Проведены исследования, в которых изучали влияние теней эритроцитов с инкапсулированными ростовыми факторами на жизнеспособность и пролиферативную активность культуры дермальных фибробластов человека (ДФЧ). С этой целью к питательной ростовой среде (ДМЕМ с антибиотиками и глутамином) добавляли 10% от объема среды теней эритроцитов, включающих в себя смесь телячьей эмбриональной сыворотки и лизатом тромбоцитов с добавлением фактора роста фибробластов. В качестве положительного контроля использовали культуральную среду ДМЕМ с добавлением 10% ТЭС, отрицательный контроль - ДМЕМ без сыворотки. Важнейшей задачей, решаемой с помощью предлагаемого способа, является постепенное высвобождение биологически активных веществ из контейнеров. В способе прототипа показано, что высвобождение антибиотиков из теней эритроцитов происходит постепенно при их внутривенном введении. Для проверки скорости высвобождения факторов роста из теней эритроцитов в условиях in vitro проведены следующие исследования. Тени эритроцитов, полученные описанным ранее способом, инкубировали с раствором 10% альбумина, разведенного 1: 1 дистиллированной водой. It was found that after 24 hours, 54.5% of encapsulated albumin is released from the erythrocyte shadow, 78% after two days, and 98% after three days. Для проверки эффективности предлагаемого способа проведены следующие исследования. Описанным выше способом получали тени эритроцитов с инкапсуляцией в них биологически активных веществ - смеси сыворотки крови и лизата тромбоцитов с добавлением фактора роста фибробластов - опыт 1. Во втором опыте использовали те же тени эритроцитов, в которых инкапсулировали, вместо биологически активных веществ, физиологический раствор. В трех пробирках размещали фибробласты в физиологическом растворе с концентрацией клеток 400 тыс./100 мкл и тщательно перемешивали их с тенями эритроцитов из опыта 1 и опыта 2. В третью пробирку вместо теней эритроцитов помещали смесь фибробластов с физиологическим раствором (контроль). Во все три пробирки добавляли по 1,25 мл раствора фибриногена (25 мг/мл), тщательно перемешивали и добавляли раствор тромбина. Немедленно заливали эти смеси на силиконированные чашки Петри. Проводили прижизненное окрашивание ядер клеток в скаффолде с применением флуорохрома Hoechst 3334 (США) обладающим высокой специфичностью к двухцепочечной молекуле ДНК (длина волны возбуждения 377 нм и длина волны эмиссии 447 нм).
