Het gebruik van gepolariseerd licht in stromingscytometrie.
De vinding heeft betrekking op een nieuwe meetmethode die op eenvoudige wijze kan worden toegepast in stromingscytometers of apparaten die op analoge wijze werken. Gedurende de laatste jaren worden stro-mingscytometers gebruikt om biologische cellen of andere 5 deeltjes met een diameter die meestal ligt tussen 0.5 um en 120 um te bestuderen. In de volgende beschrijving zullen we het woord cel gebruiken voor alle biologische cellen en andere deeltjes.
Het principe van een stromingscytometer is dat cellen gesuspendeerd in een vloeistofstroom een sterke lichtbundel passeren bijvoorbeeld een 10 gefocusseerde laserbundel. De hierbij optredende lichtsignalen verschaffen informatie omtrent de celgrootte, celstructuur en (bio-}chemische samenstelling van iedere individuele cel. Veel gemeten optische grootheden zijn (Melamed MR (editor) 1979: Flow Cytometry and sorting, John Wiley & Sons): de intensiteit van het licht verstrooid 15 onder een kleine hoek ten opzichte van de invallende lichtbundel, meestal voorwaartse lichtverstrooiing genoemd, de intensiteit van het licht verstrooid onder een hoek van ongeveer 90° ten opzichte van de invallende lichtbundel, meestal orthogonale of zijwaartse lichtverstrooiing genoemd, de fluorescentie intensiteit van al dan niet 20 gekleurde cellen, de fluorescentie depolarisatie en de lichtabsorptie van al dan niet gekleurde cellen. De voorwaartse lichtverstrooiing geeft informatie over de afmetingen van de cel (Mullaney B.L. and Dean P.N., 1970ï Biophys J. 10: - 772). Het is ook sterk afhankelijk van het verschil in brekingsindex tussen cellen en het externe medium 25 waardoor cellen met een beschadigd membraan een geringere voorwaartse lichtverstrooiing vertonen.
Orthogonale lichtverstrooiing verschaft informatie omtrent de structuur van de cellen (Visser J.W., Van den Engh G.J., van Bekkum D.W., 1980: Blood cells: 391). Indien de breedte van de belichtingsbundel kleiner 30 is dan de diameter van de cellen kan de pulsvorm van het lichtverstrooiingssignaal informatie opleveren omtrent de lengte en de vorm van de cellen.
De mogelijkheid om verschillende cellen van elkaar te onderscheiden kan worden vergroot door gelijktijdig meer van deze parameters te 35 meten. Zo kan men bijvoorbeeld door gelijktijdig de voorwaartse en zijwaartse lichtverstrooiing te meten een onderscheid maken tussen S3 ύ . 3 ο ο * ’ »» -2- menselijke cytotoxische lymfocyten, B en regel lymphocyten, monocyten en granulocyten (Terstappen L.W.M.M., de Grooth B.G., ten Napel C.H.H., van Berkel W. Greve J., 1986: ter publicatie aangeboden; Hoffman R.A., Kung P.C., Hansen W.P. Goldstein G., 198Ο: Proc. Natl. Aca. Scie. 77-5 ^914 - 4917)· Lichtabsorptie in combinatie met lichtverstrooiing wordt gebruikt in stromingscytometers om een onderscheid te maken tussen erythrocyten en thrombocyten, en tussen lymphocyten, monocyten, eosino-fiele granulocyten, basofiele granulocyten en neutrofiele granulocyten. Dit is echter alleen mogelijk nadat de cellen met een 10 serie verschillende kleurstoffen worden gekleurd waardoor de benodigde apparatuur zeer complex is (bijvoorbeeld, H-6OOO of H.l system van de firma Technicon Instruments Corporation, Tarrytown, New York, U.S.A.).
De huidige vinding heeft betrekking op de polarisatiemetingen van elastisch verstrooid licht in stromingscytometers. Een nuttige 15 toepassing van deze vinding kan worden verkregen met de configuratie zoals weergegeven in figuur 1.
Een invallende laserbundel bewegend langs de z-as is lineair gepolariseerd met elektrische veldsterkte Eo parallel aan de x-as. Het laserlicht wordt gefocusseerd in de oorsprong van het 20 coördinatensysteem met behulp van lens LI. In de oorsprong kruist de laserbundel de celstroom van de stromingscytometer welke langs de x-as bewegen.
Qrthogonale lichtverstrooiing wordt verzameld met een objectief (L2) in . een conus rond de negatieve y-as. Met een bundelsplitser (bs) wordt 50$ 25 van het licht op een photomultiplier (PM1) gericht, het resterende licht wordt door een polarisatiefilter (P) geleid en het doorgelaten licht wordt gedetecteerd met een tweede photomultiplier (PM2). Het polarisatiefilter is zodanig geplaatst dat slechts de electrische component langs de z-as van de lichtbundel wordt doorgelaten. Hierdoor 30 detecteert PM1 orthogonale lichtverstrooiing, terwijl met PM2 licht wordt gedetecteerd, dat (voor kleine waarden van de apertuur van het objectief) orthogonaal ten opzichte van de invallende bundel is verstrooid en lineair gepolariseerd is loodrecht op de polarisatierichting van de invallende bundel. De intensiteit die met PM2 wordt gedetecteerd 35 wordt gedepolariseerde orthogonale lichtverstrooiing genoemd.
Door gelijktijdige meting van de piekhoogte van de pulsen gedetecteerd 830-050 * -3- door PM1 en PM2 voor iedere afzonderlijke cel konden wij een duidelijk onderscheid maken tussen eosinofiele granulocyten (E) en neutrofiele granulocyten (N). Dit is weergegeven in een stromingscytometrische density map waarin de piekhoogte van deze pulsen tegen elkaar zijn 5 uitgezet voor iedere afzonderlijke cel (figuur 2). Met de opstelling gegeven in figuur 1 wordt de gedepolariseerde orthogonale lichtverstrooiing voornamelijk veroorzaakt door anisotrope celstructuren en multiple lichtverstrooiingsprocessen in de cel (van der Hulst H.C., I98I: Lightscattering by small particles, Dover Publications Inc., New 10 York). Dit is in overeenstemming met onze waarneming dat eosinofiele granulocyten, welke een groot aantal intracellulaire granulae bezitten, een grotere gedepolariseerde lichtverstrooiing vertonen dan neutrofiele granulocyten (figuur 2).
Meer in het algemeen heeft de vinding betrekking op het gebruik maken 15 van de polarisatietoestand van het licht verstrooid door cellen of andere deeltjes in stromingscytometers. Dit kan worden toegepast voor iedere hoek tussen het verstrooide licht en de invallende lichtbundel, bijvoorbeeld voorwaartse, zijwaartse en achterwaartse lichtverstrooiing. De polarisatierichting van de invallende lichtbundel kan gevari— 20 eerd worden in iedere richting.
Claims (20)
1. Methoden waarbij de gedepolariseerde orthogonale lichtverstrooi-ingsintensiteit wordt gebruikt om onderscheid te maken tussen eosino-fieXe granulocyten en. neutrofiele granulocyten met behulp van stro-mingscytometers of analoge apparatuur. Bij deze methode is het 5 elektrisch veld van de invallende bundel gepolariseerd langs de as van de celstroom, de voortplantingsrichting van de invallende bundel loodrecht op de celstroom en de gedepolariseerde orthogonale lichtver-strooiingsintensiteit is de orthogonale lichtverstrooiingsintensiteit gemeten met een polarisatie element tussen de celstroom en de fotode- 10 tector waarbij het polarisatie element zodanig is geplaatst dat alleen de elektrische component van het licht parallel aan de voortplantingsrichting van de inkomende lichtbundel wordt doorgelaten.
2. Methoden volgens conclusie 1, waarbij echter de gedepolariseerde orthogonale lichtverstrooiingsintensiteit wordt gebruikt om onderscheid 15 te maken tussen eosinofiele granulocyten en andere biologische celtypen.
3· Methoden volgens conclusie 1, waarbij echter de gedepolariseerde orthogonale lichtverstrooiingsintensiteit wordt gebruikt om onderscheid te maken tussen willekeurige celtypes en/of deeltjes.
4. Methoden, volgens conclusie 1 en/of 2 en/of 3 waarbij de gedepola riseerde orthogonale lichtverstrooiingsintensiteit wordt gebruikt in combinatie met de orthogonale lichtverstrooiing.
5. Methoden volgens conclusie 1 waarbij de gedepolariseerde orthogonale lichtverstrooiingsintensiteit wordt gebruikt in combinatie met een 25 of meer andere parameters die kunnen worden gemeten in stromingscytometers.
6. Methoden volgens conclusie 2 waarbij de gedepolariseerde orthogonale lichtverstrooiingsintensiteit wordt gebruikt in combinatie met een of meer andere parameters die kunnen worden gemeten in stromingscytome- 30 ters.

12. Methoden om verschillende typen cellen te kunnen onderscheiden 15 met een stromingscytometer of soortgelijk apparaat waarbij de invallende lichtbundel gepolariseerd is evenwijdig aan de as van de celstroom, de voortplantingsrichting van het invallende licht loodrecht staat op de celstroom en waarbij een polariserend element geplaatst is in de verstrooide bundel voor de detector, dat licht doorlaat met een 20 polarisatierichting die een hoek ongelijk aan 0* met de richting van de celstroom maakt. De intensiteit van de verstrooide bundel achter het polariserend element wordt gemeten. De verstrooide bundel staat loodrecht op de invallende bundel.
13· Methoden volgens conclusie 12 waarbij echter de hoek tussen de 25 invallende lichtbundel en de richting van de celstroom kan variëren tussen 0* en 180*.

19. Methoden volgens conclusies 12 tot en met 17 waarbij echter de 5 gemeten verstrooide lichtintensiteit wordt gebruikt in combinatie met één of meer andere celparameters die gemeten kunnen worden aan cellen in een stromingscytometer of analoog apparatuur.
20. Methoden om gegevens te verkrijgen van biologische cellen of andere deeltjes, waarvoor het essentieel is gebruik te maken van de 10 polarisatietoestand van het invallende licht en/of de polarisatietoestand van het licht dat door deze cellen of deeltjes wordt verstrooid, met behulp van een stromingscytometer of analoge apparatuur. In de stromingscytometer of het analoge apparaat kunnen de volgende geometrische parameters gevarieerd worden. 15 a. De hoek tussen de voortplantingsrichting van de invallende lichtbundel en de richting van de celstroom. b. De hoek tussen de voortplantingsrichting van de verstrooide bundel en de voortplantingsrichting van de invallende bundel. c. De hoek tussen de voortplantingsrichting van de verstrooide bundel 20 de richting van de celstroom. d. De hoek tussen de richting van de elektrische polarisatievector van de invallende lichtbundel en de richting van de celstroom. e.
