Supports polymériques organiques possédant des ligands mono- ou bi-dentates et leur utilisation en catalyse supportée
L'invention concerne des supports solides organiques polymériques et comprenant des ligands phosphores mono- ou bi-dentates. L'invention concerne également leur utilisation en catalyse supportée et plus particulièrement pour la réaction de couplage comme la réaction de Heck ou de Suzuki.
La réaction de Heck, découverte dans les années 1970, permet de réaliser formellement une substitution électrophile sur un alcène, une transformation impossible en chimie organique classique. Cette réaction, catalysée par des complexes de palladium(O) en présence d'une base, a été réalisée avec une grande variété d'oléfines et d'halogénures vinyliques ou aromatiques (Meijere, N. L, Angew. Chem. Int. Ed. Engl., 1994, 33, 2379). Elle présente un grand intérêt d'un point de vue synthétique et a été en particulier utilisée pour préparer des alcaloïdes (Jin, J., Weinreb, S. M., J. Am. Chem. Soc, 1991, 119, 2050). Z 2Pd<0> z

Une variante très intéressante de cette réaction est le couplage de Suzuki qui met en jeu un halogenure arylique et un organoborate généralement aromatique (Miyaura, Ν., Suzuki, N., Chem. Rev. 1995, 95, 2475). L'avantage majeur de cette réaction est qu'elle permet l'accès à des composés bisaryliques difficilement accessibles par d'autres voies. Ces derniers trouvent des applications intéressantes en chimie fine et en chimie pharmaceutique.

Le champ d'application de la réaction de Heck et du couplage de Suzuki à l'échelle industrielle serait largement étendu si des versions efficaces de ces réactions étaient disponibles en catalyse supportée.
Deux exemples viennent d'être récemment publiés : l'un concerne l'utilisation de zéolites (réaction de Heck) (Djakovitch, L., Heise, H., Kohler J. Organomet. Chem., 1999, 584, 16), et l'autre met en jeu une silice modifiée avec un ligand soufré (couplage de Suzuki) (Zhang, T.Y., Allen, M.J., Tetrahedron Lett. 1999, 40, 5813). Toutefois, la modulation de la réactivité dans les deux cas (réaction de Heck et couplage de Suzuki) passe par l'utilisation de phosphines diversement modifiées (mono- versus di-phosphines, aryl- versus alkylphosphines, ...). L'invention propose donc de nouveaux supports particulièrement utiles en tant que support en catalyse. L'intérêt majeur de cette invention est que d'une part, le recyclage de ces catalyseurs supportés ne demande aucun traitement particulier et d'autre part, il se fait sans perte d'activité catalytique.
L'invention a ainsi pour objet des supports solides organiques polymériques, de type acrylique ou amberlite, et comprenant des ligands phosphores mono- ou bi-dentates.
Selon l'invention, un support solide polymérique peut être une résine de type amberlite (Académie Press, New York 1956 ; John Wiley, New York 1958 ; Pergamon Press, 1970).
L'invention a plus particulièrement pour objet un support solide polymérique de type acrylique.
Un support solide polymérique de type acrylique peut être un copolymère acrylique tel que défini dans le brevet EP 79 842. Un tel copolymère se compose de :
- 30 à 90 %, en poids, d'un premier monomère (I) défini comme étant la maille du support et qui consiste en une N-(mét)acrylylpolyméthylèneimine de formule :

De préférence, l'invention a pour objet un support solide polymérique de type acrylique qui est un copolymère acrylique tel que défini ci-dessus. De préférence, le copolymère acrylique tel que défini ci-dessus se compose de 70 à 90 % du monomère (I), de 3 à 10 % du monomère (II) et de 10 à 20 % du monomère (III). De manière très préférentielle, ce copolymère est constitué du composé (1) pour le monomère (I) et du composé (4) pour le monomère (III), soit la résine Met-Expansin.
Le support peut être optionnellement fonctionnalisé et/ou comprendre la fixation d'un bras espaceur. Le bras espaceur peut permettre d'éloigner, si nécessaire, le point de fixation du catalyseur de la surface du support polymérique et donc de rendre le catalyseur plus accessible pour le substrat sujet à catalyse. L'introduction d'un ou plusieurs groupes fonctionnels peut par exemple faciliter la fixation du catalyseur sur ce dernier.
L'invention a donc également pour objet un support acrylique tel que défini ci-dessus,
support qui peut être optionnellement fonctionnalisé et/ou comprendre la fixation d'un bras espaceur. Pour les polyacrylamides tels que définis ci-dessus, l'introduction d'une ou plusieurs fonctions et/ou d'un bras espaceur peut se faire par addition de tout composé correspondant, contenant par exemple un (des) atome(s) de phosphore et/ou d'azote et/ou d'oxygène et/ou d'halogène et susceptible de réagir sur la fonction acide ou ester de l'agent de fonctionnalisation III. Plus particulièrement, pour la résine Met-Expansin, la réaction d'allongement ou de fonctionnalisation se fait par réaction sur la fonction acide de la β-alanine.
Ainsi un support selon l'invention peut être un copolymère tel que défini dans EP 79841 et fonctionnalisé à l'aide de l'éthylènediamine. De tels copolymères sont décrits dans le brevet EP 81408. L'invention a plus particulièrement pour objet un support correspondant à la résine Met-Expansin-NH2, résine Met-Expansin fonctionnalisée avec 1 ' éthylènediamine.
Les supports solides polymériques tels que définis ci-dessus sont particulièrement intéressants en tant que support en catalyse. L'invention a donc également pour objet l'utilisation de tels supports comme support catalytique et les catalyseurs ainsi formés. De tels catalyseurs supportés sont de préférence des complexes de métaux de transition sur un support tel que défini ci-dessus. De manière préférentielle, le métal de transition est le palladium.
L'invention a enfin pour objet l'utilisation de catalyseurs supportés tels que définis ci- dessus, pour des réactions de couplage et en particulier pour la réaction de Heck ou Suzuki.
Dans la partie expérimentale est présentée la synthèse et l'utilisation de supports polymériques organiques porteurs de phosphines liées de manière covalentes. Ces supports peuvent être utilisés sous forme de poudre fine (résine de type Met-Expansin) ou de billes (résine de type amberlite).
Les ligands phosphores monodentates (TPPS) et bidentates (PPS) sont tout d'abord fixés sur un support polymérique acrylique : Met-Expansin-NH
2 à terminaisons aminés primaires. Ce support est obtenu en deux étapes : préparation du copolymère précurseur (Met-Expansin), suivie de la fonctionnalisation par une diamine.

Ce sont des liaisons covalentes de type sulfonamides qui sont choisies comme mode de liaison ligand/Met-Expansin-NH2. L'approche ainsi développée consiste à : - protéger une phosphine possédant une fonction sulfonate sous forme thiophosphine,
- préparer la chlorosulfonylthiophosphine nécessaire pour la fixation sur Met- Expansin-NH2,
- fixer la thiophosphine sur Met-Expansin-NH2, - déprotéger la phosphine en deux étapes,
- ajouter le palladium (0) sous forme de Pd2(dba)3 pour former le complexe sur Met-Expansin,
- réaliser la réaction de Heck, et
- récupérer, par filtration, le catalyseur fixé sur Met-Expansin.
Ensuite, les mêmes ligands phosphores sont fixés, selon le schéma réactionnel décrit ci-dessus, sur un support de type amberlite (par exemple l'IRA 93) possédant des terminaisons NH2. L'avantage majeur de la méthode décrite ici est la possibilité de récupérer la totalité du catalyseur par simple filtration, et de l'utiliser plusieurs fois sans perte d'activité catalytique.
A moins qu'ils ne soient définis d'une autre manière, tous les termes techniques et scientifiques utilisés dans le présent texte, ont la même signification que celle couramment comprise par l'homme de l'art. Toutes les références mentionnées dans le présent texte (publications, demandes de brevet ou brevets) sont incorporées par référence.
Les exemples suivants sont présentés pour illustrer les procédures ci-dessus et ne doivent en aucun cas être considérés comme une limite à la portée de l'invention.

I - Préparation des catalyseurs supportés
I - A - Préparation du catalyseur Pd/TPPS-Met-Expansin-NH2
(Pd/monophosphine)
I - A - 1 - Préparation du composé TPP(S)S :
TPPS TPP(S)S
A une solution de TPPS (100 mg ; 0,175 mmol) dans le diméthylformamide (2 ml) est ajouté un excès de soufre élémentaire (56 mg ; 1,750 mmol) à température ambiante. Le mélange réactionnel est chauffé à 40° C pendant 15 minutes. La RMN du phosphore 31 révèle la présence d'un seul produit résonnant à 42 ppm
= -5 ppm). La solution est ensuite filtrée pour éliminer l'excès de soufre qui n'a pas réagi. Le filtrat est évaporé sous vide (0,01 mm Hg). La TPP(S)S est isolée sous la forme d'une poudre blanche (Rendement : voir ci-après).
I - A - 2 - Préparation du composé TPP(S)S-C1
A une suspension de TPP(S)S (précédemment préparée) dans du dichlorométhane (3 ml) sous atmosphère d'argon sont ajoutés 6 équivalents de chlorure d'oxalyle (90 μl ; 1 mmol) en présence d'une quantité catalytique de diméthylformamide (une goutte). Un dégagement gazeux (CO2) est observé, et la solution devient homogène. Le milieu réactionnel est agité pendant une heure à température ambiante. La RMN du phosphore 31
révèle la présence d'un seul produit résonnant à 40 ppm attribué à la TPP(S)S-C1. Enfin le produit est isolé sous la forme d'une poudre jaune pâle après évaporation du solvant sous vide (0,01 mm Hg).
Le rendement des deux premières étapes est estimé à 60 %.
I - A - 3 - Préparation de la résine TPP(S)S-Met-Expansin-NH :
TPP(S)S-CI TPP(S)S-Met-Expansin-NH2
A la TPP(S)S-C1 sont ajoutés la résine Met-Expansin-NH2 (1 g ; 0,7 mmol NH2) et 10 ml de pyridine. Instantanément une coloration jaune apparaît. La solution est portée à reflux de la pyridine (115° C) pendant 24 heures. La suspension est filtrée. Le solide est lavé successivement avec du toluène (3 fois 10 ml), de l'éthanol (3 fois 10 ml), de l'eau (3 fois 10 ml) et du dichlorométhane (3 fois 10 ml). La RMN du phosphore 31 du filtrat et des surnageants de lavage rassemblés et concentrés révèle la présence des traces de TPP(S)S. La résine TPP(S)S-Met-Expansin-NH2 est isolée sous forme d'une poudre marron. Le rapport chlorure de sulfonyle/fonction aminé de la résine est d'environ 1/7.
I - A - 4 - Préparation de la résine TPP(SMe S-Met-Expansin-N . :

La résine précédemment préparée est mise en suspension dans du dichlorométhane
(10 ml). Ensuite, du triflate de méthyle (1 ml ; 8,836 mmol) est ajouté à température ambiante. La suspension est chauffée à reflux du dichlorométhane (40° C) pendant 12
heures. La résine TPP(SMe)S-Met-Expansin-NH2 est isolée après filtration, lavage au tétrahydrofurane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg). La RMN du phosphore 31 du filtrat et du surnageant de lavage rassemblés et concentrés ne révèle la présence d'aucun produit phosphore.
Les atomes d'azote des liens sulfonamides ainsi que les fonctions aminés libres de la résine sont susceptibles d'être alkylés au cours de cette étape.
I - A - 5 - Préparation de la TPPS-Met-Expansin-NH, :
TPP(S e)S-Met-Expansin-NH2 TPPS- et-Expansin-NH2
La résine TPP(SMe)S-Met-Expansin-NH2 obtenue est mise en suspension dans du dichlorométhane fraîchement distillé et dégazé (10 ml), puis la tris(diméthylamino)phosphine (1 ml ; 5,502 mmol) est ajoutée à température ambiante. La suspension est laissée sous agitation magnétique pendant 3 heures. La phosphine supportée sur la résine est isolée après filtration, lavage au dichlorométhane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg).
La RMN du phosphore 31 du filtrat et du surnageant de lavage révèle la présence du tris(diméthylamino)méthylthiophosphonium [(Me2N)3PSMe]+ à 67 ppm et du tris(diméthylamino) phosphonium [(Me2N)3PH]+ à 21 ppm.
Le rendement de ces deux dernières étapes est d'environ 85 %, soit un rendement total de 50 %.
I - A - 6 - Préparation du catalyseur Pd/TPPS-Met-Expansin-NH. :
Pd/TPPS- et-Expansin-NH2
La TPPS-Met-Expansin-NH2 est mise en suspension dans du tétrahydrofurane distillé (15 ml), puis le complexe de tris(dibenzylidène-acétone)dipalladium (20 mg ; 0,022 mmol) est ajouté. Une coloration pourpre apparaît instantanément et disparaît ensuite au profit d'une couleur jaune-vert. La suspension est portée à reflux du tétrahydrofurane (67° C) pendant 12 heures. Le catalyseur au palladium est obtenu après filtration et lavage au tétrahydrofurane. La RMN du phosphore 31 du filtrat et du surnageant de lavage ne révèle la présence d'aucune trace de produit phosphore. Le catalyseur est séché sous vide (0,01 mm Hg) et est gardé sous atmosphère d'argon. La charge du palladium est estimée à 0,02 mmol par gramme de polymère.
I - B - Préparation du catalyseur Pd/PPS-Met-Expansin-NH2 (Pd/diphosphine)
I - B - 1 - Préparation du composé PP(S S :
A une solution de PPS (50 mg ; 0,058 mmol) dans du diméthylformamide (2 ml) est ajouté un excès de soufre élémentaire (56 mg ; 1,750 mmol) à température ambiante. Le mélange réactionnel est chauffé à 40° C pendant 15 minutes. La solution est ensuite filtrée pour éliminer l'excès de soufre qui n'a pas réagi. Le filtrat est évaporé sous vide (0,01 mm
Hg). La PP(S)S est isolée sous la forme d'une poudre blanche (Rendement : voir ci- après).
I - B - 2 - Préparation du composé PP(S)S-C1 :
A une suspension de PP(S)S (précédemment préparée) dans du dichlorométhane (3 ml), sous atmosphère d'argon, sont ajoutés 4 équivalents de chlorure d'oxalyle (40 μl ; 0,468 mmol) en présence d'une quantité catalytique de diméthylformamide (une goutte). Un dégagement gazeux (CO
2) est observé, et la solution devient homogène. Le milieu réactionnel est agité pendant une heure à température ambiante. Le composé PP(S)S-C1 est isolé sous la forme d'une poudre jaune pâle après évaporation du solvant sous vide (0,01 mm Hg).
Le rendement des deux premières étapes est estimé à 85 %.
I - B - 3 - Préparation de la résine PP(S)S-Met-Expansin-NH;. :
PP(S)S-Met-Expaπsin-NH
2 Au composé PP(S)S-C1, sous atmosphère d'argon, sont ajoutés la résine Met-Expansin- NH
2 (1 g ; 0,7 mmol NH
2) et 10 ml de pyridine. Instantanément une coloration jaune apparaît. La solution est portée à reflux de la pyridine (115° C) pendant 24 heures. La suspension est filtrée. Le solide est lavé successivement avec du toluène (3 fois 10 ml), de l'éthanol (3 fois 10 ml), de l'eau (3 fois 10 ml) et du dichlorométhane (3 fois 10 ml). La résine PP(S)S-Met-Expansin-NH
2 est isolée sous forme d'une poudre marron. Le rapport chlorure de sulfonyle/fonction aminé de la résine est de 1/7.
I - B - 4 - Préparation de la résine PP(SMe)S-Met-Expansin-NH 2. '
La résine précédemment préparée est mise, sous atmosphère d'argon, en suspension dans du dichlorométhane (10 ml). Ensuite, du triflate de méthyle (1,8 ml ; 16 mmol) est ajouté à température ambiante. La suspension est chauffée à reflux du dichlorométhane (40° C) pendant 12 heures. La résine PP(SMe)S-Met-Expansin-NH2 est isolée après filtration, lavage au tétrahydrofurane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg).
Les atomes d'azote des liens sulfonamides ainsi que les fonctions aminés libres de la résine sont susceptibles d'être alkylés au cours de cette étape.
I - B - 5 - Préparation de la résine PPS-Met-Expansin-NH- :
PP(SMe)S-Met-Expansiπ-NH2 PPS-Met-Expaπsiπ-NH2
La résine PP(SMe)S-Met-Expansin-NH2 est mise en suspension, sous atmosphère d'argon, dans du dichlorométhane fraîchement distillé et dégazé, puis la tris(diméthylamino)phosphine (2 ml ; 11 mmol) est ajoutée à température ambiante. La suspension est laissée sous agitation pendant 3 heures. La phosphine supportée sur la résine est isolée après filtration, lavage au dichlorométhane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg).
Le rendement des deux dernières étapes est d'environ 85 %, soit un rendement total de 75 %.
I - B - 6 - Préparation du catalyseur Pd/PPS-Met-Expansin-NH2
La résine PPS-Met-Expansin-NH2 est mise en suspension, sous atmosphère d'argon, dans le tétrahydrofurane distillé (15 ml), puis le complexe de tris(dibenzylidène- acétone)dipalladium (20 mg ; 0,022 mmol) est ajouté. Une coloration pourpre apparaît instantanément et disparaît ensuite au profit d'une coloration jaune-verte. La suspension est portée à reflux du tétrahydrofurane (67° C) pendant 12 heures. Le catalyseur au palladium est obtenu après filtration et lavage au tétrahydrofurane (3 fois 10 ml). Il est séché sous vide (0,01 mm Hg). La RMN du phosphore 31 du filtrat et du surnageant de lavage ne révèle la présence d'aucune trace de produit phosphore. Le catalyseur est gardé sous atmosphère d'argon. La microanalyse a permis de déterminer que la charge réelle du support ainsi préparé est de 0,028 mmol de palladium par gramme de polymère.
I - C - Préparation de Pd/TPPS-IRA-NH2 (Pd/monophosphine)
I - C - 1 - Préparation du composé TPP(S S : comme décrit dans I - A - 1
A une solution de TPPS (100 mg ; 0,175 mmol) dans le diméthylformamide (2 ml) est ajouté un excès de soufre élémentaire (56 mg ; 1,750 mmol) à température ambiante. Le mélange réactionnel est chauffé à 40° C pendant 15 minutes. La RMN du phosphore 31 révèle la présence d'un seul produit résonnant à 42 ppm. La solution est ensuite filtrée pour éliminer l'excès de soufre qui n'a pas réagi. Le filtrat est évaporé sous vide (0,01 mm Hg). La TPP(S)S est isolée sous la forme d'une poudre blanche (Rendement : voir ci- après).
I - C - 2 - Préparation du composé TPP(S)S-C1 : comme décrit dans I - A - 2

A une suspension de TPP(S)S (précédemment préparée) dans du dichlorométhane (3 ml), sous atmosphère d'argon, sont ajoutés 6 équivalents de chlorure d'oxalyle (90 μl ; 1 mmol) en présence d'une quantité catalytique de diméthylformamide (une goutte). Un dégagement gazeux (CO2) est observé, et la solution devient homogène. Le milieu réactionnel est agité pendant une heure à température ambiante. La RMN du phosphore 31 révèle la présence d'un seul produit résonnant à 40 ppm. Le composé TPP(S)S-C1 est isolé sous la forme d'une poudre jaune pâle après évaporation du solvant sous vide (0,01 mm Hg).
Le rendement des deux premières étapes est estimé à 60 %.
I - C - 3 - Préparation de la résine TPPfSVS-IRA-NH
TPP(S)S-CI TPP(S)S-IRA-NH2
Au composé TPP(S)S-C1, sous atmosphère d'argon, sont ajoutés la résine IRA-NH, (0,375 mg ; 1,575 mmol NH2) et 10 ml de pyridine. La solution est portée à reflux de la pyridine (115° C) pendant 36 heures. Le surnageant est enlevé. Les billes de la résine sont lavées successivement sous agitation avec du toluène (3 fois 10 ml), éthanol (3 fois 10 ml), eau (3 fois 10 ml) et dichlorométhane (3 fois 10 ml). La RMN du phosphore 31 du filtrat et des surnageants de lavage rassemblés et concentrés révèle la présence des traces de TPP(S)S-C1. La résine TPP(S)S-IRA-NH2 est isolée sous forme de billes de couleur beige foncé.
Le rapport chlorure de sulfonyle/f onction aminé de la résine est de 1/15.
I - C - 4 - Préparation de la résine TPP(SMe)S-IRA-NH: :

Ensuite du triflate de méthyle (0,5 ml ; 4,418 mmol) est ajouté à température ambiante. La suspension est chauffée à reflux du dichlorométhane (40° C) pendant 20 heures. La TPP(SMe)S-IRA-NH2 est isolée après séparation du surnageant, lavage au tétrahydrofurane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg). La RMN du phosphore 31 du filtrat et du surnageant de lavage rassemblés et concentrés ne révèle la présence d'aucun produit phosphore.
Les atomes d'azote des liens sulfonamides ainsi que les fonctions aminés libres de la résine sont susceptibles d'être alkylés au cours de cette étape.
I - C - 5 - Préparation de la résine TPPS-IRA-NH. :
TPP(SMe)S-IRA-NH2 TPPS-Met-IRA-NH2
La résine TPP(SMe)S-IRA-NH2 est mise, sous atmosphère d'argon, en suspension dans du dichlorométhane fraîchement distillé et dégazé (2 ml), puis la tris(diméthylamino)phosphine (2 ml ; 11 mmol) est ajoutée à température ambiante. La couleur des billes s'éclaircit au fur et à mesure de l'addition. La suspension est laissée à température ambiante pendant 4 heures. La phosphine supportée sur la résine est isolée après filtration, lavage au dichlorométhane (3 fois 10 ml) et séchage sous vide (0,01 mm
Hg).
La RMN du phosphore 31 du filtrat et du surnageant de lavage révèle la présence du tris(diméthylamino)méthylthiophosphonium [(Me2N)3PSMe]+ à 67 ppm et du tris(diméthylamino) phosphonium [(Me2N)3PH]+ à 21 ppm.
Le rendement de ces deux dernières étapes est d'environ 85 %, soit un rendement total de 50 %.
I - C - 6 - Préparation du catalyseur Pd/TPPS-IRA-NH,
Pd/TPPS-IRA-NH2
La résine TPPS-IRA-NH2 est mise en suspension, sous atmosphère d'argon, dans le tétrahydrofurane distillé (3 ml), ensuite le complexe de tris(dibenzylidène- acétone)dipalladium (20 mg ; 0,022 mmol) est ajouté. Une coloration pourpre apparaît instantanément et disparaît ensuite très lentement. La suspension est portée à reflux du tétrahydrofurane (67° C) pendant 18 heures. Un dépôt noir de palladium est observé. Le catalyseur au palladium est obtenu après filtration et lavage au tétrahydrofurane (3 fois 10 ml) (le palladium noir reste accroché au fritte). Il est séché sous vide (0,01 mm Hg). La RMN du phosphore 31 du filtrat et du surnageant de lavage ne révèle la présence d'aucune trace de produit phosphore. Le catalyseur est gardé sous atmosphère d'argon. La charge du palladium est estimée à 0,02 mmol (soit 0,06 mmol par gramme de polymère).
I - D - Préparation du catalyseur Pd/PPS-IRA-NH2 (Pd/diphosphine)
I - D - 1 - Préparation du composé PP(S)S : comme décrit dans I - B - 1
A une solution de PPS (50 mg ; 0,058 mmol) dans le diméthylformamide (2 ml) est ajouté un excès de soufre élémentaire (40 mg ; 1,750 mmol) à température ambiante. Le mélange réactionnel est chauffé à 40° C pendant 15 minutes. La solution est ensuite filtrée pour éliminer l'excès de soufre qui n'a pas réagi. Le filtrat est évaporé sous vide (0,01 mm Hg). Le composé PP(S)S est isolé sous la forme d'une poudre blanche (Rendement : voir ci-après).
I - D - 2 - Préparation du composé PP(S)S-C1 : comme décrit I - B - 1
A une suspension de PP(S)S (précédemment préparée) dans du dichlorométhane (3 ml) sont ajoutés, sous atmosphère d'argon, 8,8 équivalents de chlorure d'oxalyle (45 μl ; 0,516 mmol) en présence d'une quantité catalytique de diméthylformamide (une goutte). Un dégagement gazeux (CO2) est observé et le milieu réactionnel devient homogène. La solution est agitée pendant une heure à température ambiante. Le composé PP(S)S-C1 est isolé sous la forme d'une poudre jaune pâle après évaporation du solvant sous vide (0,01 mm Hg).
Le rendement des deux premières étapes est d'environ 85 %.
I - D - 3 - Préparation de la résine PP(S)S-IRA-NK. :
Au composé PP(S)S-C1 sont ajoutés, sous atmosphère d'argon, la résine IRA-NH2 (0,375 g ; 1,575 mmol NH2) et 10 ml de pyridine. Le mélange est porté à reflux de la pyridine (115° C) pendant 64 heures. La suspension est filtrée. Les billes sont lavés successivement, sous agitation, avec du toluène (3 fois 10 ml), de l'éthanol (3 fois 10 ml), de l'eau (3 fois 10 ml) et du dichlorométhane (3 fois 10 ml). La résine PP(S)S-IRA-NH2 est isolée sous forme de billes de couleur beige foncé. Le rapport chlorure de sulfonyle/f onction aminé de la résine est de 1/16.
I - D - 4 - Préparation de la résine PPrSMe S-IRA- T.
La résine précédemment préparée est mise en suspension, sous atmosphère d'argon, dans du dichlorométhane (2 ml). Ensuite du triflate de méthyle (0,5 ml ; 4,420 mmol) est ajouté à température ambiante. La suspension est chauffée à reflux du dichlorométhane (40° C) pendant 20 heures. La résine PP(SMe)S-IRA-NH2 est isolée après filtration, lavage au tétrahydrofurane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg).

La suspension est laissée à température ambiante pendant 4 heures. La diphosphine supportée sur la résine est isolée après filtration, lavage au dichlorométhane (3 fois 10 ml) et séchage sous vide (0,01 mm Hg).
Le rendement des deux dernières étapes est d'environ 85 %, soit un rendement total de 75 %.
I - D - 6 - Préparation du catalyseur Pd/PPS-IRA-NH. :
La résine PPS-IRA-NH2 est mise en suspension, sous atmosphère d'argon, dans le tétrahydrofurane distillé (3 ml), puis le complexe de tris(dibenzylidène- acétone)dipalladium (20 mg ; 0,022 mmol) est ajouté. Une coloration pourpre apparaît instantanément et disparaît lentement ensuite. La suspension est portée à reflux du tétrahydrofurane (67° C) pendant 18 heures. Un dépôt noir de palladium est observé. atmosphère d'argon. La microanalyse a permis de déterminer que la charge réelle du support ainsi préparé est de 0,036 mmol de palladium (soit 0,09 mmol par gramme de polymère).
II - Procédure générale de catalyse pour la réaction de Heck
Les réactifs choisis pour la réaction de couplage de Heck sont l'acrylate de méthyle AM comme oléfine et l'iodobenzène IB comme aromatique halogène. La triéthylamine a été utilisée comme base.
Les premières expériences d'optimisation de la réaction ont permis d'une part de choisir le toluène comme solvant et d'autre part de travailler avec la stœchiométrie suivante : deux équivalents d'AM pour un équivalent d'IB en présence de deux équivalents de triéthylamine. Des réactions avec un ou un-demi équivalent d'IB ont également été effectuées par la suite.
Dans une procédure générale de catalyse, 2 ml de toluène distillé sont ajoutés sur le catalyseur supporté sous argon. Ensuite sont additionnés dans l'ordre l'AM (90 μl, 2 mmol), la triéthylamine (279 μl, 2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 96 heures. La réaction est suivie par chromatographie sur couche mince.
Le traitement consiste à filtrer le mélange réactionnel sous atmosphère d'argon, et à laver le support avec du toluène (4 fois 20 ml) et du dichlorométhane (3 fois 10 ml). Les surnageants de lavage sont rassemblés et concentrés sous vide (20 mm Hg). Le résidu est purifié par chromatographie sur gel de silice (éluant : pentane/acétate d'éthyle, 96/4). Le produit de couplage est isolé sous forme d'un solide blanc.
Le support polymérique est séché sous vide (0,01 mm Hg) et gardé sous atmosphère d'argon. Afin de vérifier que le support palladié ne perd pas en activité catalytique, il est réutilisé dans une seconde réaction de couplage de Heck dans les mêmes conditions opératoires que ci-dessus.
II - A - Application de Pd/monophosphine fixé sur Met-Expansin-NH2 comme catalyseur supporté
Un protocole est tout d'abord établi pour la réaction de Heck en phase homogène avec PPh3, monophosphine, voir exemple 1 ci-dessous.
Exemple 1 : 2 ml de toluène distillé sont ajoutés sur le Pd2(dba)3 (9 mg, 0,01 mmol) sous argon. Ensuite sont additionnés dans l'ordre la triphénylphosphine (10,5 mg, 0,04 mmol), l'AM (180 μl, 2 mmol), la triéthylamine (279 μl, 2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 24 heures. Le rendement en produit de couplage isolé après purification est de 80 %.
Exemple 2 : 2 ml de toluène distillé sont ajoutés sur le catalyseur supporté sous argon. Ensuite sont additionnés dans l'ordre l'AM (180 μl, 2 mmol), la triéthylamine (279 μl, 2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 96 heures. Le rendement en produit de couplage isolé après purification est de 70 %.
Exemple 3 : mêmes conditions que l'exemple 2 avec le catalyseur supporté récupéré de l'exemple 2. Le rendement en produit de couplage isolé après purification est de 70 %.
II - B - Application de Pd/diphosphine fixé sur Met-Expansin-NH2 comme catalyseur supporté
Un protocole est tout d'abord établi pour la réaction de Heck en phase homogène avec Ph2P-CH2-CH2-PPh2, diphosphine, voir exemple 4 ci-dessous.
Exemple 4 : 3 ml de THF distillé sont ajoutés sur le Pd2(dba)3 (20 mg, 0,02 mmol) sous argon. Ensuite la l,2-di(biphénylphosphino)éthane (DPPE) (16 mg, 0,04 mmol) est ajoutée. La solution pourpre est portée à reflux du THF (67° C) pendant 12 heures. La solution jaune claire obtenue est concentrée sous vide (0,01 mm Hg). 2 ml de toluène distillé sont ajoutés sur le résidu. Ensuite sont additionnés dans l'ordre l'AM (180 μl, 2 mmol), la triéthylamine (279 μl, 2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 24 heures. Le rendement en produit de couplage isolé après purification est de 82 %.
Exemple 5 : 2 ml de toluène distillé sont ajoutés sur le catalyseur supporté sous argon. Ensuite sont additionnés dans l'ordre l'AM (180 μl, 2 mmol), la triéthylamine (279 μl,
2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 96 heures. Le rendement en produit de couplage isolé après purification est de 92 %.
Exemple 6 : mêmes conditions que l'exemple 5 avec le catalyseur supporté récupéré de l'exemple 5. Le rendement en produit de couplage isolé après purification est de 92 %.
Exemple 7 : mêmes conditions que l'exemple 6 avec le catalyseur supporté récupéré de l'exemple 6, mais avec un équivalent d'AM (90 μl, 1 mmol). Le rendement en produit de couplage isolé après purification est de 72 %.
Exemple 8 : mêmes conditions que l'exemple 6 avec le catalyseur supporté récupéré de l'exemple 7. Le rendement en produit de couplage isolé après purification est de 92 %.
II - C - Application de Pd/monophosphine fixé sur IRA-NH2 comme catalyseur supporté
Exemple 9 : mêmes conditions que l'exemple 8 : 2 ml de toluène distillé sont ajoutés sur le catalyseur supporté sous argon. Ensuite sont additionnés dans l'ordre l'AM (180 μl, 2 mmol), la triéthylamine (279 μl, 2 mmol) et enfin l'IB (112 μl, 1 mmol). Le milieu réactionnel est chauffé à reflux de toluène (110° C) pendant 96 heures. Le rendement en produit de couplage isolé après purification est de 95 %.
Exemple 10 : mêmes conditions que l'exemple 9 avec le catalyseur supporté récupéré de l'exemple 9. Le rendement en produit de couplage isolé après purification est de 65 %.
Exemple 11 : mêmes conditions opératoires que l'exemple 10 avec le catalyseur supporté récupéré de l'exemple 10, mais avec un équivalent d'AM (90 μl, 1 mmol). Le rendement en produit de couplage isolé après purification est de 38 %.
Exemple 12 : mêmes conditions que l'exemple 10 avec le catalyseur supporté récupéré de l'exemple 11. Le rendement en produit de couplage isolé après purification est de 72 %.
II - D - Application de Pd/diphosphine fixé sur IRA-NH2 comme catalyseur supporté
Exemple 13 : 2 ml de toluène distillé sont ajoutés sur le catalyseur supporté sous argon. Ensuite sont additionnés dans l'ordre l'AM (180 μl, 2 mmol), la triéthylamine (279 μl,
2 mmol) et enfin l'IB (112 μl, 1 mmol). Le miheu réactionnel est chauffé à reflux de toluène (110° C) pendant 96 heures. Le rendement en produit de couplage isolé après purification est de 88 %.
Exemple 14 : mêmes conditions que l'exemple 13 avec le catalyseur supporté récupéré de l'exemple 13. Le rendement en produit de couplage isolé après purification est de 92 %.
Exemple 15 : mêmes conditions que l'exemple 13 avec le catalyseur supporté récupéré de l'exemple 14, mais avec un équivalent et demi d'AM (135 μl, 1,5 mmol). Le rendement en produit de couplage isolé après purification est de 90 %.
Exemple 16 : mêmes conditions que l'exemple 13 avec le catalyseur supporté récupéré de l'exemple 15. Le rendement en produit de couplage isolé après purification est de 92 %.
Les résultats des essais catalytiques avec les différents ligands et supports sont résumés dans le tableau 1 ci-dessous.

III - Procédure générale de catalyse pour la réaction de Suzuki
La réaction de couplage de Suzuki met en jeu les réactifs suivants : l'acide phénylboronique APB comme aromatique et le para-iodoanisol IA comme aromatique halogène dans une stœchiométrie 1/1. Elle a lieu en présence de deux équivalents de dicarbonate de césium comme base dans un mélange DME/eau (6/1 ; v/v) à 90° C.
IA APB
Dans une procédure générale de catalyse, 6 ml de DME sont ajoutés sur le catalyseur supporté. Ensuite sont additionnés dans l'ordre l'APB (121 mg, 1 mmol), le dicarbonate de césium (652 mg, 2 mmol) solubilisé dans 1 ml d'eau et enfin l'IA (234 mg, 1 mmol). Le milieu réactionnel est chauffé à reflux à 90° C pendant 96 heures. La réaction est suivie par chromatographie sur couche mince.
Le traitement consiste à filtrer le mélange réactionnel et à laver le support avec du DME (4 fois 20 ml), de l'eau (3 fois 10 ml) et du dichlorométhane (3 fois 10 ml). Les surnageants de lavage sont rassemblés et concentrés sous vide (20 mm Hg). Le résidu est purifié par chromatographie sur gel de silice (éluant : pentane/acétate d'éthyle, 98/2). Le produit de couplage est isolé sous forme d'un solide blanc.
Le catalyseur supporté est séché sous vide (0,01 mm Hg).
III - A - Application de Pd/diphosphine fixé sur Met-Expansin-NH2 comme catalyseur supporté
Nous avons tout d'abord établi un protocole pour la réaction de Suzuki en phase homogène avec Ph2P-CH2-CH2-PPh2, diphosphine, voir exemple 1 ci-dessous.
Exemple 17 : l'APB (121 mg, 1 mmol) est solubilisé dans 6 ml de DME, la DPPE (12 mg, 0,03 mmol) est ajoutée. Ensuite sont additionnés dans l'ordre, le dicarbonate de césium (652 mg, 2 mmol) solubilisé dans 1 ml d'eau, l'IA (234 mg, 1 mmol) et enfin le Pd2(dba)3 (14 mg, 0,015 mmol). Le milieu réactionnel est chauffé à 90° C pendant 24 heures. La réaction est suivie par chromatographie sur couche mince. Le rendement en
produit de couplage isolé après purification est de 80 %.
Exemple 18 : 6 ml de DME sont ajoutés sur le catalyseur supporté. Ensuite sont additionnés dans l'ordre l'APB (121 mg, 1 mmol), le dicarbonate de césium (652 mg, 2 mmol) solubilisé dans 1 ml d'eau et enfin l'IA (234 mg, 1 mmol). Le milieu réactionnel est chauffé à reflux à 90° C pendant 96 heures. La réaction est suivie par chromatographie sur couche mince. Le rendement en produit de couplage isolé après purification est de 60 %.
III - B - Application de Pd/diphosphine fixé sur IRA-NH2 comme catalyseur supporté
Exemple 19 : 6 ml de DME sont ajoutés sur le catalyseur supporté. Ensuite sont additionnés dans l'ordre l'APB (121 mg, 1 mmol), le dicarbonate de césium (652 mg, 2 mmol) solubilisé dans 1 ml d'eau et enfin l'IA (234 mg, 1 mmol). Le milieu réactionnel est chauffé à reflux à 90° C pendant 96 heures. La réaction est suivie par chromatographie sur couche mince. Le rendement en produit de couplage isolé après purification est de 90 %.
Exemple 20 : mêmes conditions que l'exemple 19 avec le catalyseur supporté récupéré de l'exemple 19. Le rendement en produit de couplage isolé après purification est de 92 %.
Les résultats des essais catalytiques avec les différents ligands et supports sont résumés dans le tableau 2 ci-dessous.

Ces résines se sont révélées être de bons catalyseurs de la réaction de Heck et de Suzuki (les rendements obtenus en produits de couplage sont du même ordre de grandeur ou même supérieurs à ceux obtenus en catalyse homogène). Les résines possédant les diphosphines sont extrêmement performantes (grâce à l'effet chélate des ligands bidentates).
