Neovascularisatie Dé uitvinding heeft betrekking op het vergemakkelijken van de groei van bloedvaten,
Het is in de literatuur bekend dat het gewenst kan zijn om in lichamen houders van cellen te implanteren die 5 in een permeabele verbinding staan met de lichamen via openingen van zodanige afmetingen dat daardoor de doorgang van elementen die afstoting door het lichaam zouden kunnen veroorzaken wordt voorkomen, zie "Encapsulated Human Islet Transplants in Diabetic Rats",Altman et al. (Vol. XXX Trans Am Soc Artif Intern Organs 10 1984).
We hebben nu gevonden dat neovascularisatie kan worden vergemakkelijkt, zowel ten aanzien van het vergroten van de (bloed)vatdichtheden en wat betreft de snelheid waarmee dergelijke dichtheden worden gecreëerd, door op een afgebonden bloed-15 vat in het dierlijke lichaam een sponsachtig materiaal aan te brengen zoals hierna wordt beschreven.
In voorkeursuitvoeringsvormen omhuift het sponsachtige materiaal een capsule met een permeabele wand waarin cellen die moeten worden gevoed door de op boven beschreven wijze gecreëerde 20 neovasculaire groepering kunnen worden gebracht.
De uitvinding wordt nu toegelicht aan een voorkeursuitvoeringsvorm waarvan een doorsnede is getekend in de figuur.
Deze uitvoeringsvorm heeft de volgende opbouw en werkt op de volgende wijze.
25 In de figuur is met 10 algemeen een samenstel aange geven van spons 12, diffusiekamer 14 en (slag)ader 16.
Spons 12 is een polyurethaan isocyanaat spons.
Spons 12 wordt op de volgende wijze gemaakt. 75 Gram van een oplossing van collageen (3 mg per ml)in 0,012N HC1, zoals in de handel 30 wordt gebracht door Collagen Corporation, Palo Alto,California onder het handelsmerk VITROGEN 100, worden gemengd met 25 g gedestilleerd water en het verkregen mengsel wordt daarna gemengd met 50 g polyurethaanisocyanaatacryl’. copolymeer ("voorpolymeer") zoals onder het handelsmerk Hypol 4000 wordt verkocht door W.R.
35 Grace & Co., Lexington, Massachusetts. Dit mengsel schuimt op en $700097 Γ· *£' - 2 - ? 9 levert binnen 10 minuten een spons op die daarna in stukken 12 van de gewenste grootte en vorm kan worden gesneden.
Spons 12 wordt daarna door en door bevochtigd met een oplossing van fibrinogeen en-' heparine (10 ml van een oplossing 5 van heparine die 1000 eenheden heparine per ml bevat, waarin een pakket van 1 mg fibrinogeen, verkrijgbaar hij Sigma Corporation is opgelost).
Daarna wordt in de spons 12 een vaatje 14 geplaatst bestaande uit een holle vezel van polyurethaanisocyanaat(acrylco-10 polymeer) zoals wordt verkocht door W.R. Grace & Co. onder het handelsmerk AMICON XM-50, dat wordt gekenmerkt door permeabele wanden met een nominale grens voor het molecuulgewicht van 50.000 dalton. De uiteinden van het vaatje 14 worden afgesloten met een vloeibaar polyurethaanisocyanaatmateriaal dat eveneens in de 15 handel wordt gebracht door W.R. Grace & Co. en dat fungeert als een soort lijm ten opzichte van het materiaal van het vaatje en dat als het nat is uithardt.
Spons 12 is in de tekening weergegeven in situ in het dierlijke lichaam. Door de spons heen loopt ook een menselijke of 20 dierlijke (slag) ader 18 die werd losgemaakt en af gebonden met de af binddraden 20 en daarna door de spons 12 werd getrokken.
De werking is als volgt.
Spons 12 met daarin het lege vaatje 14 wordt door een chirurgische ingreep in een dierlijk lichaam geplaatst met 25 de losgemaakte (slag)ader 18 er.'doorheen.
Binnen zes weken is er vanuit de (slag)ader 18 die uiteraard in vloeistofcontact staat met het dierlijke hart een reeks van vaten gegroeid die dan in staat zijn om cellen in de nabijheid goed te voeden. De groei van deze vaten of neovascula-30 risatie, wordt vergemakkelijkt door het gebruikte collageen, hepa rine en fibrinogeen en wordt niet belemmerd door het materiaal van de spons dat geen nadelige invloed heeft op de groei van de -vaten.
Het dier wordt daarna door een chirurgische ingreep 35 weer geopend, één uiteinde 16 van het vaatje wordt geopend, gewenste cellen (bijvoorbeeld pancreascellen)worden er in geplaatst en het uiteinde 16 wordt weer gesloten met de bovengenoemde "lijm”.
De nieuwe reeks van aders levert voeding voor de cellen in het vaatje 14 en de grensporiëngrootte of afsnijporiëngrootte van het 0700097 - 3 - * vaatje 14 verhindert dat lichamen die afstoting., zouden kunnen veroorzaken de cellen in het vaatje 14 bereiken.
Volgens . andere uitvoeringsvormen kunnen gewenste cellen ook in het vaatje 14 worden geplaatst voordat het samenstel 5 met de spons door een chirurgische ingreep cm de ader in het , lichaam wordt aangebracht. Het losgemaakte vat van waaruit de groei optreedt kan ook een ader zijn in plaats van een slagader.
Neovascularisatie van (bloed)vaten kan ook worden gebruikt voor het ondersteunen van bijvoorbeeld huid- ,beender-, 10 trachea-, of tandtransplantatie waarbij geen vaatje 14 nodig is.

Claims (9)
1. Samenstel omvattende een sponsachtig lichaam uit een sponsachtig materiaal dat de groei van bloedvaten in dat materiaal mogelijk maakt en een daarin gemonteerd vaatje dat poriën bezit met zodanige afmetingen dat ze moleculen met een molecuul- 5 gewicht boven een vooraf bepaalde grens niet doorlaten.
2. Combinatie volgens conclusie 1 met het kenmerk dat het sponsachtige lichaam is gevormd uit acrylcopolymeer.
3. Saimenstel volgens conclusie 2 met het kenmerk dat het vaatje is gevormd uit acrylcopolymeer.
4. Samenstel volgens een der voorgaande conclusies met het kenmerk dat het sponsachtig lichaam er in opgenomen of er op aangebracht additieven bevat die de groei van bloedvaten kunnen vergemakkelijken.
5. Werkwijze voor het teweeg brengen van neovascula- 15 risatie, met het kenmerk dat men een losgemaakt bloedvat plaatst in of tegen een sponsachtig materiaal waarin de groei van bloedvaten mogelijk is.
6. Werkwijze volgens conclusie 5 met het kenmerk dat het (bloed)vat een slagader is.
7. Werkwijze volgens conclusie 5 of 6 met het kenmerk dat het sponsachtige materiaal een acrylcopolymeer omvat.
8. Werkwijze volgens conclusie 7 met het kenmerk dat het sponsachtige materiaal ingebed in het materiaal of aangebracht op het materiaal één of meer materialen heeft die de groei van 25 (bloed)vaten aanzetten of ondersteunen.
9. Werkwijze volgens conclusies met het kenmerk dat het materiaal of de materialen (die aanzetten tot de groei van bloed (vaten) of die deze groei ondersteunen bestaan uit collageen, heparine en/of fibrinogeen. 30 8700097
