Die erhaltenen Partikel haben überwiegend eine kugelförmige Gestalt.EP 0 343 934 describes magnetic particles which consist of a core made of a magnetic material and a shell made of an inorganic oxide. The preparation is carried out by emulsifying the solution of a salt or an aqueous sol of the magnetic material and a solution of a salt or an aqueous sol of the inorganic oxide in a water-immiscible liquid, forming discrete drops. The drops are converted into a gel by introducing ammonia, then separated from the liquid phase, dewatered and calcined. The particles obtained predominantly have a spherical shape.
Die aus dem Stand der Technik bekannten magnetischen Partikel haben den Nachteil, daß sie keine einheitliche Größe besitzen, Agglomerate nicht verhindert werden können und das Masseverhältnis zwischen magnetischem Kern und Hüllschicht Schwankungen unterworfen ist.The magnetic particles known from the prior art have the disadvantage that they do not have a uniform size, agglomerates cannot be prevented and the mass ratio between the magnetic core and the cladding layer is subject to fluctuations.
Dadurch zeigen die Partikel kein einheitliches Verhalten im Magnetfeld, was sich negativ auf den Trennprozeß auswirkt. As a result, the particles show no uniform behavior in the magnetic field, which has a negative effect on the separation process.
Aufgabe der Erfindung ist es, monodisperse magnetische Partikel mit einer weitestgehend einheitlichen Größe und einem einstellbaren Verhältnis zwischen magnetischem Material und übrigen Bestandteilen bereitzustellen. The object of the invention is to provide monodisperse magnetic particles with a largely uniform size and an adjustable ratio between magnetic material and other constituents.
Diese Aufgabe wird gemäß der Erfindung gelöst durch monodisperse magnetische Partikel einer Teilchengröße von 50 bis 1500 nm, mit einem kugelförmigen Kern aus Siliciumdioxid, der punktuell mit Magnetikpartikeln einer Teilchengröße von maximal 60 nm beschichtet ist und eine Deckschicht aus Siliciumdioxid besitzt. In einer besonderen Ausführungsform sind die magnetischen Partikel in bekannter Weise silanisiert. This object is achieved according to the invention by monodisperse magnetic particles with a particle size of 50 to 1500 nm, with a spherical core made of silicon dioxide, which is selectively coated with magnetic particles with a maximum particle size of 60 nm and has a cover layer made of silicon dioxide. In a special embodiment, the magnetic particles are silanized in a known manner.
Furthermore, this object is achieved according to the invention by a process for producing the magnetic particles by dispersing silica gel monospheres in deionized water at 20 ° C. in a first stage and by adding a solution of iron (II) sulfate and iron (III) sulfate with a metering rate of 0.05 to 3 mg Fe 3 0 per minute and per m2 surface area of the monospheres, a pH value of 8 being maintained by simultaneous addition of a base, spot coating with Fe 3 0 4 and in a second step to the suspension of the coated monospheres an aqueous, acetic acid tetraethyl orthosilane solution at a rate of 0.1 to 5 mg Si0 2 per min. and per m? surface the uncoated monospheres are metered in, then the suspension is adjusted to a pH of 9 within 30 minutes by adding a base and subsequently heated to 75 ° C. and then the magnetic particles obtained (monospheres magnetic beads) are separated off and washed with water and dried at 60 to 100 ° C, optionally in vacuo.
Gegebenenfalls können die erhaltenen Monospher-Magnetbeads in einer dritten Stufe nach bekannten Verfahren silanisiert werden. If appropriate, the monospher magnetic beads obtained can be silanized in a third stage by known processes.
Gegenstand der Erfindung ist auch die Verwendung der erfindungsgemäßen magnetischen Partikel für die Isolierung von Nucleinsäuren und Biotin sowie biotinylierter Proteine aus wäßrigen Lösungen. The invention also relates to the use of the magnetic particles according to the invention for the isolation of nucleic acids and biotin and biotinylated proteins from aqueous solutions.
Die als Ausgangsmaterial zu verwendenden kugelförmigen monodispersen Si02-Partikel sind an sich aus dem Stand der Technik bekannt. The spherical monodisperse Si0 2 particles to be used as starting material are known per se from the prior art.
Die grundliegenden Reaktionsbedingungen zur Herstellung von Si02- Partikeln durch hydrolytische Polykondensation sind beispielsweise aus den Publikationen von W. Stöber et al. in J. Colloid and Interface Science 26, 52(1968) und 30, 568(1969) sowie dem US-Patent 3,634,588 zu entnehmen. Die so hergestellten Partikel zeigen aber oft große Standardabweichungen für die Teilchendurchmesser und weisen eine gewisse Porosität auf.The basic reaction conditions for the production of Si0 2 particles by hydrolytic polycondensation can be found, for example, in the publications by W. Stöber et al. in J. Colloid and Interface Science 26, 52 (1968) and 30, 568 (1969) and U.S. Patent 3,634,588. However, the particles produced in this way often have large standard deviations for the particle diameters and have a certain porosity.
For the production of highly monodisperse, non-porous, spherical Si0 2 particles which have a standard deviation of not more than 5%, reference is made to EP 0 216 278, which discloses a correspondingly stopped production process based on hydrolytic polycondensation. The essence of this process, which is preferred for the production of the S0 2 particles according to the present invention, is a two-stage procedure. First, a sol or a suspension of primary particles is formed by hydrolytic polycondensation of tetraalkoxysilanes in aqueous, alkali, ammonia, medium it is then brought to the desired final size by metered addition of further tetraalkoxysilane. The process for producing highly monodisperse, non-porous spherical SiO 2 particles takes place in two steps.
For this purpose, the tetraalkoxysilane is placed in an aqueous-alcoholic-ammoniacal hydrolysis mixture and mixed thoroughly. Suitable tetraalkoxysilanes which can be used are all readily hydrolyzable ortho esters of aliphatic alcohols. The esters of aliphatic alcohols with 1-5 C atoms, such as methanol, ethanol, n- or i-propanol, and the isomeric butanols and pentanols are primarily considered here. These can be used individually or in a mixture. The silicic acid orthoesters of the C r C 3 alcohols, in particular tetraethoxysilane, are preferred. In addition to aliphatic alcohols, the hydrolysis mixture should contain from about 0.5 to about 8 mol / l ammonia and from about 1 to about 15 mol / l water on white. Aliphatic CrC 3 alcohols, preferably CrC 3 alcohols, are suitable as the alcohol component such as methanol, ethanol and n- or i-propanol. These can be present individually or in a mixture with one another in the hydrolysis mixture. The tetraalkoxysilane is preferably added to the hydrolysis mixture in one portion, the reactant being able to be present in pure form or in solution in one of the alcohols mentioned. A concentration of tetraalkoxysilane in the hydrolysis mixture between about 0.01 and about 1 mol / l can be selected to generate the primary particles. After the reactants have been brought together, the reaction begins immediately or after a few minutes, which is indicated by an immediate
Opaleszenz bzw. The reaction mixture shows opalescence or turbidity due to the resulting particles. After generally not more than 15-30 minutes, or longer in unfavorable special cases, the reaction has ended. Depending on the choice of the reactants and their concentrations in the reaction mixture, particles with an average diameter between about 0.01 and about 2 μm can be obtained in accordance with known methods In this first step of the process according to the invention, preference is given to working with reaction mixtures which contain 3-13 mol / l water, 0.5 to 4.5 mol / l ammonia, 10 to 25 mol / l alcohol and 0.1-0.5 mol / l contain tetraalkoxysilane. This gives primary particles with average diameters between 0.01 and 1 μm. At this stage, samples can be taken from the sol of primary particles in order to examine the particles for their particle size, shape accuracy and particle size distribution, for example with the aid of electron microscopy.
Durch Isolation von Teilchenproben besteht die Möglichkeit, die Porosität der Primarteilchen beispielsweise durch Gasadsorptionsmessung zu bestimmen.By isolating particle samples, it is possible to determine the porosity of the primary particles, for example by gas adsorption measurement.
It has proven to be advantageous to carry out the reaction for producing the primary particles at a higher temperature. Temperatures between 35 and 75 ° C., preferably between 40 and 65 ° C., are favorable. It was found that the particle size scatter decreases at elevated temperature, but also the mean particle size. At lower temperatures, i.e. around room temperature, larger particles with higher size scatter are obtained under otherwise identical conditions. Furthermore, undesirable agglomerate formation is also increasingly to be seen here.
- b -In the second step of the process, with uniform mixing, a slow, continuous metering of further tetraalkoxysilane into the sol of primary tech takes place. It is essential here that the rate of metering is controlled so that an immediate, complete reaction with the particles present in the sol takes place without an excess of tetraalkoxysilane being formed, from which nuclei for new primary particles can form. Controlled regrowth of the particles in the sol is achieved under this process measure of metered addition of tetraalkoxysilane, controlled in accordance with the reaction, whereby the particle size ultimately to be achieved depends on the amount of the total added tetraalkoxysilane. The amount of the total alkoxysilane to be added is in principle not critical as long as the hydrolysis mixture is present in excess, or - b -
durch eventuelles Nachgeben von Hydrolysegemisch im Überschuß gehalten wird. Ein zeitliches Limit bei der Zugabe des Tetraalkoxysilans ist nicht gesetzt; die Zugabe kann sich über Stunden bis Tage erstrecken. Auch Unterbrechungen und Wiederaufnahme des Nachwachsens sind möglich, da die Teilchen in allen Stadien ihres Wachstums stabil sind.is kept in excess by possibly adding hydrolysis mixture. There is no time limit for the addition of the tetraalkoxysilane; the addition can range from hours to days. Interruptions and resumption of regrowth are also possible since the particles are stable at all stages of their growth.
Auch im Nachwachsvorgang wird bevorzugt bei erhöhter Temperatur, etwa um 40 °C, gearbeitet. The regrowth process is preferably carried out at an elevated temperature, around 40 ° C.
Nach diesem Verfahren können monodisperse kugelförmige Kieselsäure- Partikel mit einer Partikelgröße von 50 bis 1500 nm hergestellt werden, wobei eine gewünschte Größe sehr genau einstellbar ist, da die Standardabweichung vom mittleren Teilchendurchmesser nur 2 bis 5 % beträgt.Monodisperse spherical silica particles with a particle size of 50 to 1500 nm can be produced by this method, a desired size being able to be set very precisely, since the standard deviation from the mean particle diameter is only 2 to 5%.
The punctiform coating of the silica monospheres with magnetite is carried out by precipitation from a solution of iron (II) and iron (III) salts, preferably from a solution of iron (II) and iron (III) sulfate. The molar ratio between the divalent and trivalent iron salt is 1: 1. It should be noted that the precipitation solutions must be protected against oxidation. The presence of oxidizing or reducing agents is not necessary during the precipitation.
Der pH-Wert der Monospherdispersion wird für die Fällung des magnetischen Fe304 (Magnetit) auf Werte zwischen 7 bis 9, bevorzugt 7,5 bis 8,5 eingestellt. Der pH-Wert wird während der Fällungsreaktion durch Zugabe einer Base konstant gehalten, bevorzugt wird 25%ige wäßrige Ammoniaklösung verwendet. Die Temperatur der Suspension wird auf 0 bis 40 °C eingestellt. For the precipitation of the magnetic Fe 3 0 4 (magnetite), the pH of the monosphere dispersion is adjusted to values between 7 and 9, preferably 7.5 to 8.5. Die Dosiergeschwindigkeit der Eisen(ll)-Eisen(lll)-salzlösung liegt zwi- sehen 0,05 bis 3 mg Fe304 pro Min. und pro ms Oberfläche, bevorzugt zwischen 0,2 bis 1 mg Fe304 pro Min. und pro ms Oberfläche der Monospher. Unter den angegebenen Bedingungen erfolgt eine punktförmige Abscheidung des Magnetits auf der Oberfläche der Monospher. Die Größe der Magnetitpartikel beträgt maximal 60 nm.The metering rate of the iron (II) iron (III) salt solution is between 0.05 to 3 mg Fe 3 0 4 per minute and per ms surface, preferably between 0.2 to 1 mg Fe 3 0 4 per minute and per ms surface of the monospheres. Under the specified conditions, the magnetite is deposited in a punctiform manner on the surface of the monospheres. The size of the magnetite particles is a maximum of 60 nm.

The binding capacity of the magnetic particles according to the invention is approximately 5 μg / mg beads for DNA. For the streptavidin-coated particles, the binding capacity is very dependent on the substance to be isolated.
Die erfindungsgemäßen kugelförmigen magnetischen Partikel haben den Vorteil, daß sie monodispers, unporös und in verschiedenen Größen her- stellbar sind. Die Standardabweichung vom mittleren Durchmesser beträgt weniger als 5 %. Das Masseverhältnis zwischen dem magnetischen Material und den übrigen Bestandteilen ist in engen Grenzen einstellbar. Dadurch zeigen die Partikel ein gleiches Verhalten im Magnetfeld. The spherical magnetic particles according to the invention have the advantage that they are monodisperse, non-porous and can be produced in different sizes. The standard deviation from the mean diameter is less than 5%. The mass ratio between the magnetic material and the other components can be adjusted within narrow limits. As a result, the particles show the same behavior in the magnetic field.

Die erhaltene Lösung wird mit einer Geschwindigkeit von 120 ml/Min. der Dispersion der mit Magnetit beschichteten Monospher zudosiert. Danach wird innerhalb von 30 Min. der pH-Wert mit der wäßrigen Ammoniaklösung auf 9,0 angehoben und die Dispersion auf 75 °C erwärmt und für 30 Min. auf diesem Wert gehalten. Nach dem Abkühlen auf Raumtemperatur wird die Dispersion aufgearbeitet.350 g of tetraethylorthosilane are dissolved with stirring in a closed apparatus in a mixture of 290 g of glacial acetic acid and 2275 g of completely deionized water within 30 minutes. Particles are added to 190 ml of ethanol in order to set a volume ratio of water to ethanol of 2.5: 1.5. A mixture of 57.5 g of γ-aminopropyltriethoxysilane in 57.5 g of ethanol is then added. The mixture is boiled under reflux for a further 10 hours, then the solvent is removed, the particles are washed with ethanol and redispersed in water, the dry matter content being adjusted to 20% by weight. The magnetic particles have terminal amino groups bonded to the surface. The magnetic particles are used further in the form of the aqueous dispersion.
