Abstract
Se da a conocer un metodo para identificar un producto, que implica las etapas de: (1) asociar con el producto una ligadura del marcador; y (2) detectar la ligadura del marcador en el producto, en un punto posterior en el tiempo, como un recurso para identificar el producto, por el contacto de este producto con una composicion de deteccion. La composicion de deteccion comprende una o mas primeras secuencias de nucleotidos, que codifican uno o mas factores de transcripcion, dependientes de ligaduras, naturales o sinteticos, en que dichos factores comprenden al menos un dominio de union de ligadura, al menos un dominio de union del ADN y al menos un dominio de transactivacion; y una segunda secuencia de nucleotidos, que codifica un gene informador, bajo el control regulador de un elemento de respuesta del receptor o un elemento de respuesta, modificado o sintetico, y un segundo promotor. El metodo puede tambien emplear un co-represor o un co-activador o una secuencia de nucleotidos, que codifica el co-represor o activador. La interaccion entre la ligadura del marcador y el dominio de union de la ligadura es altamente especifica e induce un cambio en la expresion del gene informador, el cambio produce una senal detectable, que identifica la presencia de la ligadura del marcador en el producto. La composicion de deteccion, una linea celular que contiene la primera y segunda secuencias de nucleotidos, los equipos que la usan y los productos marcados con ligaduras especificas del marcador, son utiles en este metodo.
