Abstract
Die Erfindung betrifft neue arylsubstituierte aminoacylierte Phosphonsäuren und deren Halbester, ein Verfahren zu ihrer Herstellung und ihre Verwendung. α-N-Acylgeschützte-β-substituierte Alkenphosphonsäurederivate werden in Gegenwart eines Rhodiumkomplex-katalysators asymmetrisch hydriert. Auch die Herstellung der Ausgangsverbindungen wird erfindungsgemäß aufgezeigt. Die neuen Verbindungen sind als Herbizid, Bakterizid, Antibiotikum oder als antivirales Mittel anwendbar.
Description
Arylsubstituierte a inoacylierte Phosphonsäuren und deren Halbester
Die Erfindung betrifft neue arylsubstituierte aminoacylierte Phosphonsäuren und deren Halbester in optisch aktiver Form sowie ein Verfahren zur Herstellung dieser Verbindungen durch asymmetrische katalytische Hydrierung geeigneter Vorstufen mit chiralen Komplexkatalysatoren. Die Erfindung behandelt darüber hinaus diese neuartigen geeigneten Vorstufen. Auch die Verwendung der neuen Verbindungen wird aufgezeigt.
Arylsubstituierte aminoacylierte Phosphonsäuren und deren Halbester sind bisher kaum bekannt geworden.
Bekannt sind optisch aktive α-Aminophosphonsäuren oder deren Derivate, die nach verschiedenen Verfahren hergestellt werden können. Neben klassischen Racemattrennungen nehmen breiten Raum stöchiometrische Verfahren ein, die sich alle optisch aktiver Auxiliare bedienen. Diese Verfahren beruhen dabei auf C-C, C-P, C-H oder C-N-Knüpfungsreaktionen unter gleichzeitiger
Einführung der (des) chiralen Zentren (Zentrums) . Der Stand der Technik ist in einem Reviewartikel von B. Dharwan und D. Redmore in Phosphorous, Sulfur and Silicon 32 (1987) , 119 bis zum Jahr 1986 und in einem weiteren von V. P. Kukhar, V. A. Soloshonov und V. A. Solodenko in Phosphorous, Sulfur and Silicon 92 (1994) , 239-264 dargestellt worden.
Es ist weiter bekannt, daß neben den auxiliargestützten Synthesen zwei weitere Verfahren zur asymmetrischen Hydrierung des α-N-Formyl-phosphonsäuredimethylesters und der Säuren bzw. von ZNHC (=CH2) PO (OR) 2-Verbindungen mit Rhodiumkomplexkataly-satoren beschrieben wurden (U. Schöllkopf, I. Hoppe und A. Thiele, Liebigs Ann. Chem. 1985, 555-559 und US-A-5 321 153) . Im ersten Verfahren ist
die Synthese zu den ungewöhnlichen α-Aminophosphonsäuren speziell zu AlaP beschrieben. Als Katalysator erwies sich nur der (+) -DIOP-Rh-Komplex geeignet, der bei schlechter Aktivität AlaP in 76 % ee ergab, wobei nur das Methyl-, nicht jedoch das Ethylphosphonat als Substrat überhaupt reagierte und die klassischen Rhodiumkomplexkatalysatoren wie DIPAMP-Rh, NORPHOS-Rh völlig versagten. Andererseits wird gemäß der U -Patentanmeldung 5 321 153 zur Hydrierung von ZNHCH(=CH)2PO(OMe)2 und Säure zu Aryl-, Alkyl- und CycloalkylVerbindungen DIPAMP eingesetzt, allerdings unter schwachem Druck. Es werden hohe chemische Ausbeuten bei optischen Ausbeuten um 90 % ee erreicht. Ein Nachteil ist allerdings die für Fünfringchelat-bildner verfahrensgemäß geringe Aktivität, so wird beispielsweise das N- [1- (Dimethoxy-phosphoryl) ] -4-phenylethenylformamid bei einem Substrat/Katalysator-Verhältnis von 144:1 bei einem Druck von 40 psi erst in 16 Stunden durchhydriert.
Auch die Knüpfung einer CH-N-Bindung durch enantioselektive Reduktion einer analogen Schiffschen Base als Vorstufe durch Aluminiumamalgam führte nur zu mäßigen chemischen und optischen Ausbeuten (67 % ee) (X.Y. Jiao, . Y. Chen und B. F: Hu, Synth. Commun. 22 (1992), 1179).
Aufgabe der Erfindung ist es, neue arylsubstituierte aminoacylierte Phosphonsäuren und deren Halbester bereitzustellen, wobei zu ihrer Herstellung unter Vermeidung der obengenannten Nachteile ein Syntheseverfahren zu entwickeln ist, welches je nach Zielprodukt mit chiralen Rhodiumkomplexkatalysatoren durch asymmetrische katalytische Hydrierung wahlweise aus geeigneten ungesättigten Vorstufen die Herstellung der arylsubstituierten aminoacylierten Phosphonsäuren oder deren Halbester in hoher Geschwindigkeit und bei der optisch aktiven Form auch mit hoher Stereoselektivität und einem hohen Substrat/Katalysator-Verhältnis erlaubt .
Diese Aufgabe wird anspruchsgemäß gelöst.
Die neuen optisch aktiven arylsubstituierten aminoacylierten Phosphonsäuren bzw. deren Halbester besitzen die allgemeine Formel I
in der

AIkoxy, ( Cx -C4 ) -AIky1oxya1ky1 , mit Heteroatomen wie N, 0, P, S und/oder
Halogen wie Cl, F, Br substituiert, wie Phenyl, Biphenyl, Naphthyl, Anthracenyl,
Heteroaryl, ggf- einfach oder mehrfach mit linear oder verzweigten (C1-C4) -Alkyl, (C1-C4) -
Alkoxy, (C1-C4) -Alkyloxyalkyl, mit Heteroatomen wie N, 0, P, S und/oder
Halogen wie Cl, F, Br substituiert, wie 2-, 3- oder 4-Pyridyl, 2- oder 3-Thienyl, 2- oder
3-Furyl,
R2 = Formyl, Acetyl , Benzoyl, Benzyloxycarbonyl (Z) , tert . - Butoxycarbonyl (BOC) und o-Nitrobenzoyl sowie R3, R4 = unabhängig voneinander H, (C-L-CJ) -Alkyl, ggf. linear oder verzweigt bedeuten.
Gegenstand der Erfindung ist ferner ein Verfahren zur Herstellung der Verbindungen der Formel I, indem erfindungsgemäß ein α-N-acylgeschütztes- ß-substituiertes Alkenphosphon-säurederivat der allgemeinen Formel II,
in der
R , R , R und R die in der allgemeinen Formel I angegebenen Bedeutungen besitzen,
in Gegenwart eines chiralen Rhodiumkomplexkatalysators der allgemeinen Formel IV,
in der
X = α-Naphthoxyalkyl wie α-Naphthoxymethyl und Y = Wasserstoff und
A = eine n-Alkyl-, iso-Alkyl- oder Cycloalkylgruppe bedeuten,
in einem Lösungsmittel oder einer Suspension bei Temperaturen zwischen -20 °C und +50 °C und einem
Wasserstoffdruck zwischen 0,05 und 2,0 MPa asymmetrisch hydriert wird.
Die vorliegende Erfindung umfaßt auch den Prozeß, der die Herstellung der Verbindungen der Formel II beinhaltet. So werden diese Verbindungen aus ß-substituierten-N-acylierten Amino-carbonsäurederivaten der allgemeinen Formel III,
in der
R = die in der allgemeinen Formel I angegebenen Bedeutungen besitzt und R = einen Benzoylrest repräsentiert, via Chloraddition unter Umsatz mit P(OR)3, (R = Alkyl) (Brovarets, V. S. et al . , Zh. Obsh. Khim. 1993, 63, 1259. U. Schmidt et al . , Synth. Commun. 1996, 26, 777, DOS 195 19 983 AI) , nachfolgender Reaktion mit Bromtrimethylsilan zum Bis (trimethylsilyl) ester
(R = Si(CH3)3) und Hydrolyse mit Wasser oder Ethanol, bzw. durch Versetzen des Umsatzproduktes mit Natronlauge sowie Extraktion mit Chloroform und nachfolgender Ansäuerung der wäßrigen Phase in Gegenwart einer Chloroformschicht, erhalten.
Erfindungsgemäß werden zur Herstellung der neuen Verbindungen der Formel I in optisch aktiver Form die prochiralen Vorstufen unter diesen milden Bedingungen in hoher Geschwindigkeit und in hoher Stereoselektivität durch den chiralen Katalysator asymmetrisch hydriert, bevorzugt hinsichtlich der Geschwindigkeit durch das Vorliegen eines sieben-gliedrigen Metallchelates und hinsichtlich der Stereoselektivität durch das Vorliegen unterschiedlicher Phosphordonatoratome, welche die Vorzugskonformationen ermöglichen.
Die Hydrierung kann in alkoholischer Lösung, bevorzugt in methanolischer Lösung, aber auch in Suspensionen erfolgen.
Die eingeführten, bei ihrer Synthese kristallin anfallenden Rhodiumkomplexkatalysatoren erlauben die Ausführung des Prozesses bei Temperaturen zwischen - 20 °C und + 50 °C.
Bevorzugt wird jedoch bei Raumtemperatur hydriert.
Die Hydrierungen werden in einem Druckbereich durchgeführt, der zwischen 0,05 und 2,0 MPa Wasserstoffdruck, vorzugsweise jedoch bei Normaldruck, liegt.
Erfindungsgemäß muß der Katalysator nicht kristallin vorliegen. Er kann auch in situ aus dem Liganden und einem geeigneten Cokatalysator wie [Rh (COD) 2] BF4 hergestellt werden. Die kationische Form des Komplexkatalysators wird bevorzugt .
Die Herstellung des Zielproduktes als Racemat erfolgt in gleicher Weise unter Anwendung des Rhodiumkatalysators in achiraler Form.
Die erhaltenen unnatürlichen Aminophosphonsäuren oder deren Halbester sind durch ihre potentielle herbicide, antibakterielle, antibiotische und antivirale Aktivität ausgezeichnet. Die Wirkung erstreckt sich sowohl auf die
Phosphonsäurederivate direkt, als auch insbesondere auf die aus ihnen hergestellten Di- und Oligopeptide .
Unter einem linearen oder verzweigten (C-L-QJ) -Alkylrest versteht man Reste wie Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, tert.-Butyl, Isobutyl, sec-Butyl.
Als linear oder verzweigte (C^d) -Alkoxyreste sind gemeint: Methoxy, Ethoxy, n-Propoxy, Isopropoxy, n-Butoxy, Isobutoxy, tert . -Butoxy, sec-Butoxy.
(C^C) -Alkoxyalkylreste bestehen aus den oben genannten linear oder verzweigten Alkylresten, welche über eine Sauerstoffunktion an Alkylenreste wie Methylen, Ethylen oder Propylen gebunden sind.
Die Erfindung wird durch die nachfolgenden Beispiele erläutert, ohne sie jedoch einzuengen.
Beispiel 1
α-N-Benzoylamino-ß-phenyl-ethylenphosphonsäure
3.3 g (0.01 mol) des α-N-Benzoylamino-ß-phenyl- ethylenphosphonsäuredimethylesters werden mit 4.3 ml (0.03 mol) Bromtrimethylsilan 2 Std. bei Raumtemperatur gerührt . Die Reaktionsmischung wird eingeengt und der Rest mit Ethanol versetzt und über Nacht stehen gelassen. Es wird wieder eingeengt. Der ölige Rückstand wird mit Wasser zur Kristallisation gebracht und aus Acetonitril umkristallisiert. Die Säure wird bei 110 °C 4 Std. über
H2S04 getrocknet. Schmp. 182 - 186 °C. Die Ausbeute beträgt

Beispiel 2
(S) -α-N-Benzoylamino-ß-phenyl-ethylphosphonsäure
Die nach Beispiel 1 hergestellte Säure wird in Gegenwart des Katalysators der Formel (S) -IV in einer Halbwertzeit von 7 min. hydriert und danach wie folgt aufgearbeitet: Die Hydrierlösung wird im Vakuum bis zur Trockne eingedampft und dreimal mit Methylenchlorid digeriert . Der zurückbleibende Feststoff wird getrocknet und kann für analytische Zwecke in Methanol gelöst werden. Die Ausbeute beträgt 96 % und der Enantiomerenüberschuß 83 %. Schmp. 180 -186 °C. C15HιeN04P (305.3), Ber. (%) : C 59.02 H 5.28 N 4.59 P 10.15, Gef. Beispiel 3
α-N-Benzoylamino-ß-phenyl- ethylenphosphonsäuremonoethylester
3,3 g (0,009 mol) des α-N-Benzoyl-ß-phenyl- ethylenphosphonsäurediethylesters werden in 28 ml 2 n
Natronlauge 3 Std. bei 75 °C gerührt. Die Reaktionsmischung wird gekühlt und 5 x mit CHC13 extrahiert. Dann wird die wäßrige Schicht mit CHC13 unterschichtet und unter intensivem Rühren langsam mit 2 n Schwefelsäure angesäuert. Die Schichten werden getrennt, die wäßrige Schicht 3 x mit CHC13 extrahiert und die vereinigten organischen Extrakte mit Na2S04 getrocknet. Schließlich wird das Lösungsmittel verdampft und der zurückbleibende ölige Rest mit Ether übergössen. Im Kühlschrank kristallisiert ein weißer Feststoff aus. Schmp. 106 - 108 °C. Die Ausbeute beträgt 2,5 g (80,6 %) . C17H18N04P x X H20 (340.3) Ber. (%) : C 60.00 H 5.62 N 4.12 P 9.10 Gef. (%) : C 60.72 H 5.62 N 4.15 P 9.12, 31P NMR: δ 15.6 ppm (CD30D)
Beispiel 4
(S) -α-N-Benzoylamino-ß-phenyl- ethylphosphonsäuremonoethylester
Der nach Beispiel 3 hergestellte Monoethylester wird in Gegenwart des Katalysators der Formel (S) -IV in einer Halbwertzeit von 11 min. hydriert und danach wie folgt aufgearbeitet : Die Hydrierlösung wird zur Trockne eingedampft. Der Rückstand wird in 0,4 n Natronlauge bei Raumtemperatur gelöst und 3 x mit CH2C12 extrahiert . Die wäßrige Lösung wird mit Essigsaureethylester überschichtet und unter kräftigem Rühren mit 1 n Schwefelsäure angesäuert. Die Schichten werden getrennt, die wäßrige Lösung noch mehrere Male mit Essigsaureethylester
extrahiert und die vereinigten Extrakte mit Na2S04 getrocknet. Nach dem Verdampfen des Lösungsmittels kristallisiert der Halbester aus.

Verfahren zur Herstellung der Verbindungen der Formel I gemäß Anspruch 1, dadurch gekennzeichnet, daß ein α-N-acylgeschütztes-ß-substituiertes

R ,1, τR-2 R und R die in der allgemeinen Formel I angegebenen Bedeutungen besitzen, in Gegenwart eines chiralen oder achiralen
Rhodiumkomplexkatalysators der allgemeinen Formel IV,
in der
X = α-Naphthoxyalkyl wie α-Naphthoxyτnethyl und Y = Wasserstoff und
A = eine n-Alkyl-, iso-Alkyl- oder Cycloalkylgruppe bedeuten,
asymmetrisch hydriert wird.
Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß in Lösung oder in Suspension hydriert wird.
4. Verfahren nach Anspruch 2 und 3 , dadurch gekennzeichnet, daß bei einer Temperatur von -20 °C bis +50 °C hydriert wird.
5. Verfahren nach Anspruch 2 bis 4, dadurch geke nzeichnet, daß bei einem Druck zwischen 0.05 und 2.0 MPa hydriert wird.
6. Verfahren nach Anspruch 2 bis 5, dadurch geke nzeichnet, daß die Hydrierung in alkoholischer Lösung, bevorzugt in methanolischer Lösung, oder Suspension erfolgt.
7. Verfahren nach Anspruch 2 bis 6 , dadurch gekennzeichnet, daß bei Raumtemperatur hydriert wird.
8. Verfahren nach Anspruch 2 bis 7, dadurch gekennzeichnet, daß bei Normaldruck hydriert wird.
9. Verfahren nach Anspruch 2 bis 8, dadurch gekennzeichnet, daß der Katalysator in kristalliner Form oder in situ gebildet aus dem Liganden und einem geeigneten Cokatalysator wie Rh(C0D)2BF4 eingesetzt wird.
10. Verfahren nach Anspruch 2 bis 9, dadurch gekennzeichnet, daß bevorzugt die kationische Form des Katalysators eingesetzt wird.
11. Verfahren nach Anspruch 2, dadurch gekennzeichnet , daß das α-N-acylgeschützte-ß-substituierte Alkenphosphonsaurederivat der Formel II aus einem ß-substituierten-N-acylierten Aminocarbonsäurederivat der allgemeinen Formel III,
i /-COOH R-CH = C 2 HI
NHR2
in der
R und R die in der allgemeinen Formel I angegebenen Bedeutungen besitzt, herstellt .
12. Verfahren nach Anspruch 11, dadurch gekennzeichnet , daß man die Verbindung der allgemeinen Formel III mit Chlor und anschließend mit P(OR)3, wobei R in diesem Fall einen (C-L-C -Alkylrest, welcher linear oder verzweigt vorliegen kann, bedeutet, umsetzt .
13. Verwendung der Verbindungen der allgemeinen Formel I sowie aus ihnen erhältliche Verbindungen als Herbizide, Bakterizide, Antibiotika oder antivirale Mittel.
