Description

Verfahren zur Herstellung von Polyestern aus Hydroxycarbonsäuren und Polyolen durch PolykondensationProcess for the preparation of polyesters from hydroxycarboxylic acids and polyols by polycondensation
Hintergrundbackground
The invention relates to a process for the production of polyesters from hydroxycarboxylic acids and polyols, the polyesters obtainable by this process and their use as depot matrix material for medicinal substances.
Vorzugsweise werden Zinnoctoat und Aluminiumtriisopro- pyloxid verwendet.WO 95/23175 describes branched-chain esters from a polyol containing at least one substituent with electrolytic properties with acid residues consisting of polyhydroxycarbon acids which has a molecular mass of up to 500,000. These polyol esters are produced by reacting an appropriately substituted polyol with one or more hydroxycarboxylic acids in dimer form or lactone form in the presence of a ring-opening catalyst suitable for polymerization, such as tin, zinc, tin chloride, zinc chloride tetra tetrachloride, aluminum chloride, tin octoate or aluminum triisopropyl oxide. Tin octoate and aluminum triisopropyl oxide are preferably used.
Vorzugsweise wird Zinnoctoat eingesetzt.US Pat. No. 5,922,338 also relates to polyester made up of lactic acid, glycolic acid and a polyol. Again, the use of a catalyst is taught to make these polyol esters. Tin octoate is preferably used.
The processes known in the prior art for the production of polyol-containing polyesters from lactic acid and glycolic acid have the disadvantage that the hydrophilic backbone component is only incorporated with an unsatisfactory yield and poor reproducibility. In addition, the use of catalysts, in particular tin catalysts, is problematic with regard to the toxicity of these compounds, all the more so since the process products are substances which are to be used as depot matrix material for medicinal substances.
Protei- nen, in vivo bedingen.Furthermore, the polyesters known in the prior art have long glycolic acid-lactic acid side chains, which slow polymer mass degradation in vivo and unfavorable release properties of high molecular weight active ingredients, such as e.g. Proteins, cause in vivo.
The invention is therefore based on the object of providing new polyesters which have faster polymer mass degradation in vivo and more favorable release properties of high molecular weight active ingredients.
These polyesters are said to be produced by a process in which the hydrophilic backbone component can be efficiently incorporated into the polymer, and in which the use of a catalyst can also be dispensed with, in the case of commercially available and inexpensive ones. Starting materials can be used and finally has good reproducibility.

Lactic acid and glycolic acid are particularly preferably used in defined proportions as hydroxycarboxylic acid components and dextran, DSS, PVA or DEAED as a polyol component in the PK process according to the invention, so that a dextran, DSS, PVA or DEAED-containing polylactite glycolite (hereinafter also referred to as PK-Dex-PLG, PK-DSS-PLG, PK-PVA-PLG or PK-DEAED-PLG) results. In step (i) of the process according to the invention, a mixture is first prepared from the aforementioned components, ie the polyols and the hydroxycarboxylic acids, the proportion of the polyol generally being in the range from 0.25-20% by weight and preferably 1-4 % By weight.
Das Gewichtsverhältnis des Polyols zu den Hydroxycarbonsäuren liegt im allgemeinen im Bereich von 1 :200 bis 1 :10 und vorzugsweise von 1 :120 bis 1 :20.The weight ratio of the polyol to the hydroxycarboxylic acids is generally in the range from 1: 200 to 1:10 and preferably from 1: 120 to 1:20.
The mixture is then heated to reduced temperature e.g. heated in the range of 120-200 ° C, preferably 140-170 ° C, particularly preferably 150-160 ° C, until the melt that forms is almost anhydrous.
In this process step, the pressure is preferably reduced to below approximately 30 mbar, particularly preferably to 10-30 mbar. Since this step serves to dewater the starting mixture, the reduced pressure generated in a simple manner by a water jet pump can initially be higher, that is to say approximately 200-300 mbar, and then slowly reduced further until a reduced pressure is reached, which, for example, in the Range of 10-30 mbar. The exact pressure conditions at this stage are not critical as long as the reduced pressure is suitable for dewatering the educts.
At the same time, the temperature is set so that the educts melt during the dewatering process. The optimal temperature depends on the choice of starting materials, but is typically between 140 and 170 ° C and in particular between 150 and 160 ° C.
The dewatering and melting process typically takes 1-4 hours. The exact duration of the process is largely uncritical. The dewatering and melting process is preferably carried out over 2-3 hours. In the next stage of the process according to the invention, the pressure is reduced further so that it is in the range from below 3 mbar, preferably below 2 mbar, particularly preferably from 0.5-1.5 mbar, while the temperature is raised to 140-180 ° C , preferably 150-170 ° C, is increased.
These reaction conditions are maintained over a period of 12-96, preferably 24-48 hours and the polyesters according to the invention are formed by a polycondensation reaction. After cooling, they are cleaned by methods known per se, for example by falling over, and then dried.
For this purpose, the polymer is dissolved, for example, in acetone or in a polar protic solvent, filtered and then washed in deionized water.
These polyesters produced by the process according to the invention are another aspect of the present invention.
After drying in an oil pump vacuum for a period of about two days, polyesters are obtained which have a weight-average molecular weight in the range from 3000 to 100000 Da and preferably 5000 to 100000 Da.
Die Glasübergangstemperatur der Polyester Tg beträgt im allgemeinen 20-50°C und vorzugsweise 22-48°C.Their number average molecular weight is generally 1000 to 10000 Da and preferably 1900 to 10000 Da, so that from this a polydispersity (Mw / Mn) of 1 to 20, preferably from 1.4 to 10.2 and particularly preferably from 1.4 to 6 , 5 or, very particularly preferably, from 1, 9 - 6.4 results. The glass transition temperature of the polyester Tg is generally 20-50 ° C and preferably 22-48 ° C.
Bevorzugt beträgt der Gewichtsanteil des Kohlenhydrats am Polymer 0,5-10 Gewichts%, besonders bevorzugt 0,9-3 Gewichts%.The polyesters according to the invention have a polysaccharide content of more than 0.25% by weight. The proportion by weight of the carbohydrate in the polymer is preferably 0.5-10% by weight, particularly preferably 0.9-3% by weight.
As is demonstrated in Example 7, the polyesters according to the invention have branched structures. Compared to the polyol-containing polyesters produced by ring opening polymerization (RP), the polyol-containing polyesters produced by the process according to the invention show an increased water absorption (exemplary embodiment 8, Figure 1).
The increased water absorption of the polycondensate results in a more pronounced swelling of the polymer in an aqueous environment compared to the polyol-containing polyester produced by ring-opening polymerization. This is particularly important when using such polymers as a parenteral depot formulation, since the improved swelling forms a larger pore structure, which enables diffusion-controlled release even of large molecules (e.g. peptides, proteins). In addition, bulk erosion is supported in connection with the branched structure of the polymer, which results in a further improvement in the release properties.
Another characteristic of the polyesters produced according to the invention is the rapid loss of mass through hydrolysis. As can be seen in exemplary embodiment 9 and Figure 2, this, for example in the case of the PK-DSS-PLG, begins immediately after incubation in aqueous buffers and extends almost linearly over a period of approximately 22 days. In contrast, a loss of mass occurs in RP-DSS-PLGs produced by ring-opening polymerization only after about 10 days. At this point, over 25% of the DSS-PLG polyester produced by the process according to the invention had already been broken down.
The hydrolytic bulk erosion that starts immediately with the polycondensate has the advantage of a more uniform release rate with regard to the release of macromolecules (e.g. peptides or proteins), since from the beginning there is a linear loss of mass and thus a uniform dissolution of the polymer matrix.
The polyesters according to the invention can thus be used as matrix material for the production of depot forms containing pharmacologically active substances. Examples of corresponding parenteral depot forms are implants, microparticles, microcapsules, pellets, powders or granules. However, the polyesters produced according to the invention can also be used to produce inhalable nanospheres for pulmonary administration or in the preparation of oral slow-release forms, for example suspensions. In the production of the corresponding depot forms, the active ingredient can be embedded (dispersed or dissolved) in the polyester according to the invention, the polyester can envelop the active ingredient or the polyester can be coated on the surface with the active ingredient.
US 4,389,330.The manufacture of such depot forms is known per se to a person skilled in the art and is described, for example, in standard works such as Sucker, Fuchs and Speiser, “Pharmaceutical Technology”, Georg Thieme Verlag Stuttgart, 1978, and in the patent literature, such as in WO 95/23175; US 5,648,096; US 4,166,800 or US 4,389,330.
Geeignete Sprühtrocknungs-Verfahren sind beispielsweise beschrieben in Takada et al. (J. Pharm Sei & Techn 49, 1995, 180) oder Baras et al. (Vaccine 18, 2000, 1495).In a preferred embodiment of the invention, the polyester is in the form of microparticles in which the active ingredient is embedded. The production of such microparticles is known from the prior art. Common techniques include coacervation, solvent extraction from an oil-water dispersion or spray drying. Suitable spray drying processes are described, for example, in Takada et al. (J. Pharm Sei & Techn 49, 1995, 180) or Baras et al. (Vaccine 18, 2000, 1495).
In a typical example, 0.5 g polyester is dissolved in 6 mL glacial acetic acid, 0.05 g pharmacologically active ingredient, e.g. Peptide, dissolved in 0.15 mL water and 3.0 mL glacial acetic acid and slowly dissolved in the polymer solution. The solution is sprayed in a Büchi 190 spray dryer at 60 ° C and dried until the microparticles can be obtained as a fine, free-flowing powder.
Depot forms in which the polyesters according to the invention are used have surprisingly rapid polymer mass degradation in vivo and therefore have favorable release properties. In particular, high-molecular-weight active substances, such as proteins, are released in an advantageous manner and without a “lag phase” when the polyesters according to the invention are used as the matrix material.
Konservierungsmittel, Stabilisatoren, Antioxidantien, Puffer, Dispergiermittel.Another aspect of the invention is therefore depot forms, comprising the polyesters produced by the process according to the invention. Said depot forms can be administered in one of the known ways of administration. So they are invented Polyester according to the invention, for example, suitable for subcutaneous, intradermal or intramuscular administration. The depot forms according to the invention can have further ingredients and auxiliaries which are known per se to the person skilled in the art, for example preservatives, stabilizers, antioxidants, buffers, dispersants.
Injektionsvorrichtungen, umfassen.Another aspect of the invention is sales packages which include said depot forms containing the polyesters of the invention and instructions for their use. In another embodiment of the invention, the sales packs can also include application aids, such as Injection devices include.

Beispiel 7: Gelpermeationschromatographische UntersuchungAfter the crude product had cooled, it was dissolved in 200 ml of acetone and insoluble constituents were filtered off. The product was precipitated in 2000 ml of deionized water and then dried in vacuo (<10 mbar) for 48 hours. Example 7: Gel Permeation Chromatographic Examination
The GPC examinations were coupled with a triple detection (TriSEC system, Viscotek). In this type of polymer analysis, after light permeation-chromatographic separation of the polymers, light scattering detection at 90 ° C (RALS), viscosity and Rl detection is carried out.
Mit dieser Methodik können Aussagen über Größen (Trägheitsradien Rg) und Strukturen (Verzweigungen) der Polymere erhalten werden. Die aussagekräftigste Darstellung der Ergebnisse ist in diesem Zusammenhang der Mark-Houwink-Plot (R. Walkenhorst; Drei- fachdetektion in der Gelpermeations-Chromatographie; GIT 11/98; H.G. Elias; Makromoleküle, 5. Auflage; Hüthig und Wepf; Basel (1985)).With this methodology, statements can be made about the sizes (radii of inertia Rg) and structures (branches) of the polymers. The most meaningful presentation of the results in this context is the Mark-Houwink plot (R. Walkenhorst; triple detection in gel permeation chromatography; GIT 11/98; HG Elias; Macromolecules, 5th edition; Hüthig and Wepf; Basel (1985 )).
The study was compared to a commercial lactide glycolide
Copolymer (Resomer RG502H, Boehringer Ingelheim) carried out. Table 1 summarizes the molecular weight data from the TriSEC tests.

In the Mark-Houwink plots, the DEAED (2%) PLG shows the lowest slope, the curve flattening particularly in the higher molecular range. This polymer should therefore have highly branched structures, especially in the high molecular weight range.
The Mark-Houwink plot of the resomer RG502H can serve as an internal comparison in this figure. This polymer, which is perse linear, shows a Mark-Houwink plot that is linear over the entire mass range. The slope of the curve of the PK-DSS-PLG is also generally lower than that of the resomer, and here, too, a further flattening of the Mark-Houwink plot can be clearly seen, particularly in the high-molecular range.
In summary, this investigation shows that the polycondensates examined have a clearly branched structure. As expected, the resomer RG 502H shows all properties of a linear polymer in this study.
Beispiel 8: WasseraufnahmeExample 8: Water absorption
The water sorption was determined using a hygroscopicity balance. The sample is placed in an air-conditioned weighing room and the mass increase caused by the water absorption is determined gravimetrically. All samples were incubated for 6-9 hours. The samples were initially exposed to a relative humidity of 97% at 25 ° C for 4-5 hours. By setting a constant gas flow of 150 - 200 mL / min, constant conditions were maintained during the measuring time. After treatment with moist air, dry air was passed over the samples to measure possible water desorption. Both the kinetics of water absorption and release as well as the absolute amount of water absorbed or released were determined.
A polyol-containing polycondensate (PK-DSS-PLG) was investigated in comparison to an RP-DSS-PLG produced by ring-opening polymerization.
The RP-DSS-PLG's water absorption is reversible, whereas the PK-DSS-PLG's water absorption is not reversible. The polycondensate absorbs significantly more water than the RP-DSS-PLG.
Das Ergebnis wird in Abbildung 1 dargestellt. The result is shown in Figure 1.
Beispiel 9: PolvmerabbauExample 9: Polymer degradation
The samples were incubated in various buffer solutions (citrate buffer pH 4.9, PBS buffer pH 7.1, 0.9% NaCl solution) over a period of 6 weeks at 37 ° C. At the test dates, the supernatant solution was decanted off, the remaining polymer rest washed with water and dried. The mass was then determined by weighing, and the molecular weight data were determined by means of GPC analysis.
The investigation was carried out in comparison with a DSS polycondensate (PK-DSS-PLG) and an RP-DSS-PLG produced by ring-opening polymerization.
The evaluation of both the molecular mass loss and the mass loss showed no differences between the media used.
With both polymers, the molecular weight reduction begins immediately after the start of the investigation. In the case of the RP-DSS-PLG, however, a loss of mass only arises after approx. 10 days. The result is shown in Figure 2.

(c) Kohlenhvdratgehalt(c) Carbon content
To determine the total carbohydrate content of the polymers, the polymers are hydrolyzed in 12 molar sulfuric acid. The quantitative determination as glucose is carried out after separation by anion exchange chromatography and subsequent electrochemical detection based on a calibration with a glucose reference solution.
