PURPOSE: A cosmetic composition containing a Centella asiatica extract exhibiting excellent effects on cell proliferation and collagen and hyaluronic acid synthesis is provided which is excellent in prevention and improvement of skin aging. CONSTITUTION: The cosmetic composition contains 0.0001 to 10.0% by weight of an extract of Centella asiatica, based on the total weight of the cosmetic composition. 인체 정상 섬유아세포를 96-웰 마이크로 플레이트에 접종시키고(2x104세포/웰), 24시간 배양하였다. 배양 후, 배지를 혈청이 함유되지 않은 디엠이엠 배지로 교체하였다. 이때 하기 표 2에 기재된 농도의 병풀 추출물을 배지와 함께 넣고 48시간 배양하였다. 표 2에서 대조군은 병풀 추출물을 첨가하지 않고 배양한 것이다. 병풀 추출물은 실시예 1에서 제조한 것을 사용하였다. 배양 마지막 24시간 전에 아스코르브산 (Ascorbic acid : 50mg/ml)을 첨가하여 콜라겐 합성을 촉진시켰다. 배양 후 각 웰을 세척하고, 혈청이 함유되지 않은 디엠이엠 배지로 교체한 후 24시간 더 배양하였다. 배양 후, 각 웰의 상층액을 모아 프로콜라겐(procollagen) 타입 I C-핍타이드(PICT)양을 키트 (일본 교토 소재의 Takara)를 이용하여 새로 합성된 콜라겐 양으로 측정하였으며, PICP 양은 ng/2x104세포로 환산하였다.Human normal fibroblasts were seeded in 96-well microplates (2 × 10 4 cells / well) and incubated for 24 hours. 인체 정상 섬유아세포를 6-웰 배양 플레이트에 접종시키고(3×104세포/웰), 48시간동안 배양하였다. 이때 배지는 10% 우태아혈청(fetal calf serum), 페니실린, 스트렙토마이신(100 ㎍/㎖) 등이 함유된 디엠이엠 배지를 사용하였다. 배양 후, 여러 농도의 병풀 추출물이 함유된 배지로 교체하고 24시간 배양하였다. 병풀추출물은 실시예 1에서 제조한 것을 사용하였다. 표 5에서 대조군은 병풀 추출물을 첨가하지 않고 배양한 것이다. 배양 후 배지 부분만 모으고 단백질을 제거하기 위하여, 트리클로로아세트산을 최종농도 10%가 되게 넣고, 24시간 4℃에서 보관하였다. 이 용액을 원심분리하여 상층액을 투석하고, NaCl을 처리하여 최종농도 0.04M이 되도록 한 후, 세틸피리디움 클로라이드를 첨가하여 히아루론산을 침전시켰다. 원심분리하여 침전물을 4M NaCl에 용해시키고, 에탄올에 재침전시킨 후, 이 침전물을 정제수에 용해시켰다. 히아루론산을 정량하기 위하여 셀룰로오스 아세테이트 스트립을 이용하여 전기영동하였다[Hata, R. & Nagai, Y.(1972) Anal.
