헤멘테리아 길리아니 거머리로부터 항응고 물질의 추출 및 정제방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 에이취, 길리아니의 조타액선 추출물중의 FXa 억제활성을 나타내는 것이다.
제2도는 DEAE상의 음이온 교환에 의한 가용성 단백질 추출물 4㎎의 크로마토그래피를 나타내는 것이다.
제3도는 헤파린 아가로스 크로마토그래피에 의한 부분정제된 FXa 억제제의 분획화를 나타내는 것이다.

Claims (7)

  1. (a) 타액을 음이온 교환수지상에서 염 농도를 증가시키면서 구배시켜 팩터(Factor) Xa 억제활성을 지닌 용출액 부분을 분리하고; (b) (a)의 분리된 분획물을 헤파린, 팩터 Xa, 또는 헤파린 및 팩터 Xa를 기본으로 하여 친화 크로마토그래피시켜 항응고 및 아미드 분해 활성을 지닌 부분을 분리한 후; (c) (b)의 분리물을 역상고압 액체 크로마토그래피시켜 팩터 xa 억제활성을 지닌 용출액 부분을 분리시킴을 특징으로 하여, 헤멘테리아 길리아니(Haementeria ghilianii)거머리의 타액으로부터 팩터 Xa억제활성을 지닌 단백질성 물질을 분리하는 방법.
  2. 제1항에 있어서, 음이온 교환수지가 DEAE-셀룰로즈인 방법.
  3. 제1항에 있어서, HPLC 수지가 N-숙시미딜에스테르 함유 수지인 방법.
  4. 헤멘테리아 길리아니 거머리의 타액으로부터 제1항 내지 제3항중 어느 한 항의 방법에 따라 분리된, 팩터Xa 억제활성을 지닌 단백질성 물질.
  5. 60pM의 농도에서 팩터 Xa를 최대의 반으로 (half-maximally)억제할 수 있으며, 분자량이 15,000 내지 20,000달톤임을 특징으로 하는, 헤멘테리아 길리아니 거머리의 타액으로부터 분리된 단백질성 물질인, 본질적으로 순수한 팩터 xa 억제물질.
  6. 팩터 Xa함유 배지를 팩터 Xa를 억제하는데 효과적인 제4항의 팩터 Xa 억제활성을 지닌 단백질성 물질 일정량과 접촉시킴을 특징으로 하여, 팩터 Xa를 억제하는 방법.
  7. 제4항의 팩터 Xa억제활성을 지닌 단백질성 물질의 항응고 유효량을 약제학적으로 허용되는 담체와 함께 투여함을 특징으로 하여, 이를 필요로 하는 환자의 혈액응고를 감소시키는 방법.
