Description
Foreliggende oppfinnelse vedrorer en fremgangsmåte for fremstilling av et antibiotikum som tilfeldig betegnes antibiotikum A/16686, ved å dyrke en inntil nå ubeskrevet stamme som har blitt karakterisert taksonomisk som en ny stamme av Actinoplanes-slekten.
Antibiotikum A/16686 er et glycopeptid-antibiotikum med basisk karakter som kan danne syreaddisjonssalter. Derfor er også de fysiologisk akseptable syreaddisjonssalter av anti-biotokum A/16686 en del av oppfinnelsen.
Por enkelhets skyld når det gjelder diskusjoner av anvendelighet, benyttes uttrykket "antibiotikum A/16686"
for å referere til et antibiotikum som er valgt fra antibiotikum A/16686 fri base og dets fysiologisk akseptable syre-addis j onssalter.
Antibiotikum A/16686 hemmer in vitro veksten av
visse patogene bakterier og spesielt grampositive bakterier. Videre vil parenteral tilforsel av antibiotikum A/16686 gi
en hoy grad av beskyttelse mot eksperimentelle infeksjoner i mus.
Ifølge foreliggende oppfinnelse er det således tilveiebragt en fremgangsmåte for fremstilling av antibiotikum A/16686 i fri baseform og fysiologisk akseptable syreaddisjonssalter derav, som i form av sitt hydroklorid har de kjennetegn som er angitt i krav 1 og i det nedenstående, og denne fremgangsmåten er kjennetegnet ved at man dyrker stammen Actinoplanes sp. ATCC 33076 i et kulturmedium som inne-
holder assimilerbare kilder av karbon, assimilerbare kilder av nitrogen og uorganiske salter under submerse aerobe betingelser inntil en vesentlig mengde av antibiotikum A/16686 er dannet, og utvinner nevnte antibiotikum.
En kultur av nevnte stamme ble isolert fra en jordprøve oppsamlet i Vaghalbod (India) og ble deponert 30. januar 1979 ved den permanente kultursamling hos ATCC (American Type Culture Collection), 1201 Parklawn Drive, Rockville, Maryland, USA, hvor den har fått aksesjonsnummer ATCC 33076.
De karakteristiske egenskaper ved Actinoplanes sp. ATCC 33076 er gitt i det nedenstående.
Morfologi
Stammen vokser godt på forskjellige media med
en orange farge på substratmycelium. Det frembringer ikke pigment. Luftmycelium er alltid fraværende.
En mikroskopisk undersøkelse av det vegetative
m; celium viser forgrenede hyf^r med en diarr.-it er på ca.
1 yum. Sporangene dannes spredt bare på potetagar og er globose med en meget ujevn overflate og en diameter fra 5,0 - 9,0 ^um. Sporangiell utlosning observeres etter brudd på veggen i sporangium. De subsfæriske sporer er motile (1,0 - 1,5 /um diameter). Analyse av cellevegg-komponenter viser meso-diaminopimelinsyre og sukker-monster av typen D (Lechevalier et al., Chemical compo-sition as a criterium in the classification of Actinb-mycestes. Adv. Applied Microbiology, 14, 1971, Academic Press N.Y.).
Dyrkningsegenskaper

For å fremstille antibiotikum A/16686 dyrkes stammen Actinoplanes sp. ATCC 33076 som nevnt under aerobe betingelser i et vandig næringsmedium som inneholder en assimilerbar karbonkilde, en assimilerbar nitrogenkilde og uorganiske salter. Nevnte kulturmedium kan være et hvilket som helst av et antall næringsmedier som vanligvis benyttes under slik dyrking, men visse media er foretrukne. Således er f.eks. foretrukne karbonkilder glukose, fruktose, mannose, sukrose, o.l. Foretrukne nitrogenkilder er soya-bonnemel, pepton, kj ottekstrakt, gjærekstrakt, trypton, aminosyrer o.l. Blant de uorganiske salter som kan tilfores kulturmediene er de vanlige opploselige salter som er i stand til å gi natrium-, kalium-, jern-, sink-, kobolt-, magnesium-, kalsium-, klorid-, karbonat-, sulfat-, nitrat-
og lignende ioner. Vanligvis fordyrkes stammen som skal frembringe antibiotikum i en rysteflaske, hvoretter kul-turen benyttes til å inokulere krukkefermentere for fremstilling av vesentlige mengder antibiotikum A/16686.
Mediet som benyttes for fordyrkningen, kan være det samme
som det som benyttes for storre fermenteringer, men andre medier kan også benyttes.
Stammen som produserer A/16686, kan dyrkes ved temperaturer mellom 20 og 37°C og fortrinnsvis ved temperaturer mellom 28 og 30°C.
Under fermenteringen kan antibiotikumproduksjonen folges ved å ta prover av væsken eller av ekstrakter av mycelie-faststoffene med hensyn til antibiotisk aktivitet. Organismer man vet er folsomme overfor antibiotikum A/16686, er nyttige for dette formål. En spesielt nyttig prove-organisme er Sarcina lutea. Bioproven utfores fortrinnsvis ved agardiffusjonsmetoden på agarplater. Maksimal antibiotisk aktivitet får man vanligvis mellom den tredje og den femte dag. Det antibiotikum som fremstilles ved fermentering av stammen Actinoplanes sp. ATCC 33076, finnes både i væsken og i myceliemassen. Utvinning av antibiotikum A/16686 kan derfor utfores ved separat ekstraksjon av buljong og mycelium.
Ekstraksjon av myceliemassen utfores fortrinnsvis med metanol, men andre lavere alkoholer og kloroform kan også benyttes. Antibiotikum A/16686 utvinnes som et råprodukt fra det ekstraherende opplosningsmiddel ved i og for seg kjente metoder. På tilsvarende måte blir også buljongen ekstrahert fortrinnsvis med n-butanol, og en ytterligere mengde urenset antibiotikum A/16686 tilveiebringes ved utfelling fra denne opplosning. Rensing av det urensede antibiotikum A/16686 oppnås ved å behandle produktet som utvinnes fra ekstraksjonsoppløsningsmidlene med en blanding av kloroform:vann 4:7:2, utskilling av det oljeaktige produktet som dannes, og overhelling av dette i vann. Denne behandling forårsaker storkning av produktet som utvinnes ved filtrering, og som renses ytterligere ved hjelp av silisiumdioksydgel-kolonnekromatografi mens man eluerer med en blanding av acetonitril:0,01N HC1 1:1. Antibiotikum A/16686 som ifolge fremgangsmåten utvinnes i form av et hydroklorid, avsaltes deretter ved kromatografering på
en kryssbundet dekstrangelkolonne.
Hvert trinn i den ovennevnte rensingsprosedyre overvåkes ved tynnsjiktskromatografi under anvendelse av et passende opplosningsmiddelsystem, såsom basisk opplosnings-system, f.eks. n-propanol:n:butanol:N:ammoniumhydroksyd 2:J>:H (ovre fase) eller metanol: 10% vandig ammoniumacetat: 10% ammoniumhydroksyd 10:9'-1, hvor et eventuelt syre-addisjons salt av antibiotikum A/16686 omdannes til den frie base. Alle de ovennevnte trinn er derfor rettet mot å isolere rent antibiotikum A/16686 kjennetegnet ved en spesiell R^-verdi i det spesielle opplosningsmiddelsystem som benyttes.
Andre rensingsmetoder kan benyttes omfattende konvensjonell ekstraksjon og adsorpsjon. Nevnte alternative fremgangsmåter kan lett settes opp, trinn for trinn,
ved å overvåke rensingen ved tynnsjiktskromatografi, som beskrevet ovenfor. Når man folger t.1.c.-flekken av antibiotikum A/16686 med en spesiell Rf-verdi, vil det således fremgå hvilke operasjoner som bor utfores for å isolere rent antibiotikum A/16686.
Om ønsket kan det rene antibiotikum A/16686-hydroklorid, som man får etter den fremgangsmåte som er nevnt ovenfor, deretter omdannes til den tilsvarende frie base eller til et annet fysiologisk akseptabelt syreaddisjonssalt ved kjente metoder.
Antibiotikum A/16686 er et antimikrobielt middel, og det er spesielt aktivt mot grampositive mikroorganismer. Nærmere bestemt er aktivitetsspekteret av antibiotikum ;;/16686 in vitro oppsummert i f Sigende tabell IV.
Tabell V gjengir resultater av prover nvor antibiotikum A/16686 ble provet på en rekke Staphylococcus aureus, Streptococcus pyogenes, Streptococcus pneumoniae og Streptococcus faecalis-stammer som var klinisk isolert.
Antibiotikum A/16686 har også vist seg å ha en hoy grad av aktivitet in vivo mot forskjellige patogene organismer. Effektiviteten av antibiotikum A/16686 går tydelig frem fra tabell VI hvor man har gjengitt ED^q-verdiene i mus mot tre forskjellige mikroorganismer.
Antibiotikum A/16686 har vist seg å ha et rela-tivt lavt toksisitetsnivå når det ble brukt på provedyr. F.eks. var LDcri-verdien når antibiotikumet ble tilfort intra-50
peritonealt på mus ca. mellom 500 og 750 mg/kg.
Parmasoytiske preparater inneholdende antibiotikum A/16686 for oral, topisk eller parenteral tilforsel, kan fremstilles på i og for seg kjent måte. Eksempler på disse preparatene er beskrevet f.eks. i Remington's Pharmaceutical Sciences 15. utgave, 1975» Mack Publishing Co., Easton, Penn-sylvania} USA. Disse omfatter tabletter, kapsler, pulvere, salver, flytende opplosninger, opplosninger for innsprøyting o.l. Doseringsenheten kan inneholde 0,5 - 99%, og fortrinnsvis 5 ~ 80$ aktiv bestanddel. Den daglige dose kan bestemmes under betraktning av flere faktorer såsom kroppsvekt, den infiserende organisme, graden av infeksjon, perioden og tilforselsmetoden.

Eksempel 1
Fermentering av stammen Actinoplanes sp. ATCC 33076
En kultur av Actinoplanes sp. ATCC 33076 fordyrkes ved å dyrke stammen i en rysteflaskekultur med folgende sammensetning:
Flaskene rystes i ca. 96 timer ved 28 - 30°C
og deretter benyttes forkulturen (1 liter) for å inokulere krukkefermentene, som hver inneholder 10 liter av føl-gende næringsmedium:
Fermenteringssatsene inkuberes aerobisk under omroring ved 28 - 30°C. Ved intervaller undersokes den antibiotiske aktivitet mikrobiologisk ved agardiffusjonsmetoden under anvendelse av Sarcina lutea som prove-organisme. Den maksimale aktivitet får man etter 72 - 120 timers fermentering.
Eksempel 2
Utvinning av antibiotikum A/ 16686
Hel fermenteringsbuljong (170 1) fremstilt som beskrevet i eksempel 1, avkjoles til 10°C og bringes til pH 3>5 ved hjelp av 18% HC1. Den resulterende sure buljong filtreres under anvendelse av et filterhjelpemiddel ("Clarcel Flow-Ma"), og myceliekaken vaskes med vann. Deretter behandles den filtrerte buljong og mycelium hver for seg.
a) Metanol (30 1) benyttes for å ekstrahere myceliemasse som, etter filtrering, igjen ekstraheres med en blanding av
metanol/vann (30 1 metanol pluss 5 1 vann). Det benyttede mycelium kastes, og de to metanolekstraktene konsentreres under vakuum ved en temperatur på lavere enn 40° til et vandig konsentrat (6 liter). Dette vandige konsentrat ekstraheres med tre porsjoner, hver på 10 liter, n-butanol, som kombineres og konsentreres til et lite volum under vakuum.
Dette konsentratet tilsettes petroleumeter, og den resulterende utfelling fraskilles ved dekantering og tilsettes en ytterligere mengde petroleumeter. Utfellingen fraskilles ved filtrering og torkes under vakuum ved værelsestemperatur til 80 g antibiotikum A/16686 som et råmateriale som har en M.I.C. mot S. pneumoniae UC 41 på 0,1 ^ug/ml.
t>) Den filtrerte buljong pluss vaskevæske (165 1) avkjoles til 10°C og justeres til pH 3,5 ved tilsats av kon-sentrert HC1 (2,5 1). Den tilveiebragte opplosning ekstraheres med n-butanol (80 1), og det organiske ekstrakt konsentreres deretter under vakuum ved en temperatur lavere enn 35°C for å gi et butanolkonsentrat (6 1). Dette konsentrat tilsettes til petroleumeter, og den oppnådde utfelling, fraskilt ved dekantering, tilsettes til en ytterligere mengde petroleumeter. Det faste stoff fraskilles ved filtrering og torkes under vakuum ved værelsestemperatur og gir 26,3 g urenset antibiotikum A/16686 med en M.I.C. mot S. pneumoniae UC 41
på 0,8^ug/ml.
Eksempel 3
Rensing av antibiotikum A/ 16686
47,7 g urenset antibiotikum A/16686 fra eksempel
2a) behandles med 1,4 1 kloroform:etanol:vannblanding (4:7:2)
(volumforhold), og det oljeaktige produkt som dannes, fraskilles fra oppløsningen ved dekantering. Ytterligere 70 ml av den ovennevnte blanding tilsettes deretter til det oljeaktige produkt, og separasjonen gjentas. Ved behandling av det oljeaktige produkt med vann (440 ml) storkner det og fraskilles ved sentrifugering og filtrering: a) Det faste stoff, som fraskilles, suspenderes i vann (170 ml), opploses ved tilsats av metanol (400 ml)
og filtreres. Deretter strippes opplbsningsmidlene under vakuum ved å tilsette n-butanol ved en temperatur som aldri er hoyere enn 35°C, og man får et butanolkonsentrat på ca. 50 ml. Ved tilsats av dietyleter (500 ml) dannes en utfelling som fraskilles ved filtrering og torkes under vakuum ved værelsestemperatur og som gir 1,012 g av et nokså rent antibiotikum A/16686 med en M.I.C. på S0pyogenes på 0,025 ^ug/ml. Det nokså rene antibiotikum A/16686 tilveiebragt på denne måten under-kastes deretter folgende rensemetoder: 1,58 g av det ovennevnte antibiotikum A/16686 karakterisert ved en M.I.C. mot S. pyogenes på 0,025 /ug/ml opploses i acetonitril: vann 1:1 og den resulterende opplosning påfores en kolonne som inneholder 430 g "Silicagel 60" (0,06 - 0,2
mm) fremstilt i den samme blanding. Kolonnen utvikles forst under anvendelse av den samme acetonitril-vann-blanding og man samler opp 70 fraksjoner på hver 20 ml hvoretter man benytter acetonitril:N/100 HC1 1:1 og opp-samler ytterligere 290 fraksjoner, hver på 20 ml. Eluering av kolonnen overvåkes ved tynnsjiktkromatografi på
"60 F^^-Silicagel-plater og ved å prove fraksjonene mot Sarcina lutea. Fraksjonene 130 - 265 slås sammen og opp-losningsmidlene strippes under vakuum med n-butanol slik at man får et n-butanolkonsentrat på 20 ml. Dette rest-volum helles over i en stor mengde etyleter og utfellingen som dannes fraskilles ved filtrering og torkes under vakuum ved værelsestemperatur over P2O5»°S man får 1,015 g antibiotikum A/16686.
0,67 g av den ovennevnte substans opploses i 24 ml vann og 76 ml metanol. Den resulterende opplosning påfbres en 3,0 x 62,0 cm kolonne som inneholder 220 g "Sephadex LH-20" fremstilt i metanol:vann 7:3. Kolonnen utvikles med den samme blanding og man samler opp 10 ml
fraksjoner. Fraksjonene 24 - 32 slås sammen og opplos-ningsmidlene strippes under vakuum ved en temperatur lavere enn 35°C med n-butanol slik at man får et restbutanol-volum på ca. 10 ml. Denne opplosning tilsettes til etyleter for å utfelle det onskede rene antibiotikum A/16686. Utfellingen fraskilles ved filtrering, vaskes med etyleter og torkes under vakuum over P2°5 ve<i værelsestemperatur og man får 0,26 g rent antibiotikum A/16686,
b) Filtratet strippes under vakuum ved en temperatur lavere enn 35°C ved tilsats av n-butanol, slik at
man får et n-butanoIkonsentrat på 50 ml. Ved tilsats av petroleumeter dannes en utfelling som fraskilles ved filtrering og torkes under vakuum ved værelsestemperatur, og dette gir 42,9 g av delvis renset antibiotikum A/16686 karakterisert med en M.I.C.-verdi mot S.pyogenes på 0,2 yug/ml. Denne antibiotiske substans suspenderes i vann (2,5 l) og opploses ved tilsats av NaOH IN opp til pH 7. Deretter ekstraheres den tilveiebragte vandige opplosning med tre porsjoner, 5 1 hver, n-butanol og denne organiske fase, etter vasking med vann, konsentreres til 200 ml under vakuum ved en temperatur lavere enn 35°C.
Ved tilsats av dietyleter til konsentratet dannes det en utfelling som frafiltreres og torkes under vakuum ved værelsestemperatur. Det torkede produkt opploses i 160 ml av det ovre lag av en n-propanol:n-butanol: IN ammoniumhydroksydblanding 2:3:4 (volumforhold). Den resulterende opplosning tilfores en 100 cm hby kolonne,
7,5 cm i diameter, som inneholder l,7kg silisiumdioksydgel 60 (0,06-0,2mm) fremstilt i den ovennevnte bland-
ing av opplosningsmidler. Kolonnen utvikles under anvendelse av den samme blanding og man samler opp 300 ml fraksjoner. Eluering av kolonnen overvåkes med tynnsjiktkromatografi. Fraksjonene 21 - 26 slås sammen og strippes under vakuum ved en temperatur lavere enn 35°C med n-butanol slik at man får et n-butanolkonsentrat på 50 ml. Ved tilsats av dietyleter dannes en utfelling som fraskilles ved filtrering, vaskes med dietyleter og torkes under
vakuum over P2°5 vec* værelsestemperatur. E"t utbytte på 1,875 g av nokså rent antibiotikum A/16686 tilveiebringes som renses ytterligere ved å folge den samme metode som er beskrevet under punkt a) ovenfor. ;Antibiotikum A/16686 som hydrokloridet er et hvitt, krystallinsk materiale som er svakt hydroskopisk og som smelter ved 224 - 226°C. Det er meget oppløselig i vann og dimetylformamid, opploselig i metanol, etanol, propanol og butanol, men er uopploselig i etyleter, pe-troleter og benzen. Elementæranalyse av antibiotikum A/l6686-hydroklorid, som på forhånd er torket ved 140°C under en inert atmosfære, angir den folgende prosentvise sammensetning (som et gjennomsnitt av flere analyser): karbon 51,73%, hydrogen 6,34%, nitrogen 9,96%, klor (totalt innhold) 5,84%, klorioner 4,74% og en rest 1%. ;Infrarodt absorbsjonsspektrum i nujol er vist i fig. 1 i de vedheftede tegninger. ;Folgende absorbsjonsmaksima observeres (i cm<->"'"): 3290-3070, 2930 og 2860 (nujol), 1765, 1630, 1510, 1455, 1375 (nujol), 1230, 1175, 1130, 1065, 1030, 1015, 980, ;840 og 820. ;Det utrafiolette absorbsjonsspekteret vises i fig. 2 i de vedheftede tegninger og har folgende absorbsjonsmaksima: ;b) i metanol som inneholder 0,1N HC1 c) i metanol som inneholder 0,1N NaOH d) i metanol som inneholder pH 7,38 buffer ;Ultrafiolett absorbsjonsspektrum ble registrert ;med et Beckmann DK-2 spektrofotometer. ;Antibiotikum A/16686 hydroklorid har en spesifikk rotasjon [ aj^ på + 49,7° (c=0,43% i DMF) ;Antibiotikum A/16686 har folgende karakteristiske reaksjon: ;R.£-verdiene for antibiotikum A/16686 i papirkromatografi fra forskjellige elueringssystemer og S. aureus ATCC 6538 som påvisningsorganisme er gitt i den folgende tabell: ;Rm: 40 cm ;Mengder: 20 ^ug av forbindelsen opplost i vannmetanol ;(2 mg/ml). ;Rf-verdiene av antibiotikum A-16686 i forskjellige tynnsjiktkromatografisystemer er gjengitt i den folgende tabell (betingelsene er angitt nedenfor tabellen). ;Rm: ca. 140 mm ;Mengder: 2,5 /ul av en opplosning (l mg/ml) av forbindelsen i acetonitril-vann 1:1 ;Visualisering: a) bioautografi på agarplater med B. sub-tilis TCC 6633; b) karbonisering ved opp-varming med a-naftol-svovelsyre; c) jod-damp; d) klor-toluidinreagens; e) UV-lys ;ved 254 nm ;1-7 silisiumdiok3ydgel-plater 60F2^i(. visualisering a, e, b, o, d, e ;8,9 - silisiumdioksydgel 60 ^254 silanert; visualisering e, ;10, 11 - cellulose F-plater ; visualisering a. ;Som bestemt ved hoyt effektive kromatografiske fremgangsmåter, består antibiotikum A/16686, isolert og renset som beskrevet i eksempel 3, av en hovedkomponent som utgjor 70 - 80% mens de gjenværende 30 - 20% kan til-skrives minst to mindre komponenter. ;Antibiotikum A/16686-hydroklorid isolert som beskrevet i de forangående eksempler er hydrokloridet av det basiske glycopeptid som inneholder klor som skiller seg fra antibiotika som tilhorer den klassiske glycopep-tidgruppen ved at den inneholder klor og i en rekke av sine aminosyrekomponenter. I virkeligheten antyder aminosyreanalyse av antibiotikum A/16686 etter syrehydrolyse i 6N saltsyre ved 110°C i 6 timer, ved hjelp av en aminosyre-autoanalysator, nærvær av folgende aminosyrer: ornitin (118 /ug/mg =0,58<y>uM/mg), aspartinsyre (29,2 ^ug/mg = 0,22 /uM/mg), treonin (68 <y>ug/mg =0,57/uM/mg), glycin i — oi -"o — , ^ ^ f — -, \—*' i —o/ "-o — > — "-O / , leudin (41 ^ug/mg = 0,31 ^uM/mg) og fenylalanin (42 ^ug/mg. =0,25 /uM/mg).
Ytterligere fire topper ble påvist under anvendelse av kolonnen for noytrale og sure aminosyrer. To av disse fire aminosyrene ble identifisert ved hjelp av gass-kromatografi-massespektroskopi etter omdanning til sine tilsvarende metylester-trifluoracetylderviater og ble gitt folgende strukturer:
Eksempel 4
Isolering av den frie base
Ved behandling av en opplosning av antibiotikum A/16686-hydroklorid (0,05 g) i vann (3 ml) - etanol (8 ml) med 1,2-epoksybutan (2 ml) dannes en utfelling som etter henstand ved lav temperatur 1 dag utskilles ved sentrifugering, vaskes med etanol og torkes over P2O5 under vakuum ved værelsestemperatur noe som gir 0,016 g av den tilsvarende frie base.
Behandling av nevnte antibiotikum A/16686 i form av dens fri base med en farmasoytisk akseptabel uorganisk eller organisk syre forer til dannelse av det tilsvar-
ende syreaddisjonssalt. Farmasoytisk akseptable syrer er ikke-toksiske syrer som passer for dannelsen av terapeutisk nyttige salter som i og for seg er kjent, f.eks. saltsyre, hydrobromsyre, svovelsyre, fosforsyre, salpetersyre, vinsyre, eddiksyre, ravsyre, melkesyre, glutaminsyre eller metansulfonsyre.
Claims (2)
1. Fremgangsmåte for fremstilling av antibiotikum A/16686 i fri baseform og fysiologisk akseptable syreaddisjonssalter derav, som i form av sitt hydroklorid er kjennetegnet ved A) at det er et hvitt krystallinsk materiale som smelter ved .224 - 226°C; B) at det er opploselig i vann i dimetylformamid; opplose-Iig i metanol; etanol, propanol og butanol; men uopploselig i etyleter, petroleumeter og benzen; C) en omtrentlig elementær sammensetning på 51,73% karbon, 6,34% hydrogen, 9,96% nitrogen, 5,84% klor (totalt innhold), 4,74% klorioner og 1% rest; D) et infrarodt absorbsjonsspektrum i nujol med folgende observerbare absorbsjonsmaksima (se fig. 1 i de vedheftede tegninger): 3290 - 3070, 2930 og 2860 (nujol), 1765,
1630, 1510, 1455, 1375 (nujol), 1230, 1175, 1130, 1065,
1010, 1015, 980, 840 og 820 cm"<1>; E) et ultrafiolett absorbsjonsspektrum med folgende ab-sorbs jonsmaksima (se fig. 2 i de vedheftede tegninger): a) i metanol: 232 nm (E^cm = 178) 265 nm (E<1>^ =107) 1 cm b) i metanol som inneholder 0,1N HC1: 231 nm ( E10/ o = 167) 1 cm 270 nm (E<19é> = 96) 1 cm c) i metanol som inneholder 0,1N NaOH: 250 nm (E<1>^ = 232 1 cm 295 nm (skulder) d) i metanol som inneholder pH 7,38 buffer: 231 nm (E<1%> = 167) 1 cm

ATCC 33076 i et kulturmedium som inneholder assimilerbare kilder av karbon, assimilerbare kilder av nitrogen og uorganiske salter under submerse aerobe betingelser inntil en vesentlig mengde av antibiotikum A/16686 er dannet, og utvinner nevnte antibiotikum.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at man utvinner nevnte antibiotikum
ved separat ekstraksjon av næringsvæsken og mycelium med et organisk opplosningsmiddel valgt fra lavere alkanoler og kloroform, behandler den antibiotiske substans utvunnet fra ekstraheringsvæskene med en blanding av kloroform: etanol : vann 4:7:2, heller det oljeaktige produkt som utskilles i vann, fraskiller det storknede produkt og renser det ytterligere ved silisiumdioksydgel-kolonnekromatografi ved eluering med en blanding av acetonitril: 0,01N HC1 1:1 slik at antibiotisk A/l6686-hydroklorid utvinnes som avsaltes ved kromatografi på en kryssbundet dekstrangel-kolonne, og ved at man, om ønsket, omdanner antibiotikum A/16686-hydroklorid videre til den tilsvarende frie base eller til et annet fysiologisk akseptabelt syreaddisjonssalt ved kjente metoder.
