STAT 6活性化遺伝子 技術分野
本発明は、 STAT 6の活性化を促進する作用を有するタンパク質、 該タンパ ク質をコードする DNA、 該 DNAの取得方法、 該 DNAを含有する組換えべク 夕一、 該組換えベクターを含有する形質転換体ならびに該タンパク質と反応する 抗体に関する。 また、 本発明は、 STAT 6の過剰な活性化または阻害が関与す る疾患の診断、 治療または予防を行う際の本発明のタンパク質、 DNAまたは抗 体の使用に関する。
また、 本発明は、 該タンパク質、 DNA、 組換えベクターおよび形質転換体を 用いて、 STAT 6の活性化を阻害または促進する物質をスクリ一二ングする方 法に関する。 背景技術
Mo s man nらは、 免疫応答において中心的な役割を担っているヘルパー T 細胞 (以下、 Thと略す) が、 異なる二つのサブセットに分類されることを提唱 した ( J . I mmu n o 1. ( 1 986) 1 36 : 2348— 2 3 57)。彼らは、 サイトカイン産生のパターンに基づき、 Th 1と Th 2の 2種類の細胞に分類し た。 Th 1細胞は、 Th 1タイプサイトカインと称されるインタ一ロイキン 2 ( I
L一 2)、 インターフェロン r (I FN—ァ)、 腫瘍壊死因子 j3 (TNF- j3) 等 を産生し、 ウィルス感染などにおける細胞性免疫を活性化する。 一方 Th 2細胞 は、 Th 2タイプサイトカインと称されるィンターロイキン 4 ( I L一 4)、 イン ターロイキン 5 (I L— 5)、 インターロイキン 10 (I L— 10)、 イン夕一口 ィキン 13 (I L— 13) 等を産生し、 寄生虫などの細胞内寄生微生物の感染や 抗原 ·アレルゲンの暴露に対する抗体産生などの液性免疫に関与する。 このよう に、 生体における種々の免疫応答を Th細胞のタイプにより分類し、 Th lと T h 2細胞のバランスによって病的な免疫反応を理解しょうとする考えが現れて、
Th 1病、 Th 2病という概念も提唱されてきている。
Th 2はアレルギー反応に関わる多くのサイトカインを産生することから、 T h 2の機能亢進が喘息などのアレルギー疾患を引き起こすと考えられている。
I L一 4は、 活性化 Tリンパ球、 好塩基球、 及び肥満細胞によつて分泌される 免疫調節サイ卜力インである。 I L— 4は B細胞の増殖や I gE、 I gG lの産 生を誘導するとともにマスト細胞の活性化、 増殖も誘導する。 また、 好酸球が血 管内皮細胞に接着、 組織浸潤する際に機能する VC AM— 1の遺伝子発現も誘導 する。 さらに、 I L一 4は、 Th 2細胞への分化、 造血前駆細胞の増殖および分 化において重要な役割を果たすことが示されている。
I L— 1 3は、 活性化 Tリンパ球、 マスト細胞、 好塩基球、 NK細胞、 樹状細 胞から分泌されるサイトカインである。 I L一 4に対してほぼ 30 %の配列相同 性を有し、 単球 ·マクロファージ、 B細胞に対して I L一 4様の活性を示す。 し かしながら、 I L一 1 3は T細胞には作用しない。
I L._4、 I L- 1 3がその細胞表面上のそのレセプターに結合すると細胞内 チロシンキナーゼが活性化され、 いくつかの細胞内タンパク質のチロシンリン酸 化を介し細胞内へシグナルが伝達される。 近年の分子生物の発展により、 I L一 4レセプ夕一からのシグナリングの機構が解明され、 主要細胞内伝達分子が同定 されてきた。 その中でも重要な分子として S T AT 6分子が見いだされた。
STAT 6は STAT (S i gn a l T r a n s d u c e r n d Ac t i v a t o r o f T r an s c r i p t i o n)ファミリ一の一員である。
S TATとは種々のサイトカインゃ成長因子レセプ夕一下流で刺激に依存して機 能する転写因子である。 ほ乳類ではこれまでに S T AT 1, 2, 3 , 4, 5 a,
5 b, 6の 7種が同定されている。 サイト力インなどのリガンドがそのレセプ夕 一へ結合すると J AKフアミリーと呼ばれるレセプター会合型チロシンキナーゼ が活性化され、 活性化を受けた J AKがレセプ夕一自身のチロシン残基をリン酸 化することによって STAT分子の活性化が引き起こされる。 活性化された S T
AT分子は二量体を形成してすみやかに核内へ移行し、 遺伝子発現を誘導する。
I L一 4レセプ夕一、 I L_ 13レセプ夕一を介して J AKが活性化され、 レセ プター上のチロシンがリン酸化される。 引き続き STAT 6が SH 2ドメインを
介してレセプターのリン酸化されたチロシン残基に結合し、 STAT 6自身がチ 口シンリン酸化されホモ二量体を形成して核内へ移行する。 S T AT 6の標的遺 伝子としては g e r m 1 i n e ィプシロン、 C D 2 3、 MH C (M a j o r H i s t o c omp a t i b i 1 i t y Comp l e x) クラス I I抗原、 S T A T 6遺伝子等が知られている。
最近になり、 STAT 6の欠損マウスが作製され、 その生理的役割が調べられ ている。 S T AT 6の欠損マウスでは、 I L一 4や I L一 1 3の主要な機能がす ベて障害されており、 S T AT 6が I L— 4ならびに I L一 1 3のシグナル伝達 において重要な分子であることが証明された。 また、 STAT 6欠損マウスでは Th 2反応が障害されており、 Th 2型サイ トカインの産生はほとんど認められ ず、 STAT 6が Th 2細胞分化においても必須な分子であることが示された。 つまりアレルギー反応の誘導に S T AT 6が重要な分子であることが証明された。 このような背景から、 S T A T 6の機能あるいは活性化を阻害することによつ て、 I L一 4ならびに I L一 1 3の機能を特異的に抑制し、 ァレルギ一性疾患 · 炎症 ·免疫疾患を抑制できる可能性がある。 S TAT 6の活性化に関するタンパ ク質はアレルギー性疾患 · 自己免疫や炎症を原因 ·症状とする疾病に対する医薬 の有望な標的である 〔たとえば、 P r o c. 細胞外からの情報は、 何らかのシグナルの形に変えて、 細胞膜を通過し細胞質 をこえて核に到達し、標的遺伝子の発現を調節して細胞の応答が引き起こされる。 そのため、 細胞外の刺激から STAT 6の活性化に至る細胞内におけるシグナル 伝達の仕組みを解明することは、 アレルギー性疾患、 自己免疫疾患や炎症症状を 呈する疾患に対する新たな医薬の開発あるいは治療法の開発に非常に重要な手段 を提供することとなり、 極めて重要な意義を有している。
しかしながら、 細胞が一定の刺激を受けてから S TAT 6の活性化に至るまで
のシグナル伝達経路には J AKキナーゼ、 STAT分子以外にこの経路を調節制 御する他の分子の存在が考えられる。従って、より効率的な創薬研究のためには、 主要な役割を果たす伝達分子を明らかにした上でそれらに焦点をしぼつた新しい 薬物スクリーニング方法を確立することが望まれる。 しかし、 STAT6を介し たシグナル伝達経路には J A K ,' S T A Τ分子以外はいまだ不明な点が多く、 新 たなシグナル伝達分子の同定とより進んだ STAT6活性化メ力ニズムの解明が 望まれていた。 発明の開示
本発明の課題は、 上記のように有用な S T AT 6の活性化を促進する作用を有 する新規な遺伝子、 タンパク質を見出し、 これを医薬、 診断薬、 医療の分野で利 用する方法を提供することにある。 即ち、 STAT6の活性化を促進する作用を 有する新規タンパク質、 該タンパク質をコードする DNA、 該 DNAを含有する 組換えベクター、 該組換えべクタ一を含有する形質転換体、 該タンパク質の製造 方法、 該タンパク質またはその部分ペプチドに対する抗体、 該抗体の製造方法を 提供する。
また、 本発明は、 該タンパク質、 DNA、 組換えベクターおよび形質転換体を 用いて、 STAT 6の活性化を阻害または促進する物質をスクリ一ニングする方 法、 該スクリーニング用キット、 該スクリーニング方法もしくはスクリ一ニング 用キッ トを用いて得られる S TAT 6の活性化を阻害または促進する物質、 該物 質の製造方法、 STAT 6の活性化を阻害または促進する物質を含有している医 薬などを提供する。
近年、 生体内で発現している遺伝子を解析する手段として、 cDNAの配列を ランダムに解析する研究が活発に行われており、 このようにして得られた c D N
Aの断片配列が EST (Expressed Sequence Tag、 たとえば
ttp//www. ncbi. nlm. nih. gov/dbEST) として、 データベースに登録され公開され ている。 しかし、 E S Tは配列情報のみであり、 その機能を推定することは困難 である。 また、 ESTは Un i Ge n e (http//www. ncbi. nlm. nih. gov/UniGene) により整備され、 これまでに約 8万種の ESTが登録されている。 しかし、 その
多くは 5 ' 端ヌクレオチド配列を欠損しており、 タンパク質翻訳開始部位を含ま ない。 そのため、 mRN Aのコード領域の決定を前提とするタンパク質の機能解 析、 プロモーターの解析による遺伝子発現制御の理解といった遺伝子機能の解析 に直結しているとは言いがたい。
一方、 遺伝子の産物、 すなわちタンパク質の機能を解明する方法の一つに、 動 物細胞を用いた一過性発現クロ一エング法がある (たとえば、 実験医学別冊 遺 伝子工学ハンドブック)。 この方法は、動物細胞発現ベクターを用いて作製した c DNAライブラリ一を、 動物細胞にトランスフエクションすることで機能的な夕 ンパク質を直接発現させ、 このタンパク質が細胞に及ぼす生物活性を指標として cDNAを同定、 クロ一ニングする方法である。 この方法では、 目的とするタン パク質産物に関する化学的情報 (アミノ酸配列や分子量) をあらかじめ必要とせ ず、 細胞内や培養液中に発現しているタンパク質の特異的生物活性を検出して c DNAクローンの同定を行うことができる。
この発現クローニングを効率良く行なうためには、 c DNAライブラリーの作 製方法を工夫する必要がある。 なぜなら、 従来より汎用されている c DNAライ ブラリー作製方法には幾つかの方法があるが (たとえば Gub b 1 e r -Ho f f ma nの方法: Ge n e 2 5 ( 1 9 83 )ォカャマーバーグの方法: M o 1. C e l l . B i o l . 2 ( 198 2 ))、 これらの方法によって作製された c DN Aは、 そのほとんどが 5 ' 末端ヌクレオチド配列を欠損したものであり、 完全長 (mRNAの全ヌクレオチド配列を含む) であることは稀であるからである。 そ の理由は、 mRNAから c DNAを作るのに使用する逆転写酵素が、 完全長の c D N Aを作る効率が必ずしも高くないからである。
さらに、 遺伝子の機能解析を試みるに際しては、 完全長 cDNAをクロ一ニン グし、 そこからタンパク質を発現させることが必須の要件である。 従って、 全体 のクロ一ンの中で、 完全長のものの割合が高いライブラリ一を作製することが、 発現クロ一ニングを効率良く行なうために必要であった。
本発明者らは、 上記課題を解決するために鋭意研究を重ねた結果、 オリゴキヤ ッピング法を用いて完全長 c DN Aライブラリ一を作製し、 N I H 3 T 3細胞を 用いた発現クローニングによる遺伝子機能アツセィ系を完成し、 該アツセィ系に
より S TAT 6の活性化を促進する作用を有するタンパク質をコードする新規 D NA (c DNA) を単離することに成功した。 この新規 DNAは、 N I H3 T 3 細胞内で発現させることにより STAT 6の活性化の促進を誘発した。 この結果 は、 この新規 DNAが S TAT 6の活性化の促進に関与するシグナル伝達分子で あることを示しており、 本発明を完成するに至った。

(b) 該遺伝子が形質転換された細胞内で発現可能となる条件下で該形質転換さ れた宿主細胞を培養する工程、

(1 5) S T A T 6の阻害剤または活性化剤としての活性について化合物をス クリーニングするためのキッ卜であって、

まず、 本発明の基本的特徴を更に明らかにするために、 本発明の完成に至る経 緯を追いながら、 本発明について説明する。 S TAT 6の活性化を促進する作用 を有する新規遺伝子を取得する目的で、 実施例に示すように、 以下の実験を実行 した。 まずヒト正常肺線維芽細胞 (三光純薬株式会社より購入) より調製した m RNAより、 オリゴキヤッビング法によって完全長 c DNAを作製し、 該 c DN Aをベクター pME 1 8 S— FL 3 (GenBank Accession AB009864) に組み込ん だ完全長 c DNAライブラリ一を作製した。 次に、 該 c DNAライブラリーを大 腸菌に導入し、 1クローンずつプラスミドを調製した。 次に、 N I H3 T 3細胞 (大日本製薬) に、 ルシフェラーゼをコードする DNAの上流に S T AT 6応答性 配列を含有するレポ一夕一プラスミド 〔例えば、 J . 本発明において S T AT 6の活性化を促進する作用を有するとは、 適切な細胞 内に遺伝子を導入し、 該遺伝子にコードされているタンパク質を過剰発現させた
時、 生理的な刺激が無い状態でも、 STAT 6が直接的あるいは間接的に活性化 される (STAT6の活性化を誘発する) ことをいう。 及び,"'または、 適切な細 胞-内に遺伝子を導入し、 該遺伝子にコードされているタンパク質を過剰発現させ た後、 生理的な刺激が細胞に入った場合、 正常レベルの STAT 6の活性化がさ らに直接的あるいは間接的に促進される(STAT 6の活性化の促進を誘発する) ことをもいう。 STAT 6の活性化は、 例えば、 S TAT 6依存レポ一夕一遺伝 子を用いたアツセィにより、 該レポ一夕一活性を対照細胞 (該レポーター遺伝子 と空べクタ一を導入した細胞) に比し上昇させる作用を測定することにより検出 できる。レポ一夕一活性の上昇は、好ましくは、 1. 5倍以上、さらに好ましくは、 3倍以上、 さらに好ましくは 6倍以上である。
発現させたいタンパク質をコードするボリヌクレオチド (例えば cDNA) を 適切な発現ベクター内にクローニングし、 該発現べクタ一と S TAT 6依存レポ 一夕一プラスミドを適切な細胞に共導入 (コ · トランスフエクシヨン) し、 一定 時間培養後、 レポ一夕一活性を測定する。 あるいは、 共導入し一定時間培養後、 刺激剤を添加しさらに培養しレポ一夕一活性を測定する。 適切な発現ベクターは 当業者にはよく知られており、 例えば、 pME I S S— FL 3、 p c DNA 3. 1 ( I n v i t i- o g e n社) などが挙げられる。 レポ一夕一遺伝子は、 当業者 がその発現を容易に検出できるものであればよく、 例えば、 ルシフェラーゼ、 ク 口ラムフエニコールァセチルトランスフェラーゼ、 3—ガラク トシダ一ゼをコ一 ドする遺伝子である。 ルシフェラーゼをコードする遺伝子を使用することが最も 好ましく、 S T AT 6依存レポ一タープラスミ ドとしては、 例えば、 STAT 6 応答配列を有するルシフェラ一ゼレポ一タープラスミ ド N 4 X 8 _ 1 u cが例示 される。 適切な細胞とは、 I L— 4あるいは I L一 1 3などの刺激により S TA T 6が活性化されるような応答を示すような細胞であり、 例えば、 N I H 3 T 3 細胞が挙げられる。細胞培養および細胞への遺伝子導入(トランスフエクシヨン) は、当業者であれば当該技術分野で公知の慣用方法により実施でき最適化できる。 好ましい方法としては、 N I H 3 T 3細胞を細胞培養用 96穴プレートに 1 X

"同一性" とは、 当該技術で知られているとおり、 配列を比較することにより 決定される、 2以上のタンパク質あるいは 2以上のポリヌクレオチドの間の関係 である。 当該技術で "同一性" とは、 またタンパク質またはポリヌクレオチド配
列の間の適合によって、 あるいは場合によっては、 一続きのそのような配列間の 適合によって決定されるような、 タンパク質またはポリヌクレオチド配列の間の 配列相関性の程度を意味する。 "同一性"および "類似性" は、 既知の方法により 容易に決定できる。 同一性を決定する好ましい方法は、 試験する配列間で最も長 く適合するように設計される。 同一性および類似性を決定するための方法は、 公 に利用可能なプログラムにコードされている。 相同性決定には、 例えば、 BLAST (Basic Local Alignment Search Tool) プログラム (例えば、 Altschul SFf Gish W, Miller W, Myers EW, Lipman DJ. , J. Mol. Biol. , 215: p403-410 (1990), Altschul SF, Madden TL, Sc affer AA, Zhang J, Miller W, Lipman DJ. , Nucleic Acids Res. 25: p3389-3402 (1997) )を利用し決定することができるが、 これに限 定されない。 BLASTのようなソフトウェアを用いる場合、 デフォルト値を用いる のが好ましい。
BLAST検索に一般的に用いられる主な初期条件は、 以下の通りであるが、 これ に限定されない。 アミノ酸置換行列とは 20種類のアミノ酸の各々のペアの類縁 性を数値化した行列であり、通常 BLOSUM62のデフォルトマトリックスが用いられ る。 このアミノ酸置換行列の理論については、 Altschul S. F. , J. Mol. Biol. , 219:555-565 (1991)に、 DNA配列の比較への適用については、 States D. J. , Gis W. , Altschul S. 期待値 (EXPECT) とは、 データベース配列に対してマッチする際の統計的有意 性に関する閾値であり、 デフォルト値は 10である。

当業者であれば、 公知の方法を用いてこのタンパク質中のアミノ酸の置換など を適宜行い、 配列番号 1等で表されるァミノ酸配列を有する夕ンパク質と同様に
STAT 6の活性化を促進する作用を有するタンパク質を作製することが可能で ある。 一つの方法としては、 該タンパク質をコードする DNAに対して、 慣用の 突然変異誘発法を使用する方法がある。 別の方法としてはたとえば部位特異的変 異法 (たとえば宝酒造株式会社の Mu t a n-S u p e r Ex p r e s s K m キッ ト) が挙げられる。 また、 タンパク質のアミノ酸の変異は自然界におい ても生じうる。 このようにアミノ酸の欠失、 置換、 付加により配列番号 1等で表 されるァミノ酸配列を有する夕ンパク質に対してアミノ酸配列が変異した変異体 であって、 S T A T 6の活性化を促進する作用を有するタンパク質及び該タンパ ク質をコードする DNAも本発明に含まれる。変異の数は、好ましくは 1 0まで、 より好ましくは 5まで、 最も好ましくは 3までが好ましい。

本発明のタンパク質としては、 ヒ卜や哺乳動物のあらゆる細胞や組織に由来す る天然のタンパク質でもよく、 化学合成タンパク質であってもよく、 また遺伝子 組換え技術によって得られたタンパク質でもよい。 タンパク質は糖鎖ゃリン酸化 などの翻訳後修飾は受けていても受けていなくても良い。
本発明のタンパク質としては、 例えば、 分泌タンパク質 (増殖因子、 サイ トカイ ン、 ホルモン等)、 タンパク質修飾酵素 (タンパク質リン酸化酵素、 タンパク質脱 リン酸化酵素、 プロテアーゼ等)、 核内タンパク質 (核内受容体、 転写因子等) 膜 タンパク質が挙げられる。 膜タンパク質は、 受容体、 細胞接着分子、 イオンチヤ
ンネル、 トランスポーター等が挙げられる。 タンパク質が膜タンパク質である場 合、 後述するスクリーニングにより選択された化合物は、 細胞内へ容易に移行す ることが予想されるため、 医薬化合物のリサーチツールとしては、 より有用であ る。

本発明の DNAは、 配列番号 2等で表されるヌクレオチド配列からなる DN A とストリンジェン卜な条件下でハイプリダイズし、 かつ S T AT 6の活性化を促 進する作用を有するタンパク質をコードする DNAも含まれる。 ス卜リンジェン 卜な条件とは、 当業者には十分理解できることであり、 たとえば、 T. Ma n i a t i s らの実験操作書 (Mo l e c u l a r C l o n i n g A L a b o r a t o r y Ma n u a l , C o l d S p r i n g H a r b o r L a b o r a t o r y 1 9 82、 1 9 8 9) に従えば容易に実施できる。
すなわち、 ストリンジェントな条件とは、 30 %ホルムアミドを含むハイプリ ダイゼ―シヨン溶液中 (5 X S S C (0. 7 5MのN a C l、 7 5mMのクェン 酸三ナ卜リゥム)、 5 Xデンハルト溶液、 0. 5。b' S D S、 1 0 0 ," g m 1の変 性せん断サケ精子 DNA) で 3 7°Cのインキュベーションをー晚行い、 その後 2 X S S C、 0. 1 ¾' S D S中、 室温で 1 0分の洗浄を 3回行い、 次いで 1 X S S C、 0. 1 ¾SD S中、 37 °Cで 1 0分の洗浄を 2回行う条件である (低ストリ ンジエンシー)。 より好ましい条件は、 40 ¾ホルムアミドを含むハイブリダィゼ —ション溶液中で 42°Cのィンキュベーションをー晚行い、 その後 2 X S S C、 0. 1 % S D S中、 室温で 1 0分の洗浄を 3回行い、 次いで 0. 2 X S S C、 1 % SDS中、 42°Cで 1 0分の洗浄を 2回行う条件である(中ストリンジエンシー)。 最も好ましい条件は、 50.%ホルムアミドを含むハイブリダィゼ一シヨン溶液中 で 42°Cのインキュベーションをー晚行い、 その後 2 X S S C、 0. 1 % S D S 中、 室温で 1 0分の洗浄を 3回行い、 次いで 0. 2 X S S C、 0. 1 %SD S中、 5 0°Cで 1 0分の洗浄を 2回行う条件である (高ストリンジエンシー)。 この際、 得られた DN Aは、 STAT 6の活性化を促進する作用を有するタンパク質をコ 一ドすることが必須である。
本発明は、 上記 (3) あるいは (4) のポリヌクレオチドのヌクレオチド配列 と高い類似性を有し、 かつ STAT 6の活性化を促進する作用を有するタンパク 質をコードするヌクレオチドを含むポリヌクレオチドを含む。 代表的には、 これ
らのヌクレオチド配列は、 上記 (3) または (4) のポリヌクレオチドのヌクレ ォチド配列の全長にわたり 9 5 %同一であり、より好ましくは 9 7 %同一であり、 最も好ましくは少なくとも 9 9 %同一である。
上記の本発明の DNAは、 前述のタンパク質を、 組換え DNA技術を用いて製 造するのに用いることができる。 本発明の DNA及びペプチドは、 概略以下のよ うにして得ることができる。
(A) 本発明のタンパク質をコードする DNAをクロ一ニングする。
(B) 夕ンパク質の全コード領域あるいはその一部をコードする DN Aを発現用 ベクターに組み込んで、 組換えベクターを構築する。
(C) 構築した組換えベクターにより、 宿主細胞を形質転換する。
(D) 得られた細胞を培養し、 該タンパク質、 またはその類縁体を発現させ、 力 ラムクロマトグラフィーにより精製する。
上記の工程中で D N A、 組換え体宿主としての大腸菌等の取り扱いに必要な一 般的な操作は、 当業者間で通常行われているものであり、 たとえば、 上記 T. M a n i a t i sらの実験操作書に従えば容易に実施できる。 使用する酵素、 試薬 類も全て市販の製品を用いることができ、 特に断らない限り、 製品で指定されて いる使用条件に従えば、 完全にそれらの目的を達成することができる。 以下に上 記 (A) 〜 (D) の工程について更に詳しく説明する。
上記 (A) における本発明のタンパク質をコードする DNAのクローニングの 手段としては、 本願明細書実施例に記載した方法の他に、 本発明のヌクレオチド 配列 (たとえば配列番号 2等) の一部を有する合成 DNAをプライマーとした P
CR法によって増幅する方法、 あるいは、 適当なベクターに組み込んだ DNAを 本発明のタンパク質の一部あるいは全領域をコードする DN A断片もしくは合成

すなわち、 本発明は、 上記の DN Aを含有する組換えベクターである。
本発明のタンパク質の発現べクタ一は、 たとえば、 本発明のタンパク質をコー ドする D N Aから目的とする D N A断片を切り出し、 該 D N A断片を適当な発現 ベクター中のプロモーターの下流に連結することにより製造することができる。 用いる発現ベクターとしては、 複製可能であれば、 大腸菌をはじめとする原核 生物由来、 酵母由来、 真菌由来、 昆虫ウィルス由来、 脊椎動物ウィルス由来いず れのベクタ一でも良いが、 宿主として使用する微生物または細胞に適したものを 選択する必要がある。 また、 発現物に応じて、 宿主細胞一発現ベクター系として は、 適切な組み合わせが選択される。
微生物を宿主として使用する場合、 これら微生物に適したプラスミ ドベクター が組み換え体 D N Aの複製可能な発現べクターとして一般に用いられる。
たとえば、 大腸菌を形質転換するためのプラスミ ドベクターとしては、 プラス ミド p BR 3 2 2や pBR 3 2 7などを用いることができる。 プラスミドベクタ 一は通常複製起源、 プロモータ一、 及び組換え体 DN Aで形質転換した細胞を選 別するのに有用な表現型を組換え体 D N Aに与えるマーカ一遺伝子等を含んでい る。 プロモーターの例としては、 3—ラクタマーゼプロモータ、 ラクトースプロ モータ一、 トリプトファンプロモータ一等が挙げられる。 マーカー遺伝子の例と しては、 アンピシリン耐性遺伝子ゃテトラサイクリン遺伝子などが挙げられる。 適した発現べクタ一の例としては、 プラスミ ド pBR 322、 P BR 3 2 7の他 に、 pUC.1 8、 pUC 1 9等が挙げられる。
酵母で本発明の DNAを発現するためには、 複製可能なベクタ一として、 たと えば YE P 24を用いることができる。 プラスミ ド YE p 24は UR A 3遺伝子 を含有しており、 この UR A 3遺伝子をマーカ一遺伝子として利用することがで
きる。 酵母細胞用の発現ベクターのプロモーターの例としては、 3—ホスホグリ セレ一卜キナーゼ、 グリセルアルデヒドー 3—ホスフエ一卜デヒドロゲナーゼ、 アルコールデヒドロゲナーゼなどの遺伝子のプロモーター等が挙げられる。
真菌で本発明の D N Aを発現するための発現ベクターに用いられるプロモ一夕 一及び夕一ミナ一ターの例としては、 ホスホダリセレートキナーゼ(PGK)、 グ リセルアルデヒドー 3—ホスフェートデヒドロゲナーゼ(GAPD)、 ァクチン等 の遺伝子プロモーター及びターミネータ一が挙げられる。 適した発現べクタ一の 例としては、 プラスミ ド p P GAC Y 2、 p B S F AHY 83等が挙げられる。 昆虫細胞で本発明の D N Aを発現させるための発現ベクターに用いられるプロ モータ一の例としては、 ポリヘドリンプロモ一夕一、 P 10プロモー夕一などが 挙げられる。
動物細胞で本発明の D N Aを発現させるための組換えべクタ一は、 一般に遺伝 子を制御するための機能配列、 たとえば、 複製起源、 本発明の DNAの上流に位 置すべきプロモーター、 リボソーム結合部位、 ポリアデニル化部位や転写終止配 列を含有している。 本発明の DNAを真核細胞内で発現させるのに用いることが できるそのような機能配列はウィルスやウイ,)レス性物質から得ることができる。 例えば、 SRひプロモーター、 SV40プロモーター、 LTRプロモーター、 C
MV (サイトメガロウィルス) プロモーター、 HS V-TKプロモーターなどがあ げられる。 これらのうち、 CMVプロモータ一、 S R αプロモーターなどを用い るのが好ましい。 また、 本発明のタンパク質をコードする遺伝子の上流位置に本 来存在するプロモーターも、 上述の宿主一ベクター系で使用するのに適している ならば使用することができる。 複製起源については、 外来性の起源、 たとえばァ デノウィルス、 ポリオ一マ、 S V40等のウィルス由来の複製起点を用いること ができる。 また、 発現べクタ一として宿主染色体に組み込まれるような性質を有 するベクターを用いる場合、 宿主染色体の複製起源を利用することができる。 適 した発現ベクターの例としては、 プラスミド p S V 2— d h f r (ATCC 3
7 146)、 pBPV - 1 (9— 1) (ATCC 37 1 1 1)、 p c DNA 3. 1
( I NV I TROGEN社)、 pME 1 8 S— FL 3等が挙げられる。
本発明は、 上記の組換えベクターを含む形質転換された細胞である。 本発明の
複製可能な組換えベクターで形質転換された微生物または細胞は、 前述の通り、 組換えべクタ一に与えられた少なくとも 1種の表現型によって形質転換されずに 残った親細胞から選別される。 表現型は少なくとも 1種のマーカー遺伝子を組換 えベクターに揷入することによって与えることができる。 また複製可能なべクタ 一が本来有しているマ一カー遺伝子を利用することもできる。 マーカー遺伝子の 例としては、 たとえば、 ネオマイシン耐性などの薬剤耐性遺伝子ゃジヒドロ葉酸 レダクターゼをコードする遺伝子などが挙げられる。 上記 (C.) において用いる宿主としては、 大腸菌をはじめとする原核生物、 酵 母、 真菌等の微生物、 及び昆虫や動物等の細胞のいずれでも良いが、 用いる発現 ベクターに適したものを選択する必要がある。 微生物の例としては、 エシュリヒ ァ コリ (E s c h e r i c h i a c o l i ) の菌株、 たとえば E. c o 1 i

(S e r r a t i a ma r c e s a n s) 等の大腸菌以外の腸内菌、 あるいは シュ一ドモナス (P s e ud omo n a s) 属の種々の菌株が挙げられる。 酵母 としては、 たとえば、 サッカロミセス セレピシェ (S a c c h a 1- omy c e s c e r e v i s i a e)、 シソ 'サッカロマイセス ポンべ (S c h i z o s a c c h a r o m y c e s p o m b e )、 ピキア ノ ス卜リス ( P i c h i a p a s t o r i s ) などが用いられる。 真菌としては、 たとえば、 ァスペルギルス ニドランス (A s p e r g i 1 1 u s n i d u 1 a n s )、 アクレモニゥム ク リソゲナム (Ac r emon i um c h r y s o g e n um) (ATCC 1 1
550 ) 等が挙げられる。
昆虫細胞としては、 たとえば、 ウィルスが Ac NPVの場合は、 夜盗蛾の幼虫 由来株化細胞 (S p o d o p t e r a f r u g i p e r d a : S f 細胞)、 T r i c h o p l u s i a n iの卵由来の H i g h F i v e TM細胞、 などが用 いられる。 動物細胞の例としては、 HEK 293細胞、 COS— 1細胞、 COS
一 7細胞、 He 1 a細胞、 チャイニーズハムスター (CH〇) 細胞等が挙げられ る。 これらの中でも、 CHO細胞および HEK 29 3細胞が好ましい。
細胞を宿主とする場合、 用いられる発現ベクターと宿主細胞の組合せは実験の 目的により異なるが、 その組合せにより、 一過的発現、 構成的発現の 2種類の発 現方式が考えられる。
上記 (C) における微生物及び細胞の形質転換とは、 DNAを強制的方法や、 細胞の貪食能により微生物や細胞に取り込ませ、 プラスミド状態あるいは染色体 に組み込まれた状態で DNAの形質を一過的あるいは構成的に発現させることで ある。 当業者であれば公知の方法によって形質転換できる (たとえば実験医学別 冊遺伝子工学ハンドブック)。たとえば動物細胞の場合、 DEAE—デキス卜ラン 法、 リン酸カルシウム法、 エレクト口ポレーシヨン法 (電気穿孔法)、 リボフェク ション法などの方法で D N Aを細胞に導入することができる。動物細胞を用いて、 本発明のタンパク質を安定に発現させる方法としては、 上記の動物細胞に導入さ れた発現べクタ一が染色体に組み込まれた細胞をクローン選択によって選択する 方法がある。 具体的には、 上記の選択マーカーを指標にして形質転換体を選択す る。 さらに、 このように選択マーカーを用いて得られた動物細胞に対して、 繰り 返しクロ一ン選択を行なうことにより本発明のタンパク質の高発現能を有する安 定な動物細胞株を得ることができる。 また、 D i h yd r o f o r a t e r e d u c t a s e (DHFR) 遺伝子を選択マーカ一として用いた場合 M e t h o t r e x a t e (MTX) 濃度を徐々に上げて培養し、 耐性株を選択することに より、 DHFR遺伝子とともに、 本発明のタンパク質をコードする DNAを細胞 内で増幅させて、 さらに高発現の動物細胞株を得ることもできる。
上記の形質転換された細胞を本発明のタンパク質をコードする D N Aが発現可 能な条件下で培養し、 本発明のタンパク質を生成、 蓄積せしめることによって、 本発明のタンパク質を製造することができる。 すなわち、 本発明は、 上記 (3) 〜 (6) に記載の単離されたポリヌクレオチドを含む形質転換された細胞を、 該 ポリヌクレオチドによりコードされているタンパク質を発現させる条件下培養し、 次いで培地から該タンパク質を回収することを含む該タンパク質の製造方法であ
上記の形質転換された細胞の培養は、 当業者に公知の方法で行なうことができ る (たとえばバイオマニュアルシリーズ 4、 羊土社)。 たとえば動物細胞の場合、 各種の動物細胞培養法、 たとえば、 シャーレ培養、 マルチトレ一式培養、 モジュ ール培養などの付着培養、 または細胞培養用担体 (マイクロキャリアー) に付着 させるか生産細胞自体を浮遊化させ浮遊培養等の公知の方法により培養を行なえ ば良い。 培地は通常良く用いられる動物細胞用の培地、 たとえば D— M E Mや R P M I 1 6 4 0等を用いれば良い。
上記培養物から本発明のタンパク質を分離精製するには、 自体公知の分離 ·精 製法を適切に組み合わせて行なうことができる。 これらの公知の分離、 精製法と しては、' 塩析ゃ溶媒沈澱法などの溶解度を利用する方法、 イオン交換クロマトグ ラフィ一などの荷電の差を利用する方法、 透析法、 限外ろ過法、 ゲルろ過法、 お よび S D S—ポリアクリルアミ ドゲル電気泳動法などの主として分子量の差を利 用する方法、 ァフィ二ティークロマトグラフィーなどの特異的親和性を利用する 方法、 逆相高速液体クロマトグラフィーなどの疎水性の差を利用する方法、 等電 点電気泳動法などの等電点の差を利用する方法などが用いられる。 たとえば、 本 発明のタンパク質は、 硫安またはエタノール沈殿、 酸抽出、 ァニオンまたはカチ オン交換クロマトグラフィー、 ホスフ才セルロースクロマトグラフィー、 疎水性 相互作用クロマトグラフィー、 ァフィ二ティクロマトグラフィー、 ヒドロキシァ パタイ トクロマトグラフィーおよびレクチンクロマ卜グラフィーを含む既知の方 法により組換え細胞培養物から回収し、精製することができる。最も好ましくは、 高性能液体クロマトグラフィ一が精製に使用される。ポリぺプチドが細胞内合成、 単離または精製の間に変性するときには、 活性なコンフォーメシヨンを再生する ために夕ンパク質をリフォールデイングするためのよく知られた技術を使用でき る。
本発明のタンパク質を他の夕ンパク質との融合夕ンパク質として製造すること ができる。 これらも、 本発明に含まれる。 この融合タンパク質を発現する際に用 いられるベクタ一としては、 該夕ンパク質をコードする D N Aを組み込むことが でき、 かつ該融合タンパク質を発現することができるベクタ一であれば、 いかな るベクターでも用いることができる。 本発明のペプチドに融合できる夕ンパク質
としては、 たとえばダル夕チオン一 S —トランスフェラーゼ (G S T )、 ヒスチジ ン残基の 6個の連続配列 (6 X H i s ) 等が挙げられる。 本発明のタンパク質を 他の夕ンパク質と融合した夕ンパク質として発現させた場合には、 融合したタン パク質に親和性をもつ物質を用いたァフィ二ティ一クロマトグラフィーを用いて 精製することができ、 有利である。 例えば、 G S Tとの融合タンパク質として生 産した場合は、 ダル夕チオンをリガンドとするァフィ二ティーク口マトグラフィ 一により精製することができる。 ' 本発明は、 上記 (7 ) のタンパク質の活性を阻害するタンパク質を含む。 たと えば、 抗体や上記 (7 ) のタンパク質の活性中心等に結合し、 活性の発現を妨げ る他のタンパク質が挙げられる。
本発明は、 前記の本発明のタンパク質あるいはその部分べプチドに反応する抗 体ならびにそのような抗体の製造方法に関する。 さらに好ましくは、 前記の本発 明のタンパク質あるいはその部分ペプチドに特異的に反応する抗体、 並びにその ような抗体の製造方法に関する。 ここでいう 「特異的」 とは、 交差反応性が少な いもの、 より好ましくはないものを意味する。 抗体は、 本発明のタンパク質を認 識し得る抗体であれば、 ポリクローナル抗体、 モノクローナル抗体、 ならびにこ れらの抗体のフラグメント、 一本鎖抗体、 ヒト化抗体の何れであってもよい。 抗 体フラグメントは、 公知の技術によって作製することができる。 たとえば、 該抗 体フラグメントには、 限定されるものではないが、 F ( a b ' ) 2フラグメント、 F a b ' フラグメント、 F a bフラグメント及び F vフラグメントが含まれる。 たとえば、 モノクローナル抗体またはポリクロ一ナル抗体は、 上記 (1 ) または ( 2 ) に記載のタンパク質を抗原またはェピトープ含有フラグメントとして非ヒ ト動物に投与することにより得られる。 本発明のタンパク質に対する抗体は、 本 発明のタンパク質あるいはそのペプチドを抗原として用い、 自体公知の抗体また は抗血清の製造法に従って製造することができる。 たとえば実験医学別冊 新遺 伝子工学ハンドブック 改訂第 3版に記載の方法が挙げられる。
ポリクローナル抗体の場合であれば、 たとえば、 本発明のタンパク質をゥサギ などの動物に本発明の夕ンパク質あるいはペプチドを注射することにより該タン パク質あるいはべプチドに対する抗体を産生させ、次いで血液を採取し、これを、
たとえば硫安沈殿、 イオン交換クロマトグラフィー、 あるいは該タンパク質を固 定化したァフィ二ティ一カラム等によって精製することで調製することができる, モノクローナル抗体の場合は、 たとえば、 本発明のタンパク質をマウスなどの 動物に免疫し、 同マウスから脾臓を抽出し、 これをすりつぶして細胞にし、 マウ スミエロ一マ細胞とポリエチレングリコールなどの試薬により融合させ、 これに よりできた融合細胞 (ハイプリ ドーマ) の中から、 本発明のタンパク質に対する 抗体を産生するクローンを選択する。 次いで、 得られたハイプリ ドーマをマウス 腹腔内に移植し、同マウス内より腹水を回収し、得られたモノクローナル抗体を、 たとえば硫安沈殿、 イオン交換クロマトグラフィー、 あるいは該タンパク質を固 定化したァフィ二ティーカラム等によって精製することで調製することができる, 得られた抗体をヒトに投与する目的で使用する場合は、 免疫原性を低下させる ために、 ヒト型化抗体あるいはヒト抗体を用いることが好ましい。 これらのヒト 型化抗体ゃヒト化抗体は、 トランスジエニックマウスまたは他の哺乳動物を用い て作製することができる。 ヒト型化抗体の一般的概説は、 たとえば、 Mo r r i s o n, S . 56 ( 1 9 98)] によって供されている。 ヒト化キメラ抗体は、 マウス抗体の V 領域とヒト抗体の C領域を遺伝子組換えにより結合し、 作製することができる。 ヒト化抗体は、 マウスのモノクローナル抗体から相補性決定部位 (CDR) 以外 の領域をヒト抗体由来の配列に置換することによって作製できる。 また、 免疫系 をヒトのものと入れ換えたマウス用いて、 該マウスを免疫して、 通常のモノクロ ーナル抗体と同様に直接ヒト抗体を作製することもできる。 これらの抗体は、 夕 ンパク質を発現するクローンを単離したり同定するのに使用できる。 また、 これ らの抗体は、 本発明のタンパク質を細胞抽出液、 または本発明のタンパク質を産 生する形質転換細胞から精製するのに使用できる。 更にこれらの抗体は、 細胞や 組織中の本発明のタンパク質を検出する EL I S Aや R I A (ラジオィムノアツ セィ)、 またはウエスタンプロット系の構築に使用できる。 このような検出系は、
動物、 好ましくは、 ヒ卜の組織または血管内流体などの身体サンプル中に存在す る本発明のタンパク質の存在量を検出する診断目的に使用することができる。 た とえば、 これらの抗体は、 アレルギー性疾患、 炎症、 自己免疫疾患、 糖尿病、 高 脂血症、 感染症 (一例として H I V感染)、 癌などの、 本発明のタンパク質の(発 現)異常に起因する S T A T 6の望ましくない活性化によって特徴付けられる疾 患の診断に使用できる。 疾患の診断の基礎を提供するために、 本発明のタンパク 質の発現についての通常の値、 すなわち標準値が確立されなければならないが、 これは当業者においては周知の技術である。 すなわち、 複合体形成のための適切 な条件下で、 ヒトあるいは動物のどちらでもよいが、 正常の被験者から得られた 体液あるいは細胞抽出物と、 本発明のタンパク質に対する抗体とを結合させ、 こ の抗体一タンパク複合体の量を化学的または物理的手段により検出し、 これを既 知量の抗原 (本発明のタンパク質) を含む標準液を用いて作成した標準曲線を用 いて、 正常サンプルから得られた標準値を算出する。 標準値と本発明のタンパク 質が関係する疾患を潜在的に患う被験者からのサンプルから得られた値と比較し. 標準値との偏差によって疾病の存在を確認することができる。 また、 これらの抗 体は、 本発明のタンパク質の機能を研究する試薬としても用いることができる。 本発明の抗体は精製され得、 次いで、 たとえば、 アレルギー性疾患、 炎症、 自 己免疫疾患、 糖尿病、 高脂血症、 感染症 (一例として H I V感染)、 癌などの、 本 発明のタンパク質の(発現)異常に起因する S T A T 6の望ましくない活性化によ つて特徴付けられる疾患の患者に投与され得る。 すなわち本発明は、 上記に記載 の抗体を有効成分として含有する医薬、 および抗体を用いた治療方法である。 こ れらの医薬は治療的使用のためにさらなる有効成分または不活性成分(たとえば、 従来の薬学的に受容可能なキャリアまたは希釈剤 (たとえば、 免疫原性アジュバ ント) と、 生理学的に無毒の安定化剤および賦形剤とともに組み合わされ得る。 これらの組み合わせは、濾過滅菌され、そして凍結乾燥により投薬バイアル中に、 または安定化水性調製物中の貯蔵物として投薬形態にされ得る。患者への投与は、 たとえば、 動脈内注射、 静脈内注射、 皮下注射などの当業者に公知の方法により 行い得る。 投与量は、 患者の体重や年齢、 投与方法などにより変動するが、 当業 者であれば適当な投与量を適宜選択することが可能である。 これらの抗体は、 本
発明のタンパク質で仲介される S T AT 6の活性化の促進を阻害し、 治療効果を 示す。
本発明の DN Aは、 細胞内シグナリングプロセスに関与する他のタンパク質を 単離、 同定、 クローン化することにも使用できる。 たとえば、 本発明のタンパク 質をコードする DNA配列は、 コードされたタンパク質を 「バイ ト (b a i t)J として用いて、 c DNAまたはゲノム DNAライブラリーから、 本発明のタンパ ク質に結合できるタンパク質をコードする他の配列 「プレイ (p r e y)」 を単離 し、 クローン化する酵母ツーハイブリッドシステム (たとえば Na t u r e, 3 40 : 245— 246 (198 9)) に用いることができる。 同様の方式で、 本発 明のタンパク質が、 他の細胞タンパク質 (たとえば STAT 6, J AK 1) に結 合できるかどうかも決定することができる。 あるいは別の方法として、 本発明の タンパク質の抗体を用いた免疫沈降法 (たとえば、 実験医学別冊新遺伝子工学ハ ンドブック) によって、 本発明のタンパク質に結合し得るタンパク質を細胞抽出 物から単離する方法が挙げられる。さらに別の方法として、上記に記載のように、 本発明のタンパク質を他のタンパク質との融合タンパク質として発現させ、 融合 タンパク質に対する抗体を用いて免疫沈降法を行ない、 本発明のタンパク質に結 合し得る夕ンパク質を単離する方法が挙げられる。
診断アツセィは、 上記した方法により、 STAT6の活性化を促進するタンパ ク質をコードするヌクレオチド配列中の変異を検出することにより疾患の診断や 該疾患への感受性を決定するための方法を提供する。さらに、このような疾患は、 個体に由来するサンプル中のタンパク質または m R N Aレベルの異常な減少また は増加を測定することを含む方法によって診断してもよい。 発現の減少または増 加は、当該技術で R N Aレべルでのポリヌクレオチドの定量によく知られた方法、 たとえば、 R T _ P C Rなどの核酸増幅、 R N a s e保護法、 ノ一ザンブ口ッ卜 法およびその他のハイブリダィゼーション法などを使用して測定できる。 宿主に 由来するサンプル中のタンパク質レべルの測定に使用され得るアツセィ技術は、 当業者によく知られている。 そのような方法には、 ラジオィムノアツセィ、 競合 的結合測定法、 ウエスタンプロット分析および E L I S Aアツセィが含まれる。 本発明の D N Aは、 本発明の夕ンパク質またはそのべプチドフラグメントをコー
ドする DN Aまたは mRN Aにおける異常を検出するのに使用できる。本発明は、 個体における上記 (1)、 (2) または (7) に記載のタンパク質の発現に関連し た疾患または疾患への感受性を診断する方法に関する。 該方法は、 タンパク質を コードするポリヌクレオチド配列における変異を、 測定することを含む。
本発明の DN Aは、 本発明の DN Aを用いることによって、 本発明の夕ンパク 質またはその部分ペプチドをコードする DNAまたは mRNAの異常を検出する ことができるので、 たとえば、 該 DNAまたは mRNAの損傷、 突然変異あるい は発現低下や、 増加あるいは発現過多などの遺伝子診断に有用である。 すなわち 本発明は、 個体における該タンパク質の発現または活性に関連した、 該個体にお ける疾病または疾病への感受性の診断方法であって、
(a) 個体のゲノムにおける上記 (1)、 (2) または (7) に記載のタンパク質 をコードするヌクレオチド配列中の変異の存在または不存在を決定し、 および Z または
( b ) 該個体に由来するサンプル中での該タンパク質の発現量を分析する、 ことを含む診断方法であって、 発現するタンパク質の量が正常の 2倍以上あるい は 1 ,' 2以下の場合に病気であると診断する方法に関する。
上記 (a) により、 S T AT 6の活性化を促進するタンパク質をコードするヌ クレオチド配列に変異がある場合は、 該変異が S T AT 6の発現または活性に関 連した疾病を引き起こす可能性がある。 あるいは、 (b) により、 被験者における 前記 (1)、 (2) または (7) のタンパク発現量を測定し正常値と異なる値を示 す場合は、 STAT 6の活性化を促進する本発明の新規タンパク質の発現量異常 が S TAT 6の発現または活性に関連した疾病の原因である可能性がある。 ここ で、 (a) の S TAT 6の活性化を促進するタンパク質をコ一ドするヌクレオチド 配列の変異の有無を測定する方法としては、 該タンパク質をコードするヌクレオ チド配列の一部をプライマーとして、 RT— PCRを行い、 その後通常のヌクレ ォチド配列決定方法によって配列を決定し、変異の有無を検出できる。あるいは、 ? 尺ー33じ?法 ( 611017 1 0 3、 5 : 8 74— 8 7 9、 1 98 9年、 実 験医学別冊新遺伝子工学ハンドブック) によっても変異の有無を調べることがで さる。
また、 (b) のタンパク発現量を調べる方法としては、 たとえば、 前記 ( 1 6) に記載の抗体を利用する方法が挙げられる。
また、 本発明は、 本発明のタンパク質による S TAT 6の活性化を阻害または 促進する化合物のスクリーニング方法に関する。
尚、 S T AT 6の活性化を阻害する化合物は、 その作用の結果、 i n v i v oまたは i n v i t r oにおいて ST AT 6の阻害剤としての活性を有するこ ととなる。 一方、 STAT 6の活性化を促進する化合物は、 その作用の結果、 i n v i v oまたは i n v i t r oにおいて S TAT 6の活性化剤としての活 性を有することとなる。 従って、 上記スクリーニング方法は、 STAT 6の阻害 剤または活性化剤としての活性についてスクリ一二ングするものであり、 上記化 合物は、 S TAT 6の阻害剤または活性化剤としての活性を有する化合物である。 上記スクリ一ニング方法は、
(a) S TAT 6の活性化を促進する作用を有するタンパク質をコードする遺伝 子および検出可能なシグナルを提供し得る成分を細胞に提供する工程、
(b) 該遺伝子が形質転換された細胞内で発現可能となる条件下で形質転換され た宿主細胞を培養する工程、
(c) 該形質転換された細胞と 1あるいは複数個の被検化合物とを接触させるェ 程、
(d) 検出可能なシグナルを検出する工程、 および、
(e) 該検出可能なシグナルにより活性化剤化合物および,''または阻害剤化合物 を単離または同定する工程、
を含む。
好ましくは、シグナルを正常より 2倍以上に増加させる化合物を活性化剤とし、 8 0 以下に減少させる化合物を阻害剤化合物として単離または同定する。
検出可能なシグナルを提供し得る成分としては、 たとえばレポーター遺伝子が 挙げられる。 レポ一ター遺伝子は、 テストを行なう転写因子の活性化を直接検出 するかわりに用いられるもので、 調べたい遺伝子のプロモータ一をレポ一ター遺 伝子につなぎ、 レポーター遺伝子の産物の活性を測定することによってプロモ一 夕一の転写活性の解析を行なうものである (バイオマニュアルシリーズ 4、 羊土
社 ( 1 994))。
レポ一夕一遺伝子としては、 その発現産物の活性または生産量 (mRNAの生 産量も含まれる) を当業者が測定可能なものであれば、 いかなるペプチド、 タン パク質をコードする遺伝子も用いることができる。 たとえば、 クロラムフエニコ ールァセチルトランスフェラーゼ、 3—ガラクトシダーゼ、 ルシフェラーゼ等の 酵素活性を測定することで利用できる。 STAT 6の活性化を評価するのに用い るレポータープラスミ ドとしては、 S T AT 6認識配列をレポーター遺伝子の上 流に組み込んだものであればよく、 たとえば CD 23や g e r m 1 i n e Cィプ シロン転写開始部位由来の配列が利用できる。 あるいは、 J . B i o l . Ch e m. 275, 26500 -26 506 (2000)、 J. Ex p. Me d. 1 90, 1 837 - 1 848 (1 999)、 J . I mm u n o 1. 1 50, 540 S - 54 1 7 (1993)、 J . I mmu n o l . 1 57, 20 58- 2065 (1996) に記載のレポ一夕一プラスミドが例示される。
宿主細胞としては、 S TAT 6の活性化の促進を検出し得る細胞であればよく、 好ましくは、 哺乳動物細胞であり、 たとえば N I H 3 T 3細胞、 He pG2細胞 等が好適に用いられる。形質転換及び培養に関しては、上記に記載の通りである。
S TAT 6の活性化を阻害または促進する化合物のスクリーニングは、 具体的 には、たとえば、一定時間培養した形質転換細胞に、被験物質を任意の量添加し、 一定時間後の該細胞が発現するレポ一夕一活性を測定し、 被験物質を添加しない 細胞のレポ一タ一活性と比較することにより、 S T A T 6の活性化を阻害または 促進する化合物をスクリーニングすることができる。 この際、 必要に応じて適切 な刺激、 例えば I L一 4添加等、 を同時に行っても良い。 レポーター活性の測定 は、 当業者に公知の方法 (たとえばバイオマニュアルシリーズ 4、 羊土社 (1 9
94)) で行なうことができる。 スクリーニングの被検物質には特に制限はなく、 低分子化合物、 ペプチドなどが挙げられる。 被検化合物は、 人工的に合成したも のであっても、 天然に存在するものであっても良い。 また単一物質でも、 混合物 でもい。 検出可能なシグナルとしては、 上記レポーター遺伝子の他に、 STAT
6の活性化によつて結果的に発現が誘導されることが知られている、 たとえば I
L一 1レセプターアンタゴニスト、 CD 23、 MHCクラス I I、 STAT 6の
遺伝子の mRNA量あるいはタンパク量を測定しても良い。 また、 核抽出物を用 いたゲル移動度シフト法等の DN Aとタンパク質の結合を検出する手法により、 活性化 STAT 6の定量を行うこともできる。 あるいは、 細胞抽出液を用いた S TAT 6のリン酸化の定量を行っても良い。
mRNA量の測定は、 たとえばノーザンハイブリダィゼ一ションゃ RT— PC R法などが挙げられる'。 夕ンパク量の測定はたとえば抗体を用いる方法が挙げら れる。 抗体は公知の方法によって作製しても良いし、 市販のもの (たとえば和光 純薬工業株式会社) を使用することもできる。
また、 以下の (a) 〜 (f) の工程により医薬組成物を製造することも可能で ある。
(a) STAT 6の活性化を促進する作用を有するタンパク質をコードする遺伝 子および検出可能なシグナルを与えることができる成分を細胞に提供する工程、
( b ) 該遺伝子が形質転換された細胞内で発現可能となる条件下で形質転換され た宿主細胞を培養する工程、
(c) 該形質転換された宿主細胞と 1あるいは複数個の化合物とを接触させるェ 程、
(d) 検出可能なシグナルを測定する工程、
(e) 該検出可能なシグナルにより活性化剤化合物および/または阻害剤化合物 を単離または同定する工程、 および
(f ) 単離または同定された化合物を医薬組成物として最適化する工程。
本発明のタンパク質は、 以下の工程により、 該タンパク質のァゴニスト、 アン 夕ゴニストまたは阻害剤を、 構造を基礎にして設計する方法に使用してもよい。
(a) まず、 タンパク質の三次元構造を決定する工程、
(b) ァゴニスト、 アンタゴニス卜または阻害剤の反応性部位または結合部位と 思われる部位の三次元構造を推論する工程、
( c ) 推論した結合部位または反応性部位に結合するかあるいは結合すると予測 される候補化合物を合成する工程、 および
(d) 該候補化合物が本当にァゴニスト、 アン夕ゴニストまたは阻害,剤であるか 否かを試験する工程。
また本発明は、 上記スクリーニングによって得られた化合物を含む。 しかしな がら、 本発明のスクリーニング方法は、 上記の方法に限定されるものではない。 さらに、 上記 (1 4 ) に記載の方法により医薬組成物を製造する方法も含む。 該候補化合物には特に制限はなく、 低分子化合物、 ペプチドなどが挙げられ、 また、 人工的に合成したものであっても、 天然に存在するものであっても良い。 上記スクリ一二ングによって得られた化合物は、 S T A T 6の活性化を阻害また は促進する作用を有しているので、 S T A T 6の望ましくない活性化あるいは不 活性化に起因する疾患を治療または予防するための医薬として有用である。 混合 物から目的化合物を単離、 精製するには、 自体公知の方法、 例えば濾過、 抽出、 洗浄、 乾燥、 濃縮、 結晶化、 各種クロマトグラフィー等を適宜組み合わせて行な うことができる。 化合物の塩を取得したい時は、 化合物が塩の形で得られる場合 にはそのまま精製すれば良く、 また遊離の形で得られる場合には、 通常の方法に より適当な溶媒に溶解または懸濁し、 所望の酸または塩基を添加し、 塩を形成さ せて単離精製すれば良い。 本発明の方法を用いて得られる化合物またはその塩を 医薬組成物として最適化する工程としては、 例えば以下のように常法により製剤 化することが挙げられる。 すなわち活性成分として有効な量の上記化合物または その薬理的に許容される塩と、 薬理的に許容される担体とを混合すれば良い。 ま た製剤化は選択された投与様式に適した形態が選ばれる。 経口投与に適した組成 物としては、 錠剤、 顆粒剤、 カプセル剤、 丸剤、 および散剤などの固体形態、 溶 液剤、シロップ剤、エリキシル剤、および懸濁液剤などの液体形態が挙げられる。 非経口投与に有用な形態としては、 無菌溶液剤、 乳剤、 および懸濁液剤が挙げら れる。 上記の担体としては、 例えばゼラチン、 乳糖、 グルコース等の糖類、 コー ン -小麦 *米 * とうもろこし澱粉等の澱粉類、 ステアリン酸等の脂肪酸、 ステア リン酸カルシウム ·ステアリン酸マグネシウム等の脂肪酸塩、 タルク、 植物油、 ステアリンアルコール ·ベンジルアルコール等のアルコール、 ガム、 ポリアルキ レングリコール等が挙げられる。これらのうち液状担体の例としては、一般に水、 生理食塩水、 デキストロースまたは類似の糖溶液、 エチレングリコール、 プロピ レングリコ一ル、 ポリェチレングリコール等のグリコール類が挙げられる。

STAT 6は、 アレルギー性疾患、 炎症、 自己免疫疾患、 糖尿病、 高脂血症、 ガン及びウィルス感染などの多種の病理学的状態におけるその関与のため、 薬物 デザィン及び治療介在のための魅力的な標的である。 多数の実験が、 STAT 6 活性の阻害が深い生理学的作用を有し得ることを示している 〔たとえば、 Na t u r e 380, 627— 630 (1 996)、 N a t u r e 380, 630 - 633 ( 19 96), I mmun i t y 4, 3 1 3 - 3 19 (1 996)、 J .

Me d. 1 87, 1 537- 1 542 (1 998)〕。 本明細書中に報告する S TAT 6の活性化を促進する作用を有する新規タンパ ク質の発見により、 異常な S TAT 6機能を阻害する新しい方法が提供された。 さらなる具体例において、 本発明は、 S TAT 6の活性化を阻害するための前記 の S T A T 6の活性化を促進する作用を有する夕ンパク質の機能を阻害する化合 物を用いる方法に関する。 上記スクリーニング方法によって得られた、 STAT 6の活性化を阻害する化合物は、 たとえばアレルギー性疾患、 炎症、 自己免疫疾 患、 糖尿病、 高脂血症、 感染症 (1例として H I V感染症)、 ガンなどの、 S TA T 6の望ましくない活性化によつて特徴つけられる疾患を治療または予防する医 薬として有用である。
逆に、 S T A T 6の活性化が T h 2細胞への分化を促進することから、 T h l が機能亢進している疾患、 例えば、 多発性硬化症やインシュリン依存型糖尿病な どの臓器特異的自己免疫疾患やリューマチの症状軽減や治療の可能性も考えられ る。 よって、 上記スクリーニング方法によって得られた、 S T A T 6の活性化を 促進する化合物はこれらの疾患の治療または予防のための医薬として有用である, 更に、本発明のタンパク質をコードする遺伝子は、癌、 自己免疫疾患、糖尿病、 高脂血症、 アレルギー性疾患、 および炎症性応答を初めとする様々な疾患の治療 を目的とした遺伝子治療にも有用である。 遺伝子治療とは、 疾病の治療を目的と して、 遺伝子または遺伝子を導入した細胞をヒ卜の体内に投与することを意味す る。 本発明のタンパク質ゃ該タンパク質をコードする DNAは、 診断目的にも使用 できる。
本発明のスクリーニング方法を用いて得られる化合物またはその塩を上述の医 薬組成物として使用する場合、 常套手段に従って実施することができる。 たとえ ば、 錠剤、 カプセル剤、 エリキシル剤、 マイクロカプセル剤、 無菌性溶液、 懸濁 液剤などとすることができる。 このようにして得られる製剤は安全で低毒性であ るので、 たとえば、 ヒ卜や哺乳動物 (たとえば、 ラット、 ゥサギ、 ヒッジ、 ブ夕、 ゥシ、 ネコ、 ィヌ、 サルなど) に対して投与することができる。 患者への投与は、 たとえば、 動脈内注射、 静脈内注射、 皮下注射など当業者に公知の方法により行 いうる。 投与量は、 患者の体重や年齢、 投与方法などにより変動するが、 当業者 であれば適当な投与量を適宜選択することが可能である。 また、 該化合物が DNA によりコードされうるものであれば、 該 DNAを遺伝子治療用べクタ一に組込み、 遺伝子治療を行うことも考えられる。 投与量、 投与方法は、 患者の体重や年齢、 症状などにより変動するが、 当業者であれば適宜選択することが可能である。 す なわち本発明は、 上記化合物を有効成分として含有する医薬に関する。
さらに、 上記化合物は、 アレルギー性疾患、 炎症、 自己免疫疾患、 糖尿病、 高 脂血症、 ウィルス性疾患、 感染症、 ガンなどの、 S T A T 6の望ましくない活性 化によって特徴つけられる疾患の治療または予防する医薬として有用である。 す なわち本発明は、 上記化合物を含むアレルギー性疾患、 炎症、 自己免疫疾患、 糖 尿病、 高脂血症、 ウィルス性疾患、 ガンなどの医薬に関する。 具体的には、 例え
ば、 慢性関節リウマチ、 変形性関節症、 全身性エリテマトーデス、 糖尿病、 敗血 症、 喘息、 アレルギー性鼻炎、 虚血性心疾患、 炎症性腸疾患、 くも膜下出血、 ゥ ィルス肝炎、 エイズ、 などに対する治療及び予防薬として有用である。
さらにまた、 本発明は、 アレルギー性疾患、 炎症、 自己免疫疾患、 糖尿病、 高 脂血症、 ウィルス性疾患、 ガンなどの医薬の製造における上記 (14) 記載の方 法により製造された医薬組成物の使用も含む。
また本発明は、 上記 (3) 〜 (6) に記載の遺伝子に対するアンチセンスオリ ゴヌクレオチドも含む。 アンチセンスオリゴヌクレオチドは、 標的とした遺伝子 配列に対して相補的な配列を持つオリゴヌクレオチドを用いて、 タンパク質への 翻訳、 細胞質への輸送、 あるいは全体的な生物活性機能に必要な他の活性等の R N Aの機能を阻害することによって、標的遺伝子の発現を抑制することができる。 この際、 アンチセンスオリゴヌクレオチドとしては、 RNAを用いても良いし、 DNAを用いても良い。 本発明の DNA配列は、 本発明のタンパク質をコードす る遺伝子から転写された mRNAとハイプリダイズし得るアンチセンスオリゴヌ クレオチドを作製するために使用できる。 一般にアンチセンスオリゴヌクレオチ ドが、その遺伝子の発現に対して抑制的に作用す'ることは公知での事実である(た とえば、 細胞工学 Vo l . 1 3 No. 4 ( 1994))。 本発明のタンパク質 をコードする遺伝子に対するアンチセンスコード配列を有するオリゴヌクレオチ ドは、 標準の方法で細胞内に導入することができ、 該オリゴヌクレオチドは、 本 発明のタンパク質をコードする遺伝子の mRNAの翻訳を効果的に遮断して、 そ の発現を遮断して、 望ましくない作用が阻害される。

スホジエステル結合が、 その活性が調節されるべき R N Aが位置する細胞の領域 に浸透する組成物の能力を高める機能を有する構造により置換される。
このような置換は、 ホスホロチォェ一卜結合、 ホスホロアミデート結合、 メチ ルホスホネ一ト結合または短鎖アルキルもしくはシクロアルキル構造を含むこと が好ましい。 オリゴヌクレオチドはまた、 少なくとも幾つかの修飾されたヌクレ ォチド型を含んでいても良い。 従って、 天然に通常見いだされるもの以外のプリ ン及びピリミジンを使用していても良い。 同様に本発明の本質的な意図が実行さ れる限り、ヌクレオチドサブユニッ トのフラノシル部分を修飾することもできる。 このような修飾の例は、 2, 一〇一アルキル—、 及び 2 ' —ハロゲン置換ヌクレ ォチドである。 本発明において有用な幾つかの糖部分の 2 ' 位の修飾の例は、 〇 H、 SH、 S CH3、 OCH3、 OCN、 または O (CH2) nCH3 (ここで nは 1から約 1 0'である)、及び同様の特性を有する他の置換基である。全てのこのよ うな類似体は、 本発明の遺伝子の m R N Aとハイブリダィズしてその R N Aの機 能を阻害する機能を果たす限り、 本発明に包含される。
本発明のオリゴヌクレオチドは、 約 3から約 50ヌクレオチドを含み、 約 8か ら約 25ヌクレオチドを含むことが好ましく、 約 12から約 20ヌクレオチドを 含むことがさらに好ましい。 本発明のオリゴヌクレオチドは、 周知の方法である 固相合成法により作製することができる。 このような合成のための装置は、 Ap p l i e d B i o s y s t emsを含む幾つかの業者により販売されている。 ホスホチォェ一ト等の他のオリゴヌクレオチドの製造も当業者に公知の方法で作 製できる。
本発明のォリゴヌクレオチドは、 本発明の遺伝子から転写される m R N Aとハ ィブリダイズできるように設計される。 与えられた遺伝子の配列に基づいてアン チセンスオリゴヌクレオチドを設計する方法は、 当業者であれば容易である 〔た とえば、 村上および牧野:細胞工学 Vo l . 13 No. 4 P 2 59 - 266

5 - 95 ( 1 996)〕。 最近の研究は、 mRNAの 5 ' 領域、 好ましくは翻訳開 始部位を含む領域に設計されたアンチセンスオリゴヌクレオチドが、 遺伝子の発
現の阻害に最も効果的であることを示唆している。 アンチセンスオリゴヌクレオ チドの長さは、 1 5から 30ヌクレオチドが好ましく、 20から 25ヌクレオチ ドがより好ましい。 ホモロジ一検索で他の mRNAとの相互作用がないこと、 ォ リゴヌクレオチド配列内で二次構造を取らないことを確認しておくことは重要で ある。 設計したアンチセンス分子が機能したかどうかの評価は、 適当な細胞を用 いて、 該細胞にアンチセンス才リゴヌクレオチドを導入し、 当業者には公知の方 法で、 対象 mRNAの量 (たとえば、 ノーザンブロッ トまたは RT— PC R法)、 あるいは対象タンパク質の量(たとえば、 ウエスタンブロッ卜または蛍光抗体法) を測定することにより、 発現抑制の効果を確認できる。
一方、 三重らせん形成 (トリプル ·ヘリックス技術) は、 核内の DN Aを標的 とした、主に転写の段階での遺伝子発現制御方法である。ォリゴヌクレオチドは、 主に転写に関与する遺伝子領域に設計され、 それにより、 転写及び本発明のタン パク質の産生を抑える。 これらの RNA、 DNA、 オリゴヌクレオチドは、 公知 の合成装置などを用いて製造することができる。
本発明のオリゴヌクレオチドは、 標的核酸配列を含む細胞に、 たとえばリン酸 カルシウム法、 リポフエクシヨン法、 エレクトロボレ一シヨン法、 マイクロイン ジェクション法などの DNAトランスフエクション法、 またはウィルスなどの遺伝 子導入ベクターの使用を含む遺伝子導入法のいずれを用いて導入してもよい。 適 切なレトロウイルスベクターを用いてアンチセンスオリゴヌクレオチド発現べク タ一を作製し、 その後、 該発現ベクターを細胞と i n V i v oまたは e x v i v oで接触させることにより、 標的核酸配列を含む細胞に導入できる。
本発明の DN Aは、 アンチセンス RNAz DNA技術またはトリプル ·ヘリッ クス技術を用いて、 本発明のタンパクを介する S TAT 6の活性化の促進を阻害 するのに使用できる。
本発明のタンパク質をコードする遺伝子のアンチセンスオリゴヌクレオチドは、 たとえばアレルギー性疾患、 炎症、 自己免疫疾患、 糖尿病、 高脂血症、 感染症 (た とえば、 H I V感染症)、 ガンなどの、 STAT 6の望ましくない活性化によって 特徴つけられる疾患を治療または予防する医薬として有用である。 すなわち、 本 発明は、 上記アンチセンスオリゴヌクレオチドを有効成分として含有する医薬で
ある。 また、 本発明のアンチセンスオリゴヌクレオチドは、 ノーザンハイブリダ ィゼ一シヨン法または P C R法を用いてそれらの疾病の検出に利用することもで きる。
本発明は、 STAT 6の活性化を阻害するリボザィムも含む。 リポザィムは、 核酸のヌクレオチド配列を認識して、核酸を切断する活性を持つ RN Aである(た とえば、 柳川弘志 実験医学バイオサイエンス 1 2、 RNAのニューエイジ)。 リ ボザィムは、 選択された標的 RNA、 たとえば本発明のタンパク質をコードする mRN Aを開裂するように製造することができる。 本発明のタンパク質をコード する DNAのヌクレオチド配列を基に、 本発明のタンパク質の mRNAを特異的 に切断するリボザィムを設計することができ、 かようなリボザィムは本発明の夕 ンパク質の m R N Aに対して相補的な配列を有し、 該 m R N Aと相補的結合し、 ついで該 mRN Aが開裂され本発明のタンパク質の発現が減少し (または完全に 発現せず)、発現減少のレベルは標的細胞内でのリボザィム発現のレベルに依存し ている。
よく用いられるリポザィムには、 ハンマーヘッド型とヘアピン型の 2種類があ り、 特にハンマ一へッ卜型リボザィムは切断活性に必要な一次構造や二次構造が よく調べられており、 当業者であれば、 本発明のタンパク質をコードする DNA のヌクレオチド配列情報のみで容易にリポザィムの設計が可能である〔たとえば、 飯田ら:細胞工学 Vol.16 No.3, P438-445 (1997)、 大川&多比良:実験医学 Vol.12
No.12 p83- 88(1994)〕。 ハンマ一ヘッドリポザィムは、 標的 RN Aと相補鎖を形成 する 2ケ所の認識部位 (認識部位 I と認識部位 I I ) と活性部位からなる構造を なし、 標的 RNAと認識部位で相補対を形成した後、 標的 RNAの NUXの配列
(N : Aまたは Gまたは Cまたは U、 :八または〇または11) の 3 ' 末端側で 切断することが知られており、 特に GUC (あるいは GUA) が一番高い活性を 持つことが知られている 〔たとえば Koizumi, Mら: Nucl. Acids
Res.17, 7059-7071 (1989) , 飯田ら :細胞工学 Vol.16 No.3, ρ438- 445 (1997)、 大 川 &多比良:実験医学 Vol.12 No.12 p83- 88 (1994)、 川崎 &多比良:実験医学

そこでまず、 本発明の DNA配列の中から GTC (または GTA) の配列を探
し出し、 その前後で数ヌクレオチドから十数ヌクレオチドの相補対をつくること ができるようにリポザィムを設計する。 設計したリボザィムの適切性の評価は、 たとえば、 大川&多比良の文献 〔実験医学 Vol.12 No.12 p83- 88 (1994) 〕 に記載 の方法によって、 作製したリボザィムが、 イン ビトロで標的 mRNAを切断で きるかどうかを調べることで評価できる。 リボザィムの調製は、 RNA分子を合 成するのための当分野で周知の方法により調製する。
別法としては、 リボザィムの配列を DNA合成機で合成し、 たとえば T 7或い は S P 6のような適切な RNAポリメラーゼプロモー夕を有する多種のベクター に組み込み、 イン ビトロで酵素的に RNAを合成させる方法が挙げられる。 こ れらのリボザィムは、 たとえばマイクロインジェクション法などの遺伝子導入方 法によって細胞内に導入できる。 あるいは別の方法として、 リボザィム DN Aを 適当な発現べクタ一に組み込んで、 株細胞、 細胞或いは組織内に導入する。 選択 された細胞中にリボザィムを導'入するのに、 適切なベクタ一を使用することがで き、 たとえばプラスミドベクター、 動物ウィルス (たとえばレトロウイルス、 ァ デノウィルス、 ヘルぺスあるいはワクシニアウィルス) ベクターがこれらの目的 に通常用いられるこれらのリボザィムは、 本発明の夕ンパク質で仲介される S T AT 6の活性化の促進を阻害する作用を有する。
本発明である S T AT 6の活性化を促進する作用を有する夕ンパク質をコ一ド する D N Aは、 オリゴキヤッピング法を用いて完全長 c D N Aライブラリーを作 製する方法および該機能を有するタンパク質の存在を示すシグナル因子を用いる 方法からなる取得方法により取得された。シグナル因子には >たとえばレポーター 遺伝子が挙げられる。
機能を有する遺伝子 (cDNA) を多数取得するためには、 不完全長のものが 多い c DNAライブラリ一を用いると効率が悪い。 したがって、 全体のクローン の中で、 完全長のものの割合が高いライブラリーが必要となる。 完全長 c DNA は遺伝子から出来る mRNAの完全なコピーのことである。 オリゴキヤッビング 法で作製した c DNAライブラリ一は、 完全長 cDNAの割合が 50〜80 %で あり、 従来の方法で作製された c DNAライブラリ一と比べて、 5〜1 0倍の完 全長 c DNAクローンの濃縮になっている(菅野純夫:月刊 BIO INDUSTRY Vol.16
No.11ρ19- 26)。 完全長 c DNAは、 遺伝子の機能解析においては、 タンパク質発 現のために必須なクロ一ンであり、 完全長 c DNAのクロ一ンそのものが活性測 定のための材料として極めて重要なものであるため、 遺伝子の機能解析を試みる に際して、 完全長 cDNAのクローニングは必須の要件である。 さらにその配列 を決定することで、 それがコ一ドする夕ンパク質の一次配列を確定するための重 要な情報となると同時に、 遺伝子の全ェクソンの配列も分かる。 すなわち、 完全 長 cDNAは、 遺伝子を同定する上で貴重な情報、 たとえばタンパク質の一次配 列、 ェクソン一イントロン構造、 mRNAの転写開始点、 プロモーターの位置な どを決めるための情報をも与える。
オリゴキヤッビング法による完全長 c DNAライブラリー作製は、 たとえば実 験医学別冊新遺伝子工学八ンドブック改訂第 3版 (1 999年) に記載の方法に 従い行うことができる。 機能を有するタンパク質の存在を示すレポーター遺伝子 は、 転写因子等のタンパク質因子が結合できる適切な発現制御配列部分 (1つま たは複数) と、 その転写因子等による活性化を測定できる構造遺伝子部分からな る。 構造遺伝子部分は、 その発現産物の活性または生産量 (mRNAの生産量も 含まれる) を当業者が測定可能なものであれば、 いかなるペプチド、 タンパク質 をコードする遺伝子も用いることができる。 たとえば、 クロラムフエニコ一ルァ セチルトランスフェラ一ゼ、 5—ガラクトシダ一ゼ、 ルシフェラーゼ等を用いる ことができ、 その酵素活性を測定することで利用できる。
オリゴキヤッピング法とは、 鈴木 *菅野 実験医学別冊 遺伝子工学ハンドブ ック改訂第 3版に記載のように、 BAP, TAP, RNAリガーゼにより、 キヤ ップ構造を合成オリゴに置換する方法である。
本発明の S TAT 6の活性化を有するタンパク質をコ一ドする DNAを取得す るためには、 イン ビトロ ( i n V i t r o) の系、 あるいは細胞を用いて (c e l 1— b a s e d) の系のどちらの方法でも良く、 好ましくは細胞を用いた系 である。 細胞は、 大腸菌をはじめとする原核生物、 酵母、 真菌等の微生物、 及び 昆虫や動物等の細胞のいずれでも良く、 好ましくは動物細胞であり、 2 93 EB
NA細胞、 N I H 3 T 3細胞が例示できる。
機能を有するタンパク質の存在を示すレポーター遺伝子としては、 本願明細書
に示した S TAT 6のレポ一ター遺伝子の他に、 たとえば CREB (cAMP responsive element binding protein) 結合配列あるレ は A P— 1 (activator protein- 1)結合配列をレポ一夕一遺伝子の発現制御配列部分に有するレポ一夕一 遺伝子が挙げられる。 たとえば、 CREBを活性化する機能を有する遺伝子を取 得したい場合は、 C R E B依存レポ一夕一プラスミ ドとォリゴキャッピング法で 作製した完全長 c D N Aクローンを細胞に共導入し、 その中からレポ一夕一活性 が上昇したプラスミ ドを選ぶことによって、 該目的を達成することができる。 ま た、 CREBを抑制する機能を有する遺伝子を取得したい場合は、 CREB依存 レポ一タープラスミドとオリゴキヤッビング法で作製した完全長 c DNAクロ一 ンを細胞に共導入し、 その中からレポ一夕一活性が減少したプラスミドを選ぶこ とによって、 該目的を達成することができる。 この場合、 細胞に何らかの刺激を 加えた状態で行なっても良い。 c DN Aクローンの細胞への導入は、 1クローン でも良いし、 複数のクローンを同時に導入しても良い。 あるいは、 完全長 cDN Aとレポーター遺伝子を細胞に導入した後、 細胞を I L一 4あるいは I L一 13 などで刺激し、 レポ一夕一活性の上昇の弱いクローンを選ぶことによって、 ST A T 6の活性化を抑制する機能を有する遺伝子を取得するためのスクリー二ング 系を構築することもできる。

(実施例 1 ) オリゴキヤッピング法を用いた完全長 c D N Aライブラリ一の作
(1 ) ヒト肺線維芽細胞 (C r yo NHL F) からの RNA調製 ヒ卜肺線維芽細胞 (C r yo NHL F :三光純薬株式会社より購入) を、 添 付のプロトコ一ルに従って培養した。 1 0 cmシャーレ 50枚まで継代培養した 後、 セルスクレーパーで細胞を回収した。 次いで、 回収した細胞から RNA抽出 用試薬 I SOGEN (二ツボンジーンより購入) を用いて全 RNAを取得した。 取得の具体的方法は、 試薬のプロ卜コールに従った。 次いで、 オリゴ— dT セ ルロース カラムを用いて、 全 RNAからポリ A + RNAを取得した。 ポリ A + RNA取得の具体的方法は、 上記 Ma n i a t i sの実験書に従った。
(2) オリゴキヤッピング法による完全長 c DNAライブラリー作製
上記ポリ A + R N Aから、 オリゴキヤッピング法により完全長 c D N Aライブ ラリーを作製した。 ォリゴキヤッピング法による完全長 c D N Aライブラリ一作 製の具体的方法は、 菅野らの方法 〔たとえば、 Ma r uy ama, K. & S u g a n o, S . Ge n e, 1 38 : 1 71 - 174 (1 994)、 S u z u k i . Y. e t a 1. Ge n e, 200 : 1 49 - 1 56 (1 997)、 鈴木 ·菅野 実 験医学別冊 遺伝子工学ハンドブック改訂第 3版〕 に従って作製した。
(3 ) プラスミ ド DNAの調製
上記実施例で作製した完全長 c DN Aライブラリ一を、 エレクトロポレーショ ン法によって大腸菌 TOP 10株に形質転換した後、 100 g 'm 1アンピシ リンを含有する L B寒天培地に塗布し、 37°Cで一晩インキュベートした。 続い て、 アンピシリン含有 LB寒天培地上で生育した大腸菌のコロニーから、 Q I A 0£1^社の<31八 6 1 1 96 U l t r a P I a sm i d K i tを用い てプラスミドを回収した。具体的方法は、 Q I Awe 1 1 96 U l t r a P 1 a s m i d K i tに添付のプロトコールに従った。
(実施例 2) STAT 6の活性化を促進する作用を有する DN Aのクローニン グ
(1) STAT 6の活性化を促進する作用を有するタンパク質をコードする c
DNAのスクリーニング
N I H3 T 3細胞 (大日本製薬より購入) を細胞培養用 96穴プレートに 1 X
104C e 1 1 sZwe 1 1 となるように、 10 % F B S存在下の I MDM培地
を用い、 24時間 37 °Cで培養した (5%C〇2存在下)。 次いで、 FuGENE
6 (R o c h e社より購入) を用いて、 STAT 6応答配列を有するルシフェラ ーゼレポータープラスミ ド N4X 8— l u c 1 00 n gと、上記実施例 1. (3) で調製した完全長 cDNA2 ^ 1を 1ゥエルに共導入した。 導入の方法は添付の プロ.トコールに従った。 S T A T 6応答配列を有するルシフェラーゼレポーター プラスミ ド N4 X 8— 1 u cは以下の通りに構築した。

C CTGGGAAGAAGAGC— 3 ' (配列番号 486)) を合成した。 合成ォ リゴヌクレオチドを各々 1 n g / a 1 となるように滅菌水で溶解し、 1 0 μ 1ず つ混ぜ、 さらに滅菌水で 32 1に容量を合わせた。 その溶液を 90 °Cで 5分間 加熱後、 .徐々に室温まで冷却し二本鎖オリゴヌクレオチド溶液を調製した。 その 溶液を T 4ポリヌクレオチドキナーゼ (宝酒造) を使用して、 添付のマニュアル に従い反応を行った後、 常法に従い反応産物を精製した。 一方、 pGL 3— P r omo t e rベクター (プロメガ) の S V40プロモーター領域を H i n d I I

Aリガ一ゼ (G I BCOz'BRL) を使用して挿入した。 得られたクローンを常 法に従い塩基配列を決定し、 ォリゴヌクレオチド断片が複数挿入されたクローン を選択した。 最大 4個の断片が挿入されたクローンが得られ、 N4X4— 1 u c. と命名した。 N 4 X 4— 1 u cプラスミ ドから、 X h o I部位と B g 1 I I部位 を用いて、 4個連結した DNA断片を切り出し精製後、 pB l u e s c r i p t
I I KS + (S t r a t a g e n e) の B amH I部位と X h o I部位へ挿入 した。 このプラスミドより、 Kp n Iと S p e Iを用いて 4個連結した DNA断 片を切り出し、 N4X4— 1 u cプラスミドの Kp n I部位と Nh e I部位に挿 入し、 最終的に N4\ 8— 1 u cを得た。
プラスミ ド N4 X 8— 1 u cおよび上記完全長 c DNAを共導入し、 48時間 37 °Cで培養後、 さらに最終濃度 0. 5 n g/m 1のマウス I L一 4 (免疫生物 研究所) を添加し 6時間培養した。 ロング夕一ムルシフェラーゼアツセイシステ ム、 ピツカジーン LT2. 0 (東洋インキ社) を用いて添付されている説明書に 従い、 S TAT 6のレポ一夕一活性 (ルシフェラ一ゼ活性) を測定した。 なおル シフェラ一ゼ活性は、 P e r k i n E 1 m e r社の W a 1 1 a c AR VOT MS T 1420 MULT I LABEL C〇 UN T E Rを用いて行った。
(2) ヌクレオチド配列の決定
上記スクリーニングを 1 1 5、 000クローン行い、 ルシフェラーゼ活性が対 照実験 (完全長 c DN Aの代わりに、 空べクタ一 pME 1 8 S— FL 3を導入し た細胞のルシフェラーゼ活性) と比べて 3倍以上上昇しているプラスミ ドを選抜 し、 まず、 クローニングされている c DNAの 5 ' 側 (シークェンスプライマ一 : 5,— CTTCTGCTCTAAAAGCTGCG— 3,(配列番号 487)と 3, 側 (シークェンスプライマ一 : 5 ' — CGACCTGCAGCTCGAGCAC A— 3 ' (配列番号 4 S 8 ) ) からそれぞれ o n e— p a s sシークェンスを行な い、 できる限り長く決定した。 なお、 ヌクレオチド配列決定のための試薬や方法 は、 Th e rmo S e q u e n a s e I I Dy e Te rm i n a t o r Cy c l e S e q u e n c i n g K i t (アマシャム フアルマシア社)、 あ るいは B i gDy e Te rm i n a t o r Cy c l e S e qu e n c i n g F S Re a dy R e a c t i o n K i t (アプライ ドバイオシステム ズ社) を用い、 AB I PR I SM 377シークェンサ一、 あるいは、 AB I PR I SM 31 00シ一クェンサーを用い、 各々キッ卜に添付されている説明 書に従って行なった。
(3) 得られたクローンのデータベース解析
得られたヌクレオチド配列について、 G e n B a n kに対する B LAS T (Basic local alignment search tool) 〔S. F. Al tschul et al. , J. ol. Biol. , 215: 403-410 (1990) 〕 検索を行なった。 その結果、 242クローンが STAT 6の活性化を促進する作用を有する新規のタンパク質をコードする 1 1 2種類の 遺伝子であった。

(実施例 3) S TAT 6の活性化の促進を阻害する化合物のスクリーニング N I H 3 T 3細胞を細胞培養用 9 6 w e 1 1プレートに、 1 X 1 04 C e 1 1 s / 1 00 1 'w e 1 1の細胞数になるように、 1 0 ¾ F B S存在下の I MD M培地にまき、 5 %C〇2存在下、 3 7°Cで 24時間培養した。 次いで、 F uG ENE 6を用いて、 上記実施例 2で得た、 配列番号 3、 1 7、 1 9、 2 1 8、 4 32若しくは 47 2の STAT 6の活性化を促進する作用を有するタンパク質を コードするヌクレオチド、 または配列番号 64のヌクレオチドを含有するプラス ミド 3 0 n gと、 S TAT 6応答配列を有するルシフェラ一ゼレポータープラス ミ ド l O O n gを lwe l 1 に共導入した。 48時間後、 プロテインキナーゼ阻 害剤であることが知られている AG 1 8、 AG490、 あるいはスタウロスポリ ン ( C A L B I O C H E Mより購入) をそれぞれ終濃度 2 0 iM, 2 0 tMf 3 0 Mになるように培養液中に加えた。 37 °Cで 30分培養後、 さらにマウス I L— 4を最終濃度 1 n g , m 1になるように添加し 6時間培養し、 ピツカジーン LT 2. 0を用いてレポーター活性を測定した。 その結果、 AG 1 8, AG 49 0, スタウロスポリンはコント口一ルプラスミ ド p c DNA3. 1 (+ ) (インビ トロジェン社) 導入ゥエルには全くレポーター遺伝子の発現を抑制しないが、 酉己 列番号 3、 1 7、 1 9、 2 1 8、 43 2若しくは 47 2の S TAT 6の活性化を 促進する作用を有するタンパク質をコードするヌクレオチド、 または配列番号 6 4のヌクレオチドを含有するプラスミ ドを導入したゥエルにおいてはレボー夕一 遺伝子の発現を抑制した (図 1〜 7 )。
同様に、 他のアミノ酸配列をコードする遺伝子でも、 同様の方法によりレポ一 ター遺伝子の発現の抑制を確認できる。 産業上の利用の可能性
本発明により、 産業上有用性の高い STAT 6の活性化を促進する作用を有す るタンパク質やそれらの遺伝子が提供された。 本発明のタンパク質やそれらの遺 伝子により、 STAT 6の過剰な活性化、 又は阻害が関与する疾患の治療や予防 に有用な化合物のスクリーニング、 さらにそのような疾患の診断薬を作製するこ とが可能である。 更に本発明の遺伝子は、 遺伝子治療に用いられる遺伝子ソース
としても有用である。 本明細書で引用した全ての刊行物、 特許及び特許出願をそのまま参考として本 明細書にとり入れるものとする。
