した。
( 1 ) T G F— ^ファミ リーによる分化増強
ヒト臍帯静脈内皮由来細胞を実施例 1 と同様に F G F添加又は無添加で Ί日間 培養した。 またこれらの細胞に対し、 T G F— 5ファミ リ一に属する増殖因子の
1種であるァクチビン A (R&D S y s t ems , I n c . 製) 1 0 0〃g mlを投与し、' その作用を調べた。 実施例 3と同様に、 平滑筋細胞マーカー遺伝 子 (力ルポニン、 SM 22 ) 及び血管内皮細胞マ一カー遺伝子 (vWF) の発 現を RT— P CR/サザンプロッ ト法により調べた。 また実施例 2と同様に、 F GF除去後のヒ ト臍帯静脈内皮由来細胞を抗 vWF抗体及び抗カルポニン抗体で 同時染色した。
結果を第 6図 a及び bに示す。 第 6図 aは、 F G F添加 (+ ) 又は FGF無添 加 (一)、 及びァクチビン A添加 (+ ) 又は無添加 (―) で 7日間培養した後の 各マーカー遺伝子の発現を示している。 第 6図 bは、 抗 vWF抗体及び抗カルポ ニン抗体で同時染色した細胞の写真を示す。これらの結果から、 ァクチビン Aが、 ヒト臍帯静脈内皮由来細胞の F G F除去後の平滑筋細胞への分化を増強すること がわかる。
(2) ァクチビン結合タンパク質による分化抑制
ヒト臍帯静脈内皮由来細胞を実施例 1と同様に F G F添加又は無添加で 5日間 培養した。 またこれらの細胞に対し、 ァクチビン Aの阻害剤であるァクチビン結 合タンパク質の 1種であるフォリス夕チン (R&D S y s t ems , I n c . 製) 1 zgZmlを投与し、 その作用を調べた。 実施例 3と同様に、 平滑筋細胞 マーカ 遺伝子 (力ルポニン、 SM22 or)、 血管内皮細胞マー—カー.遺伝子 (V WF)、 及び内部標準 (GAPDH) の発現を RT— P CR/サザンプロッ ト法 により調べた。
結果を第 7図に示す。 第 7図は、 FGF添加 (+ ) 又は FGF無添加 (一)、 及びフォリス夕チン添加 (+ ) 又は無添加 (一) で 5日間培養した後の各マーカ
一遺伝子の発現を示している。 これらの結果から、 フォリス夕チンが、 ヒト臍帯 静脈内皮由来細胞の F G F除去後の平滑筋細胞への分化を抑制することがわか る。
(3) TGF— /?ファミ リ一阻害物質による分化抑制
ヒト臍帯静脈内皮由来細胞に、 アデノウイルスベクターを用いて、 TGF—/? フアミリーの細胞内情報伝達経路の阻害物質である s ma d 7を強制発現させた 。 具体的に説明すると、 サイ トメガロウィルスプロモーターの制御下にて F L A G—ェピトープ (Nakao, A. et al. (1997) Nature 389, 631-635) とライゲートしたマ ウス Smad 7 c D N Aを有する組換え E 1欠失アデノウィルスベクター (A d CMV- S ma d 7 ) を作製し、 既に記載されている手法 (Fujii, M. et al. (I999) Mol. Biol. Cell 10, 3801-3813; Nakao, A. et al. ( 1999) J. Clin. Invest.104, 5-11.) に従つ て精製した。 すなわち、 293細胞に上記ベクターを導入後、 その培養上清中に 発現される組換え S ma d 7夕ンパク質を精製し利用した。
上述のように作製した S ma d 7発現細胞及び非発現細胞を、 実施例 1 と同様 に F GF除去培地中で 7日間培養した後、 実施例 2と同様に血管内皮細胞マーカ 一 ( vWF) 及び平滑筋細胞マーカ一 (力ルポニン) で同時染色した。
結果を第 8図に示す。 第 8図は、 Smad 7発現細胞 ( 1及び 3 ) と、 Sma d 7非発現細胞 ( 2及び 4 ) の写真を示す。 これらの結果から、 Smad 7が、 ヒ ト臍帯静脈内皮由来細胞の F GF除去後の平滑筋細胞への分化を抑制すること がわかる。
( 4 ) ME K依存的分化及び分化抑制の解除
ヒト臍帯静脈内皮由来細胞を F GF無添加培地中で 4日間培養した後、 さらに
FGF添加又は無添加で 4日間培養した。 またこれらの細胞に対し、 MAPキナ ーゼキナーゼ (MEK) 阻害剤の 1種である PD 98 059 (CALB I O CH EM社製) 50〃 gZmlを投与し、 その作用を調べた。 実施例 3と同様に、 平 滑筋細胞マーカ一遺伝子 (力ルポニン、 SM22 c 及び内部標準 (GAPDH ) の発現を RT—P CR/サザンプロヅ ト法により調べた。
結果を第 9図に示す。 第 9図は、 FGF無添加で 4日間培養 (4 ) したもの、 及び FGF添加 ( 8 +) 又は FGF無添加 ( 8—) でさらに 4日間培養したもの、 並びに PD 98059添加で培養した後 (右端のレーン) の各マーカー遗伝子の 発現を示している。 各平滑筋細胞マーカーの発現は、 : FGF除去後に経時的 (4 日、 8日) に増加したが、 FGF除去後 4日目に FGFを再度添加し、 さらに 4 日間培養したもの ( 8 ( + )) については、 各平滑筋細胞マ一カーの発現は F G F除去前のレベル近くまで減少していた。 この FGF再添加による各平滑筋細胞 マーカーの発現減少は、 P D 9 8059の投与によって阻害された (右端のレー ン)。 これらの結果から、 F G Fによる平滑筋細胞への分化抑制作用は、 MEK 依存的に起こることがわかり、 ME K阻害剤の投与によって、 平滑筋細胞への分 化抑制が解除され、 分化が増強されることがわかる。
〔実施例 8〕 骨芽細胞への分化
本実施例においては、 ヒト臍帯静脈内皮由来細胞から骨芽細胞への分化を確認 した。
ヒト臍帯静脈内皮由来細胞は、 実施例 1と同様に F GF添加培地で培養した後 、 FGF無添加培地で 3週間培養した。 その後、 血管内皮細胞マーカーであるゥ サギポリクローナル抗 vWF抗体 (D ak o製)、 及び骨芽細胞マーカーである
ャギポリクローナル抗アル力リホスファターゼ抗体 (C o r t e x B i o c h em, I n c .) を一次抗体として用いて二重標識し、 その後、 それそれテキサ スレヅ ドコンジユゲートロバ抗ゥサギ I gG (Ame r s ham) 及び F I T C コンジュゲートロバ抗ャギ I g G (Ame r s ham) を用いて二次標識した。 続いて、 共焦点レーザー走査顕微鏡 (Carl Zeiss, Jena, Germany) を用いてシグナル を検出した。
その結果を第 1 0図に示す。 第 1 0図は、 FGF添加 (+ ) 及び FGF除去後 3週目 (―) のヒ ト臍帯静脈内皮由来細胞を、 抗 vWF抗体と抗アルカリホスフ ァターゼ抗体とを用いて染色した結果を示す。 この写真から、 FGF除去後 3週 目に骨芽細胞のマーカーを発現する細胞が確認できた。 従って、 ヒ ト臍帯静脈内 皮由来細胞から、 骨芽細胞を誘導する分化細胞が得られることがわかった。 〔実施例 9〕 脂肪細胞への分化
本実施例においては、 ヒ ト臍帯静脈内皮由来細胞から脂肪細胞への分化を確認 した。
ヒト臍帯静脈内皮由来細胞は、 実施例 1と同様に F GF添加培地で培養した後 、 FGF無添加培地で 0~3週間培養した。 その後、 実施例 3と同様に、 脂肪細 胞特異的マーカー遺伝子 (リポプロテインリパーゼ) の発現を RT— P CRZサ ザンブロヅ小法に.より調べた。 リポプロティンリパーゼ遺伝子増幅用のプライ.マ ーセッ トを以下に示す :

上記プライマーセヅ トを用いて P CRを 36サイクル行った場合には、 得られ
る断片は 2 7 '6 b となる。 その後の操作は、 実施例 3と同様に行った。
その結果を第 1 1図に示す。 第 1 1図は、 F G F除去後 0〜 3週目のヒト臍帯 静脈内皮由来細胞におけるリポプロテインリパーゼの発現を示す。この結果から、 ヒト臍帯静脈内皮由来細胞は、 F G F除去後に経時的に脂肪細胞を誘導する分化 細胞に分化することがわかった。 産業上の利用可能性
本発明により、 他の種の細胞への分化能力を有する細胞及び他の種の細胞に分 化した細胞を大量に作製することが可能となる。 従って, そのような分化細胞を 作製することにより、 再生医療などにおいて有用な細胞を大量かつ簡便に得るこ とができる。 また本発明において最も好ましい血管内皮由来細胞である臍帯静脈 血管内皮由来細胞は、 液体窒素中に長期保存可能であることから、 出生時に臍帯 静脈血管内皮由来細胞を採取しておけば、 将来的に何らかの病的原因により各種 細胞 (例えば平滑筋細胞、 骨芽細胞及び脂肪細胞) の再生が必要になった際に保 存細胞を取り出して、 増殖及び分化させて使用することができる。 従って、 本発 明を利用することにより、 自己から取り出した細胞を分化させた後に体内に戻す ことができるため、 再生医療において重要な免疫学的問題が少ないことが期待さ れる。
Claims
1. 分化細胞の作製方法であって、 以下の工程 :
(a) 血管内皮由来細胞を増殖因子を含む培地中で培養する工程、 及び
(b) 培養した細胞の培地から増殖因子を除き、 又は上記の増殖因子活性を阻害 した状態で、 さらに細胞を培養する工程、
を含み、 他の種の細胞への分化能力を有する細胞及び z又は他の種の細胞に分化 した細胞を得ることを特徴とする、 上記作製方法。
2. 血管内皮由来細胞が臍帯静脈血管内皮由来細胞である、 請求の範囲第 1項に 記載の作製方法。
3. 血管内皮由来細胞が哺乳動物から採取されたものである、 請求の範囲第 1項 又は第 2項に記載の作製方法。
4. 哺乳動物がヒトである、 請求の範囲第 3項に記載の作製方法。
5. 増殖因子が繊維芽細胞増殖因子 (FGF) ファミ リーに属する因子である、 請求の範囲第 1— 4項のいずれか 1項に記載の作製方法。
6. 繊維芽細胞増殖因子が酸性繊維芽細胞増殖因子 (FGF— 1 ) である、 請求 の範囲第 5項に記載の作製方法。
-7. 他の種の細胞が、 平滑筋細胞、 骨芽細胞及び脂肪細胞からなる群より選択さ れるものである、 請求の範囲第 1一 6項のいずれか 1項に記載の作製方法。
8. 請求の範囲第 1一 7項のいずれか 1項に記載の作製方法により得られる分化 細胞。
9. 請求の範囲第 1― 8項のいずれか 1項に記載の作製方法に従って培養した細
胞を、 さらに増殖因子添加培地中で培養し、 血管内皮細胞に再分化させることを 特徴とする、 血管内皮細胞の作製方法。
1 0. 血管内皮由来細胞を分化させる方法であって、 以下の工程:
(a) 血管内皮由来細胞を增殖因子を含む培地中で培養する工程、 及び
(b) 培養した細胞の培地から増殖因子を除き、 又は上記の増殖因子活性を阻害 した状態で、 さらに細胞を培養する工程、
を含み、 他の種の細胞への分化能力を有する細胞及びノ又は他の種の細胞に分化 した細胞を得ることを特徴とする、 上記分化方法。
