Es besteht somit ein besonderes Bedürfnis nach einfachen und zuverlässigen Verfahren zur Herstellung von unsymmetrisch substituierten Biphenylen, die sich in einem wirtschaftlichen Gesamtverfahren sinnvoll einsetzen lassen. Hierzu ist ganz besonders eine hohe Selektivität bei hoher Ausbeute gefordert. Die konkret beschriebenen Umsetzungen erfassen weiterhin nicht den Einsatz von Bromarylcarbonsäure. Organomet. Chem. 390 (1990) 389) oder Zinn- (Angew. Chem. 98 (1986) 504) organischen Arylverbindung hergestellt werden. Alle unter dieses Schema fallende Synthesestrategien weisen bis zum letztendlichen Erhalt der gewünschten, nicht derivatisierten Biphenylcarbonsäure eine unökonomisch hohe Stufenzahl auf. Dies ist bedingt durch die zusätzlich zu der eigentlichen Kopplungsreaktion durchzuführende Herstellung des metallorganischen Kopplungspartners - in der Regel aus einer korrespondierenden Grignard-Verbindung - und die nach der Kopplung durchzuführende Generierung der Carboxylfunktionen aus der jeweiligen Vorlauferfunktion.
Auf Grund der einfachen und günstigen Verfügbarkeit der isomeren Brombenzoesäuren ist deren direkte Kopplung mit einer geeigneten Aryl-Grignard-Verbindung (dem regelmäßig am günstigsten zur Verfügung stehenden metallorganischen Reagenz) zum Aufbau von Biphenylcarbonsäuren wünschenswert. Eine solche Umsetzung ist bisher nicht realisiert und beschrieben worden. Ein Grund hierfür liegt sicherlich in dem Umstand, daß bei der Umsetzung einer Carbon- säure mit einem Grignard-Reagenz zunächst eine Deprotonierung der Carbonsäure stattfindet. Zugleich wird Grignard-Reagenz verbraucht und es ist eine Ausfällung der Carbonsäure (jetzt als Halogenmagnesiumsalz gebunden) zu befürchten. Hierbei würden zwei Reaktionspartner der erwünschten Reaktion in der vorgeschalteten schnelleren Übertragung eines Wasserstoffatoms der Carbonsäure auf die Grignard-Verbindung bereits verbraucht. Des weiteren wurde gefunden, daß auch auf anderem Wege hergestellte Salze der Brombenzoesäuren im Sinne des Aufbaues eines Biphenylgerustes und somit unter Bildung des Salzes einer Biphenylcarbonsäure reagieren. Diese Salze können dabei sowohl als isolierte Substanz eingesetzt oder auch in situ hergestellt und anschließend der Palladium katalysierten Kopplungsreaktion unterworfen werden. Wird das Salz einer Carbonsäure als Kopplungskomponente eingesetzt, so kann die Synthese mit einem Äquivalent der Grignard-Verbindung zum vollständigen Umsatz gebracht werden. Die als Katalysator eingesetzte Palladiumverbindung kann in einer molaren Konzentration von
0,01 bis 20 % (bezogen auf die Bromarylcarbonsäureverbindung) sinnvoll eingesetzt werden. Größere Einsatzmengen an Katalysator beeinträchtigen den Verlauf der Reaktion nicht, sind jedoch im Sinne einer ökonomischen Synthesedurchführung nicht sinnvoll. Größere Einsatzmengen an Cokatalysator beeinträchtigen den Verlauf der Reaktion nicht, sind jedoch im Sinne einer ökonomischen Synthesedurchführung und mit Blick auf die Entsorgung der Syntheserückstände nicht sinnvoll. Die eingesetzte Cokatalysatormenge ist vorzugsweise in der Größenordnung von einem bis zu wenigen mol-%. Im besonderen hat sich die Verwendung von Zinkchlorid als Cokatalysator bewährt. Als Reaktionsmedium eignen sich gegenüber Aryl- Grignard-Verbindungen inerte Lösungsmittel wie Tetrahydrofuran, andere cyclische Ether, acyclische Ether oder Acetale.
